Abstract
本发明公开了一种抗衰老中药组合物,其特征在于其原料组成及配比为:紫灵芝10‑50份、葡萄籽10‑50份、大麦1‑20份、牡丹皮5‑30份、人参1‑25份;中药组合物的提取物的制备方法为:按配比称取中药原料紫灵芝、葡萄籽、大麦、牡丹皮、人参,适当粉碎,混合,加入提取溶剂,50‑80℃加热提取0.5‑3h,提取1‑3次,过滤,合并滤液,减压浓缩至无醇味,再上大孔树脂柱纯化,收集50%‑95%酒精洗脱部分,减压浓缩至提取液与药材比为1∶0.5~1∶10,加丁二醇溶解,过滤,制得中药组合物的提取物。用该方法获得的中药组合物的提取物能强效清除自由基,抑制酪氨酸酶活性,减少黑色素生产,加快皮肤表层细胞更新,延缓肌肤衰老,添加于化妆品中,抗衰老功效显著,天然无刺激,健康好肤色。
Description
技术领域
本发明涉及一种中药组合物,具体的说是一种抗衰老中药组合物及制备方法和其提取物在化妆品中的应用。
背景技术
皮肤和其他器官一样,随着年龄不断增加,都逐渐趋向衰老。皮肤的衰老是一种正常现象,从我们出生开始,皮肤老化的现象就一直存在,皮肤老化的结果,除了使皮肤的生理功能减退外,最重要的是使人的外貌发生了变化。人们往往通过皮肤及头发的一些外在特征,来判断一个人的年龄、身份和地位。衰老使美丽减色,也会对人的心理产生负面效应,因此人人都希望保持外貌年轻、青春永驻。
引起皮肤衰老的原因很多,但归根结底都与人体中的自由基有关。生物体的衰老过程是机体的组织细胞不断产生的自由基积累的结果,自由基可以引起DNA损伤从而导致突变,诱发肿瘤和人体老化。自由基是正常代谢的中间产物,其反应能力很强,可使细胞中的多种物质发生氧化,损害生物膜。自由基形成后,它们可以进攻和损伤皮肤组织细胞结构,并引起如下变化:长链分子(如胶原蛋白、弹性纤维和染色体物质)中的累积性氧化变异,使皮肤逐渐失去弹性和张力,皱纹不断增加;粘多糖(如透明质酸)分解和细胞间质(如神经酰胺)流失,使皮肤干燥角化;惰性物质的积累和衰老色素(如脂褐素)的积累;脂质过氧化引起细胞膜和质膜的变化;动脉和毛细血管的纤维化;酶活力降低和免疫力降低,促进衰老。
预防和延缓皮肤衰老是美容皮肤科学的重要课题。开发效果明显、安全、稳定、吸收好的抗衰老活性物,已成为当前美容行业的热点,而植物来源、天然的中药抗衰老活性物是其主要研究方向。
发明内容
本发明的目的就是提供一种天然安全的强效抗衰老中药组合物以及该中药组合物提取物的制备方法。
为达到本发明的目的,本发明采用了如下技术方案:
一种抗衰老中药组合物,其特征在于包含如下重量百分比的组分:紫灵芝10-50份、葡萄籽 10-50份、大麦1-20份、牡丹皮5-30份、人参1-25份。
