Abstract
本发明提供一种双前药骨架聚合物PCPt、其纳米颗粒及其制备方法和应用,PCPt以姜黄素、顺,顺,反‑[Pt(NH3)2Cl2(OH)2]、赖氨酸二异氰酸酯和聚乙二醇单甲基醚衍生物为原料。本发明提供的主链型铂(IV)和姜黄素(Cur)双前药骨架聚合物PCPt为两亲结构,可在水溶液中自组装成纳米粒子,具有较高的载药量和载药效率,对癌细胞表现出有效的细胞内摄取和增强的细胞增殖抑制作用。PCPt、PCPt NPs或包含他们的药物递送系统是铂和Cur共基联合化疗的有前途的平台。

技术领域
本发明涉及医药领域,具体涉及一种双前药骨架聚合物PCPt、其纳米颗粒及其制备方法和应用。
背景技术
公开该背景技术部分的信息仅仅旨在增加对本发明的总体背景的理解,而不必然被视为承认或以任何形式暗示该信息构成已经成为本领域一般技术人员所公知的现有技术。
癌症仍然是一个最严重的全球公众健康问题,严重威胁了人们的健康和生活。化疗在治疗各种临床癌症疾病时仍然是被采用最广泛的方法。然而,传统的单化疗手段由于对于新肿瘤的不充分选择性、对健康组织的毒性和药物抵制,治疗效率极大降低。不像单化疗手段,使用两种或更多药物的联合化疗可以活化不同的信号方式、最大化治疗效率、减少药物用量并且能克服药物抵制。肿瘤简单药物联用虽然具有卓越的优势,但由于其在水溶液中的溶解度较低,且不同药物的药代动力学差异较大,极大地阻碍了其应用,导致治疗失败。
具有诸多优点的纳米药物传递系统被极大地探究用来癌症治疗,包括提升药物溶解力、延长血液循环次数、降低副作用和通过提升渗透滞留效应增加肿瘤累积量。在纳米载体中,聚合物纳米粒子由于组合治疗受到关注。目前,常见的聚合物共传递系统用于联合治疗通常是用物理法包裹治疗药物。在这种系统下,不可控的药物释放和过早的释放导致严重的副作用并且达不到满意的治疗效果。作为另一种方式,共价共轭不同的药物到聚合物链的末端或副链上是另一种方法去构建聚合共传递系统。共价共轭系统是惰性和稳定的在血液循环中,能够响应刺激释放药物,例如酸性PH,肿瘤还原电势和酶过度表达,因此在药物释放时提供了较好的控制并且减少了副作用。然而,这些系统有一些缺点例如合成过程复杂、不确定的药物负载比、低药物负载成分和效率。在这些系统中,常常可以观察到不同批次的载药和释药行为的变化,这些变化可能是临床转化的关键瓶颈。
发明内容
为了克服现有技术的不足,同时解决由于肿瘤的异质性和耐药性的出现,常规的单一化学疗法对于癌症的治疗效果不满意的情况,本发明提供了一种双前药骨架聚合物PCPt、其纳米颗粒(PCPt NPs)及包含PCPt或其纳米颗粒的药物递送系统,以及上述物质的制备方法和应用。本发明提供的主链型铂(IV)和姜黄素(Cur)双前药骨架聚合物PCPt为两亲结构,可在水溶液中自组装成纳米粒子,具有较高的载药量和载药效率,对癌细胞表现出有效的细胞内摄取和增强的细胞增殖抑制作用。PCPt、PCPt NPs或包含他们的药物递送系统是铂和Cur共基联合化疗的有前途的平台。
在本发明的第一方面,本发明提供了一种双前药骨架聚合物PCPt,其以姜黄素(Cur)、顺,顺,反-[Pt(NH3)2Cl2(OH)2](DHP)、赖氨酸二异氰酸酯(LDI)和聚乙二醇单甲基醚(mPEG)衍生物为原料。
在本发明的实施方式中,PCPt为两亲性三嵌段结构,其中,Pt(IV)前药(DHP)和Cur与LDI共聚形成主链,以构建疏水性嵌段,然后用亲水性聚乙二醇单甲基醚衍生物(比如mPEG5k-NCO)为终止。
在本发明的一些实施方式中,所述聚乙二醇单甲基醚衍生物为异氰酸酯官能化的聚乙二醇单甲醚(mPEG5k-NCO)。
