Abstract
本发明提供了一种载药纳米胶束制剂、及其制备方法和应用。还提供了该多肽药物及其纳米胶束制剂的制备方法。其中,所述的载药纳米胶束制剂由聚乙二醇化磷脂、癌症治疗性多肽组装形成。所述多肽药物能够于趋化因子受体CXCR4特异性结合,抑制由趋化轴CXCL12/CXCR4诱导的胰腺癌细胞的迁移。本发明的多肽药物及其胶束制剂为胰腺癌的治疗提供了可行的方法和技术。
Description
技术领域
本发明属于生物医药领域,具体涉及一种载药纳米胶束制剂、及其制备方法和应用。
背景技术
胰腺癌是恶性程度最高的消化道肿瘤,致死率极高,其五年生存率低于10%。化疗是胰腺癌治疗中除了手术治疗外最有效的治疗手段,然而即使是一线化疗药物吉西他滨,有效性也远低于临床预期。
趋化因子CXCL12及其受体CXCR4在许多癌种发生发展中起到重要作用。CXCR4在胰腺癌、乳腺癌、非小细胞肺癌以及食道癌等癌症中都有较高水平的表达,并与其淋巴结转移和不良预后相关。其中,CXCR4在胰腺癌组织中呈高表达,与胰腺癌的分化、转移、生长及预后密切相关。对正常胰腺组织、胰腺癌组织、癌旁组织和胰腺周围淋巴结组织进行免疫组织化学分析,发现CXCR4蛋白在正常胰腺组织中低表达,在胰腺癌组织、癌旁组织和胰腺周围淋巴结中高表达,表明CXCL12/CXCR4趋化轴可能参与了胰腺癌发生发展的过程,同时,CXCR4在淋巴结位置的过度表达也表示CXCR4可能与胰腺癌的淋巴结转移和远端转移有关。因此,CXCR4可以作为治疗胰腺癌的一个潜在标志物蛋白。
CXCR4拮抗剂主要有4种,小分子拮抗剂、多肽类拮抗剂、CXCR4抗体以及CXCL12衍生物。相比其他三种而言,多肽具有分子量小、结构简单、容易合成、特异性强、活性高、毒性低以及免疫原性低的特点,在疾病治疗方面具有很强的优势。同时,多肽药物也存在溶解性低、稳定性差、循环时间短等问题,阻碍了其药效发挥和生物利用度。随着纳米技术的发展,纳米药物载体在一定程度上能克服这些问题。纳米药物载体是尺寸在纳米级范围内的药物递送体系,可以提高药物的生物利用度,例如它可以控制药物在体内的释放过程,促进跨越生物屏障,具有较长的半衰期和不同的生物分布特征。此外,根据高渗透长滞留效应(EPR效应)纳米载体能够优先聚集在肿瘤组织中,从而降低对正常细胞的毒性,能够更好的治疗肿瘤。用于药物递送的常见纳米载体一般分为两类,有机纳米载体和无机纳米载体。有机纳米载体主要有脂质体、树枝状大分子、聚合物纳米粒子、聚合物胶束等,无机纳米载体有金属纳米颗粒、碳纳米管、超顺磁氧化铁纳米颗粒等。
聚合物纳米胶束作为纳米载药体系的一种,能够提高E5多肽在盐溶液中的溶解度,同时保护E5多肽,是载药体系的良好选择。但截至目前,还未见有将多肽药物及其胶束制剂用于胰腺癌的治疗。
发明内容
在阐述本发明内容之前,定义本文中所使用的术语如下:
术语“E5多肽”是指:氨基酸序列为GGRSFFLLRRIQGCRFRNTVDD(N端-C端)的多肽。
术语“CXCR4”是指:7次跨膜G蛋白偶联受体,趋化因子基质细胞衍生因子-1(CXCL12)的特异性受体。
术语“FITC”是指:异硫氰酸荧光素。
术语“CXCL12”是指:一种趋化因子,基质细胞衍生因子-1(SDF-1),一类由细胞分泌的小细胞因子或信号蛋白。
