Abstract
本发明公开了一种包裹黄芩素的纳米胶束及其应用,其可以显著改善缺血小鼠海马CA1区细胞数目的减少,抑制细胞的死亡,还可以减少海马CA1区星形胶质细胞和小胶质细胞的活化,可以显著抑制由缺血引起的炎症反应。该纳米胶束复合物对中风体外模型的神经细胞的凋亡具有一定的抑制作用。可以用于缺血性脑损伤和/或脑卒的治疗。

本发明涉及纳米医学技术领域,具体地,涉及一种包裹黄芩素的纳米胶束及其应用。
背景技术
脑卒中(Stroke)是以脑部缺血及出血性损伤症状为主要临床表现的神经系统疾病。 由于其高发病率,高死亡率和高致残率,脑卒中已经成为危害人类健康的主要疾病之一, 每年影响数百万人的生命。在临床上可分为缺血性中风或出血性中风两大类,其中缺血性中风是主要的脑中风类型,约占所有中风类型的80~85%,缺血性中风是由于血栓的形成 导致流向大脑的血液减少的情况,从而导致闭塞血管供应的组织梗死,随后一系列导致神 经元细胞死亡和神经炎症的生化反应开始发生。
目前针对中风治疗的手段有限,临床上的主要通过组织型纤溶酶原激活剂(tPA)达到溶 解血栓的效果,从而恢复大脑正常供糖供养。tPA是治疗缺血性脑卒中患者最常用的纤溶 酶原激活剂。急性缺血性脑卒中后tPA溶栓受推荐治疗窗口时间短(症状开始时间<4.5h) 限制。纤溶酶原激活剂在血管内的主要作用是通过促进纤溶酶诱导的闭塞性凝块的溶解来 维持血管的通畅。在内皮细胞基底膜-星形胶质细胞间期,纤溶酶原激活物调节血脑屏障 的通透性,调节突触对突触间隙缺血损伤的反应。急性缺血性脑中风后tPA溶栓治疗受限 于治疗窗口短,因此临床上只有少数脑中风患者能得到有效治疗。
缺血性中风会引发一系列的级联反应,包括胶质细胞的激活,从而促进中枢神经炎症 和损伤。脑缺血后炎症反应影响缺血性损伤的发展,被认为是与病变进展相关的关键因素。 永久性缺血和短暂缺血后再灌注均可诱导脑组织的炎症反应。在脑缺血的几分钟到几小时内,脑内各种不同类型的细胞,如内皮细胞、小胶质细胞、星形胶质细胞和白细胞等被诱 导产生各种炎症因子,例如肿瘤坏死因子TNF-α、白细胞介素IL-1和IL-6等,进一步加速氧化应激介导的神经元死亡。近年来,越来越多的研究显示,由于天然中草药成分的抗炎、抗氧化等功效,对脑中风的神经损伤具有一定的神经保护效果,且不良反应少,能有 效地改善患者的预后。因此,天然神经保护剂具有广阔的应用前景。
然而,天然中草药成分多为小分子结构,要发挥其对缺血性中风的神经保护效果,急 需解决药物穿越血脑屏障的问题。血脑屏障的阻碍也可能是近30年来国内外开发了上千个脑卒中神经保护剂试验失败的一大原因,因此,对天然神经保护剂包装改造对其神经保 护作用的发挥至关重要。
纳米技术是一种新兴的医学治疗方法,是在接近原子大小的尺度上对物质进行处理, 从而产生具有原子、细胞或分子功能的新结构。纳米材料由于其微小的尺寸,具有独特的 物理化学性质,如导电性、强度、耐久性和化学反应活性,并已被用于电子、化妆品和药物。纳米材料的出现也为生物医学应用提供了非凡的机遇,其在医学中的应用被称为纳米医学。随着纳米医学的蓬勃发展,科学家们研究了纳米材料在药物传递、靶向治疗和成像等领域的应用,各种纳米材料已被开发为治疗中枢神经系统疾病的有效工具,且安全性和有效性均有所提高。
聚合物胶束属于两亲性非病毒基因载体聚合物传递体系,由亲水性外部和疏水核心组 成,可以在“有机-水”溶液中自组装形成纳米级(1~200nm)尺寸。大多数情况下,胶束外壳主要是亲水性基团如聚乙二醇,而内核主要是种类繁多的疏水性物质构成。外壳控制体内药代动力学性质,而内核负责药物滞留、稳定性和药物释放特性等。聚合物胶束的独特性在于,这些共聚物结构可以将表面疏水性药物封装在核心,而外壳表面可以根据需要 进行修饰,设计或调整达到期望的功能属性。这一特定的功能吸引了世界范围内的众多科 学研究者去研究这一药物载体。聚合物纳米胶束作为纳米载体,其载体能力相比于纳米颗粒,脂质体和替他纳米载体没有显著的降低。
