Abstract

Description
背景技术
纤维化可以由损伤引起的结缔组织的增厚和瘢痕形成,其特征为成纤维细胞的过度增殖和细胞外基质(ECM)组分的积累。这种异常常见于包括肺、肝和肾等的器官中,引起组织结构破坏,并导致器官功能的严重损伤1,2。事实上,纤维化可以在几乎每个器官中发生,并且在多种慢性疾病中是导致终末期器官衰竭和死亡的主要原因3。肺纤维化的共同特点是肺的气囊(肺泡)周围的成纤维细胞过度增殖4。广泛的生物医学研究已确定,成纤维细胞数量的增加与它们在肺中过多的ECM沉积相结合,最终导致肺泡结构破坏,肺顺应性降低,并破坏气体交换功能5-7。
最常见的肺纤维化类型是特发性肺纤维化(IPF)。这种疾病最终影响整个肺叶,但它始于发生在外围区域的微小纤维化病变,并缓慢向内进展,并且这种纤维化最终可导致呼吸衰竭8,9。IPF是一种致命疾病,从诊断之时起存活时间的中位数仅为2-4年10。IPF的病理进展的机制和本质尚未被完全了解,尽管多项研究已暗示了肺泡上皮细胞的特定亚群——肺泡II型(AT2)细胞的贡献4,11。
肺的肺泡上皮由肺泡I型(AT1)和II型(AT2)细胞两者的组合构成。AT2细胞是肺泡干细胞,并且可以在肺泡稳态和损伤后修复期间分化成AT1细胞12,13。AT1细胞最终构成成人肺中肺泡表面几乎95%,是大的鳞状细胞,起到薄的气血屏障的上皮组分的作用14。在IPF组织中,异常增生的AT2细胞通常位于成纤维细胞灶点附近15,并且临床中,在IPF组织中常常观察到影响AT2细胞功能的基因突变体16,17。此外,鉴定IPF肺的分子谱的最新进展表明,在IPF肺的AT2细胞中TGFβ信号传导(许多纤维化疾病中共同的纤维化信号传导)被激活18。
肺纤维化患者的肺顺应性降低,气体交换被破坏,并且最终呼吸衰竭和死亡。据估计,在美国,IPF在65岁以上的成年人中的患病率为每200名中1名,存活时间中位数为2-4年。在中国,IPF的估计发病率为3-5/100,000,占所有间质性肺病的约65%。诊断通常在50至70岁之间做出,男女之比为1.5至2:1。患者的存活时间通常只有2-5年。
目前没有治愈IPF的药物。两种已知的药物尼达尼布和吡非尼酮对1年内用力肺活量的降低速率具有相似的作用。尽管两种药物都显示出降低死亡率的趋势,但这两种药物未能显示出存活时间的显著增加。主要原因之一是尚无肺纤维化、特别是特发性肺纤维化(IPF)的理想动物模型,以便筛选用于治疗肺纤维化、特别是特发性肺纤维化(IPF)的候选药物。
发明内容
本发明涉及用于构建肺纤维化、特别是特发性肺纤维化(IPF)的动物模型的方法,使用所述方法构建的动物模型,以及筛选用于治疗肺纤维化、特别是特发性肺纤维化(IPF)的候选药物的方法。本发明提供了一种肺纤维化、特别是特发性肺纤维化(IPF)的构建的疾病动物模型,并且所述构建的动物模型可用于研究肺纤维化、特别是特发性肺纤维化(IPF),筛选用于在动物和人类中治疗肺纤维化、特别是特发性肺纤维化(IPF)的候选药物,以及探索动物和人类的肺纤维化、特别是特发性肺纤维化(IPF)的药物靶点。
第一方面,本发明提供了一种构建肺纤维化、特别是特发性肺纤维化(IPF)的动物模型的方法,所述方法包括提高所述动物的肺泡上皮上的机械张力的步骤。
优选地,在所述提高肺泡上皮上的机械张力的步骤之前,所述动物经历肺切除术(PNX)。

本发明的特定实施方式的描述
特定实施方式和实施例的描述以说明而不是限制的方式提供。本领域技术人员将会容易地认识到可以被改变或修改以产生基本上相似的结果的各种不同的非关键性参数。
特发性肺纤维化(IPF)是一种以肺功能的进行性且不可逆的下降为特征的慢性肺病。症状通常包括逐渐出现呼吸短促和干咳。其他变化可能包括感觉疲倦和杵状甲。并发症可能包括肺动脉高压、心力衰竭、肺炎或肺栓塞。
