Abstract
本发明为一种食管癌的分子标志物及其应用,公开了一种食管鳞癌的分子标志物,为CDCA7基因和/或CDCA7基因的表达产物,CDCA7基因在食管鳞癌组织中显著高表达,能够作为食管鳞癌的分子标志物用于食管鳞癌诊断;通过检测受试者组织中CDCA7基因的表达情况,为判断其是否患有食管鳞癌、或者是否存在患有食管鳞癌的风险提供新的思路,从而指导临床医师提供相应的预防方案或者治疗方案;同时,通过采用本发明提供的CDCA7基因诊断具有更及时、更特异、更灵敏的效果,能够实现食管鳞癌的早期诊断,从而提高食管鳞癌患者的预后生存时间、降低食管鳞癌患者的死亡率。

技术领域
本发明涉及肿瘤分子生物学领域,具体地涉及CDCA7基因及其表达产物在食管鳞癌的 诊断以及治疗中的用途。
背景技术
众所周知,恶性肿瘤的一个普遍特征是细胞周期的紊乱,由于抑癌基因的失活或者原癌 基因的扩增和激活,导致癌细胞失去了很多抑制因素,这些大多数会引起细胞周期蛋白及其 激酶的表达升高,进而表现出一种不受控制的细胞周期进程。
食管癌(EC,Eophageal carcinoma)是世界范围内常见的恶性肿瘤之一,我国是食管癌 发病率及死亡率最高的国家,发病率及死亡率分别居各类恶性肿瘤的第三位及第四位。在我 国,食管癌的发病具有较强的地区特异性,主要集中在几个高发区,其中河南、山西、河北 交界的太行山区发病率最高。我国食管鳞癌的主要组织类型为鳞状细胞癌,大多数患者确诊 时已处于中晚期,治疗仍以手术结合放化疗为主,患者尤其中晚期患者治疗效果不尽人意, 预后差5年生存率仅10%左右。
与其它类型肿瘤相比,食管癌早期症状不明显,缺乏特异性,因此大多数患者确诊时已 处于中晚期,对于早期食管癌的诊断手段主要包括食管拉网脱落细胞学检查、钡餐造影、食 管镜、内镜超声等。食管拉网脱落细胞学检查虽然操作简单成本低,但是检查的敏感度偏低, 受检查较为痛苦,1980年后,随着纤维内镜、电子内镜的推广和普及,食管拉网脱落细胞学 检查的应用逐渐减少。消化道钡餐造影仅在病变发生解剖形态学明显改变时才能显示出来, 而且造影检查某局部病变,需要多方位投射,仅在某一合适方位显现,故早期食管癌的X线 征象易发生遗漏,使造影检查的准确率降低。纤维内镜和内镜超声检查虽然可以直接观察到 肿瘤组织,更好的显示病变,诊断率高达80%,但内镜检查容易使患者产生异物感、反胃等 不良反应,而且检查时间较长,患者不易接受。癌症的肿瘤标志物检测由于其无创性、价格 的合理性,以及肿瘤标志物检测的高敏感度和高特异性,在癌症的早期诊断和预后评估等方 面具有重要的临床应用价值。然而食管鳞癌(Esophageal squamouscell carcinoma,ESCC)的 早期诊断标志物及靶向治疗的靶点甚为局限,缺乏可供参考的国际标准。因此,深入探索食 管癌的发生发展机理,对于临床发现早期诊断的分子标志,制定有效的临床干预措施,提高 食管癌患者的生存率、改善患者生存质量显得至关重要。
发明内容:
因此,本发明要解决的技术问题在于提供一种食管癌早期诊断相关的新的分子标志物。
基因表达谱(gene expression profile),是指构建处于某一特定状态下的细胞或组织的非 偏性cDNA文库,通过大规模的cDNA测序,收集cDNA序列片段、定性、定量分析其mRNA群体组成,从而描绘该特定细胞或组织在特定状态下的基因表达种类和丰度信息, 这样编制成的数据表就称为基因表达谱。
肿瘤在组织形态学上被划分为多种不同的类型和亚型,而肿瘤亚型的准确诊断对肿瘤的 临床治疗具有重要意义,然而肿瘤的分型在临床上一直处于难以处理的状态,许多形态学上 相似的肿瘤,却需要不同的治疗方法,从分子生物学的角度上看,肿瘤是由于某些染色体上 DNA损伤致使细胞内基因异常表达,导致细胞生长失控、缺乏分化和异常增生的一类复杂 基因疾病。