上述抗衰老中药组合物提取物的制备方法,其特征在于:按配比称取中药原料紫灵芝、葡萄籽、大麦、牡丹皮、人参,适当粉碎,混合,加入提取溶剂,50-80℃加热提取0.5-3h,提取1-3次,过滤,合并滤液,减压浓缩至无醇味,再上大孔树脂柱纯化,收集50%-95%酒精洗脱部分,减压浓缩至提取液与药材比为1 ∶ 0.5 ~1 ∶ 10,加丁二醇溶解,过滤,制得中药组合物提取物。本发明中使用的提取溶剂可采用水、乙醇、乙醇水溶液。
本发明的第二目的是将上述抗衰老中药组合物的提取物用于抗衰老化妆品中,制出性质稳定、效果良好的抗衰老化妆品。
为实现本发明的第二目的,本发明采用了如下技术方案:将上述中药组合物的提取物用于配制化妆品,其特征在于化妆品中含有上述中药组合物提取物,其用量为0.1-25%,优选用量为1-10%。
本发明的有益效果是:紫灵芝提取物具有非常强效的抗自由基功能和良好的收敛止血、消肿生肌功效;葡萄籽提取物含有丰富的花青素、维生素E和氨基酸,是非常强效的自由基清除剂,而且还具有非常好的抗紫外线、抗辐射和抗过敏作用;大麦富富含生物类黄酮,具有非常强的抗自由基功效和抑制酪氨酸酶活性的作用,能预防雀斑及老年斑;牡丹皮含有丰富的丹皮酚,是非常强效的自由基去除剂,同时也是非常强效的酪氨酸酶抑制剂和防晒剂,具有强效的抗衰老、美白和防晒功能;人参含有人参皂苷,具有强效消除自由基和抑制酪氨酸酶活性作用,具有非常好的修复受损组织细胞、淡化疤痕、利水消肿、美白之功效。以上诸药合用,可协同增效,共奏补气生血,荣养肌肤之功,强体质之本。本发明的中药组合物提取物能强效清除自由基、抑制酪氨酸酶活性,减少黑色素生成,加快皮肤表层细胞更新,延缓肌肤衰老,添加于化妆品中,抗衰老功效显著,天然无刺激,健康好肤色。
具体实施方式
以下通过具体实施例来对本发明作进一步的说明,但这些实施例只是为了说明,不是对本发明的限制。
实施例1 中药组合物提取物(Ⅰ)的制备
取紫灵芝25g、葡萄籽30g、大麦8g、牡丹皮25g、人参12g,适当粉碎,混合,加入50%酒精1.5L ,70℃加热提取两次,每次2h,过滤,合并滤液,减压浓缩至无醇味。以上提取液再上AB-8大孔树脂柱纯化,收集70%酒精洗脱部分,在60℃条件下减压浓缩得80mL浓缩液,再加入20mL丁二醇,过滤,制得100mL中药组合物提取液。
实施例2 中药组合物提取物(Ⅱ)的制备
取紫灵芝25g、葡萄籽25g、大麦5g、牡丹皮25g、人参20g,适当粉碎,混合,加入50%酒精1.5L ,70℃加热提取两次,每次2h,过滤,合并滤液,减压浓缩至无醇味。以上提取液再上AB-8大孔树脂柱纯化,收集70%酒精洗脱部分,在60℃条件下减压浓缩得80mL浓缩液,再加入20mL丁二醇,过滤,制得100mL中药组合物提取液。
实施例3 抗衰老营养霜
表1给出含有本发明的中药组合物提取物的抗衰老营养霜配方。

