本发明公开了一种油茶籽中活性肽纳米乳的制备方法,烘干油茶籽至含水量为20%,低温压榨,过滤得到油茶籽油和残渣;将残渣进行水解方法提取油茶籽活性肽;以油茶籽油、油茶籽活性肽、表面活性剂、助乳化剂、抗氧化剂、去离子水为原料,制备纳米乳;利用超声波与高压均值机,进行均质处理,得到纳米乳。对茶油籽活性肽进行纳米包裹,提高了活性肽的稳定性和安全性,抗氧化性好,纳米乳结构方便吸收,低温压榨可避免由于温度较高引起的活性物质损耗以及茶油色泽较深等问题;步骤简单,利于实施,生产成本低,综合利用茶油籽,更加环保,方便规模化生产。

背景技术
油茶籽是油茶树的果实,油茶树是我国主要的木本油料树,被誉为“东方树”。油茶是我国主要木本油料,也是世界四大木本食用油料。油茶可食用、药用以及作为辅料,如在《中国药典》中被作为药用油脂收录(可以作为液体制剂的非极性溶剂,用于溶解固醇类激素、油溶性维生素、游离生物碱、挥发油和芳香族药物)《,纲目拾遗》、《农政全书》、《中华本草》等中医药典籍中曾记录其具有清热解毒、润燥、杀虫、息风等作用,临床用药中多以单方或配方用于治疗皮肤过敏、暗疮、疥癣、小儿红屁股、以及皮肤烧烫伤等多种外用疾病。
纳米乳液(nanoemulsion)又称微乳液(microemulsion),是由水、油、表面活性剂和助表面活性剂等自发形成,粒径为1~100nm的热力学稳定、各向同性,透明或半透明的均相分散体系纳米乳具有许多其它制剂无可比拟的优点:①为各向同性的透明液体,属热力学稳定系统,经热压灭菌或离心也不能使之分层;②工艺简单,制备过程不需特殊设备,可自发形成,纳米乳粒径一般为1~100nm;③黏度低,可减少注射时的疼痛;④具有缓释和靶向作用;⑤提高药物的溶解度,减少药物在体内的酶解,可形成对药物的保护作用并提高胃肠道对药物的吸收,提高药物的生物利用度。因此纳米乳作为一种药物载体受到广泛的关注。
发明内容
本发明要解决的技术问题是克服以上技术缺陷,提供一种油茶籽中活性肽纳米乳的制备方法。
为了达到上述发明目的,本发明采用的技术方案为:
一种油茶籽中活性肽纳米乳的制备方法,包括以下步骤:
S1,烘干油茶籽至含水量为20%,低温压榨,过滤得到油茶籽油和残渣;
S2,将残渣进行水解方法提取油茶籽活性肽;
S3,以油茶籽油、油茶籽活性肽、表面活性剂、助乳化剂、抗氧化剂、去离子水为原料,制备纳米乳;
S4,先将油茶籽油与去离子水进行活化,其余成分分散均匀后加入反应釜内进行混合,快速搅拌10-30min,得到混合物A;
常温下磁力搅拌,将活化后的油茶籽油与去离子水缓慢加入混合物A中,混合搅拌均匀,得到混合物B;
S5,利用超声波与高压均值机,对混合物B进行均质处理,得到纳米乳。
进一步的,S1中低温压榨为70-90摄氏度炼油机30-60min。
进一步的,S2中水解工艺的水解温度为50摄氏度,采用碱性蛋白酶控制ph为8,水解时间3h。
进一步的,所述表面活性剂为是吐温-80或者蓖麻油聚氧乙烯醚(40)。
进一步的,所述助乳化剂为丙三醇。

本发明的有益效果为:对茶油籽活性肽进行纳米包裹,提高了活性肽的稳定性和安全性,抗氧化性好,纳米乳结构方便吸收,茶油籽由水分、粗脂肪、淀粉、粗蛋白质、茶籽多糖、多酚类物质、黄酮类化合物、皂素和粗纤维以及少量的鞣质组成,其中茶多酚、山茶苷和角鲨烯是其特征性的生物活性物质,具有降低胆固醇、抗衰老和预防肿瘤等功效;低温压榨可避免由于温度较高引起的活性物质损耗以及茶油色泽较深等问题;步骤简单,利于实施,生产成本低,综合利用茶油籽,更加环保,方便规模化生产。
具体实施方式
为了使本发明的内容更容易被清楚地理解,下面对本发明实施例中的技术方案进行清楚、完整地描述。
实施例一
一种油茶籽中活性肽纳米乳的制备方法,包括以下步骤:
S1,烘干油茶籽至含水量为20%,低温压榨,S1中低温压榨为70摄氏度炼油机3min,过滤得到油茶籽油和残渣;
S2,将残渣进行水解方法提取油茶籽活性肽;S2中水解工艺的水解温度为50摄氏度,采用碱性蛋白酶控制ph为8,水解时间3h;
S3,以油茶籽油、油茶籽活性肽、表面活性剂、助乳化剂、抗氧化剂、去离子水为原料,制备纳米乳;所述表面活性剂为是吐温-80;所述助乳化剂为丙三醇;各原料配比的重量份数为,油茶籽油20份、油茶籽活性肽1份、表面活性剂30份、助乳化剂10份、抗氧化剂1份、去离子水50份;
S4,先将油茶籽油与去离子水进行活化,其余成分分散均匀后加入反应釜内进行混合,快速搅拌10min,得到混合物A;常温下磁力搅拌,将活化后的油茶籽油与去离子水缓慢加入混合物A中,混合搅拌均匀,得到混合物B;
S5,利用超声波与高压均值机,对混合物B进行均质处理,所述均质压力为40MPa,均质时间18min,得到纳米乳。
实施例二
一种油茶籽中活性肽纳米乳的制备方法,包括以下步骤:
S1,烘干油茶籽至含水量为20%,低温压榨,S1中低温压榨为90摄氏度炼油机30-60min,过滤得到油茶籽油和残渣;
S2,将残渣进行水解方法提取油茶籽活性肽;S2中水解工艺的水解温度为50摄氏度,采用碱性蛋白酶控制ph为8,水解时间3h;
S3,以油茶籽油、油茶籽活性肽、表面活性剂、助乳化剂、抗氧化剂、去离子水为原料,制备纳米乳;所述表面活性剂为蓖麻油聚氧乙烯醚(40);所述助乳化剂为丙三醇;各原料配比的重量份数为,油茶籽油30份、油茶籽活性肽5份、表面活性剂40份、助乳化剂20份、抗氧化剂6份、去离子水60份;
S4,先将油茶籽油与去离子水进行活化,其余成分分散均匀后加入反应釜内进行混合,快速搅拌30min,得到混合物A;常温下磁力搅拌,将活化后的油茶籽油与去离子水缓慢加入混合物A中,混合搅拌均匀,得到混合物B;
S5,利用超声波与高压均值机,对混合物B进行均质处理,所述均质压力为55MPa,均质时间22min,得到纳米乳。
实施例三
一种油茶籽中活性肽纳米乳的制备方法,包括以下步骤:
S1,烘干油茶籽至含水量为20%,低温压榨,S1中低温压榨为80摄氏度炼油机45min,过滤得到油茶籽油和残渣;
S2,将残渣进行水解方法提取油茶籽活性肽;S2中水解工艺的水解温度为50摄氏度,采用碱性蛋白酶控制ph为8,水解时间3h;
S3,以油茶籽油、油茶籽活性肽、表面活性剂、助乳化剂、抗氧化剂、去离子水为原料,制备纳米乳;所述表面活性剂为是吐温-80;所述助乳化剂为丙三醇;各原料配比的重量份数为,油茶籽油25份、油茶籽活性肽3份、表面活性剂35份、助乳化剂15份、抗氧化剂3份、去离子水55份;
S4,先将油茶籽油与去离子水进行活化,其余成分分散均匀后加入反应釜内进行混合,快速搅拌20min,得到混合物A;常温下磁力搅拌,将活化后的油茶籽油与去离子水缓慢加入混合物A中,混合搅拌均匀,得到混合物B;
S5,利用超声波与高压均值机,对混合物B进行均质处理,所述均质压力为50MPa,均质时间20min,得到纳米乳。

Claims (7)
1.一种油茶籽中活性肽纳米乳的制备方法,其特征在于,包括以下步骤:
S1,烘干油茶籽至含水量为20%,低温压榨,过滤得到油茶籽油和残渣;
S2,将残渣进行水解方法提取油茶籽活性肽;
S3,以油茶籽油、油茶籽活性肽、表面活性剂、助乳化剂、抗氧化剂、去离子水为原料,制备纳米乳;
S4,先将油茶籽油与去离子水进行活化,其余成分分散均匀后加入反应釜内进行混合,快速搅拌10-30min,得到混合物A;
常温下磁力搅拌,将活化后的油茶籽油与去离子水缓慢加入混合物A中,混合搅拌均匀,得到混合物B;
S5,利用超声波与高压均值机,对混合物B进行均质处理,得到纳米乳。
2.根据权利要求1所述的一种油茶籽中活性肽纳米乳的制备方法,其特征在于:S1中低温压榨为70-90摄氏度炼油机30-60min。
3.根据权利要求1所述的一种油茶籽中活性肽纳米乳的制备方法,其特征在于:S2中水解工艺的水解温度为50摄氏度,采用碱性蛋白酶控制ph为8,水解时间3h。
4.根据权利要求1所述的一种油茶籽中活性肽纳米乳的制备方法,其特征在于:所述表面活性剂为是吐温-80或者蓖麻油聚氧乙烯醚(40)。
5.根据权利要求1所述的一种油茶籽中活性肽纳米乳的制备方法,其特征在于:所述助乳化剂为丙三醇。
6.根据权利要求1所述的一种油茶籽中活性肽纳米乳的制备方法,其特征在于:各原料配比的重量份数为,油茶籽油20-30份、油茶籽活性肽1-5份、表面活性剂30-40份、助乳化剂10-20份、抗氧化剂1-6份、去离子水50-60份。
7.根据权利要求1所述的一种油茶籽中活性肽纳米乳的制备方法,其特征在于:所述均质压力为40-55MPa,均质时间18-22min。
