Abstract
本发明提供一种用于癌症治疗的索拉非尼‑基因共载纳米药物及其制备方法和应用,其中索拉非尼‑基因共载纳米药物包括由靶向脂质层包裹的纳米团聚体;靶向脂质层包括索拉非尼、磷脂、靶向物和胆固醇;纳米团聚体为由活性氧响应材料包裹的shUSP22。上述制备方法,包括以下步骤:采用活性氧相应材料包裹shUSP22,制得纳米团聚体;将靶向脂质层负载于纳米团聚体的表面,得索拉非尼‑基因共载纳米药物。上述应用为在制备治疗癌症药物中的应用。本发明的索拉非尼‑基因共载纳米药物能够有效提高癌细胞内活性氧水平,抑制肝癌细胞中USP22的表达,高效杀伤肝癌细胞,延长索拉非尼的血液循环时间,抑制肿瘤生长。
Description
技术领域
本发明属于抗肿瘤药物技术领域,具体涉及一种用于癌症治疗的索拉非尼-基因共载纳米药物及其制备方法和应用。
背景技术
肝细胞肝癌(HCC,以下简称肝癌)是我国目前第四位常见恶性肿瘤。受限于肝癌早筛、早诊的困难以及其复杂的生物学机制,目前肝癌患者的五年生存率仅14%-18%。近年来,随着肝癌分子生物学、药物科学的发展,针对肝癌增殖、凋亡、血管生成等分子靶向治疗在肝癌的诊治中逐渐受到重视。多激酶抑制剂-索拉非尼(Sorafenib)的问世,一定程度上延长了肝癌患者的生存期。但是肝癌复杂的生物学机制及药物的不良反应,限制了索拉非尼的临床应用和治疗效果。据相关临床研究统计,仅约40%的肝癌患者可从索拉非尼的治疗中获益。因此,如何寻找到更加精准有效的治疗靶点便成为影响肝癌治疗效果的关键因素。
泛素特异性蛋白酶22(USP22)是肿瘤干细胞的标志蛋白之一。相关研究表明USP22通过缺氧诱导因子1-α参与肿瘤干性的调节并密切调控多药耐药相关蛋白1等肿瘤干细胞相关蛋白。针对USP22进行治疗,有望逆转肝癌的肿瘤干性并克服肿瘤耐药。但目前尚缺乏可有效抑制肝癌内USP22表达或功能的小分子抑制剂或基因药物。
纳米基因药物是指纳米载体经电荷相互作用、疏水相互作用等装载基因药物后,通过主动或被动靶向将基因药物输送至靶细胞,纠正其异常基因的表达,从而到达治疗疾病的目的。针对肿瘤内乏氧、偏酸的独特微环境,设计相应的肿瘤微环境响应型纳米药物可有效提高基因药物的递送效率,避免基因药物脱靶造成的毒副作用。
由于肿瘤异常的生理代谢特点,相较于正常人体组织,肿瘤微环境中含有较高浓度的活性氧。因此,设计活性氧响应的纳米基因药物可避免基因药物的脱靶效应,并有效提高基因药物的递送效率。但肿瘤细胞内活性氧水平仍不足以引起活性氧响应性纳米基因药物在细胞内的快速、高效释放,目前还没有文献公开可行的技术方案。
发明内容
为解决上述现有技术中存在的问题,本发明提供一种用于癌症治疗的索拉非尼-基因共载纳米药物,其能够有效提高癌细胞内活性氧水平,极大的抑制肝癌细胞中USP22的表达,高效杀伤肝癌细胞,有效延长索拉非尼的血液循环时间,并有效抑制肿瘤生长。本发明还提供上述索拉非尼-基因共载纳米药物的制备方法和应用。
本发明利用活性氧响应的电荷反转型阳离子聚合物B-PDEAEA包裹USP22 shRNA(shUSP22)形成纳米团聚体,并进一步在其表面通过薄膜水化法负载了含索拉非尼的半乳糖修饰脂质层,制备得到了一种共载索拉非尼及shUSP22的肝癌靶向的自激活纳米药物(Gal-SLP)。本发明通过外源性地在纳米基因药物中加入活性氧激动剂以诱导肿瘤细胞内活性氧水平的升高,并进一步引发活性氧响应性纳米基因药物中基因的释放。