黄芩素是传统中药黄芩根中的有效药用成分,是一种疏水性的药物。黄芩素是重要的 黄酮类化合物之一,对各种神经毒性均具有很强的神经保护作用。黄芩素的多种药理活性 可以有效预防和治疗脑血管病、神经退行性病变及其他神经系统疾病。另外黄芩具有扩张血管、降压、心肌保护、内皮细胞保护以及抗动脉粥样硬化等心血管保护作用。黄芩素可 通过抑制线粒体氧化应激,对人神经母细胞瘤细胞(SH-SY5Y)具有保护作用。并有研究表 明,黄芩素对大鼠永久性大脑中动脉闭塞的损伤具有保护作用,可显著改善神经功能缺损, 减少脑组织水肿和梗塞体积。这可能为临床治疗神经退行性疾病带来新的希望。
但由于小分子药物在血液循环过程中极易被降解,同时血脑屏障阻碍了98%小分子药 物进入大脑等问题的存在,严重影响了小分子药物对缺血性脑中风的治疗效果。因此,急 需开发一种新的手段提高药物的递送效率。
乳铁蛋白(LF)是一种天然的转铁蛋白,具有抗氧化、免疫刺激、抗炎保护作用,能通 过抑制促炎细胞因子的释放和刺激抗炎细胞因子的产生来抑制炎症反应。进一步研究发 现,乳铁蛋白具有降低溶血产物对培养的神经元的毒性,减轻脑中风造成脑组织的脂质过氧化,促进小胶质细胞吞噬受损红细胞和死亡神经元,促红细胞增殖等作用。因此,乳铁蛋白对脑中风患者具有一定的治疗效果。顺铂(CDDP)负载的Fe3O4/Gd2O3杂化纳米粒子和乳铁蛋白(LF)和RGD二聚体(RGD2)结合的纳米粒,对原位脑肿瘤的治疗中,发现LF接 枝的纳米粒在LF受体的介导能够跨越BBB,进入脑部发挥治疗效果。由于低密度脂蛋白受体在脑中枢神经系统中大量表达。因此,乳铁蛋白可以作为一种很有前途的靶向分子, 广泛用于脑靶向纳米药物载体的构建。
发明内容
本发明的目的是为了克服现有技术的上述不足,提供一种包裹黄芩素的纳米胶束及其 应用。本发明通过构建一种纳米载药系统将天然神经保护剂与纳米医药结合作用的手段, 利用嵌段共聚物mPEG-PPhe胶束自组装包裹疏水药物黄芩素,既能保证药物在血液循环不被完全降解,同时能穿过血脑屏障将药物顺利递送脑部有效释药。由于聚合物胶束本身 不具有靶向血脑屏障的作用,因此乳铁蛋白作为靶向分子,帮助纳米胶束穿越血脑屏障。 本发明创新性通过温控的方式将嵌段聚合物胶束插入靶向分子乳铁蛋白的空腔中,实现纳 米胶束的表面改性,解决载药聚合物胶束穿越血脑屏障的问题。
本发明的第一个目的是提供一种包裹黄芩素的纳米胶束的制备方法、
本发明的第二个目的是提供所述制备方法制备得到的纳米胶束。
本发明的第三个目的是提供所述的纳米胶束在制备治疗缺血性脑损伤和/或脑卒中的 药物中的应用。
为了实现上述目的,本发明是通过以下方案予以实现的:
本发明公开了一种两亲性嵌段聚合物包裹黄芩素(Bai)的LF-PEG-PPhe-Bai纳米胶 束的制备方法,并将其应用于中风后的神经修复。
其制备方法为:本发明采用Fuchs-Farthing方法,以三光气为环合试剂合成苯丙氨酸 活化单体(苯丙氨酸酸酐Phe-NCA),再通过开环聚合反应得到聚苯丙氨酸(PPhe)。 之后在氮气保护下将mPEG10k-NH2与Phe-NCA反应获得两亲性嵌段聚合物;之后在体系 中加入天然活性药物黄芩素(BAI),通过两亲性嵌段共聚物的自组装将BAI包裹在胶束 体系的内部,最后通过加热的方法,将乳铁蛋白LF接枝在纳米胶束的表面。合成的纳米 胶束复合物粒子后,进行各种表征检测,确保粒子的成功。接下来进一步将纳米胶束 LF-PEG-PPhe-BAI作用于神经细胞SH-SY5Y,该复合物能够明显地抑制神经细胞的凋亡。 本发明将纳米胶束体系与靶向分子LF的作用将复合物体系输运到是脑部,并在此基础上将神经治疗中药药物一起输运到细胞内,对脑缺血后的神经细胞达到修复效果。
其中,两亲嵌段共聚物是具有两亲性的嵌段共聚物。嵌段共聚物是指在单一线性分子 中存在两种或两种以上结构不同的链段,可根据需要合成具有特定化学结构、分子量的共 聚物。具有两亲性的嵌段共聚物在溶液中可自组装成特定的超分子有序聚集体——胶束。两亲性嵌段共聚物在水中溶解后可自发形成由亲水性外壳和亲脂性内核组成的高分子胶 束。
