Abstract
本发明公开了一种近红外二区荧光探针和制备及其在骨成像中的应用,按比例,配制谷胱甘肽水溶液,作为溶液一;配制HAuCl4水溶液,作为溶液二;将上述溶液一、溶液二和超纯水按照比例混合,然后剧烈搅拌;加入NaOH溶液调节反应溶液的pH至11~12;向反应溶液中通入饱和CO,持续2~5min;反应溶液在室温下以500~600rpm搅拌24h;利用旋转蒸发仪收集沉淀,甲醇多次洗涤;最后沉淀真空干燥,得到GSH配体的Au25团簇。本发明的红外二区荧光探针Au25团簇展现出了从近红外一区到近红外二区波段优异的光学性能和天然的骨靶向能力,并且具有较高的生物安全性,在未来的骨成像中具有潜在的应用价值。
Description
技术领域
本发明属于生物医药领域,涉及一种优异生物安全性能的近红外二区荧光探针和制备方法,并将其应用于骨成像。
背景技术
医学成像技术能够提供生物体的可视化信息,并且可以实时监控生物体的生理和病理过程,在临床诊断、手术导航及预后评估中起到关键作用。荧光成像技术相较于传统的成像技术(计算机X射线断层扫描,核磁成像,正电子发射断层扫描及超声检查等)具有较低成本、较高的时空分辨率、优异的灵敏度和选择性、可实时分析和定位生物分子等优势。随着光波长越长,光的穿透深度变深,近红外二区(1000-1700nm)荧光成像相较于传统的可见光区(400-700nm)及近红外一区(700-900nm)成像具有较强的穿透深度及较高的信噪比,这些特点决定了近红外二区荧光成像在生物成像中的应用优势。
随着近红外二区成像技术的快速发展,研究者们相继研发了各种类型的发光材料用于生物成像,包括无机、有机及复合材料。首次应用于近红外二区成像的单壁碳纳米管有巨大的应用潜力,但仍局限于较低的荧光量子产率和较差的生物相容性;量子点由于可调节的结构而具有出色的光学性能,但同样面临着生物安全性的挑战;稀土掺杂纳米颗粒由于高稳定性和波长可灵活调谐性而受到广泛研究,但生物相容性差及长期的毒理性问题限制了其进一步应用。为了进一步改善材料的性能,研究者们开发出了一系列的水溶性有机红外二区荧光团。其研究最广泛的结构设计原则主要包括基于供体-受体-供体的分子结构及聚次甲基染料结构调控,但荧光团量子产率低,光稳定性差及设计结构单一限制了其进一步应用到活体成像中;半导体纳米粒子因高度离域的π共轭主链及可调制的侧链结构而具有可调节的光物理性能,但分子间强π-π相互作用引发的荧光淬灭限制了其实际应用;基于聚集诱导发光原理设计的AIEgens材料解决了材料荧光淬灭的问题,但是较低的量子产率和较差的生物安全性限制了其进一步发展。保障较好的生物安全性和较高的发光量子产率仍然是研发近红外二区荧光材料首先要考虑的两个关键问题。
发明内容
为了解决现有技术中的问题,本发明提供一种近红外二区荧光探针和制备及其在骨成像中的应用,解决现有技术中红外二区荧光材料生物安全性和发光量子产率较低的问题。
为了达到上述发明目的,本发明采用的技术方案如下:
一种近红外二区荧光探针,由以下方法制得:
(1)按比例,配制浓度为50mg/mL的谷胱甘肽(GSH)水溶液,作为溶液一;
(2)配制浓度范围为13~14mg/mL的HAuCl4水溶液,作为溶液二;
(3)将上述溶液一、溶液二和超纯水按照6:5:189的比例混合,然后剧烈搅拌;
(4)加入NaOH溶液调节反应溶液的pH至11~12;
(5)向反应溶液中通入饱和CO,持续2~5min;
(6)反应溶液在室温下以500~600rpm搅拌24h;
(7)反应溶液利用旋转蒸发仪收集沉淀,沉淀用甲醇多次洗涤;
(8)最后沉淀真空干燥,得到GSH配体的Au25团簇。
一种近红外二区荧光探针的制备方法,包括以下步骤:

本发明的有益效果为:本发明的红外二区荧光探针Au25团簇展现出了从近红外一区到近红外二区波段优异的光学性能和天然的骨靶向能力,显著靶向骨的主要成分羟基磷灰石(HAP),并且具有良好的可肾脏清除能力,能快速排出小鼠体外,具有较高的生物安全性,在未来的骨成像中具有潜在的应用价值。

具体实施方式
下面结合附图,对本发明的技术方案作进一步的描述。
