Abstract
本申请提出一种食品级油水双相负载乳液凝胶运载体系的制备方法,采用双诱导方式对负载有不同模式功能因子的乳液进行诱导,所有原料均为可食用原材料,所制备的SPI‑SBP乳液凝胶运载体系可用于营养功能因子的口服递送。采用漆酶与其他诱导方式结合的双诱导法制备,具备更好的包埋及模拟消化控释效果,保护模式功能因子抵抗严苛的消化环境,使其尽可能多的到达小肠并在小肠顺利释放。可以同时包埋水溶性和脂溶性的功能因子。漆酶与不同诱导方式的结合,起到了不同的功能因子包埋和控释效果,通过调节漆酶与不同诱导方式的组合方式,可以实现对SPI‑SBP乳液凝胶运载体系所包埋功能因子的控释调控,以达到功能因子更好地被人体吸收利用的目的。

技术领域
本申请涉及食品加工技术领域,尤其涉及一种食品级油水双相负载乳液凝胶运载体系的制备方法。
背景技术
目前现代食品工业日益注重食品对营养及健康的改善,含有功能性成分的食物正日渐流行。但是限制其发展的一个主要问题是许多天然功能因子(如某些维生素、类胡萝卜素等)通常具有自身稳定性差,对外界环境敏感,水溶性差,易发生降解等问题,通常需要借助传递体系添加到食品中提高生物利用率。目前对乳液和凝胶两大功能因子运载体系的研究较为充分。
在乳液和凝胶的基础上,研究发展了乳液凝胶递送载体。乳液凝胶是同时含有乳化分散油滴及凝胶水介质的复合凝胶体系,其特点是乳化的油滴被包裹在凝胶网络中起到支撑物的作用,油相能够包埋脂溶性功能因子,是理想的营养物包埋缓释载体。乳液凝胶的诱导方式可分为热诱导、酸诱导、酶诱导和离子诱导等。
水包油型乳液能够在水连续相中运输或溶解疏水性成分,并且能够对功能因子在贮藏、消化过程中起到一定的保护作用,但缺点是不稳定,在贮藏过程中容易发生相分离、聚集絮凝等现象,并且在消化过程中容易被破坏降解。凝胶的优点是具有一定的机械强度,能够将功能因子固定在凝胶网络中,可以一定程度上更好的保护功能因子免受极端环境的影响,但缺点是水凝胶基质难以包埋脂溶性营养素,具有一定的应用局限性。其他运载体系如脂质体及纳米颗粒也同样存在不稳定或安全性等问题。
对于乳液凝胶体系来说,不同诱导方式由于作用机理不同,形成的凝胶结构可能会有所差异,而功能因子的稳定性和释放能力将很大程度依赖传递体系的微观结构。此外,在蛋白/多糖复合基质乳液凝胶中,由于多相体系的复杂性,两种诱导方式的结合以及凝胶条件的改变可能会存在一定程度的相互影响(如离子或pH可能会影响酶活性),从而对复合乳液凝胶的凝胶结构、机械性质及消化特性产生影响,目前关于双重诱导方式对乳液凝胶结构、消化特性及营养物包埋释放特性的研究较少。
更重要的是,虽然乳液凝胶同时具备水相和油相,拥有水相和油相分别同时包埋递送不同溶解特性功能因子的潜质,但目前的相关研究报道很少。在现有技术的乳液凝胶运载体系中,油水两相的功能因子同时负载到乳液凝胶中很难实现,即便实现了,其油水两相的功能因子的包埋率和释放率也都欠佳,无法在体内发挥协同增效的作用。

本申请旨在提供一种食品级油水双相负载乳液凝胶运载体系的制备方法。将不同溶解特性(水溶性和脂溶性)的功能因子同时负载于同一食品级口服乳液凝胶递送体系中,并通过改变诱导方式,对功能因子在模拟消化环境中的释放进行调控,为不同功能因子在体内发挥协同增效的作用提供可能。
为此,本申请的第一方面实施例提出一种油水双相负载乳液凝胶运载体系的制备方法,采用诱导方式对负载有水溶性功能因子和脂溶性功能因子的乳液进行诱导,得到油水双相负载乳液凝胶运载体系。