本发明首先采用Fuchs-Farthing方法,将L-苯丙氨酸(L-Phe)在无水的环境下与三光气 反应,得到活化的氨基酸酸酐(Phe-NCA);再采用开环聚合的方式,以PEG-NH2为大分子引发物与Phe-NCA活化的氨基酸酸酐反应,得到mPEG-PPhe两亲性的纳米胶束;在此基础上,往胶束体系中加入天然活性中药黄芩素(Bai),通过两亲性嵌段聚合物在水 中自组装的特性,将BAI包裹在胶束体系中;之后通过加热的方法使胶束表面接枝上靶向 分子乳铁蛋白LF;完成纳米胶束LF-PEG-PPhe-Bai的合成。通过傅里叶红外光谱、透射 电镜、Zeta电位、核磁检测等一系列的表征方式对产物的粒子结构及形态进行确认。对复 合材料的生物学效应,,验证纳米胶束复合物对中风体外模型的SH-SY5Y神经细胞的凋 亡抑制作用。
一种包裹黄芩素的纳米胶束的制备方法,包括以下步骤:
S1.采用Fuchs-Farthing法,苯丙氨酸与三光气反应,得到Phe-NCA;
S2.取Phe-NCA和mPEG10k-NH2反应得到嵌段聚合物聚乙二醇-聚苯丙氨酸 mPEG-PPhe;
S3.嵌段聚合物聚乙二醇-聚苯丙氨酸mPEG-PPhe和黄芩素在超声下自组装,得到聚 合物胶束PEG-PPhe-Bai;
S4.聚合物胶束PEG-PPhe-Bai接枝乳铁蛋白,得到纳米胶束LF-PEG-PPhe-Bai。
优选地,步骤S1中,含有苯丙氨酸的有机溶剂溶液加热回流,与含有三光气的有机溶剂溶液混合,继续回流,直至得到澄清溶液作为有机相,用无机相洗涤有机相,保留有 机相,用无水硫酸镁干燥有机相,浓缩后用石油醚析出,纯化。
优选地,步骤S1中,以质量计算,苯丙氨酸和三光气的用量比为4~6:2~4。
更优选地,步骤S1中,以质量计算,苯丙氨酸和三光气的用量比为5:3。
优选地,步骤S2中,氮气保护下,mPEG10k-NH2溶液与Phe-NCA混合,得到澄清溶 液后,与三氯甲烷混合,充分反应后用无水乙醇沉淀,取沉淀,纯化。

进一步优选地,步骤S2中,所述充分反应为35℃条件下反应48h。

与现有技术相比,本发明具有以下有益效果:
本发明成功合成了LF-PEG-PPhe-BAI纳米胶束,并通过粒径电位以及电镜等检测发 现胶束粒径150nm左右,大小均一、稳定性良好;在体外实验中,成功建立体外神经细 胞脑缺血模型,发现最合适的给药剂量后发现LF-PEG-PPhe-BAI纳米胶束显著提高氧糖 剥夺细胞内的抗氧化酶SOD的活性,降低脂质过氧化产物含量,同时降低肿瘤坏死因子的含量来发挥神经保护作用;全脑缺血建立小鼠的缺血模型后,发现LF-PEG-PPhe-BAI 纳米胶束可以显著改善缺血小鼠海马CA1区细胞数目的减少,抑制细胞的死亡,同时也 发现LF-PEG-PPhe-BAI纳米胶束减少海马CA1区星形胶质细胞和小胶质细胞的活化,可 以显著抑制由缺血引起的炎症反应。该纳米胶束复合物对中风体外模型的神经细胞的凋亡 具有一定的抑制作用。可以用于缺血性脑损伤和/或脑卒的治疗,有很好的应用前景。

具体实施方式
下面结合说明书附图及具体实施例对本发明作出进一步地详细阐述,所述实施例只用 于解释本发明,并非用于限定本发明的范围。下述实施例中所使用的试验方法如无特殊说 明,均为常规方法;所使用的材料、试剂等,如无特殊说明,为可从商业途径得到的试剂和材料。
1、细胞株
人神经母细胞瘤细胞(SH-SY5Y细胞系)由暨南大学医学院提供,经本实验室传代培 养。
2、主要试剂
乳铁蛋白(LF)购自源叶生物;胰酶、高糖DMEM培养基、胎牛血清均为Gbcio公 司产品;24孔聚苯乙烯培养基板为美国Corning公司产品;超氧化物歧化酶试剂盒、丙二 醛试剂盒购自碧云天生物公司。
3、仪器
德国LEO公司场发射扫描电镜:LEO 1530VP,Nikon显微镜,日本Olympus公司光学倒置显微镜,Sigma32184高速冷冻离心机,Thermo CO2培养箱,江苏省金坛市医疗仪器 厂78-1磁力搅拌器,HV-85高压灭菌器,无菌操作台,恒温水浴锅等广州科桥实验技术设备有限公司。
实施例1包裹黄芩素的纳米胶束LF-PEG-PPhe-Bai的制备
一、实验方法
1、苯丙氨酸酸酐(Phe-NCA,N-羧基-α-苯丙氨酸-环内酸酐)的合成
