Abstract

本发明公开了一种用于防治美洲斑潜叶蝇的迷迭香精油杀虫剂,该杀虫剂是以迷迭香精油作为主要有效成分,配以溶剂、分散剂、保湿剂和稀释剂制成。本发明迷迭香精油杀虫剂对美洲斑潜蝇的各个虫态均具有明显杀虫效果,可用于美洲斑潜蝇的全面生物防治。相关内容目前未见报道。

Description

一种用于防治美洲斑潜叶蝇的迷迭香精油杀虫剂
技术领域
本发明属于生物农药领域,具体涉及一种用于防治美洲斑潜叶蝇的迷迭香精油杀虫剂。
背景技术
美洲斑潜蝇(Liriomyza sativae Blanchard),双翅目(Diptera),潜蝇科(Agromyzidae),是一种多食性入侵害虫,寄主范围极广,主要有丝瓜、黄瓜、冬瓜、豇豆、荷兰豆、菜心、白菜、荠菜、空心菜、菠菜、苋菜、番茄、芍药、菊花、万寿菊、百合等,其主要通过幼虫“潜道式”取食叶肉、雌成虫产卵器刺伤叶片后吸取汁液等方式造成叶片薄壁细胞损伤并引起细菌和真菌病害。
灭蝇按、阿维菌素、多杀菌素、甲氰菊酯、毒死蜱等化学合成农药常用于潜叶蝇的防治。但是过早预防性用药和超剂量用药造成了一系列3R问题(害虫再猖獗、害虫对农药抗性和农药残留对环境的污染)和对潜叶蝇的天敌种群无差别的杀伤,甚至造成其局部绝种。迷迭香是一种四季常绿的芳香植物,其起源于地中海地区,在当地有将其用作香料、调味料和药品的习俗。迷迭香成分相关研究发现,迷迭香中的一些成分具有杀幼虫、杀卵、杀成虫的特性,但是迷迭香精油对美洲斑潜蝇的相关毒力测定结果还未见报道,缺少田间应用的理论依据。
发明内容
本发明针对现有技术存在的问题,提供了一种用于防治美洲斑潜叶蝇的迷迭香精油生物杀虫剂。
为了实现上述目的,本发明采用的技术方案是:
一种用于防治美洲斑潜叶蝇的迷迭香精油杀虫剂,其各成分按体积百分数计为:迷迭香精油6-24%、溶剂6-24%、分散剂30%、保湿剂10%、稀释剂12-48%,其体积百分数之和为100%。
进一步地,用于防治美洲斑潜叶蝇卵的迷迭香精油杀卵剂,其各成分按体积百分数计为:迷迭香精油16-24%、溶剂16-24%、分散剂30%、保湿剂10%、稀释剂12-28%,其体积百分数之和为100%。
用于防治美洲斑潜叶蝇幼虫的迷迭香精油杀幼虫剂,其各成分按体积百分数计为:迷迭香精油12.5-18.75%、溶剂12.5-18.75%、分散剂30%、保湿剂10%、稀释剂22.5-35%,其体积百分数之和为100%。
用于防治美洲斑潜叶蝇蛹的迷迭香精油杀蛹剂,其各成分按体积百分数计为:迷迭香精油13.33-20%、溶剂13.33-20%、分散剂30%、保湿剂10%、稀释剂20-33.34%,其体积百分数之和为100%。
用于防治美洲斑潜叶蝇成虫的迷迭香精油杀成虫剂,其各成分按体积百分数计为:迷迭香精油6-9%、溶剂12.5-18.75%、分散剂30%、保湿剂10%、稀释剂22.5-35%,其体积百分数之和为100%。
其中,所述溶剂包括乙醇、丙酮、二甲基亚砜等常用无毒有机溶剂中的任意一种。
所述分散剂为0.05wt%的Tween 20或Tween 80的水溶液。
所述保湿剂为丁二醇、己二醇。
所述稀释剂为水。
所述迷迭香精油的制备方法包括如下步骤:
(1)立秋之后,采集迷迭香针状叶片,在清水中浸洗除去灰土等固体杂质后平铺摊开,在室温下自然风干,然后经粉碎机粉碎并过20目网筛,再转移至容器密封保存;
(2)将所得粉末转移至蒸馏装置中,然后按质量体积比1g:10mL在所得粉末中加水混匀,蒸馏4h,馏分经蛇形冷凝管冷凝后形成的油水混合物转移至分液漏斗中,静止分层,排出下层液体,保留上层淡黄色液体;
(3)用无水硫酸钠对所得淡黄色液体进行干燥处理,即得迷迭香精油,将其于棕色试剂瓶中密封并避光保存。
本发明的显著优点在于:
本发明迷迭香精油杀虫剂对美洲斑潜蝇的各个虫态均具有明显杀虫效果,可用于美洲斑潜蝇的全面生物防治。同时,与灭蝇胺、甲维盐等常用于美洲斑潜蝇防治的化学农药相比,本发明提供的迷迭香精油杀虫剂属于植物源农药,具有低毒、高效、安全环保和对环境友好的特点,在田间生产使用无任何残留成分,对靶标害虫以外的生物无任何毒害作用,可用于蔬菜、花卉等园艺产品的生产,且其储运方便、易保存,是合成化学杀虫剂的良好替代品。
具体实施方式
为了使本发明所述的内容更加便于理解,下面结合具体实施方式对本发明所述的技术方案做进一步的说明,但是本发明不仅限于此。
一种用于防治美洲斑潜叶蝇的迷迭香精油杀虫剂,其各成分按体积百分数计为:迷迭香精油6-24%、乙醇12%、0.05wt%的Tween 20水溶液30%、丁二醇10%、水12-48%,其体积百分数之和为100%。
所用迷迭香精油的制备方法包括如下步骤:
(1)立秋之后,采集迷迭香针状叶片,在清水中浸洗除去灰土等固体杂质后平铺摊开,在室温下自然风干,然后经粉碎机粉碎并过20目网筛,转移至容器密封保存;
(2)将所得粉末转移至蒸馏装置中,然后按质量体积比1g:10mL在所得粉末中加水混匀,蒸馏4h,馏分经蛇形冷凝管冷凝后形成的油水混合物转移至分液漏斗中,静止分层,排出下层液体,保留上层淡黄色液体;

