技术领域
本发明属于纳米泡制备技术领域,具体涉及于一种超声、荧光双重显影负载量子点纳米泡及其制备方法和应用。
背景技术

目前,肿瘤的临床治疗手段主要包括手术治疗、全身化学治疗等,对于中晚期癌症病人,往往治疗效果较差、预后不良,且副作用强烈、患者生活质量较低。肿瘤若能在早期发现并进行干预治疗,预后效果将有明显的提高。
肿瘤的诊断手段可分为体外诊断和体内诊断。体外诊断多为抽取患者的血液样本进行“液体活检”或者对其组织进行病理学的检查,主要为组织病理学检测、肿瘤细胞检测和肿瘤标志物的检测。这些方法虽然准确性较高,但操作繁琐,检测结果具有滞后性。体内诊断则是利用荧光、超声造影剂等材料的显影或X射线、磁共振等影像学方法对体内组织的病理形态进行实时检测,常见的是放射成像、超声成像和荧光成像。在放射成像过程中辐射会对身体正常组织造成伤害,不宜长期高频的检测评估,超声成像和荧光检测对患者的机体损伤最小、应用最为广泛。
超声造影剂是一种空泡型的制剂,多为壳-核结构,利用其两界面不同的回声反射性质,使超声波的散射回声信号增强,从而提高超声检测的分辨率及敏感程度,在超声检测中广泛使用。目前临床使用的大多是微米级的造影剂,又可称为微米泡,其粒径分布大致在1-4μm。微米泡的气液型结构可以显著增强与组织背景的对比度,从而提高回声反射的分辨率;核壳结构可以通过外壳的保护作用,缓解其在体内的破损和泄漏。常用的外壳材料包括白蛋白、表面活性剂、半乳糖、脂质类化合物等,而内核气体的选材主要包括空气、全氟化物、氮气等。在血液中,静脉注射的微米泡会很快被网状内皮系统清除掉,在血清中的半衰期只有几分钟。但由于粒径限制,微米泡难以透过细小的脉管系统,更加难以在肿瘤区域富集,从而限制了其在狭小组织中的造影能力。纳米级造影剂的结构与微米泡类似,外壳材料多为脂质、表面活性剂或聚合物高分子。在超声波作用下,纳米泡发生规律性的膨胀和压缩现象,从而产生很强的回声反射,通过传感器到达处理器实现对靶部位的显影。由于其粒径较小,常具有许多特殊的分布行为,如肿瘤区域的渗透和富集等。肿瘤的快速生长需要大量血管的生成,通过新生血管将氧气和营养物质源源不断地向肿瘤区域运送。但快速生长的肿瘤血管内皮间隙较正常血管组织大,可渗漏一定体积和粒径的物质,而正常血管组织不会出现此类现象,称为EPR效应。肿瘤血管内皮的空隙约为380-780nm,这就为纳米泡在肿瘤的被动靶向富集提供了机会。此外,纳米级造影剂粒径越小,穿透能力越强,但产生的背向散射能力越弱,相对稳定性就越弱,可通过调节成膜材料和成膜材料的比例或制备方法进一步提高背向散射能力,提高显影效果。
荧光成像常通过注射荧光性、低毒的检测物质或给予特异性的分子探针,利用电子吸光跃迁、释能发光的原理,通过灵敏的光学检测器直接检测患者体内的荧光成像行为。此类非损伤性的检测方法使用广泛,不仅可以实时成像,还可以同时检测多分子、多组织的成像情况,灵敏度较高,但目前临床可应用于人体的荧光检测物质少之又少。近年来,随着生物低毒的碳量子点(Carbon Quantum Dots,CQDs)研究的深入,CQDs极有可能成为下一个临床应用的荧光检测剂。碳量子点是一种新型的荧光性纳米级的碳材料。由类球状的分散粒组成,粒径约为10nm。CQDs具有极好的生物相容性、毒性低,非常适合用于人体荧光检测。CQDs的研究热潮来源于其良好的荧光性质CQDs荧光效果稳定、强度高,且激发波长范围宽,可以根据临床仪器的不同需要制备不同激发和发射波长的碳点,应用范围广泛。
