背景技术
对于乙型肝炎病毒(HBV:Hepatitis B Virus)而言,目前可利用的治疗方法中有效的方法少,需要开发新的治疗方法。就此而言,近年来,已知乙型肝炎病毒(HBV)或丁型肝炎病毒(HDV:Hepatitis D Virus)通过介由牛磺胆酸钠共转运多肽(NTCP:SodiumTaurocholate Cotransporting Polypeptide)侵入细胞来感染人,从而导致乙型肝炎发病。对此,作为通过抑制NTCP来治疗乙型肝炎的尝试,例如,对使用环孢菌素A(CsA:Cyclosporin A)、Myrcludex B进行了研究(例如,非专利文献1)。

发明要解决的课题
然而,上述以往的抑制剂不仅抑制HBV或HDV介由NTCP而侵入细胞,也抑制如输送胆汁酸等这样的NTCP自身的功能(例如,非专利文献2~5)。
因此,本发明所要解决的课题为提供基于环肽的新型NTCP抑制剂。
用于解决课题的手段
本申请的发明人为了解决上述课题而反复进行了深入研究,结果发现,具有特定结构的环肽抑制HBV或HDV介由NTCP而侵入细胞,从而完成了本发明。

[图6]为示出所鉴定的肽中Met密码子误读的评价的图。就有些由针对NTCP亲和性的最初筛选中鉴定得到的肽而言,尽管在翻译反应中Met不存在,但包含Met密码子。为了评价于Met密码子处错误地读取了哪个氨基酸,通过Met缺失翻译反应对各cDNA进行翻译,利用MALDI-TOF MS确定其质量。观察到与Thr(*)或对应的误读及与它们对应的K加合物(**及)。为了评价各误读对于NTCP键合的影响,通过引物组装来合成各个克隆,针对与仅固定于磁性微珠或微珠的NTCP的亲和性,就亲和选择自身进行评价(即,相关的mRNA嘌呤霉素、体外翻译、亲和选择及基于实时RT-PCR的定量化)。D1(M5I)、D4(M12L)及L4(M10T)全部基于之后验证的目的而选择。

本说明书中,“环肽”表示在分子内至少具有由4个以上氨基酸形成的环状结构。作为环肽的分子结构,除了环状结构以外,还可以具有氨基酸经由肽键连接而成的链状结构,并且,也可以具有肽结构以外的结构。
本说明书中,“环状结构”表示直链肽中相隔2个氨基酸残基以上的2个氨基酸直接键合或介由接头等键合从而在分子内形成的闭环结构。
本说明书中,“相隔2个氨基酸残基以上”表示在2个氨基酸之间存在至少2个残基的氨基酸。
环状结构中的闭环结构没有特别限定,可由2个氨基酸通过共价键合而形成。
作为2个氨基酸间的共价键,可举出例如二硫键、肽键、烷基键、链烯基键、酯键、硫酯键、醚键、硫醚键、膦醚键、偶氮键、C-S-C键、C-N-C键、C=N-C键、酰胺键、内酰胺桥联、氨甲酰键、脲键、硫脲键、胺键、及硫酰胺键等。
2个氨基酸在氨基酸的主链中键合时,可利用肽键而形成闭环结构,也可通过2个氨基酸的侧链彼此的键合、侧链与主链的键合等来形成2个氨基酸间的共价键。
环状结构不限于通过直链肽的N末端与C末端的氨基酸的键合形成,也可以通过末端的氨基酸与末端以外的氨基酸的键合、或末端以外的氨基酸彼此的键合来形成。用于形成环状结构而键合的氨基酸中的一者为末端氨基酸、另一者为非末端氨基酸的情况下,环肽具有直链肽呈尾状附着于环状结构而成的结构。
作为形成环状结构的氨基酸,除了天然氨基酸以外,还包括人工的氨基酸突变体、衍生物,可举出例如天然蛋白质L-氨基酸、非天然氨基酸、及具有氨基酸特征的本领域中已知特性的化学合成的化合物等。
蛋白质性氨基酸(proteinogenic amino acids)以本领域已知的三字母表示法而表示时,为Arg、His、Lys、Asp、Glu、Ser、Thr、Asn、Gln、Cys、Gly、Pro、Ala、Ile、Leu、Met、Phe、Trp、Tyr、及Val。
作为非蛋白质性氨基酸(non-proteinogenic amino acids),表示蛋白质性氨基酸以外的天然或非天然的氨基酸。
作为非天然氨基酸,可举出例如:主链结构与天然型不同的α,α-二取代氨基酸(α-甲基丙氨酸等)、N-烷基-α-氨基酸、D-氨基酸、β-氨基酸、α-羟酸;侧链结构与天然型不同的氨基酸(正亮氨酸、高组氨酸等);在侧链具有多余的亚甲基的氨基酸(“高”氨基酸、高苯丙氨酸、高组氨酸等);及侧链中的羧酸官能团被磺酸基取代的氨基酸(半胱氨酸等);等等。作为非天然氨基酸的具体例,可举出国际公开第2015/030014号中记载的氨基酸。
形成环状结构的氨基酸的数目为4个以上即可,没有特别限定,例如可以为5个以上、8个以上、10个以上,也可以为30个以下、25个以下、20个以下、15个以下。
作为形成环状结构的氨基酸的数目,优选为4个以上且30个以下,在4个以上且30个以下的范围内,形成环状结构的氨基酸的数目可以设为5个以上、8个以上、10个以上,也可以设为30个以下、25个以下、20个以下、15个以下。
形成环状结构的氨基酸的数目可以设为8个以上且20个以下,可以设为10个以上且20个以下,也可以设为10个以上且15个以下。
本发明中,环肽可以施加磷酸化、甲基化、乙酰基化、腺苷酰化、ADP核糖基化、及糖链加成等修饰,也可以与其他的肽、蛋白质等融合。另外,环肽可以介由适当的接头而进行生物素化。
另外,本发明中,环肽可以是将2个具有1个环状结构的环肽介由接头结构键合而成的、在分子内具有2个环状结构的二聚体,也可以具有在分子内形成有内酰胺结构的分子内内酰胺桥联结构。
作为将2个环肽连接的接头结构,没有特别限定,可以采用肽合成领域中作为将肽彼此连接的接头而周知的结构。
分子内内酰胺桥联结构可以通过构成环肽的氨基酸的侧链彼此键合而形成,例如,Lys侧链的氨基与Asp或Glu的侧链的羧基键合而形成肽键,由此形成分子内内酰胺结构,环肽可以在分子内作为桥联结构而具有另一个环结构。例如可以使DAP、DAB、及Orn代替Lys而与Asp或Glu键合。
本发明为基于环肽的新型NTCP抑制剂。特别地,本发明为针对NTCP自身的功能的抑制作用更弱的NTCP抑制剂、更强地抑制HBV或HDV与NTCP的相互作用的NTCP抑制剂、或者是兼具上述两者的NTCP抑制剂。
本发明中,认为具有Xw1-W-Xw2-W结构、或具有P-Xp1-Xp2-H结构的环肽介由Xw1-W-Xw2-W结构或P-Xp1-Xp2-H结构与NTCP键合,变构地抑制NTCP的功能的至少一部分。其中,本发明中的NTCP抑制剂优选作为用于抑制HBV或HDV向细胞内侵入的侵入抑制剂而发挥功能。
HBV的生命周期大致分成针对细胞的感染初期和感染后期的阶段。更具体而言,在感染初期,HBV与肝细胞上的NTCP键合,介由NTCP侵入细胞,被输送至核,形成cccDNA(共价闭环DNA,covalently closed circular DNA)。另外,在感染后期中,进行自cccDNA向RNA的转录、衣壳形成、反转录、包膜获得、病毒释放。
本发明的NTCP抑制剂通过抑制NTCP的功能的至少一部分来抑制HBV或HDV的感染初期的与NTCP的键合和介由NTCP的细胞侵入,可以用于治疗或预防与HBV或HDV相关的疾病。
本发明的NTCP抑制剂抑制NTCP、优选人NTCP的至少一部分。
作为与HBV或HDV相关的疾病,可举出例如急性肝炎、慢性肝炎、爆发性肝炎、肝硬化、肝衰竭、及肝细胞癌。
本发明中,环肽可以以药学上可接受的盐的形式使用。作为环肽的药学上可接受的盐,可举出例如与作为药学上可接受的碱、酸所形成的盐。

本发明的环肽可利用基于无细胞翻译系统的翻译合成方法而适当地制造。
可以通过制备编码环化的肽的核酸,并在无细胞翻译系统中翻译该核酸来制备。编码环化的肽的核酸可以使用在生物体的翻译系统中所使用的遗传密码、经过重编程的遗传密码或它们的组合,本领域技术人员可适当进行设计。核酸可以是DNA,也可以是RNA。
根据使用无细胞翻译系统的方法,使用经非天然氨基酸而氨基酰化的tRNA,除了天然氨基酸之外,还能够向肽中高效地导入非天然氨基酸。例如,使用本申请的发明人开发的人工氨基酰tRNA合成酶flexizyme,即能够用任意的天然或非天然的氨基酸对具有任意的反密码子的tRNA进行氨基酰化。因此,使用该技术,能够以使mRNA三联体形成的遗传密码编码与生物体的翻译系统不同的氨基酸的方式进行重编程(国际公开第2008/059823号)。
本发明的NTCP抑制剂可以以含有其的医药组合物的形式使用。
医药组合物的施予形态没有特别限定,可以是经口施予,也可以是非经口施予。作为非经口施予,可举出例如肌肉内注射、静脉内注射、及皮下注射等注射施予、经皮施予、以及经粘膜施予等。
作为经粘膜施予的施予途径,可举出例如经鼻、经口腔、经眼、经肺、经阴道、及经直肠等。
从代谢、排泄等药物动力学的观点考虑,可以对医药组合物中的环肽进行各种修饰。例如,可以向环肽加成聚乙二醇(PEG)、糖链从而使血中滞留时间较长,降低抗原性。
另外,可以使用聚乳酸·乙二醇(PLGA)等生物体内降解性的高分子化合物、多孔羟基磷灰石、脂质体、表面修饰脂质体、用不饱和脂肪酸制备的乳液、纳米颗粒、纳米球等作为缓释化基剂,使环肽内包于上述物质。经皮施予的情况下,可以在皮肤表面流通弱电流而使环肽透过角质层(离子电渗透(iontophoresis)法)。

以下,示出实施例来具体说明本发明,但本发明不限于以下的实施例。
<材料及方法>
材料
Fmoc保护氨基酸购自MercK Millipore。寡核苷酸购自Eurofins GenomicsJapan。只要没有特别指出,则其他试剂由Nacalai Tesque,Inc.获得。
RaPID选择
使用已知的方法(Morimoto,J.,Hayashi,Y.&Suga,H.Discovery of MacrocyclicPeptides Armed witHa Mechanism-Based Warhead:Isoform-Selective Inhibition ofHuman Deacetylase SIRT2.Angew.Chem.Int.Ed.Engl.51,3423-3427(2012);及Kawakami,T.,Murakami,H.&Suga,H.Messenger RNA-programmed incorporation of multiple N-methyl-amino acids into linear and cyclic peptides.Chem.Biol.15,32-42(2008)中记载的方法),合成氰基甲基酯(CME)或二硝基苄基酯(DBE)活化氨基酸。氨基酰化核酶flexizyme及tRNA如下制备:由PCR生成DNA模板(下述表1),使用CMC或DBE活化氨基酸进行tRNA的氨基酰化,利用其进行体外转录(Goto,Y.,Katoh,T.&Suga,H.Flexizymes forgenetic code reprogramming.Nat.Protoc.6,779-790(2011).)。具体而言,RNA分子通过基于利用PCR组装的DNA模板的T7 RNA聚合酶反应而合成(表1),利用PAGE进行纯化。在冰上添加40mM的各tRNA底物及氨基酰化核酶flexizyme、600mM的MgCl、2.5mM的活化氨基酸底物及100mM的HEPES-KOH(pH7.5)2小时(N-氯乙酰基-酪氨酸-CME、N-甲基-甘氨酸-DBE、N-甲基-丙氨酸-DBE)或6小时(N-甲基-4-O-甲基-酪氨酸-CME、N-甲基-丝氨酸-DBE、N-甲基-苯丙氨酸-CME及N-甲基-亮氨酸-DBE),然后,在乙酸钠及乙醇中沉淀并回收。将嘌呤霉素键合寡核苷酸(表1)及T4 RNA连接酶通过苯酚:氯仿提取及乙醇沉淀进行纯化,制备嘌呤霉素键合mRNA。

环肽文库的核糖体使用已知的方法(Hayashi,Y.,Morimoto,J.&Suga,H.In vitroselection of anti-Akt2 thioether-macrocyclic peptides leading to isoform-selective inhibitors.ACS Chem.Biol.7,607-613(2012).中记载的方法)合成。具体而言,为了进行最初的选择,于37℃将1.2μM的嘌呤霉素键合mRNA文库在含有25μM的L-或D-ClAc-Tyr-tRNAfMet的Met缺失FIT反应中进行30分钟翻译。将其产物于25℃孵育12分钟后,在20mM的EDTA的存在下,于37℃孵育30分钟,由此破坏核糖体-mRNA复合体。然后,于42℃将得到的肽键合mRNA使用RNaseH-反转录酶(Promega)进行1小时反转录,将缓冲液更换为50mM的Tris-HCl(pH7.4)、150mM的NaCl、0.05vol%的吐温-20及1mM的βDDM。实施His pull-down Dynabeads(Life Technologies公司,商品名)上的6代连续传代(于4℃各30分钟的反选择),接着,于4℃针对固定于相同微珠的200nM的NTCP实施30分钟亲和选择。于95℃加热5分钟,使cDNA从微珠溶出,通过在LightCycler thermal cycler(Roche公司)上实施使用了Sybr Green I的定量PCR,对来自最终反选择(阴性对照)及亲和选择步骤的分别回收进行评价。利用PCR回收经富集的DNA文库,作为生成下一轮筛选所用mRNA文库的转录反应的输入而使用。为了进行后述的聚焦选择,除去下述FIT反应而进行选择。就该FIT反应而言,缺失Met、Asp、Glu及Arg以及它们的同族氨基酰tRNA合成酶,包含6.25μM的L-或D-ClAc-Tyr-tRNAfMet以及各12.5μM的EnGluGUC-MeGly、EnGluCUC-MeAla、EnGluGCG-MeTyr(Me)、EnGluCCG-MePhe及EnGluCCU-MeSer。
为了确定最初选择的序列,将DNA试样使用DNA连接试剂盒Mighty Mix(TakaraBio公司)克隆至pGEM-T-easy载体(Promega公司),转化至感受态DH5a E.Coli,确定了来自各试样的24个克隆的序列。为了进行经聚焦的选择物的高通量测序,使用巢引物(表1)将DNA试样进行PCR扩增,使用Nucleospin色谱柱(Machery-Nagel公司)纯化,使用MiSeq高通量测序仪(Illumina公司)对序列进行确定。数据分析使用CLC Workbench软件(Qiagen公司)进行。

环肽使用Syro Wave自动合成仪基于标准的Fmoc固相合成而进行合成。为了合成N-甲基-O-甲基-酪氨酸残基,在O-甲基-酪氨酸偶联后,如下进行Fmoc去保护、得到的游离胺的硝基苯磺酰基保护、及使用了硫酸二甲酯的树脂上的甲基化。Fmoc去保护:将N-甲基-2-吡咯烷酮(NMP)及2,4,6-三甲基吡啶(33μL,0.25mmol)中的硝基苯磺酰氯(22mg,0.1mmol)添加于树脂,使反应进行15分钟,于室温洗涤树脂,将树脂用NMP洗涤5次。使用NMP中的1,8-二氮杂双环(5.4.0)十一碳-7-烯(11.25μL,0.075mmol)及硫酸二甲酯(22.25μL,0.25mmol)5分钟以实施2次甲基化,然后洗涤树脂5次。)用NMP置换。实施2次硝基苯磺酰基去保护(在NMP中使用1,8-二氮杂双环(5.4.0)十一碳-7-烯(18.75μL,0.125mmol)及2-巯基乙醇(17.5μL,0.25mmol)处理5分钟,)后,将树脂用NMP洗涤5次,继续进行固相肽的合成。最后的Fmoc去保护工序之后,通过与8当量的氯乙酰基-N-羟基-丁二酰胺于室温反应1小时,从而将各肽的N末端氯乙酰化。使用三氟乙酸(TFA)中的各三异丙基硅烷、乙二硫醇及水2.5容量%,实施自树脂的切断及侧链去保护。用溶解于二甲基亚砜(DMSO)的乙醚进行沉淀并回收肽,用N,N,N-三乙胺制成碱性并成环。使用水:乙腈梯度(含有0.1%TFA),利用Prominence HPLC系统(Shimadzu公司)实施。将最终TFA盐由5mM的HCl水溶液冷冻干燥3次而得到HCl盐,为了进行下一验证,将其溶解于DMSO,进行纯化。
表面等离子体共振
使用Biacore T200装置(GE Healthcare公司)测定了各肽相对于人NTCP的键合常数。电泳缓冲液(running buffer)使用了含有0.1容量%的DMSO及1mM的β-十二烷基-麦芽糖苷的HBS-P+(10mM的HEPES、150mM的NaCl、及0.05容量%的表面活性剂P20,pH7.4)。按照制造者的指示(基线响应:1000~3000个折射单位),将100nM的重组NTCP固定于经Ni2+处理的NTA芯片上。将肽作为分析物以60μL/分钟的流速注入,使用1:1键合模型将键合常数模型化。
重组NTCP蛋白的制备

HBV的制备和感染

HDV感染分析
如已知的方法(Kaneko,M.等人,A Novel Tricyclic Polyketide,VanitaracinA,Specifically Inhibits the Entry of Hepatitis B and D Viruses by TargetingSodium Taurocholate Cotransporting Polypeptide.Journal of virology 89,11945-11953(2015)中记载的方法)那样,由经表达质粒转染的Huh-7细胞的培养上清制备HDV(HDV表达质粒pSVLD3由Fox Chase Cancer Center的John Taylor博士提供)。在4%PEG8000的存在下,将HDV于37℃以5GEq/细胞接种16小时(图4(b)),测定细胞内HDV RNA,由此对感染进行评价。

<结果>
针对NTCP的初始RaPID选择
为了验证环肽对HBV的细胞侵入进行抑制,利用使用了经纯化的His加成NTCP(关于其结构及功能,至少一部分可以由Watashi,K.等人的Cyclosporin A and its analogsinhibit hepatitis B virus entry into cultured hepatocytes through targeting amembrane transporter,sodium taurocholate cotransporting polypeptide(NTCP).Hepatology 59,1726-1737(2014)及上述的<材料及方法>的记载中确认)作为靶标的RaPID系统实施选择,得到肽文库。得到的肽文库是由包含AUG起始密码子、4~15个连续的NNK密码子的随机区域(N=A、G、C或T、K=T或G)、Cys密码子、及编码Gly-Ser接头的3次重复的序列的准随机的DNA模板通过转录及翻译而生成的(图5(b))。使用了将各肽的起始formyl-Met)取代为L-或D-N-氯乙酰化Tyr后、与下游的Cys反应形成硫醚键的环状结构这样的进行了基因重编程的翻译系统(图5(b))。如此,制备了各自含有大于1012种环肽的2个文库,进行了筛选(一个为L-起始,一个为D-起始)。
在进行7次选择后,根据第5次、第6次、及第7次后的经富集的DNA文库的序列分析可知,10个肽为共通或同源(6个为L-文库,4个为D-文库。下述表2)。其中,在内部包含Met密码子的3个(NTCP-D1、D4、L4)由于Met被从翻译系统中排除而被Ile/Leu及/或Thr取代。上述情况已通过MALDI-TOF MS对在FIT系统中翻译的特定的肽进行测定而得到了确认(图6)。对以包含Ile、Leu或Thr密码子代替Met密码子的方式突变的这些肽的各个克隆与NTCP的亲和性进行分析,上述分析中M5I、M12L及M10T突变体分别使用了NTCP-D1、NTCP-D4及NTCP-L4的活性形态,仅观察到键合亲和性的微小变化(图6)。

为了进一步表征鉴定得到的环肽,利用不含(GS)3接头序列的固相肽方法(且针对NTCP-D1、D4、L4各自伴有作为Met突变体的活性体)合成表2所示的10个肽。利用表面等离子体共振评价上述各肽针对NTCP的键合常数(表2及图7)。3个肽以大于100nM的KD值显示出更弱的键合,与此相对,一些肽(NTCP-D2、D3、L1、L3、L4、L5及L6)以4~50nM的解离常数(KD)显示出强键合。
接着,本申请的发明人对上述10种肽针对原代人肝细胞中的HBV感染的效果进行了评价。这些分析中,利用肽将细胞进行2小时前处理,然后,在肽存在下接种HBV16小时后,进行洗涤,在不存在肽的条件下进一步将细胞培养12天(图2(a))。HBV感染通过测定培养上清中的HBs抗原而进行评价。使用了通过与NTCP键合来抑制HBV侵入的天然型环肽CsA作为阳性对照(图2(b))。在所有情况下,100μM的各肽的处理均未伴随显著的细胞损伤作用,且使HBs抗原水平(图2(b))显著减少(图2(c))。这表明,新发现的包含硫醚-大环的全部肽具有抗HBV活性。
经聚焦的文库RaPID选择
通过筛选及活性验证中的自第一轮起的肽序列的检查,本发明的申请人确认了HBV感染性分析中活性最强的肽中,NTCP-D2及D4这2个肽包含PX(I/L)H(I(T/I)W(T/I)W基序(以下,称为“W基序”),其他3个肽、即NTCP-L1、L4及D1包含(T/L)W(T/I)P基序(以下,称为“P基序”)。根据该观察,为了基于P或W基序构建4个经聚焦的肽文库(2个L-ClAc-Tyr起始及2个D-ClAc-Tyr起始),使用了FIT翻译(图8)。这些文库是使用下述遗传密码而生成的,所述遗传密码是将Met、Asp、Glu及Arg用N-甲基-Leu、Ser、Gly、Ala及Phe、及N-甲基-O-甲基-Tyr取代并进行重编程而得到的。这些文库可以用于对鉴定而得到的肽的“药物相似性”(例如,更为良好的肽酶耐性及辛醇-水分配系数)进行改善。在使用NTCP作为诱饵(bait)的文库运行7次RaPID选择后,使用高通量测序仪对自各轮选择得到的DNA文库进行分析。就自最后一轮的选择而鉴定得到的肽序列的比对而言,在全部4个文库中显示出相同序列的强富集。由此,为了进行化学合成及其后的表征(表2中的WD1-4、WL1-6、PD1及PL1),基于相对浓缩及序列多样性,选择了12个肽。利用表面等离子体共振(表2及图9)对上述各肽相对于NTCP的键合动力学进行评价,大部分显示出10~100nM的范围的KD值。
接下来,本申请的发明人针对作为目标的包含W基序及P基序的来自最初的RaPID选择的肽、以及自经聚焦的文库RaPID选择鉴定得到的12个肽的影响,以10μM的浓度对原代人肝细胞的HBV感染进行了评价(参见上述)。如图3所示,通过聚焦筛选而鉴定得到的12个环肽中的10个使HBV抗原显著减少,其中的7个显示出比在初始筛选中鉴定得到的肽更高的效力。特别地,WD1、WL2、及WL4肽未显示显著的细胞损伤效果(图3(b)),并且最显著地使HBV抗原减少(图3(a))。
具有比环孢菌素A更高的抗HBV活性的环肽
为了评价WD1、WL2、及WL4这3个肽的抗HBV活性,过量表达人NTCP基因,使用HBV易感性HepG2-hNTCP-C4细胞,通过已知的方法(Iwamoto,M.等人,Evaluation andidentification of hepatitis B virus entry inhibitors using HepG2 cellsoverexpressing a membrane transporter NTCP.Biochemical and biophysicalresearcHcommunications 443,808-813(2014)中记载的方法)实施HBV感染分析。HBV接种后,在培养上清中测定HBs抗原,使用细胞评价HBc抗原,如图2(a)所示,确认了HBV感染。该分析中,CsA使利用HBs及HBc抗原监测的HBV感染减少,但作为HBV复制抑制剂而已知的核苷类似物恩替卡韦未显示出显著的减少。使用WD1、WL2、及WL4的处置并未显著地显示细胞毒性(图1(a)),并且使细胞内的HBc(图1(b))及上清中的HBs水平降低(图1(c))。尤其是,WD1、WL2及WL4肽的抗HBV效果高于由最初的筛选而得到的D1及D2的抗HBV效果。
为了在更为生理性的条件下评价抗HBV活性,使用原代人肝细胞进行HBV感染分析,使用了纯化HBV作为接种材料(参见上述<材料及方法>)。利用各肽的处理使培养上清中的HBs抗原水平剂量依赖性地减少(图1(d))。经计算,该分析中的WD1、WL2、WL4及CsA的50%抑制浓度(IC50)分别为0.85±0.01,2.54±0.11、0.66±0.03及3.17±0.30μM。这表明,该实验中发现的硫醚-大环具有比CsA高出最大约5倍左右的抗HBV效果。
不影响NTCP转运体功能的HBV细胞吸附的抑制
接下来,本申请的发明人对在WD1、WL2及WL4环肽中观察到的抗HBV效果是否来自于对病毒向细胞侵入的抑制进行了确认。HBV生命周期由包括吸附、侵入、核内转移、及形成共价键合的环状DNA(cccDNA)的感染初期阶段、以及作为后期阶段的包括转录、衣壳形成、反转录、包膜获得、病毒释放的HBV复制过程形成(图10,Watashi,K.等人,Cyclosporin Aand its analogs inhibit hepatitis B virus entry into cultured hepatocytesthrougHtargeting a membrane transporter,sodium taurocholate cotransportingpolypeptide(NTCP).Hepatology 59,1726-1737(2014);及Locarnini,S.&Zoulim,F.Molecular genetics of HBV infection.Antiviral therapy 15Suppl 3,3-14(2010))。为了调查肽对于HBV生命周期后期阶段带来的效果,使用了在不存在四环素的条件下重现HBV复制、而由于NTCP表达缺失而不会发生感染初期阶段的Hep38.7-Tet细胞。在存在或不存在肽的条件下,通过对除去四环素后的细胞内HBV DNA进行6天的定量,对HBV复制进行评价。在上述条件下,与作为阳性对照使用的核苷类似物恩替卡韦不同,3个环肽针对HBV DNA水平几乎未显示出效果(图10(a))。

