Abstract

Description
技术领域
本发明涉及生物技术领域,特别是涉及一种α-酮戊二酸在制备药物中的用途。
背景技术
骨关节炎(osteoarthritis osteoarthritis osteoarthritis osteoarthritis,OA)是一种常见的慢性关节疾病。骨关节炎患病率高,列全球致残疾病第六位。预计到2020年,会升至致残疾病第四位。发病多见于中老年人,女性多于男性。多发于负重大的膝关节、踝关节、脊柱及频繁活动的手指关节等部位。骨关节炎主要病理改变是关节软骨的退行性变,临床症状通常包括关节疼痛、压痛、僵硬和功能受限骨关节炎,并伴有滑膜炎和继发性骨赘形成。骨关节炎导致关节软骨破坏,以及软骨下骨硬化。这些病理改变和病人关节的疼痛症状密切相关,并造成关节活动障碍和关节畸形,严重影响了患者的生活质量,引起或加剧其他的合并症,降低了预期寿命。该病发展到晚期,关节软骨大面积剥脱,软骨下骨直接受力,导致关节功能丧失并最终致残。骨关节炎的发病原因尚不完全清楚,它的发生发展是一种长期、慢性、渐进的病理过程,年龄、肥胖、机械损伤和遗传是主要的危险因素9-11。每年有2亿多人因为骨关节炎的相关症状而求医。随着老年人口迅速增多,骨关节炎对社会经济的影响将更为显著。
目前临床上大多数的治疗方法如药物止痛、物理治疗和关节腔注射等只能缓解疾病的症状,并不能阻止关节软骨破坏和继发骨赘的形成与发展。关节软骨由软骨细胞和软骨基质组成。软骨细胞是关节软骨组织中存在的唯一的细胞,在发挥维持、重塑和修复软骨这一无血管组织的功能方面发挥重要作用。软骨细胞针对不同种类,强度的生物力学刺激(剪切力,应力和压力)信号进行整合并作出反应,通过合成与分解因子的分泌,重塑细胞外基质以适应力学刺激。在这一过程中,软骨细胞通过合成(II型胶原和蛋白聚糖的生成)与分解过程(产生降解基质成分的各种酶)的平衡,实现软骨组织基质的动态平衡。总而言之,软骨组织中合成和分解过程的失衡导致了骨关节炎的发生。
α-酮戊二酸(α-ketoglutarate,简称α-KG)是三羧酸循环中的作用中间产物,也是合成多种氨基酸、蛋白质,例如谷氨酰胺(Glutamine,Gln)下游代谢产物。研究表明,Gln来源的a-酮戊二酸通过组蛋白去甲基化酶JMJD3调节巨噬细胞向M2表型分化,抑制巨噬细胞向M1表型分化,同时,α-KG抑制了p-IKKα/β的形成,抑制诱导炎症因子的表达。
目前,尚无α-KG在治疗关节炎中的报道,因此,提供一种包含α-KG的药物,对于骨关节炎的临床治疗有极大的潜在应用价值。
发明内容
鉴于以上所述现有技术的缺点,本发明的目的之一在于提供一种α-KG在制备药物中的用途,所述药物用于:1)治疗骨关节炎及相关疾病;和/或,2)抑制软骨细胞的分解表型;和/或,3)促进软骨细胞的合成表型;和/或,4)促进皮肤毛囊再生,用于解决现有技术过程中的问题。
本发明的另一目的在于提供一种药物组合物。
为实现上述目的及其他相关目的,本发明第一方面提供一种α-KG在制备药物中的用途,所述药物用于:1)治疗骨关节炎及相关疾病;和/或,2)抑制软骨细胞的分解表型;和/或,3)促进软骨细胞的合成表型;4)促进皮肤毛囊再生。
可选地,所述α-KG用于抑制骨细胞的NF-κB信号通路的激活和/或传导。
可选地,在白介素-1β(IL-1β)刺激的环境下,所述α-KG调控所述软骨细胞的分解表型和/或合成表型。
可选地,所述IL-1β刺激所述软骨细胞。
可选地,所述IL-1β的浓度为1-15ng/ml。
可选地,所述α-酮戊二酸在所述药物中的形式为二甲基-α-酮戊二酸。
可选地,所述药物进行关节腔注射。
本发明的第二方面提供一种药物组合物,所述药物组合物包括α-KG。
可选地,所述药物组合物还包括药学上的可接受的载体。
