Background
The skin is the first line of defense of the human body and has a very important barrier function. For infants, the skin structure development is not complete, and the epidermis, the dermis structure and the skin appendages have obvious difference compared with adults; such as: the literature reports that the thicknesses of the horny layer and the epidermis of the skin of the infant are both obviously lower than that of an adult, the thickness of the horny layer is 30 percent thinner than that of the adult, and the thickness of the epidermis is 20 percent thinner; the premature infant has thinner epidermis; therefore, the barrier function of infants is not mature, the skin sensitivity is much higher than that of adults, and the infants are easily influenced by the change of external environment.
In the prior art, in order to prevent adverse effects on the skin of infants caused by dry weather and other environments, skin care of infants is generally required to be carried out by using infant skin care products. However, as a common allergen in skin care products, essence in infant skin care products is easy to cause immune stress reaction of children, and the skin of infants is red and swollen, pruritus and eczema. Meanwhile, the essence is used as an important sensory index of the skin care product, has the functions of covering bad smell and endowing users with better use experience, plays an important role in most infant skin care products, and is an indispensable part of various skin care products.
Therefore, how to reduce the irritation of the essence in the infant cosmetics while keeping the effect of the essence has very important technical significance and industrial prospect. However, in the prior art, in the field of research and development of infant skin care products, research aiming at reducing the irritation of essence is lacked, and an effective and reliable infant skin care product formula system for reducing the irritation of essence is not formed. The infant skin care products in the prior art cannot meet the requirements of nonirritant and aroma and sensory requirements of the formula at the same time, and the products cannot meet the higher requirements of market consumers. Example 1
In this example, the extract was obtained by the bud of Cryptomeria fortunei (available from Jiafa lion, france)Renew) and proves that the cedar bud extract has the obvious effect of reducing the irritation of the essence in the formula of the infant skin care product.
First, the present inventors determined 26 flavor-sensitive ingredients, as shown in table 1.
TABLE 1 fragrance perfume potentially containing 26 allergenic components listed in the International fragrance Association (IFRA)
Then, on the basis, the inventor utilizes the components to carry out experimental study on the irritation of the essence allergen and the experimental study on the effect of the cedar bud extract on reducing the irritation of the essence.
Specifically, the skin experiment is performed using the 3D skin model in this embodiment. The 3D skin model has a multilayer structure, and the displayed function is similar to an in vivo mechanism, so that the response of a human body to stimulation can be predicted on a cellular and molecular level; through the 3D skin model, the efficacy of the product can be confirmed according to the dynamics of biological response, the influence of the product on the homeostasis on the molecular level and the dynamic change of genome response can be monitored, and the parameters which can not be quantified in vivo can be realized in vitro by means of the 3D skin model. In this embodiment, by designing the 3D skin model of the infant, the model is made to be highly close to the natural skin of the infant in structure, form, tissue environment and physiological characteristics, and the physiology and reactivity of the skin of the infant are realized. This skin model is also an ideal model for studying the interaction between the epidermis and the dermis. This example uses this skin model for experiments.
The primary response of the skin of infants to allergens includes inflammation, which represents a defense response of the body to stimuli, manifested by redness, swelling, heat, pain, dysfunction, and the like. The intensity of the inflammatory response actually represents the irritation of the allergen to the skin. Meanwhile, interleukin 6 (IL-6) is an important member of a cytokine network and can be quantified as an inflammatory response marker. An increase in interleukin 6, indicating the presence of inflammation; the degree of increase is closely related to the inflammatory response, with a more pronounced increase in interleukin 6 indicating a more severe inflammation. Therefore, the degree of the skin irritation of the allergen to the infant can be represented by the content of interleukin 6. This example uses the interleukin 6 content for the characterization of the irritability.
Preparation of a test sample: respectively diluting the 26 allergens to 1 mass percent by using olive oil; comparison products: 100% pure olive oil; taking the sample preparation of amyl cinnamaldehyde from Bellca Biotech, guangzhou as an example, amyl cinnamaldehyde was diluted to 1% by mass with olive oil.
Then, in this example, the test sample or the control was applied to the 3D skin model pretreated with or without cedar bud extract, and the interleukin 6 content was measured at a specific time interval. The results are shown in Table 2.
Table 2 comparison of interleukin 6 contents corresponding to 26 kinds of essence allergens and 100% purity olive oil in pretreated 3D skin models of cedar bud extracts of different concentrations;
as can be seen from Table 2, the cedar bud extract can significantly reduce the expression level of interleukin 6 caused by 26 allergens in the essence, reduce the degree of skin inflammation, and reduce the irritation caused by the essence. Moreover, the cost and the efficacy are combined, and 3 percent of the cedar bud extract is the most suitable proportion in the infant Tong Hufu product.
In order to further verify the effect of reducing the irritation of essence by the cedar bud in the skin care products for babies and children, a composition formula containing the cedar bud extract is provided, and a control test is designed; regulations allow the maximum concentration of a single essence allergen in the infant Tong Hufu product to be 0.001%, and experiments are carried out at the maximum concentration in the example; the skin care composition containing essence and cedar bud extract is obtained. Meanwhile, the effect of the cedar bud extracts in different proportions on reducing the skin irritation is tested according to a skin irritation test method in the technical Specification for cosmetic safety.
The test samples and controls are shown in Table 3 below, including sample A, B, C, D.
Table 3:
detecting according to a test method and a judgment standard of technical Specification for cosmetic safety; specifically, the skin was scored according to the following table 4 in combination with the performance of the sample applied to the skin, and graded according to the scoring results and the skin irritation intensity shown in table 5.
TABLE 4 skin irritation response score
Table 5 skin irritation intensity grading
Integral mean value | Strength of |
0—<0.5 | Has no irritation |
0.5—<2.0 | Light irritation |
2.0—<6.0 | Middle irritation |
6.0—<8.0 | Strong irritation |
The skin irritation test results for the above samples are shown in table 6 below:
table 6:
sample (I) | Results of irritation test |
Sample A | Strong irritation |
Sample B | Light irritation |
Sample C | Has no irritation |
Sample D | Has no irritation |
From the above results, it can be seen that the cedar bud extract can significantly reduce the irritation of the formulation and significantly reduce the irritation of the skin care composition under the maximum amount of added essence allergen allowed by the regulations. In particular, formulations containing 3% or more of the cedar bud extract reduce the irritation of the formulation from a strong irritation level to a non-irritating level, compared to formulations without the cedar bud extract.
It should be understood that the above-mentioned embodiments of the present invention are only examples for clearly illustrating the technical solutions of the present invention, and are not intended to limit the specific embodiments of the present invention.
