Abstract

猫杯状病毒,涉及一种病毒毒株。本发明猫杯状病毒保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为:CGMCC NO.24374,毒株名称为猫杯状病毒FCV‑H。猫杯状病毒FCV‑H的基因核苷酸序列如SEQ ID NO:1所示。猫杯状病毒FCV‑H株致病能力强,且体外增殖的第20代产物,仍保持强毒株的致病力,毒力不随传代次数的增加而降低。病毒毒价在106 . 78TCID50/mL时,即可造成100%的动物发病,可作为代表性毒株。猫杯状病毒FCV‑H株低剂量(105 . 78TCID50/mL)攻击试验动物,虽然不会导致动物发病,但亦可刺激动物产生高水平的IgG、IgM抗体。

本发明涉及一种病毒毒株。
背景技术
猫杯状病毒(FCV)是一种高度流行的家猫病原体,并且经常与猫疱疹病毒合并感染并引起猫上呼吸道疾病。尽管它最早是从新西兰猫的肠道内容物中分离出来的,但在文献中关于从肠道中鉴定FCV的报道只有零星报道,关于其作为肠道病原体作用的信息仍不清楚,大多数FCV是从采集的猫口腔拭子中分离而来,FCV在世界范围内广泛分布。有人提出,猫、老虎、猎豹、狮子等猫科动物的几乎所有成员都对FCV易感,该病毒也已从狗的粪便中分离出来,这显示FCV在不同动物物种之间表现出种间循环。成年猫和幼龄猫均会感染FCV,一岁以内的幼猫最易感,FCV感染在成年猫中为中度且不会致命,在一些幼猫病例中可能导致肺炎或严重的上呼吸道疾病。在过去十年中,FCV的毒力突变体已被确定为严重和急性毒力全身性疾病(VSD)的原因,这些疾病包括持续高烧、厌食、抑郁、面部和四肢水肿、面部溃疡或脱发、耳廓和脚、肺水肿、胰腺炎、肝坏死并且死亡率较高,这些突变的FCV分离株被称为VS-FCV,关于这种新疾病的报道越来越多。
发明内容
为了更好地研究和预防猫杯状病毒,本发明提供了一株猫杯状病毒。
本发明猫杯状病毒保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为:CGMCC NO.24374,毒株名称为猫杯状病毒FCV-H。
猫杯状病毒FCV-H的基因核苷酸序列如SEQ ID NO:1所示。
猫杯状病毒FCV-H的衣壳蛋白氨基酸序列如SEQ ID NO:3所示。
编码猫杯状病毒FCV-H衣壳蛋白的基因,该基因的核苷酸序列如SEQ ID NO:2中核苷酸序列所示。
本发明猫杯状病毒FCV-H株适于体外CRFK细胞体外增殖,且病毒滴度稳定,F1、F5、F10、F20代的毒价分别为109.29TCID50/mL、109.5TCID50/mL、109.49TCID50/mL、109.78TCID50/mL;各代次无显著差异。
猫杯状病毒FCV-H株致病能力强,且体外增殖的第20代产物,仍保持强毒株的致病力,毒力不随传代次数的增加而降低。病毒毒价在106.78TCID50/mL时,即可造成100%的动物发病,可作为代表性毒株。猫杯状病毒FCV-H株低剂量(105.78TCID50/mL)攻击试验动物,虽然不会导致动物发病,但亦可刺激动物产生高水平的IgG、IgM抗体。表明猫杯状病毒FCV-H毒株具有良好的免疫原性,具备制成疫苗的潜力。
猫杯状病毒FCV-H毒株可用于后续动物感染模型的建立以及新型疫苗的研发。

