Abstract
本发明涉及一种抗病毒性感冒中药制剂在抑制肺炎中的应用,所述抗病毒性感冒中药制剂包括黄芩、金银花、连翘、大青叶、淡竹叶、甘草浸膏、糊精、牛蒡子、薄荷油、人工牛黄和辅料。本发明的抗病毒性感冒中药制剂具有抑制流感病毒引起的肺炎的作用;具有抑制体温增高的作用;具有抑制炎症的作用;具有增强免疫功能的作用,对于肺炎的抑制,以及公共健康事业具有重大价值。

技术领域
本发明涉及抗病毒性感冒中药制剂在抑制肺炎中的应用。
背景技术
肺炎指肺泡、远端气道和肺间质的感染性炎症,可由细菌、病毒和其他病原体等因素感染引起,其中以细菌性和病毒性肺炎最为常见。现有技术中抗病毒性感冒中药制剂往往见效慢,且作用单一,而西药常常具有嗜睡作用。
因此,本发明针对上述不足,提供一种抗病毒性感冒中药制剂在解热、消炎、增强免疫,甚至抑制肺炎等多方面中的应用。
发明内容
本发明的目的是针对现有技术中的不足,提供一种抗病毒性感冒中药制剂在抑制肺炎中的应用。
为实现上述目的,本发明采取的技术方案是:
提供一种抗病毒性感冒中药制剂在抑制肺炎中的应用,所述抗病毒性感冒中药制剂包括黄芩、金银花、连翘、大青叶、淡竹叶、甘草浸膏、糊精、牛蒡子、薄荷油、人工牛黄和辅料。
优选地,所述抗病毒性感冒中药制剂在抑制流感病毒引起的肺炎中的应用。
优选地,所述抗病毒性感冒中药制剂在抑制肺炎引起的体温增高中的应用。
优选地,所述抗病毒性感冒中药制剂在抑制肺炎炎症中的应用。
优选地,所述抗病毒性感冒中药制剂在增强免疫功能中的应用。
本发明采用以上技术方案,与现有技术相比,具有如下技术效果:
本发明的抗病毒性感冒中药制剂具有抑制流感病毒引起的肺炎的作用;具有抑制体温增高的作用;具有抑制炎症的作用;具有增强免疫功能的作用,对于肺炎的抑制,以及公共健康事业具有重大价值。
具体实施方式
下面将,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动的前提下所获得的所有其他实施例,都属于本发明保护的范围。
需要说明的是,在不冲突的情况下,本发明中的实施例及实施例中的特征可以相互组合。
下面结合具体实施例对本发明作进一步说明,但不作为本发明的限定。
实施例1
观察所述抗病毒性感冒中药制剂对流感病毒引起肺炎的抑制作用:
1对小鼠流感病毒性肺炎的治疗作用
1.1小鼠病毒感染量LD50的确定:将10-2-10-8浓度的病毒液滴鼻感染小鼠,每个浓度组10只,0.05mL/只。观察动物15天内的死亡情况,确定病毒感染小鼠的LD50的量。经试验观察病毒感染小鼠的LD50为10-4。
1.2方法:取小鼠按体重随机分为6组。分别为所述抗病毒性感冒中药制剂三个剂量组、双黄连胶囊组、病毒感染对照组与正常动物对照组。除正常对照组外,将小鼠用乙醚轻度麻醉,以15个LD50流感病毒液滴鼻感染,每只0.05mL。感染前一天开始灌胃给药,每天1次,每次0.2mL/10g体重,连续5天,对照组在同等条件下蒸馏水灌胃。第6天称取小鼠体重后解剖,摘取全肺称重,计算肺指数值,并求出肺指数抑制率。
肺指数=[肺重(g)/体重(g)]×100
肺指数抑制率=(病毒对照组肺指数均值-试验组肺指数均值)÷病毒对照组肺指数均值
结果采用组间t检验进行统计学处理。
1.3结果:所述抗病毒性感冒中药制剂三个剂量组的肺指数值及肺部病变明显低于病毒对照组,高剂量、中剂量与病毒对照组比较有显著性差异。表明所述抗病毒性感冒中药制剂在所试剂量内对流感病毒引起的小鼠肺炎有明显的抑制作用(见表1)。

注:与正常对照组比较,##P<0.01,与病毒对照组比较,*P<0.05,**P<0.01。
2对流感病毒所致小鼠死亡的保护作用
2.1小鼠病毒感染量的确定:将不同浓度的病毒液滴鼻感染小鼠,每个浓度组10只,0.05mL/只滴鼻感染。观察动物15天内的死亡情况,取感染后15天内动物死亡率在90%以上的浓度为正式试验的感染量
2.2方法:取小鼠100只,按体重随机分为5组。分别为所述抗病毒性感冒中药制剂三个剂量组、双黄连胶囊组及病毒感染对照组。各给药组灌胃给药,每天1次,每次0.2mL/10g,连续7天,病毒对照组在同等条件下给予蒸馏水。给药第二天各组小鼠用乙醚轻度麻醉后,以2个LD50流感病毒液滴鼻,每只 0.05mL。感染后连续观察14天,每天记录小鼠的死亡数,计算死亡率、死亡保护率及生命延长率。
死亡率%=死亡动物数÷动物总数
死亡保护率=(对照组死亡率—实验组死亡率)÷对照组死亡率
生命延长率=(实验组平均存活天数-对照组平均存活天数)÷对照组平均存活天数
2.3结果:小鼠感染病毒14天内,所述抗病毒性感冒中药制剂三个剂量组动物的死亡数明显低于病毒对照组,存活天数明显延长,大、中剂量组与病毒对照组比较有显著差异,表示所述抗病毒性感冒中药制剂在此剂量时对流感病毒感染致小鼠死亡有明显的保护作用。
实施例2
观察所述抗病毒性感冒中药制剂对肺炎引起的体温增高的抑制作用:
1对酵母致大鼠发热的解热作用
1.1方法:取大鼠50只,体重200-220克。实验当日晨先测动物基础体温,然后以15%酵母液按2mL/100g背部皮下注射,4h后测体温,选肛温升高0.8℃以上者用于实验。随机分组,每组10只,分别灌胃给药或水10mL/kg体重。药后每0.5h测肛温一次,共测3次,以不同时间所测肛温与基础肛温差值为体温变化的指标,结果采用差值t检验进行组间比较。
1.2结果:所述抗病毒性感冒中药制剂三个剂量组在给药0.5小时开始,体温升高值明显低于模型对照组,其中高、中剂量组可持续到给药后2小时与模型组比较有显著性差异(P<0.01,P<0.05)。
2对内毒素致家兔发热的退热作用
2.1方法:取家兔45只,实验前测基础体温,取体温在38.0-39.5℃之间的动物用于实验,并将动物按体温均值分成5组,每组8只。灌胃给药0.5h后各动物耳缘静脉注射内毒素液1.0mL/kg进行致热,分别测致热后30min、1h、1.5h、 2.0h的肛温,以致热后不同时间肛温与基础体温之差值为体温变化的指标。结果采用差值t检验组间比较,进行统计学处理。
2.2结果:所述抗病毒性感冒中药制剂三个剂量组对大肠杆菌性内毒素引起的家兔体温增高有明显的抑制作用,与对照组比较有显著性差异(*P<0.05, **P<0.01),其中高剂量组作用可持续到给药后2小时,中、低剂量组可持续到给药后1小时。
实施例3
观察所述抗病毒性感冒中药制剂对急性渗出性炎症及慢性增殖性炎症的抑制作用:
1对醋酸引起小鼠腹腔毛细血管通透性的影响
1.1方法:取健康小鼠,体重18~20g,雌雄各半,随机分为5组。
①空白对照组:蒸馏水10mL/kg/d;②双黄连组:0.88g/kg/d;③所述抗病毒性感冒中药制剂高剂量组:20g生药/kg/d;④所述抗病毒性感冒中药制剂中剂量组:10g生药/kg/d;⑤所述抗病毒性感冒中药制剂低剂量组:5g生药/kg/d。
灌胃给药,每天1次,连续7天,末次给药30min后,尾静脉注射0.5%伊文思蓝生理盐水(0.1mL/10g)后,立即腹腔注射0.6%醋酸生理盐水0.2mL/只,20min后将小鼠颈椎脱臼处死,剪开腹腔皮肤肌肉,用6mL生理盐水数次洗涤腹腔,吸管吸出洗涤液,合并后加入生理盐水至10mL,3000rpm离心15min,取上清液于分光光度计590nm波长处比色测光密度(A值)。结果进行组间比较 t检验。
1.2结果:所述抗病毒性感冒中药制剂高、中、低剂量和双黄连对醋酸引起小鼠腹腔毛细血管通透性增加有明显抑制作用,与空白对照组比较有明显统计学差异(见表2)。

2对大鼠棉球肉芽肿的影响
2.1方法:取健康wistar大鼠,体重140~160g,雄性,随机分为6组。
①空白对照组:蒸馏水10mL/kg/d;②氢化可的松组:30mg(6mL)/kg/d;③双黄连组:0.432g/kg/d;④所述抗病毒性感冒中药制剂高剂量组:10g生药/kg/d;⑤所述抗病毒性感冒中药制剂中剂量组:5g生药/kg/d;⑥所述抗病毒性感冒中药制剂低剂量组:2.5g生药/kg/d。
各组动物盐酸氯胺酮麻(0.1g/kg),左右蹊部剪毛,碘酒消毒,75%酒精脱碘,各切一小口,以血管钳扩充皮下组织,每侧埋入一灭菌棉球(20mg,含青霉素160μ、链霉素0.1mg),缝合皮肤。从手术当日开始,除氢化可的松组皮下注射给药外,其余各组动物均灌胃给药,每天1次,连续7天。第8天颈椎脱臼处死动物,取出棉球,剔除脂肪组织,置60℃烤箱中烘干,称重,减去原棉球重量即为肉芽肿重量,计算肉芽肿指数(mg/100g体重),结果进行组间比较t 检验。
2.2结果:所述抗病毒性感冒中药制剂高、中剂量和氢化可的松对大鼠棉球肉芽肿增生具有明显的抑制作用,与空白对照组比较有显著性差异(P<0.05, P<0.01)。双黄连对大鼠棉球肉芽肿增生有抑制的趋势,但无统计学差异(P>0.05) (见表3)。

实施例4
观察所述抗病毒性感冒中药制剂对免疫功能的增强作用:
1所述抗病毒性感冒中药制剂对小鼠网状内皮系统吞噬功能的影响(炭粒廓清法)
1.1方法:取健康昆明种小鼠,雄性,随机分为5组。①空白对照组:蒸馏水10mL/kg/d;②双黄连组:0.88g/kg/d;③所述抗病毒性感冒中药制剂高剂量组: 20g生药/kg/d;④所述抗病毒性感冒中药制剂中剂量组:10g生药/kg/d;⑤所述抗病毒性感冒中药制剂低剂量组:5g生药/kg/d。各组动物灌胃给药7天,每天1 次,末次给药60分钟后尾静脉注射1:10碳汁10mL/kg,分别于注射后0.5分钟、 5分钟经眼眶取血(20μL/只),放入0.1%Na2CO3溶液(2mL)中混匀,在722 分光光度计675nm处比色测A值,按下列公式计算吞噬指数(K)和吞噬系数 (a),结果进行组间比较t检验。
K=(lgC1-lgC2)÷(T2-T1)
a=W÷WLS×K1/3
C为血碳粒浓度,T为时间,W为体重,WLS为肝脾合重。
1.2结果:所述抗病毒性感冒中药制剂可显著增强小鼠网状内皮系统吞噬功能,其高剂量组吞噬指数明显增加,与空白对照组比较有显著性差异(P<0.05)。
2所述抗病毒性感冒中药制剂对鸡红细胞所致小鼠溶血素抗体生成的影响。
2.1方法:取健康昆明种小鼠,雌雄各半,动物分组及给药同1.1。各组动物于给药第4天腹腔注射2%的鸡红细胞悬液0.2mL/只,给药第8天经眼眶取血 20μL/只。加入含1mL生理盐水的试管中摇匀,再加5%鸡红细胞悬液和补体各 0.5mL,37℃水浴30分钟,冰箱中终止反应,2000rpm离心10分钟,取上清液 1mL加入3mL的都氏试剂中,10分钟后在722型分光光度计540nm波长处测光密度(A值)。结果进行组间比较t检验。
2.2结果:所述抗病毒性感冒中药制剂高剂量可显著提高小鼠血清溶血素水平,与空白对照组比较有显著性差异(P<0.05),中、低剂量和双黄连亦有提高血清溶血素水平的趋势,但无统计学意义。
3所述抗病毒性感冒中药制剂对小鼠迟发性变态反应的影响
3.1方法:动物分组及给药2.1。各组动物于给药当日,在颈部脱毛皮肤上滴50%DNCB丙酮溶液2μL/只,给药第9天在腹部脱毛皮肤上滴0.25%DNCB 丙酮溶液20μL/只,24h后尾静脉注射1%伊文思蓝10mg(10mL)/kg,30分钟后将小鼠拉颈处死,取腹部蓝染皮肤剪碎,置4mL丙酮生理盐水(1:1)混合液中浸泡24小时,2000rpm离心10分,取上清液在722型分光光度计610nm波长处测光密度(A值)。结果进行组间比较t检验。
3.2结果:所述抗病毒性感冒中药制剂高、中剂量和双黄连均可明显抑制 DNCB诱导的小鼠迟发性变态反应,与空白对照组比较有非常显著性差异 (P<0.05,P<0.01)。
本发明的抗病毒性感冒中药制剂具有抑制流感病毒引起的肺炎的作用;具有抑制体温增高的作用;具有抑制炎症的作用;具有增强免疫功能的作用,对于肺炎的抑制,以及公共健康事业具有重大价值。
以上所述仅为本发明较佳的实施例,并非因此限制本发明的实施方式及保护范围,对于本领域技术人员而言,应当能够意识到凡运用本发明说明书所作出的等同替换和显而易见的变化所得到的方案,均应当包含在本发明的保护范围内。

1.一种抗病毒性感冒中药制剂在抑制肺炎中的应用,所述抗病毒性感冒中药制剂包括黄芩、金银花、连翘、大青叶、淡竹叶、甘草浸膏、糊精、牛蒡子、薄荷油、人工牛黄和辅料。
2.根据权利要求1所述抗病毒性感冒中药制剂在抑制肺炎中的应用,其特征在于,所述抗病毒性感冒中药制剂在抑制流感病毒引起的肺炎中的应用。
3.根据权利要求1所述抗病毒性感冒中药制剂在抑制肺炎中的应用,其特征在于,所述抗病毒性感冒中药制剂在抑制肺炎引起的体温增高中的应用。
4.根据权利要求1所述抗病毒性感冒中药制剂在抑制肺炎中的应用,其特征在于,所述抗病毒性感冒中药制剂在抑制肺炎炎症中的应用。