姜黄素是一种很有前景的天然抗癌成分,由于其药理学安全性和克服多药耐药性的潜力而被引入研究,不幸的是,姜黄素的应用因其较差的水溶性和不稳定性而受到限制。在本发明的实施方式中,Cur和铂(IV)的前药DHP作为形成主链型聚合物的共聚单体,以聚乙二醇单甲基醚(mPEG)衍生物比如mPEG5k-NCO作为终止剂形成的PCPt为两亲结构,克服了姜黄素水溶性差和不稳定而无法直接药用的缺陷,同时采用Pt(IV)最大程度的减少顺铂(cis-dichlorodiammineplatinum(II),cisplatin)严重副作用,由于Pt(IV)的还原反应特性,所获得的双重药物骨架的PCPt可以通过链断裂的方式响应还原条件,并同时释放生物活性的Pt(II)和Cur,实现姜黄素和顺铂的协同应用,从而更好地治疗癌症。
在本发明的第二方面,本发明提供了一种制备上述第一方面中所述的双前药骨架聚合物PCPt的方法,其包括Cur、DHP和LDI原位聚合后与聚乙二醇单甲基醚衍生物偶联。
在本发明的实施方式中,Cur、DHP和LDI的摩尔比为1-6:1-6:2-13,优选为5:4:8。
PCPt的合成图如图2所示。
本发明的制备方法制备得到的PCPt不同于采用共价共轭等方法(药物往往在聚合物链的末端或副链上)构建的聚合共传递系统,其为前药骨架的聚合物,其中药物Cur、DHP是作为聚合物主链的组成部分而不是偶联到聚合物的末端或侧链上,具有固定的Pt/Cur比,能够更好的控制组成并增加药物载量和载药效率,使成分不会爆炸式的释放,而且本发明的方法简单易操作,克服了聚合共传递方式合成过程复杂、药物负载比不确定、载药量和载药效率低、载药和释药行为批次间差异大的缺陷。
在本发明的第三方面,本发明提供了一种PCPt纳米颗粒,其由上述第一方面中所述的双前药骨架聚合物PCPt自组装得到。
在本发明的实施方式中,PCPt是一种两亲性ABA三嵌段聚合物,可以在选择性溶剂中组装。在本发明的一些实施方式中,所述自组装溶剂为水。本发明在研究中发现,PCPt的CAC值(即临界胶束浓度,是表征表面活性剂溶液物化性能的重要参数)低,PCPt NP在水溶液中更稳定,并且能够在体内长时间循环。并且,PCPt NPs为单个球形颗粒,小而均一,有利于延长血液循环和细胞内吞作用。在本发明的一些实施方式中,所述纳米颗粒的平均直径为90-110nm。
本发明的PCPt NPs具有还原敏感性,在本发明的一些实施方式中,根据DLS结果,即使在孵育72小时后,PBS 7.4中PCPt NP的平均大小和PDI均没有明显差异。但是,在存在10mM SA(抗坏血酸钠)的情况下,PCPt NP的粒径和PDI都发生了巨大变化,粒径在12小时内可以从103nm快速增加到911nm,然后逐渐减小,推测由于骨架发生降解导致。因此可预期,PCPt NPs骨架中的Pt(IV)前药可以通过在肿瘤细胞的还原性微环境下(例如DTT和/或GSH的存在下)裂解还原为活性Pt(II)药物。
游离Cur为疏水性药物,水饱和浓度较低,且游离Cur溶液不均一,通常静置10分钟即可出现明显可见的沉淀,而在本发明的实施方式中,纳米级PCPt NPs溶液是均匀的,可以维持至少一个月没有沉淀,可见,本发明的PCPt NPs形式的共轭Cur具有更高的水溶性和稳定性。此外,游离Cur很容易被降解,在本发明的一些实施方式中,发明人发现,在相同条件下孵育7h后,游离Cur含量仅保持599.9%,而PCPt NPs中94%的Cur保持正常。以上表明,本发明的PCPt NPs可以保护Cur免于被降解,而且提高了Cur的稳定性。