一种用于癌症治疗的索拉非尼-基因共载纳米药物(Gal-SLP),包括由纳米团聚体以及包括在纳米团聚体外表面的靶向脂质层;
所述靶向脂质层包括索拉非尼、磷脂、靶向物和胆固醇;
所述纳米团聚体为由活性氧响应材料包裹的基因药物shUSP22(针对人USP22基因的特异性shRNA真核表达质粒);
本发明中,含有索拉非尼的靶向物修饰的所述靶向脂质层能够有效提高癌细胞内活性氧水平,由活性氧响应材料包裹shUSP22,能够提高所述索拉非尼-基因共载纳米药物的靶向性,并使其在高浓度活性氧的癌细胞内释放基因药物,进而更加准确高效地杀死癌细胞。
优选地,所述索拉非尼、磷脂、靶向物以及胆固醇的摩尔比为1:(6-8):(1-2):(1-2),进一步优选为1:(6.5-7.5):(1.5-2):(1-1.5);作为一种具体的优选方案,所述索拉非尼、磷脂、靶向物以及胆固醇的摩尔比为1:6.9:1.8:1.2。
作为优选,所述磷脂为二油酰基磷脂酰乙醇胺(DOPE),所述靶向物为二硬脂酰基磷脂酰乙醇胺-聚乙二醇2000-半乳糖(DSPE-PEG2000-galactose),所述胆固醇为胆固醇琥珀酸单酯(CHEMS)。
作为优选,所述活性氧响应材料为聚合物B-PDEAEA。
作为优选,所述活性氧响应材料与shUSP22的N/P(即聚合物/质粒DNA的氮磷比)为5~50,进一步优选为5~25,更进一步优选为17。
作为优选,所述索拉非尼与针对人USP22基因的特异性shRNA真核表达质粒的质量比为:5~15:1。
作为优选,所述针对人USP22基因的特异性shRNA真核表达质粒中目标表达片段的DNA序列为F:GCGAAGGGTACTTGCTGTTCTA,R:CGCTTCCCATGAACGACAAGAT。
作为优选,所述纳米药物中索拉非尼的载药量为2~5%,包封率为60~90%。
一种上述用于癌症治疗的索拉非尼-基因共载纳米药物的制备方法,包括以下步骤:
(1)采用活性氧相应材料包裹shUSP22,制得纳米团聚体;
(2)将上述靶向脂质层包裹于(1)中制得的纳米团聚体的表面,得所述索拉非尼-基因共载纳米药物。
优选地,步骤(2)中采用薄膜水化法将靶向脂质层包裹于纳米团聚体表面。
优选地,所述靶向脂质层材料包括索拉非尼、磷脂、靶向物以及胆固醇;所述索拉非尼、磷脂、靶向物以及胆固醇的摩尔比为1:(6-8):(1-2):(1-2),进一步优选为1:6.9:1.8:1.2。
作为优选,所述磷脂为二油酰基磷脂酰乙醇胺,所述靶向物为二硬脂酰基磷脂酰乙醇胺-聚乙二醇2000-半乳糖,所述胆固醇为胆固醇琥珀酸单酯。
作为优选,所述活性氧响应材料为聚合物B-PDEAEA。

作为优选,本发明提供了上述索拉非尼及shUSP22共载的肝癌靶向的自激活纳米药物(Gal-SLP)的制备方法,包括:
(1)通过活性氧响应性材料B-PDEAEA包裹shUSP22,制备得到了不同N/P比的纳米团聚体;并通过USP22蛋白表达的抑制效率、生物安全性等实验确定了N/P=17为纳米复合体的最适N/P比。
(2)调整脂质层中索拉非尼、磷脂(二油酰基磷脂酰乙醇胺(DOPE))、靶向物(二硬脂酰基磷脂酰乙醇胺-聚乙二醇2000-半乳糖(DSPE-PEG2000-galactose))及胆固醇(CHEMS,胆固醇琥珀酸单酯)比例,并进一步通过薄膜水化法将含索拉非尼的肝癌靶向脂质层包裹(1)中制备得到的纳米团聚体,形成索拉非尼及shUSP22共载的肝癌靶向的自激活纳米药物(Gal-SLP)。
