Abstract

本发明涉及使用SRSF10(富含丝氨酸/精氨酸的剪接因子10)的生物活性抑制剂治疗乙型肝炎病毒感染的方法和药物组合物,特别是所述抑制剂使SRSF10保持在去磷酸化状态,并阻止或降低SRSF10的剪接活性。
发明背景
尽管存在预防性疫苗,但慢性乙型肝炎病毒(HBV)感染在世界范围内仍然是主要健康问题,因为它们涉及2.5亿人口,并且表现为原发性肝癌(肝细胞癌,HCC)的首要诱因(Petruzziello,2018)。目前临床上公认的针对HBV慢性感染的抗病毒治疗包括直接靶向由病毒DNA转录并间接增强宿主免疫反应的聚乙二醇化干扰素-α(IFN-α和/或特异性抑制HBV逆转录的核苷类似物(NUC)。这些治疗通常会导致患者血液中的病毒血症短暂降低(针对IFN)或持久降低(针对NUC)(Zoulim和Durantel,2015)。然而,很少取得肝脏病毒的清除,因此必须使用NUC进行终身治疗。因此,迫切需要开发能够直接或间接抑制HBV复制的新型抗病毒分子,这些抗病毒分子可用于组合疗法。

为了破译HBc的细胞核功能,发明人鉴定了其在人类分化的肝细胞中的细胞核伙伴。对人类肝细胞核中的HBc相互作用因子的蛋白质组学鉴定揭示了大多数RNA结合蛋白(RBPs)参与mRNA的代谢。特别地,发现了富含丝氨酸/精氨酸的剪接因子10(SRSF10)在HBc复合物中富集了近3000倍,因此突出了HBc与该核RNA结合因子的强且优先的相互作用。
为了进一步研究SRSF10在HBV复制过程中的作用,发明人使用了化合物1C8(4-吡啶酮苯并异噻唑甲酰胺(4-pyridinonebenzisothiazole carboxamide)),该化合物最近被确定为HIV-1复制的强抑制剂(Shkreta等,2017)。1C8最初被鉴定为IDC16的结构模拟物,IDC16是一种抑制主要SR因子SRSF1剪接功能的药物(Cheung等,2016)。分子分析表明,1C8并不抑制SRSF1,而是阻止SRSF10的磷酸化,从而调节其剪接活性(Shkreta等,2017)。特别地,1C8被揭示可抑制SRSF10在丝氨酸133的磷酸化。此残基的磷酸化以及丝氨酸131发生的磷酸化先前被发现会改变与其他剪接因子的相互作用(Shkreta等,2017)。体外研究表明,1C8强烈抑制HIV复制,而对细胞基因和细胞活力的影响很小。
HIV复制的抑制与剪接的病毒mRNA合成的改变有关并且与SRSF10和HIV mRNAs的结合减少有关(Shkreta等,2017)。专利申请WO2015/164956公开了苯并异噻唑衍生物化合物(属于1C8)及其在治疗HIV感染中的用途。
发明内容
在本发明中,研究HBV复制机制的发明人试图破译HBc的调节功能,通过质谱(MS)分析确定其在人类肝细胞核中的细胞伴侣。该分析鉴定了超过200种蛋白质。功能注释显示,这些因子中约有50%是RNA结合蛋白(RBPs),并且特别是SR蛋白,其中最相关的功能类别与具有高度相互连接的参与RNA剪接的因子相一致。功能分析确定SRSF10能够强有力地和差异化地调控HBV的复制。此外,靶向SRSF10蛋白活性的小化合物1C8诱导不同株型的HBV(基因型C和D)的病毒复制显著下降。
因此,本发明涉及一种用于治疗乙型肝炎病毒(HBV)感染的SRSF10活性抑制剂。
在特定的实施方案中,所述抑制剂使SRSF10维持在去磷酸化状态,并防止或降低SFR10的剪接活性。
在特定的实施方案中,SRSF10活性的抑制剂能够在被感染细胞中减少cccDNA和/或pgRNA。
本发明还涉及一种用于筛选可用于治疗乙型肝炎病毒(HBV)感染的多种候选化合物的方法,该方法包括以下步骤:(a)测试每种候选化合物抑制SRSF10活性的能力,以及(b)并正选择能够抑制所述SRSF10活性的候选化合物。
具体实施方式
在本研究中,发明人证明了SRSF10是存在于HBc核复合物中的主要因子,并且可以在HBV生命周期中发挥作用。由于1C8被证明了能够调节SRSF10的磷酸化状态,发明人直接研究了这个已经被证明能够调节HIV复制的分子是否也能够抑制HBV的复制。他们发现,1C8化合物确实能够抑制持续感染的肝细胞中HBV(基因型为D和C)的复制,强烈地将细胞内HBVRNAs减少60-70%(见图5A),并将细胞培养上清液中病毒抗原(HBs和HBeAg)和HBV病毒粒子的分泌减少60-70%(见图5B)。IC8化合物还能够抑制HBV cccDNA在新生感染环境中的建立:在HBV接种之前和接种过程中(基因型C和D)在肝细胞中添加该分子可强有力地减少cccDNA的建立(分别减少70%和50%),并且其他病毒标志物(即细胞内RNAs、HBeAg、HBsAg)在1C8处理条件下也减少了(图7A和B以及图8A和B)。
感染的肝细胞中的HBV cccDNA是所有病毒亚基因组转录本和前基因组RNA(pgRNA)的模板,负责持续的慢性感染和再激活,以确保新合成的病毒后代和cccDNA池通过细胞内核衣壳回收得到补充。在本发明的上下文中,首次表明SRSF10活性控制1)慢性感染细胞中细胞内HBV RNA(HBV总RNA和HBV前基因组RNA)的命运;和2)新生感染中cccDNA的建立。这些知识为减少HBV感染对象的新生cccDNA合成提供了机会,从而为彻底治愈慢性感染HBV患者提供了机会。

如本文所使用的术语“富含丝氨酸/精氨酸的剪接因子10”或“SRSF10”具有其本领域的一般含义。SRSF10(也称为NSSR、TASR、SRp38、TASR1、TASR2、FUSIP1、FUSIP2、SFRS13、SRrp40、SFRS13A、PPP1R149)属于SR(丝氨酸/精氨酸)蛋白家族,是一组参与premRNA剪接的RBPs(RNA结合蛋白)。它在人类中由位于1号染色体上1p36.11处的基因(ID:10772)编码。SRSF10因其在有丝分裂期间和对热休克的应答中的脱磷酸化最初被鉴定为通用剪接阻遏物(Shin等,2004;Shin等,2005)。相反,磷酸化的SRSF10可以激活剪接,并且其磷酸化状态决定其与不同的RBPs如hnRNPs(hnRNPK,F和H)和TRA2B的相互作用(Shkreta等,2017;Shkreta等,2016)。重要的是,磷酸化的SRSF10会激活与应激、DNA损伤、细胞凋亡和致癌途径相关的几种细胞转录物的选择性剪接(Shkreta等,2016;Zhou等,2014a;Zhou等,2014b)。最后,SRSF10还在病毒premRNAs的剪接中发挥作用,特别是HIV-1转录本的剪接(Shkreta等,2017)。
SRSF10活性抑制剂

确定化合物作为SRSF10活性抑制剂的能力的测试是本领域技术人员众所周知的。在一个优选的实施方案中,该抑制剂特异性地将SRSF10维持在去磷酸化状态,以充分的方式抑制SRSF10的生物活性。将SRSF10维持在去磷酸化状态和抑制SRSF10的生物活性可以通过本领域众所周知的任何测定法来确定。例如,该测定法可以在于确定药剂改变SRSF10的磷酸化状态的能力。直接测量蛋白质磷酸化的经典方法包括用放射性标记的32P-正磷酸孵育整个细胞,生成细胞提取物,用SDS-PAGE分离蛋白质,并在薄膜上显露。其他传统方法包括二维凝胶电泳,这种技术假设磷酸化会改变蛋白质的迁移率和等电点。例如在Shkreta等2016和Shkreta等2017中,描述了SRSF10磷酸化状态的测定方法,基于去磷酸化形式的SRSF10在凝胶条件下与磷酸化形式相比具有更快的迁移率。液相色谱-质谱(LC-MS)也是评估SRSF10磷酸化状态的有用工具。Shkreta等2017也使用该测定法来确定IC8促进SRSF10在丝氨酸133处的去磷酸化。
然后可以安排竞争性测定,以确定该试剂抑制SRSF10的生物学活性的能力。可以设想功能测定法,以便评估诱导或抑制HBV感染细胞中与在感染细胞中建立cccDNA和/或pgRNA相关的剪接活性的能力(参见带有1C8化合物的实例和图7-8)。
本领域技术人员可以容易地确定SRSF10活性抑制剂是否中和、阻滞、抑制、消除、降低或干扰SRSF10的生物学活性。为了检查SRSF10活性抑制剂是否与最初表征的1C8化合物以相同的方式改变SRSF10的磷酸化状态和/或抑制SRSF10的剪接活性和/或抑制HBV感染细胞中的cccDNA和/或pgRNA,可以用每种抑制剂进行检测。例如,肝细胞中的HBV感染可以通过分析病毒参数如cccDNA定量和/或pgRNA定量来测量。对于cccDNA定量,总DNA用T5核酸外切酶(New England Biolabs)消化,然后使用Taqman Fast Advanced Master Mix(LifeTechnologies)进行qPCR。对于pgRNA定量,RT-qPCR可以使用Taqman Fast AdvancedMaster Mix(Life Technologies)进行。使用商业化探针引物混合物(Life Technologies#Hs99999908_m1)将PgRNA水平标准化为GUSB。
HBV感染也可以通过病毒参数的分析来进行测量,例如通过Elisa(化学发光免疫分析试剂盒Autobio)对分泌的HBe和HBs抗原进行定量分析(分泌的HBe和HBs抗原的水平是肝细胞HBV感染的标准分泌标志物)和/或如实施例部分所述使用特异性HBV引物通过qPCR或RT-qPCR评估从感染细胞中提取的细胞内总HBV DNA或RNA。
已经确定了抑制SRSF10的小分子与SRSF10在HIV中的作用有关;这样的SRSF10抑制剂也被预期为在治疗HBV的本发明中有用。特别是将这种小分子化合物,例如通过共轭接合或制剂,靶向到肝脏,可能对治疗HBV有利。
在一个特定的实施方案中,根据本发明的活性抑制剂是小的有机分子,例如化合物1C8(或C8):4-吡啶酮苯并异噻唑甲酰胺(参见Shkreta等2016和Shkreta等2017WO2015164956)或属于1C8的苯并异噻唑衍生物化合物(参见WO2015164956)。WO2015164956中公开的所有活性化合物在此通过引用并入本文。特别是以下化合物:
所有这些化合物均显示出抗HIV活性,尤其是活性最高的1C8化合物,该化合物在残基133处抑制SRSF10的磷酸化,从而调节其剪接活性。
在一个实施方案中,本发明还涉及用于治疗乙型肝炎病毒(HBV)感染的苯并异噻唑衍生物化合物。
该苯并异噻唑衍生物化合物在WO2015164956中公开,并在此以引用方式纳入。在一个优选的实施方案中,所述化合物选自由1C8、E5、D3、C2或SL309化合物所组成的列表。
在一个特定的实施方案中,本发明还涉及用于治疗乙型肝炎病毒(HBV)感染的化合物1C8(4-吡啶酮苯并异噻唑甲酰胺)或苯并异噻唑衍生物化合物(E5、D3、C2或SL309化合物)。
药物组合物
SRSF10活性的抑制剂可以与药学上可接受的赋形剂,以及任选的缓释基质,例如生物可降解的聚合物相组合,以形成治疗组合物。
在本发明的用于口服、舌下、皮下、肌肉内、静脉内、经皮、局部、吸入或直肠给药的药物组合物中,活性成分,单独或与另一种活性成分组合,可以以单位给药形式,作为与常规药物载体的混合物,对动物和人类给药。合适的单位给药形式包括口服途径形式,例如片剂、凝胶胶囊、粉剂、颗粒剂和口服混悬剂或溶液剂、舌下和口腔给药形式、气雾剂、埋植剂、皮下、经皮、局部、腹膜内、肌肉内、静脉内、皮下、经皮、鞘内和鼻内给药形式以及直肠给药形式。
优选地,药物组合物包含赋形剂,这些赋形剂对于能够被注射的制剂是药学上可接受的。这些赋形剂尤其可以是等渗的、无菌的、盐溶液(磷酸一钠或磷酸二钠、钠、钾、氯化钙或氯化镁等或这些盐的混合物),或干燥的,特别是冻干的组合物,这些赋形剂根据情况加入无菌水或生理盐水后,可以配制注射液。
适用于注射用途的药物形式包括无菌水溶液或分散体;剂型包括芝麻油、花生油或丙二醇水溶液;以及用于临时制备无菌注射液或分散体的无菌粉剂。在所有情况下,该剂型必须是无菌的,并且必须是流动的,以便于注射。它在生产和储存条件下必须是稳定的,并且必须以防止例如细菌和真菌等微生物的污染的方式保存。
含有本发明化合物作为游离碱(free base)或药学上可接受的盐的溶液可以在水中与表面活性剂,如羟丙基纤维素适当混合制备。分散体也可以在甘油、液态聚乙二醇及其混合物和油中制备。在常规的储存和使用条件下,这些制剂含有防腐剂,以防止微生物的生长。
本发明的SRSF10活性抑制剂可以以中性或盐的形式配制成组合物。药学上可接受的盐包括酸加成盐(与蛋白质的游离氨基基团形成),它是用例如盐酸或磷酸的无机酸形成的,或用例如乙酸、草酸、酒石酸、扁桃酸酸等有机酸形成的。用游离的羧基形成的盐类也可以衍生自无机碱,例如钠、钾、铵、钙或铁的氢氧化物,以及诸如异丙胺、三甲胺、组氨酸、普鲁卡因等有机碱。
载体也可以是溶剂或分散介质,其包含例如水、乙醇、多元醇(例如甘油、丙二醇和液态聚乙二醇等)、其合适的混合物和植物油。可以例如通过使用包衣(例如卵磷脂),在分散液的情况下通过维持所需的粒径以及通过使用表面活性剂来维持适当的流动性。可以通过各种抗菌剂和抗真菌剂来预防微生物的作用,例如对羟基苯甲酸酯、氯丁醇、苯酚、山梨酸、硫柳汞等。在许多情况下,将优选包括等渗剂,例如糖或氯化钠。通过在组合物中使用延迟吸收的试剂,例如单硬脂酸铝和明胶,可以使可注射组合物的吸收延长。
通过将所需量的活性化合物与所列举的各种其他成分按要求加入适当的溶剂中,然后过滤灭菌,从而制备无菌注射溶液。通常,通过将各种已灭菌的活性成分加入到无菌载体中来制备分散体,所述无菌载体包含基本分散介质和以上列举的所需其他成分。在制备无菌注射溶液的无菌粉末的情况下,优选的制备方法是真空干燥和冷冻干燥技术,它从先前的活性成分和任何其他所需成分的无菌过滤溶液中得到其粉末。
配制后,溶液将以与剂型相容的方式和治疗有效的量给药。该制剂容易以多种剂型施用,例如上述的注射液类型,但也可以使用药物释放胶囊等。
例如,对于在水溶液中的肠胃外给药,如果需要,溶液应该适当地缓冲,并且首先用足够的盐水或葡萄糖使液体稀释剂呈等渗状态。这些特定的水溶液特别适合于静脉内、肌肉内、皮下和腹膜内给药。在这方面,可以采用的无菌水介质,将是本领域的技术人员根据本公开内容所知晓的。例如,一个剂量可以溶解在1ml等渗NaCl溶液中,并加入到1000ml皮下注射液中,或者在建议的输注部位注射。根据治疗对象的状况,剂量必然会出现一些变化。在任何情况下,负责给药的人将确定适合每个施用对象的剂量。
本发明的SRSF10活性或表达的抑制剂可配制在治疗混合物内,每剂约含0.0001至1.0毫克,或约0.001至0.1毫克,或约0.1至1.0或甚至约10毫克左右。也可以给予多个剂量。
除了为肠外给药,如静脉注射或肌肉注射而配制的本发明化合物外,其他药学上可接受的形式包括,例如片剂或其它口服给药的固体;脂质体制剂;缓释胶囊;以及当前使用的任何其他形式。
筛选方法
本发明还涉及一种筛选对治疗乙型肝炎病毒(HBV)感染有用的多种候选化合物的方法,该方法包括以下步骤:(a)测试每种候选化合物抑制SRSF10活性的能力,以及(b)并并且阳性选择能够抑制所述SRSF10活性的候选化合物。
通常,候选化合物选自有机小分子、肽、多肽或寡核苷酸组成的组。
测试候选化合物是否可以抑制SRSF10活性可以使用本领域已知的报道基因检测方法或常规方法进行测定。
例如,该方法可以包括使表达SRSF10的细胞与候选化合物接触,并测量SRSF10的磷酸化状态(例如,SRSF10剪接活性的激活或抑制),并将细胞应答与标准细胞应答进行比较。通常,标准细胞应答是在不存在候选化合物的情况下测量的。细胞应答比标准的减少表明该候选化合物是SRSF10的活性抑制剂。
已被阳性选择的候选化合物可进行进一步的选择步骤,以便进一步检测其在从患有HBV感染的施用对象中分离的肝细胞(或被HBV感染的dHeparRG细胞见Grippon等2002)上的特性。例如,已经通过上述筛选方法阳性选择的候选化合物可以被进一步选择,选择具有抑制HBV感染细胞中SRSF10剪接活性的能力或减少HBV感染细胞中cccDNA和/或pgRNA的能力。通常,筛选方法可以进一步包括以下步骤:i)使来自HBV感染患者的肝细胞与阳性选择的候选化合物接触;ii)确定所述HBV感染细胞中cccDNA和/或pgRNA的量;以及iii)将步骤ii)测定的ccDNA和/或pgRNA的量与在没有阳性选择的候选化合物的情况下进行步骤i)时测定的ccDNA和/或pgRNA的量进行比较。如上所述的步骤i)可以通过向肝细胞的培养基中加入一定量的待测候选化合物来进行。通常,准备多个培养样品,以便在不同的培养样品中添加增加量的待测试的候选化合物。通常,还制备至少一种不含候选化合物的培养样品作为阴性对照,以便进一步比较。
最后,可对已经被阳性选择的候选化合物进行进一步的选择步骤,在动物模型上进一步分析其对HBV感染的特性。通常,可以将阳性选择的候选化合物施用于动物模型,确定HBV感染的进展并与未施用候选化合物的动物模型中确定的HBV感染的进展进行比较。

实施例1
材料与方法:
野生型HepaRG、工程化HepaRG细胞的培养以及HBV感染的条件。
在补充有10%FBS(Perbio)、青霉素/链霉素(500U/mL)、谷氨酰胺(2mM)、氢化可的松Upjohn(2.5mg/L;Serb实验室)、胰岛素(5mg/L;Sigma Aldrich)的Williams培养基(ThermoFisher)中培养人肝祖HepaRG细胞。为进行分化过程,将2%的DMSO添加到上述补充了的Williams培养基中。如前所述(Gripon等,2002)分化后的HepaRG细胞(dHepaRG)感染由HepG2.2.15或HepAD38细胞制备的HBV基因型D接种物或由HepG2细胞制备的HBV基因型C(GenBank:KP017269.1),该基因型C用线性化pcDNA3-HBV-1.35-基因组单元质粒稳定转染,并进行G418抗性选择。图例中表示了感染的多重性(MOI;表示为病毒基因组当量/细胞),但如果不预处理,则为100vge/cell。
HepaRG-TR-StepTag-HBc细胞系是通过在表达盒中携带四环素抑制因子(TR)的细小病毒进行双重转导而产生的,该表达盒可以对杀稻瘟菌素(Blasticidine)具有抗性并携带StrepTag-HBc基因,其在可以抗博来霉素的表达盒中编码N端带有StrepTag的HBV核心/衣壳蛋白。将HepaR-TR-StrepTag-HBc细胞在含有100mg/ml博来霉素和10mg/ml杀稻瘟菌素的相同Williams培养基中培养。四环素也被添加到培养基中(1mg/mL)以获得转基因的表达,即StepTag-HBc。
StrepTag纯化
IBA的系统采用Strep-Tactin亲和柱树脂,是目前应用最广泛的用于蛋白质纯化检测和固定的亲和层析系统之一,并具有许多优势,包括经过生理纯化过程(在有利于蛋白质与蛋白质相互作用的缓冲液条件下),分离出的复合物纯度高。纯化所使用的程序是制造商推荐的程序,可在网上查询:www.ibalifesciences.com/strep-tactin-system-technology.html。
化学试剂。IC8由Benoit Chabot博士提供。用于抑制HBV进入的PreS1肽(HBVPreS1的第2至48个氨基酸序列,TNLSVPNPLGFFPDHQLDPAFRANSNNPDWDFNPNKDHWPWPEANKVG(SEQ ID N°1),在N端用肉豆蔻酰基修饰,并重悬于标准盐酸盐中)由Genscript(Honk-Hong)合成。核苷类似物、拉米夫定(Lamivudine)和替诺福韦(Tenofovir)由GileadSciences提供。用于确定这些分子的抗HBV活性的两种实验方案见图1。
细胞毒性测定
病毒参数的分析
从细胞培养上清液中,使用化学发光免疫分析试剂盒(Autobio,中国)按照制造商的说明,通过ELISA对分泌的HBe和HBs抗原进行定量。根据制造商的规程,分别使用MagMAx试剂盒(Thermo scientific)和MagNAPure(Roche)从细胞培养上清液中提取细胞外病毒DNA,并用DNAse-I或RNAse-A处理。分别使用Nucleospin 96Tissue或NucleoSpin 96RNA试剂盒(Macherey-Nagel)从感染的细胞中提取细胞内总HBV DNA或RNA。使用SuperScriptIII逆转录酶(Invitrogen)将RNAs转录为cDNA。使用LightCycler480(Roche)对总细胞内HBV DNA和来自RNA的cDNA进行实时PCR,并管家基因进行归一化,利用以下引物:5'-GCTGACGCAACCCCCACT-3'(HBV正义链:SEQ ID N°2)和5'-AGGAGTTCCGCAGTATGG-3'(HBV反义链:SEQ ID N°3)、5'-TGCTGGGAAGTGCCATGAG-3'(PRP正义链:SEQ ID N°4)和5'-CGGTGCATGTTTTCTCACGATAGTA-3'(PRP反义链:SEQ ID N°5)。
HBV标准品用于细胞外病毒DNA和RNA的定量。对于cccDNA的定量,将总DNA用T5核酸外切酶(New England Biolabs)在37℃下消化6小时,然后使用Taqman Fast AdvancedMaster Mix(Life Technologies)进行qPCR。用β-球蛋白定量将CccDNA归一化。对于pgRNA定量,使用Taqman Fast Advanced Master Mix(Life Technologies)使用自制的探针引物混合物进行qPCR。使用商业探针引物混合物(Life Technologies#Hs99999908_m1)将PgRNA水平归一化为GUSB。