肺的肺泡上皮由肺泡I型(AT1)和II型(AT2)细胞两者的组合构成。AT2细胞是肺泡干细胞,并且可以在肺泡稳态和损伤后修复期间分化成AT1细胞。AT1细胞最终构成成人肺中肺泡表面的几乎95%,是大的鳞状细胞,起到薄的气血屏障的上皮组分的作用14。在IPF组织中,异常增生的AT2细胞通常位于成纤维细胞灶点附近15,并且临床中,在IPF组织中常常观察到影响AT2细胞功能的基因突变体16,17。AT2生理功能异常导致进行性肺纤维化的确切病理机制仍有待阐明。
“动物模型”或“疾病动物模型”是用于研究和探索人类疾病的活的非人类动物,用于更好地理解疾病过程、病理机制的目的,并用于筛选有效药物和探索理想药物靶点的目的。
探索疾病的潜在药物靶点是药物发现中的第一步,并且也是筛选用于这个疾病的新药物的关键点。
在本发明的实施方式中,在PNX后的肺(具有显著提高的机械张力)中CDC42-GTP的表达水平被显著提高(图1A和图1B),并且这种CDC42-GTP表达的提高可以通过假体植入来抑制(图1A和图1B),在上述发现的基础上,本发明研究了缺失AT2细胞中的Cdc42基因在PNX诱导的肺泡再生期间的影响。**P<0.01,Student’s t检验。
在向动物给药他莫昔芬后,Sftpc基因启动子驱动的重组酶(Spc-CreER)被用于特异性缺失AT2细胞中的基因。CreER小鼠系统常用于诱导性基因的敲除研究。
在本发明的实施方式中,构建了一种AT2细胞中的Cdc42基因被特异性缺失的小鼠品系。在本发明中,所述小鼠被命名为Cdc42 AT2基因无效小鼠(图2)。在Cdc42 AT2基因无效小鼠的肺中,几乎没有AT2细胞分化成AT1细胞,并且在PNX后第21天没有新的肺泡形成(图3B)。
在本发明的实施方式中,某些Cdc42 AT2基因无效小鼠在PNX处理后第21天显示出显著的体重减轻和呼吸率提高(图4A和图4B)。事实上,在PNX后第60天,几乎50%的PNX处理的Cdc42 AT2基因无效小鼠达到终点安乐死的预定健康状况标准(图4B),并且到PNX后第180天,约80%的PNX处理的Cdc42 AT2基因无效小鼠达到它们的终点(图4B)。Cdc42 AT2基因无效小鼠在它们的终点时显示出肺中严重的纤维化(图4D)。
在本发明的实施方式中,PNX后的Cdc42 AT2基因无效小鼠在它们的终点时显示出,Cdc42 AT2基因无效小鼠肺中严重的纤维化(图4D与图4C相比)。从PNX后第21天开始,某些Cdc42 AT2基因无效肺的胸膜下区表现出组织增厚的迹象(图4D)。到终点时为止,致密的纤维化已进展到大多数Cdc42 AT2基因无效肺的中心。
在本发明的实施方式中,在Cdc42 AT2基因无效小鼠的肺中的致密纤维化区域中检测到胶原蛋白I(图4E),并且Cdc42 AT2基因无效小鼠中每个肺叶中表达胶原蛋白I的区域的比例在PNX处理后逐渐提高(图4F)。qPCR分析也显示从PNX后第21天起,胶原蛋白ImRNA表达水平逐渐提高(图4G)。此外,从PNX后第21天起,与PNX处理的对照小鼠相比在PNX处理的Cdc42 AT2基因无效小鼠中观察到肺顺应性的逐渐降低(图4H)。已知在肺变得纤维化时常常发生肺顺应性的降低19-24。
在本发明的实施方式中,从10月龄到24月龄的未经PNX处理的Cdc42 AT2基因无效小鼠(图5A)与Cdc42 AT2基因无效小鼠达到10月龄之前(图5C)相比,没有显示出显著的纤维化变化。在对照小鼠的肺中从未观察到纤维化变化,即使对照小鼠达到24月龄(图5B),但到12个月时,在Cdc42 AT2基因无效肺的胸膜下区中纤维化已明显开始发展,并进展到肺的中心(图5C)。
成纤维细胞灶点被认为是进行性肺纤维化的相关形态学标志物,并且被认为是在进行性肺纤维化中纤维化反应起始和/或持续的位点。成纤维细胞灶点含有增殖的α-SMA+成纤维细胞。
在本发明的实施方式中,观察到在Cdc42 AT2基因无效肺的相对正常的肺泡区域中一些α-SMA+成纤维细胞开始聚集到AT2细胞簇附近(区域1,图6A)。并且肺的致密纤维化区域充满α-SMA+成纤维细胞(区域2,图6A)。此外,在PNX后第21天,在Cdc42 AT2基因无效小鼠的肺中,α-SMA+细胞的增殖急剧增加,表明增殖的α-SMA+成纤维细胞有助于肺纤维化的发展(图6B)。