作为具体优选,一种上述用于癌症治疗的索拉非尼-基因共载纳米药物的制备方法,包括以下步骤:
(1)用HEPES缓冲溶液稀释shUSP22,得质粒DNA溶液;控制所述活性氧响应材料与shUSP22的N/P为5~25,将聚合物B-PDEAEA以不同浓度溶解在HEPES缓冲溶液中,得不同N/P比的B-PDEAEA溶液;将等体积的质粒DNA(针对人USP22基因的特异性shRNA真核表达质粒)溶液分别添加到不同N/P比的B-PDEAEA溶液中,涡旋,静置,得不同N/P比的纳米团聚体溶液。
(2)将索拉非尼、DOPE、CHEMS和DSPE-PEG2000-半乳糖溶解于有机溶剂(例如氯仿)中,旋蒸去除有机溶剂,得到透明的癌细胞靶向脂质薄膜。将该脂质薄膜置于HEPES溶液中常温水化过夜,后在冰浴中超声,得到脂质体溶液。将脂质体溶液静置,并经尼龙滤器过滤除去游离的药物聚集体;
(3)将步骤(2)中制得的经尼龙滤器过滤的脂质体溶液加入到步骤(1)中制备得到的纳米团聚体溶液中,混合后室温静置过夜,得所述索拉非尼-基因共载纳米药物。
一种上述索拉非尼-基因共载纳米药物在制备治疗癌症药物中的应用,所述癌症药物优选为肝癌药物。
在体外细胞实验中,首先验证了索拉非尼及本发明所制备得到的Gal-SLP可有效提高肝癌细胞内活性氧水平;进一步地,通过细胞毒性、凋亡检测、USP22蛋白表达水平检测等实验证明了本发明所制备的Gal-SLP能够极大的抑制肝癌细胞中USP22的表达,高效杀伤肝癌细胞。
在体内动物实验中,本发明所制备的Gal-SLP能够有效延长索拉非尼的血液循环时间,并有效抑制肿瘤生长。
本发明首先通过活性氧化响应的电荷反转型阳离子聚合物B-PDEAEA包裹USP22靶向基因药物(shUSP22),制备得到了活性氧响应的纳米团聚体。通过USP22蛋白表达的抑制效率、生物安全性等实验确定了聚合物B-PDEAEA与shUSP22的N/P=17为该纳米团聚体的最适N/P比。将含索拉非尼的半乳糖修饰的脂质层通过薄膜水化法负载于上述纳米团聚体表面,形成了肝癌靶向的shUSP22及索拉非尼共载的纳米药物(Gal-SLP)。Gal-SLP的制备流程图如图21所示。
与现有技术相比,本发明的有益效果为:
(1)本发明的活性氧响应性Gal-SLP避免了传统基因治疗载体的脱靶效应,能在高活性氧浓度的肝癌细胞内释放基因药物;
(2)本发明的表面负载有半乳糖修饰脂质体的Gal-SLP能够避免血液对索拉非尼的清除,延长索拉非尼的体内循环时间,并靶向抗去唾液酸糖蛋白受体高表的肝癌细胞递送索拉非尼及基因药物;
(3)在癌症细胞内,本发明的Gal-SLP可通过释放索拉非尼提高肿瘤内活性氧浓度,促进活性氧响应性的纳米团聚体解离,实现shUSP22的快速、高效释放,从而达到高效的基因转染;
(4)本发明的Gal-SLP具有良好的体内抗肿瘤效果与生物安全性。
综上所述,本发明的所制备的共载索拉非尼及USP22靶向基因药物的肝癌靶向的自激活纳米药物,可高效的递送shUSP22及索拉非尼到肝癌部位,避免了体内的网状内皮系统对基因药物及索拉非尼的清除,使得shUSP22及索拉非尼的体内循环周期延长;同时,Gal-SLP进入肝癌细胞后快速释放索拉非尼,引起细胞内活性氧水平的升高,继而引发shUSP22的快速释放,更大程度上实现了USP22蛋白表达的抑制,对肝癌的疗效增强。

