Abstract

Technical Field
The invention belongs to the technical field of semen collection, and particularly relates to an electric stimulation semen collection method for yaks.
Background
Artificial insemination is one of the important means of modern biotechnology, can exert excellent breeding value of male livestock to the maximum extent, improve the reproduction rate, and is widely applied to animal husbandry.
In the prior art, the common methods for animal semen collection mainly comprise a massage method and a false negative method, the two methods are commonly applied to semen collection of livestock such as cattle, sheep, pigs, chickens and the like, but the two semen collection methods are difficult to apply to most wild animals with strong wildness, violent temperament and small galleries and easy fright. With the development of the technology, people invent that the electrical stimulation method is applied to the semen collection work of male geese, and then the electrical stimulation method is widely applied to the artificial semen collection of other wild animals. Therefore, the electrical stimulation semen collection method has important significance for protecting and utilizing wild animal resources.
The problem of yak semen collection cannot be solved all the time due to the influences of factors such as strong wildness of wild yaks, hard living geographic environment and the like. In the prior art, after domestication of yaks in a Qinghai Datong breeding cattle farm in China, yak semen is successfully collected by adopting a pseudo-vaginal method and is prepared into tubule frozen semen. The artificial vagina method requires that yaks are well domesticated before artificial semen collection, and meanwhile, female yaks in estrus are matched, so that early work is very complicated. Therefore, an effective collection method for semen collection of wild yaks is needed.
Disclosure of Invention
The invention aims to provide an electric stimulation semen collection method for yaks, which is used for solving the technical problem that no method capable of effectively collecting semen of yaks exists in the prior art.
In order to achieve the purpose, the invention adopts the following technical scheme:
a method for electrically stimulating and semen collecting of yaks, which comprises the following steps:
preparing before semen collection:
selecting healthy, non-hereditary diseases, non-reproductive defects and adult yaks aged 4-8 years;
fixing the yak in a roadway or a semen collection platform, and fastening the head and the back ankle of the yak by using a rope;
shearing the abdominal hair of the male yaks by using a hair scissors, cleaning the scrotum and the periphery of the scrotum of the male yaks by using distilled water, simultaneously cleaning the inside of the scrotum, and then wiping water;
electrical stimulation semen collection:
switching on a power supply of the electrical stimulation semen collector, and regulating the voltage to 10V by a voltage regulating knob;
inserting the stimulating electrode bar coated with vaseline or soap water into rectum of yak to an insertion depth of 40-45 cm;
poking the inching switch to stimulate the sacral nerve of the male yak, simultaneously controlling the output voltage to enable the initial voltage to be 10V, the voltage rising amplitude to be 2V each time, the time for continuously outputting the voltage under each level of voltage to be 6-10s, the interval time to be 3-8s, circularly executing once, then rising the voltage by each level, and generally when the voltage rises to 24-30V, the semen of the yak can be collected.
Further, before a power switch of the electrical stimulation semen collector is turned on, a control switch of the electrical stimulation semen collector is turned off, the control mode is selected to be manual, and the voltage gear is selected to be 50V.

The present invention will be further described with reference to preferred embodiments thereof. The examples are given solely for the purpose of illustration and are not intended to limit the scope of the invention.
Examples
First, it should be noted that the working principle of adopting the electrical stimulation method for semen collection in this embodiment is as follows: the autonomic nervous system innervates the male with an erection and ejaculation, which are innervated by parasympathetic and sympathetic nerves of the sacral spinal cord, respectively. The yak ejaculation can be divided into two processes: the first is the expulsion of semen into the urogenital tract, which is controlled by the sympathetic system, and the second is the ejection of semen out of the urogenital tract, which is parasympathetic. The electric stimulation semen collection of the yak is realized by directly stimulating the ejaculation center at the waist recommendation part by adopting an electrode. The electric stimulation semen collection method is based on physiological principles such as the reproductive physiological process and characteristics of the yak, combines the modern microelectronic technology, sets working mode and operating parameters by a pulse current generator, generates cooperative spectrum pulse under the control of a host CPU, transmits the cooperative spectrum pulse to an electrode (load) inserted into the rectum of the yak by a cable to generate an electric (current) field, and directly stimulates the nerve endings of a sperm line of the yak and the ampulla of the vas deferens by generating the pulse electric (current) field which stimulates the yak regularly from weak to strong to simulate the natural semen collection process, so that the nerve excitation is transmitted to the semen center of the yak to cause the excitation of the semen center of the yak, the muscle contraction, the penis erection and the smooth muscle contraction to collect the semen of the yak, and the aim of collecting the semen of the yak is achieved.
Next, it should be noted that the structure of the electrical stimulation semen collector adopted in this embodiment is: the electro-stimulation semen collector comprises a main machine (the main machine is formed by assembling a 12V direct-current battery, a pulse current generator, an automatic operation control button and a manual operation control table), pulse current output current, electrodes, a charging power supply, a handheld control button (namely a manual switch) and a semen collection funnel.
Specifically, this embodiment provides a yak electrical stimulation semen collection method, including:
preparing before semen collection:

it should be noted that the semen collection site selected in this embodiment is a site that is dry in terrain, wide, sanitary, and quiet in environment, and is close to a production laboratory. The electrical stimulation semen collection fixing frame can be designed and installed in a semen collection field according to the size and safety of the yak body, so that the semen collection bull is properly fixed, the safety of people and livestock is ensured, and semen collection is facilitated.
Shearing the abdominal hair of the male yaks by using a hair scissors, cleaning the scrotum and the periphery of the scrotum of the male yaks by using distilled water, simultaneously cleaning the inside of the scrotum, and then wiping water;
the method is characterized in that the subtraction of the abdominal hair of the yak has the following effects: the semen collection device is convenient for observing the discharged semen, collecting the semen and preventing the dust in the abdominal hair from polluting the semen.

switching on a power supply of the electrical stimulation semen collector, and regulating the voltage to 10V by a voltage regulating knob;

When a worker toggles the electric switch to manually control the switch to keep the switch at the position of the jog switch, the duration time of the jog position is the time of continuously outputting voltage under each level of voltage, after fingers are loosened, the jog switch can automatically rebound to the middle closing position, at the moment, current cannot pass through the electrode bar, namely the interval time, and the yak is in the electrical stimulation intermission period.
Of course, it can be understood that the collection of the yak semen when the voltage is raised to 24-30V is only one embodiment of the present invention, and the interval is the output voltage required for collecting the yak semen under normal conditions. However, in practical application, due to the difference of the collected objects, some yaks may realize semen discharge at about 12V, and some yaks may need about 40V to discharge semen, and the yaks can be specifically adjusted according to the difference of the collected objects without limitation.
As an alternative embodiment, further, in order to avoid harming the body of the yak, the output voltage of the stimulation shocker should be limited within a range not higher than 46V, and the interval time should be limited within a range not lower than 3 s.
As an alternative embodiment, the sperm collection frequency of said yak is not more than 2 times per week. Preferably, the sperm collection frequency of the yaks is 1 time per week. The reason is that: after the same yak is used for semen collection twice a week, the volume and the quality of the semen of the second time are poorer than those of the semen of the first time, and meanwhile, the yak recovers the health and the speed of producing the semen is slower.
As an alternative embodiment, when the liquid discharged from the penis of the electrically stimulated male yak is non-opalescent, the non-opalescent liquid is discarded until the liquid discharged from the penis is opalescent, and semen collection is performed. Wherein the non-opalescent liquid is prostatic liquid, is generally transparent liquid, is mixed with a small amount of sperms, does not meet the requirement of sperm collection, and therefore, should be discarded.
As an alternative embodiment, when the electrode inserted into the rectum of the yak feels fecal, the stimulation electrode rod is pulled out and the fecal is cleaned, and then the stimulation electrode rod is inserted into the rectum of the yak again for electrical stimulation.
Application example
The method for collecting the semen of the yak is applied to a Longri livestock farm in Sichuan province, 30 times of semen collection is carried out on the yak in the livestock farm, and more than 2.5 million of tubule frozen semen of the yak is produced. As can be seen from the tables 1 and 2-1 to 2-3, the yak electrical stimulation semen collection method successfully collects a plurality of yak semen with the semen volume of more than or equal to 5ml in 2019 and 2020, and the qualified probability of successfully freezing and storing each semen is up to more than 90% in 2020. Therefore, by adopting the yak electrical stimulation semen collection method, the yak semen can be efficiently obtained and frozen for storage for subsequent use.

electrical stimulation semen collection:
switching on a power supply of the electrical stimulation semen collector, and regulating the voltage to 10V by a voltage regulating knob;
inserting the stimulating electrode bar coated with vaseline or soap water into rectum of yak to an insertion depth of 40-45 cm;
poking the inching switch to stimulate the sacral nerve of the male yak, simultaneously controlling the output voltage to enable the initial voltage to be 10V, the voltage rising amplitude to be 2V each time, the time for continuously outputting the voltage under each level of voltage to be 6-10s, the interval time to be 3-8s, circularly executing once, then rising the voltage by each level, and generally when the voltage rises to 24-30V, the semen of the yak can be collected.

The yak electrical stimulation semen collection method according to claim 1, wherein before a power switch of the electrical stimulation semen collection device is turned on, a control switch of the electrical stimulation semen collection device is turned off, a control mode is selected to be manual, and a voltage gear is selected to be 50V.