在本发明的第四方面,本发明提供了一种药物递送系统或药物载体,其包含上述第一方面中所述的双前药骨架聚合物PCPt或上述第三方面中所述的PCPt纳米颗粒。
在本发明的一些实施方式中,所述药物递送系统或药物载体可以通过链断裂的方式响应还原条件从而实现活性药物的释放。
本发明的PCPt或其纳米颗粒PCPt NPs可以通过链断裂的方式响应还原条件,并同时释放生物活性的Pt(II)和Cur。在本发明的实施方式中,本发明使用透析方法在PBS(pH7.4)或10mM SA中研究了PCt NP中Pt(II)和Cur的释放。其中,在pH 7.4下检测到相当缓慢的Pt释放,在72小时后仅释放了约18.8%的Pt(II),这表明PCPt聚合物和NP在中性条件下是稳定的。添加10mM SA后Pt(II)的释放速率显着增加,72h后Pt(II)的累积释放量达到93.3%,这证明Pt(II)的释放是还原依赖性的,并且PCPt NPs可以在60h降解。基于Pt(IV)破坏的还原性环境。Cur释放的结果类似于Pt(II)释放。因此,对还原反应敏感的PCPt NP在开发用于癌症联合治疗的智能载体方面具有巨大潜力。
在本发明的第五方面,本发明提供了上述第一方面中所述的双前药骨架聚合物PCPt或上述第三方面中所述的PCPt纳米颗粒或上述第四方面中所述的药物递送系统或药物载体在制备治疗肿瘤的药物中的应用;或者在制备实现联合化疗的药物中的应用。
在本发明的实施方式中,经体外细胞分析,PCPt NPs对癌症细胞比如宫颈癌HeLa细胞表现出有效的细胞内摄取和增强的细胞增殖抑制作用。并且与单药治疗相比,PCPtNPs改善了生物安全性并增强了细胞毒性。在本发明的一些实施方式中,与DHP和Cur相比,PCPt NP对HeLa细胞显示出增强的细胞毒性。对于50%的细胞抑制作用,HeLa细胞的组合指数(CI)值计算为0.74,这证实了PCPt NP中铂和Cur的协同作用。
在本发明的实施方式中,发明人利用Cur分子固有的荧光特性,研究了癌细胞对药物的摄取情况以及药物在细胞内的分布情况。成像结果表明,PCPt NPs逐渐将Cur释放到细胞质,然后扩散到细胞核。Cur的不同胞内分布可能是由于内体中PCPt NP的Pt(IV)还原为Pt(II)。这些成像结果表明,与Cur较差的水溶性和稳定性相比,PCPt NPs易于在还原环境中被癌细胞内化并释放药物。
相较于现有技术,本发明的优势在于:
本发明的PCPt两亲性三嵌段结构,药物Cur、DHP是作为聚合物主链的组成部分而不是偶联到聚合物的末端或侧链上,具有固定的Pt/Cur比和较高的载药量、载药效率,能够更好的控制组成并增加药物载量和载药效率,使成分不会爆炸式的过早释放,而且本发明的方法简单易操作,克服了现有方法过程复杂、药物负载比不确定、载药量和载药效率低、载药和释药行为批次间差异大的缺陷。本发明的PCPt可在水溶液中自组装形成PCPt NPs,PCPt NPs为单个球形颗粒,小而均一,有利于延长血液循环和细胞内吞作用,可以保护Cur免于被降解,而且提高了Cur的稳定性,同时PCPt NPs具有还原敏感性,PCPt NPs骨架中的Pt(IV)前药(DHP)可以通过在肿瘤细胞的还原性微环境下(例如DTT和/或GSH的存在下)裂解还原为活性Pt(II)药物,Cur可在酸性内体/溶酶体和酶微环境下释放,从而发挥协同化学作用。PCPt NPs能够改善生物安全性并增强癌症细胞细胞毒性。PCPt NPs是铂和Cur共基联合化疗的有前途的平台。

下面结合具体实施例,进一步阐述本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。下列实施例中未注明具体条件的实验方法,通常按照常规条件或按照制造厂商所建议的条件。
