Description
Technical Field
The invention relates to a wound flushing fluid and a preparation method thereof, in particular to a high-efficiency antibacterial and anti-inflammatory biological polysaccharide flushing fluid and a preparation method thereof, and belongs to the field of medical instruments.
Background
In the surgical operation, the wound surface or the incision is treated by using normal saline or a flushing fluid prepared by mixing sodium carboxymethylcellulose with traditional Chinese medicine components, chitosan or other saccharides in most clinical cases. Compared with normal saline, the washing liquid has the effects of increasing adhesion resistance and the like, but provides a healing environment for the wound, and the components in the washing liquid have no outstanding effects of inhibiting bacteria, diminishing inflammation and promoting the wound to heal quickly.
The water-soluble sodium carboxymethyl cellulose is natural cellulose which is nontoxic, nonirritating and nonimmunity-antigenicity-free to human bodies, and is an important cellulose derivative obtained by chemical improvement. Has excellent biocompatibility, and is widely used in medical products due to its excellent effect of preventing tissue adhesion.
Lysozyme, also known as muramidase, is an alkaline enzyme that hydrolyzes mucopolysaccharides in bacteria. Lysozyme dissolves bacteria by breaking the beta-1, 4 glycosidic bond between N-acetylmuramic acid and N-acetylglucosamine in the cell wall, breaking down the cell wall insoluble mucopolysaccharide into soluble glycopeptides, causing the contents of the broken cell wall to escape. It can be used as a natural anti-infective substance with antibacterial effect. Has antibacterial, antiviral, hemostatic, repercussive, analgesic, and tissue recovery promoting effects.
According to the invention, the lysozyme is added into the washing fluid component prepared from the sodium carboxymethylcellulose as the raw material, so that the effects of efficiently inhibiting bacteria and diminishing inflammation and promoting rapid healing of the wound are increased while the wound is cleaned and tissue adhesion is prevented, and a physical barrier can be finally formed to provide a favorable growth environment for wound healing. Through patent search, no report of adding lysozyme in biological polysaccharide flushing fluid is found at home and abroad.
Disclosure of Invention
The invention provides a biological polysaccharide flushing fluid with high-efficiency bacteriostasis and anti-inflammation and a preparation method thereof, which can clean wounds, prevent tissue adhesion, increase the effects of high-efficiency bacteriostasis and anti-inflammation on the wounds and promote rapid healing of the wounds, and finally form a physical barrier to provide a favorable growth environment for wound healing.
In order to achieve the above purpose of the present invention, the technical scheme adopted by the present invention is as follows:
the biological polysaccharide flushing fluid has the sodium carboxymethylcellulose substitution degree of 0.6-1.0. Preferably, the degree of substitution of sodium carboxymethylcellulose is 0.9.
The biological polysaccharide flushing fluid comprises, by weight, 0.5-6% of sodium carboxymethylcellulose, 0.3-5% of lysozyme, 0.1-0.9% of sodium chloride, 0.1-0.3% of a preservative, 0.01-0.1% of an acid-base regulator and the balance ultrapure water.
Preferably, the biological polysaccharide flushing fluid with high-efficiency bacteriostasis and anti-inflammation is prepared from 1.0% by weight of sodium carboxymethylcellulose, 3.0% by weight of lysozyme, 0.9% by weight of sodium chloride, 0.1% by weight of preservative and 0.04% by weight of acid-base regulator.
As a preferable mode of the invention, the biological polysaccharide flushing fluid is prepared by adding 1.0 percent of sodium carboxymethylcellulose, 3.0 percent of lysozyme, 0.9 percent of sodium chloride, 0.1 percent of preservative and 0.04 percent of acid-base regulator to 100 percent of ultrapure water in percentage by weight.
The preparation method of the biological polysaccharide flushing fluid with high-efficiency bacteriostasis and anti-inflammation comprises the following steps:
(1) dispersing sodium carboxymethylcellulose in purified water, and completely stirring to obtain a solution A;
(2) dispersing lysozyme in purified water, and fully stirring and dissolving to obtain a solution B;
(3) mixing the A, B solutions, and adding sodium chloride and antiseptic to obtain solution C;
(4) adding an acid-base regulator to regulate the pH value of the solution C to be between 5.5 and 7.5.
Sampling and measuring the performances such as pH value, viscosity and the like, and filling, sealing, packaging and sterilizing after the performances are qualified to obtain the biological polysaccharide flushing fluid with high efficiency, bacteriostasis and anti-inflammation.
In research and development, applicants found that in surgical operations, the wound surface or the surgical incision is treated by using physiological saline or a washing liquid prepared by mixing sodium carboxymethylcellulose as a raw material with traditional Chinese medicine components, chitosan or other saccharides in clinic. Compared with normal saline, the washing liquid has the effects of increasing adhesion resistance and the like, but provides a healing environment for the wound, and the components in the washing liquid have no outstanding effects of inhibiting bacteria, diminishing inflammation and promoting the wound to heal quickly. Further, the applicant finds that the carboxymethyl cellulose with the degree of substitution of 0.6-1.0 can improve the lysozyme activity and has a surprising synergistic effect.
