技术领域
本发明属于食品技术领域,具体涉及亚马逊莓在制备有助于排铅作用的食品或保健品中的用途。
背景技术
铅属于三大重金属污染物之一,是一种严重危害人体健康的重金属元素,人体中理想的含铅量为零。随着环境污染的日趋严重,自然中的铅可以通过食物、饮用水等方式进入人体,其中大部分铅累积在骨骼,剩余少部分随血液循环流动而分布到全身各组织和器官,影响血红细胞和脑、肾、神经系统功能,特别是婴幼儿吸收铅后,将有超过30%保留在体内,影响婴幼儿的生长和智力发育。
铅在人体内可累积,当人体中的铅含量达到一定程度时,会引发身体的不适,此时往往已经产生了难以逆转的身体损伤。
目前防治铅蓄积的基本原则是环境干预、健康教育和临床干预三者相结合,一般情况下,只有当血铅浓度≥0.9μmol/L时,才考虑给予药物干预。临床上的药物主要有二巯基丁二酸钠、依地酸二钠钙等,这些药物虽然排铅效果优良,但会使人体流失部分必需微量元素,且会对肾脏造成损害,副作用较大,此外,这些驱铅药物对铅蓄积造成的神经损害逆转意义不大,效果甚微。
因此,在日常生活中避免铅的摄入以及促进铅的排泄成为了避免铅蓄积及其损害的最佳途径。
此外,亚马逊莓(Acai Berry),又称巴西莓,主要分布在南美亚马逊河流域,因其卓越的抗氧化能力而被人们熟知。亚马逊莓含有不饱和脂肪酸、植物甾醇、氨基酸、维生素、矿物质以及微量元素等多种营养物质,也含有多种功能活性物质,主要包括黄酮类、花青素类、酚酸等多种多酚类物质。
已知的是,人们对亚马逊莓的活性和功能已多有研究,目前公开的包括抗氧化活性、抗炎、免疫调节、皮肤美白、解酒保肝、抗基因毒性以及抗癌等活性和功能。例如:
引用文献1公开了一种含有巴西莓提取物的组合物,在该组合物中巴西莓提取物和卡卡杜李提取物可以单独或者协同作用,减少皮肤氧化性损伤,增加皮肤胶原的产生,以及减少炎症应答;
引用文献2公开了巴西莓具有的保肝作用,其对急性肝缺血再灌注损伤、慢性肝损伤等都具有良好的治疗效果。
尽管作为保健品、食品中已经应用到亚马逊莓或其加工物,但对于亚马逊莓的各种功效的研究仍然存在可待开发的空间,另外,亚马逊莓对于生物体促排铅功能方面尚没有可证实的公开信息。

发明内容
发明要解决的问题
基于以上现有技术中对于亚马逊莓已经开展的探索,本发明在对亚马逊莓可能的生物学功效进行研究时,意外地发现了亚马逊莓或其提取物(活性成分)对促进生物体排铅具有明显的功效。
因此,本发明首要的目的在于提供一种亚马逊莓或其提取物(活性成分)的新的用途,具体而言,提供了一种亚马逊莓或其提取物(活性成分)在制备有助于排铅作用的食品或保健品中的用途。本发明以斑马鱼为研究对象,从促进铅排泄和改善由铅累积导致的贫血两个方面对亚马逊莓的促排铅功能进行验证和展示。
另外,进一步的研究也显示了亚马逊莓或其提取物(活性成分)在改善由于铅累积所导致的贫血方面也具有可验证的效果。
此外,本发明也提供了一种包含亚马逊莓或其提取物(活性成分)的食品或保健品在促进生物体排铅以及改善或缓解由于铅累积引起的贫血的用途。

4)本发明提供了一种关于亚马逊莓或其提取物成分的新的应用方式,为亚马逊莓及其类似植物的应用研究提供了新的思路。

除非另有定义,本发明所用的其他的技术和科学术语具有与本发明所属技术领域中的普通技术人员所通常理解的相同含义。
本发明所述的亚马逊莓是美国、拉美地区一种棕榈树上长出的果实,又名巴西莓(Euterpe edulis Mart.、Euterpe precatoria Mart.、Euterpe oleracea Mart.)。本发明所述的亚马逊莓包括但不限于亚马逊莓鲜果、亚马逊莓干果、亚马逊莓冻果、亚马逊莓冻干、亚马逊莓果浆或其他食品上可接受的形态。
本发明通过生物学研究发现了亚马逊莓或其提取物(有效成分)一种新的功效,并基于此研究,拓展了现有的对亚马逊莓的应用方式。本发明主要是基于如下见解而得到:
本发明以斑马鱼为研究对象,对其使用含有一定浓度的亚马逊莓提取物的水进行饲养,并在亚马逊莓提取物的安全浓度范围内,研究了水体含有铅元素情况下所饲养的斑马鱼的生物学变化,通过实际实验数据,发现了亚马逊莓对于生物体具有减少铅累积和排铅的效用。
(亚马逊莓或其提取物)
本发明中,可以直接使用上述亚马逊莓果实或其处理物。在一些典型的实施方案中,可以将该果实粉碎(任选地包括打浆、均质等处理)得到浆状物,或者将这些果实去除大部分的水分以得到基本干燥的加工物(例如干燥的块状物、粉末状物或者冷冻干燥物等),另外,也可以将亚马逊莓果实在必要的菌株的存在下进行发酵而得到发酵物。对于这些果实或其处理,可以使用一种也可以使用它们任意形式或比例的混合物。通常,这些果实或其处理物可以在例如杀菌等必要处理后直接使用,或者,也可以经冷冻保存后使用。
此外,本发明中,还可以使用亚马逊莓的提取物。所谓提取物,即通过任选的提取或加工手段分离、浓缩而得到的含有亚马逊莓活性物质或有效物质的组分。这些活性物质主要包括花青素、原花青素,多种类黄酮(黄酮类化合物)以及多种不饱和脂肪酸,包括油酸,亚油酸,棕榈酸等。
所述的亚马逊莓提取物包括但不限于其水提取物或醇提取物。在本发明一些具体的实施方案中,可以将新鲜的亚马逊果实经过制浆机制浆后,将得到的浆料与提取溶剂(水、醇或它们的混合物)进行混合,并在加热的情况下进行有效成分的提取。对于所述溶剂的用量,没有特别限制,通常可以以浆料的5~50倍(体积)的量而使用。在一些优选的实施方案中,所述加热可以在60~100℃的条件下进行,加热时间可以为1~3小时。过高的加热温度或过长的时间可能导致有效成分的氧化变质。在加热处理后,可以将固液成分分离,进一步,可以将液体成分进行干燥以得到固体(例如粉末状)或半固体(含水浆料)的提取物。此外,还可以根据需要在固液分离之前或之后对混合体系进行吸附脱色处理,也可以在得到最终的提取物后进行杀菌消毒处理等。
本发明中,对于亚马逊莓提取物的最终形态没有特别限制,例如为液体(借助所要的溶剂)、半固体(含水浆料)、粉末、颗粒或块状物(经压制)等。在一些具体的实施方案中,本发明所述的亚马逊莓提取物包含丰富的多酚类化合物、氨基酸、脂肪酸、维生素等营养物质。
(可食用组合物)
本发明进一步提供了一种可食用组合物,并且该组合物包括以上所述的亚马逊莓或其提取物。
在一些具体的实施方案中,所述组合物中,亚马逊莓或其提取物可以以干燥物、溶液或浆料(果浆)的方式而使用,优选地,以含水浆料的方式而使用。这些含水浆料通常具有8质量%以上、优选为10质量%、进一步优选为14质量%以上的可溶性固体成分,并具有粘稠的外观。
对于本发明所述组合物,除了上述必要组分以外,还可以根据最终产品的需要,可以包括其他任选的成分,可以列举的包括:
含乳成分或蛋白成分,所述含乳成分包括乳品包括以来源于生鲜牛(羊)乳的鲜乳、乳粉、乳清蛋白或奶酪等;对于蛋白成分可以来自于植物蛋白,例如大豆蛋白、花生蛋白等。
植物或植物提取成分,包括无花果、石榴、猕猴桃、橘子、橙子、菠萝、草莓、苹果、橡胶、葡萄、梨、樱桃、蓝莓、黑莓、覆盆莓和山桑子等水果或它们的提取物;洋蔥、黄瓜、番茄、花椰菜、红萝卜、菠菜、芥蓝、球芽甘蓝、大蒜、罗勒、奧勒冈草等果蔬类物质或它们的提取物;稻类(籼稻、粳稻、糯稻)、麦类(小麦、大麦、燕麦、黑麦)、玉米、高粱、粟、黍、黄米、荞麦、大豆、蚕豆、豌豆、绿豆、红小豆、芸豆等谷物或它们的提取物;核桃、开心果、腰果、榛子、扁桃、杏仁、松子、花生、瓜子、板栗、夏威夷果、白果等坚果类物质或它们的提取物;咖啡或其提取物。
动物类成分,包括牛、羊、鱼或禽类的肉制品成分。
