Abstract
The invention belongs to the technical field of animal feed, and particularly discloses a compound polysaccharide feed additive for improving the quality of cock semen, a preparation method and an application thereof, wherein the compound polysaccharide feed additive for improving the quality of cock semen is obtained by mixing agaricus blazei polysaccharide, lycium barbarum polysaccharide, seaweed selenium polysaccharide and a compound polysaccharide stabilizer; the compound polysaccharide feed additive for improving the quality of the semen of the cocks is added into basic ration of the cocks. The compound polysaccharide feed additive for improving the sperm quality of the cocks can improve the sperm quality and the oxidation resistance level of the cocks. The preparation method is suitable for preparing the compound polysaccharide feed additive for improving the quality of the semen of the cocks, and the compound polysaccharide feed additive for improving the quality of the semen of the cocks is suitable for being added into basic daily ration of the cocks to feed the cocks.

(1) Test animal selection and grouping
Selecting 40 feathers of healthy Hailan brown parent breeding cocks of 44 weeks old in a certain chicken farm in Tianjin, and dividing the cocks into a control group and a test group after balancing according to the weight, the amount of sperms, the sperm motility and the sperm density; each group had 4 replicates, each replicate 5 feather cocks.
(2) Daily ration and feeding management
Preparing a basic ration according to the NRC and the chicken feeding standard in China by matching the nutrition required by the breeding cocks, feeding the basic ration by a control group, feeding the test ration by adding 0.1 percent of the compound polysaccharide feed additive for improving the sperm quality of the cocks in the example 3 into the basic ration by a test group, and freely feeding and drinking water in the test process and carrying out conventional immunization; after the control group and the test group are both fed with the basic ration for pre-testing for 1 week, the test group starts to feed the test daily ration, the control group starts to continue to feed the basic daily ration, and the test period is 5 weeks, namely 45 weeks to 50 weeks.

the semen density calculation method comprises the following steps:
number of sperm per ml = total number of sperm per 5 squares × 5 × 10 × 1000 × dilution;
measuring the sperm motility: sucking semen diluted by 0.3% NaCl solution by using a dropper, dropping the semen on a glass slide, covering a cover glass, ensuring that no bubbles exist in the semen, observing the semen under a microscope, and calculating the number of the linearly movable sperms in a visual field;
the sperm motility calculation method comprises the following steps:
sperm motility = number of sperm moving straight ahead/total number of sperm in visual field × 100%;
fourthly, measuring the sperm teratogenesis rate: taking semen smears, performing microscopic examination on teratospermia after Giemsa staining, smearing 2 pieces of sperm on each sample, counting 500 sperms on each piece, and calculating the deformity rate;
determination criteria of teratospermia: teratospermia mainly refers to giant head, thin head, defect, double head, pear-shaped head, head protrusion, neck enlargement, mitochondria enlargement, with primary particles, no tail, tail curl and other curls;
the method for calculating the sperm teratogenesis rate comprises the following steps:
sperm aberration (%) = number of teratospermic sperm/500 × 100;
fifthly, measuring the activity of glutathione peroxidase in seminal plasma: the activity of glutathione peroxidase in seminal plasma is measured by a kit provided by Nanjing institute of bioengineering, Inc.
