However, pharyngeal swab and nasopharyngeal swab sampling are both inadequate: 1. there is an increased risk of infection: the sampling staff and the patient need to be in close contact, most viruses are infectious, and the operation greatly increases the risk of infection of the staff; 2. the virus amount is small, the false negative rate is high: (1) because the infected part is often the lower respiratory tract, the pharynx is only the channel of the air exhaled by the lung, the virus in the lung is adhered to the mucous membrane surface of the pharynx when the air is exhaled, the virus amount is less, and the virus can be wiped off by a swab less; (2) the virus excretion of a patient is sometimes fluctuating, that is, some time intervals excreted more and some time intervals excreted less, and the conventional sampling method can only take a sample at a certain time point, and the time point is likely to fall into the time interval with less virus excretion, so that the sample can not be detected due to too little virus content. This is the important reason for the high false negative rate in the clinic at present.
