Abstract

However, since a lot of herbs in nature contain huge energy and have the effects of whitening, resisting wrinkles, fading color spots and smoothing skin, the development of a skin beautifying formula which has high safety level, multiple effects and good stability and is derived from natural active ingredients of plants becomes a focus of attention of research and development institutions and manufacturing enterprises in recent years.
CN106260363A discloses a herbal tea for whitening and beautifying, which is prepared from raw materials of jujube, kelp, peach kernel, Chinese yam, medlar, lily, purslane, honeysuckle and crystal sugar, and the herbal tea has obvious effects on improving common problems of dark yellow, rough and lack of elasticity of skin, but only focuses on whitening effect, and mainly functions on inhibiting synthesis of skin melanin and promoting melanin metabolism of skin.
CN105614719A discloses a donkey-hide gelatin cake with effects of whitening skin, removing freckles, which is prepared from 28 components such as cherry, walnut, red date, honey, rose, peanut and cymbidium floribundum, and has the pharmaceutical efficacy, can effectively whiten skin, remove freckles, improve immunity of organism and improve sleep quality, but the donkey-hide gelatin is used as a main component, and is matched with other numerous plant components, so that the formula is complex and the preparation process is complex.
The variety of the herbaceous plants is wide, and the result of compounding different herbaceous plant extracts has great uncertainty, so that the plant extracts are combined into a formula which has the whitening and anti-aging effects, is reasonable, efficient and safe, and has important market value.
Disclosure of Invention
The invention aims to provide a whitening and anti-aging composition and a preparation method and application thereof, and particularly provides a whitening and anti-aging composition based on herbaceous plants and a preparation method and application thereof. In the whitening and anti-aging composition, the lily extract and the peach kernel extract can regulate and control skin pigments (melanin and yellow pigment) and inhibit pigment synthesis and transportation; the semen Persicae extract and thallus laminariae extract can promote blood microcirculation and increase haematochrome content; the kelp extract and the acerola cherry powder can reduce the oxidative stress and the inflammatory state of skin tissues, moisten the skin and relieve wrinkles.
Preferably, the whitening and anti-aging composition based on the herbaceous plants comprises 15.5-35.5 parts of lily extract, 15-35 parts of peach kernel extract, 10.5-20.5 parts of kelp extract and 9.5-20 parts of acerola powder in parts by weight.
When the components in the whitening and anti-aging composition are mixed according to the specific parts by weight, the whitening and anti-aging effect on skin is more remarkable, and the side effects of water consumption, skin dryness, barrier damage and the like caused by accelerated skin metabolism in the whitening regulation and control process can be relieved.

Preferably, the method comprises the steps of:
(1) firstly, mixing and stirring lily extract, peach kernel extract, kelp extract and acerola cherry powder uniformly according to the formula amount to obtain a mixture A;
(2) premixing collagen peptide and elastin peptide with part of the mixture A obtained in the step (1) to obtain a premix B;
(3) and (3) uniformly mixing and stirring the premix B obtained in the step (2) and the rest mixture A obtained in the step (1) to obtain the whitening and anti-aging composition.

In the invention, the preparation method ensures that the main components such as lily extract, peach kernel extract, kelp extract, acerola powder and the like are not damaged and can be fully and uniformly mixed by controlling the temperature, the humidity and the mixing time; if the temperature is too high, the main components of the herbaceous plants can be damaged, and if the temperature is too low, the main components cannot be fully mixed; when the humidity is too high, the moisture absorption of the herbaceous plants influences the water content and the properties of the mixture; if the humidity is too low, the mixing uniformity is affected; the time is too short, the mixing effect can not be achieved, and the time is too long, so that not only is energy wasted, but also excessive mixing of the powder is caused, and the components and the uniformity of the powder are damaged; within the above defined range, the best results are achieved.
In a third aspect, the present invention provides a use of the herbal-based whitening anti-aging composition according to the first aspect in preparing food, health products or cosmetics.

Example 1
The embodiment provides a whitening and anti-aging composition based on herbaceous plants, which comprises, by weight, 27 parts of lily extract, 28 parts of peach kernel extract, 16 parts of kelp extract, 13 parts of acerola powder, 10 parts of collagen peptide and 5 parts of elastin peptide.