如上所述,本发明提供了一种α-KG在制备药物中的用途和一种药物组合物,根据本发明提供的用途,所述药物用于:1)治疗骨关节炎及相关疾病;和/或,2)抑制软骨细胞的分解表型;和/或,3)促进软骨细胞的合成表型;和/或,4)促进皮肤毛囊再生。本发明还利用在IL-1β的刺激环境下,研究所述α-KG对软骨细胞的表型的调控作用。此外,本发明还模拟体内骨关节炎模型,通过关节腔注射α-KG能明显抑制骨关节炎的进展,因此,根据本发明提供的药物组合物能保护软骨组织,阻止骨关节炎进展,对于骨关节炎的临床治疗和预防有极大的潜在应用价值。此外,本发明提供的药物可以促进皮肤毛囊显著增加,对于脱发的治疗和改善也具有极大的潜在应用价值。其他的特征、优势和效果可以参考本发明权利要求书和说明书公开在内的内容。

本发明提供本发明第一方面提供一种α-KG在制备药物中的用途,所述药物用于:1)治疗骨关节炎及相关疾病;和/或,2)抑制软骨细胞的分解表型;和/或,3)促进软骨细胞的合成表型;和/或,4)促进皮肤毛囊再生。
本发明公开的一具体实施例中,所述α-KG例如用于抑制骨细胞的NF-κB信号通路的激活和/或传导,进一步地,所述骨细胞例如可以为软骨组织中的软骨细胞。所述α-KG的浓度例如为3~20mM,进一步地,基于提高所述有效抑制骨关节炎的的观点,所述α-KG的浓度为3~10mM,例如3mM、5mM、7mM、8mM。进一步地,在α-KG在药物中的形式为二甲基-α酮戊二酸,进入细胞以进行调控作用。
本发明公开的一具体实施例中,所述α-KG在IL-1β刺激的环境下,调控所述软骨细胞分解表型和/或分解表型,进一步地,所述IL-1β用于刺激软骨细胞,所述IL-1β浓度为1-15ng/ml,例如1ng/ml、5ng/ml、10ng/ml。
本发明第二方面提供一种药物组合物,所述药物组合物包括α-KG。
所述药物组合物还可以包括药学上可接受的载体。所述载体可以包括各种赋形剂和稀释剂,这些载体本身并不是必要的活性成分,且施用后没有过分的毒性。另一方面,在施用所述药物组合物所需要考虑的剂量应当取决于施用频率和模式,受治疗的受试者的年龄、性别、重量和一般状况,治疗的状况和严重性,以及给药途径而异,待治疗的任何伴随性疾病以及对于本领域技术人员显而易见的其他因素。同时,根据受治疗者的情况和其他病理状况,包含本发明的药物组合物可以与一种或多种其他的治疗活性化合物或物质组合施用或应用,本发明公开的一具体实施例中,所述药物可以进行关节腔注射。
本发明还进行IL-1β刺激抑制了软骨细胞,软骨细胞内Gln的代谢情况的实验,在本发明的一个实施例中,IL-1β刺激软骨细胞,抑制了软骨细胞Gln代谢,减少了软骨细胞内Gln的含量,从而确立了骨关节的与软骨细胞内Gln含量水平的有关。
本发明还进行IL-1β刺激抑制了软骨细胞,软骨细胞内Gln下游代谢产物α-KG的代谢情况的实验,在本发明公开的一具体实施例中,IL-1β刺激软骨细胞,抑制软骨细胞内生成α-KG的基因表达,并且软骨细胞内α-KG的含量变少,从而为补充/回补α-KG可以抑制骨关节炎的进展作出启示。
本发明还进行了α-KG对软骨细胞的分解表型、合成表现,以及对NF-κB信号的影响的实验。在本发明公开的一具体实施例中,通过全基因组测序的方法,检测到了在回补α-KG后,α-KG能明显促进软骨细胞的合成表型,抑制了软骨的分解表型,并通过q-PCR和westernblot的方法验证了这一结果。此外,通过全基因组测序的方法检测到了在回补α-KG后,利用例如在IL-1β刺激的条件下,NF-κB信号通路中很多基因的表达被抑制了,例如NF-κB信号通路中细胞核内P65的表达。因此,从而确立了α-KG可以通过抑制NF-κB信号通路,抑制了炎症的产生和对软骨细胞的表型的调控。