实施例
方法
小鼠和存活曲线记录。
Rosa26-CAG-mTmG(Rosa26-mTmG)、Cdc42 flox/flox小鼠25和Tgfbr2 flox/flox小鼠26以前已有描述。所有实验均按照北京生命科学研究所实验动物护理和使用指南(Guide forCare and Use of Laboratory Animals of the National Institute of BiologicalSciences)中的建议进行。为了监测小鼠的存活率,在PNX处理后每周称量对照和Cdc42 AT2基因无效小鼠两者的体重。一旦小鼠达到预先确定的终点标准,将这些小鼠处死。我们根据预先确定的判据定义终点27,28。
产生Spc-CreER;rtTA(Spc-CreER)敲入小鼠。
将CreERT2、p2a和rtTA元件通过酶法连接并插入到小鼠的内源SPC基因中。插入位点是内源SPC基因的终止密码子,然后在rtTA的3’末端处产生新的终止密码子。使用CRISPR/Cas9技术将所述CreERT2-p2a-rtTA片段插入到基因组中。
肺切除术(PNX)和假体植入。
将8周龄雄性小鼠每隔一天注射他莫昔芬(剂量:75mg/kg),共4次。在最后一剂他莫昔芬注射后第14天将小鼠麻醉并连接到呼吸机(Kent Scientific,Topo)。在第四肋间处切开胸壁并取出左肺叶。对于假体植入而言,将具有与左肺叶相近的尺寸和形状的软硅酮假体插入到空的左肺腔中。
肺功能测试。
苏木精和曙红(H&E)染色和免疫染色。
将肺用4%多聚甲醛(PFA)充盈,并在4℃下在4%PFA中连续固定24小时。然后将肺在30%蔗糖中冷冻保护并包埋在OCT(Tissue Tek)中。
H&E染色实验遵循标准的H&E方案。简而言之,将载片用水清洗以除去OCT。将细胞核用苏木精(Abcam,ab150678)染色2分钟,并将细胞质用曙红(Sigma,HT110280)染色3分钟。在脱水和澄清步骤后,将切片用中性树脂密封。
免疫荧光染色实验遵循已有方案的描述29。简而言之,在除去OCT后,将肺切片用3%BSA/0.1%TritonX-100/PBS封闭1小时,然后将载片与第一抗体在4℃温育过夜。在用0.1%TritonX-100/PBS清洗载片3次后,将切片与第二抗体在室温温育2小时。

对于p-SMAD2染色实验而言,在组织固定过程中在4%PFA中添加1X磷酸酶抑制剂(Bimake,B15002)。将酪胺信号放大法用于pSMAD2染色。
将人类肺组织在4℃下用4%PFA固定24小时,在30%蔗糖中冷冻保护并包埋在OCT中。所有实验均在北京生命科学研究所和北京中日友好医院的机构审查委员会的批准下进行。
统计分析。
所有数据均表示为平均值±s.e.m.(如图例中所示)。图中呈现的数据来源于不同小鼠在不同日期进行的多个独立实验。除非另有说明,否则图中呈现的大部分数据基于至少三个独立实验。样品间差异的推断的统计显著性使用双尾非配对Student’s t-检验来评估。
分离小鼠AT2细胞。
在4剂他莫昔芬注射后,如前文所述将Spc-CreER,Rosa26-mTmG小鼠的肺分离19,44。简而言之,将麻醉小鼠用中性蛋白酶(Worthington-Biochem,LS02111)和DNase I(Roche,10104159001)充盈。在BD FACS Aria II和III设备中使用单细胞选择模式利用GFP荧光分选AT2细胞。
分离人类AT2细胞。
用手术刀将人类肺组织切成小块,然后用中性蛋白酶(Worthington-Biochem,LS02111)、DNase I(Roche,10104159001)、I型胶原酶(Gibco,17100-017)和弹性蛋白酶(Worthington,2294)消化。然后在BD FACS Aria II和III设备中使用单细胞选择模式对所述的消化的悬浮液进行CD326+、HTII-280+CD45-、CD31-细胞的分拣。所有实验均在北京生命科学研究所和北京中日友好医院的机构审查委员会的批准下进行。
原代人类和小鼠AT2细胞培养和细胞拉伸测定法。
通过FACS分拣AT2细胞并将其铺于硅酮膜上24小时,然后进行拉伸实验。等轴应变系统和方法前文已有详细描述30。在使用25%的表面积变化进行的静态拉伸24小时后,对原代AT2细胞进行抗p-SMAD2抗体染色分析。使用TGFβ中和抗体(biolegend,521703)培养被拉伸的人类或小鼠AT2细胞,在培养基中加入1μg/ml TGFβ中和抗体。
定量RT-PCR(qPCR)。
使用Zymo Research RNA小量制备试剂盒(R2050)从全肺或原代AT2细胞分离总RNA。使用两步cDNA合成试剂盒(Takara,分类号6210A/B),按照制造商的建议进行反转录反应。qPCR使用CFX96 TouchTM实时PCR检测系统进行。用Gapdh mRNA水平对靶基因的mRNA水平进行归一。
用于qPCR的引物如下所列。
3D肺泡重建。
对于振动切片机切片而言,将肺用2%低熔点琼脂糖轻柔充盈至满容量。然后将肺在4%PFA中在4℃固定过夜。使用振动切片机(Leica VT100S)切出厚度为200μm的振动切片机切片。免疫染色实验按照标准的整体染色方案进行。Z堆栈图像通过Leica LSI宏观共焦显微镜和/或A1-R倒置共焦显微镜获取。
CDC42-GTP测定法。
使用CDC42活化测定生物化学试剂盒(cytoskeleton,#BK127),按照制造商提供的建议确定GTP-CDC42水平。简而言之,将整个肺叶在液氮中研磨,然后使用细胞裂解缓冲液(在试剂盒中附带)裂解。然后将细胞裂解物添加到附带的微孔板的孔中。在反应后,测量490nm处的吸光值。
在缺失Cdc42的外显子2之前和之后对Cdc42 DNA序列的片段测序,引物序列:正向:CTGCCAACCATGACAACCTAA(SEQ ID NO:1);反向:AGACAAAACAACAAGGTCCAG(SEQ ID NO:2)。
实施例1.产生一种AT2细胞中的Cdc42基因被特异性缺失的小鼠品系
1.为了构建进行性肺纤维化动物模型,通过特异性敲除肺泡II型细胞(AT2细胞)中的Cdc42基因产生了Cdc42 AT2基因无效小鼠。
为了特异性缺失AT2细胞中的Cdc42基因,将带有Spc-CreER敲入等位基因的小鼠与Cdc42 floxed(Cdc42flox/flox)小鼠杂交(图2A)。在Cdc42flox/flox小鼠中,含有Cdc42基因的翻译起始外显子的Cdc42基因的外显子2侧翼带有两个loxp位点。在Spc-CreER;Cdc42flox /flox小鼠中,AT2细胞中的Cdc42基因的外显子2在他莫昔芬处理后通过Cre/loxp介导的重组特异性缺失(图2B)。Spc-CreER;Cdc42flox/flox小鼠被命名为Cdc42 AT2基因无效小鼠。
2.在PNX处理后Cdc42 AT2基因无效小鼠的肺发展出进行性纤维化变化。
对Cdc42 AT2基因无效小鼠和对照小鼠进行了左肺叶切除(肺切除术,PNX)。在PNX处理后不同时间点分析了Cdc42 AT2基因无效小鼠和对照小鼠的肺(图4A)。我们发现在PNX后第21天,某些Cdc42 AT2基因无效小鼠显示出显著的体重减轻和呼吸频率提高。事实上,到PNX后第60天,几乎50%的PNX处理的Cdc42 AT2基因无效小鼠达到终点安乐死的预定健康状况标准(图4B),并且到PNX后第180天,超过70%的PNX处理的Cdc42 AT2基因无效小鼠(n=33)达到它们的终点(图4B)。H&E染色显示,模拟处理和PNX处理的对照小鼠的肺未显示出纤维化变化(图4C)。H&E染色显示,PNX处理的Cdc42 AT2基因无效小鼠的肺,在终点时与PNX后第21天的肺相比具有显著增加的纤维化区域(图4D)。
3.在PNX后第21天在Cdc42 AT2基因无效小鼠的肺的边缘处发展出纤维化变化。
