Technical Field
The invention relates to the technical field of plant extraction, and particularly relates to a ginseng and gimeracil fruit extracting solution and a preparation process thereof.
Background
The plants contain various active substances, such as terpenoids, aromatic compounds, aliphatic compounds, nitrogen-containing and sulfur-containing compounds and the like, and the active substances have various functional properties, such as oxidation resistance, bacteriostasis, cancer resistance, tumor inhibition, mosquito repelling and insect prevention and the like. However, the plants contain many impurities which are not digested by human body, and the impurities can bring a part of active substances out of the body, so that the efficiency of the active substances absorbed and utilized by the human body is high. In order to increase the availability of active substances, plant active substances are often extracted to increase the purity of the active substances. Among the plant extraction methods, the steam distillation method is widely used due to the advantages of simple equipment, simple operation, low extraction cost and the like, but the higher temperature of the steam distillation method easily causes thermal decomposition and hydrolysis of a target substance, thereby causing the failure of part of active substances.

Therefore, the ginseng and ji fruit extracting solution can be applied to the fields of resisting tumors, reducing blood fat and blood sugar, preventing aging and the like.
Detailed Description
In order to make the objects, technical solutions and advantages of the embodiments of the present invention clearer, the technical solutions in the embodiments of the present invention will be clearly and completely described below. The examples, in which specific conditions are not specified, were conducted under conventional conditions or conditions recommended by the manufacturer. The reagents or instruments used are not indicated by the manufacturer, and are all conventional products available commercially.
It should be noted that the embodiments and features of the embodiments in the present application may be combined with each other without conflict. The present invention will be described in detail below with reference to specific examples.
The invention provides a preparation process of a ginseng and gimbals fruit extracting solution, which comprises the following steps: adding ethanol into Ginseng radix fructus Jujubae, reflux extracting at 70-80 deg.C, and filtering to obtain residue and filtrate; then, taking the filter residue obtained in the previous reflux extraction as a raw material, and extracting according to the reflux extraction steps for three times in total; mixing the filtrates obtained in the three times to obtain a mixed solution; distilling the mixed solution at 45-50 deg.C under reduced pressure to obtain crude extractive solution; centrifuging the crude extract, and only taking the upper and middle layer solution to obtain emulsion; adsorbing the emulsion by adopting an adsorbing material, and eluting by using ethanol to obtain an active solution; distilling the active solution at 45-55 deg.C under reduced pressure until no ethanol is present to obtain extractive solution of radix Ginseng and fructus Jujubae.
In detail, the invention selects ethanol as the extraction solvent of the ginseng fruit, and the active substances are extracted from the ginseng fruit by a series of physical processes of permeation, dissolution, distribution, diffusion and the like of the ethanol in the ginseng fruit. In detail, the feed-liquid ratio of the ginsengii fruit to the ethanol is 1 (8-12), the feed-liquid ratio can ensure that the ginsengii fruit is fully contacted with the ethanol, so that active substances are dissolved out more quickly, raw materials are wasted when the feed-liquid ratio is too large, and the extraction rate of the active substances is reduced when the feed-liquid ratio is too small, so that the bioavailability is reduced. Further, in order to sufficiently dissolve the active substances in the Jiyu fruit, the Jiyu fruit can be crushed into particles of 180-220 meshes and then added with ethanol. The ginseng fruit is crushed into small particles, so that the contact area of ethanol and the ginseng fruit particles can be increased, the dissolution rate of active substances is accelerated, the extraction time is shortened, and the extraction rate of the active substances of the ginseng fruit is improved.
Reflux extracting mixture of ethanol and radix Ginseng and fructus Jujubae at 70-80 deg.C. Wherein, the movement speed of molecules can be accelerated by the temperature rise, and the permeation, diffusion and dissolution speed of the molecules can be accelerated by the structural change of cell membranes caused by the high temperature. However, since the temperature is too high, the structure of part of the active material is destroyed, and the ethanol is easily volatilized, which is not favorable for extraction of the active material, and the extraction amount is reduced, the temperature of the reflux extraction is preferably 70 to 80 ℃.
In detail, 70-90 v/v% ethanol is used in reflux extraction. According to the principle of similar solubility, the extraction amount of active substances gradually increases with the increase of ethanol concentration. However, when the concentration is more than 80 v/v%, the water content in the ethanol solvent is reduced, the polarity of the solvent is weakened, which is disadvantageous for the extraction of the ginseng fruit, and the extraction rate of the active substance is lowered. Therefore, it is reasonable to select an ethanol concentration of 70-90 v/v%.
Further, the time for reflux extraction is 70-90 minutes. The longer the reflux time, the more beneficial the extraction of the active substance. However, the reflux time is too long, and the active material is affected by the temperature, thereby causing a decrease in the extraction rate of the active material. Therefore, the reflux time is too long to facilitate the extraction of the active substance, so the reflux time is selected to be 70-90 minutes.
After the active substances in the ginseng and fruit are extracted by the ethanol, a plurality of active substances still remain in the filter residue, the filter residue is taken as the raw material, the secondary extraction is carried out by the ethanol according to the reflux extraction step, and the secondary filter residue is taken as the raw material for carrying out the tertiary extraction. Therefore, the residual substances in the filter residue can be extracted, the richness of the active substance composition is increased, the extracting solution is closer to the effective components in the natural ginseng fruit, and the physiological effect of the active substances is improved.
