Description
Technical Field
The invention relates to the technical field of water treatment, in particular to Bacillus cereus (WF 01) and application thereof in removing suspended matters in water.
Background
The microbial flocculant is a substance with flocculation activity produced by microorganisms, and the main chemical components of the microbial flocculant are macromolecular substances such as polysaccharide, protein, lipid, nucleic acid and the like, so that the microbial flocculant has good flocculation and precipitation performance due to the specific structure of the macromolecular substances. Microbial flocculants have been reported to be used in the treatment research and engineering practice of various domestic wastewater, industrial wastewater and various heavy metal wastewater. Compared with traditional chemical flocculants such as inorganic high molecular polymers (polyaluminium chloride, polyaluminium sulfate or iron) and organic high molecular compounds (polyacrylamide), the microbial flocculant has the advantages of safety, no toxicity, no secondary pollution and the like, so that the development of the novel microbial flocculant has important value for removing suspended matters in water bodies.

Application of Bacillus cereus (WF 01) in removing suspended matters in water body is provided.
An application of a fermentation liquid of Bacillus cereus (Bacillus cereus) WF01 in removing suspended matters in a water body is disclosed, wherein the culture method of the fermentation liquid comprises the following steps:
and inoculating the bacillus cereus WF01 into an improved LB culture medium for culture to prepare WF01 fermentation liquor.
Wherein, the improved LB culture medium comprises the following raw materials:
10g/L of glucose, 10g/L of peptone and 5g/L of yeast extract powder; the pH is 7.0-7.5.
Further, the conditions for culturing Bacillus cereus WF01 in the modified LB medium were: cultured at 37 ℃ for 72 h.
Example (b):
confirming whether the bacillus cereus WF01 has the efficacy of removing the suspended matters in the water body:
1) bacillus cereus WF01, which is separated from soil and has the preservation number of (CCTCC NO:2021207), extracting the total DNA of WF01, determining the 16S rDNA sequence, constructing an evolutionary tree by using MEGA6.0, determining the species of the evolutionary tree, and identifying the strain WF01 as Bacillus cereus (Bacillus cereus);
inoculating bacillus cereus WF01 into an LB culture medium, wherein the LB culture medium comprises the following raw materials:
10g/L of glucose, 10g/L of peptone and 5g/L of yeast extract powder; the pH is 7.0-7.5;
culturing at 37 deg.C for 72h to obtain WF01 fermentation liquid;
culturing Bacillus subtilis (without flocculation activity) with the same culture medium under the same culture conditions to obtain a fermentation liquid as one of control groups;
2) comparison of flocculation effects of WF01 fermentation broth
Respectively weighing 0.3g of processed (dried, ground and sieved by a standard sieve of 80 meshes) diatomite (purchased), bentonite (purchased) and loess (collected from lion mountain of Huazhong agriculture university), placing in a 250mL triangular flask, adding distilled water to 100mL, and stirring uniformly to obtain suspension;
then adding 1mL WF01 fermentation broth, 1mL Bacillus subtilis fermentation broth, 10ppm Polyacrylamide (PAM), 10ppm polyaluminium chloride (PAC), 10ppm polyferric chloride (PFC), and 0.1mL 1% CaCl2Adjusting the pH value of the metal coagulant aid to 7.0-7.5, respectively measuring the flocculation activity of the metal coagulant aid after reacting for 20min, and comparing the flocculation effects of WF01 on non-flocculation microorganisms (bacillus subtilis) and 3 traditional flocculants on different flocculation systems;
3) method for comparing flocculation effect
Taking the suspension liquid after flocculation reaction, measuring OD value at wavelength of 550nm, standing for 20min, and taking the suspension liquid without flocculant as a control group;
flocculation activity ═ A-B)/(Ax 100%
Wherein: a is the OD value of the control group suspension at the wavelength of 550nm, and B is the OD value of the experimental group suspension at the wavelength of 550 nm;
compared with other traditional flocculants, the WF01 fermentation liquor has good flocculation activity on diatomite, bentonite and loess, has no difference compared with the traditional 3 flocculants, and even has more advantages to a certain extent, so that the microbial flocculant has potential wide applicability.
Although embodiments of the present invention have been shown and described above, it is understood that the above embodiments are exemplary and should not be construed as limiting the present invention, and that variations, modifications, substitutions and alterations can be made to the above embodiments by those of ordinary skill in the art within the scope of the present invention.
