本发明要解决的技术问题是克服现有技术中缺乏植物介导合成纳米银化合物光催化降解染料废水的应用缺陷和不足,提供一种辣木籽提取物介导合成纳米银材料在染料废水处理中的应用。
本发明上述目的通过以下技术方案实现:
一种辣木籽提取物介导合成纳米银材料在染料废水处理中的应用。
辣木籽提取物介导合成纳米银材料应用于有机染料废水处理的主要作用机理如下:
有机染料的降解过程发生在AgNPs的表面上,AgNPs充当催化剂。通过TEM测定AgNPs材料的粒度在4.1-33.5nm范围内,并且XRD分析表明,生物合成的AgNP具有球状结构,较小尺寸的AgNP具有较大的表面积,可以提供更多可用的活性位点,在该位点,有机染料通过电子转移发生降解。在阳光照射下,电子被激发并从银的价带(VB)移至空导带(CB),导致形成电子空穴对,具有良好的氧化剂和还原剂,与反应混合物中的氧气发生反应时,激发的电子在催化剂(AgNPs)活性表面生成自由基,从而导致有机染料的降解。辣木籽提取物在这个过程的作用除了通过帽帮助形成稳定的纳米颗粒外,还参与电子转移,促进催化降解有机染料。
优选地,所述染料废水为含有亚甲基蓝和/或刚果红和/或4-硝基苯酚的染料废水。
本发明的辣木籽提取物介导合成纳米银材料用于染料废水处理可以催化还原剂对亚甲基蓝和/或刚果红和/或4-硝基苯酚的还原,优选地,染料废水处理的具体方法如下:
S1.将NaBH4溶液于染料废水混合;
S2.将辣木籽提取物介导合成纳米银材料加入上述混合溶液中,光催化反应,
其中S1中NaBH4溶液的浓度为0.01~0.18mol/L,S2中辣木籽提取物介导合成纳米银材料的浓度为20~6000mg/L。
其中,在反应体系中NaBH4溶液的浓度范围为0.01~0.18mol/L,NaBH4溶液浓度的影响为:NaBH4是染料降解的催化剂,浓度过低则达不到催化反应的效果,无法实现染料的有效降解。另一方面,浓度高到一定程度则达到饱和,再增加对提高降解效率无效。
辣木籽提取物介导合成纳米银材料的浓度的影响为:纳米银材料是染料废水处理的活性物质,废水中的染料降解效率与其浓度密切相关,浓度过低无法有效促成降解,降解效率与浓度一定程度下成正比。
优选地,S1中NaBH4溶液的浓度为0.02~0.10mol/L。
更优选地,S1中NaBH4溶液的浓度为0.3mol/L。
优选地,S2中辣木籽提取物介导合成纳米银材料的浓度为50~500mg/L。
更优选地,S2中辣木籽提取物介导合成纳米银材料的浓度为400mg/L。
优选地,S2中所述光催化反应的反应时间为15~70min。本发明的催化降解反应在有无太阳光的情况下均可进行,其中反应时间控制在15~75min,反应时间太短则降解不充分,反应时间太长则增加时间成本。
进一步优选地,S2中所述光催化反应的反应时间为75min。
优选地,本发明所述辣木籽提取物介导合成纳米银材料为圆球形核壳结构,其内部为聚集的银离子,外部为覆盖的辣木籽提取物分子。
优选地,所述辣木籽提取物介导合成纳米银材料的粒径范围为3.8~31.0nm,平均粒径为9.8nm。
进一步优选地,所述辣木籽提取物介导合成纳米银材料中粒径为11~15nm的纳米银颗粒占纳米银材料总颗粒数的30%,粒径为3.8~11nm(11nm以下)的纳米银颗粒占纳米银材料总颗粒数的58%。
优选地,所述辣木籽提取物介导合成纳米银化合物通过辣木籽提取物和硝酸银合成得到,其中所述辣木籽提取物和硝酸银的质量比为10:0.764。
本发明具体的辣木籽提取物介导合成纳米银材料的方法如下:
通过混合辣木籽提取物和硝酸银来合成AgNP,将反应混合物在60℃,300rpm的加热磁力搅拌器上放置8小时,每隔2小时通过分光光度计观察样品,以在350至450nm处获得标准峰,并观察到颜色变化,8小时后,将溶液以8000rpm离心20分钟,并弃去上清液。
将沉淀物悬浮在蒸馏水中,并以8000rpm的速度离心三次,每次20分钟,以完全去除未结合的化合物,之后,将获得的沉淀物在80℃的烘箱中干燥,得到AgNP。
