Abstract
本发明提供了一种基于生物信息学手段结合拉曼光谱的对海马神经元衰老基因鉴定的方法以及基于所述方法鉴定获得的具有预测和/或诊断用途的分子标记。本发明还创新性的使用体外培养海马神经元上清液的拉曼光谱的峰位强度水平作为筛选模型,用于鉴定海马神经元衰老生物标志物。本发明鉴定获得的分子标记,以及构建的鉴定模型在制备用于预测或检测海马体衰老的产品中的具有广阔的应用前景。
Description
技术领域
本发明属于医疗诊断领域,尤其是涉及一种基于拉曼光谱和生物信息学分析技术鉴定海马神经元衰老的方法和试剂盒。
背景技术
神经元是终末期分化的、衰老具有特异性的一种细胞,其中海马神经元对学习、记忆和情绪调节很重要,且对内环境变化十分敏感,再生能力微弱。已有研究团队利用单细胞RNA测序分析揭示了海马神经元的表观遗传特征,进一步提示海马神经元(hippocampalneurons,HN)突触的高度可塑性。正常的大脑是异质的,由大量专门的细胞类型构成,具有高度精确的电生理行为,能满足大脑能源供应,可参与废物清除和免疫防御。衰老的大脑具有从炎症介质的平衡状态转变为促炎症状态的属性,神经内环境的改变可能会导致人的认知功能出现严重损伤。正常和衰老的神经元中都可能存在与内环境调节相关的亚群,它们之间的差异可以导致代谢的变化、甚至突触功能的衰竭,最终导致预后的不同。
目前广为知晓的,由海马神经元衰老引发的疾病中阿尔茨海默症(Alzheimer’sDisease,AD)和帕金森病(Parkinson’s disease,PD)最为常见,而大部分AD和PD病人在发病中晚期才会出现明显地行为学表现,目前对于海马神经元衰老引发的疾病缺少有效的发病风险预测和诊断方法,尽管目前关于帕金森病的研究已经取得了许多进展,发现了许多和帕金森病相关的基因包括α-synuclein,PARK2(parkin)和PARK6(PINK1)等;但对于HN在衰老发生发展过程中的关键基因所知甚少,同时对于关键基因的验证现有技术并没有效的手段。因此开发一种准确并且有助于早期识别HN在衰老发生发展过程的诊断标记,在预防、预测、诊断神经元衰老引发的疾病诊断研究中具有重要的意义。
现有技术通过对比正常组织与癌变组织的拉曼光谱,可以找到两种组织特征吸收峰的差异,从而为癌症的最终确诊和确定肿瘤切除范围提供重要信息。Tsui等发现在1637、1585和1372cm-1处的血红蛋白相关拉曼谱带在棘细胞和棘红细胞等异形细胞中的振幅较高,提示红细胞的生化异常。一些研究证明了拉曼光谱在从单克隆丙种球蛋白病(MGUS)到无症状骨髓瘤(aMM)和有症状骨髓瘤(sMM)的过程中区分外泌体的能力,从而为患者护理提供了有用的临床指征。基于对现有技术充分的理解,目前尚未有拉曼光谱应用于海马体衰老程度分析的研究和应用,也缺乏将拉曼光谱模型结合生物信息学分析深入探讨海马体衰老相关疾病的分子标记。
发明内容
有鉴于此,本发明旨在提出一种海马体衰老相关疾病的分子标记,以及筛选海马体衰老相关分子标记的方法。
为达到上述目的,本发明的技术方案是这样实现的:
本发明的第一个方面为一种筛选海马神经元(HN)衰老相关基因的方法,所述方法包括如下步骤:
1)基于生物信息学的方法筛选出年轻HN和衰老HN差异表达关键基因;
2)体外培养海马神经元模型,检测上清相关物质的拉曼光谱特征峰,构建基于拉曼光谱的正交偏最小二乘判别分析(OPLS-DA)鉴别模型,获得鉴定海马神经元衰老的拉曼光谱:代表色氨酸的拉曼峰(755和759cm-1)、酪氨酸/脯氨酸的拉曼峰(853cm-1)、苯丙氨酸的拉曼峰(1031和1063cm-1)、核酸的拉曼峰(679、786和826cm-1)、胶原的拉曼峰(859cm-1)、脂质的拉曼峰(957、1078、1268、1285、1299和1437cm-1)、代表葡萄糖拉曼峰(920cm-1)、酪氨酸峰位(643、848、897、1603和1616cm-1)和鸟嘌呤峰位(1190、1415、1485、1491和1573cm-1);