本发明还进行了建立骨关节炎模型的实验,在本发明的一个实施例中,通过关节腔注射α-KG,并通过OARSI定量评分,确立了α-KG能明显抑制骨关节炎的进展。
本发明还进行了皮肤毛囊再生水平的实验,在本发明的一个实施例中,通过皮内注射注射α-KG,并通过HE染色观察,确立了α-KG能明显增加皮肤毛囊,可以用于改善和/或治疗脱发。
以下将通过具体的实施例对对发明进行更为详细的阐述,除非另外说明,本发明中所公开的实验方法、检测方法、制备方法均采用本技术领域常规的分子生物学、生物化学、染色质结构和分析、分析化学、细胞培养、重组DNA技术及相关领域的常规技术。这些技术在现有文献中已有完善说明。
下列实施例中未注明具体条件的实验方法,按照常规方法和条件,或按照商品说明书选择。本发明使用的α-KG,采用二甲基-α-KG,所述二甲基-α-KG进入细胞发挥调控作用,并可以购自市售的产品,其余本发明所用试剂和原料均市售可得。
实施例1小鼠软骨细胞的培养
小鼠软骨细胞取自7d龄C57BL/6小鼠(上海斯莱克动物有限公司),具体操作步骤如下:在无菌手术条件下,小心分离C57小鼠的双侧膝关节软骨,并用0.2%的中性胶原酶(Serva,德国)消化8h,离心获得细胞沉淀后用含10%FBS的改良Eagle培养基(DMEM)(Gibco,美国)培养液重悬细胞并接种于培养皿中,在37℃、5%CO2恒温孵育箱中,用高糖的DMEM培养液(含10%的FBS、100U/mL青霉素和100mg/L链霉素)进行培养。48h首次换液,观察细胞融合率达80%~90%时,用0.25%浓度的胰蛋白酶(Gibco,美国)消化传代,离心后收集细胞重悬,调整浓度接种于培养皿。按此方法换液传代,所获细胞分别记为P1、P2、P3代等。
实施例2IL-1β对包含谷氨酰胺培养液培养的软骨细胞中谷氨酰胺含量水平的影响
1实验方法
(1)标准溶液的配制
准确称量十九种氨基酸标准品,用水配制混合液,使其终浓度均为0.05μg/mL,0.1μg/mL,0.2μg/mL,0.5μg/mL,1μg/mL,2μg/mL,5μg/mL,10μg/mL,20μg/mL,50μg/mL,100μg/mL十一个标准浓度梯度。
(2)样品前处理
取样本涡旋30s,液氮快速冻存,室温溶解淬灭的细胞溶液,涡旋30s,800g离心1min,取上清液至离心管中,放置于干冰上;用500μL甲醇(-80℃)重置细胞,置于液氮中快速冻存,重复涡旋和离心步骤,合并上清液,液氮吹干,加入浓度为1μg/mL的丙氨酸-d4同位素内标1000μL,涡旋混匀,瞬离,取100μL,加入浓盐酸:正丁醇(1:3)60μL,混匀后瞬离(把盖上液滴甩下),65℃恒温放置15min衍生化,瞬离后低于45℃挥干,加入80%乙腈水100μL复溶,混匀直接进样分析。
(3)LC-MS检测程序
色谱条件:色谱柱:ACQU ITYBEH C18色谱柱(2.1×100mm,1.7μm,美国Waters公司),进样量5μL,柱温40℃,流动相A-含0.1%甲酸和0.1%七氟丁酸的乙腈,流动相B-0.1%甲酸水,流速0.2mL/min;梯度洗脱程序如下:0~1.5min,5%A;1.5~2min,5~20%A;2~7min,20~30%A;7~8.5min,30~98%A;8.5~10.5min,98%A;10.5~11min,98~5%A;11~12.5min,5%A。
MS条件:电喷雾电离(ESI)源,正离子电离模式。离子源电压3200V,溶剂温度380℃,锥孔电压20V。采用多重反应监测(MRM)进行扫描。用于定量分析的离子对见下表1。

(4)标准曲线
对标准液的浓度系列分别进行LC-MS检测,以标准品的浓度为横坐标,峰面积比值为纵坐标,以此来考察标准溶液的线性。得到的各氨基酸的线性回归方程见表。相关系数>0.99。