肿瘤的发展机制具有复杂性,研究肿瘤基因表达谱、选取基因信息是从信息学角度出发 以寻找肿瘤相关基因、发现肿瘤基因表达特征的直接手段。当前的肿瘤分类技术高度依赖病 理学工作者对肿瘤组织的主观判断,而基于微阵列技术,即使一些组织没有显著变化,利用 基因表达谱也能够对之做出早期诊断;基于基因表达谱的肿瘤分型和分类研究为理解肿瘤的 发生的机制,以及肿瘤的临床治疗提供了重要依据;研究人员可以根据基因表达谱的变化来 区分形态学上相似的肿瘤,而肿瘤类型的精确诊断将有助于制定配套的最佳治疗方案。
本发明通过对食管鳞癌患者的癌样本和配对癌旁样本进行转录组测序分析后,发现大多 数患者的肿瘤组织相对于其癌旁配对组织中CDCA7基因的表达水平增高。因此,考虑细胞 中CDCA7的表达量升高,可以促进食管鳞癌的发生和发展。同时,在食管鳞癌细胞系里面 敲除和过表达CDCA7基因后,细胞的表型发生变化,在食管鳞癌细胞系中敲低CDCA7基因后,可以抑制食管鳞癌细胞的增殖、克隆形成、侵袭和迁移,延缓细胞周期的进程;反之,在食管鳞癌细胞系中过表达CDCA7基因后,可以促进食管鳞癌细胞的增殖、克隆形成、侵 袭和迁移,加速细胞周期进程。提示CDCA7可能通过细胞周期影响食管鳞癌细胞的增殖速 度,进而促进食管鳞癌的发生发展。
基于此,本发明提供如下技术方案:
第一方面,本发明提供了一种食管癌的分子标志物,所述分子标志物为CDCA7基因和 /或CDCA7基因的表达产物。
可选地,上述的食管癌的分子标志物,所述CDCA7基因的表达产物包括CDCA7 mRNA和/或CDCA7蛋白。
进一步地,所述食管癌为食管鳞癌。
第二方面,本发明提供了第一方面所述食管癌的分子标志物在如下a-c至少一种中的用 途:
a.作为食管癌的诊断标志物,或者制备用于食管癌诊断的产品;
b.制备用于食管癌疗效监测的产品;
c.制备治疗食管癌的药物。
可选地,a-b中所述产品可以是指检测CDCA7基因mRNA水平的产品,检测方法包 括RT-PCR、实时定量PCR、高通量芯片或测序等;或检测CDCA7基因表达的试剂,也可 以是包含所述试剂的试剂盒、芯片,或者是使用所述试剂的高通量测序平台。
第三方面,本发明提供了一种用于食管癌诊断的引物组,所述引物组包括基于上述的食 管癌分子标志物设计的引物。
可选地,所述引物组包括如下引物:
CDCA7 Forward Primer,其核苷酸序列如SEQ ID NO.1所示;
CDCA7 Reverse Primer,其核苷酸序列如SEQ ID NO.2所示。
第四方面,本发明提供了一种CDCA7基因和/或其表达产物的抑制剂在制备治疗食管 癌的药物中的应用。
可选地,所述抑制剂包括抑制CDCA7基因表达的试剂、和/或抑制CDCA7基因表达产物的试剂。
所述抑制CDCA7基因表达的试剂包括抑制基因转录的试剂、抑制基因翻译的试剂;
所述抑制CDCA7基因表达产物的试剂包括抑制CDCA7基因mRNA的试剂,例如 CDCA7基因mRNA的双链核糖核酸;
所述抑制CDCA7蛋白的试剂,包括抑制CDCA7蛋白稳定性、蛋白活性的试剂、蛋白功能的试剂;
所述抑制CDCA7蛋白功能的试剂包括CDCA7抗原蛋白的肿瘤疫苗、抑制CDCA7蛋白功能的抗体,具体地,所述抗体可以是多克隆抗体,或是单克隆抗体。
本发明的有益效果:
1.本申请从转录组水平寻找食管鳞癌肿瘤组织与正常组织之间的基因表达模式的差异,通 过对差异基因表达的研究来推断基因之间的相互作用关系,细胞癌变过程中基因“开启” 或“关闭”的机制;揭示基因与食管癌的发生和发展的内在联系,首次发现了CDCA7 基因表达与食管鳞癌相关,为寻找癌变过程中可能涉及的食管癌癌变机理、发现用于预 防和早期检测的候选分子标志物、筛选抑制或减缓食管鳞癌细胞增殖、侵袭转移药物以 及基因治疗的可能靶点提供新的思路。
2.根据本发明的结果,通过检测受试者组织中CDCA7基因的表达情况,可以判断其是否 患有食管鳞癌、或者是否存在患有食管鳞癌的风险,从而指导临床医师提供相应的预防 方案或者治疗方案。
3.相比传统的检测手段,通过采用本发明提供的CDCA7基因诊断具有更及时、更特异、 更灵敏的效果,能够实现食管鳞癌的早期诊断,从而提高食管鳞癌患者的预后生存时间、 降低食管鳞癌患者的死亡率。

