Abstract
本发明公开了一种云芝糖肽工业化绿色生产工艺,包括采用低温冷冻复苏法培育、复壮筛选菌株,采用合理的发酵培养基配合比培养,发酵液的提取包括固相菌丝体提取云芝多糖和液相提取云芝多糖,液相经反渗透膜分割为二部分,分子量3万以上浓缩醇沉得到药品级成品,3万及以下部分经处理得到食品级或饲料级添加剂,形成分梯级的全部产出物合理利用。本发明技术先进,采用绿色生产工艺,降低了生产成本;在确保原工艺流程药品级产量收率不降的前提下,增加了产出云芝菌粉作为功能性食品基料和饲用添加剂及饲用蛋白粉,饲料级添加剂的应用填补了国内该项产品空白;本发明实现了资源综合利用,减少了排放,适用于工业化生产。

技术领域
本发明涉及生物技术领域,尤其涉及一种云芝糖肽工业化绿色生产工艺。
背景技术
云芝为多孔菌科真菌彩绒革盖菌[Coriohus versicolor(L.ex Fr.)]的干燥子实体。彩绒革盖菌又名杂色云芝、彩绒革盖菌、黄云芝、灰芝、多色牛肝菌、红见手、千层蘑、彩纹云芝。生于多种阔叶树(柳、杨、白桦、李、桃等)的木桩、倒木和枝上,偶生于松树树干上,分布于全国各省区,主产于东北地区。收载于《中国药典》,具有健脾利湿,清热解毒的功效,用于治疗湿热黄疸、肋痛、纳差、倦怠乏力等疾病。
我国将云芝作为天然植物入药已有千年历史,中医早有记载采用云芝对抗癌症,有着良好临床效果,至于肺癌 、气管炎症及气管扩张,亦有一定疗效。东汉时期《神农本草经》已有记载以云芝治癌,当时癌症叫做“痛肿”,医师会以云芝来加强人体抵抗力,治疗癌症。明朝李时珍的《本草纲目》都有记载和描述,云芝有安神、益寿、利关节、坚筋骨、治耳聋作用。《本草纲目》记载的云芝超过120种,李时珍将其分为六大类灵芝(古代通称灵芝),明确标定为无毒、益气、久食轻身延年。
现代科学家亦证实,云芝能明显提高人体免疫能力、调节内分泌、改善肿胀,对人体各器官机能,如中枢神经、循环系统、呼吸系统、肝脏都有良好的调节和保护作用。对于癌症,它更能有效抑制癌细胞扩散及肿瘤胀大的情况,能减轻副作用。野生云芝的提炼物能有效抑制白血病(即血癌)、肺癌、乳癌、肝癌及大肠直癌细胞的生长,其中以抑制血癌及肝癌细胞生长的效果最为明显,功效高达百分之九十,同时也可提升个人免疫力。除了抗癌,科学家亦证实云芝内的A型胚胎蛋白,有助预防癌症。现代医药学研究表明,从云芝子实体、菌丝体、发酵液中提取的多糖,均具有强烈的抗癌活性,并在日本成为一种抗恶性肿瘤的药物。国内除用于肝炎外,还试用于肝癌的预防和治疗,取得了满意的效果。对慢性支气管炎、慢性活动性肝炎等都有较好的疗效。国外的阿特索(AT-SQ从云芝中提取的以多糖为主并含少量蛋白质的制品),对机体免疫功能亦有明显增强作用。
作为商品的云芝生产有二个路径:其一为固体发酵生产,子实体直接入药或提取云芝糖肽,固体发酵控制难,易污染、劳动密集、周期长,需21天,故成本高,原料来源受限制,不利于工业化大规模生产。其二是液体深层发酵法,它具有受外界环境影响小,恒温和无菌空气通入,发酵速度快,周期为3天,便于放大等优点,目前国内外商品生产者大多采用此法,它也是工业化大量生产的根本方法。
真菌多糖作为一种天然生物大分子,食药用功效广泛,且安全无毒,云芝多糖属于杰出代表。中国顶级药用真菌专家刘波1974年版《中国药用真菌》(一经出版即传入日本,后翻译成日文在日本学术杂志公开全文刊登)等资料传入日本后,不到3年日本东京化学研究所的多位学者迅速开发出云芝PSK,并抢先申请为发明专利,全球仅日本三蓝株式会社日企独家生产经营,PSK商品名为Kerstin。中国上海师大杨庆尧教授于上世纪八十年代选取云芝COV-1,专利号CN89105471.5菌株,通过深层培养菌丝体,水提醇沉法生产云芝糖肽,称PSP。本发明团队主要骨干起始于八十年代生产云芝糖肽,提取收率1.1‰(与发酵放罐体积之比),其余都作为排放物。用该方法从菌丝体中提取云芝糖肽,不但需要投资较大的设备,而且发酵菌丝本身生产成本也较高,从投入产出法来分析,每吨云芝发酵醪液仅收得云芝糖肽一公斤,剩余物是排放物,上世纪九十年代后,经创新改革,将固液相分离后的发酵液提取胞外多糖,收率翻了一翻,但仍未解决大量排放物的问题。目前国内外研究人员致力于研究云芝糖肽的提取纯化,纯化后的多糖中不含有蛋白质及多肽类物质,主要用于药用抑癌性,而对生产过程中产生的大量对人及动物极具有益的营养成份和活性元素作为废料排放。随着国家环保政策进一步趋紧,“双碳”经济的步伐逐步加快,有效资源的综合开发利用是大势所趋,因此需提供一种云芝糖肽绿色生产工艺,对生产过程产生的大量对人及动物极具有益的营养成份和活性元素综合利用。
发明内容
本发明目的是解决上述技术问题,提供一种云芝糖肽工业化绿色生产工艺。
为了实现上述技术目的,达到上述的技术要求,本发明所采用的技术方案是:一种云芝糖肽工业化绿色生产工艺,其特征在于:包括以下步骤:
(1)菌种筛选:以COV-1云芝菌种作为出发菌株,采用非转基因技术,根据固体发酵温差大易产生变异性子实体的原理,将其进行原生质体诱变,模拟自然恶劣环境,选取生命力强的茁壮菌株复壮,提供给大生产,菌种筛选方法为低温冷冻复苏法,具体包括以下步骤:
a.菌种活化与提纯复壮:将保存的菌种提前活化,制作平板A,培养基为综合培养基,灭菌后在无菌条件下,28℃下培养菌落;继续制作平板B,挑选平板A上强壮菌丝前端接于平板B上,28℃下培养5~7天至白色绒毛状菌丝长满;
b.冻干箱预冻至-60℃~-55℃,冻干箱的搁板温度在-55℃~-50℃,将培养皿平板B放至搁板上,恒温一天,然后每小时升温一度,逐步升温至28℃;
c.制作平板C,灭菌后在无菌条件下,将冷冻复苏后的平板B菌落接至平板C上培养;
d.重复步骤a~c方法提纯培养,复壮,将强壮菌丝前端接入培养瓶中,置SPY-50双层摇床,摇床转速180~220r/min,培养温度25±1℃,培养时间4天,待白色气生菌丝生长成圈状,无杂菌,选强壮菌丝前端接入无菌培养基的试管或茄瓶中,25~28℃下培养5~7天,至白色绒毛状菌丝长满斜面,新鲜斜面置0~4℃冰箱内保存,即可提供给大生产作起始菌种,冰箱保存时间不超过30天,如需较长时间保存,需制作液体石蜡斜面孢子储存;
(2)发酵:供种后进入一级种子罐,一级种子罐培养基配方为葡萄糖3%,黄豆饼粉1.5%,酵母粉0.2%,MgSO4·H2O0.1%,KH2PO40.1%,K2HPO4 0.1%,消泡剂0.25L,计料体积300L/750L,培养得到一级罐种液;一级罐种液进入二级种子罐,二级种子罐培养基配方为葡萄糖3%,黄豆饼粉1.5%,酵母粉0.2%,MgSO4·H2O0.05%,K2HPO40.05%,消泡剂0.75L,计料体积4000L/5000L,培养得到二级罐种液;二级罐种液进入发酵大罐,发酵大罐培养基配方为玉米粉0.2%加糖化酶,玉米浆干粉0.01%,葡萄糖3%,黄豆饼粉1.5%,酵母粉0.2%,MgSO4·H2O0.0125%,K2HPO40.0125%,VB10.005%,消泡剂4.2L,计料体积40000L/50000L,发酵中止后得到发酵液;
(3)提取:发酵中止进行菌丝灭活,防止菌丝老化、自溶和杂菌利用,菌丝灭活液冷却后立即泵送入自动板框压滤进行固液分离;液相经反渗透膜切割为二部分,分子量3万及以下部分送中间罐进行下一步反渗透膜切割,分离液达中水标准排放或回用,高浓度部分送薄膜浓缩岗位进一步浓缩,至浓缩液密度1.09023~1.10687kg/L,投入经干燥破壁的菌丝体,装盘至烘箱,在60~80℃真空干燥12小时,得食品级或饲料添加剂级粗品,其主要成分为3万及以下的不含肽的云芝胞外糖,多肽,生物碱、酚类,植物固醇类,二萜及三萜类化合物,16种人体所需氨基酸,VB1、VB2、VB6、及铜、铁、钾、锌等10多种人体所需的微量元素,经30-B不锈钢万能粉碎机80~100目粉碎,检验合格后包装;分子量大于3万部分送至薄膜浓缩岗位,浓缩醇沉进入药品级,提取云芝胞外多糖;固相滤饼进入水提罐,水解温度96±2℃,水解时间6小时,加水量为菌丝体的4~5倍量,然后进入自动板框二次固液相分离,液相浓缩提取云芝胞内多糖,对固相菌丝体进行干燥,经200目破壁得云芝菌丝体蛋白粉,因云芝菌细胞壁较厚,固相菌丝体经破壁后也可再次水煮提取云芝胞内多糖或直接用作饲料级添加剂。
优选的:菌丝体提取的云芝胞内多糖粗品与云芝发酵液中提取的云芝胞外多糖粗品分开纯化,纯化达标后得云芝多糖药品级可混粉。
优选的:发酵大罐发酵中止条件为PH值4.5~5.5,镜检菌丝粗壮,呈网状,菌丝边缘及锁状联合清晰,这是菌丝生长旺盛的表现,此时放罐菌丝蛋白中A型胚胎蛋白比例最高,如出现菌丝边缘及锁状联合模糊,则立即放罐,如继续培养则进入菌丝老化自溶趋势;PH值小于4.5时菌丝细弱。
与传统工艺相比,本发明的有益效果:
1、本发明技术先进,采用绿色生产工艺,在原工艺路线上,少投入,通用共用部分不投入,降低了生产成本;在确保原工艺流程药品级产量收率不降的前提下,增加了产出云芝菌粉作为功能性食品基料和饲用添加剂及饲用蛋白粉,提高了产品产出比,实现资源综合利用,减少了废料排放,适用于工业化生产。
2、本发明采用低温冷冻复苏法培育复壮筛选菌株,排除劣质性状的菌株,选择出具有更优良生产性能的高产菌株,达到纯化与复壮菌种、保持稳定生产性能的目的。
3、本发明不仅采用醇沉法提取发酵液中的云芝胞外多糖,而且通过水提法二次处理云芝菌丝体提取云芝胞内多糖,提高了云芝糖肽的收率。
4、本发明形成了分梯级的全部产出物合理利用,生产所形成的产出物梯级为:药品级、食品级、饲料级、有机肥料级,新增产出有价值产品以四只五十吨发酵大罐,全年十一个月的新增产出物按云芝子实体折算,相当于四十亩土地的产出量;饲料级添加剂的应用填补了国内该项产品空白。

具体实施方式
下面以50T发酵大罐为例对本发明作进一步说明。
参照附图,一种云芝糖肽工业化绿色生产工艺,其特征在于:包括以下步骤:
(1)菌种筛选:以COV-1云芝菌种作为出发菌株,采用非转基因技术,根据固体发酵温差大易产生变异性子实体的原理,将其进行原生质体诱变,模拟自然恶劣环境,选取生命力强的茁壮菌株复壮。菌种筛选方法为低温冷冻复苏法,具体包括以下步骤:
a.菌种活化与提纯复壮:将保存的菌种提前活化,制作平板A,培养基为综合培养基,灭菌后在无菌条件下,28℃下培养菌落;继续制作平板B,挑选平板A上强壮菌丝前端接于平板B上,28℃下培养5~7天至白色绒毛状菌丝长满;
b.冻干箱预冻至-60℃~-55℃,冻干箱的搁板温度在-55℃~-50℃,将平板B放至搁板上,恒温一天,然后每小时升温一度,逐步升温至28℃;
c.制作平板C,灭菌后在无菌条件下,将冷冻复苏后的平板B菌落接至平板C上培养;
d.重复步骤a~c方法提纯培养,复壮,将强壮菌丝前端接入培养瓶中,置双层摇床,摇床转速180~220r/min,培养温度25±1℃,培养时间4天,待白色气生菌丝生长成圈状,无杂菌,选强壮菌丝前端接入无菌培养基的试管或茄瓶中,25~28℃下培养5~7天,至白色绒毛状菌丝长满斜面,新鲜斜面置0~4℃冰箱内保存,即可提供给大生产作起始菌种,冰箱保存时间不超过30天,如需较长时间保存,需制作液体石蜡斜面孢子储存;
(2)发酵:根据工艺流程,供种后进入一级种子罐,一级种子罐培养基配方为葡萄糖3%,黄豆饼粉1.5%,酵母粉0.2%,MgSO4·H2O0.1%,KH2PO40.1%,K2HPO4 0.1%,消泡剂0.25L,计料体积300L/750L,在温度27℃、罐压0.04~0.05MPa、通气量1:0.5、搅拌速度180~200r.p.m条件下培养48h得到一级罐种液;一级罐种液进入二级种子罐,二级种子罐培养基配方为葡萄糖3%,黄豆饼粉1.5%,酵母粉0.2%,MgSO4·H2O0.05%,K2HPO40.05%,消泡剂0.75L,计料体积4000L/5000L,在温度25℃、罐压0.04~0.05MPa、通气量1:0.5、搅拌速度180~200r.p.m条件下培养24~48h得到二级罐种液;大罐培养基按顺序投入配料罐,加水搅拌泵送入发酵大罐,灭菌后大罐培养基37~37.5m3,二级罐种液进入发酵大罐,发酵大罐培养基配方为玉米粉0.2%加糖化酶,玉米浆干粉0.01%,葡萄糖3%,黄豆饼粉1.5%,酵母粉0.2%,MgSO4·H2O0.0125%,K2HPO40.0125%,VB10.005%,消泡剂4.2L,计料体积40000L/50000L,在温度25℃、罐压0.04~0.05MPa、通气量1:0.5、搅拌速度180~200r.p.m条件下培养60~72h,发酵中止后得到发酵液。
(3)提取:发酵中止则进行菌丝灭活,防止菌丝老化、自溶和杂菌利用,灭活条件为96~100℃ 0.5~1h。灭活液冷却后立即泵送入自动板框压滤进行固液分离,液相经反渗透膜切割为二部分,分子量3万及以下送中间罐进行下一步反渗透膜切割,分离液达中水标准排放或回用,高浓度部分送薄膜浓缩岗位进一步浓缩,至浓缩液密度1.09023~1.10687kg/L,投入经干燥破壁的菌丝体,装盘至烘箱,在60~80℃真空干燥12小时得食品级或饲料添加剂级粗品,经30-B不锈钢万能粉碎机100目粉碎,检验合格后包装;分子量大于3万的送至薄膜浓缩岗位,浓缩条件为蒸汽压力<0.05MPa,温度75℃,发酵液流量2000L/h,蒸汽流量3.0m3/h ,真空度≤-0.065MPa,浓缩醇沉进入药品级;固相滤饼进入水提罐,水解温度96±2℃,水解时间6小时,加水量为菌丝体的5倍量,然后进入自动板框二次固液相分离,液相浓缩提取胞内云芝多糖,固相菌丝体进入干燥,经200目破壁得云芝菌丝体蛋白粉,因云芝菌细胞壁较厚,经破壁后也可再次水煮提取云芝胞内多糖或直接用作饲料级添加剂。
本优选实施例中,发酵大罐发酵中止的质量标准PH值4.5~5.5,镜检菌丝粗壮,呈网状,菌丝边缘及锁状联合清晰明显这是菌丝生长旺盛的表现,此时放罐菌丝蛋白中A型胚胎蛋白比例最高,如出现菌丝边缘及锁状联合模糊,则立即放罐,如继续培养则进入菌丝老化自溶趋势,PH值小于4.5时菌丝细弱。
本优选实施例中,菌丝体提取的云芝胞内多糖粗品与云芝发酵液中提取的云芝胞外多糖粗品分开纯化,纯化达标后得云芝多糖药品级可混粉。
本实施例平均发酵菌浓35~37%,菌丝胞内糖收率1.1kg/m3(成品/发酵液),胞外糖收率1.3kg/m3(成品/发酵液),合计药品级云芝糖肽收率2.4kg/m3(成品/发酵液)。
具体实施时,云芝多糖分子量3万及以下的合格检验标准,采用《中国药典》2020版云芝检验标准:
【检查】水分不超过13.0%(通则0832第二法),总灰分不超过6.0%(通则2303),酸不溶性灰分不超过4.0%(通则2302)。
【浸出物】按照水溶性浸出物测定法(通则2201)项下的热浸法测定,不少于18.0%。
【含量测定】a总糖:取粗粉约5g,精密称定,置锥形瓶中,精密加水120ml,称定重量,加热回流1小时,放冷,再称定重量,用水补足减失的重量,摇匀,用脱脂棉滤过,精密量取滤液40ml,加酚酞指示液1-2滴,用氢氧化钠试液调节pH值至中性,加稀硫酸25ml,加热回流4小时,放冷,用氢氧化钠试液调节pH值至中性,精密加入碘滴定液(0.1mol/L)25ml,逐滴加氢氧化钠试液4ml,加边加剧烈振摇,密塞,置暗处放置10分钟,加稀硫酸4ml,立即用硫代硫酸钠滴定液(0.1mol/L)滴定,至近终点时,加淀粉指示液2ml,继续滴定至蓝色消失,并将滴定的结果用空白试验校正,即得。每1ml碘滴定液(0.1mol/L)相当于9.008mg的无水葡萄糖(C6H12O6)。
b单糖:精密量取总糖项下的滤液40ml,加酚酞指示液1-2滴,用氢氧化钠试液调节pH值至中性,按总糖项下方法,自“精密加入碘滴定液(0.1mol/L)25ml”起,同法操作。每1ml碘滴定液(0.1mol/L)相当于9.008mg的无水葡萄糖(C6H12O6)。
总糖的含量减去单糖的含量,即为云芝多糖的含量。按干燥品计算,含云芝多糖以无水葡萄糖(C6H12O6)计,不少于3.2%。
绿色清洁生产“三废”的处理:
①废气:云芝多糖肽发酵过程通入的无菌空气,菌丝体生长过程排出含微量CO2空气,无毒、无尘,符合国家大气污染物综合排放标准,直接排放。
②固体废物:二次压滤的固相滤饼,为云芝多糖肽菌丝体,含有大量蛋白质,粗蛋白含量达40%已作饲用蛋白源利用。企业污水处理站板框压滤后的干泥,作为有机堆肥基料。
③废水:液相提取过程经3万以下的二次反渗透膜切割,系用切向流进行过滤原理,排放水达中水标准,可用于绿化浇灌,清洁打扫,部分生产过程回用,多余部分经企业污水站生化处理,达标排放;生产废水由厂区管理系统收集至格栅隔油集水池,去除水中粗大杂物及浮油,然后用泵提升至微电解反应器发生氧化还原反应,再投加石灰乳(辅助加入PAM)与水中部分有机物絮凝沉淀,由斜管隔油沉淀池中去处;沉淀出水进入选择反应池,经生化作用后,用泵提升至UBF厌氧反应器,废水中的有机物在厌氧条件下转化为沼气,厌氧出水经脱气沉淀池后进入预曝调节池,脱气沉淀池的污泥回流至选择反应池,厌氧沼气经水封罐排放;同时,其它废水也进入预曝调节池,混合废水经水解酸化、调节水质水量后,分批次进入CASS反应器发生好氧生物降解,好氧出水经水器收集并计量后,达标排放;UBF厌氧反应器、CASS反应器内的剩余污泥及沉淀污泥排入污泥池,由污泥泵压入板框压滤机,进一步脱水干化,干泥外运无害化处理。作为有机肥基料,清液回到预曝调节池处理。废水排放工艺流程见附图4。
饲料添加剂应用:
饲料添加剂经扬州大学推荐及养殖场应用,奶牛、猪、鸡、水产,在用户原饲料配方的基础上,取代原配方里50%蛋白源(按折纯蛋白计算),使用效果良好,原因是除营养成份蛋白源外,还有云芝的功能,“纳差”即动物胃口变好,优于酵母粉,云芝多糖的功能、增强免疫力,防病抗病的食疗功能,养殖企业饲喂不用抗生素,达到不用药,少用药的效果。云芝菌丝体A型胚胎蛋白也有增强免疫力的功能,使用云芝糖肽工业化生产的副产品,纯天然对人类安全菜篮子发挥重要作用。经查询农业农村部饲料产品目录,该项目填补了国内产品空白。
本发明的上述实施例,仅仅是清楚地说明本发明所做的举例,但不用来限制本发明的保护范围,所有等同的技术方案也属于本发明的范畴,本发明的专利保护范围应由各项权利要求限定。
Claims (3)
1.一种云芝糖肽工业化绿色生产工艺,其特征在于:包括以下步骤:
(1)菌种筛选:采用低温冷冻复苏法,选取茁壮菌株复壮,具体包括以下步骤:
a.菌种活化与提纯复壮:将保存的菌种提前活化,制作平板A,培养基为综合培养基,灭菌后在无菌条件下,28℃下培养菌落;继续制作平板B,挑选平板A上强壮菌丝前端接于平板B上,28℃下培养5~7天至白色绒毛状菌丝长满;
b.冻干箱预冻至-60℃~-55℃,冻干箱的搁板温度在-55℃~-50℃,将平板B放至搁板上,恒温一天,然后每小时升温一度,逐步升温至28℃;
c.制作平板C,灭菌后在无菌条件下,将冷冻复苏后的平板B菌落接至平板C上培养;
d.重复步骤a~c方法提纯培养,复壮,将强壮菌丝前端接入培养瓶中,置双层摇床,摇床转速180~220r/min,培养温度25±1℃,培养时间4天,待白色气生菌丝生长成圈状,无杂菌,选强壮菌丝前端接入无菌培养基的试管或茄瓶中,25~28℃下培养5~7天,至白色绒毛状菌丝长满斜面,新鲜斜面置0~4℃冰箱内保存,即可提供给大生产作起始菌种,冰箱保存时间不超过30天,如需较长时间保存,需制作液体石蜡斜面孢子储存;
(2)发酵:供种后进入一级种子罐,一级种子罐培养基配方为葡萄糖3%,黄豆饼粉1.5%,酵母粉0.2%,MgSO4·H2O0.1%,KH2PO40.1%,K2HPO4 0.1%,消泡剂0.25L,计料体积300L/750L,培养得到一级罐种液;一级罐种液进入二级种子罐,二级种子罐培养基配方为葡萄糖3%,黄豆饼粉1.5%,酵母粉0.2%,MgSO4·H2O0.05%,K2HPO40.05%,消泡剂0.75L,计料体积4000L/5000L,培养得到二级罐种液;二级罐种液进入发酵大罐,发酵大罐培养基配方为玉米粉0.2%加糖化酶,玉米浆干粉0.01%,葡萄糖3%,黄豆饼粉1.5%,酵母粉0.2%,MgSO4·H2O0.0125%,K2HPO40.0125%,VB10.005%,消泡剂4.2L,计料体积40000L/50000L,发酵中止后得到发酵液;
(3)提取:发酵中止进行菌丝灭活,防止菌丝老化、自溶和杂菌利用,菌丝灭活液冷却后立即泵送入自动板框压滤进行固液分离;液相经反渗透膜切割为二部分,分子量3万及以下部分送中间罐进行下一步反渗透膜切割,分离液达中水标准排放或回用,高浓度部分送薄膜浓缩岗位进一步浓缩,至浓缩液密度1.09023~1.10687kg/L,投入经干燥破壁的菌丝体,装盘至烘箱,在60~80℃真空干燥12小时,得食品级或饲料添加剂级粗品,经不锈钢万能粉碎机80~100目粉碎,经检验合格后包装;分子量大于3万部分送至薄膜浓缩岗位,浓缩醇沉进入药品级,提取云芝胞外多糖;固相滤饼进入水提罐,水解温度96±2℃,水解时间6小时,加水量为菌丝体的4~5倍量,然后进入自动板框二次固液相分离,液相浓缩提取云芝胞内多糖,对固相菌丝体进行干燥,经200目破壁得云芝菌丝体蛋白粉,因云芝菌细胞壁较厚,固相菌丝体经破壁后也可再次水煮提取云芝胞内多糖或直接用作饲料级添加剂。
2.根据权利要求1所述的一种云芝糖肽工业化绿色生产工艺,其特征在于:步骤(3)中菌丝体提取的云芝胞内多糖粗品与云芝发酵液中提取的云芝胞外多糖粗品分开纯化,纯化达标后得云芝多糖药品级可混粉。
3.根据权利要求1所述的一种云芝糖肽工业化绿色生产工艺,其特征在于:步骤(2)中发酵大罐发酵中止条件为PH值4.5~5.5,镜检菌丝粗壮,呈网状,菌丝边缘及锁状联合清晰。