Mixing the filtrates to obtain a mixed solution, and distilling at 45-50 deg.C under reduced pressure to obtain a crude extractive solution. The reduced pressure distillation can purify the ginseng fruit at a relatively low temperature, and the fragrance and the physiological function of the active substances of the ginseng fruit are more similar to those of natural ginseng fruit. Not only can keep the savory and mellow smell of the ginseng fruit, but also can keep the physiological characteristics of the ginseng fruit, and furthest keeps the efficacy of active substances.
The crude extract is centrifugally separated, only the solution of the upper layer and the solution of the middle layer are reserved, dissolved impurities in the lower layer can be removed, active ingredients of the ginseng fruit are reserved to the maximum degree, and the utilization rate of a human body is improved. In detail, the centrifugal rotating speed is 7500-8500r/min, and the centrifugal time is 8-15 minutes. The liquid-liquid separation is realized by utilizing the principle that components with different emulsion densities rapidly settle and separate in a centrifugal force field. Under the condition of centrifugal separation, impurity substances dissolved out from the crude extraction liquid and active substances can be separated from each other, and the purity of the active substances is improved.
And then, adsorbing the emulsion by using an adsorbing material, and further purifying and decontaminating the emulsion. In detail, the adsorbing material in the invention is macroporous adsorption resin, and the macroporous adsorption resin is a separation material combining the adsorption and screening principles. The adsorption of the macroporous adsorption resin is essentially a surface adsorption phenomenon generated by high dispersion of objects or uneven acting force on surface molecules, and the adsorption performance is caused by Van der Waals attractive force or hydrogen bond generation; meanwhile, due to the porous structure of the macroporous adsorption resin, the macroporous adsorption resin has a screening effect on substances with different molecular sizes. The invention adopts AB-8 type macroporous absorption resin, and absorbs the emulsion with the volume flow of 1.3-1.8 BV/h. The AB-8 type macroporous adsorption resin can adsorb a large amount of active substances in a short time under the volume flow, and has high desorption rate and high desorption speed. The macroporous absorption resin is eluted by ethanol to achieve the purpose of separating active substances, and then the active substances are purified and purified. Further, after the emulsion is adsorbed by an adsorbing material, 55-65 v/v% ethanol is adopted for elution to obtain active liquid. The ethanol is used as eluent, the volume fraction of the ethanol is too low or too high, the polarity of the ethanol is greatly different from that of a target compound, the chemical bond formed by the resin and the active substance cannot be damaged, and other adsorbed impurities compete for desorption to reduce the elution rate, so 55-65 v/v% of ethanol is adopted.
Then distilling the active liquid at 45-55 deg.C under reduced pressure until no ethanol is present to obtain extractive solution of radix Ginseng and fructus Jujubae. The process can retain the physiological activity of the active substance, and concentrate the active substance, so that the content of the active substance in unit volume is increased, and the physiological performance and efficacy of the active substance are improved. Further, before the active liquid is distilled under reduced pressure, the active liquid is treated with an ion exchange resin to remove salts and pigments in the active liquid. When the active substances are extracted from the ginseng fruit, a part of impurities such as pigments, salts and the like are often mixed in the solution of the ginseng fruit. Wherein the pigment is mainly derived from pigment contained in the cellulose raw material, and products generated by caramelization reaction, Maillard reaction and the like. During the extraction process of the ginseng fruit, a large amount of sodium ions are often mixed into the extracting solution, and the quality of the ginseng fruit extracting solution serving as a final product is reduced due to the existence of the sodium ions. When the extract is used as an additive to be added into other substances, the salts and the pigments can cause the problems of demulsification, deepening color and the like of a final product, and the application of the ginseng fruit extract is limited. The ion exchange resin is a high molecular compound with a network structure of functional groups, and the ion exchange resin has high purity of the obtained product due to simple separation technology and equipment, convenient operation and high production continuity degree. In the invention, cation exchange resin is adopted to desalt desorption solution, and anion exchange resin is adopted to decolor desorption solution.
The invention also provides the extracting solution of the ginseng and ji fruit, and the extracting solution of the ginseng and ji fruit prepared by the preparation process of the extracting solution of the ginseng and ji fruit contains more active substances and more abundant components, thereby improving the bioavailability of the ginseng and ji fruit. The cucurbitacin component contained in the herba senecionis extract is a highly-oxidized tetracyclic triterpenoid extracted from plants, and the cucurbitacin B, E is the main component, so that the herba senecionis extract has various biological activity functions of resisting tumors, resisting inflammation and the like. The extract of the ginseng and the fructus lycii also contains gypenoside and ginseng total saponin, and the pharmacological actions of the gypenoside and the ginseng total saponin are similar, so that the ginseng and the fructus lycii have the pharmacological actions of obviously reducing blood fat, reducing blood sugar, balancing blood pressure, resisting cancer and aging, protecting liver, improving immune function and the like. Due to the pharmacological characteristics of the Shenji fruit extract, the Shenji fruit extract can be used as an additive and widely applied to the fields of medicinal preparations, beverages, skin care products and the like, and has wide application prospect.