具体实施方式:
下面结合附图和实施例对本发明作进一步详细的说明。以下实施例仅用于说明本发明而 不用于限制本发明的范围。实施例中未注明具体条件的实验方法以及实验试剂的配制条件, 按照常规方法和条件进行,例如Sambrook等人,分子克隆:实验室手册(NewYork: ColdSpring HarborLaboratory Press,1989)中所述的条件,或按照制造厂商的说明书进行。
实施例1 CDCA7基因的差异表达
1、实验材料
155例食管鳞癌样本及其癌旁配对样本来源于山西医科大学附属肿瘤医院的胸外手术切 除的标本,手术切除的食管肿瘤组织离体后立即取材,取材时均佩戴一次性无菌手套,应用 高压灭菌的手术刀片切取。所有患者术前均病理证实,术前均未行放疗、化疗等治疗,且无 其他部位的肿瘤等。所有组织均在手术半小时内液氮保存后,移入-80度冰箱冻存。
2、转录组测序(RNA-seq)检测CDCA7基因的差异表达
转录组是指特定组织或细胞在某个时间或某个状态下转录出来的所有RNA的总和,主 要包括mRNA和非编码RNA。转录组测序是基于Illumina测序平台,研究特定组织或细胞 在某个时期转录出来的所有mRNA,是基因功能与结构研究的基础,对理解生物体的发育和疾病的发生具有重要作用。随着基因测序技术的发展以及测序成本的降低,RNA-seq凭借高通量、高灵敏度、应用范围广等优势,已成为转录组研究的主要方法。本申请依托上海明码科技有限公司,对155例食管鳞癌和癌旁正常组织进行RNA-seq分析,筛选具有显著表 达差异的基因,CDCA7是其中之一。
2.1 RNA提取与检测
采用标准提取方法从组织或细胞中提取RNA,随后对RNA样品进行严格质控,质控方 式主要是通过Agilent 2100bioanalyzer:精确检测RNA完整性。
2.2文库构建与质检
mRNA的获取主要有两种方式:一是利用真核生物大部分mRNA都带有polyA尾的结构特征,通过Oligo(dT)磁珠富集带有polyA尾的mRNA。二是从总RNA中去除核糖体RNA, 从而得到mRNA。随后在NEB Fragmentation Buffer中用二价阳离子将得到的mRNA随机打 断,按照NEB普通建库方式或链特异性建库方式进行建库。
NEB普通建库:以片段化的mRNA为模版,随机寡核苷酸为引物,在M-MuLV逆转录 酶体系中合成cDNA第一条链,随后用RNaseH降解RNA链,并在DNA polymerase I体系 下,以dNTPs为原料合成cDNA第二条链。纯化后的双链cDNA经过末端修复、加A尾并 连接测序接头,用AMPure XP beads筛选250-300bp左右的cDNA,进行PCR扩增并再次 使用AMPure XPbeads纯化PCR产物,最终获得文库。建库用试剂盒为UltraTM RNA Library PrepKit for Illumina。
2.3上机测序
库检合格后,把不同文库按照有效浓度及目标下机数据量的需求pooling后进行Illumina 测序。测序的基本原理是边合成边测序(Sequencing by Synthesis)。在测序的flow cell中加 入四种荧光标记的dNTP、DNA聚合酶以及接头引物进行扩增,在每一个测序簇延伸互补链 时,每加入一个被荧光标记的dNTP就能释放出相对应的荧光,测序仪通过捕获荧光信号, 并通过计算机软件将光信号转化为测序峰,从而获得待测片段的序列信息。
2.4信息分析
RNA-seq的核心是基因表达差异的显著性分析,使用统计学方法,比较两个条件或多个 条件下的基因表达差异,从中找出与条件相关的特异性基因。
3、结果
转录组测序结果显示,无论是采用非配对检验方法,还是配对检验方法比较,CDCA7 基因在食管鳞癌组织中的表达水平,与癌旁配对组织相比显著升高(图1)。
实施例2促进和抑制食管鳞癌细胞系中CDCA7基因的表达