3)将步骤2)的鉴定海马神经元衰老的拉曼光谱与步骤1)中的差异表达基因相对照,获得与衰老HN相关基因。
进一步,所述步骤1)包括如下步骤:a)差异表达基因分析:应用GEO数据库的在线分析工具GEO2R进行分析,筛选年轻HN和衰老HN的差异表达基因;
b)差异表达基因的功能分析:对差异基因开展GO功能注释和KEGG信号通路富集分析;
c)蛋白质互作网络与关键基因分析:通过蛋白质相互作用数据库STRING11.0探索差异基因间编码的蛋白质间的关系,并建立它们间蛋白质互作网络,以PPI网络节点前10名的蛋白所对应的基因为网络中有较高连接度的差异表达关键基因。
进一步,步骤3)获得的衰老HN相关基因为EGFR、PTK2B和DLG2、RASGRF1。
本发明的第二个方面提供根据第一个方面的方法筛选获得的海马神经元(HN)衰老相关基因在制备诊断制剂中的应用。
本发明的第三个方面提供一种预测和/或诊断试剂盒,所述试剂盒包含检测根据第一个方面的方法筛选获得的海马神经元(HN)衰老相关基因的试剂。
本发明的第四个方面提供一种检测生物标志物的试剂在制备预测和/或诊断HN衰老相关疾病的试剂盒中的应用,所述生物标志物为EGFR、PTK2B和DLG2、RASGRF1。
本发明第五个方面提供一种基于拉曼光谱检测海马神经元内环境中的代表色氨酸的拉曼峰(755和759cm-1)、酪氨酸/脯氨酸的拉曼峰(853cm-1)、苯丙氨酸的拉曼峰(1031和1063cm-1)、核酸的拉曼峰(679、786和826cm-1)、胶原的拉曼峰(859cm-1)的峰位强度的水平作为生物标志物在制备预测和/或诊断HN衰老相关疾病的试剂盒中的应用。
进一步的,所述应用还包括将代表脂质的拉曼峰(957、1078、1268、1285、1299和1437cm-1)和代表葡萄糖拉曼峰(920cm-1)的峰位强度水平作为生物标志物。
进一步,所述的第四个方法和第五个方面提供的应用中的HN衰老相关疾病为神经退行性疾病。

相对于现有技术,本发明具有以下有益效果:
本发明利用C57BL/6J小鼠海马神经元体外细胞培养技术,结合非弹性光散射过程的拉曼光谱(Raman spectroscopy,RS)技术,捕获海马神经元生存内环境中,特定的生物分子“指纹”,反映来自衰老神经细胞生存环境中,氨基酸、核酸、脂质、葡萄糖等内环境物质变化,该指纹可以用于鉴定海马体衰老程度。然后结合基因芯片数据技术,从GEO(GeneExpression Omnibus)数据库下载并整理有关衰老的生物信息分析微阵列数据,找到HN在衰老发生发展过程中的关键基因。把利用拉曼光谱技术得到的相关的生物学峰位结果即拉曼光谱指纹,和差异基因提示的生物学作用靶点相结合,为神经元衰老引发的认知障碍性疾病的病程进展的预测、诊断提供重要参考。
本发明使用拉曼光谱鉴定衰老HN培养上清液含有的生酮生糖氨基酸(如色氨酸、苯丙氨酸、酪氨酸)、核酸物质、胶原物质、脂质、糖类水平均高于年轻HN,提示衰老细胞中与维系生命能量、代谢遗传相关的合成过程减弱或中止了,这也进一步印证了生命体内的“负反馈”机制。

除有定义外,以下实施例中所用的技术术语具有与本发明所属领域技术人员普遍理解的相同含义。以下实施例中所用的试验试剂,如无特殊说明,均为常规生化试剂;所述实验方法,如无特殊说明,均为常规方法。
下面结合实施例来详细说明本发明。
实施例1:基于交叉生物信息学分析手段筛选年轻HN和衰老HN差异表达基因
1实验方法:
1.1数据来源:数据来自于NCBI.GEO(Gene Expression Omnibus,GEO,http://www.ncbi.nlm.nih.gov/geo/)数据库,数据系列号为GSE113680(物种:Mus musculus,数据包含2个培养0天的小鼠海马神经元样本数据和2个培养15天的小鼠海马神经元样本数据,均为表达谱数据,样本全部采用GPL10740.Affymetrix Mouse Gene 1.0ST Array平台检测;
设置年轻HN组和衰老HN组。
1.2差异表达基因分析:应用GEO数据库的在线分析工具GEO2R进行分析,GEO2R采用了R语言中GEOquery以及limma程序包,再以P<0.05,|logFC|>4为筛选条件,挑选年轻HN和衰老HN的差异表达基因。
1.3差异表达基因的功能分析:使用DAVID(the Database for Annotation,Visualization and Integration Discovery)在线分析工具(https://david.ncifcrf.gov/)对差异基因开展GO功能注释和KEGG信号通路富集分析。所述GO功能主要包括三个方面,分别是生物学过程(biological process,BP),细胞定位(cellularcomponent,CC)和分子功能(molecular function,MF)。
1.4蛋白质互作网络与关键基因分析:通过蛋白质相互作用数据库STRING11.0(https://sring-db.org/)探索差异基因间编码的蛋白质间的关系,并建立它们间蛋白质互作网络(protein protein interaction network,PPI)。并将PPI结果在Cytoscape软件中打开编辑,利用网络拓扑性质指标Degree.Centrality来分析节点在网络中的得分,节点得分越高越有可能是关键节点。我们以PPI网络节点前10名的蛋白(degree.TOP10)所对应的基因为网络中有较高连接度的Hub基因。
2实验结论:
2.1差异基因:根据差异基因的筛选条件,在GSE113680芯片数据中共得到差异表达基因340个,其中显著上调的差异表达基因46个,显著下调的差异表达基因294个(表1-2),火山图见图1a,图中的红色或蓝色点分别代表显着上调或下调的基因。

2.2使用DAVID进行DEG的基因本体论和途径富集分析:差异表达基因显著富集了9个KEGG通路,显著富集到38个GO-BP,27个GO-CC,16个GO-MF结果。按照P值从小到大排列,取前五GO功能结果绘图(图2和图3)。生物过程(BP)中主要过程为细胞粘附,化学突触传递,细胞内信号转导;细胞成分(CC)主要存在于树突,神经细胞体,轴突;分子功能(MF)主要发挥Ras鸟苷酸交换因子活性,鸟苷酸交换因子活性和酶结合功能;在KEGG分析中,调控的信号主要为钙信号通路,Ras信号通路,胆碱能突触信号通路(图1c-f,表3-6)。

2.3PPI网络和用DEGs鉴定关键候选基因:PPI网络见图1b,共有82个节点,109个互作关系对。拓扑得分高,可视作网络关键节点,前十名基因的表达程度值和PPI网络程度值,分别见图4和表7,上述结果提示前十名(TOP10)基因的在衰老HN组中的表达水平高于年轻HN组。

通过cytohubba插件确认Hub基因,结果显示Hub基因分别为EGFR,PTK2B,GRIN2A,TAC1,CAMK2A,DLG2,GRM5,RASGRF1,KCNJ4和PTGS2,数据显示它们之间存在较强的相互作用。GO功能提示在前10名的基因中,1个基因参与了细胞粘附,4个基因参与了化学突触传递,2个基因参与了细胞内信号转导,2个基因参与了突触可塑性的调节过程;4个基因主要存在于树突,2个基因主要存在于神经细胞体,3个基因主要存在于轴突,2个基因主要存在于树突脊,8个基因主要存在于细胞膜;1个基因主要发挥鸟苷酸交换因子活性分子功能,3个基因主要发挥酶结合分子功能,6个基因主要发挥蛋白质结合分子功能,3个基因主要发挥谷氨酸受体结合分子功能;5个基因参与调控钙信号通路,3个基因参与调控Ras信号通路,2个基因参与调控胆碱能突触信号通路,4个基因参与调控催产素信号通路,1个基因参与调控胰岛素分泌信号通路(图2a-d,图3a-f)。
实施例2:基于拉曼光谱的衰老海马体神经元鉴别模型的构建和有效性分析
1.实验方法:
1.1 C57BL/6J小鼠体外培养的正常神经元模型、衰老神经元模型建立
选择Gibco(Primary Mouse Hippocampus Neurons,1×106Viable Cells/vial)从第17天C57BL/6小鼠胚胎中分离小鼠海马神经元(Catalog No.A15587)为供体来源,快速解冻水浴后,将细胞悬浮液稀释后,以0.5×106密度接种于采用聚赖氨酸包被(4.5μg/cm2)的培养皿中,加入完整的Neurobasal/B-27培养液介质。细胞在36-38℃温度下,在5%CO2环境中培养。培养24小时后,从培养孔抽取培养基的一半,换上新鲜培养基,以后每3天半量换液。
设置正常神经元组:培养7天的神经元作为正常神经元组;
设置衰老神经元组:培养至14天的神经元作为衰老神经元组;
设置对照组:空白培养基。
1.2基于1.1构建的小鼠神经元体外培养模型的海马体神经元内环境相关指标的采集:海马神经元培养上清氨基酸、脂质、核酸和葡萄糖等物质拉曼光谱检测
(1)将5μL的海马神经元培养上清滴在氟化钙玻片(或石英玻片)上,选用共聚焦拉曼光谱仪XploRA Raman microscope测定量;测定条件为:选取785nm激光作为激发光,输出功率为10mW,物镜选取40倍,标本固定在XYZ三维平台上。拍摄过程使用×40 0.6NA尼康镜头,样品上约2×2μm的光斑大小范围接收输出功率为10mW的激光束照射,测量范围600-1800cm-1,每组测量29-30个位点,分辨率为1cm-1。应用Labspec6软件进行平滑和基线校正等数据处理,全部光谱以各自的1450cm-1拉曼峰为内标完成了强度归一化。
1.3基于拉曼光谱数据分析的正交偏最小二乘判别分析(OPLS-DA)构建年轻HN、衰老HN鉴别模型:
采用SIMCA14.1软件对基于1.2通过拉曼光谱法获得年轻HN,衰老HN和对照组的培养上清液的拉曼光谱进行有监督的正交偏最小二乘判别分析(orthogonal partialleast-squares discrimination analysis,OPLS-DA)。使用拟合优度参数R2和Q2分别对OPLS模型的性能进行评价。在零假设条件下,通过y矩阵的随机变化对模型进行200次重采样,进行模型验证。
1.4对构建的OPLS模型鉴定有效峰进行筛选和对构建的OPLS模型特异性验证
(1)为寻找分类模型中的统计学上具有显著差异的拉曼峰位作为潜在生物学标志物,我们使用了V+S分析。在综合考虑相关系数、载荷、在V+S图中与中心的距离等参数的基础上,选择Variable Importance(VIP)>1.5并且p(corr)>0.4的峰位作为潜在生物标志物。将得到的潜在生物标志物进行显著性检验,p<0.05的潜在生物标志物被认为具有统计学意义。
OPLS-DA VIP plot,它反映了模型中的峰位VIP值与相关系数的关系。图中的峰位越接近红色,标明这一峰位在鉴别模型中的相关系数越大。V+S plot可以按照VIP数值由高到低的顺序提供拉曼峰位列表,VIP>1.0并且具有生物学意义的峰位被筛选为的潜在标志物。使用Origin软件进行相关数据处理。使用IBM SPSS Statistics 20进行统计学分析,Graphpad Prism 5绘制统计相关图形。
(2)基于拉曼光谱数据建立的有监督的OPLS-DA模型的有效性可以通过记分图、排列图和ROC图进行评价。
记分图被用于评价模型是否可以鉴别样品,当图中散点样本分群明显,反映了OPLS-DA可以更好地提取光谱中的差异信息,所建立的鉴别方法很好地分别鉴别模型中的样本。
排列图被用于模型成立的判定,排列图分析显示,Q2在Y轴的截距为负值,表示OPLS-DA模型成立,且未过度拟合。
ROC图被用于评价鉴别方法的真实性,曲线下面积(AUC)越接近1,说明鉴别方法的真实性越高。
2.实验结论:
2.1利用OPLS-DA模型初筛体外培养年轻HN组和衰老HN组上清液物质潜在的特征拉曼光谱生物标志物
分别应用对照组、年轻HN组和衰老HN组培养液上清的各30、29和30张拉曼光谱数据,建立有监督的OPLS-DA模型。
图5为OPLS-DA载荷图,用于初步筛选对对照组、年轻HN组和衰老HN组鉴别模型有贡献的拉曼峰位。图中红色和蓝色峰位数字分别与核酸和蛋白质有关。载荷图与记分图在表示样本物质含量关系时存在相关性,即位于载荷图纵坐标正半轴的峰位所代表的物质在记分图横坐标正半轴的群体中含量相对更高,载荷图的纵坐标负半轴与记分图横坐标负半轴也有类似的对应关系。由图5a显示了在600-1800cm-1的范围内对照组、年轻HN组和衰老HN组的拉曼光谱图,年轻HN组和衰老HN组的光谱图形呈现出相似的形态,因此需要结合OPLS-DA方法建立的分类模型,进一步筛选出能有效鉴别对照组、年轻HN组和衰老HN组的峰位作为潜在的生物标志物。
利用OPLS-DA模型得到的,氨基酸(755、759、853、1031、1063cm-1)、核酸(679、786和826cm-1)和脂类(859cm-1)相关的峰位,在图5a中分别用橘红色、绿色和黑色竖线表示,对应拉曼光谱的峰位归属展示在表8中。

在对照组、年轻HN组和衰老HN组建立模型中,综合考虑VIP(VIP>1.5)、相关系数、载荷、在V+S图中与中心的距离等相关参数,分析得出图7。
图7a为OPLS-DA载荷图,用于初步筛选对照组、年轻HN组和衰老HN组鉴别模型有贡献的拉曼峰位。结果表明:氨基酸(759cm-1)、核酸(786cm-1)、和胶原(859cm-1)等特征峰位在三组样本的鉴别中起到重要作用。
图7d、7g、7j分别为三个两两组合模型的VIP plot,直观地反映了模型中的峰位的VIP数值的分布情况。图中显示,年轻HN组的蛋白质(1031和1063cm-1)的峰位强度高于对照组(图7d)。衰老HN组的胶原(859cm-1)、核酸(826cm-1)和氨基酸(759、853、1031和1063cm-1)峰位强度高于对照组(图7g),年轻HN组的胶原(859cm-1)、核酸(679、786cm-1)和蛋白质(755、759和853cm-1)的峰位强度低于衰老HN组(图7j),此结果与之后的统计分析结果一致(图5b)。
图7e、7h、7k分别为三个两两组合模型的VIP图,从图中可知,三个鉴别模型中相关系数大于0.4的峰位数从大到小依次为年轻HN组和衰老HN组,对照组和衰老HN组,对照组和年轻HN组,反映了衰老HN组与年轻HN组之间可能存在较大的物质差异。
图7f、7i、7l分别为三个两两组合模型的V+S plot,V+S plot可以按照VIP数值由高到低的顺序提供拉曼峰位清单,这个清单为分别确定三个两两组合模型中的潜在生物标志物提供了主要依据。
选择Variable Importance(VIP)>1.5并且p(corr)>0.4的峰位作为潜在生物标志物,同时经统计学验证后,对应色氨酸峰位(755和759cm-1),酪氨酸/脯氨酸峰位(853cm-1),代表苯丙氨酸峰位(1031和1063cm-1);核酸物质峰位(679、786和826cm-1);代表胶原物质峰位(859cm-1)峰位被筛选为对照组、年轻HN组和衰老HN组区分潜在标志物(图5b),建立起基于拉曼光谱数据建立的有监督的OPLS-DA模型。
2.2基于拉曼光谱数据建立的有监督的OPLS-DA模型的有效性
基于拉曼光谱数据建立的有监督的OPLS-DA模型的有效性可以通过用Hotelling的95%置信椭圆绘制的OPLS-DA鉴别记分图(图6a,6d,6g,6j)、OPLS鉴别模型排列图(图6b,6e,6h,6k)和OPLS鉴别模型ROC图(图6c,6f,6i,6l)进行评价。
图6中展示的OPLS-DA鉴别记分图中三组样本分群明显,对照组和衰老HN组位于X正半轴,年轻HN组位于X负半轴,反映出三组得到了区分。衰老HN组和对照组分别位于Y轴的正、负半轴,反映出两种类型的培养液得到了区分。这一区分结果表明运用有监督的OPLS-DA方法可以很好地甄别对照组、年轻HN组、衰老HN组的培养液的光谱数据,为分析三个群体的物质特征打下了基础。图6d、6g、6j分别为对照组和年轻HN组、对照组和衰老HN组、年轻HN组和衰老HN组三个模型的OPLS-DA记分图,三幅图中的两组样本分别位于X轴的正负半轴,散点图中样本分群明显,反映了OPLS-DA可以更好地提取光谱中的差异信息,所建立的鉴别方法可以识别培养液样本在成分上的差异,三个模型可以很好地分别鉴别模型中的两组样本(见图6d,6g,6j)。
为了判断OPLS-DA模型是否成立,OPLS鉴别模型排列图被用于模型成立的判定。排列图分析显示,Q2在Y轴的截距为负值,表示OPLS-DA模型成立,且未过度拟合(图6b,6e,6h,6k)。
为了验证在诊断方面的可信性,ROC图被用于评价鉴别方法的真实性,曲线下面积(AUC)越接近1,说明鉴别方法的真实性越高。ROC曲线显示在对照组、年轻HN组、衰老HN组模型中,AUC(对照组)=1,AUC(年轻HN组)=0.984483,AUC(衰老HN组)=1(图6c),在对照组和年轻HN组模型中,AUC(对照组)=1,AUC(年轻HN组)=1(图6f),在照组和衰老HN组模型中,AUC(对照组)=1,AUC(衰老HN组)=1(图6i),在年轻HN组和衰老HN组模型中,AUC(年轻HN组)=0.986207,AUC(衰老HN组)=0.986207(图6l),提示判别分析结果准确性高。
上述结果表明,对应色氨酸峰位(755和759cm-1),酪氨酸/脯氨酸峰位(853cm-1),代表苯丙氨酸峰位(1031和1063cm-1);核酸物质峰位(679、786和826cm-1);代表胶原物质峰位(859cm-1)峰位能够对海马神经元衰老组、年轻组和对照组进行有效区分,能够用于医学诊断试验或者分子标记验证模型。
实施例3建立海马神经元衰老差异基因的拉曼光谱验证模型
1.实验方法
应用统计学方法将实施例2基于拉曼光谱构建的海马神经元年轻、衰老鉴别模型与实施例1生物信息学研究获得的衰老差异基因相结合,验证与衰老差异基因相关的有生物学意义的峰位,利用OPLS-DA得到的潜在生物学标志物对应的峰位。
使用IBM SPSS Statistics 26进行统计学分析,验证OPLS结果和与差异基因有关的生物学峰位,符合正态分布的数据用x±s[平均值±标准偏差(SD)]表示,使用单因素方差分析(ANOVA)进行组间比较,LSD方法用于方差齐组之间的比较,Tamhane的T2方法用于方差不齐组之间的比较。不符合正态分布的数据表示为中值(四分位区间),应用Kruskal-Wallis检验来比较不同组间差异。P<0.05具有统计学意义,本文中的图形是使用GraphPadPrism 6软件制作的。
2.实验结论:
为验证OPLS模型中区分年轻和衰老HN的生物学峰位,我们按照OPLS模型中,VIP大于1.5的参数分析结果,进行统计验证后发现,统计结果与OPLS模型提示一致。统计结果显示衰老HN组培养上清液中氨基酸(755、759、853、1031、1063cm-1)含量高于年轻HN组,差异有显著统计学意义(P<0.01);核酸(679、786和826cm-1)含量高于年轻HN组,差异有显著统计学意义(P<0.01);胶原物质(859cm-1)含量高于年轻HN组,差异有显著统计学意义(P<0.01)(见图5a和图5b)。
我们发现OPLS模型筛选出的氨基酸,以生酮生糖氨基酸(如色氨酸、苯丙氨酸、酪氨酸)为主,与能量代谢密切相关,因此为进一步筛选区分年轻和衰老HN的生物学峰位,我们选择统计与能量代谢有关的脂质和葡萄糖对应的生物学峰位。衰老HN组中脂质(957、1078、1268、1285、1299和1437cm-1)和葡萄糖(920cm-1)的含量高于年轻HN组,差异具有显著统计学意义(P<0.01)(见图5c)。
为筛选和衰老差异基因相关的生物学峰位,选择生物信息学分析筛选出的前10个基因,包括受体酪氨酸激酶超家族成员、天冬氨酸受体、神经肽P物质、膜相关鸟苷酸激酶、谷氨酸受体、RAS鸟嘌呤释放因子、环氧合酶等。结合基因组成成分,我们将HN上清液的拉曼光谱数据,进一步统计学分析后发现,年轻HN和衰老HN培养上清液成分中,酪氨酸峰位(643、848、897、1603和1616cm-1)和鸟嘌呤峰位(1190、1415、1485、1491和1573cm-1)在年轻HN组中的强度显著低于衰老HN组,并且差异具有统计学意义(P<0.01)(见图5d、图5e)。我们筛选出与酪氨酸和鸟嘌呤相关的基因EGFR、PTK2B和DLG2、RASGRF1,结合基因属性和富集分析结果,鉴定出上述四个基因是参与HN衰老的差异基因。
以上所述仅为本发明的较佳实施例而已,并不用以限制本发明,凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。

1.一种筛选海马神经元衰老相关基因的方法,其特征在于:所述方法包括如下步骤:
1)基于生物信息学的方法筛选出年轻海马神经元和衰老海马神经元差异表达关键基因;
2)体外培养海马神经元模型,检测上清相关物质的拉曼光谱特征峰,构建基于拉曼光谱的正交偏最小二乘判别分析鉴别模型,获得鉴定海马神经元衰老的拉曼光谱:代表色氨酸的拉曼峰755和759cm-1、代表酪氨酸/脯氨酸的拉曼峰853cm-1、代表苯丙氨酸的拉曼峰1031和1063cm-1、代表核酸的拉曼峰679、786和826cm-1、代表胶原的拉曼峰859cm-1、代表脂质的拉曼峰957、1078、1268、1285、1299和1437cm-1、代表葡萄糖的拉曼峰920cm-1、代表酪氨酸的拉曼峰643、848、897、1603和1616cm-1、和代表鸟嘌呤的拉曼峰1190、1415、1485、1491和1573cm-1;
3)将步骤1)中的差异表达基因与步骤2)的鉴定海马神经元衰老的拉曼光谱相对照,获得与衰老海马神经元相关基因。
2.根据权利要求1所述的方法,其特征在于:所述步骤1)包括如下步骤:
a)差异表达基因分析:应用GEO数据库的在线分析工具GEO2R进行分析,筛选年轻海马神经元和衰老海马神经元的差异表达基因;
b)差异表达基因的功能分析:对差异基因开展GO功能注释和KEGG信号通路富集分析;
c)蛋白质互作网络与关键基因分析:通过蛋白质相互作用数据库STRING11.0探索差异基因间编码的蛋白质间的关系,并建立它们间蛋白质互作网络,以PPI网络节点前10名的蛋白所对应的基因为网络中有较高连接度的差异表达关键基因。
3.根据权利要求2所述的方法,其特征在于:步骤3)获得的衰老海马神经元相关基因为EGFR、PTK2B和DLG2、RASGRF1。
4.根据权利要求1-2任一所述的方法筛选获得的海马神经元衰老相关基因在制备诊断制剂中的应用。
5.一种预测和/或诊断试剂盒,其特征在于:包含检测根据权利要求1-2任一所述的方法筛选获得的海马神经元衰老相关基因的试剂。
6.检测生物标志物的试剂在制备预测和/或诊断海马神经元衰老相关疾病的试剂盒中的应用,其特征在于:所述生物标志物为EGFR、PTK2B和DLG2、RASGRF1。