具体实施方式
以下具体实施方式用来进一步说明本发明。
基于数据库预测及不同USP22 shRNA(简写:shUSP22,全称:针对人USP22基因的特异性shRNA真核表达质粒)预测序列在人肝癌细胞中USP22蛋白表达抑制水平的检测,本发明选定shUSP22的核苷酸序列为F:GCGAAGGGTACTTGCTGTTCTA,R:CGCTTCCCATGAACGACAAGAT;其中,质粒结构选用p-Genesil-1-shRNA。
实施例1:不同N/P的纳米团聚体的制备及表征
用HEPES缓冲溶液(10mM,pH=7.4)将质粒DNA(USP22 shRNA)稀释至DNA浓度为40μg/mL的溶液。根据预设N/P比(N/P=5、9、13、17、21、25),将B-PDEAEA以相应浓度溶解在HEPES缓冲溶液(10mM,pH=7.4)中,得不同浓度的B-PDEAEA溶液。然后,将等体积的质粒DNA溶液分别添加到不同浓度的B-PDEAEA溶液中,立即将混合物涡旋10秒钟,后静置30分钟,得不同N/P比的纳米团聚体溶液。
通过动态光散射及透射电子显微镜对纳米团聚体的粒径、zeta电位及形貌进行检测。纳米团聚体的粒径及zeta电位(图1)结果表明经B-PDEAEA包裹质粒DNA得到的不同N/P比纳米团聚体粒径为~50nm,zeta电位在+20至+25mV范围内。对N/P比为17的纳米团聚体进行投射电镜观察(图2),可知本发明制备得到的纳米团聚体尺寸分布均匀。
实施例2:不同N/P比纳米团聚体的基因负载效率及活性氧响应能力的检测
1、不同N/P比的纳米团聚体的凝胶阻滞实验
取20μL实施例1中所制备的不同N/P比纳米团聚体溶液,同时取0.4μg的shUSP22用HEPES缓冲溶液(10mM,pH=7.4)稀释至20μL作为对照。将上述7个样品上样于质量体积分数为0.8%,含Gel Red的琼脂糖凝胶中,在TBE缓冲液中120V电泳45分钟。电泳结束后,将凝胶置于紫外成像仪中进行拍摄。
电泳结果(图3)表明,实施例1中所选用的活性氧响应性聚合物B-PDEAEA在N/P比5-25之间均可高效压缩质粒DNA,并阻滞DNA的迁移。
2、不同N/P比的纳米团聚体在200μM过氧化氢中37℃孵育1小时后的凝胶阻滞实验
取20μL实施例1中所制备的不同N/P比的纳米团聚体溶液及20μL含0.4μg shUSP22的HEPES缓冲溶液(置于0.5mM过氧化氢中37℃下孵育1小时。孵育结束后,将上述7个样品上样于质量体积分数为0.8%,含Gel Red的琼脂糖凝胶中,在TBE缓冲液中120V电泳45分钟。电泳结束后,将凝胶置于紫外成像仪中进行拍摄。
凝胶电泳结果(图4)表明,不同N/P比的纳米团聚体均具有良好的过氧化氢响应性,在低浓度过氧化氢短时间孵育下即可释放其所负载的质粒DNA。
实施例3:不同N/P比纳米团聚体对Huh-7肝癌细胞系中USP22蛋白表达的抑制效率