除非另行定义,文中所使用的所有专业与科学用语与本领域熟练人员所熟悉的意义相同。本发明所使用的试剂或原料均可通过常规途径购买获得,如无特殊说明,本发明所使用的试剂或原料均按照本领域常规方式使用或者按照产品说明书使用。此外,任何与所记载内容相似或均等的方法及材料皆可应用于本发明方法中。文中所述的较佳实施方法与材料仅作示范之用。如无特殊说明,在本发明的发明内容、附图、附图说明以及实施方式中,Pt即指代Pt(II)。
实施例
实验
材料:顺铂购自山东博源药业有限公司(中国山东济南)。聚(乙二醇)单甲醚(mPEG5k-OH Mn=5000g/mol),3-(4,5-二甲基噻唑-2-基)-2,5-二苯基溴化四唑(MTT)和Hoechst 33258购自Sigma-Aldrich。可从阿拉丁试剂公司(中国上海)获得姜黄素,L-ascorbic酸钠盐(SA),六亚甲基二异氰酸酯(HDI),二月桂酸二丁基锡,L-赖氨酸二异氰酸酯(LDI)和过氧化氢溶液(H2O2)。培养基(DMEM)/高糖培养基,磷酸盐缓冲液(PBS)和胰蛋白酶购自GE Healthcare Life Sciences。胎牛血清(FBS)购自Thermo Fisher ScientificInc.(中国上海)。除非另有说明,所有化学试剂均为分析纯,无需进一步纯化。将二氯甲烷(DCM)、二甲基亚砜(DMSO)和N,N-二甲基甲酰胺(DMF)用氢化钙(CaH2)干燥2周,然后进一步纯化。
仪器:在室温(RT)下在Bruker AVANCE II 400NMR光谱仪上记录核磁共振(NMR)谱。凝胶渗透色谱仪(GPC)的测量在Wyatt Technology的DAWN(SEC-MALS)仪器上进行,以确定聚合物的摩尔质量和多分散指数。傅立叶变换红外(FT-IR)测量在Thermo ScientificNicolet IS10仪器上进行。在Cary 5000UV-Vis-NIR光谱仪上测定UV-可见吸收光谱。在日立F-4600光谱仪上记录荧光光谱。使用Malvern Zetasizer Nano ZS90仪器通过动态光散射(DLS)测量粒度和粒度分布。透射电子显微镜(TEM)图像在JEOL JEM2100电子显微镜上拍摄。铂含量的测量是在电感耦合等离子体质谱仪(ICP-MS,Optima 2000DV,PerkinElmer,美国)上进行的。共聚焦激光扫描显微镜(CLSM)(Leica SP8,德国)用于观察体外细胞荧光成像。通过流式细胞仪成像系统(BD FACSCalibur,美国)测量细胞摄取的荧光定量分析。
异氰酸酯官能化的聚乙二醇单甲醚(mPEG5k-NCO)的合成:
用HDI活化mPEG5k-OH,以合成NCO修饰的mPEG5k-OH(mPEG5k-NCO)。
具体包括:将mPEG5k-OH(3g,0.6mmol)与甲苯(100mL)共沸干燥,然后再溶解于干燥的DCM(20mL)中。将HDI(2.9mL,18mmol)与50mL干燥的DCM溶解。然后,将mPEG5k-OH溶液滴加到HDI中,随后滴加几滴二月桂酸二丁基锡。将混合物在氩气气氛下回流20小时。浓缩后,将粗产物沉淀到500mL的石油醚中。溶解和沉淀过程再重复两次之后,将最终产物在真空下干燥,并在氮气氛围下于密封小瓶中于-4℃储存,以备进一步使用。
Cur和Pt(IV)-主链前药聚合物的合成[mPEGpoly(platinum-co-Cur)-mPEG(即PCPt的合成)]
根据文献Z.Zhang,Y.Wu,G.Kuang,S.Liu,D.Zhou,X.Chen,X.Jing,Y.Huang,J.Mater.Chem.B,2017,5,2115-2125.,合成了顺,顺,反-[Pt(NH3)2Cl2(OH)2](DHP)。使用前,用水/DMF将Cur重结晶。将DHP(134mg,0.4mmol)分散在干燥的DMSO(2mL)中。将LDI(181mg,0.8mmol)溶解在干燥的DMSO(2mL)中,并在搅拌下滴加到上述混合物中。使反应在氩气下在50℃下反应8小时。冷却至室温后,加入到2mL干燥的DMSO中的Cur(184mg,0.5mmol)。然后,将反应混合物在氩气下于50℃保持24小时。随后,快速加入在DMSO溶液(10mL)中的mPEG5k-NCO(750mg,0.15mmol),并将混合物在50℃下再搅拌24h。用去离子水(MWCO=14000)透析溶液72小时,以去除DMSO和未反应的单体。冻干后,获得目标产物PCPt。PCPt的分子量和多分散指数通过GPC以DMF为洗脱液进行测定。具体的合成路线如图2所示。
PCPt纳米颗粒(PCPt NPs)的制备和表征

以芘作为荧光探针,测定PCPt NPs的临界聚集浓度(CAC)值。将不同浓度的PCPtNPs(4.9×10-5mg/mL~0.5mg/mL)在芘溶液中涡流过夜,在发射波长为373nm的荧光分光光度计上记录荧光。
PCPt纳米颗粒的稳定性
用DLS和TEM测定了PCPt NPs在不同条件下的胶体稳定性。详细地,将冻干的PCPt粉末称重,然后溶解于磷酸盐缓冲盐水(PBS)(0.01M,pH7.4),PBS(0.01M,pH 5.0)去离子水或带有10mM SA的去离子水中。PCPt NPs的浓度固定为1mg/mL。在预定的时间间隔分别通过DLS和TEM分析PCPt NP的形态、大小和大小分布。
然后,通过表征425nm处吸光度的变化,使用UV-vis光谱学研究了游离Cur和PCPtNP的生理稳定性。简而言之,将Cur和PCPt NP分散在pH 7.4的PBS缓冲液中,并在黑暗中于37℃孵育。在预定的时间点进行采样,并记录200至800nm的Cur的UV-vis吸收光谱。
体外药物释放
通过使用透析方法测量铂和Cur从PCPt NPs的释放曲线。通常,将冻干的PCPt NP(5mg)分别在pH 7.4(5mL)和带有SA(10mM)的去离子水(5mL)的PBS缓冲液中混合。然后,将悬浮液引入预溶胀的透析袋(MWCO=3500)中,并浸入上述相应溶液(35mL)中。在37℃的振荡培养箱中进行透析。在所需的时间间隔,对2mL透析液进行采样,然后将新鲜溶液(2mL)添加到透析液中。通过确定相对于其标准校准曲线的吸光度来计算Cur浓度。通过ICPMS检测铂含量。
细胞培养
人宫颈癌细胞系(HeLa)购自中国科学院上海生物化学与细胞生物学研究所。将细胞在含10%热灭活的FBS,100IU/mL青霉素-链霉素的完全的DMEM培养基中培养,并在37℃与5%CO2孵育。每2天更换新鲜DMEM培养基以保持指数增长。
细胞毒性试验
通过标准MTT测定法评估细胞毒性。选择HeLa细胞系作为模型细胞。将测试分为以下四组:顺铂、Cur、DHP和PCPt NP。简而言之,在96孔板的每个孔中,将5000个细胞接种在100μLDMEM培养基中,并将细胞培养24小时以允许细胞附着。用新鲜的DMEM补充培养基,然后将最终Pt/姜黄素浓度为6.25至216μM的每种样品药物与细胞共培养。孵育48或72小时后,将培养基改回新鲜的DMEM,然后添加20μLMTT试剂(PBS 7.4中5mg/mL)。将细胞再孵育4小时。之后,小心吸出培养基,使用150μL DMSO溶解由活细胞产生的紫色MTT-甲酰胺晶体,并使用酶标仪在490nm下测量甲瓒产物的吸光度。将没有药物共孵育的细胞设置为对照。