2实验结果
图1显示的是在IL-1β刺激软骨细胞的条件下,软骨细胞内氨基酸含量的变化。如图1所示,在IL-1β处理软骨细胞36h后,将细胞刮下,用LC-MS的方法检测软骨细胞内氨基酸含量的变化发现,在IL-1β处理的条件下,Gln含量的表达由8740.24±584.31ng/107下降至330±150.32ng/107。
实施例3IL-1β对不包含谷氨酰胺培养液培养的软骨细胞的分解表型和合成表型的影响
1实验方法
(1)小鼠软骨细胞的处理
采用实施例1中培养的P1代小鼠软骨细胞,当P1代软骨细胞贴壁生长后,通过10ng/mlIL-1β的无血清的DMEM培养液培养处理。对于无谷氨酰胺处理软骨细胞,先提前12h换无谷氨酰胺的培养液,之后再用IL-1β处理软骨细胞。
(2)实时荧光定量PCR(Real Time q-PCR)分析
1)采用实施例1中培养的P1代小鼠软骨细胞,将培养皿里培养液吸出,用PBS洗一遍。
2)Total RNA的提取
①向匀浆裂解液中加入氯仿(RNAiso Plus的1/5体积量),盖紧离心管盖,用手剧烈振荡15秒,待溶液充分乳化(无分相现象)后,再室温静置5分钟。
②12000g,4℃离心5分钟。
③从离心机中小心取出离心管,此时匀浆液分为三层:无色的上清液、中间的白色蛋白层及带有颜色的下层有机相。吸取上清液转移至另一新的离心管中(切忌吸出白色中间层)。
④向上清中加入等体积的异丙醇,上下颠倒离心管充分混匀后,在15~30℃下静置10分钟。
⑤12000g,4℃离心10分钟。一般在离心后,试管底部会出现沉淀。
3)RNA沉淀的清洗
小心弃去上清,缓慢地沿离心管壁加入75%的乙醇1ml(切勿触及沉淀),轻轻上下颠倒洗涤离心管管壁,12000g,4℃离心5分钟后小心弃去乙醇(为了更好地控制RNA中的盐离子含量,应尽量除净乙醇)。
4)RNA的溶解
室温干燥沉淀2~5分钟,加入适量的RNase-free水溶解沉淀,必要时可用移液枪轻轻吹打沉淀,待RNA沉淀完全溶解后,立即测浓度并逆转录,剩余的于-80℃保存。
5)RNA浓度测定
将(4)中的获得RNA的样品经过Nanodrop 2000仪器测试浓度。
6)反转录反应
将(5)中的RNA的样品进行反转录反应,其中,反转录的反应体系和反应条件见表3。

7)RT-PCR反应
在冰上,将对SYBR Premix Ex Taq、软骨分解和合成基因上下游引物、ROXReference Dye、cDNA template、dH2O加入试管中,混合均匀后,均匀分装至PCR管中,将PCR管放入Q-PCR仪中对产物进行检测,其中,Real Time PCR反应体系和反应条件见表4;Real-time PCR引物序列见表5。

(3)蛋白质印迹(western blot)分析
采用实施例1中培养的P1代小鼠软骨细胞,将其通过包含以下的步骤进行westernblot分析:
①收集蛋白样品:吸去培养液,4℃PBS洗2遍后吸干。每孔中加入100μl细胞裂解液,冰上裂解10min,收集至1.5ml离心管中,10,000转离心10min,收集上清。BCA蛋白定量试剂盒测定蛋白浓度,蛋白与5×上样缓冲液混合后,100℃加热10min。-80℃保存待用。
②电泳:根据蛋白大小配置SDS-PAGE凝胶,每孔加入20μg样品。恒压80伏跑至分离胶后,电压调到120V,根据蛋白marker,蛋白条带分开后,即可停止电泳。
③转膜:剪好PVDF膜和滤纸,甲醇中活化5min,再于转膜缓冲液冲洗待用。从玻璃板中取出凝胶块,用水洗去电泳液。按以下顺序组成转印系统:阴极夹板,海绵,滤纸,凝胶,PVDF膜,滤纸,海绵,阳极夹板。将整个转膜仪放入冰盒中,加入转膜缓冲液,凝胶靠近负极,PVDF膜靠正极。100V,转膜1.5h。
