The invention has the beneficial effects that: the novel auricularia polytricha strain provided by the invention has an excellent blood fat reducing effect, the extract of the novel auricularia polytricha strain can effectively reduce the contents of total cholesterol, triglyceride, low-density lipoprotein cholesterol and high-density lipoprotein cholesterol in serum, and the blood fat is effectively reduced, the effect is superior to that of a plurality of wild auricularia polytricha strains and auricularia polytricha strains which are preserved in the common microorganism center of China Committee for culture Collection of microorganisms and management and have the preservation number of CGMCC No.40184, and the novel auricularia polytricha strain has very excellent application value in preparing health care products for assisting in reducing blood fat or medicines for reducing blood fat.

Detailed Description
The raw materials and equipment used in the invention are known products and are obtained by purchasing commercial products.
Example 1 preparation of aqueous extract of Auricularia polytricha of the invention
Taking 30g of auricularia polytricha MEK1 (preserved in China general microbiological culture Collection center with the preservation number of CGMCC NO.23834), placing in a round-bottomed flask, adding 1000mL of water, standing for 1 hour, heating and refluxing for 3 hours, filtering while hot, washing a filter and filter residues with a small amount of hot water, placing the filter residues and filter paper in the flask, adding 1000mL of water, heating and refluxing for 3 hours, filtering while hot, combining the filtrates, and concentrating under reduced pressure at 60 ℃ to 100mL to obtain the extract.
Example 2 preparation of aqueous extract of Auricularia polytricha of the invention
Taking 30g of auricularia polytricha MEK1 (preserved in China general microbiological culture Collection center with the preservation number of CGMCC NO.23834), placing in a round-bottomed flask, adding 1000mL of water, standing for 1 hour, heating and refluxing for 1 hour, filtering while hot, washing a filter and filter residues with a small amount of hot water, placing the filter residues and filter paper in the flask, adding 1000mL of water, heating and refluxing for 1 hour, filtering while hot, combining the filtrates, and concentrating under reduced pressure at 60 ℃ to 100mL to obtain the extract.
Example 3 preparation of aqueous extract of Auricularia polytricha of the present invention
Taking 30g of auricularia polytricha MEK1 (preserved in China general microbiological culture Collection center with the preservation number of CGMCC NO.23834), placing the auricularia polytricha MEK1 in a round-bottomed flask, adding 1000mL of water, standing for 1 hour, heating and refluxing for 5 hours, filtering while hot, washing a filter and filter residues with a small amount of hot water, placing the filter residues and the filter paper in the flask, adding 1000mL of water, heating and refluxing for 5 hours, filtering while hot, combining the filtrates, and concentrating under reduced pressure at 60 ℃ to 100mL to obtain the extract.
Comparative example 1 preparation of aqueous extract of Auricularia polytricha
The extract of Auricularia polytricha MEK2 (wild isolated Auricularia polytricha) was prepared in the same manner as in example 1, except that 30g of the extract was used.
Comparative example 2 preparation of aqueous extract of Auricularia polytricha
The extract of Auricularia polytricha MEK3 (wild isolated Auricularia polytricha) was prepared in the same manner as in example 1, except that 30g of the extract was used.
Comparative example 3 preparation of aqueous extract of Auricularia polytricha
An extract was prepared from 30g of auricularia polytricha MEK5 (deposited in China general microbiological culture Collection center, accession number CGMCC NO.40184) by the method of example 1.
The beneficial effects of the present invention are demonstrated by the following experimental examples.
Experimental example 1 Effect of Auricularia polytricha aqueous extract on Mixed type hyperlipidemia animal model
1. Method for producing a composite material
1.1 model feed
The maintenance feed is added with 20.0% of cane sugar, 15% of lard, 1.2% of cholesterol, 0.2% of sodium cholate, and proper amounts of casein, calcium hydrophosphate, stone powder and the like. In addition to crude fat, moisture, crude protein, crude fat, crude fiber, crude ash, calcium, phosphorus, calcium of the model feed: phosphorus is required to meet the national standards for maintaining feed.
1.2 Experimental procedures
Male SD rats (200 ± 20g) were randomly divided into 5 groups, including three dose groups (0.5, 1, 2g of crude drug/kg), a blank control group and a model control group, 10 rats per group.