结合指数(CI)的计算公式如下:
ICx,a和ICx,b是单独使用以产生x%细胞杀伤力的药物A(DHP)和药物B(Cur)的浓度。Ca,x是与Cur组合达到相同x%作用的DHP的浓度,而Cb,x是与DHP组合达到相同x%细胞杀伤的Cur的剂量。小于,等于和大于1的CI值分别表示协同作用,加和作用和拮抗作用。
体外细胞吸收
CLSM跟踪细胞对游离Cur和PCPt NP的摄取。将HeLa细胞接种到6孔板中,在2mLDMEM培养基中以每孔2×105个细胞的密度接种,并孵育过夜。然后加入游离的Cur和PCPtNP溶液(500μL),分别培养0.5或4h。Cur的最终浓度固定为2μg/mL。在用CLSM表征之前,将细胞用冷PBS 7.4冲洗3次以去除药物,用4%多聚甲醛(在PBS 7.4中固定)固定20分钟,然后用Hoechst 33258(10μg/mL)将细胞核染色10分钟,最后,用冷PBS 7.4洗涤细胞3次,并用CLSM记录。
此外,通过流式细胞术对体外游离Cur和PCPts的细胞摄取进行定量。将HeLa细胞以每孔3×105个细胞的浓度在2mL DMEM培养基中种植到6个孔板中。孵育24小时后,加入溶于500μLPBS中的游离Cur和PCPts(Cur的终浓度固定为15μg/mL),再培养1或4h。然后,除去原始培养基,并用PBS洗涤细胞3次。用胰蛋白酶使细胞分离,通过离心收集。最后,将细胞重悬于500μL PBS中,并通过流式细胞仪进行研究。
结果和讨论
PCPt的合成和表征
Cur具有抗感染、抗炎和抗癌的特性。Cur可以减轻顺铂引起的副作用,并增强顺铂的抗癌作用。如图2所示,在该研究中,将Pt(IV)前药(DHP)和Cur与LDI共聚以构建疏水性嵌段,然后用亲水性mPEG5k-NCO终止,从而获得两亲性三嵌段PCPt。
单体和共聚物的化学结构通过1H NMR光谱表征。1H NMR结果证实了DHP和mPEG5k-NCO的制备(图9)。图3a显示了PCPt共聚物的1H NMR光谱,并且可以清楚地观察到Cur、Pt(IV)、LDI和PEG的所有特征信号。另外,在图10中还提供了所有单体和共聚物的FT-IR光谱。通过GPC测定,PCPt的分子量和分子量分布分别为17.4kDa和1.51(图3b)。此外,由于Pt(IV)固有的还原敏感性,GPC还用于表征在10mM L-抗坏血酸钠(SA)条件下PCPt的降解。如图3c所示,在与10mM SA孵育12小时后,GPC曲线变宽,并且出现了一系列低分子量物质,这表明PCPt的还原活化解离。
自组装行为
PCPt是一种两亲性ABA三嵌段聚合物,可以在选择性溶剂中组装。首先,进行1HNMR光谱研究自组装行为。如图4a所示,与DMSO-d6中的1H NMR谱图相比,一个在3.51ppm处的信号分配给了mPEG5k(-OCH2CH2-)的峰,而PCPt的所有其他峰在D2O中消失了,这说明PCPt可以在水溶液中自组装成PCPt NP。为了进一步研究自组装性能,使用芘作为荧光探针测量了PCPt的CAC值。确定PCPt NP的CAC为7.94×10-3mg/mL(图4b)。由于低的CAC值,PCPt NP在水溶液中更稳定,并且能够在体内长时间循环。接下来,通过透析方法制备PCPt NP。通过DLS分析得到的PCPt NP的平均粒径和粒径分布分别约为100nm和0.19(图4c)。结果表明,PCPt NPs小而均一,有利于延长血液循环和细胞内吞作用。如图4d所示,TEM结果表明PCPtNP是单个球形颗粒,与DLS结果一致。所有这些结果为自组装过程提供了明显的证据。
