本发明提供了一种防治饱和脂肪酸诱导的代谢综合征的药物。本发明通过实验证明,2‑溴十六烷酸能够抑制高饱和脂肪酸饮食引起的内质网应激、炎性反应、细胞凋亡以及胰岛素抵抗,可用于制备预防和对抗肥胖或高脂饮食引起的代谢综合症。

技术领域:
本发明涉及一种防治饱和脂肪酸诱导的代谢综合征的药物。
背景技术:
因摄入脂肪过多造成的血脂增高,引起诸如脂肪肝、高血糖、高胰岛素血症、高血压、糖耐量下降、胰岛素抵抗以及肥胖等病理改变,统称为代谢综合症。研究表明,血液中饱和脂肪酸浓度增高是引起代谢综合症的主要元凶。
内质网是细胞内重要细胞器,可参与蛋白质的折叠与修饰。饱和脂肪酸会诱发组织细胞发生内质网应激(即内质网正常功能紊乱),引起组织慢性炎症反应、胰岛素抵抗以及细胞凋亡等病理损伤。因此,内质网应激在代谢综合症的发病过程中处于关键核心地位(Ozcan,U.,et al.Science.2004)。已知活化内质网膜上的三个跨膜蛋白激活下游标记物ATF4以及CHOP,进而促进分子伴侣的合成,能够恢复内质网的内稳态,缓解内质网应激(Maurel,M.,et al.Am J Physiol Cell Physiol.2013)。
因此,抑制饱和脂肪酸诱导的内质网应激表达的化合物,或者可以抑制饱和脂肪酸诱导的病理损伤的物质,可以制备用于预防和治疗高脂饮食引起的代谢综合症的药物。
棕榈酸常被用于诱导内质网应激研究饱和脂肪酸对细胞的损伤研究。
但迄今为止,未见有关2-溴十六烷酸对饱和脂肪酸诱导的内质网应激及其病理损伤作用的报道。
发明内容:
本发明的目的是提供一种抑制饱和脂肪酸诱导的内质网应激、炎性反应、凋亡和胰岛素抵抗等代谢综合征的药物。
本发明首次发现,2-溴十六烷酸能够达到上述发明目的。
2-溴十六烷酸(式I)是已知化学结构的化合物,英文名2-bromohexadecanoicacid,分子式CH3(CH2)13CH(Br)CO2H,CAS登录号18263-25-7。
本发明用以下四个实验充分证实了2-溴十六烷酸的上述新功能。四个实验均采用了国际公认的饱和脂肪酸处理模型、实验方法和检验标准:
1、2-溴十六烷酸可以减轻饱和脂肪酸诱导的内质网应激
为验证2-溴十六烷酸对饱和脂肪酸诱导的内质网应激的抑制效应,我们体外培养成肌细胞系C2C12,诱导形成成熟肌管后,分为对照组、棕榈酸(作用浓度为500μM)单独处理组、2-溴十六烷酸(作用浓度为500μM)单独处理组、棕榈酸及2-溴十六烷酸合并处理组;处理肌管12小时后,收集细胞样本提取蛋白质,利用Western blot方法分析了2-溴十六烷酸对内质网应激标志性蛋白ATF4及CHOP表达的影响。
实验结果表明,对照组肌管内ATF4及CHOP表达非常低,棕榈酸处理12小时后诱导肌管内ATF4及CHOP表达急剧增加,说明诱导了明显的内质网应激反应;2-溴十六烷酸与棕榈酸同时处理时,可以明显抑制棕榈酸诱导的ATF4及CHOP表达的增加幅度,具有显著抑制棕榈酸诱导的内质网应激反应的作用。

2、2-溴十六烷酸可以减轻饱和脂肪酸诱导的炎性反应
为验证2-溴十六烷酸对饱和脂肪酸诱导的炎性反应,我们体外培养成肌细胞系C2C12,诱导形成成熟肌管后,收集对照组、棕榈酸(作用浓度为500μM)单独处理组、2-溴十六烷酸(作用浓度为500μM)单独处理组、棕榈酸及2-溴十六烷酸合并处理组蛋白质,利用Western blot方法分析了2-溴十六烷酸对炎性反应标志物TNF-alpha及IL-6表达的影响。