④.封闭:取出PVDF膜,浸泡与5%的脱脂牛奶中,5%的脱脂牛奶用1x TBST配置,500ul tuween 20溶于500ml 1xTBS形成TBST。室温摇床封闭1h。
⑤一抗孵育:封闭过后,倒掉封闭液,1x TBST洗膜,5min/次,洗5次。根据一抗稀释浓度,稀释一抗,之后4°一抗孵育过夜。
⑥二抗孵育:回收一抗,1x TBST洗膜,5min/次,洗5次,二抗室温摇床孵育1h。
⑦扫膜显影:通过化学发光系统曝光扫膜。
(4)细胞核蛋白抽提
采用实施例1中培养的P1代小鼠软骨细胞,将其通过包含以下的方法抽提细胞核蛋白,用于观察NF-KB信号通路P65的变化:
①试剂放置在冰上,混匀。取适量的细胞浆蛋白抽提试剂I,在使用前1min内加入PMSF及蛋白酶抑制剂。取适当量的细胞核蛋白裂解液,在使用前1min内加入PMSF及蛋白酶抑制剂。
②用PBS洗一遍细胞。
③用EDTA溶液处理细胞,并用移液枪吹打细胞。离心收集细胞,吸尽上清,留下细胞沉淀。
④每20μl细胞沉淀加入200μl添加了PMSF的细胞浆蛋白抽提试剂I。
⑤最高速剧烈震荡10s,把细胞沉淀完全分散开。
⑥冰浴10-15min。
⑦加入细胞浆蛋白抽提试剂II 11μl。最高速剧烈震荡5s,冰上放置1min。
⑧最高速剧烈震荡5s,4℃,16,000g,离心5min。
⑨立即吸取上清至一预冷的1.5ml离心管中,即为细胞浆蛋白。
⑩完全吸尽残余的上清,加入50μl添加了PMSF的细胞核蛋白裂解液。
2实验结果
图2显示的是在缺乏Gln的条件下,用IL-1β刺激软骨细胞时,软骨细胞的分解表型和合成表型情况。如图2所示,在无Gln的条件下,IL-1β刺激软骨细胞,经q-PCR检测发现分解表型基因包括MMP3、MMP13、ADAMTS5、NOS2等显著高于Gln存在情况下这些软骨分解表型基因的表达(图2A)。IL-1β刺激软骨细胞24h后,分解表型的蛋白的表达情况和基因的情况表现出一致性,即,无Gln时,分解表型的蛋白表达更高(图2B)。而且在不同的时间段,例如IL-1β刺激6h、12h、24h、36h都发现在无Gln时,软骨分解表型基因,例如MMP3、MMP13、ADAMTS5、NOS2表达比有谷氨酰胺时更高(图2C-F)。在蛋白水平上,经western blot检测也发现无谷氨酰胺时,IL-1β刺激软骨细胞在不同时间点都促进了软骨细胞分解表型基因,例如MMP3、MMP13、NOS2的蛋白表达(图2G)。而对于软骨细胞的合成基因,例如SOX9、COL2A1、ACAN,经q-PCR检测发现在无Gln的条件下,IL-1β刺激12h、24h时,SOX9、COL2A1、ACAN基因的表达降低了(图2H-J),而在6h和36h时无显著差异。这可能是由于在IL-1β刺激后,抑制了谷氨酰胺转运体,在后期有和无谷氨酰胺的差异不是很大。在蛋白水平上,经western blot检测发现在无谷氨酰胺的条件下,在IL-1β刺激软骨细胞36h时,SOX9、COL2A1蛋白的表达比有谷氨酰胺时更低(图2K)。
图3显示的是在IL-1β刺激软骨细胞的条件下,软骨细胞内Gln的代谢情况。如图3所示,在IL-1β刺激软骨细胞的条件下,经q-PCR检测软骨细胞中Gln转运基因的表达情况发现,IL-1β刺激导致Gln转运体例如SLC1A5,SLC38A2、SLC7A5基因的表达降低了,而且具有时间依耐性,刺激时间越长,其表达越低(图3A),进一步地,检测了谷氨酰胺酶GLS和谷氨酰胺合酶GS的表达的情况,经q-PCR和western blot检测发现谷氨酰胺酶GLS和谷氨酰胺合酶GS的表达明显降低了,而且刺激时间越长,表达越低(图3A-B)。