近红外二区成像系统的搭建:
近红外二区成像系统搭建在一个光学平台上。首先,利用光纤耦合的808nm二极管激光器提供激发光,由4.5mm的准直镜准直光线,光线通过850nm和1000nm的短通滤波片过滤;然后,采集图像利用的是液氮冷却的二维InGaAs阵列(美国普林斯顿,320×256像素);最后,收集光路利用的是1000nm和1300nm的长通滤波片,收集大于1300nm的发光。活体成像过程中,成像平台处的面内激光功率密度为140mW/cm2,曝光时间为300ms。
下面结合具体实施例对本发明作进一步的说明。
实施例1
用于骨成像的近红外二区荧光探针的制备方法,包括如下步骤:
按比例,配置50mg/mL的GSH水溶液作为溶液一;配制出浓度范围为13~14mg/mL的HAuCl4水溶液,如将51mg分子量为339.79的HAuCl4加入3.75mL的水中,得到溶液二;按照6:5:189的比例,将4.5mL的上述溶液一和3.75mL溶液二一起加入到141.75mL的超纯水中,剧烈搅拌,然后加入NaOH调节反应溶液的pH至11~12;持续2~5min向反应溶液中通入饱和CO;在室温下以500~600rpm搅拌24h;利用旋转蒸发仪收集沉淀,沉淀用甲醇多次洗涤,然后真空干燥,得到GSH配体的Au25团簇。
实验例1
一种安全的近红外二区荧光探针的结构表征及光学性质
Au25SG18团簇(实施例1制备)的晶体结构由25个Au原子和18个谷胱甘肽配体共同组成,如图1所示。
Au25SG18团簇(实施例1制备)的分子量通过ESI-MS测定,得到三个不同价态的分子离子峰,如图2所示,三者皆为Au25SG18,表明团簇结构的准确性,且具有较高的纯度。
Au25SG18团簇(实施例1制备)溶于超纯水中,采用655nm的激发波长,测定其在近红外区的发射光谱,如图3所示。Au25SG18团簇在近红外区域具有两个特征发射峰,分别位于840nm和1050nm。
实验证明,实施例1制备的近红外二区荧光探针具有原子精确的结构以及较高的纯度,并且在近红外区域具有双重PL发射峰。
实验例2
近红外二区荧光探针Au25团簇的骨靶向性能研究
将Au25SG18(实施例1制备)溶于超纯水中,配置0.5mg/mL的Au25SG18水溶液,然后取200μL,分别加入20mg、30mg、40mg的HAP中进行搅拌,搅拌时间分别为0h、1.6h、5h、7h,再用荧光光谱仪分别测定搅拌后上清液的荧光发射光谱,激发波长为400nm。如图4所示,Au25SG18在可见光波长范围内的荧光发射强度会随搅拌时间的增长而减弱,且随着HAP含量的加大,搅拌时间增长,Au25SG18被HAP吸附的百分比也增加,骨吸附率最高接近100%。
实验证明,实施例1制备的近红外二区荧光探针Au25SG18具有优异的骨靶向能力。
实验例3
近红外二区荧光探针Au25团簇的骨成像实验
选取21-23g的C57BL/6雄性小鼠,将Au25团簇PBS溶液(200μL,5.3mg/mL)通过尾静脉注射到雄性小鼠体内,在异氟烷气体麻醉状态下分别在不同时间点1h、4h、24h、2周对小鼠背部进行近红外二区骨成像,如图5所示。注射Au25团簇1h后,荧光信号主要集中在小鼠的肾脏和脊柱部位,说明Au25团簇具有骨靶向能力,并且主要通过小鼠的肾脏进行排泄。4h后,肾脏的荧光信号几乎消失,脊柱的荧光信号也明显下降,表明具有较好的可肾脏清除能力。24h后,脊柱荧光虽有降低,但依旧清晰可见。2周后,小鼠体内已经检测不到任何的荧光信号,表明此时Au25团簇已经完全排出小鼠体内,不会在小鼠体内积累,具有良好的生物安全性。
实验证明,实施例1制备的近红外二区荧光探针Au25团簇具有良好的靶向骨的能力,可以进行荧光骨成像,并且具有优异的可肾脏清除能力,不会对机体造成长期的安全隐患。
实验例4
近红外二区荧光探针Au25团簇的药代动力学研究
将雄性小鼠(C57BL/6,21-23g)随机分为3组(每组3只):
血药浓度实验:
随机选取雄性小鼠(C57BL/6,21-23g)1组(每组3只)
