The preparation method comprises the following steps: weighing the raw materials according to the mass parts, mixing in a mixer, then crushing by a crusher, performing secondary crushing by an ultrafine crusher, sieving to ensure that 90% of the fineness enters the mixer through 80 meshes; adding the composite vegetable material and the premix into a mixer for secondary mixing; then the mixture enters a modulator, steam is added for heating up for modulation, and the mixture enters a bulking machine for bulking (the bulking temperature is 120-130 ℃); and (3) drying the expanded particles in a dryer (the drying temperature is 115-120 ℃), and then spraying the expanded particles in spraying equipment by adding fish oil, rapeseed oil and a feed flavoring agent. And after the spraying is finished, the obtained feed particles are cooled to room temperature in a cooling area, and then products which are too small or too large are filtered out through a finished product sieve to obtain the finished pet cat feed.
The beneficial effects of the present invention are demonstrated by the following experiments.
Experiment-examination of influence effect of eucommia ulmoides leaf chlorogenic acid on pet cat blood pressure
1. Design of experiments
18 cats in 2 years old or so are selected, the weight, body type and living environment of each cat are basically consistent, wherein 3 cats with normal blood pressure level are used as a blank control group, the other 15 cats with similar and higher blood pressure level are divided into five groups to be used as experimental groups after other abnormal factors are eliminated, the first group 3 is a model group, the second group 3 is a captopril group, the third group 3 is an eucommia bark group, the fourth group 3 is a hawthorn fruit group, and the fifth group 3 is a sophora flower bud group, and the marks are made respectively.
Feeding cat food for the blank control group and the model group every day for the cats to eat freely; cats in the captopril group are regularly fed with captopril medicaments according to the amount of 5mg/kg/d every morning, and cat food is fed every day for free feeding in the rest of the time; the eucommia ulmoides group is fed with cat food containing the eucommia ulmoides extract prepared in the second embodiment every day for cats to eat freely, and the eucommia ulmoides extract is added into the cat food according to the amount of 10g per kilogram; feeding cat food containing fructus crataegi extract (flavone content 13% (UV)) into fructus crataegi group every day for cat to feed freely, and adding fructus crataegi extract into cat food according to 10g per kg; the sophora japonica bud group is fed with cat food containing sophora japonica bud extract (rutin content is 92% (UV)) every day for cats to eat freely, and the sophora japonica bud extract is added into the cat food according to the amount of 10g per kilogram. All cats were kept in the same environment for a five week trial period.
2. Test item and method
Daily food intake was recorded for each cat and blood pressure, body weight and plasma AngII levels were measured in weeks.
3. Analysis of test results, with reference to FIGS. 1 to 3
As a result of measuring systolic blood pressure by a cuff method, as shown in fig. 1, SBP values of 5week of the eucommia ulmoides leaf group, the hawthorn group, the sophora japonica bud group and the captopril group were significantly decreased (p <0.01) compared with the model group, the SBP difference between 0week and 5week of the eucommia ulmoides leaf group was 30.54mmHg, the SBP difference between 0week and 5week of the hawthorn group was 21.81mmHg, the SBP difference between 0week and 5week of the sophora japonica bud group was 19.32mmHg, and the hypotensive effect of the eucommia ulmoides leaves was significant.
As shown in fig. 2, the weight of cats in all groups increased with time, but the weight of captopril group was lighter than that of the blank group and the natural plant extract groups, indicating that the natural plant extracts had little effect on cats and that captopril had some toxic side effects on cats.
Angiotensin ii (AngII) is formed by the enzyme angiotensin-enzyme catalyzing angiotensinogen, and AngII levels are determined by Angiotensin Converting Enzyme (ACE) activity. The inhibition of MAPK signal path can improve angiotensin II dependent hypertension and reduce organ damage and superoxide anion production, and is a common inhibition mechanism of antihypertensive drugs for dogs. Therefore, the method can be used for evaluating the influence of the eucommia ulmoides leaf chlorogenic acid on Angiotensin Converting Enzyme (ACE) and MAPK pathways by detecting the level of the AngII in the blood plasma. Referring to FIG. 3, the plasma AngII level (501.72ng/mL) of captopril group was significantly lower than that of the model group (566.4 ng/mL; p <0.05), the plasma AngII level (411.5ng/mL) of folium Eucommiae group was significantly lower than that of the model group (566.4 ng/mL; p <0.01), the plasma AngII level (417.0ng/mL) of fructus crataegi group was significantly lower than that of the model group (566.4 ng/mL; p <0.01), and the plasma AngII level (427.9ng/mL) of flos Sophorae group was significantly lower than that of the model group (566.4 ng/mL; p < 0.01). The result shows that the eucommia ulmoides leaf chlorogenic acid has obvious effect of inhibiting the generation of AngII.
Effect investigation applied to Experimental two-pet dog feed
1. Design of experiments
Selecting 12 dogs of 2 years old or so, wherein the weight, body type and living environment of each dog are basically consistent, wherein 3 dogs with normal blood pressure levels are used as a blank control group, the rest 9 dogs with similar blood pressure levels and higher blood pressure levels are divided into three groups after other abnormal factors are eliminated and used as an experimental group, the first group is 3 model groups, the second group is 3 captopril groups, and the third group is 3 eucommia ulmoides leaf chlorogenic acid groups which are respectively marked.
Feeding dog food for free feeding of dogs in the blank control group and the model group every day; the dogs in the captopril group regularly feed captopril medicaments according to the amount of 4.5mg/kg/d every morning, and dog food is put in every day for the dogs to freely feed the dogs at the rest time; the dog food prepared in the seventh embodiment is put into the eucommia ulmoides leaf chlorogenic acid group every day for free feeding of dogs. All dogs were kept in the same environment for five weeks.
2. Test item and method
The daily feed intake of each dog was recorded and the blood pressure, body weight and plasma AngII levels of the dogs were measured in units of one week.
3. Analysis of test results, with reference to FIGS. 4 to 7
Systolic blood pressure was measured by the tail sleeve method and as shown in figures 4 and 5,. DELTA.SBP is the difference between systolic blood pressure on day 1 and systolic blood pressure at 3 or 5 weeks after the start of the experiment. The delta SBP values at 3 weeks and 5 weeks of the chlorogenic acid group and the captopril group are significantly higher than those of the blank group (p <0.01), the delta SBP values at 3 weeks and 5 weeks of the chlorogenic acid group and the captopril group are significantly lower than those of the model group (p <0.01), and the diastolic blood pressure and the systolic blood pressure of the dogs have the same change in the feeding period, and the results show that the chronic feeding of the feed containing the eucommia ulmoides oliv chlorogenic acid can lead to the gradual reduction of the systolic blood pressure and the diastolic blood pressure of the dogs.
As shown in fig. 6, the body weight of dogs in all groups increased over time, with no significant difference in body weight gain for each group over the 5-week trial period.
As shown in figure 7, the plasma AngII level (475.4ng/mL) of the eucommia ulmoides leaf chlorogenic acid group is significantly lower than that of the model group (533.0 ng/mL; p <0.01), and the plasma AngII level (449.8ng/mL) of the captopril group is significantly lower than that of the model group (533.0 ng/mL; p <0.01), which indicates that the eucommia ulmoides leaf chlorogenic acid has the effect of inhibiting ACE and MAPK from inhibiting AngII.
It can be seen from the above tests that:
(1) chlorogenic acid serving as a natural ingredient with rich content in folium cortex eucommiae has a remarkable effect of reducing the blood pressure of the pet cat and dog, has no obvious side effect, and is suitable to be added into a feed which is daily eaten by the pet cat and dog as a daily health-care means of the pet cat and dog.
(3) The eucommia ulmoides leaf extract rich in chlorogenic acid is matched with various medicinal material extracts which can be used as both medicine and food, the scientific proportion is adopted, the eucommia ulmoides leaf extract has a prevention and treatment effect on hypertension common to pet cats and dogs, and compared with chemical medicines, the eucommia ulmoides leaf extract has no toxic or side effect.

Claims (10)

1. Application of folium Eucommiae chlorogenic acid in preparing anti-hypertension pet feed or feed additive is provided.
2. A natural plant antihypertensive feed additive is characterized by being prepared from the following raw materials in parts by weight: 50-150 parts of eucommia ulmoides leaf extract, 50-150 parts of mulberry leaf extract, 50-150 parts of phaseolus calcaratus extract, 50-100 parts of fingered citron extract, 50-150 parts of almond extract, 50-100 parts of gordon euryale seed extract, 50-150 parts of spina date seed extract, 0-100 parts of hawthorn extract, 0-100 parts of sophora flower bud extract and 0-100 parts of cassia seed extract.
3. The natural plant antihypertensive feed additive according to claim 2, wherein the mass content of chlorogenic acid in the eucommia ulmoides leaf extract is 55-95%.
4. The natural plant antihypertensive feed additive according to claim 2, wherein the folium cortex eucommiae extract is obtained by processing folium cortex eucommiae through the following steps: soaking folium Eucommiae in the extractant for 1.5-2.5 hr, wherein the mass ratio of folium Eucommiae to extractant is 1: 6-1: 15, treating with 300-600W ultrasonic wave for 0-50min, concentrating the filtrate, and purifying with resin chromatography column or C18Purifying with chromatographic column, and lyophilizing or spray drying to obtain dried extract, wherein the extractant is water, ethanol or mixture of two.
5. The natural plant antihypertensive feed additive according to claim 2, which is prepared from the following raw materials in parts by weight: 120 parts of eucommia leaf extract, 50 parts of mulberry leaf extract, 55 parts of phaseolus calcaratus extract, 50 parts of fingered citron extract, 75 parts of almond extract, 80 parts of gordon euryale seed extract and 100 parts of spina date seed extract.
6. Use of a natural plant antihypertensive feed additive of any of claims 2-5 in a feed, wherein the feed is for pet cats and pet dogs.
7. The use of the natural plant antihypertensive feed additive of claim 6 in feed, wherein the feed contains 10-25mg of eucommia ulmoides leaf chlorogenic acid per edible part.
8. The use of a natural plant antihypertensive feed additive in feed according to claim 6, wherein the amount of feed additive in pet cat feed is 0.47% to 1.2%.
9. The use of a natural plant antihypertensive feed additive of claim 6 in feed, wherein the amount of feed additive in the pet dog feed is 0.17% to 1.0%.
10. The pet cat feed is characterized by comprising the natural plant antihypertensive feed additive as defined in any one of claims 2 to 5, and specifically comprising the following raw materials in parts by weight: 1-7 parts of natural plant antihypertensive feed additive, 15-25 parts of chicken meal, 18-26 parts of fish meal, 1-5 parts of fish oil, 3-8 parts of rapeseed oil, 3-8 parts of composite vegetable material, 18-26 parts of starch, 1-5 parts of beer yeast powder, 1-5 parts of premix, 8-15 parts of soybean protein concentrate and 2-6 parts of feed flavoring agent.