稳定性表征
本实施例中,PCLt NPs的粒径和多分散指数(PDI)通过DLS在各种生理条件下在不同时间进行体外表征(图5a和5b)。根据DLS结果,即使在孵育72小时后,PBS 7.4中PCPt NP的平均大小和PDI也没有明显差异。但是,在存在10mM SA的情况下,PCPt NP的粒径和PDI都发生了巨大变化,粒径在12小时内从103nm快速增加到911nm,然后逐渐减小,这可能是由于骨架的降解。接下来,通过TEM进一步研究刺激响应性,相应的尺寸和形态变化。在图5c中,对于在PBS 7.4中孵育24h的PCPt NP,可以清楚地观察到平均直径约为108nm的球形纳米颗粒。相反,在10mM SA中孵育24小时后,形态和分布发生了很大变化,这进一步证实了PCPtNPs的还原敏感性。
游离的姜黄素是一种疏水性抗癌药物,其在水中的饱和浓度约为0.6μg/mL,纳米级姜黄素可表现出更高的溶解度和稳定性(C.Tian,S.Asghar,Y.Xu,Z.Chen,M.Zhang,L.Huang,J.Ye,Q.Ping,Y.Xiao,Colloids Surf.B,2018,165,45-55.和Y.Wu,X.Wang,Int.J.Food Prop.,2017,20,3295-3307.)。如图5d所示,纳米级PCPt NPs溶液是均匀的,可以维持一个月没有沉淀,而游离姜黄素溶液是不均一的,静置10分钟后可以清楚地观察到可见的沉淀。相对于游离Cur、PCPt NPs形式的共轭Cur显示出更高的水溶性和稳定性。更重要的是,纳米颗粒的生理稳定性是药物递送中至关重要的问题。为了增加PCPt NP和PBS(pH7.4)中游离Cur的生理稳定性,可通过紫外可见光谱法监测其吸光度的变化。图11显示了PCPt NP的UV-Vis光谱和荧光光谱。用Cur在λmax=425nm附近的吸光度评估其化学稳定性。如图5e和图12所示,在黑暗中孵育7小时后,游离Cur的降解速度确实很快,并且Cur含量仅保持59.9%。相比之下,PCPt NPs的吸光度变化要比游离Cur慢得多,在相同条件下7h后PCPt NPs中94%的Cur保持正常。这些结果表明,纳米颗粒的缀合和形成可以保护游离的Cur免于降解,并且Cur的稳定性显着提高。
铂和姜黄素释放
为了实现联合化疗,将顺铂衍生物与姜黄素共聚以产生密码释放系统(PCPtNPs)。Pt和Cur的载药量(DLC)分别约为9.7%和9.1%。结果与进料比例一致。由于Pt(IV)被共轭到PCPt的骨架上,PCPt NPs的药物释放行为将取决于还原。使用透析方法在PBS(pH7.4)或10mM SA中研究了PCt NP中Pt和Cur的释放。如图6a所示,在pH 7.4下检测到相当缓慢的Pt释放,在72小时后仅释放了约18.8%的Pt,这表明PCPt聚合物和NP在中性条件下是稳定的。不出所料,添加10mM SA后Pt的释放速率显着增加,72h后Pt的累积释放量达到93.3%,这证明Pt的释放是还原依赖性的,并且PCPt NPs可以在60h降解。基于Pt(IV)破坏的还原性环境。Cur释放的结果类似于Pt释放(图6b),在还原条件(10mM SA)下,PCPt NPs的Cur释放明显增强。与10mM SA孵育72小时后,PCPt NPs释放出约83.3%的Cur,在pH 7.4时仅释放14%的Cur。因此,对还原反应敏感的PCPt NP在开发用于癌症联合治疗的智能载体方面具有巨大潜力。
体外细胞毒性和细胞摄取
