Abstract
本发明提供了一种核桃油中诱导期的快速检测方法,该方法包括以下步骤:步骤一,设置傅里叶变换红外光谱仪的工作参数;步骤二,对PE薄膜进行清洗;步骤三,核桃油油样预处理;步骤四,采用涂膜法制样,并进行检测;步骤五,对所得红外谱图进行处理;步骤六,通过所建立红外模型得到核桃油的IP值。本发明的检测方法效率高,成本低,可定量检测出核桃油中30~130min内的IP值,具有良好的准确性。本发明的检测方法可大幅缩短检测时间至3min以内。本发明的检测方法单次消耗检测样品减少至100μL。本发明的检测方法不使用有毒有害化学试剂,不产生有害废弃物,对环境友好。

技术领域
本发明属于食用油技术领域,涉及核桃油,具体涉及一种核桃油中诱导期的快速检测方法。
背景技术
核桃油是一种功能性食用油,多不饱和脂肪酸(PUFAs)占核桃油总脂肪酸的65%以上,主要是亚油酸和亚麻酸,两者同属于人体必须脂肪酸,亚麻酸在冠心病的预防中起着重要作用。核桃油还富含多酚、生育酚、植物甾醇、角鲨烯等微量有益成分,具有多种生理功能,如促进婴幼儿大脑发育,调节血糖和血脂、抗炎、改善记忆、抑制低密度脂蛋白氧化和动脉粥样硬化。因此,核桃油被认为是一种营养丰富的天然产品,通常在销售和消费时不添加任何抗氧化剂或其他物质。然而,由于核桃油PUFAs含量高,极易氧化。因此,评估核桃油的氧化稳定性十分重要,可以进一步确定核桃油质量和货架期。
氧化稳定性是评价食用油品质的重要指标之一。它代表油脂抵抗自动氧化的能力,反映油脂的耐贮存性能。一般用食用油抗氧化的时间表示,即在氧化动力学中油脂由氧化初始阶段过渡到扩张阶段的临界时间,这段时间被称为诱导期(IP),且被认为受到许多外部因素(如温度、光线、氧气压力)和内部因素(如脂肪酸组成、活性成分含量等)的影响。
IP是通过将核桃油油样在一定温度下加热,并测定随着时间变化的某一物理或化学参数来确定的。最初的方法是测量油脂在一定的温度下随时间变化的过氧化值,并使用切线法来确定氧化起始和传播阶段的拐点,与拐点对应的时间即为IP值。目前常用的食用油氧化稳定性测定方法有活性氧法和Rancimat法。前者用碘量滴定法测定过氧化值达到100meq/kg所需的时间。后者通过蒸馏水收集氧化过程中产生的挥发性酸,测量水中电导率随时间的变化从而确定IP值。此外,由意大利Velp scientific公司开发的油脂氧化酸败仪也可以测量食用油的IP值,其基本原理是一定温度下给予密封腔内恒定的氧气压力(6bar),通过检测油样的耗氧量确定IP值。然而,上述方法测定时间长,从几个小时到几个月不等,且消耗较多样品和试剂。基于此,Jiang et al.(2020)利用电子自旋共振(ESR)光谱技术,根据自由基生成的趋势来确定油脂的IP值,该方法与Rancmat法得到的IP值呈高度线性相关(r分别为0.996和0.985)。他们的研究表明,ESR光谱技术为评价食用油的氧化稳定性提供了一种新方法。但该方法操作较为复杂,仪器较为昂贵。
发明内容
针对现有技术存在的不足,本发明的目的在于,提供一种核桃油中诱导期的快速检测方法,解决现有技术中检测方法耗时长,样品和试剂需求量大的问题。
为了解决上述技术问题,本发明采用如下技术方案予以实现:
一种核桃油中诱导期的快速检测方法,该方法包括以下步骤:
步骤一,设置傅里叶变换红外光谱仪的工作参数:
光谱范围:6000~400cm-1,分辨率4cm-1,扫描信号累加次数32次,温度设定为25℃,相对湿度为20%;
步骤二,对PE薄膜进行清洗:
取大小为8cm×8cm,厚度为0.005~0.03mm的PE薄膜,使用50mL去离子水淋洗其表面,用氮气将残余水珠吹扫掉后,再用20mL无水乙醇淋洗其表面,用氮气吹扫干;
步骤三,核桃油油样预处理:
将核桃油油样通过0.22~0.45μm有机系尼龙滤膜抽滤至一干净的容器中;
步骤四,采用涂膜法制样,并进行检测:
用移液枪移取100μL核桃油油样,滴在PE薄膜表面,并均匀涂抹油样,使其形成油膜。光谱通过FTIR光谱仪采集,以空白PE膜为背景光谱获得油膜的光谱,平行测定三次;
步骤五,对所得红外谱图进行处理:
根据峰值处的特征吸收,将获得核桃油红外光谱的有效路径长度归一化为固定路径长度;
步骤六,通过所建立红外模型得到核桃油的IP值:

本发明与现有技术相比,具有如下技术效果:
(Ⅰ)本发明的检测方法效率高,成本低,可定量检测出核桃油中30~130min内的IP值,具有良好的准确性。
(Ⅱ)本发明的检测方法可大幅缩短检测时间至3min以内。
(Ⅲ)本发明的检测方法单次消耗检测样品减少至100μL。
(Ⅳ)本发明的检测方法不使用有毒有害化学试剂,不产生有害废弃物,对环境友好。

具体实施方式
需要说明的是,本发明中:
IP是Induction period的缩写简称,即诱导期。
FTIR是Fourier Transform infrared spectroscopy的缩写简称,即傅里叶变换红外光谱。
PUFAs是Polyunsaturated fatty acids的缩写简称,即多不饱和脂肪酸。
ESR是Electron spin resonance spectroscopy的缩写简称,即电子自旋共振光谱技术。
PE薄膜是polyethylene film的缩写简称,即聚乙烯薄膜。
PLS是Partial least squares的缩写简称,即偏最小二乘法。
SD是Standard deviations的缩写简称,即标准差。
CV是The coefficient of variation的缩写简称,即变异系数。
R2是Acoefficient of determination的缩写简称,即决定系数。
RMSEC是The root means standard error of calibration的缩写简称,即校正误差均方根。
RMSEP是The root mean square error of prediction的缩写简称,即预测误差均方根。

