Abstract
The invention discloses a medical pressure-sensitive adhesive with good air permeability and application of ectoin in improving the air permeability of the medical pressure-sensitive adhesive, and provides a medical pressure-sensitive adhesive containing ectoin and a wound dressing. The invention finds a new effect that the ectoin can improve the air permeability of the medical pressure-sensitive adhesive, and can relieve or improve the problems of skin irritation, allergy and the like caused by poor air permeability of the medical pressure-sensitive adhesive by adding the ectoin into the medical pressure-sensitive adhesive. Compared with the existing medical pressure-sensitive adhesive, the medical pressure-sensitive adhesive added with the ectoin has the advantages of good air permeability, no residue, no sensitization and the like, and can effectively prevent the skin allergy problem caused by the medical pressure-sensitive adhesive.
Description
Technical Field
The invention relates to a new application of ectoin, namely a new application of improving the air permeability of a medical pressure-sensitive adhesive, and also relates to a medical pressure-sensitive adhesive containing ectoin and having good air permeability and a wound dressing, belonging to the technical field of medical materials.
Background
The medical pressure-sensitive adhesive is an adhesive material which can be adhered under slight pressure and is easy to peel, is called pressure-sensitive adhesive, is commonly called non-setting adhesive, is called pressure-sensitive adhesive for short, and comprises three types of water-soluble pressure-sensitive adhesive, solvent-based pressure-sensitive adhesive and hot-melt pressure-sensitive adhesive. The medical pressure-sensitive adhesive is widely applied to the fields of various adhesive tapes, bandages, infusion pastes, adhesive bandages, plasters, etc., and plays an important role in medical care.
Skin allergies are a common skin problem and are often caused by irritation of the skin. The air permeability is a main factor influencing the skin allergy to the medical pressure-sensitive adhesive, because the medical pressure-sensitive adhesive raw material with poor air permeability contains no polar substances, the effects of moisture absorption and sweat release cannot be achieved, and the skin is easily stimulated to cause allergy because sweat cannot be discharged; the residual glue or solvent of the medical pressure-sensitive adhesive is left on the skin and is possibly another factor causing the skin to be allergic to the medical pressure-sensitive adhesive, the adhesive force of the medical adhesive tape is not too strong, otherwise, glue is easy to remain on the skin, or skin pain is easy to cause when the medical adhesive tape is peeled off; the holding power cannot be too poor, otherwise the adhesive is easy to fall off. Therefore, it should be kept within an appropriate range.
Disclosure of Invention
Aiming at the problem of poor air permeability of the pressure-sensitive adhesive, the problem to be solved urgently is to improve the air permeability of the pressure-sensitive adhesive. Ectoin, namely tetrahydropyrimidine, tetrahydromethylpyrimidine carboxylic acid, and the english alias Ectoine. The amino acid is an annular amino acid, exists in a salt-tolerant microorganism, the living environment of the microorganism has the characteristics of high UV radiation, dryness, extreme temperature and high salinity, and the tetrahydro-methyl pyrimidine carboxylic acid can protect the structures of proteins and cell membranes in the environment, has a protective effect on adversity cells and prevents the microorganism from being damaged. The research shows that the ectoin is an amphoteric organic molecule with water locking function and a 'compatible solute', has good repairing and protecting effects on human skin, improves the immune protection capability of skin cells, increases the cell repairing capability or regenerates damaged skin. The effect of ectoin on improving the air permeability of the pressure-sensitive adhesive is not reported at present.
In the process of research, the inventor surprisingly discovers that the air permeability of the medical pressure-sensitive adhesive is greatly improved after the ectoin is added, and the performance of the ectoin in the aspect of improving the air permeability of the medical pressure-sensitive adhesive shows an unexpected effect. Therefore, the invention provides a new application of ectoin in improving the air permeability of the medical pressure-sensitive adhesive.
Furthermore, the ectoin is added in the form of raw materials when the medical pressure-sensitive adhesive is prepared, and is mixed with other raw materials to prepare the medical pressure-sensitive adhesive. Tests prove that the effect is better when the addition amount of the ectoin is 1-5% of the total mass of the raw materials of the medical pressure-sensitive adhesive. Wherein, the raw material of the medical pressure-sensitive adhesive refers to the sum of raw materials containing ectoin.
Furthermore, the ectoin may be any ectoin produced by any method, and is preferably ectoin produced by a microbial fermentation method. The method has low cost, is convenient for industrial production, and the obtained ectoin is safer.
Furthermore, the medical pressure-sensitive adhesive can be various types of medical pressure-sensitive adhesives, such as water-soluble pressure-sensitive adhesives, solvent-based pressure-sensitive adhesives, hot-melt pressure-sensitive adhesives and the like. When the medical pressure-sensitive adhesive is prepared, except that the ectoin is added into the raw materials, other preparation steps are carried out according to the existing method. Various types of medical pressure-sensitive adhesives show the same effect of enhancing air permeability after the addition of ectoin.
Furthermore, the invention also provides a medical pressure-sensitive adhesive with good air permeability, which contains the ectoin. Preferably, the content of the ectoine is 1-5% of the total mass of the raw materials of the medical pressure-sensitive adhesive. Wherein, the raw material of the medical pressure-sensitive adhesive refers to the sum of raw materials containing ectoin.
Furthermore, the medical pressure-sensitive adhesive contains ectoine, and the addition of the ectoine can obviously improve the air permeability of the medical pressure-sensitive adhesive on one hand, so that the stimulation of sweat on skin is reduced; on the other hand, the ability of the skin to resist external stimulation can be improved due to the repair and protection effects of ectoin on the skin. The two aspects take effect together, and can well improve the problems of skin irritation, allergy and the like caused by the medical pressure-sensitive adhesive.
Further, the type of the medical pressure-sensitive adhesive and the origin of the ectoin are the same as those described above.
Furthermore, the invention also provides a wound protection dressing which comprises a carrier and the medical pressure-sensitive adhesive with good air permeability. The wound protecting dressing may be adhesive tape, bandage, transfusion plaster, wound plaster, adhesive plaster, etc.
The invention finds a new effect that the ectoin can improve the air permeability of the medical pressure-sensitive adhesive, and can relieve or improve the problems of skin irritation, allergy and the like caused by poor air permeability of the medical pressure-sensitive adhesive by adding the ectoin into the medical pressure-sensitive adhesive. Compared with the existing medical pressure-sensitive adhesive, the medical pressure-sensitive adhesive added with the ectoin has the advantages of good air permeability, no residue, no sensitization and the like, and can effectively prevent the skin allergy problem caused by the medical pressure-sensitive adhesive.
Detailed Description
The present invention is described in further detail below with reference to specific examples. The following description is exemplary only and is not intended to limit the scope of the present invention.
Example 1 study on improvement of permeability of medical pressure-sensitive adhesive by Excretion
Selecting a hot-melt medical pressure-sensitive adhesive as a research object, taking the raw material of the medical pressure-sensitive adhesive, adding the ectoine into the medical pressure-sensitive adhesive according to the condition that the addition amount of the ectoine in the total raw material (the raw material comprises the ectoine) of the medical pressure-sensitive adhesive is 0wt%, 1 wt%, 3wt%, 5wt% and 7wt%, and then preparing the medical pressure-sensitive adhesive according to the method reported in patent CN 104449414A.
The medical pressure-sensitive adhesive with different ectoine contents is coated on a polyurethane non-woven fabric to prepare a pressure-sensitive adhesive tape, and the performance of the pressure-sensitive adhesive tape, including initial viscosity, permanent viscosity, 180-degree peel strength, water vapor permeability and peel integrity, is tested at 25 ℃, and the result is shown in table 1.