制备方法:A相80℃溶解混合均匀,B相80℃溶解混合均匀,环聚二甲基硅氧烷乳化前加入B相,乳化时B相加入A相并均质5min,降温至50℃时加入C相并均质2分钟,陈化24h后包装,即可。
对比例1 普通膏霜
表2给出不含有本发明的中药组合物提取物的普通膏霜配方。

制备方法:A相80℃溶解混合均匀,B相80℃溶解混合均匀,环聚二甲基硅氧烷乳化前加入B相,乳化时B相加入A相并均质5min,降温至50℃时加入C相并均质2分钟,陈化24h后包装,即可。
实施例4 实验方法和抗衰老功能评价
1、试样的配制
将中药提取物配成浓度为1%的样品溶液,进行下面实验。
2、对酪氨酸酶活性抑制能力评价
按照表3加样,不加酪氨酸酶,将1%熊果苷作为一组样品进行对比。将溶液置于37℃水浴中恒温10min;加入酪氨酸酶,将溶液置于37℃水浴中恒温10min后,于475nm下测量其吸光度值,并与1%熊果苷进行比较。

按式(1)计算样品对酪氨酸酶的相对抑制率 (I,简称抑制率,下同)。

式中:A 1、A 2、A 3、A 4分别为1组~4组溶液的吸光度。
3、对超氧阴离子自由基清除能力评价
取0.05mol/L pH=8.2的Tris-HCl缓冲液4.5mL,于25℃水浴锅中预热20min。再加入1mL试样和0.4mL 25mmol/L的邻苯三酚溶液,混匀后,于25℃水浴中反应5min,加入8mol/L的HCl 1.0mL终止反应。以Tris-HCl 缓冲液作参比,在299nm处测吸光度值。空白对照用1mL试样的溶剂来替代样品。按照式(2)计算清除率(D)。

式中A 1为空白对照的吸光度值;A 2为药物的吸光度值。
4、对羟自由基的清除能力评价
在25mL比色管中依次加入2mmol/L FeSO43mL,1mmol/L H2O2 3mL,摇匀,接着加入6mmol/L水杨酸3mL,摇匀,于37℃水浴加热15min后取出,测其吸光度;分别加入一定浓度的待测液,摇匀,继续水浴加热15min,取出测其吸光度。下式为待测液对羟自由基(·OH)的清除率:

式中A1为未加药品前反应体系的吸光度值;A2为药品清除·OH后体系的吸光度值;A3为空白对照清除·OH后体系的吸光度值。
5、实验结果
抗衰老功能评价实验如表4所示。

实验表明,本发明的中药组合物能强效清除自由基,并有效地抑制酪氨酸酶活性,具有良好的抗衰老功能。
实施例5 人体试用评价实验
1、实验方法
实施例3及对比例1制备的膏霜分别进行人体试用评价。选取30-50岁的志愿者40名,均为女性。使用方法为每日洁面后使用,每天两次(早晚各一次),左脸使用按实施例3制备的膏霜,右脸使用按对比例1制备的膏霜。实验期为一个月,使用一个月后观察左脸和右脸的肤色。
2、实验结果
使用一个月后发现,有35名使用者使用了按实施例3制备的膏霜的左脸的肤色明显好于使用了按对比例1制备的膏霜的右脸的肤色,有效率达87.5%。

1.一种抗衰老中药组合物,其特征在于包含如下重量百分比的组分:紫灵芝10-50份、葡萄籽 10-50份、大麦1-20份、牡丹皮5-30份、人参1-25份;中药组合物的提取物的制备方法为:按配比称取中药原料紫灵芝、葡萄籽、大麦、牡丹皮、人参,适当粉碎,混合,加入提取溶剂,50-80℃加热提取0.5-3h,提取1-3次,过滤,合并滤液,减压浓缩至无醇味,再上大孔树脂柱纯化,收集50%-95%酒精洗脱部分,减压浓缩至提取液与药材比为1 ∶ 0.5 ~1 ∶10,加丁二醇溶解,过滤,制得中药组合物提取物;提取溶剂为水、乙醇、乙醇水溶液。
2.权利要求1所述的中药组合物的提取物的制备方法为:按配比称取中药原料紫灵芝、葡萄籽、大麦、牡丹皮、人参,适当粉碎,混合,加入提取溶剂,50-80℃加热提取0.5-3h,提取1-3次,过滤,合并滤液,减压浓缩至无醇味,再上大孔树脂柱纯化,收集50%-95%酒精洗脱部分,减压浓缩至提取液与药材比为1 ∶ 0.5 ~1 ∶ 10,加丁二醇溶解,过滤,制得中药组合物提取物;提取溶剂为水、乙醇、乙醇水溶液。
3.权利要求1所述的中药组合物的提取物在配制化妆品中的应用,其特征在于化妆品中含有上述中药组合物提取物,其用量为0.1-25%,优选用量为1-10%。