超声成像和荧光检测对患者身体的损伤最小,应用最广泛,但发明人发现,各种单一的检测手段都不具有较高的可信度,检测结果容易受到体内各种不同因素的影响。
发明内容
基于上述现有技术的不足,本发明提供一种超声、荧光双重显影负载碳量子点纳米泡及其制备方法和应用,本发明通过将超声造影剂及荧光检测剂两者相结合,设计并制备了碳量子点偶联的纳米级脂质体纳米泡作为新型肿瘤检测试剂,从而达到一次给药,多维映证的效果,有效提高检测效率和准确性。基于上述研究成果,从而完成本发明。
本发明的技术方案如下:
本发明的第一个方面,提供一种超声、荧光双重显影负载量子点纳米泡,所述纳米泡为脂质体纳米泡,所述脂质体纳米泡表面负载有量子点;
更具体的,所述脂质体纳米泡为(2,3-二油酰基-丙基)-三甲胺(DOTAP)阳离子脂质体。
所述脂质体纳米泡的平均粒径为224.6±2.381nm,平均PDI为0.245±0.0626,平均ζ电位值为46.2±1.29mV。
本发明的第二个方面,提供超声、荧光双重显影负载碳量子点纳米泡的制备方法,所述制备方法包括:
S1、将DOTAP及胆固醇溶于氯仿中,待溶剂完全挥发后成膜,加入缓冲液后搅拌使脂质体成形均匀;
S2、向步骤S1中加入全氟化物,超声处理得脂质体纳米泡,向其中加入量子点即得。
本发明的第三个方面,提供上述纳米泡在制备肿瘤检测和诊断试剂中的应用。

上述技术方案通过将超声造影剂及荧光检测剂两者相结合,设计并制备了碳量子点偶联的纳米级脂质体纳米泡作为新型肿瘤检测试剂,优化了其处方组成及制备工艺,并对其粒径、电位指标进行评价。同时设计了体内超声成像以及活体成像实验,以推进碳量子点偶联的脂质体纳米泡的应用检测范围、安全性、有效性的完善评价,试验证明本发明制备的纳米泡能进入到了肿瘤腔隙,并在瘤内有很强的超声信号。具有较高的负电位的纳米泡更加稳定,不易聚集,能更好的靶向到肿瘤部位,体内成像也表明该纳米泡可精确靶向肿瘤,且其体内存留时间短,可被快速代谢,因此具有良好的临床应用价值。

本发明的一个具体实施方式中,提供一种超声、荧光双重显影负载量子点纳米泡,所述纳米泡为脂质体纳米泡,所述脂质体纳米泡表面负载有量子点;
更具体的,所述脂质体纳米泡为(2,3-二油酰基-丙基)-三甲胺(DOTAP)阳离子脂质体。
所述脂质体纳米泡的平均粒径为224.6±2.381nm,平均PDI为0.245±0.0626,平均ζ电位值为46.2±1.29mV。
其中,所述脂质体纳米泡包载气体可以为全氟化物、空气、氮气中的任意一种或多种;
所述量子点可以为碳量子点、CdS、CdSe、CdTe、ZnSe、InP和InAs中的任意一种或多种;由于碳量子点具有极好的生物相容性、毒性低,非常适合用于人体荧光检测,因此优选为碳量子点。
本发明的又一具体实施方式中,提供超声、荧光双重显影负载碳量子点纳米泡的制备方法,所述制备方法包括:
S1、将DOTAP及胆固醇溶于氯仿中,待溶剂完全挥发后成膜,加入缓冲液后搅拌使脂质体成形均匀;
S2、向步骤S1中加入全氟化物,超声处理得脂质体纳米泡,向其中加入量子点即得。