术语“PANC-1”是指:人胰腺癌细胞;PANC-1是源自于胰腺癌导管细胞,其倍增时间为52小时。术语“Capan-1”是指:人胰腺癌肿瘤细胞系Capan-1。
术语“BxPC-3”是指:人原位胰腺腺癌细胞系BxPC-3。
术语“MiaPaCa-2”是指:人胰腺癌细胞系MiaPaCa-2。
术语“T3M4”是指:人胰腺腺癌细胞系T3M4。
术语“Transwell实验”是指:细胞迁移实验。
因此,本发明的目的在于克服现有技术中的缺陷,提供一种载药纳米胶束制剂、及其制备方法和在胰腺癌治疗中的应用。
为实现上述目的,本发明的第一方面提供一种载药纳米胶束制剂,所述的载药纳米胶束制剂由聚乙二醇化磷脂、癌症治疗性多肽组装形成;其中:
所述的聚乙二醇化磷脂由作为亲水嵌段的聚乙二醇通过共价键与作为疏水嵌段的磷脂分子上的含氮碱基结合形成;并且
所述的癌症治疗性多肽是能够与趋化因子受体CXCR4结合,从而抑制胰腺癌细胞迁移的多肽;
优选地,所述载药纳米胶束制剂为溶液形式或冻干形式。
根据本发明第一方面的载药纳米胶束制剂,所述的癌症治疗性多肽选自以下一种或多种:以极性氨基酸为主的多肽、以疏水氨基酸为主的多肽、兼有极性氨基酸和疏水性氨基酸的多肽;
优选地,所述的癌症治疗性多肽由5-50个氨基酸组成,更优选为10-30个氨基酸,进一步优选为15-25个氨基酸。
根据本发明第一方面的载药纳米胶束制剂,所述的癌症治疗性多肽为E5多肽或FITC标记的E5多肽;
优选地,所述的E5多肽被探针标记或纳米颗粒包载;
更优选地,其中:
所述的探针选自以下一种或多种:荧光分子、量子点、放射性元素、辣根过氧化物酶、碱性磷酸酶;

根据本发明第一方面的载药纳米胶束制剂,所述聚乙二醇化磷脂分子中的聚乙二醇亲水嵌段分子量为500-10000,优选为1000-5000,进一步优选为2000-4000;
所述载药纳米胶束的粒径为10-100nm,优选为10-50nm,更优选为10-30nm;
所述聚乙二醇化磷脂与癌症治疗性多肽的摩尔比为2-10:1,优选为4-5:1。
本发明的第二方面提供了第一方面所述的载药纳米胶束制剂的制备方法,所述的方法包括以下步骤:
(1)分别制备聚乙二醇化磷脂分子溶液和E5多肽分子溶液,混匀,水浴孵育,静置,得多肽的载药纳米胶束溶液;
(2)优选地,将步骤(1)静置后所得载药纳米胶束溶液进行除菌、过滤。
根据本发明的第二方面的制备方法,其中,在步骤(1)中:
溶剂为以下一种或多种:磷酸盐缓冲液、超纯水、生理盐水,优选为磷酸盐缓冲液和超纯水,最优选为超纯水;
聚乙二醇化磷脂溶液浓度为1-50mg/mL,优选为1-30mg/mL,进一步优选为2-20mg/mL;
E5多肽溶液浓度为0.1-15mg/mL,优选为0.1-10mg/mL,进一步优选为0.5-5mg/mL;
水浴孵育温度为30-65℃,优选为40-60℃,进一步优选为40-55℃;
水浴孵育时间为20-60min,优选为20-50min,进一步优选为25-40min;
静置温度为4-30℃,优选为10-30℃,进一步优选为静置温度为20-25℃;
静置时间为2-48小时,优选为2-36小时,进一步优选为4-24小时;
聚乙二醇化磷脂与E5多肽的摩尔比为:40-50:1-20,优选为2-10:1,进一步优选为4-5:1。
根据本发明的第二方面的制备方法,其中,在步骤(2)中:所述过滤膜孔径为0.1-0.8μm,优选为0.1-0.6μm,进一步优选为0.1-0.4μm,最优选为0.22μm。
本发明的第三方面提供了第一方面所述的载药纳米胶束制剂或按照第二方面所述的方法制备的载药纳米胶束制剂制备用于治疗癌症药物中的应用:
优选地,所述癌症为胰腺癌;
更优选地,所述胰腺癌表征具有表达或过表达CXCR4的胰腺癌细胞或胰腺癌组织。
根据本发明的第三方面的应用,所述胰腺癌细胞的种类选自以下一种或多种:PANC-1、Capan-1、BxPC-3、MiaPaCa-2、T3M4,优选为PANC-1或MiaPaCa-2。
根据本发明一个特别优选的实施方式,提供一种能够针对CXCR4靶点的多肽E5及其载药纳米胶束,所述多肽通过和CXCR4结合来抑制胰腺癌细胞的迁移,达到治疗胰腺癌的目的。
本发明多肽药物及其胶束制剂可以具有但不限于以下有益效果:
1.载药纳米胶束提高了E5多肽在生理溶液中的溶解性和生物稳定性,提高了E5多肽和CXCR4靶点的结合能力,从而提高对胰腺癌细胞迁移的抑制能力。
2.多肽E5与表达CXCR4的胰腺癌细胞具有较高的亲和力,说明可以作为CXCR4拮抗剂用于治疗胰腺癌(图2)。E5多肽自身安全性较好,在60微摩尔浓度范围内无明显细胞毒性。(图3)
3.多肽药物及其胶束制剂经透射电子显微镜显示,该胶束成球形结构,粒径较均一,所制备的胶束粒径分布在10-30nm(图4)。
4.多肽E5载药纳米胶束能够抑制胰腺癌细胞系PANC-1和MiaPaCa-2的细胞迁移,在聚乙二醇化磷脂分子浓度为200微摩尔每升时,能够明显抑制胰腺癌细胞的迁移(图6)。

下面通过具体的实施例进一步说明本发明,但是,应当理解为,这些实施例仅仅是用于更详细具体地说明之用,而不应理解为用于以任何形式限制本发明。
本部分对本发明试验中所使用到的材料以及试验方法进行一般性的描述。虽然为实现本发明目的所使用的许多材料和操作方法是本领域公知的,但是本发明仍然在此作尽可能详细描述。本领域技术人员清楚,在上下文中,如果未特别说明,本发明所用材料和操作方法是本领域公知的。
以下实施例中使用的材料、试剂和仪器如下:
材料:
人源胰腺癌细胞系PANC-1购自:中国医学科学院基础医学研究所细胞资源中心。
人源胰腺癌细胞系MiaPaCa-2购自:中国典型培养物保藏中心细胞库。
小鼠来源骨髓基质细胞购自:青旗(上海)生物技术发展有限公司。
试剂:
E5多肽购自安徽省国平药业有限公司,纯度为98%。
聚乙二醇化磷脂购自美国Avanti Polar Lipids(Alabama)公司。
所使用的E5多肽和聚乙二醇化磷脂分子均在实验前用超纯水配置成合适浓度的母液。
无菌超纯水溶液,水质参数为电阻率18.2MΩ.cm@25℃。
PBS缓冲液、DMEM高糖培养基、1640培养基、胰酶以及胎牛血清购自美国Gibco公司。
CCK-8试剂盒购自日本DOJINDO公司。
结晶紫溶液购自索莱宝公司。
仪器:
Transwell小室购自美国R&D公司。
连续光谱多功能酶标仪购自美国Molecular Devices公司,型号为SpectraMaxi3。
流式细胞仪购自美国BD公司,型号为C6。
透射电子显微镜(TEM)购自日本日立公司,型号HT7700。
实施例1:
本实施例用于说明本发明多肽药物及其胶束制剂的制备方法。
(1)聚乙二醇化磷脂分子溶液的制备方法:将聚乙二醇化磷脂粉末溶解于适量超纯水中,完全溶解后得到1mM的聚乙二醇化磷脂分子溶液。
(2)E5多肽分子溶液:将E5多肽的粉末溶解于适量超纯水中,超声5分钟得到500μM的E5多肽分子溶液。
(3)将步骤(1)和步骤(2)中两种分子溶液混匀,水浴孵育,静置,得多肽的载药纳米胶束溶液。
(4)将步骤(3)所得载药纳米胶束溶液静置后除菌,再用0.22μm滤膜过滤。
实施例2:细胞表面CXCR4表达量的测定
利用流式细胞术测定细胞表面的CXCR4表达量时,将细胞分装进1.5mL离心管中,每管50万左右细胞,实验组孵育趋化因子受体CXCR4的抗体(一抗,鼠抗人),将细胞吹打均匀,室温下孵育1小时,然后将细胞悬液离心,弃去上清,用PBS重悬,再次离心,如此重复三次,洗去非特异性结合的抗体,然后加入200μL配置好的有AF488标记的山羊抗鼠二抗溶液,对照组也加入同样的二抗溶液,将细胞吹打均匀,一起在室温下孵育30min,然后将细胞悬液离心,弃去上清,用PBS重悬,再次离心,如此重复三次,洗去非特异性结合的抗体,最后用200μL PBS缓冲液将细胞重悬。如图1所示,利用CXCR4抗体筛选出了一株CXCR4低表达的细胞株MS-5,两株CXCR4高表达的细胞株PANC-1和MiaPaCa-2。MS-5、PANC-1和MiaPaCa-2的CXCR4表达量分别为:4.0%、51.1%和39.7%。说明MS-5细胞为CXCR4阴性细胞,PANC-1和MiaPaCa-2为CXCR4阳性细胞。
实施例3:E5多肽与CXCR4亲和力测定
利用流式细胞术来检测E5多肽和不同趋化因子受体CXCR4表达量细胞系的结合能力。将细胞分装进1.5mL离心管中,每管50万左右细胞,离心后弃去上清。配制浓度分别为2、4、6、8、10μM的FITC-E5多肽溶液,分别加入50μL至收集了细胞的离心管,对照组不加多肽,加入200μL PBS缓冲液,用移液枪轻轻吹打至细胞悬浮均匀,在37℃下孵育2h。将孵育好的细胞加入适量PBS缓冲液重悬,离心,弃去上清,用适量的PBS缓冲液重悬细胞,再次离心,重复3次,洗去非特异性结合的多肽,最后用200μL的PBS缓冲液重悬,得到测试样品。如图2所示,过表达CXCR4的细胞系PANC-1和MiaPaCa-2与E5多肽的结合能力很强,CXCR4阴性细胞系MS-5与E5多肽的结合能力基本可以忽略不计,说明E5多肽与CXCR4有较高的结合能力。
实施例4:E5多肽载药纳米胶束形貌分析
利用透射电子显微镜对E5多肽载药纳米胶束(M-E5)形貌进行表征。将胶束溶液稀释至20μM,滴10μL液体在铜网上,静置过夜,滴加2wt%的醋酸双氧铀溶液,染色1min后,用滤纸吸取液体,自然晾干后,在透射电子显微镜下观察拍照。由图4可知,载药纳米胶束的尺寸在20nm左右。
试验例1:E5多肽的细胞毒性实验
用CCK-8法检测E5多肽对不同细胞系的细胞活力的影响。在96孔板(Corning)中,用对应的培养基培养细胞,PANC-1和MS-5细胞4×103每孔,MiaPaCa-2细胞6×103每孔,将96孔板放置于37℃、5%CO2的培养箱种培养24h。用相应的培养基配制不同浓度的E5多肽溶液,对96孔板中的细胞进行换液,培养48h后,每孔向每个孔中加入10μL体积的CCK-8溶液,在37℃下孵育1h,用连续光谱多功能酶标仪(SpectraMax i3,美国)测定在450nm波长下的吸光度(OD)值,计算细胞的存活率。每个样品设置三个平行复孔发,实验结果为三次实验平均值。如图3所示,E5多肽自身安全性较好,在60微摩尔浓度范围内无明显细胞毒性。
试验例2:游离E5多肽对胰腺癌细胞迁移的抑制能力评价
利用Transwell实验探究游离E5多肽对胰腺癌细胞迁移的抑制能力。向24孔板中加入不含或含有CXCL12蛋白的完全培养基。配制含有不同浓度E5多肽的无血清培养基。收集细胞PANC-1和MiaPaCa-2,每种细胞各1×105个,每种细胞各200μL于1.5mL离心管中,加入配置好的含有E5多肽的培养基,在37℃下孵育1h,再次将细胞重悬,向孔径为8μm的Transwell小室中加入100μL的细胞悬液,悬于对应的下室,放置于37℃、5%CO2条件的培养箱中培养24h。
将小室放入0.1%的结晶紫溶液中,染色10min,用清水冲洗干净,然后用棉签将小室滤膜内侧的细胞擦拭干净,将小室晾干,在显微镜下拍照成像。拍照后加入冰醋酸脱色10min,吸取每个样品200μL于96孔板中,用连续光谱多功能酶标仪测定在530nm波长下的吸光度(OD)值,从而计算细胞迁移率。如图5,加入1μM、10μM和50μM的E5多肽后,可以看到,当E5多肽浓度为50μM的时候迁移数量有较为显著的降低。说明E5多肽能够抑制胰腺癌细胞的迁移。
试验例3:E5多肽载药纳米胶束(M-E5)对胰腺癌细胞迁移的抑制能力评价
利用Transwell实验探究M-E5对胰腺癌细胞迁移的抑制能力。向24孔板中加入不含或含有CXCL12蛋白的完全培养基。配制含有不同浓度M-E5的无血清培养基。收集细胞PANC-1和MiaPaCa-2,每种细胞各1×105个,每种细胞各200μL于1.5mL离心管中,加入配置好的含有M-E5的培养基,在37℃下孵育1h,再次将细胞重悬,向孔径为8μm的Transwell小室中加入100μL的细胞悬液,悬于对应的下室,放置于37℃、5%CO2条件的培养箱中培养24h。
将小室放入0.1%的结晶紫溶液中,染色10min,用清水冲洗干净,然后用棉签将小室滤膜内侧的细胞擦拭干净,将小室晾干,在显微镜下拍照成像。拍照后加入冰醋酸脱色10min,吸取每个样品200μL于96孔板中,用连续光谱多功能酶标仪测定在530nm波长下的吸光度(OD)值,从而计算细胞迁移率。如图6,按照体系中PEG-PE分子的浓度计,当M-E5浓度为200μM,即E5多肽浓度为50μM时,细胞迁移数量有较为明显的降低。说明E5多肽载药纳米胶束能够抑制胰腺癌细胞的迁移。
尽管本发明已进行了一定程度的描述,明显地,在不脱离本发明的精神和范围的条件下,可进行各个条件的适当变化。可以理解,本发明不限于所述实施方案,而归于权利要求的范围,其包括所述每个因素的等同替换。
Claims (10)
1.一种载药纳米胶束制剂,其特征在于,所述的载药纳米胶束制剂由聚乙二醇化磷脂、癌症治疗性多肽组装形成;其中:
所述的聚乙二醇化磷脂由作为亲水嵌段的聚乙二醇通过共价键与作为疏水嵌段的磷脂分子上的含氮碱基结合形成;并且
所述的癌症治疗性多肽是能够与趋化因子受体CXCR4结合,从而抑制胰腺癌细胞迁移的多肽;
优选地,所述载药纳米胶束制剂为溶液形式或冻干形式。
2.根据权利要求1所述的载药纳米胶束制剂,其特征在于,所述的癌症治疗性多肽选自以下一种或多种:以极性氨基酸为主的多肽、以疏水氨基酸为主的多肽、兼有极性氨基酸和疏水性氨基酸的多肽;
优选地,所述的癌症治疗性多肽由5-50个氨基酸组成,更优选为10-30个氨基酸,进一步优选为15-25个氨基酸。
3.根据权利要求1或2所述的载药纳米胶束制剂,其特征在于,所述的癌症治疗性多肽为E5多肽或FITC标记的E5多肽;
优选地,所述的E5多肽被探针标记或纳米颗粒包载;
更优选地,其中:
所述的探针选自以下一种或多种:荧光分子、量子点、放射性元素、辣根过氧化物酶、碱性磷酸酶;
所述的纳米颗粒选自以下一种或多种:脂质体、聚合物胶束、树枝状大分子、金纳米颗粒。
4.根据权利要求3所述的载药纳米胶束制剂,其特征在于:
所述E5多肽的氨基酸序列为:GGRSFFLLRRIQGCRFRNTVDD,所述荧光探针标记的E5多肽的氨基酸序列为:FITC-GGRSFFLLRRIQGCRFRNTVDD;
优选地,所述载药纳米胶束制剂是由聚乙二醇化磷脂分子与E5多肽通过静电相互作用组装形成的包埋了E5多肽的纳米胶束。
5.根据根据权利要求1至4中任一项所述的载药纳米胶束制剂,其特征在于:
所述聚乙二醇化磷脂分子中的聚乙二醇亲水嵌段分子量为500-10000,优选为1000-5000,进一步优选为2000-4000;
所述载药纳米胶束的粒径为10-100nm,优选为10-50nm,更优选为10-30nm;

6.根据权利要求1至5中任一项所述的载药纳米胶束制剂的制备方法,其特征在于,所述的方法包括以下步骤:
(1)分别制备聚乙二醇化磷脂分子溶液和E5多肽分子溶液,混匀,水浴孵育,静置,得多肽的载药纳米胶束溶液;
(2)优选地,将步骤(1)静置后所得载药纳米胶束溶液进行除菌、过滤。
7.根据权利要求6中所述的载药纳米胶束制剂的制备方法,其特征在于,在步骤(1)中:
溶剂为以下一种或多种:磷酸盐缓冲液、超纯水、生理盐水,优选为磷酸盐缓冲液和超纯水,最优选为超纯水;
聚乙二醇化磷脂溶液浓度为1-50mg/mL,优选为1-30mg/mL,进一步优选为2-20mg/mL;
E5多肽溶液浓度为0.1-15mg/mL,优选为0.1-10mg/mL,进一步优选为0.5-5mg/mL;
水浴孵育温度为30-65℃,优选为40-60℃,进一步优选为40-55℃;
水浴孵育时间为20-60min,优选为20-50min,进一步优选为25-40min;
静置温度为4-30℃,优选为10-30℃,进一步优选为静置温度为20-25℃;
静置时间为2-48小时,优选为2-36小时,进一步优选为4-24小时;
聚乙二醇化磷脂与E5多肽的摩尔比为:40-50:1-20,优选为2-10:1,进一步优选为4-5:1。
8.根据权利要求6中所述的载药纳米胶束制剂的制备方法,其特征在于,在步骤(2)中:所述过滤膜孔径为0.1-0.8μm,优选为0.1-0.6μm,进一步优选为0.1-0.4μm,最优选为0.22μm。
9.权利要求1至5中任一项所述的多肽药物及其胶束制剂或按照权利要求6至8中任一项所述方法制备的多肽药物及其胶束制剂在制备用于治疗癌症药物中的应用;
优选地,所述癌症为胰腺癌;
更优选地,所述胰腺癌表征具有表达或过表达CXCR4的胰腺癌细胞或胰腺癌组织。
10.根据权利要求9中所述的应用,其特征在于,所述胰腺癌细胞的种类选自以下一种或多种:PANC-1、Capan-1、BxPC-3、MiaPaCa-2、T3M4,优选为PANC-1或MiaPaCa-2。
