本发明属于污染物降解领域,尤其涉及一种利用厌氧铁氨氧化降解全氟化合物的方法及应用。所述降解方法为:在厌氧铁氨氧化过程中进行全氟化合物的降解,具体方式为:在污染底泥中引入铁离子和铁异化还原菌进行厌氧铁氨氧化反应,并添加电子供体,进行全氟化合物的降解。该方法能够有效实现对PFOA和PFOS的去除,当加入氨氮时,这种去除PFOA和PFOS的效果最为明显。而且,随着时间的延长,最终能够实现全氟化合物的完全去除。该方法能从根本上消除PFCs对环境及生物危害,且过程简单、成本低、不会造成二次污染。

背景技术
本发明背景技术中,公开的信息仅仅旨在增加对本发明的总体背景的理解,而不必然被视为承认或以任何形式暗示该信息构成已经成为本领域一般技术人员所公知的现有技术。
由碳原子与氟原子组成的全氟化合物(PFCs)是新兴的持久性有机污染物,稳定性较强,同时具备较强的电负性,能经受光照和高温,具有疏水、疏油以及降低水表面的张力等独特性质,目前被广泛应用于涂料、化工、皮革、纺织、炊具制造、合成表面活性剂、包装材料等领域。作为环境中持久性强的有机污染物,全氟辛烷磺酰基化合物(PFOS)被加入了《斯德哥摩尔公约》,PFCs的去除已成为环境学领域研究的热点对象。
由于PFCs本身的持久性分布和应用的广泛性等特点,目前已经在各类环境介质和生物体中被检出,如土壤、水体、大气、海洋,甚至在人体血清中均有检出,且其具有的免疫毒性、生殖性毒性、生物蓄积性、肝脏毒性等,对全人类的生命健康和全球生态系统造成了较大的威胁。鉴于目前对理论的研究和实际使用处在发展中阶段,而在工农业的生产中大量地使用了会严重污染水体和土壤的PFCs表面活性剂,长此以往会造成严峻的环境问题,且目前并未出具较为严格的适合PFCs生产和使用的规范,因此对PFCs在水体和底泥中去除的研究迫在眉睫。
目前常见的PFCs去除方法包括吸附分离法、超声降解技术、电化学氧化法、光化学法、膜处理工艺等物理方法、化学技术及微生物处理技术。但是申请人发现物理方法操作简单,成本低、经济且有效性高,但不能从根本消除目标的污染物质,仅是把污染物从一相转移到另一相,并且还容易造成二次污染;化学方法能够改变PFCs的化学结构,通过将PFCs转化为F-、PFCAs等物质,有效的将其降解,但费用较高、操作条件苛刻,同时也会存在二次污染等问题;生物法成本较低,操作简单,且无二次污染,但降解过程较为复杂,且PFCS可生化性差,结构复杂,C-F键能高,难以被微生物直接利用。
因此,提供一种能够高效降解PFCs,在根本上消除PFCs对环境及生物危害,且过程简单、成本低、不会造成二次污染的方法将对于水体和土壤/底泥中PFCs的治理具有重要意义。
发明内容
为了弥补现有技术的不足,本发明的目的是提供一种利用厌氧铁氨氧化降解全氟化合物的方法及应用,本发明采用厌氧铁氨氧化协同全氟化合物的降解的方式,并通过向厌氧铁氨氧化体系中添加电子供体,能够有效提高PFCs的降解效率。
本发明第一方面提供一种利用厌氧铁氨氧化降解全氟化合物的方法,在厌氧铁氨氧化过程中进行全氟化合物的降解;
具体为:
在污染底泥中引入铁离子和铁异化还原菌进行厌氧铁氨氧化反应,并添加电子供体,进行全氟化合物的降解。
环境中厌氧铁氨氧化过程的研究多侧重于氨氮的去除,而对PFCs等微量有机污染物的去除还未有报道,本发明是首次发现,厌氧铁氨氧化过程耦合铁离子氧化还原的电子传递机制,可以强化对全氟化合物的去除,而添加电子供体来提高环境中PFCs的降解效率将是一种提可行的方案。
本发明还发现,在流域基质厌氧铁氨氧化过程中引入电子供体会增加有机碳的含量,并且会改变有机碳赋存形态,增加腐殖酸类、弗里酸类有机质含量能够促进铁离子的还原,并且影响厌氧铁氨氧化过程中铁离子去除底泥中PFCs的过程,提高PFCs的降解效率。
进一步的,所述污染底泥先通过自然风干,并剔除植物腐烂的根、石块等杂质再进行全氟化合物的降解。
进一步的,所述铁异化还原菌为希瓦氏菌或地杆菌属,优选为希瓦氏菌;
进一步的,所述电子供体为活性炭、蔗糖、腐殖酸、有机酸、氨氮中的一种或多种,而去除PFCs的过程会受到例如氧化还原电位、酸碱环境、能量、催化剂等很多因素的影响,因此,综合考虑多种因素,所述电子供体优选为氨氮。
进一步的,所述电子供体的添加量为污染底泥质量的5-10%,优选为5%。
进一步的,通过含铁化合物的形式将铁离子引入体系中;所述含铁化合物包括铁盐或铁矿石。
优选的,所述铁矿石包括磁铁矿、赤铁矿、褐铁矿、镜铁矿、菱铁矿、黄铁矿中的一种或多种;
优选的,所述铁盐包括氯化铁、硝酸铁、柠檬酸铁、氢氧化铁中的任意一种或多种;
进一步的,污染底泥与含铁化合物的体积比为,进一步优选为(0.5-3):1,优选为1:1。本发明第二方面提供一种厌氧铁氨氧化降解全氟化合物的方法在全氟化合物脱除方面的应用。
与现有技术相比,本发明取得了以下有益效果:
(1)本发明所提供的利用厌氧铁氨氧化降解全氟化合物的方法,能够有效实现对PFOA和PFOS的去除,氨氮作为电子供体,这种去除PFOA和PFOS的效果最为明显。而且,随着时间的延长,最终能够实现全氟化合物的完全去除,本发明所提供的方法为自然流域底泥中全氟化合物的去除提供新思路。
(2)本发明所提供的利用厌氧铁氨氧化降解全氟化合物的方法,能从根本上消除PFCs对环境及生物危害,且过程简单、成本低、不会造成二次污染。