在PNX后第21天,Cdc42 AT2基因无效小鼠的肺开始显示出纤维化变化。Spc-Cdc42flox/-肺在肺的边缘处已显示出致密的纤维化变化(图4D)。H&E染色显示,Cdc42 AT2基因无效肺的纤维化区域的组织学变化重现了人类IPF肺的组织学变化。
4.纤维化肺中胶原蛋白I沉积的表征和肺顺应性分析。
在PNX后第21天,将从对照和Cdc42 AT2基因无效小鼠收集的肺用抗胶原蛋白I抗体染色(图4E)。与对照组的肺相比,在Cdc42 AT2基因无效小鼠的肺的致密纤维化区域中检测到更强的胶原蛋白I的免疫荧光信号。从PNX后第21天至PNX后第60天,Cdc42 AT2基因无效小鼠的肺中致密胶原蛋白I的区域逐渐增加(图4F)。qPCR分析显示,在Cdc42 AT2基因无效小鼠的肺中,从PNX后第21天至第60天,胶原蛋白I mRNA的表达水平逐渐提高(图4G)。*P<0.05,***P<0.001;****P<0.0001,Student’s t检验。
在PNX后,Cdc42 AT2基因无效小鼠的肺的肺顺应性逐渐降低。
5.在未经PNX处理的Cdc42 AT2基因无效小鼠中,从12月龄左右开始发生进行性肺纤维化。
从2月龄开始,将对照和Cdc42 AT2基因无效小鼠,14天内给于4剂他莫昔芬。在10、12、16或24个月时收集未经PNX处理的对照和Cdc42 AT2基因无效小鼠的肺(图5A)。分析未经PNX处理的对照和Cdc42 AT2基因无效小鼠的肺,并发现在Cdc42 AT2基因无效小鼠达到10月龄之前没有显著的纤维化变化(图5B和5C)。到12个月时,在Cdc42 AT2基因无效肺的胸膜下区中明显开始发展出纤维化,并向肺的中心进展(图5C)。因此,AT2细胞中Cdc42的缺失导致了未经PNX处理的Cdc42 AT2基因无效小鼠从12月龄左右开始发生进行性肺纤维化。
6.在Cdc42 AT2基因无效小鼠的肺中α-SMA+成纤维细胞灶点的发展的表征。
成纤维细胞灶点被认为是进行性肺纤维化的相关形态学标志物,并被认为是进行性肺纤维化中纤维化反应起始和/或持续的位点31。成纤维细胞灶点含有增殖的α-SMA+成纤维细胞32。在PNX后第21天,将Cdc42 AT2基因无效小鼠的肺用抗α-SMA的抗体染色(图6A)。在Cdc42 AT2基因无效肺的相对正常的肺泡区域中,一些α-SMA+成纤维细胞开始聚集到AT2细胞簇附近(区域1,图6A)。并且肺的致密纤维化区域充满α-SMA+成纤维细胞(区域2,图6A)。此外,通过使用抗α-SMA和增殖标志物Ki67两者的抗体进行免疫染色,在PNX后第21天,在Cdc42 AT2基因无效小鼠的肺中α-SMA+细胞的增殖急剧增加。这些结果表明增殖的α-SMA+成纤维细胞促进了肺纤维化的进展(图6B)。**P<0.01,Student’s t检验。
实施例2.Cdc42 AT2基因无效小鼠的序列表征
对Spc-CreER,Cdc42flox/-小鼠进行基因组纯化和PCR扩增。然后纯化Cdc42的flox和缺失条带,并使用如下的引物测序:CTGCCAACCATGACAACCTAA(SEQ ID NO:1);AGACAAAACAACAAGGTCCAG(SEQ ID NO:2)。
在缺失Cdc42基因的外显子2之前或之后,Cdc42 DNA序列的片段示出在图9中。
实施例1和实施例2证实了Cdc42 AT2基因无效小鼠确实是进行性肺纤维化、特别是IPF的疾病动物。下面的实施例显示了Cdc42 AT2基因无效小鼠的特点以及Cdc42 AT2基因无效小鼠的用途。
实施例3.在PNX处理后肺泡再生期间或在正常肺泡稳态条件下,Cdc42为AT2细胞的分化所必需
我们对对照和Cdc42 AT2基因无效小鼠进行了PNX,并在PNX后第21天分析了肺泡再生和AT2细胞分化。如图3A中所示,将对照和Cdc42 AT2基因无效小鼠的200μm肺切片用针对GFP、Pdpn和Prospc的抗体进行免疫染色。在PNX后第21天,在未植入假体的对照肺中观察到许多新分化的AT1细胞和新形成的肺泡(图3B)。然而,在Cdc42 AT2基因无效肺中,在PNX后第21天,几乎没有AT2细胞分化成AT1细胞并且没有新肺泡形成(图3B)。观察到Cdc42 AT2基因无效肺的外围区域中,肺泡被严重过度拉伸(图3B)。