顺铂广泛用于临床中各种实体恶性肿瘤的治疗。但是,成功应用顺铂的主要障碍是对正常细胞的严重副作用以及无法靶向癌细胞。此外,与单药治疗相比,本发明的PCPtNP对于联合癌症治疗的最理想效果是改善了生物安全性并增强了细胞毒性。然后,通过MTT分析评估顺铂、DHP、Cur和PCPt NP对肿瘤细胞系Hela细胞的细胞毒性。如图5所示,所有药物均以剂量和时间依赖性方式诱导HeLa细胞的细胞毒性。随着浓度的增加,细胞活力逐渐降低,孵育时间延长。在图7a中显示的生存力值表明,在培养48小时后,顺铂对HeLa细胞系表现出最高的细胞毒性,IC50值约为22.5μM。游离Cur在相同浓度下表现出最大的细胞活力。此外,与顺铂相比,用DHP培养的HeLa细胞在6.25–216μM的浓度范围内具有更高的细胞活力,这表明Pt(IV)具有更高的安全性。在相同条件下,PCPt NPs的抗癌活性低于顺铂。原因是将Pt(IV)前药还原为活性Pt(II)需要更多时间,也可能是由于药物释放相对缓慢。重要的是,与DHP和Cur相比,PCPt NP对HeLa细胞显示出增强的细胞毒性。特别是,对于50%的细胞抑制作用,HeLa细胞的组合指数(CI)值计算为0.74,这证实了PCPt NP中铂和Cur的协同作用。孵育72小时,观察到相似的趋势(图7b)。
利用Cur分子固有的荧光特性,使用CLSM对HeLa细胞中游离Cur和PCPt NP的细胞摄取和细胞内分布进行成像。如图8所示,可以清楚地看到,与游离Cur共孵育0.5h后,主要在细胞质中仅检测到弱的黄色荧光(Cur)信号,在PCPt NP中也发现了类似共孵育的HeLa细胞。胞内摄取的游离Cur和PCPt NPs表现出时间依赖性。延长4h的孵育时间后,游离Cur的吸收得到增强,不仅在细胞核中而且在细胞质中均观察到荧光。值得注意的是,对于与PCPtNPs孵育的HeLa细胞,荧光密度从0.5h显着增加到4h,Cur荧光大部分位于细胞的核周围和核区域。换句话说,PCPt NPs逐渐将Cur释放到细胞质,然后扩散到细胞核。Cur的不同胞内分布可能是由于内体中PCPt NP的Pt(IV)还原为Pt(II)。这些成像结果表明,与Cur较差的水溶性和稳定性相比,PCPt NPs易于在还原环境中被癌细胞内化并释放药物。结果表明,PCPt NPs对HeLa细胞表现出有效的细胞内摄取和增强的细胞增殖抑制作用。

Claims (8)
1.一种双前药骨架聚合物PCPt,其组成原料包括Cur、DHP、LDI和mPEG衍生物;
所述PCPt为两亲性三嵌段结构,其中,Cur、DHP、LDI共聚形成主链,构建疏水性嵌段,聚乙二醇单甲基醚衍生物为亲水性链终止剂;
所述聚乙二醇单甲基醚衍生物为异氰酸酯官能化的聚乙二醇单甲醚;
Cur、DHP和LDI的摩尔比为1-6:1-6:2-13。
2.一种制备权利要求1所述的双前药骨架聚合物PCPt的制备方法,其包括由Cur、DHP和LDI原位聚合后与聚乙二醇单甲基醚衍生物偶联。
3.根据权利要求2所述的方法,其特征在于,Cur、DHP和LDI的摩尔比为5:4:8。
4.一种PCPt纳米颗粒,其由权利要求1所述的双前药骨架聚合物自组装得到。
5.根据权利要求4所述的PCPt纳米颗粒,其特征在于,所述自组装溶剂为水。
6.根据权利要求4或5所述的PCPt纳米颗粒,其特征在于,所述纳米颗粒的平均直径为90-110nm。
7.一种药物递送系统或药物载体,其包含权利要求1所述的双前药骨架聚合物PCPt或权利要求4至6中任一项所述的PCPt纳米颗粒。
8.权利要求1所述的双前药骨架聚合物PCPt或权利要求4至6中任一项所述的PCPt纳米颗粒或权利要求7中所述的药物递送系统或药物载体在制备治疗肿瘤的药物中的应用;或者在制备实现联合化疗的药物中的应用。