统计学分析。使用GraphPad Prism软件6.0用非参数Mann-Whitney检验进行统计分析。显著性如下:*P<.05,**P<.01,***P<.001,****P<.0001。
结果:
SRSF10是HBV核心/衣壳蛋白的主要相互作用物
HBV的核心/衣壳蛋白(HBc)是一种结构蛋白,在细胞质区间内对HBV前基因组RNA的封装起作用。封装后,pgRNA被逆转录为松弛的环状DNA(rcDNA),这是在病毒粒子中发现的HBV的基因组形式。除这种结构功能外,HBc还具有潜在的其他非结构性调节功能,它在高度复制的肝细胞中积累,特别是在受感染细胞的核中。为了深入了解HBc的核功能,我们对四环素诱导的HepaRG-TR-StrepTag-HBc细胞核中的StepTag-HBc相关蛋白复合物进行了质谱分析。该程序的示意图如图2所示。
进行了三个独立的实验,以提高分析的统计能力。在核酸酶(-)和核酸酶(+)的条件下,分别发现了44个和58个HBc相互作用蛋白(p值<0.005,且富集因子>4)。在两组条件之间有37种蛋白质是共有的。这些蛋白质的列表在图3中给出。
许多HBc相互作用蛋白被发现是参与RNA代谢的RNA结合蛋白(preRNA成熟、剪接、RNA运输……)。在这些蛋白质中,我们对SFSF10产生了兴趣,SFSF10是最丰富的StrepTag-HBc相关因子之一,即使存在核酸酶的作用下也是如此(图4)。正如引言部分所指出的,该蛋白质也被发现对HIV复制很重要。而有趣的是,先前已描述了一些具有抑制特性的小分子,其中包括1C8。
1C8在复制HBV的dHepaRG细胞中的抗HBV特性
我们用两种不同的实验方案分析了1C8的潜在抗HBV特性(图1)。在持续感染(HBV基因型D;MOI为100vge/cell)的dHepaRG中,每2-3天用10mM的1C8或对照分子治疗3次,我们发现1C8是唯一能够降低60-70%细胞内HBV RNA积累的药物,而FDA批准的核苷类似物(NUC;拉米夫定(Lamivudine)(3TC)和替诺福韦(Tenofovir)(TDF))不能影响这种病毒参数(图5A)。测试的分子均无法降低HBV的cccDNA。1C8还能够将细胞培养上清液中的病毒抗原(HBs和HbeAg)和HBV病毒粒子(按图5B中的分泌的HBV DNA测量)的分泌减少60-70%;这次也发现了NUCs对HBV病毒粒子的产生有预期的强抑制作用(图5B)。1C8不能抑制丁型肝炎病毒在相同细胞中的复制(图5C)。1C8对通过发光细胞活力测试法测试的40mM以下的细胞没有毒性(图5D),因此证明了抗病毒作用对HBV复制的特异性。
HBV以各种基因型存在。在评估宿主靶向制剂(HTA;即抑制细胞功能的分子,这对病毒复制很重要,从而导致抗病毒作用)时,如1C8,重要的是检查HTA的抗病毒特性在基因型之间是否保守。因此,我们评估了1C8对一种主要在亚洲流行的基因型C型病毒复制的影响。我们发现,1C8保留了对该病毒株的抗病毒特性,尽管功效略有降低(图6)。
1C8也损害了原始(naive)dHepaRG中HBV感染的建立