接着,对WD1、WL2、及WL4针对各种HBV株及相关的HDV的效果进行评价。如图4(b)及图4(c)所示,3个肽显著地抑制HDV(如HBV那样,NTCP依赖性地侵入细胞)的感染,对于HCV(非NTCP依赖性地侵入细胞)的感染未造成影响(Yan,H.等人,Sodium taurocholatecotransporting polypeptide is a functional receptor for human hepatitis B andD virus.eLife 1,e00049(2012))。作为结论,可知上述肽抑制NTCP依赖性的病毒感染。为了评价上述肽的临床应用的可能性,针对各种不同的HBV分离株,对效力最强的2个肽即WD1及WL4的抗HBV效果进行了调查。除了迄今为止观察到的针对HBV基因型D的抗病毒效果(图1)以外,WD1及WL4对于进行了试验的全部HBV基因型(A、B及C基因型)也是有效的(图4(d))。此外,这些肽也显著抑制了对核苷类似物具有耐性的、具有L180M/S202G/M204V突变的HBV分离株及具有疫苗逃逸G145R取代的HBV分离株的感染。因此,可以得出结论,本验证中鉴定的硫醚-环肽针对在临床上成为问题的各种HBV株广泛具有活性。
Claims (13)
1.牛磺胆酸钠共转运多肽(NTCP:Sodium Taurocholate CotransportingPolypeptide)抑制剂,其包括:具有Xw1-W-Xw2-W结构(Xw1及Xw2各自独立地为T、S、I、L或V、或者其N-烷基氨基酸)、或具有P-Xp1-Xp2-H结构(Xp1为任意的氨基酸,Xp2为I、L或V、或者其N-烷基氨基酸)的环肽或者其药学上可接受的盐。
2.如权利要求1所述的NTCP抑制剂,其中,环肽具有Xw1-W-Xw2-W-Xw3-W结构(Xw1及Xw2与上述定义相同,Xw3为T、S、I、L或V、或者其N-烷基氨基酸)。
3.如权利要求1或2所述的NTCP抑制剂,其中,Xw1为T或L或者其N-烷基氨基酸,Xw2为T或I或者其N-烷基氨基酸。
4.如权利要求1所述的NTCP抑制剂,其中,环肽具有P-Xp1-Xp2-H-Xp3-F(Xp1及Xp2与上述定义相同,Xp3为I、L或V、或者其N-烷基氨基酸)结构。
5.如权利要求4所述的NTCP抑制剂,其中,Xp2及Xp3各自独立地为I或L或者其N-烷基氨基酸。
6.如权利要求1~5中任一项所述的NTCP抑制剂,其中,环肽具有N-CO-CH2-S结构。
7.如权利要求6所述的NTCP抑制剂,其中,所述N来源于色氨酸的氨基。
8.如权利要求6或7所述的NTCP抑制剂,其中,所述S来源于半胱氨酸的巯基。
9.如权利要求1~8中任一项所述的NTCP抑制剂,其中,环肽具有由10~20个氨基酸构成的环结构。
10.如权利要求1~9中任一项所述的NTCP抑制剂,其中,NTCP为人NTCP。
11.如权利要求1~10中任一项所述的NTCP抑制剂,其是抑制乙型肝炎病毒(HBV)或丁型肝炎病毒(HDV)向细胞内侵入的侵入抑制剂。
12.医药组合物,其含有权利要求1~11中任一项所述的NTCP抑制剂。
13.如权利要求12所述的医药组合物,其用于治疗或预防与乙型肝炎病毒或丁型肝炎病毒相关的疾病。
