The invention belongs to the field of cosmetics, and particularly relates to a melanin hair dye and a using method thereof.
Background
Hair dye refers to cosmetic capable of changing hair color and having special application. The hair dyes that have been most used for a long time are permanent and semi-permanent hair dyes. However, as an oxidation dye which is a main component of the oxidation hair dye, a conventional permanent hair dye is also called an oxidation hair dye. The specific dyeing mechanism of the hair dye is that oxidation dye intermediate with low relative molecular mass and other components penetrate into the hair, undergo oxidation polymerization in the hair to generate macromolecular dye and are locked in the hair. Oxidative hair dyes are mostly composed of 3 reactive compounds, namely, dye intermediates, couplers (also called couplers) and oxidizing agents, and particularly p-phenylenediamine has been found to be an allergic substance which causes allergic dermatitis and is banned by some european countries. Recent scientific research also shows that phenylenediamine and some derivatives thereof have accumulative property, rust degeneration and mutation property, and can cause keratitis, nephritis, leukemia and the like. Meanwhile, most of the permanent hair dyes in the current market are two-agent hair dyes, and the permanent hair dyes need to be blended before hair dyeing, so that the hair dyeing process is complicated.
The plant type hair dye uses one plant extract or a mixture of a plurality of plants as a raw material of the hair dye, wherein the mixture of the plurality of plant extracts is used for preparing most of the hair dyes, the extraction process is complicated, and the plurality of hair dyes are prepared only primarily and no products are produced. It is reported that the domestic health department has not approved the plant hair dyes made of plant extracts completely, and most of the commercially available 'pure plant hair dyes' are gimmicks made of some ingredients containing plants, wherein some chemical ingredients such as PPD are mixed.
Melanin is divided into synthetic melanin and natural melanin, and natural melanin refers to a natural biological pigment extracted from plants, animals and microorganisms, and has a wide variety of sources, and is the most extensive pigment in the known biological pigments at present. Melanin from natural sources is insoluble in acid and water, but has greater solubility in lye where p H is greater than 8.5. The melanin is insoluble in ethanol, methanol, acetone, ether and other common organic solvents, the chelation capacity of the melanin to the metal ions with the valence of 2 and 3 is very strong, and at present, a plurality of black hair dyes achieve the purpose of coloring by utilizing the chelation of the melanin and the metal ions.
The existing hair dye has unstable dyeing color and is easy to have the problems of hair color change or fading in the hair washing process.
Disclosure of Invention
The invention mainly aims to provide a melanin hair dye, which is a novel three-agent type hair dye prepared by taking melanin naturally produced by culturing melanocytes as a raw material of the hair dye. Under certain conditions, melanin can be complexed with metal ions to form a macromolecular compound, but the diameter of the melanin molecule is large, so that the melanin molecule is not beneficial to permeating into hair. Therefore, the agent A is used as a hair pretreatment agent, and the penetrant is used to open the disulfide bonds in the hair, thereby facilitating the penetration of the following metal ions and coloring agents into the hair. The metal ions in the agent B enter human hair and are firstly complexed with amino acid in the hair, the agent C enters the hair and is complexed with the metal ions to form macromolecular melanin, hair scales are closed, and the pigment is locked inside a hair rod.
In order to achieve the purpose, the invention adopts the following technical scheme:
a melanin hair dye comprises a combination of agent A, agent B and agent C; the agent A is prepared from the following components in percentage by mass: 1-20% of penetrating agent, 0.1-2% of ascorbic acid, 0.1-2% of thickening agent, 0.01-10% of alkaline agent, 0.3-2% of preservative, 0.0-3% of essence and the balance of water;
the agent B is prepared from the following components in percentage by mass: 1-10% of metal ions, 0.1-2% of ascorbic acid, 0.1-2% of thickening agent, 0.01-1% of citric acid, 0.01-10% of alkali, 0.3-2% of preservative, 0.0-3% of essence and the balance of water;
the agent C is prepared from the following components in percentage by mass: 0.1-2% of ascorbic acid, 0.1-2% of melanin granules, 0.1-2% of thickening agent, 0.01-10% of alkaline agent, 0.3-2% of preservative, 0.0-3% of essence and the balance of water;
the melanin granules are made from primary human melanocytes, trypsin and citric acid.