在6孔板接种150000个Huh-7细胞。培养过夜后,向细胞培养基中加入实施例1制备的不同N/P比纳米团聚体,其中shUSP22浓度为8μg/孔。将负载shUSP22质粒的Lipofectamine 2000(Lipo2000)和支链聚乙烯亚胺(PEI,25kDa)设置为阳性对照,其中Lipo2000与质粒的混合比例为2μL:1μg;PEI与质粒DNA的N/P为7。孵育48小时后,弃去培养基,经PBS溶液洗涤后用胰酶收集细胞并向细胞沉淀从加入100μL含蛋白酶抑制剂的RIPA裂解液。经BCA法对蛋白浓度进行定量并加入对应体积的Loading buffer后,进行蛋白凝胶电泳及转膜。按照USP22及β-actin的分子量,裁剪得到相应的蛋白条带。含5%脱脂奶粉的封闭液封闭1小时后,将条带与对应的一抗稀释液(USP22,Abcam,Cat.ab109435,稀释比为1:2000;β-actin,Proteintech,Cat.66009-1-Ig,稀释比为1:10000)孵育过夜,后与对应的二抗稀释液(HRP-conjugated Affinipure Goat Anti-Mouse IgG(H+L),Proteintech,Cat.SA00001-1,稀释比为1:10000;HRP-conjugated Affinipure Goat Anti-Rabbit IgG(H+L),Proteintech,Cat.SA00001-2,稀释比为1:10000)常温孵育2小时。最后,利用ECL化学发光检测试剂对蛋白条带进行可视化观察。
蛋白凝胶电泳结果(图5中a)显示,不同N/P比纳米团聚体均能有效的降低USP22表达。对USP22相对表达量进行统计发现(图5中b),在Huh-7肝癌细胞系中,N/P为17的纳米团聚体能降低~40%的USP22表达,显著高于商业化质粒转染试剂Lipo2000及PEI。
实施例4:不同N/P比纳米团聚体的体外生物安全性评估
在96孔板中按3000细胞/孔的密度接种Huh-7细胞。培养过夜后,向培养基中加入负载了阴性对照(NC,negative control)质粒的不同N/P比纳米团聚体,其中NC质粒的浓度为0.5μg/孔。孵育48小时后,将培养基替换为110μL含有10μL CCK-8溶液的新鲜培养基并于37℃下继续孵育2小时。随后,通过酶标仪对每个孔450nm吸光度进行测量。
体外安全性评估实验结果(图6)显示,任一N/P比的纳米团聚体处理48小时后,Huh-7的细胞活性均大于85%;表明在DNA治疗浓度下,该活性氧响应性纳米团聚体在体外不具有明显的生物毒性。
实施例5:共载索拉非尼及USP22靶向基因药物的肝癌靶向的自激活纳米药物(Gal-SLP)及单载USP22靶向基因药物的肝癌靶向的活性氧响应纳米药物(Gal-LP)的制备
1、Gal-SLP及Gal-LP的制备
索拉非尼/DOPE/CHEMS/DSPE-PEG2000-半乳糖的摩尔比固定为1.0:6.9:1.8:1.2。将索拉非尼(0.23mg),DOPE(2.53mg),CHEMS(0.43mg)和DSPE-PEG2000-半乳糖(1.84mg)溶解于2mL氯仿中,旋蒸去除有机溶剂,得到透明脂质薄膜。将该脂质膜置于1mL HEPES(10mM,pH 7.4)溶液中常温水化过夜,后在冰浴中超声10分钟得到脂质体溶液。将脂质体溶液于4℃静置2小时,并经0.22μm尼龙滤器过滤除去游离的药物聚集体。将上述的脂质体溶液加入到实施例1中制备得到的N/P为17的纳米团聚体溶液(含20μg质粒DNA)中,充分混合后在室温静置过夜,得到Gal-SLP溶液。
