Abstract
本发明公开了一种基于相干探测的多光子超深度组织成像设备及其检测成像方法,其将多光子成像技术与光学相干层析成像技术相结合,实现对被测样本的超深度、高分子特异性成像;设备包括超短脉冲飞秒激光光源、光调制单元、光学相干显微单元,其中光调制单元调制输入激光生成连续谱激光,光学相干显微单元用于对生物样本进行相干探测,系统可以同时探测组织的OCT和SH‑OCT信号,获取组织特定结构和其周围环境的分子微环境信息,结合图像分析方法可以对组织结构与功能进行定量表征。本发明能够反映组织超深度结构与功能信息,具有超深度、强分子特异性识别能力、高时空分辨率、快速成像等优势,兼具组织结构功能信息和分子特异性信息。
Description
技术领域
本发明属于光学显微成像技术领域,具体涉及一种基于相干探测的多光子超深度组织成像设备及其检测成像方法。
背景技术
生物组织的大深度成像能帮助我们更全面地检测组织的各种病理变化。例如,在人体皮肤中,表皮层底部细胞的功能失调是产生黑色素瘤、基底细胞癌、鳞状细胞癌等皮肤癌的主要原因,这些细胞会在癌症第二阶段渗透到皮肤更深的真皮层,最后到达皮下脂肪。研究表明,如果在早期阶段发现癌症并及时治疗,那么存活率可以达到97%。对皮肤表皮层和真皮层的高分辨成像,有助于早期发现和实时监控癌细胞的转移,结合及时的医学治疗,能够有效降低皮肤癌的死亡率。然而,常规二次谐波成像在组织散射等因素的影响下成像深度只能达到表皮层,扩展二次谐波的成像深度对监控和治疗皮肤癌等疾病有重大意义。
光学相干层析成像(OCT)通过探测生物组织内部不同深度的后向散射或反射的光信号和参考光产生的干涉信号,获得样品内部微观结构的层析图像,可以实现微米量级分辨率和毫米量级成像深度的在体成像,在生物医学领域发挥重要作用。由于光学相干层析成像的对比度来源于生物折射率的变化,但由于不同病理状态下由细胞代谢引起的折射率变化不大,因此该成像方式缺乏对分子的特异性识别能力。
发明内容
鉴于上述,本发明提出了一种基于相干探测的多光子组织超深度成像设备及其检测成像方法,其将多光子显微技术与光学相干显微技术相结合,实现对组织样本的超深度成像与定量表征。
本发明采用以下技术方案实现:
一种基于相干探测的多光子超深度组织成像设备,包括飞秒激光光源、光调制单元、光学相干显微单元以及计算机,其中:
所述飞秒激光光源利用钛蓝宝石激光器产生超短脉冲飞秒激光;
所述光调制单元对超短脉冲飞秒激光进行调制,进而将调制后的激光送入光学相干显微单元;
所述光学相干显微单元利用输入的调制光对生物样本进行二次谐波-相干探测,获得毫米级大深度的组织图像;
所述计算机对所得组织图像进行定量分析,获得反映组织病理特征的结构与功能信息。
上述技术方案中,进一步地,所述光调制单元包括:
光隔离器,用于限制激光单方向传输,防止激光反向进入激光器对激光器造成损伤;
起偏器,用于产生特定偏振方向的偏振光;
半波片,用于调整偏振光的偏振方向;
透镜L1,用于将调整偏振方向后的激光耦合进单模光纤;
单模光纤,用于扩展激光的光谱宽度,产生连续谱激光;
透镜L2,用于将连续谱激光扩束为具有适配光斑尺寸的激光。
更进一步的,所述单模光纤产生的连续谱激光的脉冲光谱宽度为45nm。
进一步地,所述光学相干显微单元包括分光镜、二维扫描振镜、透镜L3、二向色分束器、滤光片、彩色滤光片、反射镜、二向色镜、四分之一波片、棱镜对色散补偿器、非线性晶体、OCT光谱仪以及SH-OCT光谱仪,其中:所述分光镜用于将输入的调制光分成两路相同的激光,其中一路依次经非线性晶体、棱镜对色散补偿器、四分之一波片打到二向色镜上并原路返回,另一路经二维扫描振镜和透镜L3打到生物样本上并原路返回,进而分光镜将两路返回的激光进行合成,合成光束经二向色分束器再分成两路激光,其中一路经滤光片送入OCT光谱仪,另一路经彩色滤光片送入SH-OCT光谱仪;所述OCT和SH-OCT光谱仪将输入光的光谱转换成电信号后送至计算机。