From the above results, it can be seen that ectoin has no influence on the initial tack of the medical pressure-sensitive adhesive, but has influences on the tack, peel property and air permeability of the medical pressure-sensitive adhesive. From the aspect of viscosity, when the addition amount of the ectoine is 1-5%, the viscosity is equivalent, and is slightly reduced compared with the case of not adding the ectoine; when ectoin was continuously added, the decrease in the permanent adhesion was significant. From the viewpoint of peeling property, the peel integrity after addition of ectoin was improved, but the peel strength was significantly reduced, wherein the reduction in peel strength was significant when the addition amount exceeded 5%. From the aspect of air permeability, the water vapor transmission rate of the medical pressure-sensitive adhesive can be obviously improved after the addition of the ectoine, and the water vapor transmission rate tends to increase along with the increase of the addition amount of the ectoine. The properties of all aspects are integrated, the performance of the medical pressure-sensitive adhesive is not obviously reduced when the addition amount of the ectoine is 1-5%, the air permeability of the medical pressure-sensitive adhesive is greatly improved, and the performance is excellent.
The pressure-sensitive adhesive tapes of numbers 1 to 5 were subjected to a patch test by the following method:
subject: 50 healthy volunteer subjects, male and female halves. The subjects were 23-53 years of age. Exclusion criteria: during pregnancy, lactation or parturition for half a year; ② acute or chronic diseases with the possibility of interfering with the test results or increasing the risk of participation of the subject; ③ the drugs which can interfere the test result are taken orally or externally (such as glucocorticoid, non-retention anti-inflammatory drug, antihistamine drug and immunosuppressant); fourthly, the patient is diagnosed with chronic allergic dermatosis or skin tumor within about 12 months and is taken; the spot-stuck part has sunburn, uneven skin color, tattoo, acne, eczema, psoriasis, cloth mark and other skin disease subjects which may influence the observation.
Test materials: the pressure-sensitive adhesive tapes of nos. 1 to 5 described above.
The test method comprises the following steps: the test subjects were divided into 5 groups of 10 persons each, and one pressure-sensitive adhesive tape was used for each group. Cutting each pressure sensitive adhesive tape into 25mm × 25mm, applying to the arm curve side of the subject, lightly pressing with palm to uniformly apply on the skin, with the same application position for each subject, removing the pressure sensitive adhesive tape at intervals of 24h, and observing the skin state. The skin condition interpretation criteria were referenced to International Contact Dermatitis Research Group, ICDRG) scoring criteria, as shown in table 2.
After the adhesive tape is attached and removed for 24 hours, the pressure sensitive adhesive tape added with the ectoin has no adhesive residue on the arm of the testee, and the pressure sensitive adhesive tape not added has adhesive residue on the arm of the testee. The skin test results are shown in table 3.
From the above results, it can be seen that the addition of ectoin can prevent the occurrence of sensitization reaction of the medical pressure-sensitive adhesive.
Example 2
The hot-melt medical pressure-sensitive adhesive in example 1 was replaced with a water-soluble pressure-sensitive adhesive and a solvent-based pressure-sensitive adhesive, and the effects of the addition of ectoin on the air permeability and skin irritation of the pressure-sensitive adhesive were studied in the same manner as in example 1, and the expression rule was the same as that in example 1.

5. Use according to claim 1, characterized in that: the medical pressure-sensitive adhesive comprises a water-soluble pressure-sensitive adhesive, a solvent-type pressure-sensitive adhesive and a hot-melt pressure-sensitive adhesive.
6. The medical pressure-sensitive adhesive with good air permeability is characterized in that: the medical pressure-sensitive adhesive contains ectoin.

The medical pressure-sensitive adhesive as claimed in claim 6, wherein: the content of the ectoin is 1-5% of the total mass of the raw materials of the medical pressure-sensitive adhesive, wherein the raw materials of the medical pressure-sensitive adhesive refer to the sum of the raw materials containing the ectoin.
8. The medical pressure-sensitive adhesive as claimed in claim 6, wherein: the ectoin is prepared by a microbial fermentation method.
9. A wound protection dressing is characterized in that: the medical pressure-sensitive adhesive with good air permeability, which comprises a carrier and any one of claims 6 to 8.
10. The wound care dressing of claim 9, wherein: the types of the plaster include adhesive tapes, bandages, transfusion plasters, adhesive bandages, adhesive plasters and plaster plasters.