傅里叶变换红外光谱(FTIR)具有简单、快速、无损、精度高、重现性好、数据处理能力强等优点。它可以很好地弥补传统IP值测量方法的不足。使用涂膜法避免了样品的交叉污染,提高了检测精度。
目前,傅里叶变换红外光谱已广泛应用于食用油质量指标的定性和定量领域。许多研究利用该技术建立食用油的检测模型,包括过氧化值、游离脂肪酸、水分、碘值等,以及各种油脂的氧化稳定性、掺假和分类等。这是基于傅里叶变换红外光谱可以提供有关有机分子结构的详细信息。油脂是甘油三酯的复杂混合物,甘油三酯由一个甘油和三个脂肪酸组成,而甘油的分子特征恒定,故观察到的光谱变化是由整体脂肪酸组成及其官能团的变化引起。
核桃油的红外光谱特征主要归因于油酸、亚油酸、亚麻酸、甘油三酯和氧化过程中各种过氧化物的存在。在3029~2767cm-1范围内的红外光谱归因于过氧化物的伸缩振动和脂肪酸含量的变化。在3473cm-1左右出现的弱宽峰,与多不饱和脂质氧化过程中所生成脂质氢过氧化物的O-H伸缩振动有关。核桃油中亚油酸和亚麻酸的含量高,存在大量不饱和双键(-HC=CH-),1785~400cm-1光谱范围内的各个特征吸收峰与甘油三酯的共轭键和羰基脂肪基团(CH2和CH3)有关。1747cm-1代表甘油三酯的酯羰基官能团的伸缩振动,条带面积减少,表明样品的降解程度增加。核桃油在氧化过程中双键的变化体现在1654cm-1,1649cm-1,914cm-1和723cm-1等波段。当核桃油的IP值降低时,说明氧化程度加深,自身的氧化稳定性下降,此时位于3473cm-1的特征峰吸光度呈现上升趋势,说明初级氧化产物氢过氧化物含量上升,1747cm-1处的特征吸收峰下降也说明核桃油氧化程度逐渐加深。1654cm-1和1649cm-1特征峰呈现先下降后上升,随后再下降的趋势,说明在氧化过程中脂肪酸的双键在逐渐裂解,914cm-1和723cm-1的特征峰呈现下降趋势也证明了这一点,而中间的短暂上升则可能是因为共轭烯烃等氧化产物的出现导致双键含量在较短时间内的增高。此外,1550~650cm-1是油脂的指纹区,可提供关于核桃油的独特信息。本发明所建立模型的光谱区域集中在1640~440cm-1。
本发明基于FTIR光谱技术,结合使用PE膜进行检测,通过建立的PLS模型预测核桃油的IP值。在本发明中,对PE薄膜进行清洗,可清洗掉PE薄膜上的杂质,使获得的结果更稳定;对油样进行抽滤预处理,可以有效滤除油中的不溶性杂质,降低红外的噪音干扰信号,得到更为纯净的图谱,从而更加有利于分析。相较于传统方法,所建立模型的RMSEP值较低(验证集和盲样集分别为2.32和3.61),R2值较高(0.9847),预测精度较高。在精密度实验中,在精度评价方面,预测值的SD和CV分别为4.13和4.19%,证明所建模型具有良好的重现性。同时该法具有快速(小于1min)、操作简单、样本量小(100μL)、绿色环保等优点,克服了传统方法存在的耗时长,所需样品量、试剂量大等问题,有效节约成本,提供了一种准确、环保的核桃油IP值快速检测方法。
为说明不同IP值核桃油的红外谱图变化及模型特征,申请人测定了核桃油的IP值,扫描了其红外光谱,优化了红外-IP模型,具体试验包括:
原料:冷榨核桃油(山西礼品核桃,冷榨法制取,3500转离心20min得到);PE膜
仪器与设备:Vertex 70傅里叶变换红外光谱仪,德国布鲁克公司;OXITEST油脂氧化酸败仪,意大利VELP公司。
软件:TQ分析软件7.2版本。
检测流程:采用基于PE膜的FTIR光谱技术对不同氧化程度的核桃油进行检测,光谱范围:6000~400cm-1,分辨率4cm-1,扫描信号累加次数32次,温度设定为25℃,相对湿度为20%。
红外光谱:为了说明不同IP值核桃油的红外谱图变化,扫描了不同IP值的核桃油的红外光谱,结果见图1。
IP值从高到低,表示核桃油氧化程度加深。由图1可知,在3500cm-1左右,出现一个弱宽峰,这个峰与氢过氧化物的O-H伸缩振动有关,吸光度增加。代表碳碳双键的723cm-1,914cm-1、1654cm-1以及3008cm-1处的吸光度有不同程度的下降,表示双键浓度下降,脂肪酸降解,核桃油的氧化程度加深。而1654cm-1和1649cm-1处的峰值出现了一个有趣的变化,呈现出先下降后上升再下降的趋势。这可能是由于氧化初期顺式双键不断降解所致。随着氧化程度的加深,不断产生共轭烯烃等次生氧化产物,导致峰的吸光度增加。但最终双键的降解仍起着主要作用,峰值仍呈下降趋势。表明红外光谱与IP之间存在一定的关系,建模具有可行性。
建模优化:为了说明红外-IP模型的特征,使用TQ软件进行分析,结果见表1。PLS是红外建模的常用方法之一,其优点是可以使用整个或部分光谱区域,而不是单一的某个峰,当光谱信息与测量值存在一定关系,就可以建立模型。利用光谱数据建立核桃油IP值的PLS预测模型,对不同光谱区域和光谱预处理方法进行优化,以选择最佳的模型参数,提高模型精度和稳定性。光谱预处理方法包括原光谱、标准正态变换(SNV)、导多元散射校正(MSC)、Norris导数滤波(NF)、Savitzky Golay平滑滤波(SF)、一阶导数和二阶导数及其之间组合;同时,光谱范围的选择对模型精度至关重要。根据核桃油的FTIR光谱分析结果和核桃油本身特点,选择建模光谱范围主要为1785~400cm–1,将其划分为若干子段,反复筛选优化,并为了防止遗漏信息,适当扩大范围筛选。由于建模条件与光谱范围之间会相互影响,综合考虑,交叉选择进行模型优化。模型的效果以决定系数(R2)、校正误差均方根(RMSEC)和交叉验证误差均方根(RMSECV)为指标进行评价。

由表1可知,不同光谱预处理和波段选择的建模结果不同,选择模型以R2值最高,RMSEC、RMSEP和偏差最低为原则,因此选择通路长度固定,原光谱和不平滑为光谱预处理方法,波段为1640~440cm-1。
图2为校正集测定IP值和预测IP值的关系图。图3为验证集测定IP值和预测IP值的关系图。图4为盲样中测定IP值和预测IP值的关系图。从图2至图4可知,所建立模型的R2值为0.9847,RMSEC值为3.38;PE–FTIR光谱法与Oxitest油脂氧化酸败仪测定的验证样品IP值一致性良好,R2值为0.9878,RMSEP值为2.32;盲样实验中R2值为0.9948,RMSEP为3.61,进一步验证了所建方法的可行性。结果表明该方法可用于分析核桃油IP值,具有良好的预测能力。
遵从上述技术方案,以下给出本发明的具体实施例,需要说明的是本发明并不局限于以下具体实施例,凡在本申请技术方案基础上做的等同变换均落入本发明的保护范围。
实施例1:
本实施例给出一种核桃油中诱导期的快速检测方法,该方法包括以下步骤:
步骤一,设置傅里叶变换红外光谱仪的工作参数:
光谱范围:6000~400cm-1,分辨率4cm-1,扫描信号累加次数32次,温度设定为25℃,相对湿度为20%;
步骤二,对PE薄膜进行清洗:
取大小为8cm×8cm,厚度为0.01mm的PE薄膜,使用50mL去离子水淋洗其表面,用氮气将残余水珠吹扫掉后,再用20mL无水乙醇淋洗其表面,用氮气吹扫干;
步骤三,核桃油油样预处理:
将核桃油油样通过0.22μm有机系尼龙滤膜抽滤至一干净的容器中;
步骤四,采用涂膜法制样,并进行检测:
用移液枪将100μL的核桃油油样滴在PE薄膜表面,并均匀涂抹核桃油油样,形成油膜。光谱通过FTIR光谱仪采集,以空白PE膜为背景光谱获得油膜的光谱,平行测定三次;
样品为冷榨法获得的晋龙品种核桃毛油以及60℃加速氧化获得的不同氧化程度的核桃油,并将其随机按照1:1或1:1:1的比例混合得到最终样品,总计10个。
步骤五,对所得红外谱图进行处理:
根据峰值处的特征吸收,将获得核桃油红外光谱的有效路径长度归一化为固定路径长度;
步骤六,通过所建立红外模型得到核桃油的IP值:

步骤三,核桃油油样预处理:
将核桃油油样通过0.45μm有机系尼龙滤膜抽滤至一干净的容器中;
步骤四,采用涂膜法制样,并进行检测:
用移液枪将100μL的核桃油油样滴在PE薄膜表面,并均匀涂抹核桃油油样,形成油膜。光谱通过FTIR光谱仪采集,以空白PE膜为背景光谱获得油膜的光谱,平行测定三次;
样品为冷榨法获得的香玲品种核桃毛油以及60℃加速氧化获得的不同氧化程度的核桃油,并将其随机按照1:1或1:1:1的比例混合得到最终样品,总计10个。
步骤五,对所得红外谱图进行处理:
根据峰值处的特征吸收,将获得核桃油红外光谱的有效路径长度归一化为固定路径长度;
步骤六,通过所建立红外模型得到核桃油的IP值:
所述的红外模型的建立过程为:校正集90个样品,验证集20个样品,测定平行三次,偏最小二乘法建模,建模条件控制通路长度类型、数据格式和是否光滑,光谱波数范围为1730~440cm-1,主成分因子为10;R2值为0.9828,RMSEC为3.68,RMSEP为3.53。

通过所建立红外模型得到核桃油的IP值。
对本实施例的样品进行检测,检测结果如表3所示。由表3可知,本方法检测结果的标准差小于3mg/kg,与使用油脂氧化酸败仪直接测定样品的IP值相比,模型所得到的IP值相差不大,检测结果接近真实值。

步骤一,设置傅里叶变换红外光谱仪的工作参数:
光谱范围:6000~400cm-1,分辨率4cm-1,扫描信号累加次数32次,温度设定为25℃,相对湿度为20%;
步骤二,对PE薄膜进行清洗:
取大小为8cm×8cm,厚度为0.005~0.03mm的聚乙烯薄膜,使用50mL去离子水淋洗其表面,用氮气将残余水珠吹扫掉后,再用20mL无水乙醇淋洗其表面,用氮气吹扫干;
步骤三,核桃油油样预处理:
将核桃油油样通过0.22~0.45μm有机系尼龙滤膜抽滤至一干净的容器中;
步骤四,采用涂膜法制样,并进行检测:
用移液枪移取100μL核桃油油样,滴在PE薄膜表面,并均匀涂抹油样,使其形成油膜;光谱通过FTIR光谱仪采集,以空白PE膜为背景光谱获得油膜的光谱,平行测定三次;
步骤五,对所得红外谱图进行处理:
根据峰值处的特征吸收,将获得核桃油红外光谱的有效路径长度归一化为固定路径长度;
步骤六,通过所建立红外模型得到核桃油的诱导期值:
