3.3 stability test

Preparing 6 parts of cordycepin standard solution with the concentration of 18 mu g/mL 40mL according to the method shown in 1.1.1, respectively adding the cordycepin standard solution into 6 parts of 10mL dandelion cordyceps sinensis beverage, uniformly mixing, measuring the chromatographic peak area of cordycepin according to the chromatographic condition of 1.1.3, calculating the cordycepin content in the mixed sample according to a cordycepin standard curve, and according to the formula: the recovery rate is (standard sample measurement value-sample measurement value) ÷ standard amount is multiplied by 100%, and the standard recovery rate of the cordycepin is calculated to be 91.28%; the standard recovery rate of adenosine is calculated to be 92.33% and the standard recovery rate of chlorogenic acid is calculated to be 107.72% by the same measuring method, which shows that the method provided by the invention is accurate and reliable and can be used for detecting effective components in beverages.
The above results show that: the precision, reproducibility and recovery rate of the method provided by the invention meet the requirements of the method, and the solution is placed at room temperature for 10h, and the result after measurement is stable. Therefore, the method can be applied to the content determination of adenosine, cordycepin and chlorogenic acid in the health-care beverage, and provides a basis for market supervision.

The invention adopts DPPH method to measure the free radical scavenging activity of the cordyceps sinensis and dandelion drink. The maximum absorption wavelength of the DPPH ethanol solution is 517 nm. When a free radical scavenger is added into a DPPH standard solution, lone pair electrons are paired, the absorbance at 517nm is reduced, and the change degree and the free radical scavenging degree are in a linear relation, so the method can be expressed by the scavenging rate, and the higher the scavenging rate is, the stronger the scavenging capacity of the substance is.
1. Preparation of DPPH Standard solution
Adding a 100mL brown volumetric flask into a 4mgDPPH standard sample, dissolving with absolute ethyl alcohol, fixing the volume to a scale to obtain a DPPH standard solution with the concentration of 0.004%, and storing in a refrigerator at 4 ℃ in a dark place for later use.

As can be seen from Table 4, the DPPH free radical clearance rate of the commercially available brand herbal tea beverage is 90.18%, and when the addition amount of the vitamin C is 0.1g/100mL, the DPPH free radical clearance rate of the cordyceps dandelion beverage is 94.86%; when the addition amount of the vitamin C is 0.15g/100mL, the DPPH free radical clearance rate of the cordyceps sinensis and dandelion beverage is 95.25%, the color protection effect is comprehensively considered, and the addition amount of the vitamin C is selected to be 0.1g/100mL finally. The beverage prepared under the condition has obvious antioxidant effect.