试验例1:迷迭香精油杀卵剂对美洲斑潜蝇卵的毒力测定
将羽化笼中经过饥饿处理、正在交配且交配时间超过5分钟的2日龄成虫经过2mL离心管移入装有打顶芽的菜豆苗(10日龄)的尼龙网养虫笼内取食交配4小时,然后将被感染菜苗转移至空养虫笼内。3天后,用直径1cm的打孔器在叶子上以虫卵为中心进行打孔,并将叶饼浸入迷迭香精油含量为240 ml/L、160 ml/L、106.67 ml/L、71.11 ml/L和47.41 ml/L的迷迭香精油杀卵剂中各5秒钟,每个处理重复三次,并采用不含迷迭香精油的空白试剂作为对照;然后将处理后的叶饼放入内铺湿润滤纸的9cm直径的培养皿中,并进行密封,再置于26℃、相对湿度60%的养虫室内静待幼虫孵化,统计孵化卵数并计算卵的死亡率,其计算方法为:卵的死亡率=(卵总数-孵化的卵数)/卵总数×100%。
试验例2:迷迭香精油杀幼虫剂对美洲斑潜蝇3龄幼虫的毒力测定
试验例1中剩余虫卵继续放置至3龄幼虫从叶子钻出来时,马上收集作为试验用虫。取部分试验用虫,分别浸入迷迭香精油含量为187.5 ml/L、125 ml/L、83.33 ml/L、55.56 ml/L和37.04 ml/L的迷迭香精油杀幼虫剂中各5秒钟,每个处理重复三次,并采用不含迷迭香精油的空白试剂作为对照;然后将其移入内铺湿润滤纸的9cm直径的培养皿中,分别于处理24h和48h后观察记录死亡情况,并计算死亡率,其计算方法为:幼虫的死亡率=死虫数/总虫数×100%(死亡标准是:幼虫对细针尖的刺激无反应)。
试验例3:迷迭香精油杀蛹剂对美洲斑潜蝇虫蛹的毒力测定
将试验例2剩余试验用虫继续进行饲养,然后收集在2h内由幼虫化成的蛹,取部分虫蛹分别浸入迷迭香精油含量为200 ml/L、133.33 ml/L、88.89 ml/L、59.26 ml/L、39.51 ml/L的迷迭香精油杀蛹剂中各5分钟,每个处理重复三次,并采用不含迷迭香精油的空白试剂作为对照;然后移出,统计羽化蛹数并计算蛹的死亡率,其计算方法为:蛹的死亡率=(蛹总数-羽化的蛹数)/蛹总数×100%。
试验例4:迷迭香精油杀成虫剂对美洲斑潜蝇2日龄成虫的毒力测定
待试验例3剩余蛹孵化成虫后,对其进行饥饿处理,然后采用瓶膜法分别对2日龄雌雄成虫进行毒力测定,每个处理重复三次,并采用不含迷迭香精油的空白试剂作为对照。所用迷迭香精油杀成虫剂中迷迭香精油的含量为:90 ml/L、60 ml/L、40 ml/L、26.67 ml/L和17.78 ml/L。处理24h和48h后观察记录死亡情况,并计算死亡率,其计算方法为:成虫的死亡率=死虫数/总虫数×100%(死亡标准是:成虫对毛笔的触动无反应)。
各试验例数据经过SPSS 22.0处理,并计算半致死浓度LC50、95置信限等相关参数,结果见表1-5。校正死亡率计算方法如下:

实施例结果表明,迷迭香精油杀虫剂对美洲斑潜蝇的各个虫态都有明显杀虫效果,其中成虫、幼虫较卵和蛹对迷迭香精油杀虫剂更敏感,而雌蝇较雄蝇更敏感。其中,160ml/L、240ml/L的迷迭香精油杀卵剂的校正死亡率分别达到了(70.47±4.83)%和(91.97±4.19)%;125ml/L、187.5ml/L的迷迭香精油杀幼虫剂的48h校正死亡率分别达到(81.97±4.19)%和100.00%;133.33ml/L、200ml/L的迷迭香精油杀成虫剂的校正死亡率分别达到了(78.24±5.57)%、(97.65±2.32);90ml/L、60ml/L的迷迭香精油杀成虫剂的48h校正死亡率分别为(81.97±4.19)%和(93.52±5.02)%。
试验例5:同浓度迷迭香精油稀释剂和迷迭香精油杀成虫剂的室内防效对比
分别配制相同迷迭香精油含量的迷迭香精油稀释剂(将迷迭香精油用乙醇稀释制得)和迷迭香精油杀虫剂,然后参照试验例1-4的方法分别处理含卵叶饼、3龄幼虫、蛹和2日龄成虫,并计算死亡率和校正死亡率,结果见表6。

结果表明,同浓度迷迭香精油杀虫剂对美洲斑潜叶蝇卵、幼虫、3龄幼虫和2日龄成虫的校正死亡率要显著高于同浓度迷迭香精油稀释剂。
以上仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。

1.一种用于防治美洲斑潜叶蝇的迷迭香精油杀虫剂,其特征在于:所述杀虫剂中各成分按体积百分数计为:迷迭香精油6-24%、溶剂6-24%、分散剂30%、保湿剂10%、稀释剂12-48%,其体积百分数之和为100%。
2.根据权利要求1所述的迷迭香精油杀虫剂,其特征在于:所述溶剂包括乙醇、丙酮、二甲基亚砜中的任意一种。
3. 根据权利要求1所述的迷迭香精油杀虫剂,其特征在于:所述分散剂为0.05wt%的Tween 20或Tween 80的水溶液。
4.根据权利要求1所述的迷迭香精油杀虫剂,其特征在于:所述保湿剂为丁二醇、己二醇。
5.根据权利要求1所述的迷迭香精油杀虫剂,其特征在于:所述稀释剂为水。
6.根据权利要求1所述的迷迭香精油杀虫剂,其特征在于:所述迷迭香精油的制备方法包括如下步骤:
(1)立秋之后,采集迷迭香针状叶片,在清水中浸洗后平铺摊开,在室温下自然风干,然后粉碎并过20目网筛;
(2)按质量体积比1g:10mL在所得粉末中加水混匀,蒸馏4h,馏分经冷凝管冷凝后形成的油水混合物转移至分液漏斗中,静止分层,保留上层淡黄色液体;