将Au25团簇PBS溶液(200μL,5.3mg/mL)通过尾静脉注射到小鼠体内,再在注射后的不同时间点通过小鼠眼眶静脉窦取血,结合电感耦合等离子体质谱(ICP-MS)测定血液中Au元素的浓度,并绘制出血药浓度曲线,如图6a所示。
根据实验计算结果,最终得到Au25团簇的血液循环半衰期约为52min。
生物分布实验:
将雄性小鼠(C57BL/6,21-23g)随机分为2组(每组3只):
1天组,
7天组。
所有小鼠均通过尾静脉注射Au25团簇PBS溶液(200μL,5.3mg/mL),并分别在注射1天和7天后解剖小鼠并取出主要器官,将取出的器官保存在10%组织固定液中,然后通过ICP-MS分析各个脏器中Au元素的浓度,从而分析Au25团簇在小鼠体内各个脏器的生物分布,如图6b所示。
Au25团簇主要分布于肾脏,其次是脾、肝、心,说明肾脏是Au25团簇的主要排泄途径。但在7天后,各个脏器中团簇的含量都趋近于零,说明Au25团簇几乎全部排出小鼠体外,不会在体内大量积累。
排泄实验:
随机选取雄性小鼠(C57BL/6,21-23g)1组(每组3只)
所有小鼠均通过尾静脉注射Au25团簇PBS溶液(200μL,5.3mg/mL),收集注射48h内的小鼠排泄的尿液和粪便,然后利用ICP-MS测定尿液和粪便中Au元素的浓度,最后得到Au25团簇的排泄曲线,如图6c-d所示。
Au25SG18团簇在小鼠体内的排泄主要通过尿液,并且主要集中在注射后的前12h内排泄,排泄率可达87%以上。
实验证明,实施例1制备的近红外二区荧光探针Au25团簇具有较长的血药浓度半衰期,且可以通过肾脏快速清除,即使我们使用高达100mg/kg的给药剂量,仍不会对机体造成长期安全隐患。
实验例5近红外二区荧光探针Au25团簇的的毒理学研究
将雄性小鼠(C57BL/6,21-23g)随机分为2组(每组8只):
对照组,
Au25团簇实验组(实施例1制备)。
将Au25团簇(200μL,5.3mg/mL)静脉注射到小鼠体内,在注射后第1、7和30天,通过小鼠眼眶静脉窦采集血液样本进行血液学分析。将血液样本分为两份,一份加入EDTA稀释,并使用血细胞计数仪进行血常规检测,另一份血液样本经过离心分离血清,用于血生化分析。
血常规检测和血生化检测是毒性研究的重要方法,为研究Au25团簇对机体的毒性,我们进一步检测了注射Au25团簇后小鼠的毒理学变化,其血常规和血生化各项指标检测结果如图7所示。结果显示,实验组小鼠的各项指标均与对照组小鼠相似,且在30天内都在正常水平范围内,均未显示异常,表明Au25团簇不会使机体产生炎症以及相关脏器的功能异常等不良反应。因此,本发明的实验研究表明,Au25团簇不会对小鼠造成明显的急性以及长期的毒性,具有较好的生物安全性。
Claims (3)
1.一种近红外二区荧光探针,其特征在于,由以下方法制得:
(1)按比例,配制浓度为50mg/mL的谷胱甘肽水溶液,作为溶液一;
(2)配制浓度范围为13~14mg/mL的HAuCl4水溶液,作为溶液二;
(3)将上述溶液一、溶液二和超纯水按照6:5:189的比例混合,然后剧烈搅拌;
(4)加入NaOH溶液调节反应溶液的pH至11~12;
(5)向反应溶液中通入饱和CO,持续2~5min;
(6)反应溶液在室温下以500~600rpm搅拌24h;
(7)反应溶液利用旋转蒸发仪收集沉淀,沉淀用甲醇多次洗涤;
(8)最后沉淀真空干燥,得到GSH配体的Au25团簇。
2.一种近红外二区荧光探针的制备方法,其特征在于,包括以下步骤:
(1)按比例,配制浓度为50mg/mL的谷胱甘肽水溶液,作为溶液一;
(2)配制浓度范围为13~14mg/mL的HAuCl4水溶液,作为溶液二;
(3)将上述溶液一、溶液二和超纯水按照6:5:189的比例混合,然后剧烈搅拌;
(4)加入NaOH溶液调节反应溶液的pH至11~12;
(5)向反应溶液中通入饱和CO,持续2~5min;
(6)反应溶液在室温下以500~600rpm搅拌24h;
(7)反应溶液利用旋转蒸发仪收集沉淀,沉淀用甲醇多次洗涤;
(8)最后沉淀真空干燥,得到GSH配体的Au25团簇。