在一些实施例中,所述诱导方式包括单诱导方式或双诱导方式,当采用单诱导方式对所述乳液进行诱导时,得到单诱导乳液凝胶运载体系;当采用双诱导方式对所述乳液进行诱导时,得到双诱导乳液凝胶运载体系。
在一些实施例中,双诱导方式为:在所述乳液中加入GDL、MTG、氯化钙或氯化镁中的任一种,充分搅拌后再加入漆酶,充分搅拌后放入培养箱中保温静置一段时间后,得到双诱导乳液凝胶运载体系。
在一些实施例中,单诱导方式为:在所述乳液中加入漆酶,充分搅拌后放入培养箱中保温静置一段时间后,得到单诱导油水双相负载乳液凝胶运载体系。
在一些实施例中,所述乳液为SPI-SBP乳液,SPI-SBP乳液的制备方法,包括如下步骤:
A1,将SPI粉末分散于去离子水中,充分搅拌后,放入冰箱内冷藏一段时间至SPI充分水化,得到SPI水溶液;
A2,将SBP粉末加入到SPI水溶液中,充分搅拌至溶解完全,得到SPI-SBP水溶液,放入冰箱内冷藏一段时间后,在80-90℃下预热10-20min,随后立即放置于冰水浴中迅速冷却至室温,向其中加入水溶性功能因子核黄素粉末,充分搅拌,得到乳液的水相;
A3,将脂溶性功能因子β-胡萝卜素溶于MCT中,搅拌至充分溶解,得到乳液的油相;
A4,向水相中添加油相,使用高速剪切机剪切后制成粗乳液,再使用高压均质机均质后,得到SPI-SBP乳液。
在一些实施例中,所述水溶性功能因子为核黄素粉末,脂溶性功能因子为β-胡萝卜素。
在一些实施例中,所述油水双相负载乳液凝胶运载体系为SPI-SBP双诱导乳液凝胶运载体系,制备方法包括如下步骤:称取一定量的SPI-SBP乳液,加入质量百分比为0.3%-1.8%的GDL、5-30U/g蛋白的MTG、5-50mmol/L氯化钙或5-50mmol/L氯化镁中的任一种,充分搅拌后,再加入15-25U/g底物的漆酶,充分搅拌后,放入培养箱中保温静置一段时间后,得到SPI-SBP双诱导乳液凝胶运载体系。
在一些实施例中,所述油水双相负载乳液凝胶运载体系为SPI-SBP单诱导乳液凝胶运载体系,制备方法包括如下步骤:称取一定量的SPI-SBP乳液,加入15-25U/g底物的漆酶,充分搅拌后,放入培养箱中保温静置一段时间后,得到SPI-SBP单诱导乳液凝胶运载体系。
本申请的第二方面实施例提出一种由上述的制备方法制备出的油水双相负载乳液凝胶运载体系。
本申请的第三方面实施例提出一种该乳液凝胶运载体系的制备方法在食品加工领域中的应用。
本申请的第四方面实施例提出一种油水双相负载乳液凝胶运载体系中不同功能因子包埋率的测定方法,包括如下步骤:
B1,样品前处理:将双诱导乳液凝胶运载体系进行冻干处理,称取一定量双诱导乳液凝胶冻干样品浸泡在去离子水中,浸泡至使乳液凝胶充分吸水溶胀,使未包埋的模式营养物充分游离出来,得到样品-水共混体系;
B2,β-胡萝卜素包埋率的测定:将样品-水共混体系中加入正己烷,混合均匀,离心,收集上清液,在450nm处测定吸光度,根据β-胡萝卜素标准曲线计算样品中上清液的β-胡萝卜素的含量,β-胡萝卜素的包埋率计算公式如下:
B3,核黄素包埋率的测定:将样品-水共混体系离心,收集上清液,在445nm处测定吸光度,根据核黄素标准曲线计算样品中上清液的核黄素的含量,核黄素的包埋率计算公式如下:
本申请的第五方面实施例提出一种SPI-SBP双诱导乳液凝胶运载体系中功能因子在模拟消化液环境中的释放率的测定方法,包括如下步骤:
C1,模拟口腔、胃部、肠道三种消化环境构建消化体系,利用制备好的SPI-SBP双诱导乳液凝胶运载体系,分别配置与模拟唾液、模拟胃液和模拟肠液对应的三种消化液样品,将待测的三种消化液样品的pH值调至中性;
C2,β-胡萝卜素释放率的测定:分别取1mL消化液样品加入1mL正己烷,混合均匀后离心,取上清液,利用紫外分光光度计在波长450nm下测定上清液的吸光值,通过β-胡萝卜素标准曲线计算消化液样品中β-胡萝卜素的含量;
核黄素释放率的测定:分别取1mL消化液样品进行离心,取上清液,利用紫外分光光度计在波长445nm下测定上清液吸光值,通过核黄素标准曲线计算消化液样品中核黄素的含量;
功能因子的释放率计算公式如下:
本技术方案可带来的有益效果:
1、所有原料均为可食用原材料,因此所制备的SPI-SBP乳液凝胶运载体系可用于营养功能因子的口服递送,作为营养强化剂的有效载体。
2、SPI-SBP乳液凝胶运载体系采用漆酶与其他诱导方式结合的双诱导法制备,与乳液体系及漆酶单诱导体系相比,具备更好的包埋及模拟消化控释效果,保护模式功能因子抵抗严苛的消化环境,使其尽可能多的到达小肠并在小肠顺利释放,是一种具有更强控释能力的新型功能因子递送体系。
3、因SPI-SBP乳液凝胶运载体系同时存在水相和油相,所以可以同时包埋水溶性和脂溶性的功能因子,通过试验证明,该乳液凝胶运载体系对水溶性和脂溶性功能因子都具备良好的胃肠控释能力,同时适用于水溶性和脂溶性功能因子口服包埋递送,是一种具备水相和油相双负载能力的运载体系,为不同溶解特性功能因子在体内的协同增效提供了新方法。
4、漆酶与不同诱导方式的结合,起到了不同的功能因子包埋和控释效果,因此,通过调节漆酶与不同诱导方式的组合方式,可以实现对SPI-SBP乳液凝胶运载体系所包埋功能因子的控释调控,以达到功能因子更好地被人体吸收利用的目的。
5、本案制备出的乳液凝胶运载体系最终的呈现方式是固体,便于储存和运输,稳定性好,加工工艺简单,容易实现工业化生产,成本低,没有引入有毒化学药品,可大剂量用于保健食品中。

具体实施方式
下面详细描述本申请的实施例,所述实施例的示例在附图中示出。下面通过参考附图描述的实施例是示例性的,旨在用于解释本申请,而不能理解为对本申请的限制。
下面参考附图1-3描述本申请实施例的一种食品级油水双相负载乳液凝胶运载体系的制备方法。
以下描述所用到的原料的缩写:
SPI:大豆分离蛋白
SBP:甜菜果胶
MCT:中链甘油三酯
GDL:葡萄糖酸-δ-内酯
MTG:转谷氨酰胺酶
本申请的实施例提出一种食品级油水双相负载乳液凝胶运载体系的制备方法,采用诱导方式对负载有水溶性功能因子和脂溶性功能因子的乳液进行诱导,得到油水双相负载乳液凝胶运载体系。所述诱导方式包括单诱导方式或双诱导方式。
以下通过具体的实施例对本案进行进一步描述。
所述乳液为SPI-SBP乳液,所述油水双相负载乳液凝胶运载体系为SPI-SBP双诱导乳液凝胶运载体系。
1、SPI-SBP乳液的制备方法:
如图1所示,将SPI粉末分散于去离子水中,使得SPI水溶液的浓度为6.0%(w/w),在室温下磁力搅拌4h后,将样品放在4℃冰箱过夜(16-18h),以保证大豆分离蛋白充分水化,得到SPI水溶液。次日,将SBP粉末以2.0%(w/w)的浓度加入水化好的SPI水溶液中,经磁力搅拌器充分搅拌4h至溶解,从而获得相应浓度的SPI-SBP水溶液,并放置于4℃冰箱过夜(16-18h)。次日将SPI-SBP分散液在85℃下预热15min,随后立即置于冰水浴中迅速冷却至室温,向其中加入核黄素粉末(水溶性功能因子),使其在水中的浓度为0.6mg/ml,在室温下避光充分搅拌,作为乳液的水相。将β-胡萝卜素(脂溶性功能因子)以0.2%(w/w)的比例溶于已加热至45℃的MCT中,磁力搅拌至β-胡萝卜素充分溶解,部分难溶颗粒采用超声手段帮助溶解,作为乳液的油相。向水相中添加20%(w/w)的油相,使用高速剪切机以10000r/min的转速剪切3min制成粗乳液;再使用高压均质机30MPa均质压力均质五次,得到SPI-SBP乳液样品。
在一些实施例中,所选用的水溶性功能因子和脂溶性功能因子可以不限于核黄素粉末和β-胡萝卜素。
2、SPI-SBP双诱导乳液凝胶运载体系的制备方法:
称取一定量新鲜的SPI-SBP乳液样品,分别加入GDL(0.3%-1.8%,w/w)、MTG(5-30U/g蛋白)、氯化钙(5-50mmol/L)或氯化镁(5-50mmol/L),进行对比试验,充分搅拌1min后加入一定量漆酶(20U/g底物),再充分搅拌1min,随后放入40℃培养箱中保温静置4h。然后将经过保温的SPI-SBP双诱导乳液凝胶运载体系移入4℃冰箱过夜备测。
3、SPI-SBP单诱导乳液凝胶运载体系的制备方法:
称取一定量新鲜的SPI-SBP乳液样品,加入一定量漆酶(20U/g底物),充分搅拌1min,随后放入40℃培养箱中保温静置4h。然后将经过保温的SPI-SBP单诱导乳液凝胶运载体系移入4℃冰箱过夜备测。
4、运载体系中不同模式功能因子包埋率的测定方法:
(1)样品前处理:将负载有功能因子的乳液凝胶运载体系样品进行冻干处理,乳液凝胶样品冻干前先切成约0.2cm×0.2cm×0.2cm的小立方体。称取0.5g乳液凝胶冻干样品放于10mL去离子水中,在室温下浸泡24h使乳液凝胶充分吸水溶胀,使未包埋的模式营养物充分游离出来,得到样品-水共混体系。
将负载有功能因子的乳液样品同样进行前处理,用于进行对比实验,乳液样品无需在冻干前切成小立方体,因其本身就是液态。
(2)样品中β-胡萝卜素包埋率的测定:将所得样品-水共混体系加入10mL正己烷,在涡旋仪上充分振荡混合均匀后,在10000r/min下离心10min,收集上清液,在450nm处测定吸光度,实验过程注意避光。根据β-胡萝卜素标准曲线计算样品中上清液的β-胡萝卜素的含量,β-胡萝卜素的包埋率计算公式如下:
(3)样品中核黄素包埋率的测定:将所得样品-水共混体系在10000r/min转速下离心10min,上清液在445nm处测定吸光度,实验过程注意避光。根据核黄素标准曲线计算样品中上清液的核黄素含量,核黄素的包埋率计算公式如下:
对比实验:将漆酶分别与GDL、MTG、氯化钙或氯化镁双诱导方式制备的SPI-SBP双诱导乳液凝胶运载体系对β-胡萝卜素和核黄素在乳液凝胶运载体系中包埋率的影响进行比较,并引入未诱导的乳液和漆酶单诱导的乳液凝胶运载体系进行对比试验。实验结果见表1。

注:Laccase代表漆酶单诱导制备的SPI-SBP乳液凝胶运载体系,L&GDL,L&MTG,L&CaCl2,L&MgCl2分别代表漆酶与GDL,MTG,CaCl2和MgCl2双诱导制备的SPI-SBP乳液凝胶运载体系,添加量分别为漆酶:20U/g底物,GDL:0.9%(w/w),MTG:20U/g,CaCl2:50mmol/L,MgCl2:40mmol/L。