脂肪成分,所述脂肪可以包括饱和脂肪酸、多不饱和脂肪酸、单不饱和脂肪酸、OPO结构脂、DHA、EPA、ARA、磷脂中的至少一种,更具体地说所述脂肪包括红花籽油、核桃油,花生油,大豆油,阿甘油、橄榄油、茶油、美藤果油、橄榄油、椰子油、紫苏油、深海鱼油、可可油、棕榈油、牛油、奶油、猪油、中链甘油三酯或卵磷脂等。
功能性添加组分,包括维生素(维生素A、β-胡萝卜素、维生素D3、维生素E、维生素K1、维生素B1、维生素B2、维生素B6、维生素B12、维生素C、泛酸、叶酸、烟酸、胆碱、肌醇、生物素中的一种或多种)、淀粉、改性淀粉、氨基酸(L-赖氨酸-L-谷氨酸、L-谷氨酸、L-精氨酸、L-色氨酸、L-谷氨酰胺、牛磺酸、L-缬氨酸、L-异亮氨酸或L-亮氨酸等)、中药或中药提取物、膳食纤维(菊粉、魔芋粉、低聚半乳糖、低聚果糖、低聚异麦芽糖、大豆多糖、环糊精、抗性糊精或大豆纤维等)。
微量元素补充剂,可以包括有机酸类的金属离子盐,例如柠檬酸钙、L-乳酸钙、磷酸氢钙、葡萄糖酸钾、柠檬酸钠、葡萄糖酸亚铁、碘化钾、葡萄糖酸锌、亚硒酸钠、葡萄糖酸铜、硫酸铬、葡萄糖酸锰和葡萄糖酸镁中的一种或多种。
任意食品上可接受的辅料,包括但不限于抗氧剂、矫味剂、稳定剂、乳化剂、食用甜味剂、食用色素等。
在一些优选的实施方案中,所述可食用组合物包括亚马逊莓或其提取物、抗氧化性维生素(维生素A、维生素C、维生素E、维生素B6等)和蛋白组分。
对于所述亚马逊莓或其提取物在组合物中的含量,原则上没有特别限定。在本发明一些具体的实施方案中,所述组合物中从具有期望的排铅效果的角度考虑,以干重计,所述亚马逊莓或其提取物的含量为0.3质量%以上,优选为1质量%以上,进一步优选为5质量%,对于其含量的上限没有特别限制,通常可以为60质量%以下,优选为30质量%以下,进一步优选为20质量%以下。
(食品或保健品)
进一步,本发明提供了一种基于亚马逊莓或其提取物,或者是基于上述可食用组合物的食品或保健品。
对于本发明的食品或保健品的具体形态,没有特别限定,在一些具体的实施方案中,所述食品或保健品可以是粉末状可冲调食品(速溶咖啡、谷物粉,坚果粉或藕粉等)、烘焙类食品(面包类、蛋糕类或饼干类烘焙食品等)、饮料(碳酸类饮料、果蔬汁饮料、功能饮料、茶类饮料、乳饮料或酒精饮料等)、乳及乳制品(来源于生鲜牛(羊)乳的鲜乳、乳粉、乳清粉、发酵乳、奶酪或炼乳等)、面食制品(挂面、方便面、馒头、包子、饺子或馄饨等)等。
在其他一些优选的实施方案中,本发明所述的保健品为口服制剂,所述口服制剂包括但不限于片剂、丸剂、颗粒剂、粉剂、茶剂、胶囊剂或口服液等。
(对于避免铅累积及其导致的贫血的用途)
进一步,本发明首次提出了可以将亚马逊莓或其提取物用于生物体排铅或降低生物体内铅蓄积的用途,因此,上述的可食用组合物以及食品或保健品也可以用于上述用途。因此,本发明认为,对于这些食品或保健品的食用可以有效降低生物体,尤其是人体铅累积导致生理不适的发生。
对于本发明所述的铅累积,指的是源自过量铅摄入所导致的生物体铅蓄积现象。所述过量铅摄入主要来自于:暴露于含铅环境,例如含铅污染物环境、汽车尾气、含铅的颜料和涂料等;含铅食品的摄入,例如含铅食品如松花蛋、膨化食品、铁皮罐装饮料、爆米花,以及较长时间保存在含铅管道内的自来水等;一些含铅化妆品、染发剂的使用等。
通常铅累积后可能对生物体,尤其是人体的神经系统、心血管系统、生殖系统、免疫系统的功能、胃肠道、肝肾和脑产生损伤,进而导致各种病理症状。本发明研究发现,本发明的食品或保健品尤其对于铅累积所引起的贫血具有预防、缓解或改善效果。
另外,本发明的食品或保健品原则上适合所有人群,尤其适合于针对铅元素暴露风险较高的人群,对于针对不同特性的人群也可以对食品或保健品中的组分进行相应的调整。
实施例
为更清楚地表述本发明的技术方案,下面结合具体实施例进一步说明,但不能用于限制本发明,此仅是本发明的部分实施例。除非另有说明,本发明中使用的仪器、试剂、材料、实验动物等均可通过常规商业手段获得。
本发明以斑马鱼为研究对象,在摸索了合适的应用浓度的基础上,从促进铅排泄和改善贫血两个方面对亚马逊莓的促排铅功能进行验证和展示。
实施例1:最大检测浓度(MTC)测定
1.检测材料
1.1样品配制信息
亚马逊莓果酱,均质后原液加入,-20℃冻存。
1.2实验动物
斑马鱼均饲养于28℃的养鱼用水中(水质:每1L反渗透水中加入200mg速溶海盐,电导率为450~550μS/cm;pH为6.5~8.5;硬度为50~100mg/L(CaCO3),由杭州环特生物科技股份有限公司提供,实验动物使用许可证号为:SYXK(浙)2012-0171,饲养管理符合国际AAALAC认证(认证编号:001458)的要求。
转基因运动神经元荧光NBT品系斑马鱼,以自然成对交配繁殖方式进行。年龄为受精后6小时(6hpf)的斑马鱼用于亚马逊莓果酱促排铅功效最大检测浓度(MTC)测定。
1.3仪器、耗材与试剂
解剖显微镜(SZX7,OLYMPUS,Japan);CCD相机(VertA1,上海土森视觉科技有限公司,China);精密电子天平(CP214,OHAUS,USA);6孔板(Nest Biotech,China)。
乙酸铅三水合物(批号D2101282,上海阿拉丁生化科技股份有限公司,China)。
2.检测方法
随机选取6hpf转基因运动神经元荧光NBT品系斑马鱼于6孔板中,每孔(实验组)30尾。分别水溶给予亚马逊莓果酱(浓度见表1),同时设置正常对照组和模型对照组,每孔容量为3mL。除正常对照组外,其余各实验组均水溶给予乙酸铅三水合物建立斑马鱼铅蓄积模型。每天换液,28℃处理4天后,测定亚马逊莓果酱对铅蓄积模型斑马鱼的MTC。
3.检测结果
在本实验条件下,亚马逊莓果酱对铅蓄积模型斑马鱼的MTC为20.0μL/mL。详见表1。

实施例2:促排铅功效测定
1.检测材料
1.1样品配制信息
亚马逊莓果酱,均质后原液加入,-20℃冻存。
阳性对照:二巯基丁二酸,白色粉末,批号C1810021,上海阿拉丁生化科技股份有限公司,-20℃冻存。用DMSO配成12.5mg/mL母液,-20℃冻存。
1.2实验动物
斑马鱼均饲养于28℃的养鱼用水中(水质:每1L反渗透水中加入200mg速溶海盐,电导率为450~550μS/cm;pH为6.5~8.5;硬度为50~100mg/L(CaCO3),由杭州环特生物科技股份有限公司提供,实验动物使用许可证号为:SYXK(浙)2012-0171,饲养管理符合国际AAALAC认证(认证编号:001458)的要求。
野生型AB品系斑马鱼,以自然成对交配繁殖方式进行。年龄为6hpf的斑马鱼用于亚马逊莓果酱促排铅功效评价。
1.3仪器、耗材与试剂
解剖显微镜(SZX7,OLYMPUS,Japan);CCD相机(VertA1,上海土森视觉科技有限公司,China);精密电子天平(CP214,OHAUS,USA);6孔板(Nest Biotech,China);电感耦合等离子体质谱仪(ICP-MS,热电XII型,Thermo,USA)。
乙酸铅三水合物(批号D2101282,上海阿拉丁生化科技股份有限公司,China),硝酸(批号20210609,杭州高晶精细化工有限公司,China);二甲基亚砜(DMSO,批号BCCD8942,Sigma,Switzerland)。
2.检测方法
随机选取6hpf野生型AB品系斑马鱼于6孔板中,每孔(实验组)均处理30尾斑马鱼,设置6个平行实验。水溶给予亚马逊莓果酱(浓度见表2),阳性对照二巯基丁二酸浓度为125μg/mL,同时设置正常对照组和模型对照组,每孔容量为3mL。