The test samples (example 1, concentration adjusted to different doses) were orally administered to the three dose groups daily in a volume of 10ml/kg, the blank control group and the model control group were simultaneously administered with the same volume of distilled water, the blank control group was administered with the maintenance feed, and the model control group and the three dose groups were administered with the model feed, and the body weights were regularly weighed. The administration was continued for 30 days. Blood was collected on day 31 without fasting from the tail tip, and serum was centrifuged to determine the levels of TC, TG and LDL-C, HDL-C in the serum.
In the same manner, the drugs of comparative examples 1 to 3 were administered to 10 mice each in 3 dose groups (0.5, 1, 2g crude drug/kg).
2. Results
The results are shown in table 1, after the rats are fed with the model feed for 30 days, the serum concentrations of TC, TG and LDL-C of the rats in the model control group are remarkably increased (p is less than 0.01) compared with that in the blank control group, and the concentration of HDL-C is reduced (p is less than 0.01) compared with that in the blank control group. The 3 dose groups of the extracts of the auricularia polytricha MEK1(CGMCC NO.23834) and MEK4 can obviously reduce the concentrations of TC, TG and LDL-C (p is less than 0.05, p is less than 0.01) in the serum of rats and increase the concentration of HDL-C (p is less than 0.05, p is less than 0.01) in the serum, are dose-dependent and have good blood fat reducing effect. The MEK1(CGMCC NO.23834) has better blood lipid-lowering effect than MEK 4.

Remarking: **: the difference was very significant compared to the blank control group (p < 0.01); and (delta): significant difference compared to model control group (p < 0.05); and delta: compared with the model control group, the difference is very significant (p <0.01)
In conclusion, the new auricularia polytricha strain provided by the invention has an excellent blood fat reducing effect, the extract of the new auricularia polytricha strain can effectively reduce the contents of total cholesterol, triglyceride, low-density lipoprotein cholesterol and high-density lipoprotein cholesterol in serum, and can effectively reduce blood fat, the effect is superior to that of a plurality of wild auricularia polytricha and the auricularia polytricha strain which is preserved in China general microbiological culture Collection center and has the preservation number of CGMCC No.40184, and the new auricularia polytricha strain has very excellent application value in the preparation of health care products for assisting in reducing blood fat or medicines for reducing blood fat.
Claims (10)
1. An Auricularia polytricha strain (Auricularia polytricha) is characterized by being preserved in the China general microbiological culture Collection center of China Committee for culture Collection of microorganisms with the preservation number of CGMCC NO. 23834.
2. An aqueous extract of Auricularia polytricha which is an aqueous extract of the Auricularia polytricha strain of claim 1.
3. The aqueous extract of auricularia polytricha as claimed in claim 2, which is prepared by a process comprising the steps of:
(1) adding water into the auricularia polytricha strain, and heating and refluxing for 1-5 hours;
(2) filtering while the solution is hot to obtain filtrate.
4. The aqueous extract of auricularia polytricha as claimed in claim 3, wherein said method further comprises the steps of:
(3) and (3) adding water into the filter residue obtained by filtering, heating and refluxing for 1-5 hours, filtering while the filter residue is hot to obtain a filtrate, and combining the filtrate with the filtrate obtained in the step (2).
5. Use of the Auricularia polytricha strain or extract thereof of claim 1 in the preparation of a food, health food or pharmaceutical product.
6. A food, health food or pharmaceutical characterized in that it is a food, health food or pharmaceutical preparation prepared by adding a food adjuvant to the powder of the Auricularia polytricha strain of claim 1 and/or its extract as an active ingredient, or a pharmaceutical preparation prepared by adding a pharmaceutically acceptable adjuvant to the food or health food.
7. The food, health food or pharmaceutical product according to claim 6, wherein the active ingredient is an aqueous extract of Auricularia polytricha strain.
8. The food, health food or pharmaceutical product of claim 7, wherein said aqueous extract of the strain of Auricularia polytricha is prepared by a process comprising:
(1) adding water into the auricularia polytricha strain, and heating and refluxing for 1-5 hours;
(2) filtering while the solution is hot to obtain filtrate.
9. The food, health food or pharmaceutical product according to claim 8, wherein the method further comprises the steps of:
(3) and (3) adding water into the filter residue obtained by filtering, heating and refluxing for 1-5 hours, filtering while the filter residue is hot to obtain a filtrate, and combining the filtrate with the filtrate obtained in the step (2).
10. The food, health food or pharmaceutical of claim 6, wherein the health food is a health food for assisting in lowering blood lipid; the medicine is used for reducing blood fat.