The preparation method of the whitening anti-aging composition based on the herbaceous plants comprises the following steps:
(1) mixing Bulbus Lilii extract, semen Persicae extract, thallus laminariae extract, and acerola cherry powder at 23 deg.C and humidity of 38% for 15min to obtain mixture A;
(2) taking 3/5 amount of the mixture A obtained in the step (1), and then premixing with collagen peptide and elastin peptide, wherein the premixing temperature is 28 ℃ to obtain premix B;
(3) and (3) mixing and stirring the premix B obtained in the step (2) and the rest mixture A obtained in the step (1) at the temperature of 28 ℃ and the humidity of 50% for 20min to obtain the whitening and anti-aging composition.
Example 2
The embodiment provides a whitening and anti-aging composition based on herbaceous plants, which comprises the following components, by weight, 15.5 parts of a lily extract, 15 parts of a peach kernel extract, 10.5 parts of a kelp extract, 9.5 parts of acerola powder, 3 parts of collagen peptide and 2.5 parts of elastin peptide.
The preparation method of the whitening anti-aging composition based on the herbaceous plants comprises the following steps:
(1) mixing Bulbus Lilii extract, semen Persicae extract, thallus laminariae extract, and acerola cherry powder at 20 deg.C and humidity of 35% for 10min to obtain mixture A;
(2) taking 1/2 amount of the mixture A obtained in the step (1), and then premixing with collagen peptide and elastin peptide, wherein the premixing temperature is 25 ℃ to obtain premix B;
(3) and (3) mixing and stirring the rest mixture A obtained in the step (1) and the premix B obtained in the step (2) at the temperature of 30 ℃ and the humidity of 55% for 15min to obtain the whitening and anti-aging composition.
Example 3
The embodiment provides a whitening and anti-aging composition based on herbaceous plants, which comprises, by weight, 35.5 parts of lily extract, 35 parts of peach kernel extract, 20.5 parts of kelp extract, 20 parts of acerola powder, 15 parts of collagen peptide and 9.5 parts of elastin peptide.
The preparation method of the whitening anti-aging composition based on the herbaceous plants comprises the following steps:

Application example 1
The application example provides a liquid beverage, and the preparation raw materials of the liquid beverage comprise, by weight: the herbal-based whitening anti-aging composition of example 1 comprises 20 parts, 15 parts of erythritol, 50 parts of purified water, 0.1 part of sweetener and 1 part of acid regulator.
The preparation method comprises the following steps:
(1) preparation: putting the whitening and anti-aging composition based on the herbaceous plants into a preparation tank, adding 50 parts of purified water, controlling the temperature to be 75 ℃, stirring at a constant temperature for 3min, then adding other residual materials, adding the residual purified water, stirring for 10min until the mixture is uniformly mixed, and then sieving by a 200-mesh sieve into a storage tank;
(2) washing the bottle: cleaning the glass bottle by using an automatic cleaning machine;
(3) filling: the liquid is separately filled by a full-automatic filling machine,
(4) and (3) sterilization: and (4) sterilizing the bottled beverage after filling by adopting a high-pressure steam sterilization pot. Sterilizing at 115 + -2 deg.C for 20min, cooling;
(5) and (4) lamp inspection: and cooling and airing the sterilized product, and performing light inspection to obtain the liquid beverage.
Application examples 2 to 6
This application example provides five liquid beverages whose formulation compositions differ from application example 1 only in that the herbal based whitening anti-aging composition of example 1 was replaced with the herbal based whitening anti-aging compositions of examples 2-6, and all other conditions were maintained. The preparation process is as in application example 1.
Comparative application examples 1 to 8
The present application example provides eight liquid beverages whose formulation compositions differ from those of application example 1 only in that the herbal-based whitening anti-aging composition of example 1 was replaced with the herbal-based whitening anti-aging compositions of comparative examples 1 to 8, and all other conditions were maintained. The preparation process is as in application example 1.