除正常对照组外,其余各实验组均水溶给予乙酸铅三水合物建立斑马鱼铅蓄积模型。每天换液,28℃处理4天后,收集斑马鱼样本,用ICP-MS检测斑马鱼体内铅含量,分析斑马鱼体内铅含量,以上述指标的统计学分析结果评价亚马逊莓果酱促排铅功效。
统计学处理结果采用mean±SE表示。用SPSS26.0软件进行统计学分析,p<0.05表明差异具有统计学意义。
3.检测结果
在本实验条件下,亚马逊莓果酱具有促排铅功效。详见表2和图1。

实施例3:对铅累积引起的贫血的改善功效测定
1.检测材料

乙酸铅三水合物(批号D2101282,上海阿拉丁生化科技股份有限公司,China),二甲基亚砜(DMSO,批号BCCD8942,Sigma,Switzerland);甲基纤维素(批号B2006074,上海阿拉丁生化科技股份有限公司,China);邻联茴香胺(批号MKCC7501,Sigma,USA);无水乙醇(批号20210514,国药集团化学试剂有限公司,China);无水乙酸钠(批号F20090306,国药集团化学试剂有限公司,China);过氧化氢(批号G2023089,上海阿拉丁生化科技股份有限公司,China)。
2.检测方法
随机选取6hpf野生型AB品系斑马鱼于6孔板中,每孔(实验组)均处理30尾斑马鱼,水溶给予亚马逊莓果酱(浓度见表3),阳性对照二巯基丁二酸浓度为125μg/mL,同时设置正常对照组和模型对照组,每孔容量为3mL。除正常对照组外,其余各实验组均水溶给予乙酸铅三水合物建立斑马鱼铅蓄积模型。每天换液,28℃处理4天后,用邻联茴香胺进行染色,染色完成后每个实验组随机选取10尾斑马鱼置于解剖显微镜下拍照,用NIS-Elements D3.20高级图像处理软件分析并采集数据,分析斑马鱼心脏及鳃部的红细胞数量(染色强度),以上述指标的统计学分析结果评价亚马逊莓果酱贫血改善功效。
其中,统计学处理结果采用mean±SE表示。用SPSS26.0软件进行统计学分析,p<0.05表明差异具有统计学意义。
3.检测结果
在本实验条件下,亚马逊莓果酱具有明显的贫血改善功效。详见表3、图2和图3。

综上,亚马逊莓果酱具有促排铅功效,可以表现为改善铅累积引起的贫血。
产业上的可利用性
本发明提供的亚马逊莓或其提取物的用途在工业上可以广泛应用。
Claims (10)
1.一种亚马逊莓或其提取物在制备有助于排铅作用的食品或保健品中的用途。
2.根据权利要求1所述的用途,其特征在于,
所述亚马逊莓包括亚马逊莓鲜果、亚马逊莓干果、亚马逊莓冻果、亚马逊莓冻干、亚马逊莓果浆或其他食品上可接受的形态。
3.根据权利要求1或2所述的用途,其特征在于,所述亚马逊莓提取物包括水提取物和醇提取物;所述亚马逊莓提取物为液体、固体或半固体。
4.根据权利要求1至3中任一项所述的用途,其特征在于,所述用途还包括改善铅累积导致的贫血。
5.根据权利要求1至4中任一项所述的用途,其特征在于,所述食品或保健品为粉末状可冲调食品、烘焙类食品、饮料、乳及乳制品、面食制品或口服制剂。
6.根据权利要求5所述的用途,其特征在于,所述口服制剂包括片剂、丸剂、颗粒剂、粉剂、茶剂、胶囊剂和口服液。
7.一种食品或保健品用于有助于排铅作用的用途,其特征在于,所述食品或保健品含有亚马逊莓或其提取物。
8.根据权利要求7所述的用途,其特征在于,
所述亚马逊莓包括亚马逊莓鲜果、亚马逊莓干果、亚马逊莓冻果、亚马逊莓果泥、亚马逊莓冻干或其他食品上可接受的形态;
所述亚马逊莓提取物包括水提取物和醇提取物;
所述亚马逊莓提取物为液体、固体或半固体。
9.根据权利要求7或8所述的用途,其特征在于,所述食品或保健品还含有其他植物或其提取物、含乳成分、动物类成分、功能性添加组分、微量元素补充剂和/或任意的在食品上可接受的辅料。
10.根据权利要求7至9中任一项所述用途还包括改善铅累积所导致的贫血。