1、siRNA设计合成
针对CDCA7基因的siRNA序列:
CDCA7-siRNA1序列:5’-GCCCTCAGAGAATTCTGTGACTGAT-3’;
CDCA7-siRNA2序列:5’-CATCCGTGACCCTTCCGCATATAAT-3’;
CDCA7-siRNA3序列:5’-GGTTGGTGGGCAATGTAATACTTAA-3’;
NC-siRNA序列:5’-TTCTCCGAACGTGTCACGTAA-3’;
以上siRNA序列与阴性对照siRNA序列(siRNA-NC)由汉恒生物科技(上海)有限公司 提供。所选用的干扰载体为pHBLV-U6-Scramble-ZsGreen-Puro,重组载体的构建步骤按照常 规分子克隆方法进行。
2、CDCA7外源重组质粒的构建
设计针对转录本ID为NM_145810.2的CDCA7基因的CDS区的引物,所选用的过表 达载体为pHBLV-CMV-MCS-3FLAG-EF1-ZsGreen-T2A-PURO,重组载体的构建步骤按照常 规分子克隆方法进行。
3、慢病毒包装
复苏后的293T细胞传两代后,当密度达到80-90%左右时1:2传代至10cm皿中,放置 于37□、5%CO2培养箱内过夜培养;待细胞达90%融合时,换新鲜培养基7ml。
准备两个15ml离心管A和B,将恢复至室温的Opti-MEM,分别加入A、B管中,每 管1.5ml;A管中加入P3000:48ul,包装载体pMD2.G,psPAX2分别6μg,目的质粒12μg, 轻轻混匀;向B管中加入Lipofectamine 3000:42μL;A、B管室温孵育5min;A管加入B 管,轻轻混匀,室温室温孵育15min;将A+B孵育后形成的脂质体-DNA复合物,加入到 换过液的细胞中(沿培养皿缓慢轻柔加入,防止吹落细胞),37□、5%CO2培养箱内继续 培养。
转染48h后收取病毒(培养上清),重新补上新鲜的DMEM完全培养基,细胞放回37□、 5%CO2培养箱内继续培养,收取的细胞上清,置于4□保存。转染72h几批再次收取病毒, 并将两次收取的上清混匀,用0.45μm的滤膜过滤除去细胞和细胞碎片等。
病毒浓缩,将上一步过滤后的上清加入病毒浓缩管中(小于15ml),3200g,50min,4□离心,离心完成后,病毒浓缩管中部可见深红色的病毒浓缩液,约200μL,转移至1.5mlEP管中,封口膜封好,置于-80□保存。
4、食管鳞癌细胞的培养与转染
4.1细胞培养
食管鳞癌细胞系KYSE150、KYSE450和KYSE180接种于含有10%FBS的DMEM培 养液中,放置于37□、5%CO2培养箱内培养,待细胞达80%融合时,0.25%胰蛋白酶消 化传代。
4.2细胞转染
将食管鳞癌细胞按1×104/孔接种到24孔细胞培养板中,在37□、5%CO2培养箱中细 胞培养24h,加入含有终浓度为5μg/mL Polybrene的完全培养基,根据CDCA7敲除和过表达慢病毒的感染复数(Multiplicity of infection,MOI)值,计算所需要的慢病毒原液的体积。 MOI值分别设定了30、50和200三个梯度,慢病毒原液体积=(转染时细胞数目×MOI)/慢 病毒梯度;加入病毒后,置于37□、5%CO2的细胞培养箱中培养;
12~24h后,移去转染细胞的病毒液体,加入0.2mL完全培养基,继续在37□、5%CO2的细胞培养箱中培养;根据细胞状态及融合度,将细胞扩大培养,同时加含有适当浓 度的嘌呤霉素的完全培养基,继续培养;利用qPCR和Western Blot检测转入基因的过表 达效率。
5、利用QPCR和Western Bolt实验检测CDCA7基因的过表达和干扰效率
5.1 QPCR实验检测CDCA7基因的mRNA表达水平
5.1.1 Trizol法提取总RNA
