Abstract
The invention discloses a health beverage composed of cordyceps militaris and dandelion and a preparation method thereof, the beverage is prepared by taking cordyceps militaris sporocarp and dandelion as raw materials through extraction, concentration, addition and the like, and each 100mL of the health beverage comprises 1-10mL of cordyceps militaris extract, 1-5mL of dandelion extract, 1-5g of sweetener, 0.01-0.5g of color fixative and the balance of drinking water. The cordyceps militaris and dandelion health-care beverage provided by the invention has the advantages of obvious antioxidant effect, fresh and cool taste and appropriate warm and cold, and is very suitable for the consumption requirements of modern consumers on quick pace of life and high diet requirements. The invention utilizes the traditional Chinese medicine characteristics of the cordyceps militaris and the dandelion, fully exerts the anti-inflammatory and antioxidant effects thereof, does not cause side effects after long-term administration, namely, does not cause cold and cool caused by the independent eating of the dandelion after long-term administration, and has the advantages of easily obtained raw materials, obvious effect and very wide market prospect.
Description
Technical Field
The invention belongs to the field of biological food, and particularly relates to a health-care beverage consisting of cordyceps militaris and dandelion and a preparation method thereof.
Background
The cordyceps sinensis is a rare Chinese medicament in China, is sweet in taste and warm in nature, and has the effects of tonifying lung and kidney, stopping bleeding and reducing phlegm. Nowadays, the yield is less and less due to the damage of the growing environment caused by excessive mining. Compared with the prior art, the cordyceps militaris with similar efficacy can be produced in an industrial way. The 2014 approved cordyceps militaris as a new food raw material by the national committee of health care in China, and can be directly eaten and applied to the field of foods. The Cordyceps militaris mainly contains active ingredients such as Cordyceps polysaccharide, cordycepic acid, cordycepin, adenosine, pentostatin, ergosterol, SOD, unsaturated fatty acid, protein, trace elements and the like. Research shows that the cordyceps militaris has the functions of enhancing immunity, resisting oxidation, improving liver function, promoting hematopoietic function of bone marrow cells, reducing blood pressure, reducing blood fat and the like, and has obvious effect in treating serious diseases such as leukemia, pulmonary fibrosis, renal failure and the like. The cordyceps militaris produced in an industrialized mode is golden in color, delicious in taste and rich in fungus fragrance. The existing cordyceps militaris mostly appears in the forms of sporocarp raw materials, capsules and other products, and has the defects of inconvenient carrying, slow absorption, difficult acceptance of mouthfeel and the like.
Dandelion, also known as herb of veronica, milkwort, etc., of the Compositae, Taraxacum, is sweet in nature, slightly bitter and cold. Has the functions of clearing away heat and toxic material, eliminating swelling, dissipating blood stasis, inducing diuresis and treating stranguria. The dandelion contains active ingredients such as the carotene, the triterpene, the sterol, the flavone, the chlorogenic acid, the volatile oil, the unsaturated fatty acid, the polysaccharide, the protein and the like, and is clinically used for treating surgical acute and chronic infections such as upper respiratory tract infection, gastritis, appendicitis, sore, furuncle, carbuncle, swelling and the like; meanwhile, the dandelion also has the effects of resisting oxidation, reducing blood pressure, reducing blood sugar, enhancing immunity, delaying senescence and the like. At present, most of dandelion in the market is obtained by artificial cultivation, and the whole cleaned and dried dandelion is used as a tea substitute, so that a lot of inconvenience is caused in eating.
The cordyceps militaris contains a large amount of nucleoside components, has unique fresh and fragrant mushrooms, and is abandoned by many people due to dislike of the flavor. The dandelion contains a large amount of alkaloid, organic acid, flavone and other ingredients, tastes cool and bitter, and is not suitable for being eaten singly in large amount.

The purpose of the invention is as follows: aiming at the problems in the prior art, the invention provides a health beverage consisting of cordyceps militaris and dandelion, which is rich in nutrition, fresh and cool in taste and suitable for warmth and coldness, can effectively remove free radicals, prevent human body aging and prevent various diseases, and better meets the continuously upgraded consumption requirements of modern consumers.
The invention also provides a preparation method of the health-care beverage consisting of the cordyceps militaris and the dandelion.
The technical scheme is as follows: in order to achieve the purpose, the health-care beverage composed of the cordyceps militaris and the dandelion is mainly prepared by taking cordyceps militaris and dandelion extract as main raw materials and adding sweetening agents, color fixative and water as auxiliary materials; the health beverage per 100mL comprises Cordyceps militaris extract 1-10mL, herba Taraxaci extract 1-5mL, sweetener 1-5g, color fixative 0.01-0.5g, and drinking water in balance.