实验结果表明,对照组肌管内TNF-alpha表达较低,棕榈酸处理12小时后诱导肌管内TNF-alpha及IL-6表达急剧增加,说明诱导了明显的炎性反应;2-溴十六烷酸与棕榈酸同时处理时,可以明显抑制棕榈酸诱导的TNF-alpha及IL-6表达的增加幅度,具有显著抑制棕榈酸诱导的炎性反应的作用。

3、2-溴十六烷酸可以减轻饱和脂肪酸诱导的细胞凋亡
为验证2-溴十六烷酸对饱和脂肪酸诱导的细胞凋亡,我们体外培养成肌细胞系C2C12,诱导形成成熟肌管后,收集对照组、棕榈酸(作用浓度为500μM)单独处理组、2-溴十六烷酸(作用浓度为500μM)单独处理组、棕榈酸及2-溴十六烷酸合并处理组蛋白质,利用Western blot方法分析了2-溴十六烷酸对细胞凋亡标记物Caspase 3剪切体表达的影响。
实验结果表明,对照组肌管内几乎不表达Caspase 3剪切体,棕榈酸处理12小时后诱导肌管内Caspase 3剪切体表达增加,说明诱导了明显的细胞凋亡;2-溴十六烷酸与棕榈酸同时处理时,可以明显抑制棕榈酸诱导的Caspase 3剪切体的增加幅度,具有显著棕榈酸诱导的细胞凋亡的作用。

4、2-溴十六烷酸可以减轻饱和脂肪酸诱导的胰岛素抵抗
为验证2-溴十六烷酸对饱和脂肪酸诱导的细胞凋亡,我们体外培养成肌细胞系C2C12,诱导形成成熟肌管后,收集经胰岛素处理或不处理对照组、棕榈酸(作用浓度为500μM)单独处理组、2-溴十六烷酸(作用浓度为500μM)单独处理组、棕榈酸及2-溴十六烷酸合并处理组蛋白质,利用Western blot方法分析了2-溴十六烷酸对胰岛素诱导的下游磷酸化AKT表达的影响。
实验结果表明,对照组细胞胰岛素处理后磷酸化AKT迅速增多,而棕榈酸处理细胞经胰岛素处理后磷酸化AKT增加幅度显著降低,说明诱导了明显的胰岛素抵抗;2-溴十六烷酸与棕榈酸同时处理时,可以明显增加胰岛素处理后磷酸化AKT的表达水平,具有显著逆转棕榈酸诱导的胰岛素抵抗的作用。

本发明的有益效果:
本发明通过实验首次发现和证实,2-溴十六烷酸具有以下作用:
1、缓解饱和脂肪酸诱导的内质网应激;
2、抑制饱和脂肪酸诱导的炎性因子的表达;
3、抑制饱和脂肪酸诱导的细胞凋亡;
4、抑制饱和脂肪酸诱导的胰岛素抵抗。
由于本领域公知抑制内质网应激的发生是防止和改善肥胖或高脂饮食诱导的代谢综合症的先决条件。因此,本发明提供了一种可用于制备预防和治疗饱和脂肪酸诱导的代谢综合征的药物2-溴十六烷酸。

具体实施方式
以下实验中所用的实验动物及材料来源如下:
C2C12细胞系,购自国家生物医学实验细胞资源库。
受试药物:2-溴十六烷酸,购买自西格玛奥德里奇(上海)贸易有限公司,纯度:97%,货号238422。
其它材料来源:胎牛血清、高糖DMEM培养基、马血清、ATF4、CHOP、TNF-alpha、IL-6、Caspase 3剪切体及磷酸化AKT抗体购自Cell Signaling Technology公司;
主要仪器设备:伯乐电泳仪,电转槽。
实验一:2-溴十六烷酸对饱和脂肪酸诱导的内质网应激的影响
1.实验目的:
考察2-溴十六烷酸对饱和脂肪酸诱导的内质网应激的影响。
2.实验方法:
体外培养成肌细胞系C2C12,诱导形成成熟肌管后,分为对照组、棕榈酸(作用浓度为500μM)单独处理组、2-溴十六烷酸(作用浓度为500μM)单独处理组、棕榈酸及2-溴十六烷酸合并处理组,处理12小时后提取蛋白质,利用Western blot方法检测内质网应激标记物ATF4和CHOP的表达。
2.1饱和脂肪酸处理体外细胞模型的制备
C2C12细胞接种至细胞培养板中,利用生长液(高糖DMED+10%胎牛血清+1%双抗)培养至95%密度,换分化液(高糖DMED+2%马血清+1%双抗)培养四天后诱导成熟肌管,分为四组:对照组、棕榈酸(作用浓度为500μM)单独处理组、2-溴十六烷酸(作用浓度为500μM)单独处理组、棕榈酸及2-溴十六烷酸合并处理组,处理12小时后提取细胞总蛋白。
2.2蛋白的提取、检测与定量
A.将收集的细胞沉淀加入一定体积的蛋白裂解液,冰上裂解20-30min;
B.待样品充分裂解后,4℃下10,000~14,000x g离心5分钟,吸取上清液;
C.利用Bradfford法检测蛋白浓度。
2.3SDS-PAGE聚丙烯酰胺凝胶电泳
A.按照上述配方配制12%的分离胶和5%的积层胶;
B.电泳前将每组蛋白样品加入5×凝胶加样缓冲液,沸水变性5~10min;
C.蛋白冷却后按照每孔总蛋白30μg上样;
D.在积层胶按照80V恒压电泳,进入分离胶后加大电压180V,电泳至凝胶底部;
E.停止电泳,取下凝胶,放入电转缓冲液中浸润平衡5min。2.3引物设计2.4转膜(湿转)
A.取8×6cm大小的厚滤纸和NC膜,在预冷的电转缓冲液中浸润平衡5min;
B.在Bio-Rad湿式电转仪的正极板上按照夹心法依次放置转膜垫、滤纸、NC膜、凝胶、滤纸及转膜垫,尽量赶净各层间的气泡,盖负极极板;
C.以60V的恒压进行电转,时间控制在60-90min;
D.电转结束后,将膜置于TBS中漂洗3次,每次5min。
2.5抗原抗体孵育反应
A.封闭:将NC膜放入杂交袋内,加入5%的脱脂奶粉,室温轻摇2h;
B.一抗反应:倒掉封闭液,按加入用5%的脱脂奶粉稀释的ATF4或CHOP抗体,4℃
轻摇过夜;
C.漂洗:将NC膜取出,用TBST洗3次,每次10min;
D.二抗反应:将NC膜放入杂交袋内,加入5%的脱脂奶粉稀释的二抗,在室温孵育2h;
E.漂洗:取出NC膜,用TBST洗3次,每次10min。
2.6ECL显色
A.按每平方厘米印迹膜使用0.1-0.2ml配置发光液:根据计算的发光液需要量,用洁净吸管或枪头分别吸取等体积的A液和B液,于洁净EP管内混匀备用;
B.用平头镊子将印迹膜取出,吸水纸吸去多余的漂洗液;将印迹膜的置于洁净保鲜膜上,膜的正面(蛋白面)朝上,将配制的ECL发光液转移到印迹膜上,充分覆盖膜的表面,室温孵育5分钟;
C.用平头镊夹持印迹膜,祛除多余的发光检测液后,平放在新的干净保鲜膜上,上面再覆盖一层保鲜膜,轻轻驱赶保鲜膜与印迹膜之间的气泡,用胶带将包裹好的印迹膜固定在X片暗盒内;
D.在暗室中取一张X光片置于包裹的膜上,合上暗盒,曝光时间可由几秒至数小时不等(操作者可根据经验自行决定),之后立即进行显影和定影。对微弱信号,曝光时间可延长至数小时;也可用配备化学发光滤片的照相器材直接记录蛋白膜的化学发光图像。E.显影结束后可以通过分辨X光胶片上印迹膜的四周边界,从而验证目的蛋白的大小和位置。
3.数据分析方法
数据以平均值±标准差表示,数据分析采用组间方差分析。***P<0.001,###P<0.001,#P<0.05表示具有显著性统计学差异。
4.实验结果
如图1,图2和图3所示。实验结果表明,对照组肌管内ATF4及CHOP表达非常低,棕榈酸处理12小时后诱导肌管内ATF4增加6.84倍(***P<0.001),CHOP表达增加10.83倍(***P<0.001),说明诱导了明显的内质网应激反应;2-溴十六烷酸单独处理与对照组类似,肌管内ATF4及CHOP表达水平较低,但与棕榈酸同时处理时,ATF4增加4.54倍(***P<0.001),CHOP表达增加2.16倍,与棕榈酸单纯处理组相比,2-溴十六烷酸与棕榈酸同时处理,ATF4增加幅度减轻33%(##P<0.05),CHOP表达增加幅度减轻81%(###P<0.001);具有显著棕榈酸诱导的内质网应激反应的作用。