利用动态光散射(图7)及透射电镜(图8)对制备得到的Gal-SLP溶液中Gal-SLP的粒径大小和Zeta电位进行观察。经薄膜水化法制备得到的Gal-SLP粒径为95.6±5.2nm,zeta电位为-5.6±0.8mV,且粒径分布均一。
Gal-LP的制备按上述Gal-SLP制备流程。其中,在脂质膜的制备中不添加索拉非尼,即可制备得到Gal-LP溶液。
2、Gal-SLP中索拉非尼载药量及包封率的测定
为了确定Gal-SLP中索拉非尼的载药量和包封率,将上述制备得到的Gal-SLP溶液冻干并重新溶解于乙腈中进行高效液相分析。结果显示Gal-SLP中索拉非尼的载药量为3.6%,包封率为74.5%,表明Gal-SLP可高效负载非水溶性的索拉非尼。
3、Gal-SLP中索拉非尼的释放特性
采用透析法对Gal-SLP中索拉非尼的释放特性进行表征。按照上述1所述方法得到Gal-SLP溶液(2mL,负载0.5mg索拉非尼)密封在透析袋(MWCO=3500Da)中,并在60mL含1%吐温80的不同pH(pH=5.0或7.4)PBS进行透析。在预设的时间点,取100μL透析液进行高效液相色谱仪分析。高效液相色谱仪分析结果(图9)显示经Gal-SLP负载的索拉非尼具有缓释的特点;同时在酸性环境(拟肿瘤微环境)下,索拉非尼的释放速率加快。
实施例6:Gal-SLP对肝癌细胞系Huh-7增殖抑制、诱导凋亡及USP22表达的敲低效率检测
1、Gal-SLP对肝癌细胞系Huh-7杀伤能力的评估
在96孔板中按3000细胞/孔的密度接种Huh-7细胞。培养过夜后,向培养基内加入不同浓度的索拉非尼及实施例5中1所制备的Gal-LP、Gal-SLP。孵育48小时后,将培养基替换为110μL含有10μL CCK-8溶液的新鲜培养基并于37℃下继续孵育2小时。随后,通过酶标仪对每个孔450nm吸光度进行测量。
肿瘤细胞杀伤实验结果(图10)表明,相较于单一的索拉非尼(图中为Sorafenib,下同)或USP22基因治疗(Gal-LP),联合索拉非尼及USP22基因治疗(Gal-SLP)其对肝癌细胞的杀伤能力更强。
2、Gal-SLP对肝癌细胞系Huh-7诱导凋亡能力的评估
在6孔板中按100000细胞/孔的密度接种Huh-7细胞。培养过夜后,向培养基内加入索拉非尼及实施例5中1所制备的Gal-LP、Gal-SLP,其中索拉非尼及shUSP22的浓度分别为5μM及5μg/孔。孵育36小时后,收集细胞并用5μL碘化丙啶溶液(PI,1μg/mL)进行染色。染色15分钟后,用流式细胞仪对PI阳性的肝癌细胞进行检测。
凋亡实验结果(图11)显示,相较于单一的索拉非尼或Gal-LP,Gal-SLP其诱导肝癌细胞凋亡的能力更强。
3、Gal-SLP对肝癌细胞系Huh-7中USP22蛋白表达抑制的评估
在直径为6cm的细胞培养皿中按200000细胞/孔的密度接种Huh-7细胞。培养过夜后,向培养基内加入索拉非尼及实施例5中1所制备的Gal-LP、Gal-SLP,其中索拉非尼及shUSP22的浓度分别为5μM及8μg/皿。孵育48小时后,按实施例3中所述,通过蛋白凝胶电泳对不同处理后肝癌细胞内USP22及β-actin表达进行检测。
蛋白凝胶电泳的结果(图12)显示Gal-LP能有效抑制肝癌细胞内USP22的表达;同时,Gal-SLP处理后的肝癌细胞几乎不表达USP22。
实施例7:在Huh-7肝癌细胞内Gal-SLP自激活现象的观察
1、不同处理下Huh-7肝癌细胞内活性氧水平的检测