应该指出,以下详细说明都是例示性的,旨在对本发明提供进一步的说明。除非另有指明,本文使用的所有技术和科学术语具有与本发明所属技术领域的普通技术人员通常理解的相同含义。
需要注意的是,这里所使用的术语仅是为了描述具体实施方式,而非意图限制根据本发明的示例性实施方式。如在这里所使用的,除非上下文另外明确指出,否则单数形式也意图包括复数形式,此外,还应当理解的是,当在本说明书中使用术语“包含”和/或“包括”时,其指明存在特征、步骤、操作、器件、组件和/或它们的组合。
实验方法:
从李村河采集底泥样品,自然风干后,剔除植物腐烂的根、石块等。通过烘干利用差量法计算土壤含水率,并称量每组土壤质量、计算土壤干重。将所取底泥混匀后,和铁矿石按照体积比1:1放置在6组1L聚丙烯烧杯中,编号为(1)-(6),每组约0.5L。分别向每组装置中加入约4.5mg/kg土样浓度的全氟辛酸(PFOA)和全氟辛烷基磺酸(PFOS),并接种典型铁异化还原菌希瓦氏菌。
分别向(1)-(5)装置中混合加入5%质量比的活性炭粉末、蔗糖、腐殖酸、有机酸、氨氮各10g,并设置(6)为空白对照组,探究不同电子供体对Feammox过程去除底泥全氟化合物的影响。分别称量256.75mg蔗糖、188mg硫酸铵、382.85mg硝酸钾、35mg磷酸二氢钾溶于800ml自来水中,配制为一级A浓度废水,模拟自然河流生态体系。
实验装置的运行维护与周期性取样:
每隔5天,利用标准取样方法取水样,并通过0.45μm的滤膜于50ml聚丙烯管中,测定六组装置中PFOA和PFOS浓度,及铁离子浓度、氟离子浓度、氨氮浓度、pH值、溶解性有机碳等浓度。均匀取底泥15g左右,利用冷冻干燥机干燥72小时后放置于-20℃下冷冻保存,用作后续各种物质和浓度的测定。将装置中的废水倾倒后加入重新配制的国家一级A浓度废水至装置的1L位置,维持实验装置的正常运行。以2个月为试验周期,试验周期结束后取底泥样品送样测定有机碳形态及微生物群落结构。
实验物质的测定:
(1)氟离子
称取不同周期下(1)-(6)号装置的土样各5g于50ml聚丙烯管中,加入25ml去离子水,利用超声波清洗机超声20min,水浴恒温振荡器振荡10min,利用离心机在6000rap/min下离心10min,将上清液通过0.45μm的滤膜,利用离子色谱法测定土样中氟离子参数。
(2)氨氮
利用可见分光光度法测定水样和土样中的NH4 +浓度。测定水样中的NH4 +浓度时,取水样100μL于50ml移液管中,并定容至刻度线处,用移液枪加入1ml酒石酸钾钠和1.5ml纳氏试剂,摇匀静置10min后利用可见分光光度计在420nm波长下测定不同周期内装置(5)的吸光度,代入NH4 +标线求出其最终浓度。测定土样中NH4 +浓度时,首先称取74.55g氯化钾溶解于1L的容量瓶中,配置成氯化钾标准溶液,之后称取不同周期装置(5)的土样各5g于50ml聚丙烯管中,加入25ml氯化钾标准溶液,利用水浴恒温振荡器振荡1h后离心或在4℃下静置4h,取上清液通过0.45um滤膜过滤,过滤后的上清液测定其NH4 +浓度时方法步骤同水样。
称取3.819g经100℃干燥过的氯化铵(NH4Cl)溶于水中,移入1000ml容量瓶中并稀释至标线,此时溶液每毫升含1.00mg氨氮,配置成为铵标准贮备溶液。移取5.00ml铵标准贮备液于500ml容量瓶中,用水稀释至标线,此时溶液每毫升含0.010mg氨氮,配置成为铵标准使用液。分别吸取0.00、0.10、0.20、0.60、1.00、1.40和2.00ml铵标准使用液于50ml比色管中,加水稀释至标线,加入1.0ml酒石酸钾钠和1.5ml纳氏试剂,混匀并放置10min后,在波长420nm处测定吸光度,绘制成为以氨氮含量(mg)对校正吸光度的标准曲线。