实施例4α-酮戊二酸对包含谷氨酰胺培养液培养的软骨细胞分解表型和合成表型的影响
1实验方法
(1)小鼠软骨细胞的处理
将实施例1中获得的P1代的小鼠软骨细胞,分别通过空白的DMEM培养液、包含10ng/ml IL-1β的DMEM培养液,以及包含10ng/ml IL-1β和7mMα-KG的DMEM培养液培养处理,形成相应的ctrl组、IL-1β组、IL-1β+αKG组。
(2)实时荧光定量PCR(Real Time q-PCR)分析
将ctrl组、IL-1β组、IL-1β+αKG组分别采用与实施例3中q-PCR实验相同的实验条件,进行q-PCR检测。
(3)蛋白质印迹(western blot)分析
将ctrl组、IL-1β组、IL-1β+αKG组分别采用与实施例3中western blot实验相同的实验条件,进行进行western blot检测。
2实验结果
图4显示的是在IL-1β刺激软骨细胞的条件下,软骨细胞内Gln下游代谢产物α-KG的含量情况。如图4所示,与ctrl组相比,IL-1β组的软骨细胞内α-KG的含量明显下降,即,说明在IL-1β刺激软骨细胞后,软骨细胞内α-KG的含量降低(图4A),此外,进一步地,通过q-PCR检测了形成α-KG相关的基因例如DLUD、IDH1、IDH2、IDH3a、IDH3b、IDH3g、GOT1、GOT2、GPT1、GPT2、CCBL1、CCBL2的表达情况。结果发现IL-1β刺激软骨细胞36h后,这些基因的表达都被抑制了(图4B),而且具有时间依赖性,IL-1β刺激的时间越长,其表达量越低(图4C)。
图5显示的是ctrl组、IL-1β组、IL-1β+αKG组的软骨细胞的分解表型的情况。如图5所示,通过全基因组测序的方法(全基因组测序服务由上海博豪生物提供),检测了全基因组的表达,与ctrl组相比比,IL-1β组的软骨细胞的分解表型,例如金属蛋白酶MMP2、MMP3、MMP8、MMP9、MM912、MMP13、MMP27以及蛋白聚糖酶ADAMTS5、ADAMTS7、ADAMTS12、ADAMTS15、ADAMTS16、ADAMTS17和诱导软骨凋亡的基因NOS2的表达增加(图5A),与此同时,IL-1β+αKG组的软骨细胞的分解表型,如金属蛋白酶MMP2、MMP3、MMP8、MMP9、MM912、MMP13、MMP27、蛋白聚糖酶ADAMTS5、ADAMTS7、ADAMTS12、ADAMTS15、ADAMTS16、ADAMTS17和诱导软骨凋亡的基因NOS2的表达被抑制了。为了进一步验证该全基因组测序的结果,IL-1β,IL-1β+α-KG处理软骨细胞24H后,通过q-PCR的检测方法,分析了软骨分解表型基因例如MMP3、MMP13、ADAMTS5、NOS2的表达情况,结果发现α-酮-戊二酸具有能够逆转IL-1β的作用,抑制了软骨分解表型基因MMP3、MMP13、ADAMTS5、NOS2的表达(图5B)。同时,通过western blot的检测方法,分析了其在蛋白水平上的表达情况,结果同样发现α-KG软骨抑制分解表型基因例如MMP3、MMP13、ADAMTS5、NOS2蛋白的表达(图5C)。而且在不同的时间段,例如6h、12h、24h、36h回补α-KG时,通过q-PCR和western blot的检测方法发现在每个时间点,回补α-KG都能抑制的软骨细胞的分解表型基因MMP3、MMP13、ADAMTS5、NOS2基因和蛋白的表达(图5D-H)。
