本发明公开了一种藤本囊托羊蹄甲的扦插繁殖方法,包括以下步骤:S1:选取扦插枝条:选择插穗枝条,将每根插穗枝条的顶端剪成平口,底端剪成羊蹄状,插穗用橡皮筋绑好,用多菌灵水溶液消毒,消毒后的插穗放置在阴凉环境中保湿,备用;S2:制作育苗袋:将育苗基质分装在降解免脱无纺布育苗袋中,用甲霜恶霉灵溶液浇透,然后用薄膜盖,备用;S3:扦插:将插穗用生根剂浸泡,浸泡好的插穗扦插于育苗基质中,使插穗与育苗基质紧密接触;扦插完成后,用多菌灵水溶液淋透,搭盖小拱棚、覆盖塑料薄膜和遮阴网85~90%。本发明解决了藤本囊托羊蹄甲遗传性状不稳定、长势参差不齐、繁殖效率低的问题。

豆科(Leguminosae)羊蹄甲属植物多为乔木,灌木或攀援藤本。全世界约600种,遍布于热带地区。我国有40种,4亚种,11变种,主产华南和西南地区。羊蹄甲属中的多种植物在世界范围内作为药用植物用于镇痛、抗菌消炎、促进伤口愈合、免疫调节、抗氧化、抗肿瘤、抗抑郁及治疗糖尿病等。羊蹄甲属植物含有丰富的黄酮类、萜类、生物碱类、木脂素类、甾体类等成分。
囊托羊蹄甲(Bauhinia touranensis Gagnep.)为豆科(Leguminosae)羊蹄甲属植物。木质藤本,有卷须,生于山坡或灌丛中,产于云南南部、贵州和广西西南部,在闽粤地区有栽培,越南也有分布。《中国中药资源志要》和《广西药用植物名录》中记载其功效为驱风活络,外用治疮疖,药用部位为茎。囊托羊蹄甲的提取物具有较好的神经细胞保护作用。从产自云南普洱的囊托羊蹄甲茎枝的95%乙醇提取物中分离得到39个化合物,包括3个新化合物、4个新天然产物及其他32个己知化合物。分别鉴定为touranlactone、glochidiolide、phyllanthurinolactone、glochidionlactoneA、menisdaurillide、aquilegiolide、黑麦草内醋、tourancyclohexenol、5-羟基-2-吲哚酮、oxndole、L-色氨酸、tourancyanphenoll、tourancyanphenollⅡ、2-cyanomethylphenol、touranphenol、2-hydroxyphenylaceticacid methyl ester、没食子酸甲酯、没食子酸乙酯、5,7-二羟基色原酮、儿茶素、表儿茶素没食子酸酯、柚皮素、高北美圣草素、北美圣草素、木犀草素、金丝桃苷、阿福豆苷、杨梅苷、槲皮苷、异槲皮苷、6-hydroxy-3',4',5,5',7-pentamethoxyflavone、4-轻基苯甲醛、对香豆酸乙酯、反式对香豆酸乙酯、松脂酚、(一)-丁香脂素、hedyotolC、片胡萝卜苷、片谷甾醇。藤本囊托羊蹄甲生长速度快,攀缘性强,适宜在边坡绿化、矿山修复等地运用,叶淡绿色,嫩叶淡紫红色,呈羊蹄状,花呈白带淡绿色,具有较强的观赏性。
藤本囊托羊蹄甲目前尚未由人工引种栽培,藤本囊托羊蹄甲的繁育技术也未见有相关的报道。
发明内容
本发明要解决的技术问题是提供一种藤本囊托羊蹄甲的扦插繁殖方法,摒弃现有技术中采用种子繁殖的方式,能够达到提高生根率、平均根长、平均生根数及综合指标的效果,解决藤本囊托羊蹄甲遗传性状不稳定、长势参差不齐、繁殖效率低的问题。
为了解决以上技术问题,本发明采用以下技术方案:
一种藤本囊托羊蹄甲的扦插繁殖方法,包括以下步骤:
S1:选取扦插枝条:选择一年生枝条作为插穗,将每根插穗枝条的顶端剪成平口,底端剪成羊蹄状,将插穗按30~50根为一捆用橡皮筋绑好,用800~1000倍水稀释的多菌灵水溶液消毒5~15min,消毒后的插穗放置在阴凉环境中保湿18~24小时,备用;
S2:制作育苗营养袋:将育苗基质分装在降解免脱无纺布育苗袋中,用500~1000倍水稀释的25%甲霜恶霉灵溶液浇透,然后用薄膜盖2~3天,备用;所述的育苗基质由包括以下重量份的原料制备而成:黄心土3~7份、泥炭土1~3份、椰糠1~3份、芥末粉1~2份、灯芯草粉1~2份、救必应渣1~2份、黄芪渣1~2份、烟草渣1~2份;
S3:扦插:将经步骤S1处理后的插穗用生根剂浸泡,浸泡好的插穗扦插于育苗基质中,使插穗与育苗基质紧密接触;扦插完成后,用800~1000倍水稀释的多菌灵水溶液淋透,搭盖小拱棚,覆盖塑料薄膜和遮阴网,覆盖率为85~90%。
进一步,所述的步骤S1中,插穗枝条呈细圆柱形、四方形或中圆柱形。
进一步,所述的步骤S1中,每根插穗主枝条长度为15~20cm,侧枝长度为5~8cm,侧枝含有4~6个芽眼。
进一步,所述的步骤S3中,生根剂由包括以下重量份的原料制备而成:生根粉5~10份、Na2SeO3 0.01~0.03份、Ce(NH4)2(NO3)6 0.01~0.03份、维生素C磷酸酯镁0.01~0.03份。

1、本发明的藤本囊托羊蹄甲的扦插繁殖方法属于无性繁殖,打破常规,摒弃现有技术中采用种子繁殖的方式,解决了繁殖效率低、繁殖后移栽成活率低、生长慢等问题,形成了藤本囊托羊蹄甲的扦插繁殖育苗技术体系,通过扦插繁殖获得的新植株,能够有效保留母体本身的所有特性,保持藤本囊托羊蹄甲的遗传稳定性,并且扦插繁殖的新植株生长更快,可更高效的投入到生产实践中。扦插过程中经过枝条选取、生根液浸泡等特殊处理,能够提高繁殖系数,缩短繁殖周期,提高藤本囊托羊蹄甲质量,对于推动藤本囊托羊蹄甲产业的发展具有重要意义,适合推广应用。
2、本发明根据藤本囊托羊蹄甲生根的特点,采用黄心土、泥炭土、椰糠为主要原料制备育苗基质,能够为藤本囊托羊蹄甲提供最基本的生根环境,满足生根营养需求;育苗基质还添加了芥末粉、灯芯草粉、救必应渣、黄芪渣、烟草渣,其中,烟草渣中的特殊物质如烟碱能杀虫驱虫,有效驱除地下害虫;单宁能杀菌除螨,抑制土传病害,有效防治地下线虫;酚、醛、醌、脂类芳香物质及有机酸,属天然植物生长调节剂,促进作物根系和叶芽生长;单糖、二糖、多糖等糖类物质、纤维素、木质素、腐植酸等在改良土壤中有着独特作用;黄芪有广泛的抗菌作用;救必应能抑制金黄色葡萄球菌,溶血性链球菌及痢疾(弗氏)、伤寒、绿脓杆菌;芥末只是含有丰富的异硫氰酸盐,异硫氰酸盐能抑制微生物生长;灯芯草具有独特的香味,能驱赶昆虫,灯芯草还具有一定的吸附性,能保持根部干爽,避免长时间浸泡引起腐根、烂根;芥末、灯芯草、救必应、黄芪、烟草渣混合使用,使得到的基质具有广谱的抗菌性,能够提高作物的抗病能力,减少腐根、烂根现象。
3、生根粉的主要成分是吲哚乙酸和萘乙酸钠,是植物生长调理剂,本发明通过合理控制生根剂浸泡时间,能够进一步促进根系发育、提高生根率,并缩短繁殖周期、提高繁殖系数。生根剂中添加了Na2SeO3,Na2SeO3中含有硒元素,能够对植物光合速率、气孔导度、蒸腾速率及水分利用效率均有积极作用,能够增进植物体内抗氧化酶的活性,有效减缓叶片衰老,同时调整植物叶层分布,增加光能利用效率,改善叶片内部组织结构。