5.实验结论
2-溴十六烷酸可显著抑制棕榈酸诱导的内质网应激。
实验二:2-溴十六烷酸对饱和脂肪酸诱导的炎性反应的影响
1.实验目的:
考察2-溴十六烷酸对饱和脂肪酸诱导的炎性反应的影响。
2.实验方法:
具体方法同实验一,区别在于利用Western blot方法检测炎性反应标记物TNF-alpha及IL-6的表达。
3.数据分析方法
数据以平均值±标准差表示,数据分析采用组间方差分析。***P<0.001,###P<0.001,#P<0.05表示具有显著性统计学差异。
4.实验结果
如图1,图4所示,实验结果表明,对照组肌管内TNF-α表达非常低,棕榈酸处理12小时后诱导肌管内TNF-α增加7.37倍(***P<0.001),说明诱导了明显的炎性反应;2-溴十六烷酸单独处理与对照组类似,肌管内TNF-α表达较低,但与棕榈酸同时处理时,TNF-α增加1.48倍,与棕榈酸单纯处理组相比,2-溴十六烷酸与棕榈酸同时处理后TNF-α增加幅度减轻79%(###P<0.001),具有显著棕榈酸诱导的炎性反应的作用。
5.实验结论
2-溴十六烷酸可显著抑制棕榈酸诱导的炎性反应。
实验三:2-溴十六烷酸对饱和脂肪酸诱导的细胞凋亡的影响
1.实验目的:
考察2-溴十六烷酸对饱和脂肪酸诱导的细胞凋亡的影响。
2.实验方法:
具体方法同实验一,区别在于利用Western blot方法检测炎性反应标记物Caspase 3剪切体(Cleaved Caspase 3)的表达。
3.数据分析方法
数据以平均值±标准差表示,数据分析采用组间方差分析。***P<0.001,###P<0.001表示具有显著性统计学差异。
4.实验结果
如图1和图5所示,实验结果表明,对照组肌管内几乎不表达Caspase 3剪切体,棕榈酸处理12小时后诱导肌管内Caspase 3剪切体迅速增加18.2倍(***P<0.001),说明诱导了明显的细胞凋亡反应;2-溴十六烷酸单独处理组也无Caspase 3剪切体的表达,与棕榈酸同时处理时,Caspase 3剪切体的表达量与棕榈酸单纯处理组相比减少87%(###P<0.001),说明2-溴十六烷酸可以抑制棕榈酸诱导的细胞凋亡的作用。
5.实验结论
2-溴十六烷酸可显著抑制棕榈酸诱导的细胞凋亡。
实验四:2-溴十六烷酸对饱和脂肪酸诱导的胰岛素抵抗的影响
1.实验目的:
考察2-溴十六烷酸对饱和脂肪酸诱导的胰岛素抵抗的影响。
2.实验方法:
具体方法同实验一,区别在于实验分组为八组,四组同实验一,另外四组是在实验一分组的基础上、收集样本前10分钟加入100nmol胰岛素刺激,之后提取蛋白利用Westernblot方法检测磷酸化AKT蛋白的表达。
3.数据分析方法
数据以平均值±标准差表示,数据分析采用组间方差分析。***P<0.001,###P<0.001表示具有显著性统计学差异。
4.实验结果
如图6和图7所示,实验结果表明,对照组细胞胰岛素处理后磷酸化AKT迅速增加19.85倍,说明对照组细胞胰岛素敏感性较强;而棕榈酸处理细胞经胰岛素处理后磷酸化AKT增加幅度为11.53倍,与对照组相比诱导了明显的胰岛素抵抗(***P<0.001);2-溴十六烷酸单纯处理即可明显提高细胞胰岛素敏感性,胰岛素刺激后磷酸化AKT迅速增加28.9倍;2-溴十六烷酸与棕榈酸同时处理时,胰岛素处理后磷酸化AKT表达水平仍然增加20.63倍,与对照组水平相当,和棕榈酸处理比较,显著逆转棕榈酸诱导的胰岛素抵抗的作用(###P<0.001)。
5.实验结论
2-溴十六烷酸可显著抑制棕榈酸诱导的胰岛素抵抗。