在正常稳态条件下,AT2细胞进行缓慢的自我更新并分化成AT1细胞,以建立新的肺泡。为了检测Cdc42是否为稳态期间AT2细胞分化所需,我们在小鼠为2月龄时缺失了AT2细胞中的Cdc42,并分析AT2细胞的命运直至小鼠达到12月龄。在12个月时收集对照和未经PNX处理的Cdc42无效小鼠的肺(图3C)。图像示出了用抗GFP、Pdpn和Prospc的抗体染色的肺切片的200μm Z-投影的最大强度。在12月龄对照小鼠的肺中,我们观察到许多新肺泡的形成(图3D)。然而,在12月龄Cdc42无效小鼠(未经PNX)的肺中,我们观察到增大的肺泡,并且没有任何新的AT1细胞形成(图3D)。
实施例4.AT2细胞中Cdc42的丧失在PNX处理的小鼠中引起进行性肺纤维化
在PNX后,对Cdc42 AT2基因无效小鼠和对照小鼠进行了更长时间的观察(图4A)。令人吃惊的是,在PNX后第21天,一些Cdc42 AT2基因无效小鼠显示出显著的体重减轻和呼吸频率提高。事实上,在PNX后第60天,几乎50%的PNX处理的Cdc42 AT2基因无效小鼠达到终点安乐死的预定健康状况标准(图4B),并且到PNX后第180天,约80%的PNX处理的Cdc42AT2基因无效小鼠达到它们的终点(图4B)。
PNX处理对照小鼠和Cdc42 AT2基因无效小鼠的H&E染色显示,Cdc42 AT2基因无效小鼠在它们的终点时,肺中出现了严重的纤维化(图4D与图4C相比)。为了确定在PNX后Cdc42 AT2基因无效小鼠开始发生肺纤维化的时间点,在PNX后各个不同时间点使用H&E染色分析了Cdc42 AT2基因无效小鼠的肺(图4D)。到PNX后第21天,一些Cdc42 AT2基因无效肺的胸膜下区表现出组织增厚的迹象(图4D)。到终点时为止,致密的纤维化已进展到大多数Cdc42 AT2基因无效肺的中心(图4D)。我们在PNX后和老年Cdc42 AT2基因无效小鼠中观察到的情况类似于IPF的特征性进展,其中纤维化病变首先发生在肺的外周,随后向内进展到肺叶的中心。
除了在Cdc42 AT2基因无效小鼠的肺的这些致密纤维化区域中检测到胶原蛋白I的强烈免疫荧光信号(图4E)之外,我们还观察到在Cdc42 AT2基因无效小鼠中,每个肺叶中表达胶原蛋白I的区域的比例在PNX后逐渐提高(图4F)。我们的qPCR分析也显示从PNX后第21天起,胶原蛋白I mRNA表达水平逐渐提高(图4G)。此外,从PNX后第21天起,与PNX处理的对照小鼠相比在PNX处理的Cdc42 AT2基因无效小鼠中观察到肺顺应性的逐渐降低(图4H),考虑到已知在肺变得纤维化时常常发生肺顺应性的降低19-24,这是一个有趣的发现。
实施例5.AT2细胞中Cdc42的丧失在未经PNX处理的老年小鼠中导致进行性肺纤维化
由于在12月龄Cdc42 AT2基因无效小鼠中发现AT2分化受损和肺泡增大(图3D),因此分析了从10月龄到24月龄的未经PNX处理的对照和Cdc42 AT2基因无效小鼠的肺(图5A)。在对照小鼠的肺中从未观察到纤维化变化,即使对照小鼠达到24月龄(图5B)。在Cdc42 AT2基因无效小鼠达到10月龄之前我们没有发现显著的纤维化变化(图5C)。观察至12个月时,在Cdc42 AT2基因无效肺的胸膜下区中纤维化明显开始发展,并在12个月后进展到肺的中心(图5C)。
总而言之,在PNX处理的小鼠中,AT2细胞中Cdc42的丧失导致进行性肺纤维化。此外,在未经PNX处理的Cdc42 AT2基因无效小鼠中,这种进行性肺纤维化表型开始发生于12月龄左右。所有这些结果证实在小鼠中,AT2细胞中Cdc42的缺失导致IPF样进行性肺纤维化,因此建立了IPF样进行性肺纤维化的小鼠模型,其可用于研究人类IPF疾病。
实施例6.在Cdc42 AT2基因无效小鼠的肺中α-SMA+成纤维细胞灶点的发展