本发明合理的控制了Na2SeO3的添加量,适量的硒可以通过控制植物叶片气孔导度,减少气孔阻力,进而提高CO2通量,达到提升植物光合速率的效果;硒还可以促进植物对Ca、Mg、Mn、Zn、P、K等元素的吸收,这些元素在促进及参与光合色素形成的过程中通常起到重要作用,同时有研究表明硒能够通过影响辅酶因子的吸收和分布对叶绿素合成相关酶的活性起到一定促进作用;气孔的蒸腾作用作为植物水分吸收与运输的主要动力,同时承担着矿质元素营养的运输与吸收以及保持植物体温的重要作用,在一定程度上影响着植物叶片的气体交换,间接地对光合作用起到促进作用。生根剂中还添加了Ce(NH4)2(NO3)6,Ce(NH4)2(NO3)6中含有的铈元素,可以提高植物的叶绿素含量,增强光合作用,促进种子萌发,提高种子发芽率,促进根系发育,促进幼苗生长,增加根系对养分吸收,同时还可以增强幼苗的抗病、抗寒、抗旱能力;铈元素在促进光合作用的同时,能够促进植体对硒元素的吸收。生根剂中还添加了维生素C磷酸酯镁,维生素C磷酸酯镁是维生素C的衍生物,它具有维生素C所有的功能,又克服了维生素C稳定性差的缺陷,且极易被吸收,能够清除自由基,参与植物抗衰老过程,对延缓植物衰老起到重要作用。维生素C磷酸酯镁与Na2SeO3、Ce(NH4)2(NO3)6配合使用时,产生协同作用,维生素C磷酸酯镁能够清除自由基,巩固了Na2SeO3、Ce(NH4)2(NO3)6的作用,同时,维生素C磷酸酯镁中镁元素的存在,能够加强酶促反应,促进光合作用和养分运输,提升吸收率,最终协同促进藤本囊托羊蹄甲的生根。
4、采用本发明的扦插繁殖方法对藤本囊托羊蹄甲进行生根培养,培养后的藤本囊托羊蹄甲栽培成活率高,生长快,提高产量。

S3:扦插:将经步骤S1处理后的插穗用生根剂浸泡30min,浸泡好的插穗扦插于育苗基质中,扦插深度为将插穗插入基质的2/3处,扦插温度控制在20℃之间;使插穗与育苗基质紧密接触;扦插完成后,用1000倍水稀释的多菌灵水溶液淋透,搭盖小拱棚,覆盖塑料薄膜和遮阴网,覆盖率为90%;扦插枝条生根完成之前浇水的频率为5d/次,生根完成之后的浇水频率为8d/次;所述的生根剂由包括以下重量份的原料制备而成:河南神润生物技术科技有限公司的速效生根粉10份、Na2SeO3 0.03份、Ce(NH4)2(NO3)6 0.03份、维生素C磷酸酯镁0.01份;生根剂用蒸馏水稀释至浓度为30g/L。
实验例
分别采用实施例1~9的繁殖方法处理藤本囊托羊蹄甲,不同处理对囊托羊蹄甲生根率、平均根长、平均生根数、综合指标、成活率和生长量的影响,结果如下表所示。其中,综合指标的计算公式为:综合指数=生根率*平均根数*平均根长。
对照组:直接用种子进行栽培,具体步骤如下:
1、基质准备:将基质装在育苗盘中,用500~1000倍水稀释的25%甲霜恶霉灵溶液浇透,然后用薄膜盖2~3天,备用;所述的育苗基质由包括以下重量份的原料制备而成:黄心土6份、泥炭土1份、椰糠1份、芥末粉2份、灯芯草粉2份、救必应渣1份、黄芪渣1份、烟草渣2份。
2、种子准备:采摘成熟种子,种皮未变褐色之前即采摘,遵循“即采即播”原则;
3、播种方法:将步骤2的种子播种于步骤1的基质中,并用基质将种子覆盖,覆盖厚度为种子直径的2~3倍;最后用800倍水稀释的25%甲霜恶霉灵溶液浇透。
分别将实施例1~9和对照组培养的藤本囊托羊蹄甲移栽至土壤中。观测和记录移栽后藤本囊托羊蹄甲的成活率和年生长量。

由表1可知,采用本发明的繁殖方法对藤本囊托羊蹄甲进行无性繁殖,对枝条生根没有出现抑制作用,出根率均在20%以上,最高达到77.5%,说明采用本发明的繁殖方法对藤本囊托羊蹄甲进行无性繁殖培育是可行的。进一步对各项指标采用公式评分法进行评价,综合评分越高说明生根效果越好,其中,综合指标评定分值最高的是实施例2,其次是实施例8,分值最少的是实施例4。由实施例1~9的数据可知,枝条形状上看,四方形插穗移栽成活率最低,其次是中圆柱形,细圆柱形插穗的成活率最高。由实施例1~9与对照组的数据可知,采用实施例1~3、实施例7~9的繁殖方法,采用的插穗枝条为细圆柱形、中圆柱形,藤本囊托羊蹄甲的成活率和生长量指标均优于对照组;采用实施例4~6的繁殖方法,采用的插穗枝条为四方形,四方形插穗本身移栽成活率极低,经本发明的方法处理后,成活率也能达到40%以上,虽然其成活率低于对照组,但其生长量还是大于对照组。说明本发明的繁殖方法可以完全替代直接引种栽培方法,来提高藤本囊托羊蹄甲的各项生长指标,从而提高产量。
以上所述仅为本发明的优选实施例而已,并不用于限制本发明,对于本领域的技术人员来说,本发明可以有各种更改和变化。凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (8)
1.一种藤本囊托羊蹄甲的扦插繁殖方法,其特征在于,包括以下步骤:
S1:选取扦插枝条:选择一年生枝条作为插穗,插穗枝条呈细圆柱形;将每根插穗枝条的顶端剪成平口,底端剪成羊蹄状,将插穗按30~50根为一捆用橡皮筋绑好,用800~1000倍水稀释的多菌灵水溶液消毒5~15min,消毒后的插穗放置在阴凉环境中保湿18~24小时,备用;
S2:制作育苗营养袋:将育苗基质分装在降解免脱无纺布育苗袋中,用500~1000倍水稀释的25%甲霜恶霉灵溶液浇透,然后用薄膜盖2~3天,备用;所述的育苗基质由包括以下重量份的原料制备而成:黄心土3~7份、泥炭土1~3份、椰糠1~3份、芥末粉1~2份、灯芯草粉1~2份、救必应渣1~2份、黄芪渣1~2份、烟草渣1~2份;
S3:扦插:将经步骤S1处理后的插穗用生根剂浸泡,浸泡好的插穗扦插于育苗基质中,使插穗与育苗基质紧密接触;扦插完成后,用800~1000倍水稀释的多菌灵水溶液淋透,搭盖小拱棚,覆盖塑料薄膜和遮阴网,覆盖率为85~90%;所述的生根剂由包括以下重量份的原料制备而成:生根粉5~10份、Na2SeO30.01~0.03份、Ce(NH4)2(NO3)60.01~0.03份、维生素C磷酸酯镁0.01~0.03份。
2.根据权利要求1所述的藤本囊托羊蹄甲的扦插繁殖方法,其特征在于:所述的步骤S1中,每根插穗主枝条长度为15~20cm,侧枝长度为5~8cm,侧枝含有4~6个芽眼。
3.根据权利要求1所述的藤本囊托羊蹄甲的扦插繁殖方法,其特征在于,所述的生根粉为河南神润生物技术科技有限公司的速效生根粉。
4.根据权利要求3所述的藤本囊托羊蹄甲的扦插繁殖方法,其特征在于,所述的生根剂用蒸馏水稀释至浓度为10~30g/L。
5.根据权利要求1所述的藤本囊托羊蹄甲的扦插繁殖方法,其特征在于:所述的步骤S3中,浸泡时间为15~45min。
6.根据权利要求1所述的藤本囊托羊蹄甲的扦插繁殖方法,其特征在于:所述的步骤S3中,扦插深度为将插穗插入基质的2/3处。
7.根据权利要求1所述的藤本囊托羊蹄甲的扦插繁殖方法,其特征在于:所述的步骤S3中,扦插枝条生根完成之前浇水的频率为3~5d/次,生根完成之后的浇水频率为6~8d/次。
8.根据权利要求1所述的藤本囊托羊蹄甲的扦插繁殖方法,其特征在于:所述的步骤S3中,扦插温度控制在15~20℃之间。