结论:本案所制备的乳液、漆酶单诱导乳液凝胶运载体系和双诱导乳液凝胶运载体系均对功能因子具有较好的包埋效果。但与乳液及漆酶单诱导乳液凝胶运载体系相比较,双诱导乳液凝胶运载体系对功能因子具有更好的包埋效果。虽然从数据上看漆酶单诱导乳液凝胶运载体系中β-胡萝卜素的包埋率比其他双诱导乳液凝胶运载体系的包埋率数值略高,但差距微乎其微,没有显著性,可以认为单诱导方式和双诱导方式对β-胡萝卜素的包埋率效果相当。而对于核黄素包埋率来说,差距很明显,可以看出双诱导乳液凝胶运载体系对β-胡萝卜素的包埋率远高于单诱导乳液凝胶运载体系和乳液。
四种双诱导乳液凝胶运载体系中,L&MTG双诱导乳液凝胶运载体系对于油水两相模式营养物均有最高的包埋率,L&MgCl2双诱导乳液凝胶运载体系对β-胡萝卜素的包埋率较低,L&GDL双诱导乳液凝胶运载体系对核黄素包埋率较低。
本案选取了较为典型的β-胡萝卜素和核黄素作为代表,实际经过大量实验发现,对比乳液及单诱导乳液凝胶运载体系来说,双诱导乳液凝胶运载体系对油水双相功能因子具有更好的包埋效果。
5、SPI-SBP双诱导乳液凝胶运载体系中功能因子在模拟消化液环境中的释放率的测定方法:
5.1体外模拟消化体系的建立
乳液凝胶运载体系的体外模拟消化体系的实验方案通过模拟口腔、胃部、肠道三个部分的消化环境构建此消化体系,分别配置模拟唾液(SSF),模拟胃液(SGF)和模拟肠液(SIF)三种消化液,三种消化液的主要制备方法如表2所示。

具体不同阶段消化实验的操作步骤如下:
(1)模拟口腔:
将黏蛋白(0.3g/L)加入至5mL的模拟口腔消化液(SSF)中混合,最后加入25μL氯化钙(0.3mol/L)和1mL的去离子水,混合均匀后用盐酸和氢氧化钠溶液调节pH至7.0,得到调配后的模拟唾液。
取5g的乳液(用于对比实验)或乳液凝胶运载体系样品与新鲜调配后的模拟唾液混合,其中乳液凝胶运载体系样品预先切成约0.2cm×0.2cm×0.2cm的小立方体,在37℃的水浴摇床中以100rpm振荡5min,口腔消化结束时用4mol/L HCl调节pH至2.5。得到模拟口腔的消化液样品。
(2)模拟胃部:
取7.5mL的模拟胃液(SGF)加入1.5mL去离子水,随后加入胃蛋白酶(25000U/mL),最后加入5μL氯化钙(0.3mol/L),用盐酸调节pH到2.5,得到调配后的模拟胃液。
将新鲜调配后的模拟胃液加入到口腔消化结束后的混合液中,在37℃的水浴摇床中以100rpm振荡120min,胃相消化结束时用4mol/L NaOH调节pH至7.0。得到模拟胃部的消化液样品。
(3)模拟肠道:
用11mL的模拟肠液(SIF)混合均匀后加入5.0mL胰酶溶液(2mg/mL),最后加入2.5mL 160mmol/L的胆盐溶液和40μL 0.3mol/L CaCl2后,用氢氧化钠调节pH到7.0,得到调配后的模拟肠液。
将新鲜调配后的模拟肠液加入到胃相消化结束后的混合液中,在37℃的水浴摇床中以100rpm振荡120min。得到模拟肠道的消化液样品。
5.2不同模式功能因子在模拟消化过程中释放率的测定
(1)在测定释放率前,将上述所有的消化液样品用一定浓度的盐酸或氢氧化钠调至中性。实验过程中注意避光操作。功能因子的释放率公式如下:
(2)β-胡萝卜素释放率的测定:分别取1mL消化液样品加入1ml正己烷,在涡旋仪上充分振荡混合均匀后,在10000r/min转速下离心10min,利用紫外分光光度计在波长450nm下测定上清液吸光值,通过β-胡萝卜素标准曲线计算消化液样品中β-胡萝卜素的含量。
(3)核黄素释放率的测定:分别取1mL消化液样品在10000r/min转速下离心10min。利用紫外分光光度计在波长445nm下测定上清液吸光值,通过核黄素标准曲线计算消化液样品中核黄素的含量。
对比实验:将漆酶分别与GDL、MTG、氯化钙或氯化镁双诱导方式制备的SPI-SBP双诱导乳液凝胶运载体系在口腔、胃相、肠相中β-胡萝卜素和核黄素的消化释放率进行比较,并引入未诱导的乳液和漆酶单诱导的乳液凝胶运载体系进行对比试验。实验结果见图2-3。图中P1、P2、P3分别代表口腔、胃相、肠相的消化过程。
结论:本案中所制备的乳液、单诱导乳液凝胶运载体系和双诱导乳液凝胶运载体系对功能因子均表现出较好的缓释特性。但对比乳液体系,各诱导条件下乳液凝胶运载体系能够减缓功能因子在胃相的释放,且对水溶性及脂溶性功能因子在不同消化阶段呈现出不同的缓释特性。脂溶性β-胡萝卜素在胃相具有更低的消化释放率(10%左右),而水溶性核黄素在胃相消化结束时消化释放率较高(10%-20%)。此外,不同双诱导乳液凝胶运载体系之间功能因子消化释放规律也有所差异。对于脂溶性功能因子,不同诱导方式的乳液凝胶运载体系在胃相消化过程中未表现出体系间的显著差异,但是L&MTG以及L&GDL双诱导乳液凝胶运载体系在肠相消化结束时的β-胡萝卜素释放率显著低于其他体系(60-65%),因此可以在有效吸收部位(小肠)作用更长的时间,达到更好的递送效果,如图2所示。对于水溶性功能因子核黄素,随着时间的延长,样品间释放率的差距逐渐减小,均表现出了良好的释放效果,如图3所示。
本申请中该乳液凝胶运载体系以及制备方法所适用的应用领域为食品加工领域,所需原料均为可食用原料。除此之外,还可以应用于其他领域。

Claims (5)
1.一种油水双相负载乳液凝胶运载体系的制备方法,其特征在于,包括如下步骤:称取一定量的SPI-SBP乳液,加入质量百分比为0.3%-1.8%的GDL、5-30 U/g蛋白的MTG、5-50mmol/L氯化钙或5-50 mmol/L氯化镁中的任一种,充分搅拌后,再加入15-25U/g底物的漆酶,充分搅拌后,放入培养箱中保温静置一段时间后,得到SPI-SBP双诱导乳液凝胶运载体系。
2.根据权利要求1所述的制备方法,其特征在于,所述SPI-SBP乳液的制备方法包括如下步骤:
A1,将SPI粉末分散于去离子水中,充分搅拌后,放入冰箱内冷藏一段时间至SPI充分水化,得到SPI水溶液;
A2,将SBP粉末加入到SPI水溶液中,充分搅拌至溶解完全,得到SPI-SBP 水溶液,放入冰箱内冷藏一段时间后,在80-90℃下预热10-20min,随后立即放置于冰水浴中迅速冷却至室温,向其中加入水溶性功能因子,充分搅拌,得到乳液的水相;
A3,将脂溶性功能因子溶于MCT中,搅拌至充分溶解,得到乳液的油相;
A4,向水相中添加油相,使用高速剪切机剪切后制成粗乳液,再使用高压均质机均质后,得到SPI-SBP乳液。
3.根据权利要求2所述的制备方法,其特征在于,所述水溶性功能因子为核黄素粉末,脂溶性功能因子为β-胡萝卜素。
4.一种由权利要求1-3任一项所述的制备方法制备出的油水双相负载乳液凝胶运载体系。
5.一种权利要求1-3任一项所述的制备方法在食品加工领域中的应用。
