Abstract
本发明公开了一种防治家禽腺、肌胃炎的药物组合物及其制备方法与应用。该药物组合物含有活性成分,所述活性成分由黄芪、蒲公英、白术、黄精、山楂、杜若、茯苓、荔枝草、甘草、马齿苋、鱼腥草、维生素C、异麦芽寡糖制成。该组方诸药合用,协同增效,共具清热解毒、化湿行气、强脾健胃、燥湿止泻之功效。可有效用于防治家禽腺、肌胃炎疾病,具有原料来源广泛、安全、环保,制备工艺简单、生产成本低等诸多优点。

技术领域
本发明涉及兽用中药领域,特别涉及一种防治家禽腺、肌胃炎疾病的药物组合物。
背景技术
腺、肌胃炎疾病病是家禽饲养过程中一种常见消化道疾病,病灶主要集中在腺胃和肌胃,通常为地方流行,一年四季均可发生,高温高湿季节发病率最高,主要呈现慢性发病的特点。养殖场一旦出现该病,造成的损失较大。家禽腺、肌胃炎主要临床表现为病鸡饮食减少,精神沉郁,体重下降,发病中后期病鸡极度消瘦,最后因衰竭而死亡。临床上主要以家禽发育不良、生长受阻、消瘦、衰竭死亡、整齐度差、饲料粪便等为特点,剖解可以看见腺胃肿大如球,腺胃乳头扁平甚至到消失、指压可流出清亮液体,腺胃壁水肿、增厚,腺胃黏膜有溃疡、脱落等主要特征。家禽腺、肌胃炎并不是一种特定的传染病或疾病,是在多因素的作用下导致的一种临床病症,且发病非常普遍,可以造成家禽的死淘率增加、料肉比下降等,给家禽养殖业造成了巨大的经济损失。
由于家禽腺、肌胃炎的发病原因较为复杂,且目前尚无特效药物能治疗。当前,在没有治疗特效药的情况下。中药防控就成了唯一的途径。中药因其成分的复杂性和多样性,可以多机制、多位点地有效防控家禽腺、肌胃炎疾病。治疗家禽腺、肌胃炎中药在畜牧行业拥有比较广阔的市场,我国畜禽产品消费巨大,国家政策也在大力扶持畜牧业的发展。而中药防控家禽腺、肌胃炎药物危害小、成本低,安全环保,保障动物源性食品安全。
发明内容
为了解决上述技术问题,本发明提供一种防治家禽腺、肌胃炎的药物组合物。
所述中药组合物,含有活性成分,所述活性成分由黄芪、蒲公英、白术、黄精、山楂、杜若、茯苓、荔枝草、甘草、马齿苋、鱼腥草、维生素C、异麦芽寡糖制成。
进一步,所述活性成分按重量份计,由黄芪40-60份、蒲公英30-60份、白术20-40份、黄精30-70份、山楂40-80份、杜若50-80份、茯苓20-50份、荔枝草60-90份、甘草40-80份、马齿苋30-60份、鱼腥草20-60份、维生素C 8-16份、异麦芽寡糖10-20份制成。
进一步,所述活性成分按重量份计,由黄芪45-55份、蒲公英36-52份、白术25-35份、黄精40-60份、山楂50-70份、杜若60-74份、茯苓30-40份、荔枝草64-82份、甘草50-70份、马齿苋40-55份、鱼腥草30-50份、维生素C10-14份、异麦芽寡糖12-18份制成。