收集新鲜的细胞沉淀于无菌的RNA-free EP管中,加入适量Trizol,反复吹打至细胞沉 淀消失,室温静置5min;向混合物中加入1/5体积氯仿,上下颠倒混匀4~6次,室温静置 5min,12000rpm,4□离心15min;离心后,混合物分为三层,依次为:无色液体(上层)、 白色蛋白层(中间)、粉红色的有机相(下层),将上清液小心转移至新的RNA-free EP管 中,注意避免吸入白色的中间层;随后加入等体积的异丙醇,上下颠倒充分混匀,在-20□静 置0.5-1h后,4□,12000rpm,离心10min;弃掉上清液,1mL用DEPC水配置的75%乙 醇洗沉淀,12000rpm,4□离心5min,室温自然干燥2~3min,加入30~50μL RNase free water溶解沉淀;使用Nanodrop检测RNA的纯度和浓度。
5.1.2反转录
使用PrimeScriptTM RT Master Mix(Perfect Real Time)试剂盒进行反转录,将RNA反 转录为cDNA,反应体系为20μL。反应条件:37□15min,85□5s,4□。反应体系如下表:

5.1.3荧光定量PCR(qPCR)
使用TBPremix Ex TaqTM(Tli RNaseH Plus)试剂盒进行qPCR,按照以下表格将对应试剂加入EP管中,混匀后依次分装在按顺序摆放的8连管中,每个样品设置三 个复孔,反应体系为25μL。反应条件:预变性:95□10min;PCR反应:95□15s,60□1 min,共40个PCR循环;Melt Curve。所用引物:Forward Primer(SEQ ID NO:1):CTTGTCATCAATGCCGTCAG;Reverse Primer(SEQ ID NO:2): CAGTTGCAGATTCCTCGACA;反应体系如下表:

5.1.4统计学处理
以对照组中的管家基因GAPDH为内参,采用△△Ct法计算目的基因的相对表达量。所有实验进行三次独立重复试验,结果数据都是以平均值±标准差的方式来表示,采用SPSS13.0统计软件来进行统计分析的,两者之间的差异采用t检验,认为当P<0.05时,结 果有意义。
5.2 Western Bolt实验检测CDCA7基因的蛋白表达水平
5.2.1细胞总蛋白的提取:
收取新鲜的细胞沉淀于无菌的EP管中,加入适量的RIPA裂解液(含蛋白酶抑制剂), 并置于冰上裂解1h,10min震荡一次;裂解结束后,12000rpm,4□离心30min,然后将上清蛋白转移至新的无菌EP管中。
5.2.2 BCA法测蛋白浓度:
按照说明书,使用BCA试剂盒中的A液和B液,以50:1的比例配置所需的工作液; 蛋白标准液的稀释:用预冷的1×PBS缓冲液将浓度为2mg/mL的蛋白标准液稀释后浓度 为0.5μg/μL;按照说明书在96孔板的第一列加入稀释后的蛋白标准品,依次为0、1、2、 4、8、12、、16、20μL,随后每孔补1×PBS缓冲液至20μL;第二列重复第一列;在96孔 板的第三列,每孔加入1μL的待测蛋白样品,每孔补1×PBS缓冲液至20μL;每个样品三 个复孔;每孔加入200μL的工作液,37□震荡30min;使用自动酶标仪在570nm波长下 检测每孔的光密度值(Optical density,OD),根据标准蛋白的OD值及上样量绘制标准曲 线,待测样品的蛋白浓度即可根据标准曲线而得知。
5.2.3蛋白变性及制备蛋白样品:
按照每孔50μg的蛋白上样量,加入4×蛋白上样缓冲液,并用1×PBS缓冲液将所有样 品的补至相同体积,将混合物于100□煮沸10min,-80□保存备用。
5.2.4电泳(SDS-PAGE)、转膜、显影