在6孔板中按100000细胞/孔的密度接种Huh-7细胞。培养过夜后,向培养基中分别加入索拉非尼及实施例5中1所制备的Gal-LP及Gal-SLP,其中索拉非尼及shUSP22的浓度分别为0.5μM及8μg/孔。孵育6小时后,将培养基替换成含10μM二氢乙锭的新鲜培养基并继续孵育30分钟。孵育结束后,收集细胞并通过流式细胞仪对细胞内活性氧水平进行定量分析。
细胞内活性氧检测结果(图13)显示,索拉非尼及Gal-SLP均可在短时间内引发肝癌细胞内活性氧水平的升高。
2、在Huh-7肝癌细胞内Gal-SLP的基因释放过程的检测
在激光共聚焦培养皿中接种20000个Huh-7细胞。培养24小时后,将培养基替换成含实施例5中1所制备的Gal-LP及Gal-SLP的新鲜培养基。其中Gal-LP及Gal-SLP制备中所用到的活性氧响应性聚合物B-PDEAEA及质粒DNA分别通过化学修饰的方法标记有FITC及Cy5;培养基中质粒DNA的浓度为1μg/皿。在孵育2、4、8小时后,用Hoechst33342对细胞核进行染色并在激光共聚焦显微镜下进行观察。在Huh-7肝癌细胞内Gal-LP、Gal-SLP的基因释放实验结果(图14)显示Gal-SLP能够更快的释放DNA进入细胞核。
实施例8:经荧光探针标记的Gal-SLP在荷瘤小鼠体内的分布评估
细胞膜近红外荧光探针(DiR)经疏水作用包裹入实施例5中1所制备的Gal-SLP中得到Gal-SLP/DiR,对Gal-SLP在荷瘤小鼠体内的分布进行标记。向肿瘤体积~200mm3的荷瘤小鼠静脉注射Gal-SLP/DiR,其中DiR的注射剂量为每只小鼠2μg。在不同的时间点,采用小动物活体光学及X光成像系统对Gal-SLP/DiR的体内分布进行观察。
活体成像结果(图15)显示Gal-SLP/DiR的荧光强度在小鼠体内衰减速度较慢,表明Gal-SLP具有良好的体内长循环特性;同时,小鼠肿瘤部位的荧光强度明显强于小鼠其他器官,这表明表面修饰有半乳糖的Gal-SLP具有良好的肝癌靶向性。
实施例9:游离索拉非尼及Gal-SLP负载的索拉非尼的药代动力学研究
将游离索拉非尼(溶于聚氧乙烯蓖麻油/乙醇=1:1(v:v)的溶液及实施例5中1所制备的Gal-SLP通过尾静脉注射入ICR小鼠体内,其中索拉非尼的注射剂量为5mg/kg。在预设的时间点,从小鼠眼眶静脉丛中取50μL血,并向其中加入1mL乙腈。经充分搅拌、超声及离心后,取乙腈溶液中上清,并经0.22μm滤器去除难溶性颗粒。
利用高效液相色谱仪对上清中索拉非尼的浓度进行测定。药代动力学研究(图16及表1)表明Gal-SLP的曲线下面积(AUC0-t)为58.7±8.98μg*h/mL,Gal-SLP具有良好的体内长循环特性,能够保护索拉非尼免于快速的血浆清除。

实施例10:Gal-SLP的体内抗肿瘤效果研究
将肿瘤平均体积为75mm3的荷瘤小鼠随机分成4组,每隔一天分别尾静脉注射生理盐水(Saline)、索拉非尼(溶于聚氧乙烯蓖麻油/乙醇=1:1(v:v)的溶液)及实施例5中1所制备的Gal-LP及Gal-SLP,共注射6次。其中索拉非尼和shUSP22的注射浓度分别为5mg/kg和0.5mg/kg。每隔一天记录小鼠的体重及肿瘤体积。第16天,以人道主义的方式处死小鼠并将小鼠的肿瘤取出进行称重。
