s1, loading the crude product containing the gardenia raw material extract into a raw material separation cylinder 2, adding ethyl acetate, methanol and water eluent into an elution cylinder 1 through a liquid inlet pipe 4 attached with an identification sticker, and eluting the gardenia raw material, wherein the ethyl acetate: methanol: the proportion of water is 15-20: 20-3.5: 0.05-0.25, in the elution process, starting a driving motor 801 to drive a linkage shaft 802, so as to drive a raw material separation barrel 2 to rotate, and detecting the crocetin glycoside content in the gardenia by HPLC chromatography after sufficient elution is obtained;
s2, reserving the elution concentrated solution obtained in the step S1 in the elution cylinder 1, taking out the gardenia raw material in the raw material separation cylinder 2 by using the lifting assembly 7, reserving the elution concentrated solution, continuously filling acetonitrile water solution with the concentration of 15-22% into the elution cylinder 1 through the liquid inlet pipe 4, continuously eluting until a secondary concentrated solution is obtained, and detecting the crocetin glycoside content in the gardenia through HPLC chromatography;
s3, taking out the secondary concentrated solution through a drawing tube 6 to a preparation collecting cylinder 9, fusing the secondary concentrated solution with 40-60% methanol water solution under the pressure of 5-15.0 Mpa, removing a small amount of impurities in front and back, and collecting the preparation for promoting the synthesis of the gardenia crocein.
Specifically, the elution pressure in the elution cylinder 1 used in the step S2 is 0.05 to 0.55 Mpa.
Example 2
Pouring the melted culture medium into a sterile plate by adopting a plate coating method to prepare a sterile plate, cooling and solidifying, selecting the gardenia endophytic bacteria strain for culturing, dropwise adding a certain amount of strain suspension on the surface of the plate, uniformly dispersing the bacteria strain to the whole surface of the plate by using a sterile glass coating rod, culturing, and separating and purifying various strains;

Extracting DNA of the endophyte, formulating quality control indexes, adopting a transposase method to segment the DNA and connect the barcodes after meeting the standard, mixing all libraries in proportion, connecting a sequencing joint, and finally accurately quantifying the DNA library by using the Qubit. After library construction was complete, a concentration and volume of DNA library was added to each individual Flow cell and the Flow cells were transferred to a gridios X5 sequencer for real-time single molecule sequencing.
Strains which regulate crocin glycosidyl genes are selected by gene sequencing and comparative genomic analysis. Thereafter, the growth curves of these several strains in gardenia were dynamically monitored. After strain 1 dies down, the number of strain 2 is rapidly reduced through a curve, and then the two strains are found to have a synergistic relationship after being researched, so that a design experiment is obtained, a single strain and double strains are combined in a gardenia plant for experiment, the result is judged by measuring the quality of gardenia fruits and the content of a pigment glycoside medium, after the strain is determined, the bacterial colony is selected and inoculated in a new LB liquid culture medium for shake flask culture, and the amplification culture is carried out.
