诸位发明人现已发现,天然抗微生物肽可用作鉴定与CD206选择性相互作用的试剂的起点,特别是结合候选化合物的计算机模拟分析与CD206模型结合使用时。有利地,被鉴定为CD206,尤其是CDD206的糖识别域4(CRD4)和糖识别域5(CRD5)的选择性结合剂的分子可以用作药学试剂,这些药学试剂刺激肿瘤相关巨噬细胞的吞噬活性,并使M1/M2巨噬细胞群朝着M2含量降低的群体极化。显而易见,这类试剂将有利地激活对肿瘤细胞的吞噬作用,同时减少免疫抑制性M2巨噬细胞。值得注意的是,诸位发明人发现结合CRD4-5的试剂中的至少一些(如果不是全部的话)将选择性地与这些结构域结合并诱导CD206的构象变化,这可能向巨噬细胞提供特异性信号。
例如,一种活性化合物(RP182)衍生自模拟微生物细胞表面蛋白的天然抗微生物肽,并通过巨噬细胞群向活化的/吞噬性M1表型的转变在激活免疫细胞方面显示出显著作用。然后,如下更详细地显示的,对另外的合成类似物进行工程化,并筛选CD206结合、构象变化的诱导以及对各种细胞群(特别是巨噬细胞)的生物效应。在他们的研究过程中,诸位发明人发现这类化合物的靶受体是CD206。在验证性实验中,RP182被证明具有远高于各种天然配体的亲和力。此外,CD206的新型3D建模的使用允许鉴定特异性靶结构域,并预测构象变化(已通过实验验证,如下更详细地显示的)。
容易理解的是,与CD206结合,尤其是与糖识别域4(CRD4)和糖识别域5(CRD5)结合的试剂在制造可免疫刺激/激活肿瘤相关巨噬细胞并减少在肿瘤微环境中具有免疫抑制功能的M2巨噬细胞的数量的药物组合物中是特别有益的。
基于较早的考虑(未显示),诸位发明人首先证实了RP182(一种衍生自天然抗微生物肽的化合物)确实与CD206结合。为此,进行了SPR分析,其中定量了RP182与CD206的结合。从图1可以看出,RP182与CD206选择性并紧密结合,而对照肽(RP426)的结合亲和力明显较低。图2描绘了另外的验证性实验的结果,其中进行了细胞热位移测定(CETSA)以证明靶接合。在此,将阴性对照(PBS)与RP182和对照肽RP426进行比较。可以看出,Tagg值(来自重复进行的两个独立实验的平均值和SD)清楚地表明与RP182的特异性相互作用超出了对照。
为了验证各种合成肽将模拟抗微生物肽的概念,进行了结构建模,并且图3中提供了用于比较的示例性结果,其中合成肽在顶部,抗微生物肽在底部。计算机模拟进一步评估了大量肽片段的其他变体,并对选定的肽进行了活性筛选。值得注意的是,在该过程中,已经确定通过用具有其他类似特征的其他氨基酸替代丙氨酸来降低疏水性产生了预测的对多种可能靶标(最初包括NFkB、TRAIL、CD206等)的亲和力的提高。当CD206被确定为靶标,使用ClusPro模型生成了对预测亲和力的计算机模拟计算(也参见后面使用CD206的3D模型产生的图)。图4描绘了选定的肽和计算的结合能,以及这些肽的结构预测。图4所示的合成序列具有SEQ ID NO:1、SEQ ID NO:2、SEQ ID NO:3、SEQ ID NO:4和SEQ ID NO:5的肽序列。图5列出了与RP182相似的、天然来源的另外的示例性肽。图5所示的序列具有根据SEQ ID NO:6、SEQ ID NO:7、SEQ ID NO:8、SEQ ID NO:9、SEQ ID NO:10、SEQ ID NO:11、SEQ ID NO:12、SEQ ID NO:13、SEQ ID NO:14、SEQ ID NO:15、SEQ ID NO:16、SEQ ID NO:17、SEQ ID NO:18、SEQ ID NO:19、SEQ ID NO:20、SEQ ID NO:21、SEQ ID NO:22、SEQ ID NO:23、SEQ IDNO:24、SEQ ID NO:25、SEQ ID NO:26、SEQ ID NO:27和SEQ ID NO:28的肽序列。因此,应当理解,所有这些肽序列及其类似物均被认为适合用于本文。例如,最适合免疫调节肽结构/功能范例结构域的宿主防御蛋白发现于URL:aps.unmc.edu/AP/main.php utilizingMolly font。提供对该数据库的调查,列出了431种肽中的129种,占29.93%,其结构域与活性所需的结构决定簇一致。对RP182进行优化,使其具有最大两亲性、疏水性和正电荷密度,如Molly字体所示。RP426是测试疏水性对于活性的重要性的对照。Molly字体描述于:“Structure/Function Link Between Cytokine Domains and Natural and DesignedLytic Peptides:Medical Promise[细胞因子结构域与天然和设计的裂解肽之间的结构/功能联系:医学前景](2012).”Jesse M.Jaynes和Gregory C.Bernard,在Small Wonders:Peptides for Disease Control[小奇迹:用于疾病控制的肽],21-45,American ChemicalSociety[美国化学学会],通过引用并入本文。然后以类似的方式从URL:www.ncbi.nlm.nih.gov的蛋白质数据库中找到的蛋白质中选择毒力和胶原蛋白部分中的肽。
已经描述了17个不同的人类C型凝集素组,参见“C-type lectins in immunityand homeostasis.[在免疫和体内平衡中的C型凝集素]”Brown,GD等人Nat Rev Immunol.[自然免疫学评论]2018年6月;18(6):374-389。所有已知成员的完整列表可以在URL:www.imperial.ac.uk/research/animallectins/ctld/mammals/humanvmousedata.html的动物凝集素数据库中找到。从该资源中,列举了88种不同的C型凝集素人类蛋白。在这88种蛋白质中,选出28种用于计算机模拟研究,因为它们具有可用的晶体结构以及3种不同的toll样受体。利用ClusPro,研究了晶体结构与所示生物类似物的单个10聚体序列的结合。根据与每个类别中的10聚体序列最相似的序列选择生物类似物。图6左侧的表显示了结果,首先按照组排序,其次按照归一化聚类结合能(NCBE)的平均值排序。显示了每种受体/配体组合的三个最低的结合能(以kcal/mol为单位)。第一部分比较了所有肽与该公开中描述的28种选定的人类C型凝集素的计算机模拟结合。另外的列显示了以下结果,首先按照具有列出的-BE的组排序,其次按照归一化聚类结合能(NCBE)的平均值排序。
然后使用ClusPro,研究了28种人类C型凝集素和TLR的晶体结构与RP182的结合。图7左侧的两个表显示了以下结果,首先按照组排序,其次按照归一化聚类结合能(NCBE)的平均值排序。最差结合蛋白/配体对的前十对显示在右侧图中。
接下来,为了证实与其他生物类似物相比,RP182与MRC1/CD206结合的选择性,诸位发明人使用SAXS衍生的全长CD206结构,重复了上述生物类似物和RP182的对接研究。最后一部分数据来自使用晶体结构PDB 5XTS进行的计算机模拟测量,该晶体结构描述于“Structural Insights into the pH-Dependent Conformational Change and CollagenRecognition of the Human Mannose Receptor[对人类甘露糖受体的pH依赖性构象变化和胶原识别的结构洞察].”Hu Z等人Structure[结构].2018年1月2日;26(1):60-71。结果如图8所示。可以看出,RP182对全长CD206的结合亲和力也高于根据其与免疫激活或巨噬细胞清除的相关性而选择的任何天然肽。
在进一步的工作中,开发了CD206的各种结构模型,并推导出了最佳拟合模型以拟合小角度X射线数据。参见图9。然后将该模型如下用于后续的肽设计工作,并将其示于图10中。在此,人巨噬细胞甘露糖受体1蛋白(也称为MMR-1或CD206)是单通路I型膜信号传导蛋白。关键的拓扑特征是大的胞外区(1371aa),然后是螺旋跨膜(TM)结构域和相对较小的胞质区(46aa)。人CD206的蛋白质序列(1,456aa)从UniProt(UniProt ID P22897-1 NCBI ID:NP_002429.1)获得,并包含两个N-末端结构域(蓖麻毒蛋白-B型凝集素和II型纤连蛋白)、然后是八个C型凝集素结构域(编号1至8)、以及一个TM结构域和一个胞质结构域。人CD206的3D结构信息仅适用于非常小的区段(644-787aa;C型凝集素4结构域)。在这项工作中,诸位发明人使用迭代线程和组装改进软件(the Iterative Threading and ASSEmblyRefinement software)I-TASSER(Web服务器版本;URL:zhanglab.ccmb.med.umich.edu/I-TASSER/用于模型构建)倡导的最可靠(基于CASP实验)方法中的一种构建了CD206的完整3D模型。I-TASSER使用分层方法,该方法使用多线程方法从RCSB-PDB(www.rcsb.org)鉴定3D模板。最后,通过迭代模板片段组装模拟构建了全长模型。为了进一步帮助建模,诸位发明人提供了使用序列相似性作为距离限制输入而鉴定的距离限制二硫键信息。I-TASSER鉴定的顶部线程模板为5ao5、3jav、5ao6和4igl。这些模型的归一化B因子值(表明模型的局部精度)在零附近波动。在I-TASSER中,模型置信度由C分数测量。C分数在-5与2之间,值越高表示置信度越高。前4个模型具有以下C分数:-0.35、-1.93、-2.89和-2.97。
分析了前4个模型,并且基于小角度X射线散射模型(SAXS;参见A)和I-TASSER置信度和C分数的比较的排名最高的I-TASSER模型被确定为CD206的可能结构折叠。中间分图详细展示了I-TASSSER一级模型。纤连蛋白II型结构域似乎被蓖麻毒蛋白的c-末端区域和C型凝集素-1结构域部分包埋和包围。其他C型凝集素结构域似乎是紧密堆积的。螺旋跨膜结构域后面是胞质结构域的两个短螺旋区域。右分图是在Pymol中生成的表面模型。
然后将如此获得的模型用于鉴定RP肽靶向的特异性CD206结构域,如图11示例性所示。值得注意的是,从图12中可以看出,该模型预测了RP182对接和CD206受体的构象变化。为了证实构象变化确实不是伪像,诸位发明人在存在和不存在RP182的情况下使用了CD206的电子显微镜检查,典型结果如图13所示。
另外的体外实验表明,配体与糖识别域4(CRD4)和糖识别域5(CRD5)的结合具有各种期望的效果,包括(a)与CD206的牢固且选择性结合(SPR、微尺度热泳、细胞热位移测定),(b)与CD206+细胞的牢固且选择性结合,(c)对CD206分子构象变化的诱导,(d)对吞噬活性的刺激,以及(e)M2巨噬细胞的减少,如下所述。
更具体地,图14描绘了示例性化合物(RP182)与CD206+CD11b+细胞结合的FACS结果,而图15描绘了选择性刺激M2极化的巨噬细胞的吞噬作用、自噬和凋亡的结果。从图中可以看出,在用RP-182处理2小时后,RP-182诱导了早期和晚期吞噬作用,而没有影响CD206水平。对极化成M1和M2并被抗Rab5等染色的BMDM进行免疫荧光法,RP182(而不是对照肽RP-426和MART-1)在M2极化巨噬细胞中诱导吞噬作用。此外,RP182诱导凋亡(通过半胱天冬酶裂解测量;左图)和自噬(通过抗LC-3测量;右图),选择性杀死小鼠和人M2极化巨噬细胞,但不使M2复极化成M1巨噬细胞。用RP-182处理M2巨噬细胞后,作为M1功能的量度的产量都有增加。
图16描绘了示出RP182刺激M2样巨噬细胞和TAM的吞噬作用的示例性结果。在图17中,在RP182处理后,显著显示了标记的KPC细胞的吞噬作用,并且图18中的结果表明,RP-182的抗M2巨噬细胞活性是CD206依赖性的。值得注意的是,即使自噬被阻断,RP182处理的凋亡活性仍然很强,这表明该活性是不依赖于内化的信号传导事件,如从图19的结果可以看出。最后,图20显示了RP-182对鼠M2巨噬细胞的细胞活力的剂量反应曲线,并且图21表明RP182处理导致M2巨噬细胞活力降低。

Claims (20)
1.一种筛选药学试剂的方法,该方法包括:
建模或鉴定作为CD206的糖识别域4(CRD4)和糖识别域5(CRD5)的选择性结合剂的试剂;
定量该试剂对CD206的该糖识别域4(CRD4)和糖识别域5(CRD5)的亲和力;以及
在确认该试剂与该CRD4和CRD5结合后,在药物配制品中使用该试剂。
2.根据权利要求1所述的方法,该方法进一步包括在建模或鉴定步骤中使用CD206的分子模型的步骤。
3.根据权利要求1所述的方法,该方法进一步包括在结合该试剂后鉴定CD206的构象变化的步骤。
4.根据权利要求1所述的方法,该方法进一步包括将该试剂与巨噬细胞体外结合的步骤。
5.根据权利要求1所述的方法,该方法进一步包括体外测试该试剂对吞噬活性的诱导的步骤。