成纤维细胞灶点被认为是进行性肺纤维化相关的形态学标志物,并且被认为是在进行性肺纤维化中纤维化反应起始和/或持续的位点。成纤维细胞灶点含有增殖的α-SMA+成纤维细胞。在PNX后第21天将Cdc42 AT2基因无效小鼠的肺用抗α-SMA的抗体染色(图6A)。观察到在Cdc42 AT2基因无效肺的相对正常的肺泡区域中,一些α-SMA+成纤维细胞开始聚集到AT2细胞簇附近(区域1,图6A)。并且肺的致密纤维化区域充满α-SMA+成纤维细胞(区域2,图6A)。此外,通过使用抗α-SMA和增殖标志物Ki67两者的抗体进行免疫染色,我们的结果显示在PNX后第21天,在Cdc42 AT2基因无效小鼠的肺中α-SMA+细胞的增殖急剧增加。这些结果表明在Cdc42 AT2基因无效小鼠的肺中,增殖的α-SMA+成纤维细胞促进了肺纤维化的发展(图6B)。
实施例7.由受损的肺泡再生引起的高机械张力导致进行性肺纤维化
在Cdc42 AT2基因无效小鼠中肺纤维化被PNX处理极大加速(图4),这一事实表明在肺纤维化和机械张力诱导的肺泡再生之间存在密切联系。
由PNX引起的肺泡丧失显著提高了作用于肺泡上皮上的机械张力。随后在正常小鼠中发生的肺泡高效再生最终将机械张力的强度降低到PNX处理前的水平;然而,由于Cdc42 AT2基因无效细胞不能分化成AT1细胞,并因此不能再生新的肺泡(图3A和3B),因此Cdc42 AT2基因无效小鼠的肺泡上皮持续经历升高的机械张力,这导致了纤维化的进行性发展(图4)。
实施例8.在AT2细胞中高机械张力激活TGFβ/SMAD信号传导的正反馈回路

因此,AT2细胞中TGFβ/SMAD信号传导的正反馈回路将是用于筛选肺纤维化、特别是特发性肺纤维化(IPF)的候选药物的理想药物靶点。
实施例9.在AT2细胞中降低TGFβ信号传导减弱了肺纤维化的进展
为了进一步评估在PNX后第21天对照和Cdc42 AT2基因无效小鼠的肺中TGFβ信号传导的活性,我们使用抗p-SMAD2的抗体进行了免疫染色实验,p-SMAD2是经典TGFβ信号传导活性的指示物(图8A)。尽管在对照肺中几乎没有AT2细胞有核p-SMAD2表达,但无效Cdc42肺中的许多AT2细胞有核p-SMAD2表达(图8A),表明在PNX后第21天Cdc42 AT2基因无效小鼠的AT2细胞中TGFβ信号传导被强烈激活。IPF样本中的许多AT2细胞也有核p-SMAD2表达,证实了在IPF肺的AT2细胞中TGFβ信号传导的激活(图8B)。
众所周知,TGFβ配体与TGFBR2的结合对TGFβ/SMAD信号传导的活化是必需的33。我们构建了AT2细胞中的Tgfbr2和Cdc42两个基因均被缺失的Tgfbr2&Cdc42 AT2双重基因无效小鼠。我们对Cdc42 AT2基因无效和Tgfbr2&Cdc42 AT2双重基因无效小鼠进行了左肺切除,并在PNX后对这些小鼠进行了180天的观察(图8C)。引人注目的是,到PNX后第180天,Tgfbr2&Cdc42AT2双重基因无效小鼠存活率为80%,而到此时,Cdc42 AT2基因无效小鼠存活率低于40%(图8D)。这些结果提供了令人信服的证据,表明AT2细胞中激活的TGFβ信号传导驱动Cdc42 AT2基因无效小鼠中肺纤维化的发展。TGFβ配体Tgfb1是TGFβ/SMAD信号传导的下游靶点之一。通过qPCR分析,我们发现Tgfb1的表达水平在Cdc42 AT2基因无效细胞中显著提高,但在Tgfbr2&Cdc42双重基因无效的AT2细胞中没有显著提高(图8E)。这表明由于TGFβ/SMAD信号传导的提高,Cdc42 AT2基因无效细胞产生更多的TGFβ配体。**P<0.01,Student’s t检验。
这个实施例表明Cdc42 AT2基因无效小鼠可用于IPF样进行性肺纤维化的新的药物靶点的发现,并且AT2细胞中的TGFβ信号传导是一个理想靶点。
结果:
本文为了研究受损的肺泡再生的长期影响,我们在左肺叶切除后,对Cdc42 AT2基因无效和同窝仔畜对照(Control)小鼠进行了更长时间的观察(图4A)。令人吃惊的是,我们发现在PNX后第21天后,一些Cdc42 AT2基因无效小鼠显示出显著的体重减轻和呼吸率提高。事实上,到PNX后第60天,几乎50%的PNX处理的Cdc42 AT2基因无效小鼠达到终点安乐死的预定健康状况标准(图4B),并且到PNX后第180天,超过70%的PNX处理的Cdc42 AT2基因无效小鼠达到它们的终点(图4B)。
PNX处理对照和Cdc42 AT2基因无效小鼠的H&E染色显示,Cdc42 AT2基因无效小鼠在它们的终点时,肺中出现了严重的纤维化(图4D与图4C相比)。为了确定在PNX后Cdc42AT2基因无效小鼠开始发生肺纤维化的时间点,我们在PNX后各个不同时间点使用H&E染色分析了Cdc42 AT2基因无效小鼠的肺(图4D)。到PNX后第21天,一些Cdc42 AT2基因无效肺的胸膜下区表现出组织增厚的迹象(图4D)。到终点时为止,致密的纤维化已进展到大多数Cdc42 AT2基因无效肺的中心。
除了在PNX后第21天在Cdc42 AT2基因无效小鼠的肺的这些致密纤维化区域中检测到胶原蛋白I的强烈免疫组织学信号(图4E)之外,从PNX后第21天到PNX后第60天,Cdc42AT2基因无效小鼠的肺中的致密胶原蛋白I的区域逐渐增加(图4F)。qPCR分析显示从PNX后第21天到PNX后第60天,Cdc42 AT2基因无效小鼠的肺中胶原蛋白I mRNA表达水平逐渐提高(图4G)。*P<0.05,***P<0.001;****P<0.0001,Student’s t检验。
此外,与PNX处理的对照小鼠相比,在PNX处理的Cdc42 AT2基因无效小鼠中肺顺应性显著降低(图4H),考虑到已知在肺发生纤维化时常常发生FVC降低和肺顺应性的降低19-24,这是一个有趣的发现。
我们还分析了未经PNX处理的对照和Cdc42 AT2基因无效小鼠的肺,并发现在Cdc42 AT2基因无效小鼠达到10月龄之前没有显著的纤维化变化(图5A-5C)。到12个月时,在Cdc42 AT2基因无效肺的胸膜下区中纤维化明显开始发展,并朝向肺的中心进展(图5C)。
综上,这些结果表明在PNX处理的小鼠中,AT2细胞中Cdc42的丧失导致进行性肺纤维化。此外,在未经PNX处理的Cdc42 AT2基因无效小鼠中,这种进行性肺纤维化表型开始发生于12月龄左右。
成纤维细胞灶点被认为是进行性肺纤维化相关的形态学标志物,并且被认为是在进行性肺纤维化中纤维化反应起始和/或持续的位点31。成纤维细胞灶点含有增殖的α-SMA+成纤维细胞32。因此,我们将Cdc42 AT2基因无效小鼠的肺用抗α-SMA以及细胞增殖标志物Ki67的抗体染色(图6A),来表征纤维化肺中各种不同的基质细胞类型的增殖。在Cdc42 AT2基因无效肺的相对正常的肺泡区域中,一些α-SMA+成纤维细胞开始聚集到AT2细胞簇附近(区域1,图6A)。并且肺的致密纤维化区域充满α-SMA+成纤维细胞(区域2,图6A)。这个分析揭示出所有纤维化肺均含有增殖的α-SMA+成纤维细胞(图6A和6B),表明这些小鼠基质细胞促进了肺纤维化的发展。**P<0.01,Student’s t检验。
所有这些结果证实,AT2细胞中Cdc42的缺失导致在小鼠中IPF样进行性肺纤维化,因此建立了IPF样进行性肺纤维化的小鼠模型,其可用于研究人类IPF疾病。
5.讨论
如上所示,在肺损伤后,AT2细胞中Cdc42的丧失导致进行性肺纤维化。我们在这里观察到的肺纤维化的渐进式发展明显类似于在IPF患者中发生的病理过程,其中纤维化最初始于肺的外围区域,随后缓慢向内前进,最终影响整个肺叶。
所有这些结果证实,AT2细胞中Cdc42的缺失导致在小鼠中IPF样进行性肺纤维化,因此建立了IPF样进行性肺纤维化的小鼠模型,其可用于研究人类IPF疾病。

43.检测权利要求1-23中的任一项所述的动物模型的方法,所述方法使用在SEQ IDNO:4所示的序列的基础上设计的一对引物。
44.权利要求43所述的方法,其中用于检测所述动物模型的引物如下所示:
