an essential oil composition for inhibiting a novel coronavirus comprises 40Wt% of lemon essential oil, 25Wt% of lavender essential oil, 18Wt% of pogostemon cablin essential oil and the balance of peppermint essential oil, wherein the lavender essential oil comprises 5-11Wt% of geraniol, 1-3Wt% of 4-terpineol or terpineol-4-ol, 1-4Wt% of 1, 8-cineole, 4-12Wt% of monoterpene linalool, 2-6Wt% of linalyl acetate and 0.5-4Wt% of borneol, and the pogostemon cablin essential oil comprises 15-25Wt% of pogostenol, 5-7Wt% of alpha-guaiene, 8-10Wt% of beta-guaiene and not less than 5Wt% of eicosene.
Example 8:
preparation of essential oil dilution sample: an equal amount of the essential oil composition of example 1-example 7 was taken, and a sample with a concentration of 35mol/L was labeled as sample 1-7, and the samples 1-7 were diluted at a ratio of 1:80 and emulsified conventionally to obtain an equal amount of emulsified essential oil diluted samples 1-7 for use.
And (3) affinity determination: the affinity of the S (Spike protein) protein of SARS-CoV-2 virus and human angiotensin converting enzyme 2 (ACE-2) is determined by Surface Plasmon Resonance (SPR), the SARS-COV-2 pseudovirus stock solution used in this example was purchased from Nanjing Kingsry Biotech, Inc., and the receptor solution containing human angiotensin converting enzyme 2 (ACE-2) was prepared by the laboratory of the research center of the applicant and derived from the oral and nasal mucosa of human body.
The receptor solution containing human angiotensin converting enzyme 2 (ACE-2) is placed on the surface of the sensor chip, SARS-CoV-2 sample solution with the concentration of 1200nM and the equal volume of distilled water mixture are injected, and the natural affinity curve A is obtained after 600s of reaction.
The receptor solution containing human angiotensin converting enzyme 2 (ACE-2) is placed on the surface of a sensor chip, SARS-CoV-2 sample solution with the concentration of 1200nM and a 75% medical alcohol mixture with the same volume are injected, and a reverse affinity curve B is obtained after reaction for 600 s.
Placing a receptor solution containing human angiotensin converting enzyme 2 (ACE-2) on the surface of a sensor chip, injecting a SARS-CoV-2 sample solution with the concentration of 1200nM and an essential oil diluted sample 1-7 with the same volume, respectively mixing to obtain a mixture, and uniformly reacting for 600s to obtain an affinity curve C-curve I after the essential oil diluted sample 1-7 is added, which is shown in detail in figure 1.
As can be seen from FIG. 1, the binding affinity of the S (Spike protein) protein of SARS-CoV-2 virus and angiotensin-converting enzyme 2 (ACE-2) is very high without the intervention of other substances, as shown in curve A, the maximum resonance signal (RU) is more than 110, but after the sample is diluted by adding 75% medical alcohol and any oil of the invention, the resonance signal (RU) is greatly reduced to below 15, as shown in curve B; it can be seen that the addition of 75% medical alcohol can kill SARS-CoV-2 virus, and because of the problems of mixing time and uniformity, the virus can not be completely killed in a short time, and the SARS-CoV-2 which is not in contact with 75% medical alcohol in time can be continuously combined with human angiotensin converting enzyme 2 (ACE-2) in the solution, so that a very low resonance signal (RU) can be detected.
As can be seen from the graph A and the graph B in FIG. 1, the higher the resonance signal (RU) is at the same concentration, the stronger the binding affinity of the S (Spike protein) protein of SARS-CoV-2 virus to human angiotensin-converting enzyme 2 (ACE-2) is. After the essential oil diluted samples 1-7 are respectively added, because the mixture ratio of the essential oil diluted samples 1-7 is different, the components and the content are different, and the extreme value of the resonance signal (RU) of the generated resonance curve C-I is not uniform, and the reason is caused by the change of the content of the effective component capable of being combined with the S protein to cause the change of the concentration difference, but as can be seen from figure 1, the resonance signal (RU) of the curve C-I is always between 45-80, and is higher than the resonance signal value of the medical alcohol participating effect of 75 percent, but is lower than the resonance signal value of the samples 1-7 without the essential oil diluted samples, which shows that the SARS-CoV-2 virus has good inhibition under the effect of the essential oil with the current concentration.
Thereby verifying the conclusion that the binding affinity of SARS-CoV-2 virus S protein and human angiotensin converting enzyme 2 (ACE-2) is obviously inhibited and hindered, and the reason for the rapid decrease of the binding affinity of SARS-CoV-2 virus S protein and human angiotensin converting enzyme 2 (ACE-2) is two: one is that the S protein epitope is effectively modified, so that effective combination and replication can not be carried out, and pathogenicity is lost; the second is that S protein has been killed and dies. Based on the above two possibilities, both of which are the expected results of the present invention, the goal of inhibiting the transmission/infection of new coronaviruses to new receptors has been fully achieved. The curve result of the Surface Plasmon Resonance (SPR) experiment is enough to confirm that the essential oil provided by the invention contains specific components and active ingredients in a content range, so that the binding affinity of SARS-CoV-2 virus S protein and human angiotensin converting enzyme 2 (ACE-2) can be effectively reduced, the infection capacity of SARS-CoV-2 on human bodies is reduced, and the effect of inhibiting or even killing SARS-CoV-2 is achieved.
It is worth to be noted that, in the qualitative judgment of the Surface Plasmon Resonance (SPR) experiment by adding the above essential oil, only the essential oil composition provided by the present invention has strong inhibitory effect on the binding affinity of the SARS-CoV-2S protein and human angiotensin converting enzyme 2 (ACE-2), but whether the SARS-CoV-2 can be effectively killed and the efficiency, time and proportion of killing can be effectively verified, and the applicant has not performed quantitative demonstration yet.
The above description is only a preferred embodiment of the present application and is not intended to limit the present application, and various modifications and changes may be made to the present application by those skilled in the art. Any modification, equivalent replacement, improvement and the like made within the spirit and principle of the present application shall be included in the protection scope of the present application.

Claims (5)

1. An essential oil composition for inhibiting a novel coronavirus, comprising: the lemon essential oil comprises, by weight, not less than 40% of lemon essential oil, 18-25% of lavender essential oil, 15-18% of pogostemon cablin essential oil and the balance of peppermint essential oil, wherein the limonene content is higher than 30% by weight, the sabinene content is higher than 14% by weight and the citronellal content is higher than 10% by weight.
2. An essential oil composition for inhibiting a novel coronavirus according to claim 1, wherein: the lemon essential oil also comprises 1Wt% of alpha-thujene, 1.2 Wt% of alpha-pinene, 5-8 Wt% of beta-pinene, 1.3-2 Wt% of (E) -beta-ocimene, 0.1-0.2 Wt% of beta-caryophyllene, 0.2Wt% of gamma-terpineol, 0.2Wt% of delta-elemene and 2.5-4 Wt% of geranial.
3. An essential oil composition for inhibiting a novel coronavirus according to claim 1 or 2, wherein: the lavender essential oil contains 5-11Wt% of geraniol, 1-3Wt% of 4-terpineol or terpineol-4-ol, 1-4Wt% of 1, 8-cineole, 4-12Wt% of monoterpene linalool, 2-6Wt% of linalyl acetate and 0.5-4Wt% of borneol.
4. An essential oil composition for inhibiting a novel coronavirus according to claim 3, wherein: the patchouli essential oil contains 15-25Wt% of patchouli alcohol, 5-7Wt% of alpha-guaiene, 8-10Wt% of beta-guaiene and no less than 5Wt% of eicosene.
5. An essential oil composition for inhibiting a novel coronavirus according to claim 4, wherein: the peppermint essential oil comprises 3-6Wt% of menthol, 2-4Wt% of menthone, 0.3-1Wt% of menthyl acetate, 9-12Wt% of 1, 8-cineole and not less than 15Wt% of menthofuran.