The whitening and anti-aging compositions prepared in examples 1 to 8 and comparative examples 1 to 8 were subjected to an intracellular tyrosinase inhibition test, an intracellular melanin inhibition test, an extracellular melanin transport inhibition test, a zebrafish embryo melanin inhibition test, and a skin elasticity test, and specifically tested as follows:
test example 1: intracellular tyrosinase inhibition assay
1.1 sample preparation
Diluting the sample composition with serum-free medium to a concentration of 0.45 mg/ml;
1.2 cell culture
Mouse B16 melanoma cell grows to fusion state, is digested with pancreatin-EDTA for 5min and then dispersed by blowing, the cell suspension is sucked into a centrifuge tube, is centrifuged for 5min at 1000r/min by a centrifuge, the supernatant is discarded, and a culture medium is added to prepare the cell suspension. The cell suspension is diluted to a certain concentration with the culture medium. B16 cells in logarithmic growth phase are inoculated in a 96-well plate, and the cell density is 7 multiplied by 104cfu/ml, 100. mu.L per well, was plated and labeled on the upper lid of the cell plate. Culturing is carried out in an incubator for 24 h.
After 24h of culture, the cell plates were carefully pipetted to remove the supernatant and divided into a blank group, a positive control group, and a sample group. Adding the prepared sample into the sample group, adding culture solution containing alpha-arbutin into the positive control group, and adding serum-free culture medium containing no medicine into the blank group. The addition amount is 100 mu L/hole, 4 multiple holes are made for each concentration, and after the addition is finished, the culture is carried out in an incubator for 72 hours.
1.3 determination of the intracellular tyrosinase inhibition Rate
And (3) discarding supernatant of the cell plate after incubation for 72h, adding 150 mu L of 1% TritonX-100 solution into each well, quickly freezing and storing at-80 ℃ for 30min, then thawing at room temperature to completely break the cells, pre-heating at 37 ℃ for 5min, adding 60 mu L of 0.5% L-DOPA solution, reacting at 37 ℃ for 2h, and measuring the absorbance (OD) value at 490nm of an enzyme-labeling instrument. And calculating the tyrosinase activity inhibition rate.
Tyrosinase activity inhibition (%) - (1-sample average absorbance/blank average absorbance) × 100%. The results are shown in Table 1.
Test example 2: intracellular melanin inhibition assay
2.1 sample preparation
Dissolving the sample composition with 1 × PBS to high concentration, filtering for sterilization, and diluting with cell culture solution to concentration of 0.45 mg/ml;
2.2 cell culture
Mouse B16 melanoma cell grows to fusion state, is digested with pancreatin-EDTA for 5min and then dispersed by blowing, the cell suspension is sucked into a centrifuge tube, is centrifuged for 5min at 1000r/min by a centrifuge, the supernatant is discarded, and a culture medium is added to prepare the cell suspension. The cell suspension is diluted to a certain concentration with the culture medium. Inoculating cells in logarithmic growth phase into 6-well plate at cell density of 1 × 104cfu/mL, 2mL per well, was plated and labeled on the upper lid of the cell plate. Culturing is carried out in an incubator for 24 h.
After the cells were cultured for 24 hours, the cells were divided into a sample group and a blank group. Sample group cell culture solution was changed to culture solution containing test sample, blank group was added to culture solution 2mL per well at 37 ℃ with 5% CO2Culturing for 48h in an incubator; the culture medium was changed after 48h, and 3ml of culture medium containing the sample was added to the sample group, and 5% CO was added to the blank group at 37 deg.C2Culturing for 72h in an incubator; culturing for 72 hr, changing culture solution, adding 3ml of culture solution containing sample into sample group, adding culture solution into blank group, continuing to culture at 37 deg.C and 5% CO2Culturing in an incubator for 48 h.
2.3 determination of the inhibitory Rate of intracellular Melanin
After the cell culture in 2.2 was completed, the cells were washed twice with PBS, the PBS was aspirated, and 200. mu.L of 0.25% trypsinized cells were added to each well for digestion for 5 min. The cells were blown up by adding 2ml PBS in two portions, collected in a centrifuge tube and centrifuged at 4000rpm for 5 min. The PBS residual liquid in the centrifuge tube is completely absorbed as much as possible, 200 mu L of melanin extracting solution is added into each tube, the mixture is shaken uniformly and is put into a water bath with the temperature of 80 ℃ to be heated for 1 hour. After cooling, the tube wall liquid drops are properly centrifuged and uniformly blown, 150 mu L of solution in each centrifuge tube is absorbed and transferred into a 96-well plate, and an enzyme-linked immunosorbent assay (ELIASA) detects the absorbance at 405nm of each hole.
