Abstract
本发明涉及lncRNA及其在肿瘤诊断、治疗和预后判断中的应用。具体而言,本发明涉及lncRNA AK131315或其表达产物或其促进剂在制备抗肿瘤产品中的应用,以及相应的产品。还提供了lncRNA AK131315或其表达产物在肿瘤诊断、抗肿瘤药物筛选、治疗方案选择和/或预后判断中的应用。本发明的AK131315可有效用于临床抗肿瘤治疗方案的选择和临床预后评估,以及用于个性化治疗和靶向治疗,具有较好的临床应用前景。
Description
技术领域
本发明属于生物技术和医学领域。具体而言,本发明涉及一种新型长链非编码RNA(Long NonCoding RNA,lncRNA)——AK131315,其具体序列和相关产品,及其在肿瘤诊断、治疗和预后判断中的用途和应用方法。
背景技术
天然免疫应答,尤其是I型干扰素(Interferon,IFN)免疫应答,在癌症、病毒感染性疾病以及自身免疫性疾病中均扮演不可或缺的角色。具体而言,干扰素(Interferon,IFN)是具有多种重要功能的细胞因子家族,可参与体内抗炎、抗肿瘤、抗病毒等一系列重要的功能。1969年,Ion Gresser进行了用鼠源IFN治疗荷瘤小鼠的实验,首次揭示了IFN具有一定的抗肿瘤效应。此后,越来越多的研究聚焦到IFN的抗肿瘤效应。接下来的临床实践也证明IFN具有较好的抗肿瘤效应。1986年,FDA批准IFNα2作为首个人类免疫治疗药物,用于治疗癌症。除了治疗肿瘤,现在IFNα已经被用于多种疾病中。由此,I型IFN的调控机制具有重要的研究价值和科学意义。
现在很多研究发现在IFN治疗后,肿瘤病人的预后有所不同,有些病人具有良好预后,生存期有了显著提高,而有些肿瘤病人对IFN治疗不敏感,由此又延伸出多种抗癌药物和IFN联合用药以提高治疗效果的治疗方案。针对病人对IFN的耐药性来探究其原因具有重要的意义。
此前已经有研究报道过miR-26a的表达水平以及RIG-I的表达水平和IFN的治疗效果有关。然而,现有技术中并无关于lnc RNA影响IFN治疗效果的报道。
因此,寻找影响IFN治疗效果的lnc RNA有助于筛选出对在临床上对IFN治疗敏感的病人,且对病人选择合适的治疗方式具有一定意义。
发明内容
本发明正是提供了一种新型lncRNA——AK131315在肿瘤免疫调节和诊断治疗中的应用。
在本发明中涉及新型lncRNA AK131315及其表达产物或前体,所述AK131315可包括如下序列:
(a)SEQ ID NO:1所示的序列;
(b)在严格条件下与(a)限定的序列杂交的序列;
(c)与(a)或(b)中序列具有90%以上序列相同性的序列;和
(d)(a)或(b)中序列在非人哺乳动物中的同源序列。
在本发明的一些方面中,提供了lnc RNA AK131315或其表达产物、或其促进剂在制备抗肿瘤产品中的应用。
在一些实施方式中,所述产品提高I型干扰素(例如IFN-α与IFN-β)水平或活性,和/或活化I型干扰素信号传递途径中的信号传导或效应基因或蛋白(例如MAVS、STING、TBK、IKK)的水平或活性。
在一些实施方式中,所述产品还包含I型干扰素或与I型干扰素联用,例如所述I型干扰素选自:IIFN-α(例如IFNα-2a、IFNα-2b及IFNα-2c)、IFN-β(例如IFNβ-1a、IFNβ-1b)、或其组合。
在一些实施方式中,所述产品用于治疗选自下组的一种或多种肿瘤:实体瘤,例如肝癌、肺癌、黑色素瘤、乳腺癌、卵巢癌、前列腺癌、肾癌、肠癌、头部颈部癌、皮肤癌、膀胱癌、胰腺癌;非实体瘤,例如血液系统肿瘤(如白血病)、神经系统肿瘤(如胶质瘤)。
在一些实施方式中,所述对象是哺乳动物,例如人、非人灵长类动物、大鼠、小鼠、猫、犬、猪、马、牛或羊。
在本发明的一些方面中,提供了一种药物组合物,其包含:
(A)lnc RNA AK131315或其表达产物或其促进剂;以及
(B)药学上可接受的载剂。
在一些实施方式中,所述产品用于治疗选自下组的一种或多种肿瘤:实体瘤,例如肝癌、肺癌、黑色素瘤、乳腺癌、卵巢癌、前列腺癌、肾癌、肠癌、头部颈部癌、皮肤癌、膀胱癌、胰腺癌;非实体瘤,例如血液系统肿瘤(如白血病)、神经系统肿瘤(如胶质瘤)。
在一些实施方式中,所述产品还包含I型干扰素或与I型干扰素联用,例如所述I型干扰素选自:IFN-α(例如IFNα-2a、IFNα-2b及IFNα-2c)、IFN-β(例如IFNβ-1a、IFNβ-1b)、或其组合。
在本发明的一些方面中,提供了检测样品中lnc RNA AK131315或其表达产物的水平的物质在制备用于肿瘤诊断、治疗方案选择和/或预后判断的产品(例如检测试剂盒)中的应用。
在一些实施方式中,所述产品用于指示被检测对象是否罹患对I型干扰素治疗敏感的肿瘤、被检测对象是否适于用I型干扰素治疗和/或预测被检测对象在治疗后的预后效果。
在本发明的一些方面中,提供了一种试剂盒,其包含:
(A')检测获自对象的样品中lnc RNA AK131315或其表达产物水平的物质;例如针对所述序列的特异性引物、抗体;和/或
(B')用于检测的器材、设备,例如选自下组的一种或多种物质:容器、使用说明书、阳性对照物、阴性对照物、缓冲剂、助剂或溶剂。
在一些实施方式中,试剂盒用于通过包括但不限于下组的方法检测lncRNA或其前体:实时定量反转录PCR、生物芯片检测法、DNA印迹法、RNA印记法、RNA免疫共沉淀、原位杂交法。
在一些实施方式中,检测试剂盒检测结果显示对象样品中AK131315水平高于对照水平,则表明该对象所罹患的肿瘤对I型干扰素治疗敏感、适于用I型干扰素进行治疗和/或I型干扰素治疗预后良好;反之,对象样品中AK131315水平低于对照水平,则表明该对象所罹患的肿瘤对I型干扰素治疗不敏感或敏感性低、不适于用I型干扰素治疗和/或I型干扰素治疗预后不佳。
在一些实施方式中,所述对照选自:健康对象(例如健康群体的平均水平)、对象本身的正常组织(例如病灶旁正常组织)、对象先前测定的水平(例如未治疗前、治疗先前阶段时)。
在一些实施方式中,检测试剂盒用于评估检测对象对治疗(尤其是I型干扰素治疗)的应答。
本发明中还提供了一种肿瘤诊断、治疗方案选择和/或预后判断的方法,所述方法包括:
(a)检测对象中lncRNA AK131315的水平;
(b)将其与对照进行比较,若对象样品中AK131315水平高于对照水平,则该对象所罹患的肿瘤对I型干扰素治疗敏感、适于用I型干扰素进行治疗和/或I型干扰素治疗预后良好;反之,对象样品中AK131315水平低于对照水平,则表明该对象所罹患的肿瘤对I型干扰素治疗不敏感或敏感性低、不适于用I型干扰素治疗和/或I型干扰素治疗预后不佳;
(c)基于(b)中结果,对检测对象进行肿瘤诊断、治疗方案选择和/或预后判断。
在本发明的一些实施方式中,还提供了相应的肿瘤治疗方法,所述方法包括给予有需要的对象本发明的lnc RNA AK131315或其表达产物或其促进剂、或者本发明的药物组合物。
本领域的技术人员可对前述的技术方案和技术特征进行任意组合而不脱离本发明的发明构思和保护范围。本发明的其它方面由于本文的公开内容,对本领域的技术人员而言是显而易见的。

具体实施方式
本发明涉及一种新型长链非编码RNA(lncRNA)在促进抗肿瘤治疗和诊断中的的功能和用途,属于生物医学材料技术领域。本发明的AK131315可有效用于临床肿瘤治疗方案的选择和临床预后评估,以及用于个性化治疗和靶向治疗,具有较好的临床应用前景。
本发明人经收集肿瘤组织(例如肝癌)和癌旁组织,检测发现AK131315的表达水平具有较大的差异。在小鼠的荷瘤实验中,我们发现,单纯过表达lnc AK131315对肿瘤组织的生长没有任何影响,而当使用干扰素治疗过表达lnc AK131315的荷瘤小鼠时,IFN的效应有了较大程度的提高,过表达lnc AK131315的荷瘤组织的体积在IFN治疗后相较于对照组,有了极为显著的减小。在IFN治疗组中过表达lnc AK131315的荷瘤组织中ISG的表达水平有了更加显著的升高。我们还发现,当在肝癌细胞系中过表达lnc AK131315时,加入IFN后细胞凋亡的水平有了显著的升高。当干扰lnc AK131315时,IFN诱导的凋亡效应有了显著减弱。
基于此,本发明提供了长链非编码RNA AK131315有效促进IFN对肿瘤治疗效果的新用途。AK131315表达量检测可用于肿瘤的治疗和预后等给出提示信息,或对肿瘤的治疗策略等给出指导性信息。针对AK131315表达量的调控,包括过表达和抑制表达等改变AK131315量的干预措施,可应用于肿瘤的诊断和/或治疗。
本文中提供的所有数值范围旨在清楚地包括落在范围端点之间的所有数值及它们之间的数值范围。可对本发明提到的特征或实施例提到的特征进行组合。本说明书所揭示的所有特征可与任何组合物形式并用,说明书中所揭示的各个特征,可以任何可提供相同、均等或相似目的的替代性特征取代。因此除有特别说明,所揭示的特征仅为均等或相似特征的一般性例子。

lncRNA及其杂交和同源序列
如本文所用,术语“本发明的lncRNA”或“lncRNA AK131315”或“AK131315”可互换使用,均是指在恶性肿瘤(例如肝癌)中特异性高表达、且能促进I型干扰素活性和/或敏感性的lncRNA分子。根据本申请中的内容,该lncRNA分子在肿瘤治疗,尤其是I型干扰素治疗中可起到促进I型干扰素诱导的凋亡的重要作用,可用于有效提高抗肿瘤治疗敏感性和效果。
本发明的lncRNA分子可为:
(a)哺乳动物基因组中特异性表达基因序列如SEQ ID NO:1(GenBank:AK131315.1)所示的特定长链非编码RNA分子的基因;
(b)在严格条件下与(a)限定的序列杂交的序列;
(c)与(a)或(b)中序列具有80%以上序列同一性的序列;和
(d)(a)或(b)中序列在非人哺乳动物中的同源序列。
如本文所用,术语“严格条件”是指:(1)在较低离子强度和较高温度下的杂交和洗脱,如0.2×SSC,0.1%SDS,60℃;或(2)杂交时加有变性剂,如50%(v/v)甲酰胺,0.1%小牛血清/0.1%Ficoll,42℃等;或(3)仅在两条序列之间的同一性至少在50%,优选55%以上、60%以上、65%以上、70%以上、75%以上、80%以上、85%以上或90%以上,更优选是95%以上时才发生杂交。例如,所述序列可为(a)中所限定序列的互补序列。
本发明的lncRNA全长序列或其片段通常可以用PCR扩增法、重组法或人工合成的方法获得。对于PCR扩增法,可根据本发明所公开的有关核苷酸序列来设计引物,扩增而得有关序列。当序列较长时,常常需要进行两次或多次PCR扩增,然后再将各次扩增出的片段按正确次序拼接在一起。
应理解,本发明的lncRNA AK131315优选获自人,获自其它动物的与人lncRNAAK131315高度同源(如具有70%以上、75%以上、80%以上,更优选85%以上如85%、90%、95%、98%甚至99%或以上的序列相同性)的其它lncRNA也在本发明优选考虑的等同范围之内。比对序列相同性的方法和工具也是本领域周知的,如BLAST。
lncRNA的促进剂
本发明中还涉及lncRNA AK131315的“促进剂”。术语“促进剂”是指可提高AK131315的水平或活性的物质。可用于本发明中的促进剂包括但不限于:AK131315表达载体、外源性AK131315、AK131315的脂质体包裹RNA等等。
本发明的AK131315或其促进剂可产生促进抗肿瘤治疗敏感性和效果的作用,提高I型IFN水平及其诱导肿瘤细胞凋亡的能力,从而可进一步用于抗肿瘤治疗。此外,通过提高抗肿瘤药物(例如I型干扰素的)敏感性,在肿瘤治疗中联用本申请的AK131315或其促进剂,可减少该药物的用量或次数,从而降低毒副作用,提高患者的顺应性。
可采用本领域中已知的方法和平台获得本申请的促进剂,并对其效果进行检验。
药物或药物组合物及其治疗应用
本发明还提供了一种药物或药物组合物,其中含有有效量的本发明的AK131315或其促进剂,以及药学上或免疫学上可接受的载体。如本文所用,术语“活性物质”或“本发明的活性物质”可互换使用,是指AK131315、其促进剂。
在一些实施方式中,药物或药物组合物包含AK131315或其促进剂,其可用于产生促进抗肿瘤作用的效果,提高I型IFN水平。
如本文所用,术语“药学上可接受的”成分是适用于人和/或动物而无过度不良副反应(如毒性、刺激和变态反应)的,即有合理的效益/风险比的物质。如本文所用,术语“有效量”是指可对人和/或动物产生功能或活性的且可被人和/或动物所接受的量。
如本文所用,术语“药学上可接受的载体”指用于治疗剂给药的载体,包括各种赋形剂和稀释剂。该术语指这样一些药剂载体:它们本身并不是必要的活性成分,且施用后没有过分的毒性。合适的载体是本领域普通技术人员所熟知的。在《雷明顿药物科学》(Remington’s Pharmaceutical Sciences,Mack Pub.Co.,N.J.1991)中可找到关于药学上可接受的赋形剂的充分讨论。
在组合物中药学上可接受的载体可含有液体,如水、盐水、甘油和乙醇。另外,这些载体中还可能存在辅助性的物质,如填充剂、崩解剂、润滑剂、助流剂、泡腾剂、润湿剂或乳化剂、矫味剂、pH缓冲物质等。通常,可将这些物质配制于无毒的、惰性的和药学上可接受的水性载体介质中,其中pH通常约为5-8,较佳地,pH约为6-8。
本发明的组合物中的活性物质占组合物总重量的0.001~99.9wt%;优选为组合物总重量的1~95wt%,较优选为5~90wt%,更优选10~80wt%。余量为药学上可接受的载体以及其它添加剂等物质。
如本文所用,术语“单位剂型”是指为了服用方便,将本发明的组合物制备成单次服用所需的剂型,包括但不限于各种固体剂(如片剂)、液体剂、胶囊剂、缓释剂。
在本发明的另一优选实施方式中,所述组合物为单位剂型或多剂型,且其中活性物质的含量为0.01~2000mg/剂,优选0.1~1500mg/剂,更优选1~1000mg/剂。在本发明的另一个优选例中,每天施用1~6剂本发明的组合物,优选施用1~3剂;最优选的,每天服用的剂量为1剂。
应理解,所用活性物质的有效剂量可随待施用或治疗的对象的严重程度而变化。具体情况根据对象的个体情况(例如对象体重、年龄、身体状况、所需达到的效果)来决定,这在熟练医师可以判断的范围内。
本发明的组合物,可以为固态(如颗粒剂、片剂、冻干粉、栓剂、胶囊、舌下含片)或液态(如口服液)或其它合适的形状。给药途径可采用:(1)直接裸DNA/RNA注射法;(2)将AK131315相关cDNA、mRNA与转铁蛋白/多聚L-赖氨酸复合物连接,以增强其生物效应;(3)cDNA、mRNA和蛋白质与带正电荷的脂类形成复合物,以克服磷酸骨架负电荷所致的穿越细胞膜的困难;(4)用脂质体包裹cDNA、mRNA和蛋白质后介导进入细胞,既有利于大分子的顺利进入又免受细胞外各种酶的水解作用;(5)cDNA、mRNA和蛋白质与胆固醇结合使其胞浆保持时间增加10倍;(6)用免疫脂质体转运cDNA、mRNA和蛋白质可使其特异性转运至靶组织和靶细胞;(7)将cDNA、mRNA和蛋白质体外转染给转载细胞(如成纤维细胞)也可较好地将相关药物载入靶细胞内;(8)电打孔(electroporation),即借助于电流将RNA、cDNA、mRNA等导入靶细胞。
在一些实施方式中,本发明中还提供了抗肿瘤的方法,所述方法包括给予有需要的对象治疗有效量的AK131315或其促进剂。优选,在给予本申请的AK131315或其促进剂之前、之时或之后给予其他抗肿瘤药物,优选I型干扰素,例如IFNα(如IFNα2)、IFNβ等。
检测试剂盒及检测应用
本发明中还提供了一种用于对象肿瘤诊断、治疗方案选择和/或预后判断的检测试剂盒。在一些实施方式中,试剂盒包括检测获自对象的样品中AK131315水平的物质,例如针对AK131315的试剂(包括但不限于特异性引物、抗体等)和/或用于检测的器材、设备等,例如选自下组的一种或多种物质:容器、使用说明书、阳性对照物、阴性对照物、缓冲剂、助剂或溶剂。
在一些实施方式中,试剂盒用于通过包括但不限于下组的方法检测lncRNA或其前体:实时定量反转录PCR、生物芯片检测法、DNA印迹法、RNA印记法、RNA免疫共沉淀、原位杂交法。

实施例
下面结合具体实施例,进一步阐述本发明。应理解,这些实施例仅用于说明本发明而不用于限制本发明的范围。本领域技术人员可对本发明做出适当的修改、变动,这些修改和变动都在本发明的范围之内。
下列实施例中未注明具体条件的实验方法,可采用本领域中的常规方法,例如参考《分子克隆实验指南》(第三版,纽约,冷泉港实验室出版社,New York:Cold SpringHarbor Laboratory Press,1989)或按照供应商所建议的条件。DNA的测序方法为本领域常规的方法,也可由商业公司提供测试。
除非另外说明,否则百分比和份数按重量计算。除非另行定义,文中所使用的所有专业与科学用语与本领域熟练人员所熟悉的意义相同。此外,任何与所记载内容相似或均等的方法及材料皆可应用于本发明方法中。文中所述的较佳实施方法与材料仅作示范之用。
实施例1:AK131315基因的qRT-PCR检测
采用qRT-PCR技术检测了肝癌样品中AK131315的表达情况。
用研磨机研磨队列1(Cohort 1)的肝癌肿瘤组织样品以及相应的癌旁组织(获自海军军医大学医学免疫学国家重点实验室),加入Trizol试剂,按照Trizol试剂操作步骤进行RNA提取得到肝癌组织RNA。
提取细胞总RNA后,取1μg RNA经高效反转录试剂盒进行反转录,反转录体系如下:
反转录程序为:37℃15min,85℃5s,4℃+∞。
队列2(Cohort 2)和队列3(Cohort 3)的肝癌cDNA样品粉末购买自上海芯超生物科技公司(cohort2:cDNA-HLivH090Su01;cohort3:cDNA-HLivH087Su02;配备有相应的对照癌旁组织cDNA样本),使用无菌水溶解后得到cDNA样品。
以反转录产物为模板,采用如下引物进行实时荧光定量PCR检测AK131315的表达量,使用的是SYBRGREEN RT-PCR试剂盒(Takara公司)并在ABI 7500实时定量PCR仪上完成:

扩增体系如下:
qRT-PCR反应参数:95℃15秒,55℃10秒,68℃读取荧光,72℃30秒,40循环。RNA的相对定量使用2-ΔΔCt法计算(GAPDH为内参)。
实验结果如图1A-C所示。结果显示:与相应正常组织相比,AK131315的表达水平在肝癌组织中极显著降低。
上述结果提示:AK131315与恶性肿瘤密切相关,可作为恶性肿瘤诊断或治疗的靶标。
实施例2:AK131315促进IFN对荷瘤小鼠肿瘤的治疗效果
将AK131315序列(SEQ ID NO:1)插入到lnc RNA表达载体pCDF1载体(购自SystemBiosciences目录号:CD100A-1–CD111B-1插入位点:CDS区域)中,从而得到了表达AK131315的载体及其对照载体(pCDF1空载)。
然后,在HEK293T细胞中使用慢病毒载体表达系统(Lentivector ExpressionSystems,System Biosciences)包装成慢病毒,感染SMMC-7721细胞(购自富衡生物,目录号:FH0071)构建出稳定表达lnc AK131315的SMMC-7721——AK-7721以及相应的对照细胞(使用pCDF1空载进行慢病毒包装)。
将1×107个AK-7721和对照细胞打入裸鼠(购自:必凯生物,6周龄,雄性)皮下进行荷瘤实验。接着使用PBS或IFNα2b对荷瘤小鼠进行治疗,每周测量小鼠的荷瘤大小。同时qRT-PCR定量检测IFNα2b治疗组荷瘤组织中多种ISG的表达水平,使用的是SYBRGREEN RT-PCR试剂盒(Takara公司)并在ABI 7500实时定量PCR仪上完成。实验流程可参见图2A。

qRT-PCR反应参数为:95℃15秒,55℃10秒,68℃读取荧光,72℃30秒,40循环。RNA的相对定量使用2-ΔΔCt法计算(GAPDH为内参)。所用GAPDH qRT-PCR定量引物分别如SEQID NO:4和SEQ ID NO:5所示(同实施例1)。
荷瘤实验结果如图2B所示。结果显示:接种稳定表达lnc AK131315的AK-7721细胞形成的肿瘤对IFNα2b的治疗更敏感。经过IFNα2b治疗后AK-7721细胞形成的荷瘤相较于对照细胞体积显著减小。
qRT-PCR实验结果如图2C所示。结果显示:AK-7721细胞形成的荷瘤在IFNα2b治疗后,组织中的多种ISG基因的表达水平显著或极显著提高。
以上结果表明高表达的lnc AK131315可提高干扰素的肿瘤治疗效果,为提高干扰素对肿瘤治疗效果提供了新思路和途径。
实施例3:AK131315促进IFNα诱导的肝细胞系凋亡
在两种肝癌细胞系SMMC-7721和BEL-7402(购自富衡生物,FH0079)中过表达AK131315(使用转染试剂瞬转),在细胞中加入转染试剂进行转染的同一时间加入IFN(IFNα2b,购自凯因益生公司,加入浓度:4000U/ml)。在IFN作用72h后,消化并收集细胞,进行PI/Annexin V染色,而后进行流式细胞术分析细胞的凋亡情况。
IFNα作用36h后,采用针对AK131315的如下siRNA序列对两种肝癌细胞系中的AK131315表达进行干扰:
AK131315干扰RNA-1(SEQ ID NO:14):
GCAUUAACUCUUCCACUUATT;
无关对照干扰RNA(SEQ ID NO:15):
UUCUCCGAACGUGUCACGUTT。
利用INTERFERin转染试剂,按照操作说明分别用小干扰RNA-1、2和对照干扰RNA对IFN作用36h后的两种肝癌细胞进行转染(转染浓度为40nM)。
干扰36h后将细胞消化下来进行PI/Annexin V染色,而后进行流式细胞术分析细胞的凋亡情况。
IFN对AK131315过表达肿瘤细胞治疗效果的实验结果如图3A和3B所示。结果显示:过表达AK131315后,IFN诱导的细胞凋亡水平极显著上升。
干扰AK131315后的肝癌细胞凋亡结果如图3C和3D所示。结果显示:干扰AK131315后IFN诱导的细胞凋亡水平显著或极显著下降。
以上实验结果表明,AK131315对于IFN诱导肝癌细胞系的凋亡具有促进作用,对其进行干扰表达后能够实现对IFN治疗效果的削弱。因此,AK131315可应用于肝癌免疫相关干预治疗或诊断预后检测。

Claims (4)
1.长链非编码RNA的促进剂在制备促进人对象中IFNα-2b对肝癌治疗效果的产品中的应用,其中所述长链非编码RNA的序列如SEQ ID NO: 1所示。
2.如权利要求1所述的应用,其中,所述产品还包含IFNα-2b或与IFNα-2b联用。
3. 检测人对象样品中长链非编码RNA的表达产物的水平的物质在制备用于所述人对象肝癌治疗方案选择和/或预后判断的产品中的应用,其中所述长链非编码RNA的序列如SEQ ID NO: 1所示,且其中所述产品用于指示被检测对象是否罹患对IFNα-2b治疗敏感的肝癌、被检测对象是否适于用IFNα-2b治疗和/或预测被检测对象在IFNα-2b治疗后的预后效果。
4.如权利要求3所述的应用,其中,所述产品是检测试剂盒。