本发明的发明原理为:
多光子成像技术(MPM)是一种新型的非线性光学成像技术,该技术以飞秒激光作为光源,基于生物组织在光源作用下受激产生的荧光和二次谐波等非线性光学效应,具有高分辨率、大成像深度、低细胞损伤和可实现三维成像等特点。
二次谐波成像技术(SHG)作为多光子成像技术的一种,利用生物组织中非中心对称物质的非线性光学效应,能对样品进行高分辨率成像。二次谐波成像可以直接成像各向异性的生物结构,例如膜、结构蛋白和微管集合等,还被用于研究组织的生理动力学,例如监测小鼠生长的肿瘤中胶原蛋白的修饰、记录神经元细胞中动作电位的变化等,二次谐波成像逐渐成为一种用于生物医学研究的强大成像手段。
二次谐波信号强度取决于激发光功率的平方,高数值孔径的显微物镜可以增强谐波转换效率。然而,在诸如皮肤等高散射组织样品中,散射降低了二次谐波信号的空间限制;同时,非聚焦区域对二次谐波信号有显著贡献。这些因素降低了二次谐波的成像分辨率,并限制了常规二次谐波成像的成像深度。
本发明方法创造性地将二次谐波成像技术(SHG)与光学相干层析成像(OCT)结合,可以将二次谐波成像的样品结构敏感性与光学相干层析成像的超深度探测特性相结合,兼具了二次谐波成像与光学相干层析成像的优点,可以在无需高数值孔径严格要求的情况下进行组织超深度成像,极大地扩展了二次谐波成像的成像深度,结合图像分析方法对组织结构进行定量表征,有助于对皮肤癌等疾病的诊断。
本发明与现有技术方案相比,具有如下优点:
1.本发明通过多光子成像技术对生物样本进行探测,同时引入频域相干探测技术实现微弱信号沿轴向分布的同步探测,有望极大地提高成像速度。
2.频域相干探测的引入,极大扩展了对组织样本成像的深度,获取组织的大深度微环境信息有助于监控疾病的病理变化过程。
3.多光子成像技术的引入,增强了成像的分子特异性识别能力,能直接成像各向异性的生物结构,例如膜、结构蛋白和微管集合等。
4.本发明能同时探测组织的OCT和SH-OCT信号,获取组织特定结构和其周围环境的分子微环境信息,可以更全面地检测组织的病理变化。
5.可以结合图像分析方法对成像结果进行定量表征,量化组织的结构和功能特性,可以更客观准确地评价组织的病理特征。

本发明基于相干探测的多光子超深度组织成像设备,包括飞秒激光光源、光调制单元、光学相干显微单元以及计算机;光调制单元用于调制输入激光生成超连续谱激光,输入至光学相干显微单元;光学相干显微单元用于对组织样本进行二次谐波-相干探测。
光调制单元包括光隔离器、起偏器、半波片、透镜L1~L2、单模光纤、反射镜;起偏器用于产生特定偏振方向的偏振光;半波片用于调整入射偏振光的偏振方向;透镜L1用于将调整偏振方向后的激光耦合进单模光纤;单模光纤用于扩展激光的光谱宽度,产生连续谱激光;透镜L2用于将连续谱激光扩束为具有适配光斑尺寸的激光;反射镜用于将激光输入到光学相干显微单元;光学相干显微单元为频域光学相干显微系统。
上述基于相干探测的多光子超深度组织成像设备的检测成像方法,通过SH-OCT光谱仪获取组织特定结构的超深层图像,通过OCT光谱仪获取该结构周围微环境信息,可用于全面地检测组织样本的病理变化,所述方法过程如下:
步骤1:飞秒激光光源发射的激光经过光调制单元和光学相干显微单元,获得被测组织样本某点的强度信息;
步骤2:通过二维扫描振镜对被测组织样本进行二维扫描,结合强度信息即可得到组织样本的三维信息;
步骤3:采用图像分析方法对组织结构与功能进行定量表征,可用于定量地评价组织样本的病理特征。
