需氧益生菌0.25千克,兼性菌0.15千克,厌氧菌0.55千克,光合细菌0.04千克,放线菌0.01千克。
优选地,将所述需氧益生菌、所述兼性菌、所述厌氧菌、所述光合细菌和所述放线菌进行混合,并混合均匀形成混合菌,所述混合菌放置在培养基中培养。培养基选用淀粉硝酸盐培养基,又称高氏一号培养基,其主要成分为:可溶性淀粉2.0g硝酸钾0.1g磷酸氢二钾0.05g氯化钠0.05g硫酸镁0.05g硫酸亚铁0.001g琼脂2g水100ml。先把可溶性淀粉放在培养器中,用5毫升水调成糊状后,倒入95毫升水,搅匀后加入其他药品,使它溶解。在烧杯外做好记号,加热到煮沸时加入琼脂,不停搅拌,待琼脂完全溶解后,补足失水。调整pH值到7.2~7.4,分装后灭菌,备用。培养基的比例可以根据实际培养混合菌的用量进行同比例增加。

为了确保发酵后菌落总数大于1×1010cfu/g,优选地,所述混合菌在所述培养基中发酵4至6天,且所述混合菌的发酵温度为34℃至37℃。经过相关设备检测得到,当发酵时间为4至6天、发酵温度控制在34℃至37℃时,可以使菌落总数大于1×1010cfu/g,具体的时间和温度,需要根据培养场地的外部环境而定,具体菌落总数依据测试结果而定。