其中,所述步骤S1中,
所述DOTAP与胆固醇的质量比为0.5-5:1,进一步优选为1-3:1,更进一步优选为1:1,通过对脂质与胆固醇的用量比例进行筛选,使得最终制备得到的脂质体纳米泡性质稳定且粒径较小。
所述缓冲液可以为PBS缓冲液;
所述步骤S2中,所述全氟化物可以为全氟戊烷,全氟戊烷对纳米泡的粒径影响较为明显,随着用量的增加纳米泡的粒径逐渐增大。全氟戊烷在常压下沸点为29.2℃,气化后是一种安全性较高的惰性气体。
所述胆固醇与全氟戊烷的质量体积比为25:0.5-5(μg/μL),优选为25:1;
所述超声处理具体条件包括:在100-1000w(优选150-250W)的功率超声处理0.5s-60min,优选为1s。
所述量子点为碳量子点、CdS、CdSe、CdTe、ZnSe、InP和InAs中的任意一种或多种;由于碳量子点具有极好的生物相容性、毒性低,非常适合用于人体荧光检测,因此优选为碳量子点。
所述碳量子点浓度控制为8-12mg/mL,优选为10mg/mL。
本发明的又一具体实施方式中,提供上述纳米泡在制备肿瘤检测和诊断试剂中的应用。
本发明的又一具体实施方式中,所述肿瘤检测和诊断试剂具体为具有超声造影和荧光检测功能的试剂。
本发明的又一具体实施方式中,提供一种肿瘤检测和诊断试剂,所述肿瘤检测和诊断试剂包含上述纳米泡。
以下通过实施例对本发明做进一步解释说明,但不构成对本发明的限制。应理解这些实施例仅用于说明本发明而不用于限制本发明的范围。
实施例
实验方法
1.1成泡设备的筛选
将DOTAP及胆固醇以一定的比例和用量溶解在氯仿中,进行旋转蒸发,使溶剂完全挥发后成膜,加入PBS溶液,在40℃水浴条件下磁力搅拌1h使脂质体成形均匀。加入5μL的全氟戊烷,在超声清洗仪(最大功率为650W)或超声破碎仪(最大功率为950W)中以不同的功率、不同的时间超声,得脂质体纳米泡,在粒径电位分析仪中检测其粒径大小。
1.2脂质体胆固醇比例的筛选
将DOTAP与胆固醇以不同的比例(w/w=0.5、1、2、3,即62.5μg-375μg:125μg)分别溶解在氯仿中,按上述方法制备脂质体。加入5μL的全氟戊烷,在超声破碎仪中以20%的功率超声1s,得脂质体纳米泡,在Zetasizer Nano ZS粒径电位分析仪中检测其粒径大小。
1.3全氟戊烷用量的筛选
控制DOTAP及胆固醇的比例不变制备脂质体。加入不同量(5、10、15、20μL)的全氟戊烷,在超声破碎仪中以20%的功率超声1s,得脂质体纳米泡,用粒径电位分析仪检测其粒径大小。
1.4脂质体成形溶剂的筛选
控制DOTAP及胆固醇的比例不变,旋转蒸发后分别加入PBS溶液1mL及配置好的甘油:PBS=1:9(v/v)的溶液1mL,在40℃水浴条件下磁力搅拌1h使脂质体均匀。加入5μL全氟戊烷,在超声破碎仪中以20%的功率超声1s,得脂质体纳米泡,用粒径电位分析仪检测其粒径大小。
1.5不同检测温度对纳米泡粒径的影响
根据上述探索结果选取最佳工艺参数和制备流程制备纳米泡,即DOTAP:胆固醇=1-3:1(125-375μg:125μg),全氟戊烷5-20μL,经超声破碎仪超声1s,功率20%。在不同的温度下(25、37℃)对同一纳米泡的粒径大小进行检测。
1.6负载碳量子点脂质体纳米泡的制备工艺与评价
根据上述探索结果选取最佳工艺参数和制备流程,制备纳米泡。在脂质体溶液中滴加不同量的碳量子点(10mg/mL),室温避光搅拌30min,即得负载碳量子点脂质体纳米泡。室温避光透析12h后,检测碳量子点脂质体纳米泡的ζ电位。
1.7体内实验
1.7.1体内超声成像实验
取荷瘤小鼠,腹腔注射4%的水合氯醛麻醉,用超声检测仪检测小鼠肿瘤处呈像情况。尾静脉注射纳米泡lip-CQDs(同1.6)注射液100μL,并记录时间.5min后,采用超声检测并记录肿瘤影像学结果,重复实验3次。
1.7.2体内的生物分布的测定
为研究药物的体内生物分布和肿瘤靶向效果,将荷瘤小鼠随机分组(n=3),用4%水合氯醛麻醉。小鼠静脉注射0.2ml lip-CQDs(同1.6)。5min后,通过Xenogen IVIS Lumina系统获取实时图像。
2.实验结果
2.1成泡设备的筛选
2.1.1超声清洗的成泡筛选结果
在实验中发现,利用超声清洗仪作为成泡设备时,纳米泡的粒径随超声功率的增大而增大如图1A,但随超声时间的延长变化规律不甚清晰,呈减小、增大、减小而后增大的趋势,如图1B。在超声破碎成泡的过程中,设计不同的破碎功率及时间参数以考察超声破碎对纳米泡粒径的影响。结果显示,脂质体纳米泡的粒径随超声破碎功率的增加而增大(图1C),随超声时间的增加先减小后增大,粒径趋势如图1D。本实验在1s处取得纳米泡粒径的最小值。
2.2脂质体胆固醇比例的筛选结果
对脂质与胆固醇比例进行筛选,以得到稳定且粒径较小的脂质体纳米泡。结果证明当胆固醇与脂质DOTAP的质量比为1:1时,粒径最小,趋势如图2A。说明当DOTAP脂质与胆固醇的质量比为1:1时,脂质体既可利用流动性包裹液态全氟戊烷,又可拥有刚性保证气体内核的稳定。而胆固醇比例更小时稳定性不足;比例更大时,多余的胆固醇或可使两脂质体连接融合,或可形成多层的大粒径脂质体。
2.3全氟戊烷用量的筛选结果
全氟戊烷的用量对纳米泡的粒径影响较为明显,随着用量的增加纳米泡的粒径逐渐增大(图2B)。全氟戊烷在常压下沸点为29.2℃,气化后是一种安全性较高的惰性气体,其优良的相变温度和物理化学性质适用于本实验的纳米成泡工艺流程。
2.4脂质体成形溶剂的筛选结果
脂质体成形溶剂的浓度、pH、温等因素在脂质体脱模成形过程中起重要的作用,对其粒径和稳定性均有影响。根据实验结果,可知甘油:PBS=1:9(v/v)溶液制备的纳米泡粒径更大,是因为甘油的黏度改变了粒子的运动分散特性,使测量结果较真实值偏大。
2.5不同检测温度对纳米泡粒径的影响
DOTAP阳离子脂质体的相变温度为35-38℃。故而参考其他研究,本实验的旋蒸、水浴、偶联温度均设定为40℃以大于其相变温度。然而却很少有研究关注和强调其检测温度。通过实验发现在25℃时,其粒径较大;而同一样本的脂质体纳米泡在37℃检测时粒径明显减小。
2.6脂质体纳米泡的制备工艺与评价
根据以上工艺参数和制备方法的探索,采用本实验最佳参数和工艺制备DOTAP阳离子脂质体纳米泡,其粒径及电位数据。实验所得DOTAP脂质体纳米泡的平均粒径为224.6±2.381nm,平均PDI为0.245±0.0626,平均ζ电位值为46.2±1.29mV,见图3A。由电镜图(图3B),也可以看出纳米泡大小均一,形态圆整。
2.7负载碳量子点脂质体纳米泡的制备工艺与评价
将一定浓度的DOTAP纳米泡与不同量的碳量子点(200-235μL)以电荷作用吸引连接,得到碳量子点在纳米泡表面偶联的极值,负载碳量子点ζ电位结果如图4,随着碳量子点用量的增加,ζ电位由正值转变为负值,在达到一定剂量后,ζ电位趋于平稳。在纳米泡溶液里加入225μL的碳量子点(10mg/mL)可近似达到纳米泡偶联极值,此时纳米泡偶联碳量子点的效率最高。
2.8体内超声成像实验
以随机分组法将小鼠分为2组建模,每组10只。观察注射纳米泡前后小鼠肿瘤处B超影像,结果如图5。在未注射溶液前,乳腺癌模型小鼠的肿瘤部位影像较暗,为黑色区域,这是因为乳腺癌皮下瘤为实体瘤,回声较弱。在尾静脉注射纳米泡5min后,超声成像可见显著的变化。这说明纳米泡溶液进入到了肿瘤腔隙,并在瘤内有很强的超声信号。具有较高的负电位的纳米泡更加稳定,不易聚集,能更好的靶向到肿瘤部位。在肝癌模型小鼠中也验证纳米泡的效果,注射前在肝脏部位可见一处囊性肿瘤,但不易与周围组织区分。在注射纳米泡之后,我们发现肿瘤区域呈像变得更加清晰。
2.9体内的生物分布的测定
为了实时观察药物和纳米颗粒在主要器官中的实时分布,我们采用体内成像实验对相关结果进行了分析。结果显示,在Hepa 1-6荷瘤小鼠体内,Lip-CQDs可在大约5min后,富集于小鼠肝脏处,其他器官处几乎没有荧光,结合超声造影结果,可证明Lip-CQDs可以精确的靶向肿瘤。肝脏处的荧光在1h后基本消失,则证明Lip-CQDs在体内存留的时间短,可以被快速代谢。
最后应该说明的是,以上所述仅为本发明的优选实施例而已,并不用于限制本发明,尽管参照前述实施例对本发明进行了详细的说明,对于本领域的技术人员来说,其依然可以对前述实施例所记载的技术方案进行修改,或者对其中部分进行等同替换。凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。上述虽然对本发明的具体实施方式进行了描述,但并非对本发明保护范围的限制,所属领域技术人员应该明白,在本发明的技术方案的基础上,本领域技术人员不需要付出创造性劳动即可做出的各种修改或变形仍在本发明的保护范围以内。
Claims (6)
1.一种超声、荧光双重显影负载量子点纳米泡,其特征在于,所述纳米泡为脂质体纳米泡,所述脂质体纳米泡表面负载有量子点;
所述脂质体纳米泡为(2,3-二油酰基-丙基)-三甲胺DOTAP阳离子脂质体;
所述脂质体纳米泡的平均粒径为224.6±2.381nm,平均PDI为0.245±0.0626,平均ζ电位值为46.2±1.29mV;
所述脂质体纳米泡包载气体为全氟化物;
所述全氟化物为全氟戊烷;
所述量子点为碳量子点;
所述超声、荧光双重显影负载碳量子点纳米泡的制备方法,包括:
S1、将DOTAP及胆固醇溶于氯仿中,待溶剂完全挥发后成膜,加入缓冲液后搅拌使脂质体成形均匀;
S2、向步骤S1中加入全氟化物,超声处理得脂质体纳米泡,向其中加入量子点即得;
所述超声处理的条件为采用超声破碎仪进行处理;

所述超声处理具体条件包括:在100-1000w的功率超声处理0.5s-60 min;
所述碳量子点浓度控制为8-12mg/mL。