如图1所示,本实施方式中由超短脉冲飞秒激光器(输出光中心波长800nm,相干长度5nm)发出的光,经光隔离器(限制激光单方向传输)后通过起偏器和半波片获得特定偏振方向的线偏振光,该线偏振光通过透镜L1汇聚后,耦合入一个单模光纤(输出光的脉冲光谱宽度为45nm),经单模光纤产生连续谱激光,经透镜L2扩束后,通过反射镜传向分光镜,透镜L1和L2相同,焦距f=20mm;分光镜(分光比90/10,其中90%进入样品臂,10%进入参考臂)将入射光分为参考光和样品光;在样品臂光路中所用透镜L3与光调制单元中的透镜L1和L2相同,焦距f=20mm;在参考臂光路中激光先通过一个非线性晶体(β-BaB2O4)产生SH波,再经过棱镜对色散补偿器(BK7玻璃制成的棱镜对对插)、四分之一波片,通过二向色镜(反射90%的SH波和5%的基波)反射作为干涉的参考光。从参考臂和样品臂返回的光在分光镜处汇合并发生干涉,通过二向色分束器分为两路光,其中一路携带二次谐波通过彩色滤光片经反射镜进入SH-OCT光谱仪,另一路通过滤光片进入OCT光谱仪。在光谱仪中,干涉光经衍射光栅(1200lines/mm)分光,不同的光谱被透镜(焦距f=50mm)聚焦到光电二极管阵列的不同位置上,并由光电二极管阵列采集。
为了实现信号的高灵敏、快速采集,本实施方式采用傅立叶域OCT技术,通过测量来自组织光信号的干涉光谱来检测光的回波时间延迟。探测系统将光谱仪与高速、高动态范围CCD相机结合使用。CCD中光电二极管阵列记录了干涉图样的光谱,可以通过傅立叶变换提取光信号的回波时间延迟。由于所有反射光都被立即测量,因此检测灵敏度得到了极大的提高。
图2展示了对人体皮肤的成像结果;人体皮肤的表皮层含有大量细胞,真皮层主要包含胶原纤维(如图2中的a),成像结果清楚地展示了皮肤的分层结构(如图2中的b),并且可以同时获得表皮层(如图2中的c)和真皮层(如图2中的d)的高分辨率图像;相比于传统成像方式成像深度只能到达真皮层浅层(如图2中的e(I)),本实施方式将成像深度扩展至真皮层深部(如图2中的e(II))。
相干层析探测的引入,极大扩展了对组织的成像深度。例如,在人体皮肤中,表皮层底部细胞的功能失调是产生黑色素瘤、基底细胞癌、鳞状细胞癌等皮肤癌的主要原因,这些细胞会在癌症第二阶段渗透到皮肤更深的真皮层,最后到达皮下脂肪。研究表明,如果在早期阶段发现癌症并及时治疗,那么存活率可以达到97%。对皮肤表皮层和真皮层的高分辨成像,有助于早期发现和实时监控癌细胞的转移,结合及时的医学治疗,能够有效降低皮肤癌的死亡率。本系统可以同时获取皮肤表皮层和真皮层的结构信息,并具有极高的时空分辨率和快速成像等优势,能够全面并及时地检测皮肤的病理变化,有助于早期发现和实时监控癌细胞的转移。结合及时的医学治疗,能够有效降低皮肤癌的死亡率。
在得到图2组织样品图像后,可以定量表征组织的功能与结构特性。如图3所示,表皮层代谢辅酶NADH和FAD相对量的代谢成像以及它们的微环境可以阐明与癌变相关的代谢变化,代谢成像最常用的光学方法是“氧化还原比”,即FAD和NADH的强度之比。氧化还原比对细胞代谢率和血管供氧量的变化敏感,氧化还原比的降低通常表明细胞代谢活性增加。真皮层胶原纤维的取向与排列有序度等结构变化反映了病变对组织结构的重塑,疾病的病理水平影响了胶原纤维的变化程度。这些功能与结构特征为癌症等疾病的生物医学解读提供了客观的、全面的、多角度的信息,有助于对癌症等疾病的诊断。
综合上述实施案例可见,本发明通过多光子与相干探测结合的方式对组织样本进行探测,能实现组织的超深度成像。频域相干探测技术的引入实现微弱信号沿轴向分布的同步探测,有望极大地提高成像速度。多光子成像技术的引入,增强了成像的分子特异性识别能力。多光子成像与相干探测相结合,使得本发明能获取组织超深度结构信息,获取更深层的分子微环境信息用以检测组织的病理变化。