图6显示的是ctrl组、IL-1β组、IL-1β+αKG组的软骨细胞在处理24h后,软骨细胞的合成表型的情况。如图6所示,通过全基因组测序的方法,检测了全基因组的表达,与ctrl组相比,IL-1β组的软骨细胞的合成表型基因,如SOX9、COL2A1、ACAN和一些软骨的其他胶原,如COL9A1、COL9A2、COL9A3、COL11A1、COL11A2的表达下降,且软骨去分化基因COL3A1的表达的增加,与此同时,IL-1β+αKG组的软骨细胞的合成基因的表达,如SOX 9、COL2A1、ACAN和一些软骨的其他胶原,如COL9A1、COL9A2、COL9A3、COL11A1、COL11A2等的表达增加,即,说明在回补α-KG后,α-KG具有能够逆转IL-1β的作用(图6A)。为了进一步验证该全基因组测序的结果,通过q-PCR的检测方法,分析了IL-1β+αKG组的软骨细胞的软骨细胞的合成基因,例如SOX9、COL2A1、ACAN的表达情况,结果发现在IL-1β刺激软骨细胞的同时,回补α-KG能够促进软骨细胞的合成基因SOX9、COL2A1、ACAN的表达(图6B-D)。同时,通过western blot的检测方法,分析了IL-1β+αKG组的软骨细胞在蛋白水平上的表达情况,结果同样发现回补α-KG能够促进软骨细胞的合成基因,例如SOX9、COL2A1蛋白的表达,且在6h、12h、24h、36h都有相似的结果(图6E)。
图7显示的是ctrl组、IL-1β组、IL-1β+αKG组的软骨细胞的NF-KB信号通路的激活情况。如图7所示,通过全基因组测序的方法,检测了全基因组的表达,与ctrl组相比,IL-1β组的软骨细胞的NF-KB信号通路相关的基因,如Nfkbia、Relb、Lbp、Pik3r1、Tnfsf11、Birc3、Traf2、Stat1、MMP2、xiap、Ccl2、Ccl12、IL-1β、IL-1α等的表达被抑制(图7A),为进一步验证是否无谷氨酰胺时能够激活NF-KB信号通路,将软骨细胞无谷氨酰胺培养12h后,接着用IL-1β刺激0min、15min、30min、60min,结果发现无谷氨酰胺时,NF-KB信号通路中P65的表达明显增加了(图7B),与此同时,观察了IL-1β+αKG组的软骨细胞在0min、15min、30min、60min时分离细胞核内NF-KB信号通路中P65的表达情况,结果发现在0min、15min、30min、60min时,IL-1β+αKG组的软骨细胞的细胞核内NF-KB信号通路中P65的表达明显被抑制了(图7C)。即说明在IL-1β刺激软骨细胞时,回补α-酮戊二酸能抑制NF-κB信号通路。
实施例5小鼠膝关节骨关节炎模型(DMM)
1实验方法
(1)构建小鼠膝关节DMM模型
从上海斯莱克实验动物有限公司购买雄性C57BL/6小鼠(n=30),并饲养至8周龄,其中,动物实验操作方法由同济大学医学院伦理委员会审查批准。
将饲养至8周龄后的雄性C57BL/6小鼠麻醉,并在膝关节内侧做纵向切口,沿髌韧带内侧打开关节腔,钝性分离髁间区的脂肪垫,找到连接内侧半月板的半月板胫骨韧带并横断,压迫止血后关闭切口,并继续饲养,分别于术后4周、8周、12周(每组n=8)麻醉并处死小鼠,分离出膝关节做组织学检测,形成治疗组。
将饲养至8周龄后的雄性C57BL/6小鼠麻醉,并在膝关节内侧做纵向切口,沿髌韧带内侧打开关节腔,钝性分离髁间区的脂肪垫,找到连接内侧半月板的半月板胫骨韧带暴露,但不切断,压迫止血后关闭切口,并继续饲养,分别于术后4周、8周、12周(每组n=8)麻醉并处死小鼠,分离出膝关节做组织学检测,形成假手术组。
(2)小鼠膝关节关节腔内注射α-KG进行组织学染色实验
在小鼠DMM手术3周后,分别对治疗组小鼠和假手术组小鼠进行关节腔内注射,每周1次,共5次。小鼠麻醉后,用酒精擦拭膝关节处皮肤消毒,治疗组的关节腔用胰岛素针注射PBS稀释的α-KG10μl,剂量为1ul二甲基-α-KG溶于9ul PBS。进行假手术并注射PBS作为对照。注射5次后,即DMM手术后8周时取材,做组织学染色观察关节软骨变化。
(3)组织学检测和评价
在小鼠DMM手术3周后,sham组注射PBS、DMM组注射PBS组、DMM注射α-酮戊二酸组。取相应的小鼠膝关节,并通过以下方法进行小鼠膝关节标本处理:
①小鼠膝关节于4%多聚甲醛固定24h。摇床中10%EDTA脱钙10d。
②石蜡包埋,在胫骨平台前中部做冠状位切片或胫骨平台内侧做矢状位切片。
③切片做HE染色、番红(Safranin-O)/固绿(fast green)染色。
其中,Safranin-O/fast green染色包括以下过程:
①石蜡切片脱蜡至水。
②Safranin-O染20min。
③1%醋酸漂洗1次。
④固绿染色3min。
⑤水洗后梯度酒精脱水,二甲苯透明及封片。
关节软骨的破坏程度通过Safranin-O/fast green染色情况来观察,通过OARSI评分来定量评价,评价标准下表6。

(4)统计分析
sham组、DMM组、DMM+αKG组的评分采用不配对的样本T检验。使用统计软件为SPSS11.0,P<0.05认为具有统计学差异。
2实验结果
图8显示的是通过DMM手术的方式模拟了骨关节炎时,α-KG对于抑制骨关节炎的进展情况。如图8所示,8周后,相比于PBS组,DMM组出现了严重的关节破坏,关节炎的进展比较明显,与此同时,DMM+αKG组的软骨组织没有明显破坏,骨关节炎进展不明显(图8A)。同时,通过OARSI评分来定量评价关节软骨的破坏程度,结果发现,DMM+αKG能够明显抑制骨关节炎的进展(图8B)。
实施例6小鼠皮肤毛囊再生水平
(1)实验方法
取12个月衰老的小鼠后肢背部的皮肤,治疗组的皮内注射PBS稀释的α-KG,剂量为1ul二甲基-a-酮戊二酸溶于4ul PBS。进行假手术并注射PBS作为对照。注射一次后,4周时取材,做组织学染色观察皮肤毛囊变化。
(2)实验结果
图9显示了小鼠后肢背部的皮肤毛囊染色的组织学切片,观察α-KG对于促进皮肤毛囊再生的的情况,如图9所示,4周后,相比于PBS组,注射二甲基-a-酮戊二酸的小鼠皮肤毛囊显著增加。
综上所述,本发明有效克服了现有技术中的种种缺点而具高度产业利用价值。上述实施例仅例示性说明本发明的原理及其功效,而非用于限制本发明。任何熟悉此技术的人士皆可在不违背本发明的精神及范畴下,对上述实施例进行修饰或改变。因此,举凡所属技术领域中具有通常知识者在未脱离本发明所揭示的精神与技术思想下所完成的一切等效修饰或改变,仍应由本发明的权利要求所涵盖。
Claims (9)
1.α-酮戊二酸在制备药物中的用途,所述药物用于:
1)治疗骨关节炎及相关疾病;和/或,
2)抑制软骨细胞的分解表型;和/或,
3)促进软骨细胞的合成表型;和/或,
4)促进皮肤毛囊再生。
2.如权利要求1所述的α-酮戊二酸在制备药物中的用途,其特征在于,所述α-酮戊二酸用于抑制骨细胞的NF-κB信号通路的激活和/或传导。
3.如权利要求1所述的α-酮戊二酸在制备药物中的用途,其特征在于,在白介素-1β刺激的环境下,所述α-酮戊二酸调控所述软骨细胞的分解表型和/或合成表型。
4.如权利要求3所述的α-酮戊二酸在制备药物中的用途,其特征在于,所述白介素-1β刺激所述软骨细胞。
5.如权利要求3所述的α-酮戊二酸在制备药物中的用途,其特征在于,所述白介素-1β的浓度为5~15ng/ml。
6.如权利要求1所述的α-酮戊二酸在制备药物中的用途,其特征在于,所述α-酮戊二酸在所述药物中的形式为二甲基-α-酮戊二酸。
7.如权利要求1所述的α-酮戊二酸在制备药物中的用途,其特征在于,所述药物进行关节腔注射。
8.一种药物组合物,其特征在于,所述药物组合物包括α-酮戊二酸。
9.如权利要求8所述的药物组合物,其特征在于,所述药物组合物还包括药学上的可接受的载体。