The preparation method of the health-care beverage consisting of the cordyceps militaris and the dandelion comprises the following steps:
(1) selecting fresh cordyceps militaris sporocarp and dandelion respectively, cleaning, and preparing cordyceps militaris water extract and dandelion water extract respectively for later use;
(2) mixing Cordyceps militaris extract and herba Taraxaci extract, adding sweetener and color fixative, drinking water, and sterilizing to obtain health beverage composed of Cordyceps militaris and herba Taraxaci.

The final beverages prepared according to the invention were tasted, scored and analyzed by orthogonal design software (using L) by 5-20 panelists by setting criteria including sensory criteria such as color and taste, detailed tabulation in Table 116(45) Orthogonal experimental design, which is analyzed by orthogonal design software, and the detailed data are shown in table 2) to obtain an optimal formula;
preparing the beverage according to the optimized formula, sterilizing and packaging to obtain the cordyceps militaris dandelion beverage; measuring the content of effective components in the Cordyceps militaris and Taraxacum mongolicum beverage by high performance liquid chromatography; and measuring the antioxidant effect of the cordyceps militaris dandelion beverage by adopting a spectrophotometer method.
The invention prepares a health beverage consisting of cordyceps militaris and dandelion, wherein the cordyceps militaris enters lung and kidney two meridians, has sweet and mild nature and taste, can tonify kidney yang, benefit lung yin, and has better curative effects on patients with lung and kidney deficiency, soreness and weakness of waist and knees, impotence and premature ejaculation, chronic cough and asthma. Cordyceps militaris has effects of regulating organism immunity, resisting oxidation, and relieving fatigue. The dandelion has sweet, bitter and cold properties when entering liver and stomach channels, has the efficacies of oxidation resistance, blood fat reduction, wide antiviral spectrum and the like, has good treatment effect on respiratory system inflammation, but is not suitable for people with deficiency-cold spleen and stomach due to the cold and cold characteristics. The cordyceps militaris and the dandelion are used in a compatible way, one is warm and the other is cold, so that yin cannot be damaged due to warm dryness, yang qi cannot be damaged due to bitter cold, and the audience of consumption is obviously increased.

The invention utilizes the traditional Chinese medicine characteristics of the cordyceps militaris and the dandelion to fully exert the anti-inflammatory and antioxidant effects of the cordyceps militaris and the dandelion, does not cause side effects after long-term administration, namely cold and cool caused by the fact that the dandelion is eaten alone after long-term administration, and has the advantages of easily available raw materials and obvious effect.

The preparation method of the cordyceps militaris and dandelion beverage provided by the invention has the advantages of high standardization degree, easiness in industrial production, stable product quality and accordance with modern food supervision requirements.
The cordyceps militaris and dandelion beverage provided by the invention takes cordyceps militaris sporocarp and dandelion as main raw materials, is matched with the sweetening agent and the color fixative, has fresh taste, is rich in various active ingredients beneficial to a human body, can effectively remove free radicals in the human body, prevents aging and prevents various diseases, and is suitable for sub-health people to eat. The invention adopts a spectrophotometer method to measure the clearance rate of the cordyceps militaris dandelion beverage on DPPH free radicals and calculates the oxidation resistance. Test results show that the beverage prepared by the invention has 94.86% of DPPH free radical clearance rate and obvious antioxidant effect.