反应机理如图1所示。向三口瓶中依次加入10g L-苯丙氨酸和150mL乙酸乙酯,加热回流,再将溶有6.0g三光气的50mL乙酸乙酯溶液缓慢滴加至三口瓶中,继续回流, 直溶液澄清。反应完成后将反应瓶放入冰箱冷冻降温,接着用4℃饱和碳酸氢钠洗涤三次, 4℃饱和食盐水洗涤一次,将上层有机相转移至单口瓶中,加入20g无水硫酸镁干燥,过 滤,将滤液旋蒸浓缩至50mL,往单口瓶中加入200mL石油醚,冷冻加速析出,恢复室 温,过滤得到粗品,粗品用乙酸乙酯和石油醚重结晶,得到8.0g苯丙氨酸酸酐(Phe-NCA) 白色片状晶体,产率为70%。
2、聚乙二醇-聚苯丙氨酸(mPEG-PPhe)的合成
在50mL的反应瓶中加入3.6649g mPEG10k-NH2,加热抽真空除水,在氮气保护下加入4mL DMF将mPEG10k-NH2溶解,再加入3.50g苯丙氨酸酸酐(Phe-NCA),等溶液澄 清后加入40ml三氯甲烷,在35℃条件下反应48h,将反应液缓慢加入大量无水乙醇中沉 淀,离心,依次用无水乙醇洗涤三次,无水乙醚洗涤三次,真空干燥,得到嵌段聚合物聚 乙二醇-聚苯丙氨酸mPEG-PPhe。
3、嵌段聚合物聚乙二醇-聚苯丙氨酸mPEG-PPhe包裹黄芩素
称取20mg嵌段聚合物(mPEG-PPhe)和3mg黄芩素(Bai)溶解于1mL DMSO中 得到混合溶液,在探头式超声作用下逐滴加入到20mL蒸馏水(pH=7.4)至混合溶液中, 自组装成胶束,并继续超声2min,然后用截留分子量为5kDa的透析袋透析除去有机溶 剂。收集透析袋中的胶束溶液,用0.45μm水相滤头过滤除去大的聚集体,之后用截留分 子量5kDa的超滤管浓缩,得到聚合物胶束PEG-PPhe-Bai,于4℃保存备用。
4、复合物表面修饰(LF)
将7mg的乳铁蛋白(LF)溶解在1ml的0.635mg/mL的聚合物胶束PEG-PPhe-Bai溶 液中,加蒸馏水配成10ml的溶液;在65℃油浴中加热10min后,放在-20℃冰箱中5~8 min进行骤冷降温,得到乳铁蛋白修饰的聚合物胶束LF-PEG-PPhe-Bai,之后置于4℃中 冷藏保存。
二、实验结果
图2显示了LF-PEG-PPhe-Bai纳米胶束的合成示意图,以三光气为环合试剂合成苯丙 氨酸活化单体(Phe-NCA),再通过开环聚合反应得到聚苯丙氨酸(PPhe)。之后在氮气 保护下将氨基化的mPEG10k-NH2与Phe-NCA反应获得两亲性嵌段聚合物,之后通过两亲 性嵌段共聚物的自组装将Bai包裹在胶束体系的内部,最后,乳铁蛋白在高温作用下打开 空间结构,聚合物与之混合,PEG-PPhe-Bai聚合物疏水部分插入乳铁蛋白的疏水结构域 内,再通过瞬间骤冷,乳铁蛋白空间结构再次形成,将嵌段聚合物包裹在空间结构空隙中,得到包裹黄芩素的纳米胶束LF-PEG-PPhe-Bai。
实施例2红外光谱(FTIR)检测
一、实验方法
收集实施例1制备得到的Phe-NCA、mPEG-PPhe的干燥样品作为待测样品,加入一定量的KBr,研磨后上机测定。
二、实验结果
如图3所示,分别对Phe-NCA和mPEG-PPhe进行红外光谱检测,在对红外光谱图进行分析时发现,Phe-NCA样品的红外光谱图在3280波长的位置出现酰胺键特有波峰,同 时在1770波长位置出现酯键波峰,表明Phe-NCA被成功合成。同样在对mPEG-PPhe样 品进行红外光谱检测时也在3500波长的位置出现酰胺键特有波峰,并在3297分别出现了 波长的位置出现氨基峰,两种物质特殊化学键的吸收峰,表明成功合成了以上两种物质。
实施例3纳米粒径和Zeta电位检测
用马尔文粒径仪分别测量空白胶束(实施例1制备得到的mPEG-PPhe),包裹黄芩素后的载药胶束(实施例1制备得到的PEG-PPhe-Bai)以及包封乳铁蛋白的载药胶束(实 施例1制备得到的LF-PEG-PPhe-Bai)的粒径大小。
一、实验方法
取2mL稀释到适当的浓度的合成实施例1的产物mPEG-PPhe、PEG-PPhe-Bai和 LF-PEG-pPhe-Bai作为待测样品,通过马尔文粒径仪,检测样品粒径和Zeta电位。
二、实验结果
结果如下图4所示,实施例1制备的PEG-PPhe空白胶束的粒径基本集中在100nm 左右;在自组装包裹黄芩素后,实施例1制备的载药胶束PEG-PPhe-Bai的粒径大约增大 至150nm左右;进一步包裹乳铁蛋白之后,实施例1制备得到的LF-PEG-PPhe-Bai的粒 径主要集中在120~170nm之间,粒径稍有点增大,变化并不明显。
进一步对聚合物胶束的电位进行检测,结果如图5所示,实施例1制备的PEG-PPhe和PEG-PPhe-Bai胶束的带正电荷,电位在27mV左右,而乳铁蛋白带负电荷,电位在-18 mV左右。当实施例1制备的PEG-PPhe-Bai胶束被乳铁蛋白包封之后,电位下降,实施 例1制备的LF-PEG-PPhe-Bai胶束的最终电位为17.41mV,表明胶束体系较稳定。
实施例4透射电镜检测
利用透射电镜对包裹了黄芩素的胶束PEG-PPhe-Bai进行形态特征进行观察。
一、实验方法
取适宜浓度实施例1制备的PEG-PPhe-Bai溶液作为待测样品,吸管吸取溶液,滴一滴溶液于碳网上,自然干燥后上机,检测实施例1制备的PEG-PPhe-Bai纳米胶束的形貌。
二、实验结果
电镜检测结果如下图6所示,实施例1制备的胶束复合物PEG-PPhe-Bai在电镜下呈现较规则的圆形,表面光滑平整,具有良好的分散性,较均匀的粒径分布,大多粒径尺寸 分布在100nm~150nm左右,与实施例3的粒径的数据相吻合,进一步证明结果的可靠 性。
实施例5体外稳定性实验
一、实验方法
配置pH 7.4的磷酸盐(PBS)溶液备用。20mL实施例1制备的LF-PEG-PPhe-Bai 胶束溶液分别加入透析袋(MWCO=5000Da)中,外液中加入1L pH为7.4的PBS缓 冲溶液。将其置于摇床中,37℃,300rmp。在设定的时间间隔,取出2mL溶液。通过 马尔文粒径仪器测定胶束的粒径变化。
二、实验结果
结果如图7所示。在接下来的为期7天的胶束稳定性实验结束后,发现实施例1制备的LF-PEG-PPhe-Bai胶束具有良好的稳定性,在PH=7.4的磷酸盐缓冲液中,胶束的粒径 基本没有发生明显的变化,只有在前5d时粒径稍有增大,但也基本稳定在150nm左右。
实施例6芩素载药率和包封率检测
一、实验方法
分别配置不同浓度的黄芩素溶液,使用紫外分光光度计测定其在最大吸收波长280nm 的紫外吸收光谱,并依据其吸收峰绘制标准曲线,计算曲线的线性拟合方程与拟合度。使 用高速离心机将最终合成的包裹黄芩素的粒子悬液使用高速离心机离心,11000rpm/min, 10℃,30min。取上清真空悬液浓缩后,紫外分光光度计测定其黄芩素的OD值,重复3 次取平均值,并依据其在绘制标准曲线的波长处的吸光值换算黄芩素含量。按以下公式计 算黄芩素的载药率(Drug loading Efficient)和包封率(EncapsulationEfficient):
二、实验结果
根据测定的回归曲线,在对黄芩素的载药量和包封率进行计算发现,结果如图8所示, 黄芩素的载药率为12.32%,包封率为57.5%,载药量和包封率相对较高。
实施例7CCK8检测氧糖剥夺后细胞毒性
利用氧-糖剥夺(oxygen glucose deprivation/OGD)处理SH-SY5Y细胞模拟体外脑缺血 的病理特征,为了确定合适的氧糖剥夺时间,分别检测OGD处理0、2、4、6、8小时后细胞的活性。
一、实验方法
收集对数期SH-SY5Y细胞制成一定浓度的细胞悬液,每孔加入100μL,调整细胞调密度至104每孔,5%CO2,37℃孵育,至细胞单层铺满孔底的60%时,分别设置不同的缺 糖缺氧时间为0、2、4、6、8h,待复糖复氧12h之后,每孔加入10μL CCK8溶液,继 续培养2h后,立即在酶联免疫检测仪OD 450nm处测量各孔的吸光值。确定细胞活力为 60%时的OGD时间为最佳的缺糖缺氧时间,即确定缺血性中风体外模型。
二、实验结果
结果如图9所示,OGD处理4小时后,细胞的存活率为60%左右,而6和8小时后, 细胞的存活率太低,不利于后期的实验研究。因而后续选用OGD处理细胞4小时作为脑 缺血的细胞模型。
实施例8纳米胶束复合物的使用剂量筛选
一、实验方法
为了挑选纳米药物的最佳给药浓度,用含有不同浓度的纳米药物(实施例1制备的LF-PEG-PPhe-Bai胶束)去处理SH-SY5Y细胞24h。通过CCK8检测不同药物浓度处理 下的细胞活性。根据细胞活性确定最佳的体外给药剂量。
首先,将对数生长期的SH-SY5Y细胞以每孔5×103个接种在96孔板里,培养过夜。用PBS洗三遍,每个孔里分别加入200μl含有不同浓度黄芩素(0、2.5、5、10、25、50、 100μM)的DMEM培养基处理细胞24h。通过CCK-8法得到SH-SY5Y细胞在不同条件 下的细胞活性。首先,向上述实验组的96孔板的每个孔里分别加入10μl的CCK-8,在 37℃孵育2h后用酶标仪在450nm的波长下测定每个孔的吸光值,求出细胞的活性。
二、实验结果
如图10所示,当黄芩素的浓度为5μM时,LF-PEG-PPhe-Bai胶束对OGD后SH-SY5Y 细胞的增殖效果最好。随着浓度的增大,纳米胶束毒副作用开始有所体现,因而我们选择 黄芩素浓度为5μM时的胶束浓度作为我们后期体外模型实验的给药剂量。
实施例9细胞匀浆SOD及MDA检测
SOD为生物体系中存在的一种抗氧化金属酶的重要组成成员,它能够催化超氧阴离 子自由基歧化生成氧和过氧化氢,在机体氧化与抗氧化平衡中起到至关重要的作用,与很 多疾病的发生、发展密不可分。MDA是膜脂过氧化最重要的产物之一,影响线粒体呼吸链复合物及线粒体内关键酶的活性,并加剧细胞膜的损伤。本实施例探究实施例1制备得到的LF-PEG-PPhe-Bai纳米胶束对OGD 4h后的SH-SY5Y细胞是否具有抗氧化的效果。
一、实验方法
取对数生长期的SH-SY5Y细胞接板,OGD 4h处理后加入各组药物,分别为对照组(不进行OGD处理+100μL基础培养基),缺血组(OGD处理4h+100μL基础培养基), 黄芩素组(OGD处理4h+含5μM黄芩素的100μL基础培养基)以及纳米胶束组(OGD 处理4h+含5μM实施例1制备得到的LF-PEG-PPhe-Bai纳米胶束100μL基础培养基)于 培养箱中培养24h;吸去培养液,收集细胞进行裂解,检测细胞内SOD以及MDA的含 量。
按照SOD试剂盒说明书进行操作,加入各试剂后,37℃孵育30min后于450nm测 定吸光度值。按照MDA试剂盒的操作要求配制空白管、标准管和各浓度梯度纳米药物组 的测定管,95℃水浴80min,流水冷却,4000rpm离心10min,收集上清液,523nm,1cm光径,蒸馏水调零,测各管的吸光度值。
二、实验结果
结果如图11所示,与对照组相比,缺血组的SOD酶活性显著性降低,而MDA含量 明显增多,对比相同剂量的黄芩素和LF-PEG-PPhe-Bai纳米胶束,这两种药物都可以提高 SOD酶活性和降低MDA的含量,但是LF-PEG-PPhe-Bai纳米胶束对SOD酶活性提高以 及降低MDA含量的作用都要比黄芩素更显著,表明实施例1制备得到的LF-PEG-PPhe-Bai 纳米胶束对氧糖剥夺细胞具有更显著的抗氧化效果。
实施例10 TNF-α生物学活性检测方法
肿瘤坏死因子(TNF家族)指一组可以引起细胞死亡(凋亡)的细胞因子,TNF-α 是一种多向性的促炎性细胞因子,由多种细胞分泌,包括脂肪细胞、活化的单核细胞、巨 噬细胞等。
一、实验方法
取对数期SH-SY5Y细胞进行接板,培养过夜,OGD 4h后加入各组药物,分别为对 照组(不进行OGD处理+100μL基础培养基),缺血组(OGD处理4h+100μL基础培养 基),黄芩素组(OGD处理4h+含5μM黄芩素的100μL基础培养基)以及纳米胶束组 (OGD处理4h+含5μM实施例1制备得到的LF-PEG-PPhe-Bai纳米胶束的100μL基础 培养基)培养24h,收集细胞培养液,在300×g离心10min,吸取上清液;酶标板中加入 300μL的洗液浸泡30s,弃掉洗液后,在吸水纸上拍干;向标准品孔加入100μL稀释的 标准品,向空白孔加入100μL的培养基,向样品孔加入100μL细胞上清液;每孔加入 50μL稀释的检测抗体;使用封板膜封板,300r/min振荡,室温孵育2h,每孔加入300μL 洗液洗板6次,吸水纸拍干,每孔加入100μL稀释的辣根过氧化物酶标记的链霉亲和素; 使用新的封板膜封板,300r/min振荡后,室温孵育45min;每孔加入300μL洗液洗板6 次,再次吸水纸上拍干,每孔加入100μL显色底物TMB,避光,室温孵育30min;每孔 加入100μL终止液在30min之内,使用酶标仪进行双波长检测,测定450nm最大吸收波 长和570nm波长下的OD值。以标准品浓度为横坐标,OD值为纵坐标,进行回归拟合生成标准曲线。根据标准曲线计算TNF-α含量。
二、实验结果
在对TNF-α含量进行检测时发现(图12),缺血组的TNF-α的表达显著高于对照组(P<0.01),而用黄芩素(黄芩组)处理后,能够减少TNF-α的表达,但对其结果进行生 物学统计时发现,其效果并不具有显著性差异,而当用同剂量的LF-PEG-Pphe-Ba纳米胶 束处理(纳米胶素组)能够显著降低TNF-α的表达。说明实施例1制备得到的 LF-PEG-Pphe-Bai胶束可以减少促炎细胞因子的产生,具有抗炎作用。
实施例11海马CA1区胶质细胞的变化
星形胶质细胞和小胶质细胞活化情况最能体现炎症反应的强弱。我们分别检测两种细 胞的阳性蛋白,观察胶质细胞活性情况。
一、实验方法
所有实验动物被随机分为假手术组(只分离双侧颈总动脉,不夹闭),缺血组,黄芩素组和纳米胶束组:
其中,缺血组,黄芩素组和纳米胶束组采用双侧颈总动脉夹闭方法建立全脑缺血小鼠 的动物模型。小鼠术前禁食不禁水12h,用1.4%的戊巴比妥钠(0.1mg/kg)腹腔注射麻醉 后,修剪颈部正中部皮毛后做0.8~1cm正中切口,切开皮肤和皮下组织,用玻璃分针分离二腹肌和胸锁乳突肌,暴露双侧颈动脉,然后小心分离双侧颈总动脉及迷走神经约0.5~1cm,以无创微动脉夹夹闭双侧颈总动脉阻断血流30min后松开,恢复脑部供血。最后缝 合切口,消毒,用青霉素进行肌注抗炎。假手术小鼠除不夹闭双侧颈总动脉外,其他操作 与模型组一致。术后小鼠麻醉苏醒前需予以白炽灯或孵育箱保温,防止低温休克的发生,室温维持在26~28℃为宜。
术后小鼠修复2d后,开始进行药物处理治疗。黄芩素组和纳米胶束组分别通过尾静 脉注射相同剂量的黄芩素和实施例1制备的得到的LF-PEG-PPhe-Bai纳米胶束,而假手术组和缺血组进行同体积的生理盐水。所有组别给药周期为10d,结束后开始进行小鼠行为学检测。
1、HE染色
为了进一步验证LF-PEG-PPhe-Bai纳米胶束在缺血体外模型中是否具有神经保护作 用。利用HE染色来观察双侧颈总动脉夹闭30min对海马CA1区神经细胞的损伤情况。
具体做法:
将玻片放置于玻璃载片盒中,二甲苯Ⅰ溶液中脱蜡5~10min,换用二甲苯Ⅱ溶液,再脱蜡5~10min,无水乙醇脱水5min,90%乙醇脱水2min,80%乙醇再脱水2min,70% 乙醇再次脱水2min,最后蒸馏水2min。
样品处理后的玻片放置于原来的玻璃载片盒中不移动,苏木素染色液染色10min,浸 自来水中冲洗去多余的染色液,约10min,再用蒸馏水洗涤10s,之后伊红染色液染色2min,再按照下列顺序进行脱水、透明、封片:60%乙醇溶液10s,70%乙醇溶液10s,80% 乙醇溶液10s,90%乙醇溶液10s,无水乙醇溶液10s,二甲苯Ⅰ溶液透明5min,二甲苯 Ⅱ溶液透明5min,最后用中性树胶封片。显微镜下观察,细胞核呈蓝色,而细胞浆呈粉 红色或红色。
2、免疫组化检测胶质细胞活化情况
为了进一步验证LF-PEG-PPhe-Bai纳米胶束在脑缺血小鼠模型中的抗炎症效果,利用 免疫组织化学来观察海马CA1区胶质细胞的活化现象。分别利用GFAP和IBA-1标志蛋白分别检测星形胶质细胞和小胶质细胞的活化情况。
具体做法:
药物处理后的小鼠大脑组织用石蜡包埋,石蜡切片脱蜡至水,依次将切片放入二甲苯 I 15min,二甲苯II 15min,无水乙醇I5min,无水乙醇II 5min,85%酒精5min,75%酒精5min,最后用蒸馏水清洗。组织切片置于EDTA抗原修复缓冲液修复。自然冷却后用 PBS洗涤,之后用3%BSA均匀覆盖组织,室温封闭30min。吸去封闭液,添加一抗(GFAP, Abcam,1:5000和IBA-1,Abcam,1:5000),4℃孵育过夜。用PBS洗涤,吸去洗涤液,添加 二抗,避光室温孵育50min。洗涤,封片。镜检拍照,采集图像。
二、实验结果
1、海马CA1区细胞数目的变化
如图13所示,假手术组的小鼠海马CA1区神经元细胞排列紧密、结构表现为锥体神经元萎缩、核固缩和不规则排列。与假手术组相比,缺血组CA1区神经元数量明显减少。 而经过黄芩素和LF-PEG-PPhe-Bai纳米胶束处理后,CA1区神经元细胞数目增多。以上结 果说明,缺血30min对小鼠海马CA1区神经元的损伤较为严重,而LF-PEG-PPhe-Bai纳 米胶束可以逆转脑缺血损伤。
2、海马CA1区胶质细胞活化情况
结果如图14和15所示,与假手术组相比,缺血组小鼠海马CA1区星形胶质细胞和小胶质细胞活化的数目明显增多,而黄芩素组处理后,并没有减少活化的数目,但是实施例1制备的LF-PEG-PPhe-Bai纳米胶束处理后,CA1区星形胶质细胞和小胶质细胞活化的 数目明显减少。在对染色后的图片,平均光度值进行统计时,也呈现出相同的结果,与假 手术组相比,缺血组小鼠海马CA1区胶质细胞的光度值明显升高,表明胶质细胞被活化, 而黄芩素和纳米胶束处理后均有明显的降低。以上结果说明,缺血30min激活小鼠海马 CA1区星形胶质细胞和小胶质细胞活化,而实施例1制备的LF-PEG-PPhe-Bai纳米胶束可 以逆转海马CA1区的炎症反应。
结果发现,缺血30min会明显增强海马CA1区胶质细胞的活化,而LF-PEG-PPhe-Bai纳米胶束表现出显著的抗炎症的效果。对于脑缺血而言,不管是体内还是体外,实施例1 制备的LF-PEG-PPhe-Bai纳米胶束均表现出明显的神经保护效果。因而本研究可以为脑缺 血的神经修复提供新的治疗思虑
最后所应当说明的是,以上实施例仅用以说明本发明的技术方案而非对本发明保护范 围的限制,对于本领域的普通技术人员来说,在上述说明及思路的基础上还可以做出其它 不同形式的变化或变动,这里无需也无法对所有的实施方式予以穷举。凡在本发明的精神和原则之内所作的任何修改、等同替换和改进等,均应包含在本发明权利要求的保护范围 之内。
Claims (9)
1.一种包裹黄芩素的纳米胶束的制备方法,其特征在于,包括以下步骤:
S1.采用Fuchs - Farthing法,苯丙氨酸与三光气反应,得到Phe - NCA;
S2.取Phe - NCA和mPEG10k - NH2反应得到嵌段聚合物聚乙二醇 - 聚苯丙氨酸mPEG -PPhe;
S3.嵌段聚合物聚乙二醇 - 聚苯丙氨酸mPEG - PPhe和黄芩素在超声下自组装,得到聚合物胶束PEG - PPhe - Bai,利用嵌段共聚物mPEG - PPhe胶束自组装包裹疏水药物黄芩素;
S4.聚合物胶束PEG - PPhe - Bai接枝乳铁蛋白,含有聚合物胶束PEG - PPhe - Bai和乳铁蛋白的混合溶液先在60~68℃孵育8~12min,之后在 - 18~ - 22℃孵育5~8min,得到纳米胶束LF - PEG - PPhe - Bai,将嵌段聚合物mPEG - PPhe胶束插入靶向分子乳铁蛋白的空腔中。
2.根据权利要求1所述的制备方法,其特征在于,步骤S1中,含有苯丙氨酸的有机溶剂溶液加热回流,与含有三光气的有机溶剂溶液混合,继续回流,直至得到澄清溶液作为有机相,用无机相洗涤有机相,保留有机相,用无水硫酸镁干燥有机相,浓缩后用石油醚析出,纯化。
3.根据权利要求1所述的制备方法,其特征在于,步骤S2中,氮气保护下,mPEG10k - NH2溶液与Phe - NCA混合,得到澄清溶液后,与三氯甲烷混合,充分反应后用无水乙醇沉淀,取沉淀,纯化。
4.根据权利要求1所述的制备方法,其特征在于,步骤S3中,超声处理下,含有嵌段聚合物聚乙二醇-聚苯丙氨酸mPEG - PPhe和黄芩素的有机溶液逐滴加入水中,纯化,收集粒径小于0 .45μm且分子量大于5kDa产物,浓缩。
5.权利要求1~4任一所述制备方法制备得到的纳米胶束。
6.权利要求5所述的纳米胶束在制备治疗缺血性脑损伤和/或脑卒中的药物中的应用。
7.根据权利要求6所述的应用,其特征在于,所述纳米胶束为治疗缺血性脑损伤和/或脑卒中的神经元细胞保护剂。
8.根据权利要求6所述的应用,其特征在于,所述纳米胶束为治疗缺血性脑损伤和/或脑卒中的神经炎症药物中的应用。
9.根据权利要求6所述的应用,其特征在于,所述脑卒中为缺血性脑卒中。