结合图像分析方法(如《Mappingmetabolic changes by noninvasive,multiparametric,high-resolution imagingusing endogenous contrast》中的细胞代谢表征,《Rapid three-dimensionalquantification of voxel-wise collagen fiber orientation》中的胶原纤维空间取向定量方法,《Automated quantification of three-dimensional organization offiber-like structures in biological tissues》中的胶原纤维有序度定量方法等)对组织结构与功能进行定量表征,可以客观准确地评价组织的病理特征。本发明具有超深度、强分子特异性识别能力、高时空分辨率、快速成像等优势,兼具组织浅层与深层、定性与定量、结构与功能信息,能够实现对组织病理变化的全面检测。
上述对实施例的描述是为便于本技术领域的普通技术人员能理解和应用本发明。熟悉本领域技术的人员显然可以容易地对上述实施例做出各种修改,并把在此说明的一般原理应用到其他实施例中而不必经过创造性的劳动。因此,本发明不限于上述实施例,本领域技术人员根据本发明的揭示,对于本发明做出的改进和修改都应该在本发明的保护范围之内。
Claims (8)
1.一种基于相干探测的多光子超深度组织成像设备,其特征在于,包括飞秒激光光源、光调制单元、光学相干显微单元以及计算机,其中:
所述飞秒激光光源利用钛蓝宝石激光器产生超短脉冲飞秒激光;
所述光调制单元对超短脉冲飞秒激光进行调制,进而将调制后的激光送入光学相干显微单元;
所述光学相干显微单元利用输入的调制光对生物样本进行二次谐波-相干探测,获得毫米级大深度的组织图像;
所述计算机对所得组织图像进行定量分析,获得反映组织病理特征的结构与功能信息。
2.根据权利要求1所述的基于相干探测的多光子超深度组织成像设备,其特征在于:所述光调制单元包括:
光隔离器,用于限制激光单方向传输,防止激光反向进入激光器对激光器造成损伤;
起偏器,用于产生特定偏振方向的偏振光;
半波片,用于调整偏振光的偏振方向;
透镜L1,用于将调整偏振方向后的激光耦合进单模光纤;
单模光纤,用于扩展激光的光谱宽度,产生连续谱激光;
透镜L2,用于将连续谱激光扩束为具有适配光斑尺寸的激光。
3.根据权利要求2所述的基于相干探测的多光子超深度组织成像设备,其特征在于:所述单模光纤产生的连续谱激光的脉冲光谱宽度为45nm。
4.根据权利要求1所述的基于相干探测的多光子超深度组织成像设备,其特征在于:所述光学相干显微单元包括分光镜、二维扫描振镜、透镜L3、二向色分束器、滤光片、彩色滤光片、反射镜、二向色镜、四分之一波片、棱镜对色散补偿器、非线性晶体、OCT光谱仪以及SH-OCT光谱仪,其中:所述分光镜用于将输入的调制光分成两路相同的激光,其中一路依次经非线性晶体、棱镜对色散补偿器、四分之一波片打到二向色镜上并原路返回,另一路经二维扫描振镜和透镜L3打到生物样本上并原路返回,进而分光镜将两路返回的激光进行合成,合成光束经二向色分束器再分成两路激光,其中一路经滤光片送入OCT光谱仪,另一路经彩色滤光片送入SH-OCT光谱仪;所述OCT和SH-OCT光谱仪将输入光的光谱转换成电信号后送至计算机。
5.根据权利要求4所述的基于相干探测的多光子超深度组织成像设备,其特征在于:所述非线性晶体用以产生二次谐波,经过非线性晶体的激光通过二向色镜反射作为干涉的参考光与经过生物样本的激光发生干涉,所述棱镜对色散补偿器由棱镜对对插组成,用于补偿色散。
6.根据权利要求1所述的基于相干探测的多光子超深度组织成像设备,其特征在于:所述光学相干显微单元采用的是频域光学相干显微系统。
7.根据权利要求1所述的基于相干探测的多光子超深度组织成像设备,其特征在于:所述超短脉冲飞秒激光的中心波长800nm,相干长度5nm。。
8.如权利要求1-7任一项所述基于相干探测的多光子超深度组织成像设备的检测成像方法,其特征在于:通过SH-OCT光谱仪获取组织的超深度图像,通过OCT光谱仪获取该组织周围的微环境信息,采用图像分析方法对组织结构与功能进行定量表征。