下面将结合本发明实施例中的附图,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动的前提下所获得的所有其他实施例,都属于本发明保护的范围。
需要说明的是,在不冲突的情况下,本发明中的实施例及实施例中的特征可以相互组合。
具体实施方式一:本实施方式猫杯状病毒保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为:CGMCC NO.24374,毒株名称为猫杯状病毒FCV-H。
本实施方式猫杯状病毒FCV-H于2019年2月在中国农业科学院哈尔滨兽医研究所,由临床发病猫分离获得。经测序,猫杯状病毒FCV-H的基因核苷酸序列如SEQ ID NO:1所示;猫杯状病毒FCV-H的衣壳蛋白氨基酸序列如SEQ ID NO:3所示;编码猫杯状病毒FCV-H衣壳蛋白的基因,该基因的核苷酸序列如SEQ ID NO:2中核苷酸序列所示。
实施例1
猫杯状病毒FCV-H不同代次生长曲线的测定:
初始代次(F0代)FCV-H以1:104的比例接种CRFK细胞,72h后收集细胞培养物,经-20℃反复冻融3次,4000g离心30min,收集上清获得F1代的产物。如此连续培养至F20代。取F1、F5、F10、F20代的产物,经101~1011倍稀释制成病毒液,接入长满单层CRFK细胞的96孔板中,每孔接种100μL病毒液,每个稀释度接种8孔,同时设置未接毒阴性对照,于37℃培养箱中培养72h,每天观察细胞病变情况,记录出现CPE的孔数,根据Reed-Muench法计算病毒滴度(TCID50)。
F1、F5、F10、F20代的产物的毒价分别为109.29TCID50/mL、109.5TCID50/mL、109.49TCID50/mL和109.78TCID50/mL。以感染复数(MOI)为0.01感染CRFK细胞,37℃吸附1h后,弃病毒液,用无血清的1640培养基清洗一遍后更换为含2%FBS的1640培养基,37℃培养箱培养。分别在12h、24h、36h、48h、60h、72h和84h收集培养上清,重复3份,分别测定F1、F5、F10、F20代次病毒在每个时间点培养上清中的病毒含量。各代次猫杯状病毒FCV-H株一步生长曲线如图1所示,4个代次的病毒滴度在第48小时即进入平台期;各个代次之间差异不显著。
实施例2
猫杯状病毒FCV-H攻毒试验:
合格动物的筛选:中华田园猫购自国内宠物交易市场,45~85日龄,体重0.4~0.9kg。采集结膜及口腔拭子,将拭子放入含1mL DMEM培养基中(含100μg/mL青霉素,100μg/mL链霉素),经37℃处理1h后,漩涡振荡混匀,拭子悬液12000g/min离心5min,取上清用0.22μm的滤器过滤后,按照Fine Quick快速病毒DNA/RNA柱式提取试剂盒说明书提取RNA,反转录为cDNA,采用RT-PCR方法检测猫只中的FCV携带情况。采集幼猫的静脉血液,3000r/min离心15min后,吸取血清,按照Feline VacciCheck Antibody Test Kit试剂盒说明书方法检测血清中FCV抗体情况。采用RT-PCR方法对采集的猫结膜及口腔拭子进行检测(扩增174bp的片段),按照Feline VacciCheck Antibody Test Kit试剂盒检测血清中的FCV抗体水平。进而选出FCV抗原、抗体均为阴性的中华田园猫。
将FCV抗原、抗体均为阴性的中华田园猫分为4组,分成高剂量组、中剂量组、低剂量组和阴性对照组,将F20代毒效价为107.78TCID50/mL的猫杯状病毒FCV-H进行稀释,以106.78TCID50/mL、105.78TCID50/mL进行攻毒,分组及攻毒情况见表1,为了防止组间交叉污染,各组分开饲养。

每日早晚喂食和喂水各1次,同时观察实验组和对照组猫的食欲情况、精神状况、结膜及口腔部分的变化,并测量体重和体温(直肠),用GraphPad Prism软件绘制体温、体重的变化曲线;用SPSS Statistics软件对体温、体重变化进行显著性分析。
攻毒后临床症状:高剂量组试验猫在攻毒后第3天体温开始持续升高(≥39.5℃),直到第9天恢复正常,在第4天体温达到峰值,期间精神抑郁,食欲略减,在第6天出现口腔溃疡,第9天指球/掌球上开始溃烂,试验猫存活率为100%,接种后第14天猫的症状减轻,体重较对照组无显著差异。中剂量组试验猫在攻毒后第7天,舌面出现少量溃疡灶,第9天指球/掌球出现溃烂,第3天体温开始升高(≥39.5℃),并持续到第7天,第4天体温达到峰值,观察期间无动物死亡,体重变化不明显。低剂量组试验猫在攻毒后第8天舌尖出现溃疡灶,舌侧溃疡灶红肿,第12天症状减轻,无指球/掌球溃烂症状,体温体重均正常。对照组在观察期间体温体重均正常,舌面无溃疡灶。感染猫杯状病毒FCV-H的猫,口腔溃疡(如图2所示),指球/掌球溃烂(如图3所示),各实验组感染猫杯状病毒FCV-H的猫体温变化如图4所示、体重变化如图5所示。
病毒载量及毒血症的测定:感染后每日采集口腔拭子,用于病毒载量的测定;隔日采集静脉血液300μL/只,取200μL用于检测毒血症,剩余全血经3000r/min离心15min后,吸取血清保存-20℃冰箱。将采集的拭子及血液提取RNA,反转录为cDNA,采用本实验室建立的实时荧光定量PCR方法进行检测,扩增体系:2×SYBR qPCR Master Mix 10μL,FCV-F0.2μL,FCV-R 0.2μL,cDNA 2μL,RNase-free H2O补充至20μL。反应条件如下:95℃30sec;40个循环:95℃10sec,57℃30sec;95℃15sec,60℃1min,95℃15sec。采集的所有样本分别做3个重复孔,同时设置阴性对照。
荧光定量PCR结果显示,高剂量组3只试验猫在攻毒后第1~13天均在口腔拭子中检测到FCV,第14天检测到2只猫带毒,第15天检测到1只猫带毒,第16天均未检测到排毒;中剂量组所有试验猫在接种后第1天可以检测到FCV,并持续排毒到第15天;低剂量组试验猫在攻毒后第5天检测到1只猫排毒,在第6~15天,低剂量组中的2只猫可在口腔拭子中检测到FCV,检测结果见表3。期间试验组的所有猫未有死亡现象,对照组均未检测到FCV。