Detailed Description
The present invention is further illustrated by the following examples.
Example 1
1. Preparation of cordyceps militaris concentrated solution
Step 1: extraction of cordyceps militaris sporocarp
Selecting 10 g of dried cordyceps militaris sporocarp, adding 100mL of drinking water, extracting at 85 ℃ for 1h, filtering, repeating the extraction process for 2 times on filter residues, combining the three filtrates, and removing the filter residues;
step 2: filtration of cordyceps militaris extracting solution
Centrifuging the filtrate for 10min at 800rpm by using a decanter centrifuge to remove solid impurities to obtain clear Cordyceps militaris extractive solution;
and step 3: concentrating
Taking a cordyceps militaris extracting solution, and concentrating the extracting solution to 100mL by adopting a rotary evaporation method under the conditions of 0.1MPa and 80 ℃;
2. preparation of dandelion concentrated solution
Weighing 10 g of dried dandelion according to the method provided in the step 1, extracting and concentrating to obtain 100mL of dandelion concentrated solution;
3. blending
Taking 6mL of cordyceps militaris extract, 1mL of dandelion extract, 3g of sucrose, 0.2g of xylitol, 0.07g of arabinose content and 0.1g of vitamin C; supplementing 100mL of drinking water, and sterilizing at 121 ℃ for 10min by adopting a high-temperature heating device; packaging to obtain the health beverage consisting of Cordyceps militaris and herba Taraxaci. The whole preparation process flow of the cordyceps militaris dandelion beverage is shown in figure 1.
Example 2
Example 2 was prepared identically to example 1, except that: taking 1mL of cordyceps militaris extract, 1mL of dandelion extract, 1g of sweetener sucrose and 0.01g of vitamin C; supplementing drinking water to 100mL, sterilizing, and making into health beverage comprising Cordyceps militaris and herba Taraxaci.
Example 3
Example 2 was prepared identically to example 1, except that: taking 5mL of cordyceps militaris extract, 5mL of dandelion extract, 5g of sweetener xylitol and 0.25g of citric acid; supplementing drinking water to 100mL, sterilizing, and making into health beverage comprising Cordyceps militaris and herba Taraxaci.
Example 4
3. Blending
Example 2 was prepared identically to example 1, except that: taking 10mL of cordyceps militaris extract, 3mL of dandelion extract, 4g of sweetener sucrose, 1g of molasses and 0.5g of citric acid; supplementing drinking water to 100mL, sterilizing, and making into health beverage comprising Cordyceps militaris and herba Taraxaci.
Test example 1
According to the preparation method of example 1, the cordyceps sinensis concentrated solution prepared in the steps 1 and 2 with different volumes is added, the sweetener is added, and the volume is supplemented to 100mL by drinking water. By using L16(45) Orthogonal test design, which is comprehensively judged by 10 testers through sensory indexes such as color, aroma, taste and the like, and the specific standard is detailed in table 1. Orthogonal design software is used for analysis, and specific data are detailed in table 2 to obtain the optimal formula.

As can be seen from Table 2, the total evaluation of the prepared beverage was the highest when 100mL of the beverage contained 1mL of the dandelion extract, 6mL of the cordyceps extract, 3g of sucrose, 0.2g of xylitol, and 0.07g of molasses. The influence of each component on the comprehensive evaluation of the beverage is as follows: dandelion > cordyceps > sucrose > xylitol > molasses.
4. Measurement of amount of color fixative added
The effect of the color fixative in different amounts was measured according to the preparation method of example 1, vitamin C was added in different weights so that the content of vitamin C in the beverage was 0.01g/100mL, 0.05g/100mL, 0.1g/100mL, 0.15g/100mL, 0.5g/100mL, respectively, and the beverage was exposed to light at room temperature after sealing, and the change in the beverage was observed after 5 days, and the results are shown in Table 3.

As can be seen from Table 3, when the vitamin addition amount is more than 0.1%, the cordyceps sinensis and dandelion drink has a good color protection effect. In combination with the DPPH radical scavenging rate, vitamin C is selected to be added in an amount of 0.1g/100 mL.

Detection of effective ingredients in the health beverage consisting of cordyceps militaris and dandelion prepared in example 1
1. Establishment of a Standard Curve
The content of effective components in the beverage is detected by high performance liquid chromatography with cordycepin, adenosine and chlorogenic acid as indexes.
1.1 cordycepin and adenosine standard curves:
1.1.1 preparation of cordycepin Standard solution
Weighing cordycepin standard samples with different weights, and preparing into 3 μ g/ml, 6 μ g/ml, or,
9 mug/ml, 12 mug/ml and 18 mug/ml series of standard solutions are put into a refrigerator at 4 ℃ for standby.
1.1.2 adenosine Standard solution preparation
Adenosine series standard solutions were prepared as described in 1.1.1 and placed in a refrigerator at 4 ℃ for future use.
1.1.3 chromatographic conditions
The chromatographic column is CNW C18(4.6mm × 250mm, 5 μm), methanol as mobile phase, pure water (15: 85), flow rate of 1.0ml/min, detection wavelength of 260nm, column temperature of 30 deg.C, and sample introduction of 10 μ L after filtration with 0.25 μm filter membrane.
1.1.4 preparation of cordycepin and adenosine standard curve
And (3) determining chromatographic peaks corresponding to cordycepin and adenosine with different concentrations according to the chromatographic conditions of 1.1.3. And respectively taking the concentrations of cordycepin and adenosine as horizontal coordinates and the chromatographic peak area as vertical coordinates, and calculating to obtain a standard curve of cordycepin: y 0.3474x +0.0718, r 0.9998(n 5); adenosine standard curve: y is 0.3321x-0.1145 and r is 0.9999(n is 5). The high performance liquid chromatogram of cordycepin and adenosine in the cordyceps militaris dandelion beverage prepared in the embodiment 1 of the invention is shown in fig. 2.
1.2 chlorogenic acid standard curve:
1.2.1 chlorogenic acid Standard solution preparation
Weighing different weight chlorogenic acid standard samples, using methanol as solvent, preparing into 3 μ g/ml, 6 μ g/ml, 9 μ g/ml, 12 μ g/ml, 18 μ g/ml series standard solutions, and placing into 4 deg.C refrigerator for use.
1.2.2 chromatographic conditions: the chromatographic column is CNW C18(4.6mm × 250mm, 5 μm), acetonitrile as a mobile phase, 0.1% phosphoric acid (11: 89), a flow rate of 1.0ml/min-1, a detection wavelength of 328nm, a column temperature of 30 ℃, and a sample introduction amount of 10 μ L after filtration through a 0.25 μm filter.
1.2.3 chlorogenic acid Standard Curve preparation
Determining chromatographic peaks corresponding to chlorogenic acids with different concentrations according to chromatographic conditions of 1.2.2, and calculating to obtain a standard chlorogenic acid curve by taking the concentration of the chlorogenic acids as an abscissa and the area of the chromatographic peak as an ordinate: y is 0.2565x-0.303 and r is 0.9933(n is 5). The high performance liquid chromatogram of chlorogenic acid in the cordyceps militaris dandelion beverage prepared in example 1 of the invention is shown in fig. 3.
2. Determination of content of effective components in cordyceps sinensis and dandelion beverage
And (3) determining chromatographic peaks of cordycepin, adenosine and chlorogenic acid of the prepared beverage respectively according to chromatographic conditions shown in 1.1.3 and 1.2.3, and respectively calculating the cordycepin content of the beverage to be 5.13mg/100mL, the adenosine content to be 3.73mg/100mL and the chlorogenic acid content to be 21.46mg/100mL through a standard curve.
3. Methodology validation
3.1 precision test
According to the method shown in 1.1.1, preparing a cordycepin standard solution with the concentration of 12 mu g/ml, continuously measuring the peak area of 6 times of chromatographic analysis according to the chromatographic condition of 1.1.3, and calculating the peak area RSD value to be 1.12%. The RSD value of the adenosine peak area is 1.58% and the RSD value of the chlorogenic acid peak area is 0.66% which are measured by the same method, which shows that the liquid chromatograph used by the invention is stable and reliable and can be used for measuring the content of cordycepin, adenosine and chlorogenic acid in the beverage.
3.2 reproducibility test
According to the method shown in 1.1.1, 6 parts of cordycepin standard solution with the concentration of 12 mu g/ml are respectively prepared, the chromatographic peak areas of the 6 parts of solution are continuously measured according to the chromatographic condition of 1.1.3, the RSD value of the peak area is calculated to be 1.49%, the RSD value of the peak area of adenosine is measured to be 1.68% by the same method, and the RSD value of the peak area of chlorogenic acid is 1.04%, so that the method used by the invention is accurate and reliable, and can be used for measuring the contents of cordycepin, adenosine and chlorogenic acid in the beverage.
3.3 stability test
Preparing a cordycepin standard solution with the concentration of 12 mu g/ml according to a method shown in 1.1.1, measuring the chromatographic peak area at room temperature every 2 hours according to a chromatographic condition of 1.1.3, measuring for 6 times, calculating the peak area RSD value of cordycepin to be 1.25%, measuring the peak area RSD value of adenosine to be 0.50% and the peak area RSD value of chlorogenic acid to be 0.34% by the same method, and showing that cordycepin, adenosine and chlorogenic acid are stable at room temperature and the detection result is stable and reliable.
3.4 recovery rate by adding standard
Preparing 6 parts of cordycepin standard solution with the concentration of 18 mu g/mL 40mL according to the method shown in 1.1.1, respectively adding the cordycepin standard solution into 6 parts of 10mL dandelion cordyceps sinensis beverage, uniformly mixing, measuring the chromatographic peak area of cordycepin according to the chromatographic condition of 1.1.3, calculating the cordycepin content in the mixed sample according to a cordycepin standard curve, and according to the formula: the recovery rate is (standard sample measurement value-sample measurement value) ÷ standard amount is multiplied by 100%, and the standard recovery rate of the cordycepin is calculated to be 91.28%; the standard recovery rate of adenosine is calculated to be 92.33% and the standard recovery rate of chlorogenic acid is calculated to be 107.72% by the same measuring method, which shows that the method provided by the invention is accurate and reliable and can be used for detecting effective components in beverages.
The above results show that: the precision, reproducibility and recovery rate of the method provided by the invention meet the requirements of the method, and the solution is placed at room temperature for 10h, and the result after measurement is stable. Therefore, the method can be applied to the content determination of adenosine, cordycepin and chlorogenic acid in the health-care beverage, and provides a basis for market supervision.