更进一步,所述活性成分按重量份计,由黄芪50份、蒲公英48份、白术32份、黄精50份、山楂60份、杜若68份、茯苓36份、荔枝草76份、甘草60份、马齿苋48份、鱼腥草44份、维生素C 12份、异麦芽寡糖16份制成。
本发明的另一目的是提供一种防治家禽腺、肌胃炎疾病的药物组合物的制备方法,包括如下步骤:
(1)按重量配比称取各药物原料;
(2)将步骤(1)中称得的各重量中药原药材粗粉碎,先用蒸馏水将药材浸泡0.5h后,煎煮3次,第一次2小时,第二次和第三次各1.0小时,合并滤液并浓缩成稠膏,备用;
(3)将步骤(1)中称取的维生素C、异麦芽寡糖与适量的麦芽糊精或葡萄糖混合均匀再与步骤(2)中的稠膏混合均匀制备成药,即得。
其中,上述制备方法制得的防治家禽腺、肌胃炎疾病的药物组合物的剂型可以是:可溶性粉剂、颗粒剂或软膏。
进一步,所述制备方法制得的防治家禽腺、肌胃炎疾病的药物组合物散剂,其药物粒径小于70微米。
本发明还提供了所述中药组合物在防治家禽腺、肌胃炎疾病中的应用。具体为将所述中药组合物制备成防治家禽腺、肌胃炎疾病的药物。
本发明还提供了所述的防治家禽腺、肌胃炎的药物组合物在用于制备防控家禽腺、肌胃炎疾病饲料方面的应用。

本发明药物组合物中的各药物组分及其用量都是根据中兽医方剂辩证论治理论,通过大量的临床试验反复验证实践得来的。本发明专利处方是结合家禽腺、肌胃炎疾病的发病机理、发病原因、临床症状,治疗原则在中医辨证论治理论指导下,经过大量的临床效果验证得出的精当处方,对家禽腺、肌胃炎疾病有很好的预防和治疗效果。家禽腺、肌胃炎按西医辩证病因复杂,没有定论,可能由霉菌毒素、沙门氏菌、网状内皮等免疫抑制病毒甚至遗传性疾病等引起,是个多病因引起的综合症。西医研究的主要是病因,找不到明确病因,治疗起来就无从下手,效果可见一斑;中医把一年分五季,春生、夏长、长夏化、秋收、冬藏。每个季节不同,环境因素不同,多发病也不同,辩证用药就有区别。中医认为家禽腺肌胃炎主要与消化有关,而消化主要是指脾胃,脾主运化,胃主受纳。脾升胃降,升清降浊,为体内气机运动的枢纽,脾胃配合共同完成食物的消化吸收。脾五行属土,喜燥恶湿,外环境过于潮湿,加之家禽阳弱卫外不足,湿气易由外入侵体内,困于脾,湿为阴邪,损伤脾阳,影响脾的运化功能;脾胃配合,脾升清,胃才能降浊,脾不好,自然影响到胃,饲料吃到胃里,胃气不降,则饲料蓄积在胃里,甚至到嗉囊,久不消化,胃胀反酸,食欲低下,湿浊内生于脾胃,造成肌胃、腺胃肿大损伤、溃疡。脾胃虚弱无力运化水谷则拉饲料便,久之不消化的饲料特别是玉米会损伤肠黏膜,造成肠炎症状,所以肌腺胃不好,饲料消化少。食物在胃里蓄积酸败,排出的粪便自然酸臭,肠道经常消化吸收这些不洁之物,就会湿热内生,所以后期粪便会变得腥臭,粘腻。脾胃为后天气血化生之源,脾胃有病则气血不足,所以病鸡表现腿细爪子白。中兽医治疗家禽腺、肌胃炎以脾胃为中心,五脏同调,最终达到脏腑和谐,气血调畅,阴阳平衡,标本兼治的效果。本发明专利药物健脾促采食,改善消化系统的血液循环,对防治家禽腺、肌胃炎效果显著。中医治疗家禽腺、肌胃炎疾病不是某个单纯的作用与某个单纯的靶点,中医讲究君臣配伍,整体布局。
在本发明专利中,杜若和荔枝草作为君药,其中杜若功效疏风消肿、理气健脾、芳香化浊,解毒中和,主治脾虚湿泻;荔枝草功效清热解毒,凉血散瘀,利水消肿,主治感冒发热,痔疮出血,肾炎水肿,白浊,痢疾,痈肿疮毒,湿疹瘙痒,跌打损伤。黄芪、白术、黄精、山楂作为臣药,其中黄芪功效补气固表、托毒排脓、利尿、生肌,主治气虚乏力、久泻脱肛、自汗、水肿、子宫脱垂、慢性肾炎蛋白尿、糖尿病、疮口;白术功效燥湿利水、止汗、安胎,主治脾虚食少、腹胀泄泻、痰饮眩悸、水肿、自汗、胎动不安;黄精功效补气养阴、健脾、润肺、益肾,主治脾胃气虚、体倦乏力、胃阴不足、口干食少、肺虚燥咳、劳嗽咳血、精血不足、腰膝酸软、内热消渴;山楂功效消食健脾、行气散瘀,主治肉食积滞、胃脘账满、泻痢腹痛、瘀血经闭、产后瘀阻、心腹刺痛、疝气作痛。蒲公英、茯苓、马齿苋、鱼腥草作为佐药,其中蒲公英功效清热解毒、消肿散结、利尿通淋,主治疔疮肿毒、乳痈、瘰疬、目赤、咽痛、肺痈、肠痈、湿热黄疸、热淋涩痛;茯苓功效利水渗湿、健脾、宁心,主治水肿尿少、痰饮眩悸、脾虚食少、便溏泄泻、心神不安、惊悸失眠;马齿苋功效清热解毒、凉血止血、止痢,主治热毒血痢、痈肿疔疮、湿疹、丹毒、便血、痔血、崩漏下血;鱼腥草功效清热解毒、消痈排脓、利尿通淋,主治肺痈吐脓、痰热喘咳、热痢、热淋、痈肿疮毒。甘草作为使药,甘草养阴血,补脾胃。诸药合用,共具清热解毒、化湿行气、强脾健胃、燥湿止泻之功效。维生素C帮助病鸡迅速恢复食欲,修复肠胃黏,异麦芽寡糖能促进肠道内有益菌的增殖,抑制肠道内有害菌及腐败物质的形成,增加维生素的含量,提高机体免疫力,促进食物消化、吸收,维持肠道正常功能,改善腹泻与便秘,增加钙、镁等矿物质的吸收,有利于机体合成B族维生素。

黄芪40份、蒲公英30份、白术20份、黄精30份、山楂40份、杜若50份、茯苓20份、荔枝草60份、甘草40份、马齿苋30份、鱼腥草20份、维生素C 8份、异麦芽寡糖10份。
(2)将步骤(1)中称得的各重量中药原药材粗粉碎,先用蒸馏水将药材浸泡0.5h后,煎煮3次,第一次2小时,第二次和第三次各1.0h,合并滤液并浓缩成稠膏,备用。
(3)将步骤(1)中称取的维生素C和异麦芽寡糖与适量的麦芽糊精和葡萄糖混合均匀再与步骤(2)中的稠膏混合均匀制备成颗粒。
(4)本发明药物使用方法:用于家禽腺、肌胃炎疾病的预防与治疗,混饮:每1吨水添加本发明中药0.5kg,自由饮水,连用7-10日。
实施例2本发明药物制备及使用
(1)称取含有下述重量的原料:
黄芪60份、蒲公英60份、白术40份、黄精70份、山楂80份、杜若80份、茯苓50份、荔枝草90份、甘草80份、马齿苋60份、鱼腥草60份、维生素C16份、异麦芽寡糖20份。按实施例1的制备方法制备和使用。
实施例3本发明药物制备及使用
(1)称取含有下述重量的原料:
黄芪45份、蒲公英36份、白术25份、黄精40份、山楂50份、杜若60份、茯苓30份、荔枝草64份、甘草50份、马齿苋40份、鱼腥草30份、维生素C10份、异麦芽寡糖12份。按实施例1的制备方法制备和使用。
实施例4本发明药物制备及使用
(1)称取含有下述重量的原料:
黄芪55份、蒲公英52份、白术35份、黄精60份、山楂70份、杜若74份、茯苓40份、荔枝草82份、甘草70份、马齿苋55份、鱼腥草50份、维生素C14份、异麦芽寡糖18份。按实施例1的制备方法制备和使用。
实施例5本发明药物制备及使用
(1)称取含有下述重量的原料:
黄芪50份、蒲公英48份、白术32份、黄精50份、山楂60份、杜若68份、茯苓36份、荔枝草76份、甘草60份、马齿苋48份、鱼腥草44份、维生素C12份、异麦芽寡糖16份。按实施例1的制备方法制备和使用。
对比例1
用于治疗鸡腺肌胃炎的中药口服液,按如下重量份组成:黄芪80份、党参60份、白术40份、茯苓60份、当归40份、山楂80份、桂枝40份、神曲60份、麦芽60份、大黄60份、苍术60份、玄明粉80份。
上述用于治疗鸡腺肌胃炎的中药口服液的制备方法如下:
(1)按上述比例称取中药药物,粗粉碎;
(2)将步骤(1)称取的黄芪、党参、白术、茯苓、当归、山楂,加入8~10倍量水,温浸半小时,煎煮提取2次,每次1小时,合并煎液,过滤,浓缩备用;
(3)在步骤(2)提取煎液后的药物残渣中加入桂枝、神曲(包煎)、麦芽、大黄、苍术,加入6~8倍量水,温浸半小时,煎煮提取2次,每次1小时,合并煎液,过滤,浓缩备用;
(4)合并步骤(2)和(3)制备的中药浓缩液,加入步骤(1)中称取的玄明粉,搅拌均匀,充分溶解,再加入乳酸调节中药浓缩液的pH值为3.0~5.0,补充水至每毫升口服液含药材2.4克。
对比例2
一种中药组合物,按如下重量份组成:黄芩13份、黄连8份、黄芪13份、苍术7份、厚朴13份、甘草13份、板蓝根13份、淫羊藿13份、鱼腥草4份、蒲公英7份、桔梗4份;
上述中药组合物的制备方法如下:
按上述比例称取中药药物,混合,粗粉碎;加10倍量水煎煮2次,每次1小时,煎煮液浓缩至浓度为1g/mL(即每1mL相当于原生药1g),得中药提取液;
对比例3
黄芪50份、蒲公英48份、白术32份、黄精50份、山楂60份、杜若68份、茯苓36份、荔枝草76份、甘草60份、马齿苋48份、鱼腥草44份。
(1)按上述比例称取中药药物;
(2)将步骤(1)中称得的各重量中药原药材粗粉碎,先用蒸馏水将药材浸泡0.5h后,煎煮3次,第一次2小时,第二次和第三次各1.0h,合并滤液并浓缩成稠膏,备用.
(3)称取适量的麦芽糊精和葡萄糖混合均匀再与步骤(2)中的稠膏混合均匀制备成颗粒。
为表明本发明药物对家禽腺、肌胃炎疾病的防治效果,对本发明药物进行临床疗效观察试验,以下通过试验例来进一步阐述本发明药物的有益效果。
试验例1
为了考察本发明药物对家禽腺、肌胃炎疾病的防治效果,2021年12月在大北农动保研究院动物实验室做本发明药物对肉鸡腺、肌胃炎治疗效果试验。
1试验使用药品及仪器
1.1试验用品:实施例1-实施例5制备的药物、对比例1药物、对比例2药物、对比例3药物、市场用于治疗家禽腺、肌胃炎疾病的常用药物。
1.2试验动物:8日龄海兰褐肉鸡,购自华北某种鸡场。
2试验方法
2.1模型感染组织液的制备方法
将无菌PBS与确定换腺胃炎的病鸡腺胃按照5:1的比例进行研磨破碎,在反复冻溶4次后,进行差速离心(10000r/min离心10min、12000r/min离心10min)取上清液,分别依次通过0.45μm、0.22μm滤器过滤除菌后-80℃保存备用。
2.2模型感染组织液接种
将EID50=10-5~10-6/0.2mL感染组织液用PBS 10倍稀释后口服分别接种需要感染的鸡群。
2.3鸡腺胃炎动物发病模型的评价
确定鸡发病模型成功的关键是:腺胃轻度肿胀,外表面有斑块;腺胃中度增大,峡部扩张明显;腺胃异常肿大。腺胃组织病理学腺上皮细胞空泡变性坏死,细胞核细胞核固缩,共同在扩张的腺腔内形成坏死碎片;导管上皮细胞增生取代腺上皮细胞;多灶性淋巴细胞浸润等。
2.4试验动物的分组和造模,把新进的6日龄的210只肉鸡平均分为11组,每组20只,饲养2天后,试验1组至试验9组用模型感染组织液口服造模,造模成功后,试验1组-试验5组分别使用实施例1-实施例5制备的药物,按照每吨水添加500g,自由饮水,连续使用7天;试验6组使用对比例1药物,按照每吨水添加1000g对比例1药物,自由饮水,连续使用7天;试验7组使用对比例2药物,按照每吨水添加1000g对比例2药物,自由饮水,连续使用7天;试验8组使用对比例3药物,按照每吨水添加500g,自由饮水,连续使用7天;试验9组使用市场用于治疗家禽腺、肌胃炎疾病的药物穿苦颗粒,按照每吨水添加500g,自由饮水,连续使用7天;试验10组作为阳性对照组,不给任何药物;试验11组作为阴性对照组,不造模,不给药。试验11组与试验1-10组隔离饲养,其他饲养条件基本一致。给完药后第三天,对试验1组-试验11组进行剖解观察,并记录试验结果。
2.5试验结果及分析
表1本发明药物对肉鸡腺、肌胃炎治疗效果的影响
从表1可以看出,试验9组模型组各项指标与试验10组空白对照组相比均有明显差异性,表明腺胃炎动物模型是比较成功的。试验组1-8组肉鸡的治愈率都明显大于阳性对照组,说明本发明药物和对照药物能够显著提高肉鸡腺胃炎的治疗效果,且试验1-5组的治愈率明显高于对比例组和对照药物组,试验5组明显优于其他试验组,对肉鸡腺胃炎疗效作用最好,采用的是实施例5制备的药物,实施例5是本发明的最优处方。
试验例2
为了进一步证明本发明药物对鸡腺胃炎的治疗作用,2021年12月,在吉林某肉鸡场做本发明药物对肉鸡腺肌胃炎、生长性能的影响。
1材料与方法
1.1试验材料本发明药物实施例5制备的药物
1.2试验动物临床诊断为腺胃炎症状且体重相近的30日龄肉鸡两栋
2试验方法
2.1试验分组
临床诊断为腺胃炎症状且体重相近的30日龄肉鸡两栋,试验随机分为2组,每栋1组,两组肉鸡分别隔离饲养,自由采食和饮水。试验组将本发明实施例5制备的药按1%加入到日粮中,自由采食,连续使用10d;对照组饲喂基础日粮。
2.2疗效判定:试验结束后,每组随机取5只鸡,颈部放血屠宰,立即剖开腹腔取腺胃、肌胃组织,固定于10%福尔马林溶液中。通过常规石蜡切片制作,伊红和苏木素(HE)染色,光学显微镜观察肌胃、腺胃的形态,判定本发明药物对肉鸡腺胃炎的治疗效果。
2.3生长性能测定:每只鸡在试验前和试验结束后空腹12h后称重,并记录每天的投料量,计算各组试验肉鸡平均日增重(ADG)、平均日采食量(ADFI)和料肉比(F/G),综合以上数据评判其生长性能。
2.4免疫指标测定:试验结束后,每组随机取5只鸡,用含促凝剂采血管,空腹无菌翅静脉采血4mL,3500r/min离心15min,取血清放置于-4℃冰箱保存备用。采用双抗体夹心法酶联免疫吸附试验(ELISA)测定血清中免疫球蛋白A(IgA)、免疫球蛋白G(IgG)和免疫球蛋白M(IgM)含量。
2.5试验结果及分析
表2本发明药物对肉鸡生产性能的比较
从表2可以看出,使用本发明药物后,日增重和采食量明显增加,料肉比明显降低,说明本发明药物可以提高患腺胃炎疾病的肉鸡的生长性能,降低料肉比。

从表3可以看出,试验组的IgG、IgM、IgA明显高于对照组。说明本发明药物能够明显提高患腺胃炎疾病肉鸡的免疫力。
以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明技术原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (10)
1.一种防治家禽腺、肌胃炎的药物组合物,含有活性成分,其特征在于,所述活性成分由黄芪、蒲公英、白术、黄精、山楂、杜若、茯苓、荔枝草、甘草、马齿苋、鱼腥草、维生素C、异麦芽寡糖制成。
2.根据权利要求1所述的防治家禽腺、肌胃炎的药物组合物,其特征在于,所述活性成分由黄芪40-60份、蒲公英30-60份、白术20-40份、黄精30-70份、山楂40-80份、杜若50-80份、茯苓20-50份、荔枝草60-90份、甘草40-80份、马齿苋30-60份、鱼腥草20-60份、维生素C8-16份、异麦芽寡糖10-20份制成。
3.根据权利要求2所述的防治家禽腺、肌胃炎的药物组合物,其特征在于,所述活性成分由黄芪45-55份、蒲公英36-52份、白术25-35份、黄精40-60份、山楂50-70份、杜若60-74份、茯苓30-40份、荔枝草64-82份、甘草50-70份、马齿苋40-55份、鱼腥草30-50份、维生素C10-14份、异麦芽寡糖12-18份制成。
4.根据权利要求3所述的防治家禽腺、肌胃炎的药物组合物,其特征在于,所述活性成分由黄芪50份、蒲公英48份、白术32份、黄精50份、山楂60份、杜若68份、茯苓36份、荔枝草76份、甘草60份、马齿苋48份、鱼腥草44份、维生素C 12份、异麦芽寡糖16份制成。
5.一种权利要求1-4任一项所述的防治家禽腺、肌胃炎的药物组合物的制备方法,其特征在于,包括如下步骤:
(1)按重量配比称取各药物原料;
(2)将步骤(1)中称得的各重量中药原药材粗粉碎,先用蒸馏水将药材浸泡0.5h后,煎煮3次,第一次2小时,第二次和第三次各1.0小时,合并滤液并浓缩成稠膏,备用;
(3)将步骤(1)中称取的维生素C、异麦芽寡糖与适量的麦芽糊精或葡萄糖混合均匀再与步骤(2)中的稠膏混合均匀制备成药,即得。
6.根据权利要求5所述的防治家禽腺、肌胃炎的药物组合物的制备方法,其特征在于,其制得的剂型包括可溶性粉剂、颗粒剂或软膏。
7.根据权利要求6所述的防治家禽腺、肌胃炎的药物组合物的制备方法,其特征在于,所述可溶性粉剂的药物粒径小于70微米。
8.权利要求1-4任一项所述的防治家禽腺、肌胃炎的药物组合物在用于制备防治家禽腺、肌胃炎疾病药物方面的应用。
9.权利要求1-4任一项所述的防治家禽腺、肌胃炎的药物组合物在用于制备防控家禽腺、肌胃炎疾病饲料方面的应用。
10.含有权利要求1-4任一项所述的防治家禽腺、肌胃炎的药物组合物的饲料。