(3)pH
测定的土样pH时,以去离子水为浸提剂,加入去离子水构成2:1的水土比,搅拌2min,静置60min后,利用pH计测定其pH值。
(4)溶解性有机碳
称取不同周期下储存的(1)--(6)号装置的土样各5g于50ml聚丙烯管中,加入25ml去离子水,利用超声波清洗机超声20min,水浴恒温振荡器振荡10min,利用离心机在6000rap/min下离心10min,将上清液通过0.45um的滤膜,利用燃烧-非色散红外吸收法在TOC测定仪中测定其浓度。
对于水样中TOC浓度测定时,可将通过0.45um滤膜的上清液直接通过TOC测定仪进行测定。
(5)PFOA和PFOS
提取土样中PFOA和PFOS时,将不同周期冷冻干燥贮存下的土样各取5g于50ml聚丙烯管中,加入3ml甲醇溶液并利用超声波清洗机超声30min,利用离心机在4000rap/min下离心10min,提取上清液;重复上述操作后提取上清液;最后加入4ml甲醇溶液并利用超声波清洗机超声30min,利用离心机在4000rap/min下离心10min,提取上清液。将三次提取的上清液混合后通过0.45μm滤膜,将通过滤膜的液体寄出测定其PFOA和PFOS的值。
利用固相萃取仪提取水样中的PFOA和PFOS,首先于萃取柱中加入6ml 0.5%氨水/甲醇溶液,静置2min排入废水瓶;加入6ml甲醇,静置2min排入废水瓶;再加入6ml去离子水,静置2min排入废水瓶;打开废水阀,通过100ml容量瓶中的水样,待水样全部通过后,加入去离子水润洗容量瓶。最后分两次加入5ml甲醇并收集,将收集到的液体移入1ml试管中,贴好标签后邮寄送出测定。
(6)微生物群落结构
称取土样各5g进行高通量测序测定微生物群落结构。
(7)有机碳形态分析
15g土样进行EEM测定三维荧光光谱分析其碳形态。
结果分析:
1、不同环境影响因子对去除PFCs的效果分析:
(1)PFCs的去除效果

通过对连续不同周期水样中PFOA的含量进行测定,数据如表1所示,对数据结果绘制如图1所示。由图1可见,六组装置中的PFOA除(2)号蔗糖和(6)号空白样外,其余装置的PFOA均呈现下降趋势。

通过连续不同周期土样中PFOA的含量进行测定,数据如表2所示,对数据结果绘制如图2所示。由图2可见,六组装置中的PFOA均成下降趋势,说明所加入的不同电子供体在Feammox过程中均对PFOA的降解去除效果起到了不同的作用。其中氨氮对Feammox过程去除PFCs具有最明显的提升作用,其次为活性炭和腐殖酸,说明外加电子供体提高了PFCs的去除。