在第二种实验方法中,我们试图确定1C8是否会像进入抑制因子(Li and Urban,2016)或核心组装调节因子(CAMs)(Berke等人,2017)一样,破坏dHepaRG细胞中HBV感染的建立。进入抑制因子,如PreS1肽(例如Myrcludex),通过阻止HBV病毒体与其细胞受体人类牛磺胆酸钠共转运肽(hNTCP)之间的相互作用,抑制HBV进入肝细胞。CAMs主要是为了抑制衣壳化过程而开发的,也被证明是通过作用于后进入水平来阻止cccDNA的建立,在某种程度上干扰了核苷酸向新感染肝细胞核的进入后运输。相反,临床使用的NUCs不会干扰HBV感染的建立。由于1C8的抗HBV抑制机制尚不清楚,我们在HBV接种前和接种过程中用该分子处理细胞,以确定其是否可以阻止cccDNA池的建立以及随后其他病毒成分的合成/产生。我们发现,在10mM的1C8能够抑制50%的HBV基因型D的cccDNA的建立(图8A)和70%HBV基因型C的cccDNA的建立(图8A)。
这种1C8诱导的抑制作用低于进入抑制剂获得的抑制作用,但高于NUC(替诺福韦;TDF)获得的抑制作用。在抑制cccDNA的建立后,其他病毒标志物(即细胞内RNAs、HbeAg、HbsAg)在1C8处理的条件下也减少了(图7A和B和图8A和B)。在平均时间内,采用相同的实验设定(即接种前24h和接种过程中的处理),1C8无法阻断HDV感染的建立(对照品Entry Inh.可抑制HDV感染),因此再次表明了1C8对HBV的作用具有特异性(图7C和8C)。
实施例2
材料与方法:
原代人类肝细胞(PHH)的培养,以及HBV感染的情况。
如前所述(Leycluse和Alexandre,2010年)原代人类肝细胞(PHH)由来自具有法国部级授权的Centre Léon Bérard(里昂)的人类肝脏切除物新鲜的制备。它们在Williams培养基(Thermofisher)中培养,补充有5%的FBS(Perbio)、青霉素/链霉素(500U/mL)、谷氨酰胺(2mM)、氢化可的松Upjohn(2.5mg/L;Serb实验室)和胰岛素(5mg/L;Sigma Aldrich)。PHH按先前描述的方法(Gripon等,2002),用HBV基因型D接种物以100vge/cell从HepAD38细胞(GenBank:KP017269.1)制备。感染16小时后,洗涤细胞。感染后6天,用浓度为10μM的替诺福韦(TDF)、拉米夫定(3TC)或1C8处理细胞。每次处理后收获上清液,感染后13天收获细胞。
病毒参数分析。
如实施例1所述,分析分泌的HBe和HBs抗原以及细胞内HBV RNA。
统计学分析。使用GraphPad Prism软件6.0用非参数Mann-Whitney检验进行统计学分析。显著性如下:*P<.05,**P<.01,***P<.001,****P<.0001。
结果
1C8可抑制原代人类肝细胞中的HBV复制
对持续感染的PHH也同样测试了1C8的抑制作用。为此,从两个不同供体获得两个不同批次的PHH感染HBV(HBV基因型D;MOI为100vge/cell),然后用10mM 1C8或对照分子按照两种不同方案进行处理(图9A和D)。对分泌的病毒抗原HBe和HBs的分析表明,用1C8进行治疗会导致病毒复制的稳健下降(>60%的减少)(图9B和D)。正如预期的那样,这些参数不受3TC的影响,3TC是一种靶向HBV pgRNA逆转录的核苷类似物。细胞内病毒RNAs(总RNA和pgRNA)的定量进一步表明,正如先前在HepaRG细胞中观察到的(图6A),将1C8加入到HBV感染的PHH中导致病毒RNAs的量显著减少(图9C)。

Claims (9)

7.一种用于筛选可用于治疗乙型肝炎病毒(HBV)感染的多种候选化合物的方法,该方法包括以下步骤:(a)测试每种候选化合物抑制SRSF10活性的能力,以及(b)并且阳性选择能够抑制所述SRSF10活性的候选化合物。
