Description

Traditional Chinese medicine mixture and application thereof in preparation of medicine for treating cat type II diabetes
Technical Field
The invention relates to a traditional Chinese medicine mixture for reducing blood sugar, belonging to the field of traditional Chinese medicines.
Background
Diabetes has been well treated and controlled since 1921 when insulin was discovered and successfully used clinically by Banting et al, canada, and more than 40 antidiabetics are now approved for the treatment of diabetes. Most of the existing therapeutic drugs act through a single way, the therapeutic effects are limited, and adverse reactions in the aspects of hypoglycemia, gastrointestinal tract reaction, liver and kidney function damage, anaphylactic reaction, cardiac burden aggravation and the like exist. In addition, due to the short-acting property of insulin, patients need to inject insulin once or twice a day to maintain blood sugar, and multiple injections bring great inconvenience, pain and other discomforts to the patients. At present, type 1 and type 2 diabetes cannot be completely cured, diabetics still have wide market demands on safe and effective diabetes treatment medicines, so that research and development of new antidiabetic medicines are greatly promoted, development of long-acting type 2 diabetes treatment medicines which act on new targets and can avoid side effects of traditional antidiabetic medicines is promoted, and injection times are reduced, so that the hot point of research and development at home and abroad is formed.
Cats as companion animals are raised daily in China, and according to the white paper book in the pet industry in China in 2021, the number of cats in cities and towns in China in 2021 reaches 5806 thousands, and the cats are the largest pets. The incidence rate of the feline type II diabetes is high, and no specific medicine exists at present due to the particularity of the pathogenesis of the feline type II diabetes. The mechanism of diabetes mellitus of human beings and other mammals is relatively complex, and is caused by the damage of metabolism-related organs such as liver, kidney and the like, so that the relative balance of the body to glycogen synthesis and metabolism is disturbed, and the damage of islet B cells is caused. The chronic pancreatitis and the pancreatic injury are easy to induce secondary feline diabetes, because the capacity of insulin secretion of islet cells after the pancreatic injury is reduced, the insulin in blood cannot decompose blood sugar, and after the cats eat high-sugar food, the high-sugar intolerance is easy to cause, so that the cats react on other metabolic organs such as liver, kidney and the like to become vicious circle. Due to the particularity of cat diabetes, most of the traditional Chinese medicines used by human beings have the action sites of liver, kidney and other metabolic organs, and cannot directly act on and repair the pancreas of the primary part, so that the effect on cats is not obvious, and the bottleneck exists in the research and development of cat hypoglycemic drugs, so that a hypoglycemic drug which can be applied to the field of cat diabetes is urgently needed.
The traditional Chinese medicine has various advantages as a hypoglycemic medicament. Firstly, taking the traditional Chinese medicine for reducing blood sugar can be realized by carving more with one arrow: namely reducing blood sugar, reducing weight, blood pressure, blood fat, uric acid and the like.
Secondly, in the blood sugar reducing treatment, when several blood sugar reducing western medicines are used in combination, even a large amount of insulin blood sugar is used, the condition that the control is still poor, the integral coordination effect of the traditional Chinese medicines is exerted, the complex factors causing the blood sugar rise are well controlled, and the blood sugar is likely to drop. Meanwhile, the traditional Chinese medicine can help to reduce insulin resistance and promote the recovery of the pancreatic island function to a certain extent. In addition, if the traditional Chinese medicine is used for conditioning the diabetic patients with large blood sugar fluctuation, the blood sugar is stable, and the occurrence of hypoglycemia can be greatly reduced.
Moreover, for some patients who take western medicines with side effects of reducing blood sugar, the traditional Chinese medicines can relieve the symptoms of the adverse reactions, such as diarrhea, abdominal distension, vomiting and the like.
In addition, the treatment of special diabetes, such as hepatic diabetes, pancreatic diabetes and the like, is complicated, and the blood sugar reducing effect is more obvious if the traditional Chinese medicine is used for treating the special diabetes.
The existing traditional Chinese medicine combined hypoglycemic drugs have no obvious effect of inhibiting blood sugar of cats, and the hypoglycemic effect can be more purposefully produced only by finding the drug which can directly act and repair pancreatic cells and assisting other components with balance and repair effects.

Medulla Junci (stem marrow or whole grass of medulla Junci of Juncus of Juncaceae) named as herba Medicaginis, medulla Junci, and herba Hyperici Japonici. Cutting stems from late summer to autumn, drying in the sun, taking out stem pith, straightening and bundling into small bundles. Is in the shape of a thin cylinder, and has white or light yellow-white surface and thin longitudinal lines. Light weight, soft quality, slight elasticity, easy breaking, white cross section, slight smell and light taste. Rush is slightly cold, sweet and bland in nature and taste; it enters heart, lung, small intestine and bladder meridians. It is produced in the edge of wetland or marsh, and the peeled rush is called "rush" and the un-peeled rush is called "rush". Rush is a traditional Chinese medicine which is commonly used at present, and because the yield is relatively high, the pretreatment process of the medicine is relatively simple, the extraction rate of active ingredients is high, and the cost is low. The essential components of the compound include amino acid compounds, carbohydrate compounds, terpenoid compounds, phenolic compounds, sterol compounds, ester compounds, nobiletin compounds, juncus effusus quinio, taro alkene and the like, and the compound has good use value in application. In the prior art, the traditional Chinese medicine composition is used for treating small cold, clearing heat, relieving exterior syndrome, causing discomfort in throat, mosquito bite and the like.
The effective components of the Chinese herbal medicine are the key of the application of the Chinese herbal medicine, and whether the Chinese herbal medicine can be widely and massively used depends on the types and the content of the effective components, and the Chinese herbal medicine has no medical value and commercial value. The effective components are the key to the use of the Chinese herbal medicines, and not all the components have use values.
The rush can prevent and treat human diseases caused by bacterial infection, respiratory tract infection and external infection. Meanwhile, the medicine can also be used for promoting urination, so that the internal heat is reduced; the main treatment is as follows: gonorrhea; edema; poor urine, bitter urine; jaundice due to damp-heat; discomfort, insomnia; the infant cries at night; laryngopharyngitis; oral ulcer; and (5) wounding. Wherein the brass compounds have antitussive, expectorant, antiasthmatic, and antibacterial activities. Protecting liver, removing liver toxicity, resisting fungi, and treating acute and chronic hepatitis and liver cirrhosis.
The antibacterial action of rush is utilized to treat inflammation in pet, particularly cat pancreas, so that chronic pancreatitis is improved, sterol and lipid compounds contained in rush can repair damage of islet cells caused by inflammation of pancreas, and the activity of the islet cells for secreting insulin is recovered, so that sufficient insulin is released to assist organs such as liver and the like to utilize glucose in blood, reduce blood sugar, and improve various complications caused by diabetes.
Spica Prunellae is cold in nature, and has sweet, pungent and slightly bitter taste, and has effects of clearing liver fire, cooling blood, arresting hemorrhage, resolving hard mass, relieving swelling, eliminating phlegm, relieving cough, and clearing away heat and toxic materials, and is suitable for treating human diseases such as lymphoid tuberculosis, acute mastitis, goiter, metrorrhagia, dizziness, pulmonary tuberculosis, facial distortion, bone and muscle pain, leukorrhagia, acute infectious icterohepatitis and bacillary dysentery. Modern pharmacological research shows that selfheal has the effects of lowering blood pressure and expanding blood vessels, rutin contained in selfheal has the effects of resisting inflammation, reducing vascular permeability, reducing brittleness, reducing liver fat and repairing pancreas, has strong clearing capacity on pancreatic focus cells, can help apoptosis of pancreatic inactive cells and promote growth of new cells, and is favorable for assisting in completing balance of pancreatic function level and realizing normal metabolic pathway, so that the activity of islet cells is restored, and the insulin secretion is increased.
The invention is prepared by weighing 5 parts of kudzuvine root, 10 parts of mulberry leaf, 10 parts of astragalus root, 15 parts of lotus seed, 30 parts of selfheal and 30 parts of rush, then crushing by a crusher, carrying out enzymolysis by cellulase after water dissolving, releasing active ingredients such as polysaccharide, polypeptide, alkalis and the like, filtering, removing impurities, precipitating by ethanol, and drying.
The invention consists of two major components, the first is the components of kudzuvine root, mulberry leaf, astragalus and lotus seed, which can rapidly and directly repair the damage of liver and spleen, promote the synthesis of hepatic glycogen and metabolize glucose in vivo. The second kind is selfheal and rush components, which can radically resist pancreatic inflammation, repair pancreatic injury, promote the recovery activity of islet cells, and normally secrete insulin to metabolize blood sugar. Wherein, one of the two components is not available, if the first component is not available, the blood sugar can not be directly and effectively controlled, and if the second component is not available, the functional injury can not be radically and effectively treated. Clinical data show that the traditional Chinese medicine can obviously reduce fasting blood glucose value of type II diabetic cats after being taken for 4 weeks, and can improve the glucose tolerance of type II diabetic cats after 4 weeks of medicine intervention. Establishment of model for cat type II diabetes
Taking 40 adult cats with the weight of more than 2kg, without limitation on varieties and public and female, firstly determining the fasting blood sugar of the adult cats by a Roche blood sugar monitor and test paper, judging that the adult cats have no diabetes before molding if the adult cats do not reach 11.1mmol/L, feeding high-fat and high-sugar feed every day, wherein the ingredients of the feed are as follows: 55 parts of basic cat food, 10 parts of milk powder, 10 parts of lard oil, 10 parts of fish oil and 15 parts of cholesterol. Cats were fed daily ad libitum and their fasting blood glucose values were measured weekly. The determination standard for successful molding is as follows: the blood glucose values are all higher than 11.1mmol/L after 3 times of continuous measurement. A total of 29 out of 40 cats successfully established a type II diabetes model.
Example 5
Influence of mixed traditional Chinese medicine intervention on fasting blood glucose
Successfully molded cats were divided into 4 groups, and the groups and numbers are shown in table 1. Wherein 1 group of traditional Chinese medicines is the traditional Chinese medicine prepared in the embodiment 1, 2 groups of traditional Chinese medicines are the traditional Chinese medicines prepared in the embodiment 2, and 3 groups of traditional Chinese medicines are the traditional Chinese medicines prepared in the embodiment 3. The administration mode is oral administration, once daily, the blank control group is non-administration group, and three groups of animals still continuously take high fat and high sugar feed during administration period. The fasting blood glucose level was measured weekly for 4 weeks after the dry dosing. The fasting glucose changes for each group of animals are recorded in table 2. As can be seen from the table, after the group of 1 Chinese medicine is administrated for 4 weeks, the fasting blood glucose of 8 cats is reduced, and 6 cats are reduced to be below the threshold value (11.1 mmol/L). After the traditional Chinese medicine 2 groups are administrated for 4 weeks, the fasting blood glucose of 5 cats is reduced, wherein 1 cat is reduced to be below the threshold value, and the rest 3 cats show the increase of the fasting blood glucose. After the traditional Chinese medicine 3 groups are administrated for 4 weeks, the fasting blood glucose of 4 cats is reduced, but the fasting blood glucose of all 4 cats is not reduced to be below the threshold value, and the rest 4 cats show the increase of the fasting blood glucose. The 5 cats in the blank control group were not administered with intervention, and all showed an increase in fasting blood glucose. The change trend of the blood sugar of each group is shown in the attached drawing, and the result proves that the fasting blood sugar of the cats can be reduced after the traditional Chinese medicine group in the embodiment 1 of the scheme is dried. Compared with an experimental group without rush and selfheal, the traditional Chinese medicine composition can only control blood sugar, but cannot reduce blood sugar, so that the traditional Chinese medicine composition cannot have a good treatment effect on diabetes.