通过对连续不同周期水样中PFOS的含量进行测定,数据如表3所示,对数据结果绘制如图3所示。由图3可见,在六组装置中除(2)号蔗糖和(6)号空白样外,其余装置中的PFOS均随实验的运行呈现出降低趋势,说明底泥中PFCs向水中的释放逐渐减弱。

通过对不同周期土样中PFOS的含量进行测定,数据如表4所示,对数据结果绘制如图4所示。由图4可见,各个装置中对PFOS的去除表现出不同的去除效果。不同于PFOA,氨氮、腐殖酸、蔗糖对Feammox过程中PFCs的去除效果提升最明显。
2、氟离子的效果分析:

PFCs降解后会释放氟离子,通过对不同周期土样(表6)和水样(表5)中氟离子浓度进行测定,我们由测定结果可以看出蔗糖的添加可实现全氟化合物的脱氟降解。
3、氨氮的效果分析:

通过对连续不同周期水样中装置(5)的氨氮测定,得到的数据记录在表7中,不同周期水样中氨氮变化趋势如图5所示,显示氨氮浓度随周期的延长,不断呈现下降趋势,验证了Feammox过程。结合水样中装置(5)的PFCs变化趋势,发现两者均呈现出相同的下降趋势,说明在装置(5)中加入氨氮,可作为电子供体参与到Feammox过程,其电子传递过程对Feammox过程去除PFCs起到促进作用。
通过对不同周期土样中装置(5)的氨氮测定,得到的数据记录在表7中,不同周期土样中氨氮变化趋势如图6所示,可见随周期的延长,土样的氨氮浓度呈现出降低趋势,而土样中装置(5)的PFCs同样呈现出降低趋势,说明在Feammox过程中氨氮对起到促进作用。

如表8、9所示,随着模拟实验的进行,TOC被不断消耗,电子供体促进了Feammox过程对PFCs的降解和去除。投加蔗糖增加溶解性有机碳含量,为PFCs的降解提供共代谢碳源。
微生物群落结构分析
称取冷冻干燥的土样5g送样至北京诺禾致源公司,利用Illumina Miseq平台进行高通量测序分析并对数据进行处理分析。

实验通过分析各实验装置底泥微生物的16SrRNA序列,得到不同电子供体对于底泥中物种丰富度和微生物群落组成的变化情况,在种族间距为3%的水平下,对不同样本在97%一致性阈值下的OTUs数和α多样性分析指数进行统计,见表10,(1)号到(6)号装置的底泥微生物分别有62884、64740、65989、65455、61378和65324条带,与(6)号对照组的相比,(1)号到(5)号实验组的微生物物种多样性明显降低,这可能是由于各类电子供体的存在直接或间接改变了环境条件,并改变了微生物群落结构。

Claims (10)
1.一种利用厌氧铁氨氧化降解全氟化合物的方法,其特征在于:在厌氧铁氨氧化过程中进行全氟化合物的降解;
具体方式为:
在污染底泥中引入铁离子和铁异化还原菌进行厌氧铁氨氧化反应,并添加电子供体,进行全氟化合物的降解。
2.如权利要求1所述的方法,其特征在于:所述污染底泥先通过自然风干,并剔除植物腐烂的根、石块杂质后再进行全氟化合物的降解。
3.如权利要求1所述的方法,其特征在于:所述铁异化还原菌为希瓦氏菌或地杆菌属,优选为希瓦氏菌。
4.如权利要求1所述的方法,其特征在于:所述电子供体为活性炭、蔗糖、腐殖酸、有机酸、氨氮中的一种或多种,优选为氨氮。
5.如权利要求1所述的方法,其特征在于:所述电子供体的添加量为污染底泥质量的5-10%,优选为5%。
6.如权利要求1所述的方法,其特征在于:通过含铁化合物的形式将铁离子引入体系中;所述含铁化合物包括铁盐或铁矿石。
7.如权利要求6所述的方法,其特征在于:所述铁矿石包括磁铁矿、赤铁矿、褐铁矿、镜铁矿、菱铁矿、黄铁矿中的一种或多种。
8.如权利要求6所述的方法,其特征在于:所述铁盐包括氯化铁、硝酸铁、柠檬酸铁、氢氧化铁中的任意一种或多种。
9.如权利要求6所述的方法,其特征在于:污染底泥与含铁化合物的体积比为(0.5-3):1,优选为1:1。
10.权利要求1-9任一项所述的厌氧铁氨氧化降解全氟化合物的方法在全氟化合物脱除方面的应用。
