Abstract
The invention provides a hair tonic, which is prepared from the following raw materials: 45-50 wt% of geranic acid, 25-32 wt% of choline, 20-25 wt% of PBS buffer solution and 0.001-0.1 wt% of keratinocyte growth factor. According to the invention, the ratio of the geranic acid to the choline is adjusted, the geranic acid and the choline are used as carriers of the keratinocyte growth factor, the permeability of the skin can be increased, and a relatively stable ionic environment is provided by matching with a PBS (phosphate buffer solution), so that the keratinocyte growth factor can permeate the skin, the activity of the keratinocyte growth factor is ensured, the proliferation and differentiation of hair follicle stem cells and hair matrix cells are further stimulated, the hair follicle neogenesis and maturation are promoted, and the hair growth is realized. The results of the examples show that the hair tonic provided by the invention is uniformly applied to the skin of an adult black mouse which shaves back hair, and the shaved part mostly restores to the length and density before shaving within three weeks.
Description
Technical Field
The invention belongs to the technical field of daily cosmetics, and particularly relates to hair tonic and a preparation method thereof.
Background
Seborrheic alopecia, known as androgenetic alopecia, accounts for about 90% of alopecia patients, has a alopecia rate and range varying from person to person, and has only mild alopecia, or can be persistent for many years, or can reach the alopecia of old people in short years, and generally progresses fastest in about 30 years. The products for preventing alopecia and growing hair in China are brought into the scope of special product production license management, and the research progress of alopecia and growing hair is slow in the last decades due to a series of reasons.
The existing medicine for treating alopecia is minoxidil liniment, which is applied to an affected part of a head, can expand subcutaneous blood vessels of the head after being absorbed by the scalp, improve microcirculation around hair follicles, reduce inflammatory cell infiltration around the hair follicles, and delay aging of epithelial stromal cells, so that the atrophic hair follicles are enlarged and the growth of hairs is promoted. However, the external medicine of the liniment needs to be administrated through the skin, and the epidermis, particularly the horny layer is the main barrier layer of the skin, and researches show that only a few medicines can enter the skin, the stability is poor, the biological activity is difficult to maintain for a long time, and the liniment is difficult to play a good hair growth effect.

Detailed Description
The invention provides hair tonic, which is prepared from the following raw materials: 45 to 50 weight percent of geranic acid, 25 to 32 weight percent of choline, 20 to 25 weight percent of PBS buffer solution and 0.001 to 0.1 weight percent of keratinocyte growth factor.
The raw material of the hair tonic provided by the invention comprises 45-50 wt% of geranic acid, preferably 48-50 wt%, more preferably 48.192-48.197 wt%, and more preferably 48.195 wt%. In the invention, the purity of the geranic acid is preferably more than or equal to 85 percent. In the present invention, when the purity of the geranic acid is less than 85%, the purification of the geranic acid is preferably performed in the present invention. The purification operation is not particularly limited, and the purity of the geranic acid can be improved to more than 85%. The source of the geranic acid is not particularly limited in the present invention, and commercially available products known to those skilled in the art may be used. In the invention, the geranic acid can perform double decomposition reaction with choline to obtain the ionic liquid, so that the skin transmittance of the keratinocyte growth factor (KGF-2) is promoted, and the hair growth effect is improved.
The raw material of the hair tonic provided by the invention comprises 25 wt% -32 wt% of choline, preferably 29 wt% -32 wt%, more preferably 29.075 wt% -29.077 wt%, and more preferably 29.076 wt%. The source of choline is not particularly limited in the present invention, and commercially available products well known to those skilled in the art may be used. In the invention, the choline can perform double decomposition reaction with the geranic acid to obtain the ionic liquid, so that the skin transmittance of KGF-2 is promoted, and the hair growth effect is improved.
The raw material of the hair tonic provided by the invention comprises 20 wt% -25 wt% of PBS buffer solution, preferably 22 wt% -25 wt%, more preferably 22.723 wt% -22.725 wt%, and more preferably 22.724 wt%. The source of the PBS buffer is not particularly limited in the present invention, and commercially available products known to those skilled in the art may be used. In the invention, the PBS buffer solution can not only dissolve KGF-2, but also provide a relatively stable ionic environment, thereby avoiding the inactivation of KGF-2 and playing a role in protecting KGF-2.
The raw material of the hair tonic provided by the invention comprises 0.001 wt% -0.1 wt%, preferably 0.001 wt% -0.05 wt%, more preferably 0.001 wt% -0.005 wt%, and even more preferably 0.003 wt% of keratinocyte growth factor. The source of the keratinocyte growth factor in the present invention is not particularly limited, and commercially available products known to those skilled in the art may be used. In the invention, the keratinocyte growth factor can stimulate the proliferation and differentiation of hair follicle stem cells and hair matrix cells, thereby promoting the neogenesis and maturation of hair follicles and realizing the hair growth effect.

The invention also provides a preparation method of the hair tonic in the technical scheme, which comprises the following steps:
(1) mixing geranic acid and choline to perform double decomposition reaction to obtain ionic liquid;
(2) drying the ionic liquid obtained in the step (1) to obtain oily liquid;
(3) mixing a keratinocyte growth factor with a PBS buffer solution to obtain a growth factor solution;

The drying operation is not particularly limited in the present invention, and may be a drying operation known to those skilled in the art. In the present invention, the drying is to remove moisture in the ionic liquid.
The invention mixes the keratinocyte growth factor and the PBS buffer solution to obtain the growth factor solution. The preparation method provided by the invention is easy to operate, simple in process and low in cost, and is very suitable for rapid and large-scale production.

(2) drying the ionic liquid obtained in the step (1) at 60 ℃ for 48h in vacuum to obtain oily liquid;
(3) dissolving 0.005 wt% KGF-2 in 22.724 wt% PBS buffer solution to obtain growth factor solution;
(4) and (3) uniformly mixing the oily liquid obtained in the step (2) and the growth factor solution obtained in the step (3) to obtain the hair tonic.
Example 2
(1) 48.197 wt% of geranic acid (with the purity of 90%) and 29.077 wt% of choline are taken to be stirred in a round-bottom flask at room temperature until no carbon dioxide gas is generated, so that ionic liquid is obtained;
(2) drying the ionic liquid obtained in the step (1) at 60 ℃ for 48h in vacuum to obtain oily liquid;
(3) dissolving 0.001 wt% KGF-2 in 22.725 wt% PBS buffer solution to obtain growth factor solution;
(4) and (3) uniformly mixing the oily liquid obtained in the step (2) and the growth factor solution obtained in the step (3) to obtain the hair tonic.
Example 3
(1) 48.192 wt% of geranic acid (with the purity of 90%) and 29.075 wt% of choline are taken to be stirred in a round-bottom flask at room temperature until no carbon dioxide gas is generated, so that ionic liquid is obtained;
(2) drying the ionic liquid obtained in the step (1) at 60 ℃ for 48h in vacuum to obtain oily liquid;
(3) dissolving 0.010 wt% KGF-2 in 22.723 wt% PBS buffer solution to obtain growth factor solution;
(4) and (3) uniformly mixing the oily liquid obtained in the step (2) and the growth factor solution obtained in the step (3) to obtain the hair tonic.
Comparative example 1
(1) 23.477 wt% of geranic acid (the purity is 90%) and 28.815% of choline are taken to be stirred in a round-bottom flask at room temperature until no carbon dioxide gas is generated, so as to obtain ionic liquid;
(2) drying the ionic liquid obtained in the step (1) at 60 ℃ in vacuum for 48h to obtain oily liquid;
(3) dissolving 0.005 wt% KGF-2 in 47.703 wt% PBS buffer solution to obtain a growth factor solution;
(4) and (3) uniformly mixing the oily liquid obtained in the step (2) and the growth factor solution obtained in the step (3) to obtain the hair tonic.
The hair tonic prepared in example 1 and control groups 1 to 3 (wherein the control group 1 is a PBS buffer solution, the control group 2 is a mixed solution of an ionic liquid composed of choline and geranic acid and the PBS buffer solution, and the control group 3 is a growth factor solution, that is, KGF-2 is dissolved in the PBS buffer solution) were applied to animal experiments to examine the effects thereof.
The results of uniformly applying 4 drops of the solutions of example 1 and control groups 1-3 to the skin of an adult black mouse whose back hair has been shaved off are shown in fig. 1-4, in which fig. 1 is a comparison graph of a black mouse without the product of control group 1 and with the product of control group 1 for 7 days, 14 days and 21 days; FIG. 2 is a graph comparing mice that were not coated with control 2 product, and that were coated with control 2 product for 7 days, 14 days, and 21 days; FIG. 3 is a graph comparing mice that were not coated with control group 3 product, and that were coated with control group 3 product for 7 days, 14 days, and 21 days; FIG. 4 is a graph comparing mice without the product of example 1 applied, with the product of example 1 applied for 7 days, 14 days and 21 days.
As can be seen from fig. 1 to 4, the hair growth effect of the shaved portion of the mice of example 1 was the best and the back hair was largely restored to the length and density before shaving in three weeks as compared with the control groups 1 to 3, demonstrating that the hair tonic of the present invention can increase the hair growth rate.
The products prepared in examples 1-3 and comparative example 1 are respectively applied to the dry scalp of 12 people, 1mL of each product is applied and massaged to promote drug absorption, and the application is performed twice a day for two months. The length and density of the hair were measured and the data are shown in table 1.

As can be seen from Table 1, after the hair growth oil is applied on the product for two months, compared with the comparative example 1, the hair growth oil disclosed by the invention can better promote the hair follicle to grow and mature, and the excellent hair growth effect can be realized within the range of the proportion of each component of the hair growth oil provided by the invention.
As can be seen from the above examples, the hair tonic provided by the present invention has an excellent hair tonic effect.

2. The hair tonic of claim 1, prepared from the following raw materials: 48 to 50 weight percent of geranic acid, 29 to 32 weight percent of choline, 22 to 25 weight percent of PBS buffer solution and 0.001 to 0.05 weight percent of keratinocyte growth factor.
3. The hair tonic of claim 2, prepared from the following raw materials: 48.192-48.197 wt% of geranic acid, 29.075-29.077 wt% of choline, 22.723-22.725 wt% of PBS buffer solution and 0.001-0.005 wt% of keratinocyte growth factor.
4. The hair tonic of any one of claims 1 to 3, wherein the geranic acid has a purity of 85% or more.
5. A process for the preparation of a hair tonic according to any one of claims 1 to 4, comprising the steps of:
(1) mixing geranic acid and choline to perform double decomposition reaction to obtain ionic liquid;
(2) drying the ionic liquid obtained in the step (1) to obtain oily liquid;
(3) mixing a keratinocyte growth factor with a PBS buffer solution to obtain a growth factor solution;
(4) mixing the oily liquid obtained in the step (2) with the growth factor solution obtained in the step (3) to obtain hair tonic;
the steps (1) - (2) and the step (3) have no sequence.
6. The production method according to claim 5, wherein the mixing in the step (1) is performed under stirring.
7. The method of claim 5, wherein the metathesis reaction in step (1) terminates without carbon dioxide gas generation.
8. The method according to claim 5, wherein the temperature of the drying in the step (2) is not more than 70 ℃.
9. The method according to claim 8, wherein the temperature for drying in the step (2) is 50 to 60 ℃.
10. The method according to claim 5, wherein the drying time in the step (2) is not less than 36 h.