为了检测感染FCV的试验猫病毒血症存在情况,隔日采集静脉血液用于检测毒血症,荧光定量PCR结果显示,高剂量组试验猫在攻毒后第2天有2只猫可在血液中检测到FCV,第4~8天高剂量组3只试验猫均可在血液中检测到FCV,第10天可在2只猫血液中检测到FCV;中剂量组试验猫在接种后第2天有2只猫可以在血液中检测到FCV,第4~10天3只猫在血液中均检测到FCV,从第12天开始血液中病毒载量逐渐降低;低剂量组其中1只猫自攻毒起至结束未在血液中检测到FCV,而另1只猫在第6~8天可在血液中检测到FCV,结果见表4。结果表明,FCV感染能够引起毒血症状。

实施例3
IgG及IgM抗体水平的检测:
血清中IgG的检测:采用间接ELISA方法检测实施例2中实验组猫(高剂量组试验猫、中剂量组试验猫、低剂量组试验猫)血清中的IgG抗体水平,步骤如下:
(1)计算实验需要的酶标板孔数,同时设置阳性对照、阴性对照以及空白对照,所有样品均做3个重复。
(2)包被:用纯化的猫杯状病毒FCV-H作为包被抗原,猫杯状病毒FCV-H毒效价为107.78TCID50/mL,用pH9.6的碳酸盐包被缓冲液按1:3000稀释猫杯状病毒FCV-H,然后在酶标板的每个微孔中加100μL,空白组只加100μL的碳酸盐包被缓冲液,轻轻混匀后,4℃过夜孵育。
(3)洗涤:将孔内的液体弃去,每孔加100μL的PBST洗涤缓冲液,静置5min后弃去,在吸水纸上拍去多余残留液体,重复3次。
(4)封闭:每个反应孔中加入300μL提前配置好的5%脱脂牛奶,37℃封闭2h。
(5)结束后弃去脱脂奶粉,然后洗涤,同步骤(3)。
(6)加样:将实验组猫血清按1:200进行稀释,猫杯状病毒阳性血清与猫杯状病毒阴性血清也按此稀释度进行稀释,每个反应孔中加入100μL,轻轻混匀后,37℃孵育1h。
(7)洗涤:同步骤(3)。
(8)加酶标抗体:将辣根过氧化物酶标记的山羊抗猫IgG按1:5000进行稀释,向每个反应孔中加入100μL,轻轻混匀后,37℃孵育1h。
(9)洗涤:同步骤(3)
(10)显色:在避光条件下,向各反应孔中加入100μL TMB显色液,37℃孵育20分钟。
(11)终止:显色液孵育完毕,每个反应孔加50μL 2M硫酸终止反应。
(12)测定:在酶标仪上,于450nm波长处测各孔的OD值。
(13)通过测定标记酶的OD450值来检测IgG抗体含量,并用GraphPad Prism软件绘制抗体规律变化曲线图。
其中,猫杯状病毒阳性血清与猫杯状病毒阴性血清均由由哈尔滨兽医研究所保藏和提供。
血清中IgM的检测:采用间接ELISA方法检测实验组猫血清中的IgM抗体水平,107.78TCID50/mL的病毒原液与pH9.6的碳酸盐缓冲液按1:1000进行稀释,包被酶标板;实验组猫血清样本品按1:100进行稀释;辣根过氧化物酶标记的山羊抗猫IgM按1:20000进行稀释,其余环节与IgG的检测相同。
本实施例检测结果显示在攻毒后的第6天,即出现典型临床症状后,感染猫杯状病毒FCV-H的猫IgG滴度呈阳性,并且在感染发生后长时间保持在高水平,这表明试验猫在感染猫杯状病毒FCV-H后6到10天内对猫杯状病毒FCV-H产生了体液免疫反应。在感染的中度阶段显示IgG水平有所增加,在猫恢复正常后IgG达到最高水平,高水平的IgG一直持续到第30天,因此表明记忆免疫水平增加。而IgM水平在感染后第6天开始大幅度产生,这表明IgM产生较快以应对猫杯状病毒FCV-H的早期感染。然后在第14天试验猫开始康复时IgM滴度开始下降,即IgM水平在感染的第一周增加,在第二周达到高峰,之后开始大幅下降,IgG和IgM抗体水平变化结果如图6~8所示。在感染期间IgM和IgG抗体的产生以及康复阶段IgG水平的增加也表明IgM和IgG已经中和了体内的猫杯状病毒FCV-H,并且IgG抗体在抵抗猫杯状病毒FCV-H感染发挥了作用并产生记忆性免疫应答。IgG和IgM抗体的结果说明猫在感染猫杯状病毒FCV-H后有强烈的体液免疫反应。