Further, the preparation method of the melanin granules comprises the following steps:
s1, melanocyte culture: culturing primary human melanocyte until cell fusion degree reaches 90%, digesting cells with trypsin digestive juice with mass fraction of 0.25%, centrifuging at 800rpm/min for 5min, suspending cells with PBS, and centrifuging at 50-200mJ/cm2The suspension was irradiated at a UVB dose of (1: 2-1: 3 for 24-48h to obtain melanocytes for later use;
s2, obtaining melanin granules: repeatedly freezing and thawing the melanocyte obtained in S1 for 3-4 times, collecting liquid, filtering, acidifying with citric acid, centrifuging, and obtaining precipitate, namely melanin granules.
Further, the penetrating agent is at least one selected from the following: l-cysteine and glutathione.
Further, the metal ion is at least one selected from the group consisting of: ferric chloride, magnesium chloride, zinc sulfate, copper sulfate, ferrous chloride, aluminum chloride, alum, and copper nitrate.
Further, the thickener is at least one selected from the group consisting of: hydroxyethyl cellulose (HEC), carboxymethyl cellulose (CMC), hydroxypropyl methyl cellulose (HPMC), xanthan gum, guar gum, sodium alginate, sodium hyaluronate, gelatin, and sodium alginate.
Further, the alkaline agent is at least one selected from the following: ammonia, ammonium bicarbonate, sodium hydrogen phosphate, sodium hydroxide, potassium hydroxide, ethanolamine, triethanolamine, arginine, and lysine.
Further, the use method of the melanin hair dye comprises the following steps: uniformly coating a certain amount of hair dye A on hair, sealing with a preservative film, and dyeing at 25-40 deg.C for 20-50 min; then uniformly coating a certain amount of hair dye B on the hair, sealing with a preservative film, and dyeing at 25-40 deg.C for 20-50 min; then uniformly coating a certain amount of hair dye C on hair, sealing with preservative film, and dyeing at 25-40 deg.C for 20-50min
Compared with the prior art, the invention has the following technical effects:
the hair dye has firm and stable hair dyeing effect, the hair color is close to the hair dyeing result of the hair dye sold in the market, the beauty concept of the public is met, the hair dye is natural and harmless, the hair dye can be stored for 3 years under the sealing condition at normal temperature, the hair dyeing time is short, and the hair dyeing temperature is low.

Fig. 1 is a graph showing the comparison of the effects of natural human black hair before dyeing, and after dyeing using the melanin hairdye of the present invention, wherein a is before dyeing, b is natural human black hair, and c is after dyeing.
Fig. 2 shows the results before and after dyeing with a commercial hair dye, wherein a is before dyeing and B is after dyeing.
Detailed Description
The invention is described in detail below with reference to the following figures and specific examples, which should not be construed as limiting the invention. The technical means used in the following examples are conventional means well known to those skilled in the art, and materials, reagents and the like used in the following examples can be commercially available unless otherwise specified.
Example 1
A melanin hair dye comprises the following preparation steps:
(1) culturing melanocytes: culturing primary human melanocyte (P3) obtained by normal separation with M254 culture medium, digesting the cells (0.25% trypsin) when the cell fusion degree reaches 90%, centrifuging (800rpm/min, 5min), suspending the cells with PBS, irradiating the suspension with UVB dose of 100mJ/cm2, continuously culturing at the ratio of 1:2 after centrifugation for 24h, and placing in a refrigerator at-80 ℃ for later use.
(2) Obtaining of melanin granules: and (2) repeatedly freezing and thawing the melanocytes obtained in the step (1) for 3-4 times, collecting liquid, filtering, acidifying by adding citric acid, and centrifuging to obtain precipitates, namely melanin granules. (3) Preparing a melanin hair dye:
the hair dye comprises three formulations, namely, formulation A: adding 5 parts of L-cysteine, 0.6 part of ascorbic acid (VC), 1 part of hydroxyethyl cellulose, 0.3 part of preservative, 4 parts of ammonia water and the balance of deionized water in sequence by weight part of 100 parts, and uniformly mixing.
And (2) agent B: adding 4 parts of alum, 0.5 part of ascorbic acid (VC), 1 part of hydroxyethyl cellulose, 0.3 part of preservative (phenoxyethanol), 0.01 part of citric acid and the balance of deionized water in sequence by weight of 100 parts, and uniformly mixing.
C, agent C: adding 0.5 part of ascorbic acid (VC), 2 parts of melanin granules, 1 part of hydroxyethyl cellulose, 0.3 part of preservative (phenoxyethanol), 6 parts of ammonia water and the balance of deionized water in sequence by weight of 100 parts, and uniformly mixing.
The use method of the melanin hair dye comprises the following steps: firstly, uniformly coating the agent A on hair, wrapping the hair with a preservative film, and preserving the temperature for 30min at 35 ℃; then smearing the agent B on hair, wrapping with preservative film, and keeping the temperature at 40 deg.C for 30 min; finally, the agent C is smeared on the hair, wrapped by a preservative film and kept at the temperature of 35 ℃ for 30 min.
Example 2
A melanin hair dye comprises the following preparation steps:
(1) culturing melanocytes: culturing primary human melanocyte (P4) obtained by normal separation with M254 culture medium, digesting the cells (0.25% trypsin) when the cell fusion degree reaches 90%, centrifuging (800rpm/min, 5min), suspending the cells with PBS, irradiating the suspension with UVB dose of 150mJ/cm2, continuously culturing at a ratio of 1:2 after centrifugation for 36h, and placing in a refrigerator at-80 ℃ for later use.
(2) Obtaining of melanin granules: and (2) repeatedly freezing and thawing the melanocytes obtained in the step (1) for 4 times, collecting liquid, filtering, acidifying by adding citric acid, and centrifuging to obtain precipitates, namely melanin granules.
(3) Preparing a melanin hair dye:
the hair dye comprises three formulations, namely, formulation A: adding 4 parts of L-cysteine, 0.8 part of ascorbic acid (VC), 0.8 part of guar gum, 2 parts of preservative (phenoxyethanol), 3 parts of essence, 0.2 part of sodium hydroxide and the balance of deionized water in sequence by weight of 100 parts, and uniformly mixing.
And (2) agent B: adding 4 parts of ferric chloride, 0.3 part of ascorbic acid (VC), 0.8 part of guar gum, 2 parts of preservative (phenoxyethanol), 3 parts of essence, 0.02 part of citric acid and the balance of deionized water in sequence by weight of 100 parts, and uniformly mixing.
C, agent C: adding 0.6 part of ascorbic acid (VC), 1 part of melanin particles, 0.8 part of guar gum, 2 parts of preservative (phenoxyethanol), 3 parts of essence, 0.3 part of sodium hydroxide and the balance of deionized water in sequence by weight of 100 parts, and uniformly mixing.
The use method of the melanin hair dye comprises the following steps: firstly, uniformly coating the agent A on hair, wrapping the hair with a preservative film, and preserving the temperature at 33 ℃ for 30 min; then smearing the agent B on hair, wrapping with preservative film, and keeping the temperature at 30 ℃ for 30 min; finally, the agent C is smeared on the hair, wrapped by a preservative film and kept at the temperature of 35 ℃ for 30 min.
Example 3
A melanin hair dye comprises the following preparation steps:
(1) culturing melanocytes: culturing primary human melanocyte (p4) obtained by normal separation with M254 culture medium, digesting the cells (0.25% trypsin) when the cell fusion degree reaches 90%, centrifuging (800rpm/min, 5min), suspending the cells with PBS, irradiating the suspension with UVB dose of 150mJ/cm2, continuously culturing at a ratio of 1:3 after centrifugation for 48h, and placing in a refrigerator at-80 ℃ for later use.
(2) Obtaining of melanin granules: and (2) repeatedly freezing and thawing the melanocytes obtained in the step (1) for 3 times, collecting liquid, filtering, acidifying by adding citric acid, and centrifuging to obtain precipitates, namely melanin granules.
(3) Preparing a melanin hair dye:
the hair dye comprises three formulations, namely, formulation A: adding 4 parts of glutathione, 0.4 part of ascorbic acid (VC), 0.8 part of sodium carboxymethylcellulose, 1.5 parts of preservative (phenoxyethanol), 2 parts of essence, 3 parts of ammonia water and the balance of deionized water in sequence by weight of 100 parts, and uniformly mixing.
And (2) agent B: adding 4 parts of ferric chloride, 0.7 part of ascorbic acid (VC), 0.8 part of sodium carboxymethylcellulose, 1.5 parts of preservative (phenoxyethanol), 2 parts of essence, 0.02 part of citric acid and the balance of deionized water in sequence by weight of 100 parts, and uniformly mixing.
C, agent C: adding 0.6 part of ascorbic acid (VC), 2 parts of melanin, 0.8 part of sodium carboxymethylcellulose, 1.5 parts of preservative (phenoxyethanol), 2 parts of essence, 5 parts of ammonia water and the balance of deionized water in sequence by weight of 100 parts, and uniformly mixing.
The use method of the melanin hair dye comprises the following steps: firstly, uniformly coating the agent A on hair, wrapping the hair with a preservative film, and preserving the temperature for 30min at 35 ℃; then smearing the agent B on hair, wrapping with preservative film, and keeping the temperature at 30 ℃ for 30 min; finally, the agent C is smeared on the hair, wrapped by a preservative film and kept at the temperature of 25 ℃ for 50 min.
Hair dyeing and evaluation
(1) Compared with the hair dye sold in the market, the hair dye of the invention
Washing equivalent white hair with shampoo, mixing two commercially available hair dyes, namely, the Cardamom pleasant hair dye cream glittering pearl black A agent and the B agent uniformly, spreading the mixture on the hair, and washing off the loose hair with clear water after 40 min.
An equal amount of white hair was taken, and after being washed with shampoo, the hair dye of the present invention was used to dye hair according to the procedure of example 3.
Then, white hair, natural black hair of human, the effect dyed with the melanin hair dye prepared in example 3, and the effect dyed with a commercially available hair dye were observed under a hair mirror.
The results are shown in figure 1, and it can be seen from the results that the product has ideal hair dyeing effect on white hair, has small damage to the hair, and the dyed hair is closer to natural black hair of a human body.
(2) Hair dyeing and evaluation
(1) Dyeing the hair: the melanin hairdyes of examples 1 to 3 were applied to commercially available hair strands, respectively, according to the procedure of example 3, and the hair was dyed. Dyeing and dyeing of commercially available Hair dyes, comparative example 1 was followed by washing with commercially available 2-in-1 shampoo (Anlishing two-in-one shampoo) and drying naturally to give four "freshly dyed" hair strands.
The above four "just dyed" hair strand colors were tested using a YS3010 spectrocolorimeter manufactured by Chongqing Chonggao laboratory instruments ltd. The results of 10 measurements were averaged out, and the results are shown in Table 2.
(2) Evaluation of degree of discoloration: the hair tresses just dyed were washed 10 times and 20 times with commercially available 2 in 1 shampoos, respectively, and then naturally dried to give "wash 10 times" and "wash 20 times". The color difference of hair tresses "washed 10 times", "washed 20 times" and "immediately after dyeing" was evaluated by visual observation of 20 participants according to the following evaluation criteria, and the results were averaged as shown in table 2.
5 evaluation criteria for discoloration:
5-little color difference immediately after dyeing and after fading;
4, the color difference between the dyed cloth and the faded cloth is small;
3-moderate color difference immediately after dyeing and after fading;
2-the color difference between the dyed and faded is large;
the color difference immediately after 1-dyeing and after fading was very large.