Determination of antioxidant function of the material in the health beverage comprising cordyceps militaris and dandelion prepared in example 1
The invention adopts DPPH method to measure the free radical scavenging activity of the cordyceps sinensis and dandelion drink. The maximum absorption wavelength of the DPPH ethanol solution is 517 nm. When a free radical scavenger is added into a DPPH standard solution, lone pair electrons are paired, the absorbance at 517nm is reduced, and the change degree and the free radical scavenging degree are in a linear relation, so the method can be expressed by the scavenging rate, and the higher the scavenging rate is, the stronger the scavenging capacity of the substance is.
1. Preparation of DPPH Standard solution
Adding a 100mL brown volumetric flask into a 4mgDPPH standard sample, dissolving with absolute ethyl alcohol, fixing the volume to a scale to obtain a DPPH standard solution with the concentration of 0.004%, and storing in a refrigerator at 4 ℃ in a dark place for later use.
2. Determination of the ability of a sample to scavenge DPPH free radicals
Respectively taking 1.0mL of commercially available brand herbal tea beverage and 1.0mL of cordyceps sinensis and dandelion beverage, putting the beverages in a 10mL test tube, adding 3.0mL of LDPPH standard solution, uniformly mixing, reacting at room temperature in a dark place for 30min, taking absolute ethyl alcohol as a blank, measuring the light absorption value at a wavelength of 517nm, and calculating the DPPH free radical scavenging rate:

As can be seen from Table 4, the DPPH free radical clearance rate of the commercially available brand herbal tea beverage is 90.18%, and when the addition amount of the vitamin C is 0.1g/100mL, the DPPH free radical clearance rate of the cordyceps dandelion beverage is 94.86%; when the addition amount of the vitamin C is 0.15g/100mL, the DPPH free radical clearance rate of the cordyceps sinensis and dandelion beverage is 95.25%, the color protection effect is comprehensively considered, and the addition amount of the vitamin C is selected to be 0.1g/100mL finally. The beverage prepared under the condition has obvious antioxidant effect.

The results in table 4 show that the cordyceps militaris and dandelion beverage provided by the invention takes cordyceps militaris sporocarp and dandelion as main raw materials, is matched with a sweetening agent and a color fixative, has fresh taste, is rich in various active ingredients beneficial to a human body, can effectively remove free radicals in the human body, prevents aging and prevents various diseases, and is suitable for sub-health people to eat.
Claims (5)
1. A health beverage comprising Cordyceps militaris and herba Taraxaci, is characterized in that the health beverage is mainly prepared from Cordyceps militaris and herba Taraxaci extract as main raw materials, and sweetener, color fixative and drinking water as adjuvants; the health beverage per 100mL comprises Cordyceps militaris extract 1-10mL, herba Taraxaci extract 1-5mL, sweetener 1-5g, color fixative 0.01-0.5g, and drinking water in balance.
2. The health beverage comprising cordyceps militaris and dandelion according to claim 1, wherein the sweetener is one or more of sucrose, xylitol and acesulfame.
3. The health beverage comprising Cordyceps militaris and herba Taraxaci according to claim 1, wherein the color fixative is preferably one or more of vitamin C and citric acid.
4. A preparation method of the health beverage consisting of cordyceps militaris and dandelion according to claim 1, which is characterized by comprising the following steps:
(1) selecting fresh cordyceps militaris sporocarp and dandelion respectively, cleaning, and preparing cordyceps militaris water extract and dandelion water extract respectively for later use;
(2) mixing Cordyceps militaris extract and herba Taraxaci extract, adding sweetener and color fixative, drinking water, and sterilizing to obtain health beverage composed of Cordyceps militaris and herba Taraxaci.
