Background
In life, various factors of continuous contact between a human body and the outside, including respiration, external pollution, radiation irradiation and the like, continuously generate free radicals in the human body. Oxygen free radicals are active harmful substances released in the oxidation process in a human body, and randomly grab electrons of other molecules in the body to destroy the structures of cells, DNA, RNA and proteins, so that the functions of cell tissues and organs in the body are reduced, the function of an immune system in the body is reduced, and various diseases are caused and even die. Recent studies have been increasingly conducted to show that anti-oxidation is an important means for preventing aging because excessive free radicals produced by the human body decompose cells and tissues, thereby affecting the normal metabolic functions of the human body and causing various health problems.

Antioxidant Activity test
DPPH is also called 1, 1-diphenyl-2-trinitrophenylhydrazine, is a very stable free radical with a nitrogen center, and the stability of DPPH is that due to the steric hindrance of 3 benzene rings with resonance stabilizing effect, unpaired electrons on the nitrogen atom in the middle cannot play an electron pairing effect. The absolute ethanol solution of DPPH is purple, has a maximum absorption peak at the wavelength of 517nm, and the absorbance and the concentration are in a linear relation. When the free radical scavenger is added, DPPH can be combined or replaced, so that the number of free radicals is reduced, the absorbance is reduced, the color of the solution is lightened, and the capability of scavenging the free radicals can be evaluated according to the free radical scavenger. Namely, the antioxidant capacity was calculated by measuring the DPPH-removing effect of the sample at a wavelength of 517 nm.
DPPH test solution: dissolving DPPH solid 1mg in 24mL of 95% absolute ethanol, performing ultrasonic treatment for 5min, and shaking. The DPPH solution is diluted with 0.5mL of 95% absolute ethanol to make the absorbance between 0.6 and 1.0. Sample solution of antioxidant composition: samples of the antioxidant compositions of examples 1-3 and comparative examples 1-3 were each made up to a concentration of 1mg/mL using 95% absolute ethanol solvent.
Adding 1.0mL of DPPH solution into a cuvette, adding 0.5mL of 95% absolute ethanol, diluting and mixing, and determining A value as A0(A0Must be between 0.8 and 1.0). mu.L of 95% absolute ethanol was taken in a cuvette, and 100. mu.L of a sample solution of the antioxidant composition (examples 1 to 3 and comparative examples 1 to 3) was added thereto, and 1.0mL of DPPH solution was further added thereto so that the total volume of the mixed reaction solution became 1.5mL, and the A value was measured. The average value was calculated in triplicate for each sample.
DPPH clearance calculation formula:
Figure BDA0003136453860000111
(A0is the value without the addition of the sample of antioxidant composition; a is the value after addition of the sample of the antioxidant composition)
The antioxidant composition free radical scavenging data is as follows:
DPPH-free radical scavenging data for antioxidant compositions
Figure BDA0003136453860000112
Figure BDA0003136453860000121
As can be seen from the test data in the above table, comparative example 3, in which the radical scavenging rate test was carried out without adding any of the cosmetic compositions, apparently did not have any radical scavenging ability; while examples 1-3 added incremental amounts of antioxidant compositions, the DPPH-radical clearance data was also increasing; the content of the antioxidant composition contained in example 1 is low, but the clearance rate is remarkably different from that of comparative example 3 without adding any antioxidant composition, which shows that the added antioxidant composition can play a good antioxidant role; under the condition of the same basic formula, the siegesbeckia pubescens Makino extract and the lycopus sieboldii fruit extract in the antioxidant composition are respectively added in the comparative example 1 and the comparative example 2, compared with the comparative example 3 (without adding any antioxidant composition), the DPPH-free radical clearance rate data is obviously different, and the fact that the antioxidant effect can be achieved by independently adding the two is shown. Compared with the DPPH and free radical scavenging rate data of example 2 (added with the antioxidant composition), the antioxidant composition obviously has better DPPH and free radical scavenging capacity than the antioxidant composition added with the antioxidant composition and has independent effect, and the synergistic effect of the antioxidant composition and the DPPH and free radical scavenging capacity is proved to be obvious. In conclusion, the antioxidant composition has an antioxidant effect.