The cell melanin inhibition (%) × (1-sample average absorbance value/blank average absorbance value) × 100%. The results are shown in Table 1.
Test example 3: extracellular melanin transport inhibition assay
3.1 sample preparation
Dissolving the sample composition with 1 × PBS to high concentration, filtering for sterilization, and diluting with cell culture solution to concentration of 0.45 mg/ml;
3.2 cell culture
Mouse B16 melanoma cell grows to fusion state, is digested with pancreatin-EDTA for 5min and then dispersed by blowing, the cell suspension is sucked into a centrifuge tube, is centrifuged for 5min at 1000r/min by a centrifuge, the supernatant is discarded, and a culture medium is added to prepare the cell suspension. The cell suspension is diluted to a certain concentration with the culture medium. Inoculating cells in logarithmic growth phase into 6-well plate at cell density of 1 × 104cfu/mL, 2mL per well, was plated and labeled on the upper lid of the cell plate. Culturing is carried out in an incubator for 24 h.
After the cells were cultured for 24 hours, the cells were divided into a sample group and a blank group. Sample group cell culture solution was changed to culture solution containing test sample, blank group was added to culture solution 2mL per well at 37 ℃ with 5% CO2Culturing for 48h in an incubator; the culture medium was changed after 48h, and 3ml of culture medium containing the sample was added to the sample group, and 5% CO was added to the blank group at 37 deg.C2Culturing for 72h in an incubator; culturing for 72 hr, changing culture solution, adding 3ml of culture solution containing sample into sample group, adding culture solution into blank group, continuing to culture at 37 deg.C and 5% CO2Culturing in an incubator for 48 h.
3.3 detection of extracellular melanin content
After the cell culture in 3.2 is finished, completely absorbing the culture solution in the culture medium to the greatest extent in a centrifuge tube, adding 200 mu L of melanin extracting solution into each tube, shaking uniformly, and heating in a water bath at 80 ℃ for 1 h. After cooling, the tube wall liquid drops are properly centrifuged and uniformly blown, 150 mu L of solution in each centrifuge tube is absorbed and transferred into a 96-well plate, and an enzyme-linked immunosorbent assay (ELIASA) detects the absorbance at 405nm of each hole.
Extracellular melanin transport inhibition (%) × (1-sample average absorbance value/blank average absorbance value) × 100%. The results are shown in Table 1.
Test example 4: zebra fish embryo melanin inhibition assay
4.1 sample preparation
Preparation of a test substance: the test composition was directly dissolved in fish embryo culture solution to prepare a test solution with a concentration of 1 mg/ml.
4.2 culture and detection
Selecting healthy zebra fish embryos, and culturing at the density of no more than 1 fish embryo in 200 mu L of fish embryo culture solution at 28 ℃ until the fertilized time is 6-8 h. The 96-well plate is divided into 3 groups, namely a blank control group (fish embryo culture solution), a positive control group (phenylthiourea working solution) and a test object group (test object), and corresponding fish embryo culture solution is added. Subsequently, 24 fish embryos multiplied by 3 are randomly selected to be put into a 96-well plate and placed in an incubator at 28 +/-1 ℃ to be cultured for 48 +/-1 h after fertilization. Covering the fish embryos with 2-4% of methyl cellulose, and taking pictures of the fish embryos under a stereoscopic microscope according to uniform shooting parameters. The head and yolk back regions of each fish embryo were labeled with analytical software, and the "average intensity of measurement" in the "measurement" column was selected, and the "average signal intensity" in the test results was selected as the melanin signal intensity. And calculating the melanin inhibition rate.
The inhibition ratio of the zebra fish embryo melanin is [ (the level of the normal zebra fish embryo melanin-the level of the zebra fish embryo melanin in a sample treatment group) × 100/(the level of the normal zebra fish embryo melanin-the level of the zebra fish embryo melanin in a positive control group) ]%. The results are shown in Table 1.

The data of the examples 1 to 3 show that the whitening and anti-aging composition based on the herbaceous plants provided by the invention has the intracellular tyrosinase inhibition rate of 34.63% -37.68%, the intracellular melanin inhibition rate of 24.26% -27.06%, the extracellular melanin transport inhibition rate of 19.35% -21.63% and the zebra fish embryo melanin inhibition rate of 34.63% -40%, which indicates that the composition has significant whitening and anti-aging and skin-health effects on human skin; as can be seen from examples 1 and 4 to 6, when collagen peptide and elastin peptide are added to the composition provided by the present invention, the intracellular tyrosinase inhibition rate, the intracellular melanin inhibition rate, the extracellular melanin transport inhibition rate, and the zebra fish embryo melanin inhibition rate of the composition can reach the highest, and the effect is the best; as can be seen by comparing examples 2-3 with examples 7-8, the range of further preferred components of the present invention, the effect is better relative to the effect outside the preferred range; as can be seen from comparison of example 1 with comparative examples 1 to 8, when any one of lily extract, peach kernel extract, kelp extract and acerola powder is absent from the composition, the intracellular tyrosinase inhibition rate, the intracellular melanin inhibition rate, the extracellular melanin transport inhibition rate, and the zebrafish embryo melanin inhibition rate of the composition are significantly decreased.
Test example 5: skin moisture improving and firming skin test
The liquid beverages prepared in application examples 1 to 6 and comparative application examples 1 to 8 were evaluated for skin moisture improving and skin firming effects:
130 subjects of 20-45 years old were randomly selected and divided into 3 groups of 10. The liquid beverages prepared in application examples 1-3, application examples 4-6 and comparative application examples 1-8 were drunk by 13 groups of test subjects, 150mL was drunk daily for 30 days, and the test subjects were not allowed to take other articles for keeping skin moisture and skin elasticity and cosmetics for influencing result determination during the test period without changing the original dietary habits during the test period.
The skin moisture content was measured by a skin moisture content tester Corneometer CM825 on day 0 and day 30, respectively, and the moisture content of the skin before and after drinking was evaluated by repeating the measurement 3 times with the probe in the same area to be measured, and the average value was taken. The results are shown in Table 2.
The skin condition on day 0 and day 30 was measured using the skin elastometer Cutomerma 580, respectively, the test site was the cheekbone, the probe repeated 3 times in the same test area during the measurement, and the average was taken. The test parameter is R2 (the ratio of the springback amount Ua of the skin without negative pressure to the maximum stretch amount Uf with negative pressure), the closer R2 is to 1, the better the elasticity of the skin, the average change value of R2 is R2 on day 30 to R2 on day 0, and the average change rate of R2 is R2/R2 on day 0. The results of the specific skin elasticity tests are shown in table 2.

The results show that: the herbal-based whitening and anti-aging composition and the prepared beverage have good skin moisture improving and skin tightening effects, wherein six components, namely lily extract, peach kernel extract, kelp extract, acerola powder, collagen peptide and elastin peptide, are mutually promoted in the effect, and have unexpected synergistic effect.
In conclusion, the whitening and anti-aging composition based on the herbaceous plants is scientific and reasonable in proportion through selection of six active functional components and a large number of experimental demonstrations, the whitening and anti-aging effects of the composition are synergistically enhanced, and the components are mainly herbaceous plant components, so that the safety, the effectiveness and the skin feeling of the composition are improved. The invention can bring good economic benefit for enterprises and has wide application prospect.
The applicant states that the present invention is illustrated by the above examples, but the present invention is not limited to the above detailed methods, i.e., it does not mean that the present invention must be implemented by relying on the above detailed methods. It should be understood by those skilled in the art that any modification of the present invention, equivalent substitutions of the raw materials of the product of the present invention, addition of auxiliary components, selection of specific modes, etc., are within the scope and disclosure of the present invention.
Claims (10)
1. A herbal-based whitening anti-aging composition is characterized by comprising a lily extract, a peach kernel extract, a kelp extract and acerola powder.
2. The herbal-based whitening anti-aging composition according to claim 1, comprising 15.5-35.5 parts by weight of lily extract, 15-35 parts by weight of peach kernel extract, 10.5-20.5 parts by weight of kelp extract and 9.5-20 parts by weight of acerola powder.
3. The herbal-based whitening anti-aging composition according to claim 1 or 2, wherein the components of the herbal-based whitening anti-aging composition further comprise collagen peptides and/or elastin peptides;
preferably, the collagen peptide is 3-15 parts by weight;
preferably, the weight part of the elastin peptide is 2.5-9.5 parts;
preferably, the herbal plant-based whitening anti-aging composition comprises 25-30 parts by weight of lily extract, 25-30 parts by weight of peach kernel extract, 15-18 parts by weight of kelp extract, 12-15 parts by weight of acerola powder, 3-15 parts by weight of collagen peptide and 2.5-9.5 parts by weight of elastin peptide.
4. The preparation method of the herbal-based whitening anti-aging composition according to any one of claims 1 to 3, wherein the preparation method comprises: mixing and stirring uniformly lily extract, peach kernel extract, kelp extract and acerola powder according to the formula amount to obtain the whitening anti-aging composition.
5. The preparation method of the herbal-based whitening anti-aging composition according to claim 4, wherein the method comprises the steps of:
(1) firstly, mixing and stirring lily extract, peach kernel extract, kelp extract and acerola cherry powder uniformly according to the formula amount to obtain a mixture A;
(2) premixing collagen peptide and elastin peptide with part of the mixture A obtained in the step (1) to obtain a premix B;
(3) uniformly mixing and stirring the premix B obtained in the step (2) and the rest mixture A obtained in the step (1) to obtain the whitening and anti-aging composition;
preferably, the stirring temperature in the step (1) is 20-25 ℃, and the stirring time is 10-20 min;
preferably, the humidity is controlled to be 35% -45% during mixing and stirring in the step (1);
preferably, the temperature of the pre-mixing in the step (2) is 20-30 ℃;
preferably, the part of the mixture A obtained in the step (1) in the step (2) accounts for 50 to 70 percent of the total amount of the mixture A obtained in the step (1);
preferably, the stirring temperature in the step (3) is 25-30 ℃, and the stirring time is 15-25 min;
preferably, the humidity is controlled to be 45% -55% during the mixing and stirring in the step (3).
6. Use of the herbal-based whitening anti-aging composition according to any one of claims 1 to 3 in foods, health products or cosmetics;
preferably, the food product comprises a tabletted candy, a gel candy, a liquid beverage or a solid beverage.
7. The tablet candy is characterized by comprising the following preparation raw materials in parts by weight: the herbal-based whitening anti-aging composition according to any one of claims 1 to 3, 30 to 60 parts, 10 to 17 parts of microcrystalline cellulose, 15 to 28 parts of isomalt, 0.01 to 1 part of sodium carboxymethylcellulose, 1 to 5 parts of silicon dioxide, and 1 to 5 parts of magnesium stearate.
8. The gel candy is characterized by comprising the following preparation raw materials in parts by weight: 2-15 parts of the herbal-based whitening anti-aging composition according to any one of claims 1-3, 3-10 parts of a gel, 40-60 parts of a sweetener, 1-4 parts of a collagen casing, 3-7 parts of a fruit juice concentrate, 0.2-0.3 part of a flavoring essence, and 8-30 parts of purified water.
9. The liquid beverage is characterized in that the preparation raw materials of the liquid beverage comprise, by weight: the herbal-based whitening anti-aging composition according to any one of claims 1 to 3, 10 to 30 parts, erythritol 10 to 20 parts, purified water 30 to 60 parts, sweetener 0.01 to 0.2 parts, and acid modulator 0.01 to 4 parts.
10. The solid beverage is characterized in that the solid beverage is prepared from the following raw materials in parts by weight: 30-60 parts of the herbal-based whitening anti-aging composition according to any one of claims 1-3, 0.01-1 part of sucralose, 0.01-4.5 parts of flavoring essence and 15-50 parts of resistant dextrin.