将洗干净的玻璃板,晾干并安装好;按说明书配置适量10%的分离胶和5%的浓缩胶; 首先配置下层分离胶,于玻璃板夹缝中加入适量10%的分离胶,再加入1mL的异丙醇使分 离胶表面保持水平;待分离胶凝固后,倒掉异丙醇,用三蒸水清洗分离胶平面2~3次;用干 净的滤纸吸干残留在胶表面的三蒸水,加入适量5%的浓缩胶,插入齿梳;待浓缩胶凝固后, 拔掉齿梳,加入适量的电泳液,将制备好的蛋白样品按顺序依次上样,并在特定泳道加5μL 蛋白Marker,恒压80V电泳2~3h;准备合适大小的PVDF膜,将膜浸泡在甲醇中,并用转膜液平衡PVDF膜;同时用转膜液浸泡滤纸海绵,转膜夹子中按照负极-正极的顺序依次放置“海绵-滤纸-SDS凝胶-PVDF膜-滤纸-海绵”,注意每层之间避免有气泡存在;恒压100V、 4□转膜2h;转膜结束后,将PVDF用5%的脱脂牛奶在水平摇床上室温封闭1h;弃去封 闭液,在PVDF膜中加入封闭液稀释好的一抗,4□层析柜中过夜孵育;次日,TBST洗膜 3次,每次10min;加入适量的TBST稀释的荧光二抗,室温孵育2h;TBST洗膜3次, 每次10min;显影。
5.2.5统计学处理
将蛋白条带的灰度值使用Image J软件进行分析,以GAPDH为内参,将CDCA7蛋白条带的灰度值进行归一化处理。结果数据都是以平均值±标准差的方式来表示,采用SPSS13.0统计软件来进行统计分析的,两者之间的差异采用t检验,认为当P<0.05时,结 果有意义。
6、结果
利用慢病毒转染的方法,在KYSE180细胞系中转染CDCA7外源过表达病毒,KYSE150和KYSE450细胞系中转染CDCA7敲除病毒,利用QPCR和Western Bolt实验对CDCA7的 表达进行验证,结果显示在KYSE180细胞系中成功过表达了CDCA7基因,在KYSE150和 KYSE450细胞系中成功敲低了CDCA7基因(图2和图3)。
实施例3 CDCA7基因的表达水平对食管鳞癌细胞周期和细胞增殖能力影响的测定
将前面实施例中构建的稳定过表达CDCA7基因KYSE180细胞系分为两个实验组,组1:转染NC的KYSE180细胞组;组2:转染CDCA7-OE的细胞组;稳定敲除CDCA7基因 的KYSE150细胞系分为三组,组1:转染siRNA-NC的KYSE150细胞组;组2:转染CDCA7- siRNA1的细胞组;组3:转染CDCA7-siRNA2的细胞组;稳定敲除CDCA7基因的KYSE450 细胞系分为三组,组1:转染siRNA-NC的KYSE450细胞组;组2:转染CDCA7-siRNA1 的细胞组;组3:转染CDCA7-siRNA2的细胞组。
1、MTT实验检测食管鳞癌细胞增殖能力
以每孔5000个细胞接种于48孔板中,设置五个时间点,每个时间点5个复孔,分别对 培养24h、48h、72h、96h和120h活的细胞数量进行检测;检测前加入1/10培养基体积的浓度为5mg/ml的MTT溶液,37℃5%CO2继续培养4h;4h后慢慢吸掉细胞培养液,每 孔加入200μL DMSO,室温避光摇床孵育15min,当紫红色络合物完全溶解后,酶标仪检 测激发光490nm波长下的吸光度值,连续检测5天。连续收集4天检测的吸光度值,以吸 光度为纵坐标,时间点为横坐标绘制细胞生长曲线。
2、硬克隆实验检测食管鳞癌细胞增殖能力
将每组细胞按0.1×104个细胞/2mL/孔接种细胞接种于6孔板中,每组细胞设置3个重 复,置于37□、5%CO2的恒温培养箱中培养10天左右,一般情况下,总数大于50个细胞, 即可判定为一个克隆。在显微镜下观察为可以肉眼可见白色的小点,即可停止培养,吸走孔 内原有培养基,加入4%的多聚甲醛室温固定15min,PBS清洗去除残留的甲醛,加入结晶 紫溶液室温染色20min,PBS清洗至克隆被清晰染色,背景尽可能干净透明为止。
3、流式细胞试验检测食管鳞癌细胞的周期
将各组细胞收取沉淀,70%冰乙醇固定过夜。离心弃掉固定液,PBS清洗2次后将细胞过滤(300μL尼龙网膜),离心弃上清,按照试剂盒说明书加入适量RNA酶和碘化丙啶(Propidium Iodide,PI)染料。避光30min后上机检测,分析各组细胞中G1、S、G2/M期 的细胞含量。
4、结果
在食管鳞癌细胞系KYSE180中过表达CDCA7基因,MTT和硬克隆实验结果显示, 与对照组相比,过表达CDCA7基因能够显著促进食管鳞癌细胞的增殖(p<0.001);流式 细胞实验结果显示,与对照组相比,过表达CDCA7基因后,G0/G1期细胞减少,S和G2/M 细胞增多,显著促进食管鳞癌细胞周期的进程(p<0.001)(图4)。
相反的,在食管鳞癌细胞系KYSE150和KYSE450中敲低CDCA7基因,MTT和硬克 隆实验结果显示,与对照组相比,敲低CDCA7基因能够显著抑制食管鳞癌细胞的增殖(p <0.001);流式细胞实验结果显示,与对照组相比,敲低CDCA7基因后,细胞阻滞G0/G1 期,S和G2/M细胞减少,食管鳞癌细胞周期被抑制(图5)。
实施例4抑制CDCA7基因表达对裸鼠成瘤的影响
1、实验材料
实验所用的NU-Foxn1nu nude mice免疫缺陷小鼠购置于北京维通利华公司,一般为4-5 周龄雌性裸鼠。将所购小鼠分成两组:CDCA7-NC组和CDCA7-SH组,每组8只。
2、裸鼠皮下成瘤实验
胰酶消化对数生长期细胞,离心后生理盐水调整细胞密度至3×107个细胞/ml,分别用1 ml注射器吸取100ul(3×107个细胞)细胞悬液,注射至裸鼠腹部皮下,形成皮丘;SPF饲 养室饲养3-4周,连续观察裸鼠瘤体的形成情况,待完全成瘤后处死裸鼠,测量并记录裸鼠 皮下肿瘤形成的数量和体积。
3、结果
动物体内裸鼠荷瘤实验结果显示,与对照组相比,敲低CDCA7基因能够显著抑制食管 鳞癌细胞在体内的增殖(图6)。
实施例5 CDCA7基因表达水平对细胞周期相关蛋白的影响
1、实验材料
将前面实施例中构建的稳定过表达CDCA7基因KYSE180细胞系分为两个实验组,组1:转染NC的KYSE180细胞组;组2:转染CDCA7-OE的细胞组;稳定敲除CDCA7基因 的KYSE150细胞系分为三组,组1:转染siRNA-NC的KYSE150细胞组;组2:转染CDCA7- siRNA1的细胞组;组3:转染CDCA7-siRNA2的细胞组;稳定敲除CDCA7基因的KYSE450 细胞系分为三组,组1:转染siRNA-NC的KYSE450细胞组;组2:转染CDCA7-siRNA1 的细胞组;组3:转染CDCA7-siRNA2的细胞组。
2、QPCR实验检测细胞周期相关蛋白基因的mRNA表达水平
方法同实施例2中5.1部分。
3、Western Bolt实验检测细胞周期相关蛋白基因的蛋白表达水平
方法同实施例2中5.2部分。
4、结果
在食管鳞癌细胞系KYSE180中过表达CDCA7基因,QPCR和Western Bolt实验结果显示,细胞周期S期的相关蛋白CCNA2基因在无论是在mRNA水平还是蛋白水平,表达 升高,而细胞周期G2/M期相关蛋白CCNB1基因在无论是在mRNA水平还是蛋白水平,表 达降低(图8)。
相反的,在食管鳞癌细胞系KYSE150和KYSE450中敲低CDCA7基因,QPCR和 WesternBolt实验结果显示,细胞周期S期的相关蛋白CCNA2基因无论是在mRNA水平还 是蛋白水平,表达降低,而细胞周期G2/M期相关蛋白CCNB1基因无论是在mRNA水平还 是蛋白水平,表达升高(图9)。
细胞周期S期相关蛋白CCNA2和G2/M期相关蛋白CCNB1的表达水平与CDCA7的 表达水平具有相关性,而其他周期蛋白表达水平与CDCA7的表达水平无关,提示我们, CDCA7基因有可能通过影响细胞周期中的S期和G2/M期,来影响食管鳞癌细胞的周期进 程,进而促进食管鳞癌的发生发展。
实施例6CDCA7基因的表达水平对食管鳞癌细胞侵袭迁移能力影响的测定
1、实验材料
将前面实施例中构建的稳定过表达CDCA7基因KYSE180细胞系分为两个实验组,组1:转染NC的KYSE180细胞组;组2:转染CDCA7-OE的细胞组;稳定敲除CDCA7基因 的KYSE150细胞系分为三组,组1:转染siRNA-NC的KYSE150细胞组;组2:转染CDCA7- siRNA1的细胞组;组3:转染CDCA7-siRNA2的细胞组;稳定敲除CDCA7基因的KYSE450 细胞系分为三组,组1:转染siRNA-NC的KYSE450细胞组;组2:转染CDCA7-siRNA1 的细胞组;组3:转染CDCA7-siRNA2的细胞组。
2、划痕实验检测食管鳞癌细胞迁移能力
将各组稀释细胞接种于含10%FBS的完全培养基中,分别加入无菌6孔板各孔,并保 证细胞数为1×106,置于37℃、5%CO2培养;各组细胞均长至90%以上融合,用100μl 枪头在细胞中划出一道整齐的裸露带,1×PBS轻轻清洗被划起的细胞,加入基础培养基 继续培养;倒置显微镜观察找出各培养孔中宽度一致的地方,并做标记且拍照记录;37℃、 5%CO2继续培养,观察各组细胞同一位置不同时间点的迁移情况。
3、Trnaswell实验检测食管鳞癌细胞侵袭、迁移能力
Transwell侵袭实验:提前4h将BD Matrigel胶与基础培养基按1:6稀释,在每个小室中加入100μL;避免有气泡产生,置于37□、5%CO2的培养箱中备用;用0.25%胰蛋 白酶消化各组细胞,终止消化、离心后,用基础培养基重悬细胞沉淀并计数,在24孔板的 小室加入含10%FBS的完全培养基,上室加入200μL的细胞悬液,其中含有5×104个细 胞,避免产生气泡,置于37□、5%CO2的培养箱中培养;培养24~48h后,用棉签将小 室里面底部的细胞擦干净,放入4%的多聚甲醛中固定20min,在三蒸水中涮一下,然后 用结晶紫染色30min,显微镜下观察穿过小室底部膜的细胞,随机选取5个视野拍照并计 数。
Transwell迁移实验:小室内不需加入BD Matrigel胶,其他均与侵袭实验操作一致。
4、结果
在食管鳞癌细胞系KYSE180中过表达CDCA7基因,划痕和Transwell实验结果显示,与对照组相比,过表达CDCA7基因能够显著促进食管鳞癌细胞的侵袭迁移能力(p<0.001)(图8);相反的,在食管鳞癌细胞系KYSE150和KYSE450中敲低CDCA7基因,划痕和 Transwell实验结果显示,与对照组相比,敲低CDCA7基因能够显著抑制食管鳞癌细胞的侵 袭迁移能力(p<0.001)(图9)。
Claims (10)
1.一种食管癌的分子标志物,其特征在于,所述分子标志物为CDCA7基因和/或CDCA7基因的表达产物。
2.如权利要求1所述的食管癌的分子标志物,其特征在于,所述CDCA7基因的表达产物包括CDCA7 mRNA和/或CDCA7蛋白。
3.如权利要求1-2任一所述的分子标志物,其特征在于,所述食管癌为食管鳞癌。
4.权利要求1-3任一所述食管癌的分子标志物在如下a-c至少一种中的应用:
a.作为食管癌的诊断标志物,或者制备用于食管癌诊断的产品;
b.制备用于食管癌疗效监测的产品;
c.制备治疗食管癌的药物。
5.如权利要求4所述的应用,可选地,a-b中所述产品可以是指检测CDCA7基因mRNA水平的产品;或检测CDCA7基因表达的试剂。
6.一种用于食管癌诊断的引物组,其特征在于,所述引物组包括基于权利要求1-3任一所述的食管癌分子标志物设计的引物,所述引物组包括如下引物:
CDCA7 Forward Primer,其核苷酸序列如SEQ ID NO.1所示;
CDCA7 Reverse Primer,其核苷酸序列如SEQ ID NO.2所示。
7.一种CDCA7基因和/或其表达产物的抑制剂在制备治疗食管癌的药物中的应用。
8.如权利要求7所述的应用,其特征在于,所述抑制剂包括抑制CDCA7基因表达的试剂、和/或抑制CDCA7基因表达产物的试剂。
9.如权利要求8所述的应用,其特征在于,所述抑制CDCA7基因表达的试剂包括抑制基因转录的试剂、抑制基因翻译的试剂。
10.如权利要求9所述的应用,其特征在于,所述抑制CDCA7基因表达产物的试剂包括抑制CDCA7基因mRNA的试剂,例如CDCA7基因mRNA的双链核糖核酸;包括抑制CDCA7蛋白的试剂,例如抑制CDCA7蛋白稳定性、蛋白活性的试剂、蛋白功能的试剂;包括抑制CDCA7蛋白功能的试剂,例如CDCA7抗原蛋白的肿瘤疫苗、抑制CDCA7蛋白功能的抗体。