在实验观察期内,小鼠体重并无明显变化(图17);同时,小鼠肿瘤体积变化曲线、肿瘤重量(图18、图19)表明Gal-SLP具有杰出的体内抗肿瘤效应,其肿瘤生长抑制率可达82.7±4.9%,而索拉非尼及Gal-LP的肿瘤生长抑制率分别为53.4±15.5%和55.0±7.8%。
实施例11:Gal-SLP的体内安全性研究
通过尾静脉向ICR小鼠体内注射生理盐水、索拉非尼(溶于聚氧乙烯蓖麻油/乙醇=1:1(v:v)的溶液)及实施例5(1)中所制备的Gal-LP及Gal-SLP,共注射3次。其中索拉非尼和shUSP22的注射浓度分别为5mg/kg和0.5mg/kg。注射结束后,取小鼠血进行血常规及肝肾功能检测。
小鼠血常规及肝肾功能检测结果(图20)显示Gal-LP及Gal-SLP对小鼠的血液系统及肝肾功能均没有明显的损伤。
综上所述,本发明经活性氧响应性聚合物B-PDEAEA包裹shUSP22及薄膜水化法制备得到了共载索拉非尼及USP22靶向基因药物(shUSP22)的肝癌靶向的自激活纳米药物(Gal-SLP)。该种纳米药物能够高效的抑制肝癌细胞内USP22蛋白的表达,同时抑制肝癌细胞的增殖,诱导其凋亡。体内抗肿瘤效果及体内安全性评价实验表明,本发明所制备的Gal-SLP具有良好的体内抗肿瘤效果与生物安全性,具有较高的临床转化前景。

Claims (8)
1.一种用于癌症治疗的索拉非尼-基因共载纳米药物,其特征在于,包括纳米团聚体以及包裹在纳米团聚体外的靶向脂质层;
所述靶向脂质层包括索拉非尼、磷脂、靶向物和胆固醇;
所述纳米团聚体为由活性氧响应材料包裹的针对人USP22基因的特异性shRNA真核表达质粒;
所述磷脂为二油酰基磷脂酰乙醇胺,所述靶向物为二硬脂酰基磷脂酰乙醇胺-聚乙二醇2000-半乳糖,所述胆固醇为胆固醇琥珀酸单酯;
所述活性氧响应材料为聚合物B-PDEAEA;
所述针对人USP22基因的特异性shRNA真核表达质粒中目标表达片段的DNA序列为:F:GCGAAGGGTACTTGCTGTTCTA,R:CGCTTCCCATGAACGACAAGAT。
2.根据权利要求1所述的用于癌症治疗的索拉非尼-基因共载纳米药物,其特征在于,所述索拉非尼、磷脂、靶向物以及胆固醇的摩尔比为1:(6-8):(1-2):(1-2)。
3.根据权利要求1所述的用于癌症治疗的索拉非尼-基因共载纳米药物,其特征在于,所述活性氧响应材料与针对人USP22基因的特异性shRNA真核表达质粒的N/P为5~25。
4.根据权利要求1所述的用于癌症治疗的索拉非尼-基因共载纳米药物,其特征在于,所述索拉非尼与针对人USP22基因的特异性shRNA真核表达质粒的质量比为:5~15:1。
5.一种如权利要求1-4中任一项所述的用于癌症治疗的索拉非尼-基因共载纳米药物的制备方法,其特征在于,包括以下步骤:
(1) 采用活性氧响应材料包裹针对人USP22基因的特异性shRNA真核表达质粒,制得纳米团聚体;
(2) 将癌细胞靶向脂质层包裹于步骤(1)中制得的纳米团聚体的表面,得所述索拉非尼-基因共载纳米药物。
6.根据权利要求5所述的用于癌症治疗的索拉非尼-基因共载纳米药物的制备方法,其特征在于,步骤(2)中采用薄膜水化法将癌细胞靶向脂质层包裹于纳米团聚体表面。
7.一种如权利要求1-4中任一项所述的索拉非尼-基因共载纳米药物在制备治疗癌症药物中的应用。
8.根据权利要求7所述的应用,其特征在于,所述癌症药物为肝癌药物。