As can be seen from the results in Table 1, the melanin hair dye of the present invention has a superior dyeing effect and is excellent in washing fastness, and can be used as a semi-permanent or permanent type hair dye.
While preferred embodiments of the present invention have been described, additional variations and modifications in those embodiments may occur to those skilled in the art once they learn of the basic inventive concepts. Therefore, it is intended that the appended claims be interpreted as including preferred embodiments and all such alterations and modifications as fall within the scope of the invention.
It will be apparent to those skilled in the art that various changes and modifications may be made in the present invention without departing from the spirit and scope of the invention. Thus, if such modifications and variations of the present invention fall within the scope of the claims of the present invention and their equivalents, the present invention is also intended to include such modifications and variations.
Claims (7)
1. A melanin hair dye is characterized by comprising a combination of an agent A, an agent B and an agent C;
the agent A is prepared from the following components in percentage by mass: 1-20% of penetrating agent, 0.1-2% of ascorbic acid, 0.1-2% of thickening agent, 0.01-10% of alkaline agent, 0.3-2% of preservative, 0.0-3% of essence and the balance of water;
the agent B is prepared from the following components in percentage by mass: 1-10% of metal ions, 0.1-2% of ascorbic acid, 0.1-2% of thickening agent, 0.01-1% of citric acid, 0.01-10% of alkali, 0.3-2% of preservative, 0.0-3% of essence and the balance of water;
the agent C is prepared from the following components in percentage by mass: 0.1-2% of ascorbic acid, 0.1-2% of melanin granules, 0.1-2% of thickening agent, 0.01-10% of alkaline agent, 0.3-2% of preservative, 0.0-3% of essence and the balance of water;
the melanin granules are made from primary human melanocytes, trypsin and citric acid.
2. The melanin hair dye according to claim 1, wherein the melanin granules are prepared by a method comprising:
s1, melanocyte culture: culturing primary human melanocyte until cell fusion degree reaches 90%, digesting cells with trypsin digestive juice with mass fraction of 0.25%, centrifuging at 800rpm/min for 5min, suspending cells with PBS, and centrifuging at 50-200mJ/cm2The suspension was irradiated at a UVB dose of (1: 2-1: 3 for 24-48h to obtain melanocytes for later use;
s2, obtaining melanin granules: repeatedly freezing and thawing the melanocyte obtained in S1 for 3-4 times, collecting liquid, filtering, acidifying with citric acid, centrifuging, and obtaining precipitate, namely melanin granules.
3. The melanin hair dye according to claim 1, wherein the penetrating agent is at least one selected from the group consisting of: l-cysteine and glutathione.
4. The melanin hair dye according to claim 1, wherein the metal ions are at least one selected from the group consisting of: ferric chloride, magnesium chloride, zinc sulfate, copper sulfate, ferrous chloride, aluminum chloride, alum, and copper nitrate.
5. The melanin hair dye according to claim 1, wherein the thickener is at least one selected from the group consisting of: hydroxyethyl cellulose, carboxymethyl cellulose, hydroxypropyl methyl cellulose, xanthan gum, guar gum, sodium alginate, sodium hyaluronate, gelatin, and sodium alginate.
6. The melanin hair dye according to claim 1, wherein the alkaline agent is at least one selected from the group consisting of: ammonia, ammonium bicarbonate, sodium hydrogen phosphate, sodium hydroxide, potassium hydroxide, ethanolamine, triethanolamine, arginine, and lysine.
7. The method of using a melanin hair coloring agent of claim 1, comprising the steps of: uniformly coating the hair dye A on hair, sealing with preservative film, and dyeing at 25-40 deg.C for 20-50 min; uniformly coating the hair dye B on hair, sealing with a preservative film, and dyeing at 25-40 deg.C for 20-50 min; then uniformly coating the hair dye C on the hair, sealing with a preservative film, and dyeing at 25-40 deg.C for 20-50 min.