其中,植物介导的AgNP合成涉及通过次级代谢产物(例如酚酸和类黄酮)将Ag+还原为Ag0。植物介导的AgNP合成过程中可分为三个阶段。Ag0小核的“成核”形成,小核的“生长相”分组,以及成簇的核被植物次生代谢产物氧化的“加帽”,从而稳定所形成的AgNPs。
优选地,本发明的辣木籽提取物的主要有效成分为没食子酸、黄酮和酚酸,通过如下方法提取得到:
辣木籽先被研磨成粉状,加水室温提取5个小时。然后以1:10的体积比例往提取物水溶液中加入石油醚进行萃取;
去除石油醚部分后,往余下水相部分以1:8的比例加入乙醇进行处理,然后滤纸过滤,滤液真空冷冻干燥后得到辣木籽提取物。
与现有技术相比,本发明的有益效果是:
本发明提供了一种辣木籽提取物介导合成纳米银材料在染料废水处理中的应用,有机染料的降解过程发生在AgNPs的表面上,AgNPs为球状结构,较小尺寸的AgNP具有较大的表面积,可以提供更多可用的活性位点,在该位点,在阳光和/或催化剂NABH4存在的情况下,有机染料通过电子转移发生降解,对亚甲基蓝MB最大降解率分别为纯水91.4%,废水为82.4%,对刚果红CR最大降解率分别为纯水86.2%,废水82.7%。

具体实施方式
下面结合具体实施方式对本发明作进一步的说明,但实施例并不对本发明做任何形式的限定。除非另有说明,本发明实施例采用的原料试剂为常规购买的原料试剂。
实施例1
一种辣木籽提取物介导合成纳米银材料在亚甲基蓝染料废水处理中的应用。
染料废水处理的具体方法如下:
S1.将NaBH4溶液与含亚甲基蓝的染料废水混合,其中亚甲基蓝的浓度为15μM;
S2.将辣木籽提取物介导合成纳米银材料加入上述混合溶液中,光催化反应,反应时间分别为15min、30min、45min、60min、75min,
其中S1中NaBH4溶液的浓度为0.03mol/L,S2中辣木籽提取物介导合成纳米银材料的浓度为400mg/L。
其中辣木籽提取物介导合成的纳米银材料通过辣木籽提取物和硝酸银合成得到,其中所述辣木籽提取物和硝酸银的质量比为100mg:7.64mg。
具体合成方法如下:
(1)通过混合10mL辣木籽提取物(10mg/mL)和90ml硝酸银(0.5mM)合成AgNP。
(2)将反应混合物在60℃,300rpm的加热磁力搅拌器上放置8小时。每隔2小时通过分光光度计观察样品,以在350至450nm处获得标准峰,并观察到颜色变化。
(3)8小时后,将溶液以8000rpm离心20分钟,并弃去上清液,将沉淀物悬浮在蒸馏水中,并以8000rpm的速度离心三次,每次20分钟,以完全去除未结合的化合物,将获得的沉淀物在80℃的烘箱中干燥,并保存在4℃用于进一步的实验。
其中,辣木籽提取物的主要有效成分为没食子酸、黄酮和酚酸,通过如下方法提取得到:
AgNPs的表征:
由于存在游离氨基或半胱氨酸残基,金属纳米颗粒(例如AgNPs)具有与植物蛋白结合的能力,辣木籽提取物中存在的还原剂,例如酚类化合物和多糖,颜色的变化与辣木籽提取物的温度和浓度直接相关。
将辣木籽提取物与硝酸银溶液混合后,通过观察反应混合物的颜色变化可以证明辣木籽提取物生物合成AgNPs的反应进程,所得溶液从黄色到红棕色的颜色变化表明形成了AgNP(图1)。由于生物合成的AgNPs的表面等离振子共振现象,观察到从黄色到黑色的增加的孵育时间(0至8h),颜色强度增加。之后,没有观察到进一步的颜色变化,表明生物合成的AgNPs性能稳定性,同时也表明负责减少银离子的各种植物分子的可用性。
(1)紫外-可见(UV-Vis)光谱
为了监测AgNPs的生物合成,在300至600nm的波长范围内对生物合成的AgNPs的胶体溶液进行了UV-Vis光谱分析,以确定最大吸光度。在每个间隔观察吸收和颜色变化,直到观察到深褐色。在每个间隔处显示的峰代表纳米颗粒的稳定性和合成。表面等离子体激元吸收的形状和位置完全基于粒子合成,包括粒子之间的间隙和环境介电常数。使用分光光度计研究反应动力学,并使用origin 9.1软件分析接收的数据。
本实施例的AgNPs生物合成每60分钟反应后进行一次UV-Vis吸收光谱分析,得出420nm处的吸光度,表明AgNP的形成(图2)。图1表明银纳米颗粒根据其尺寸显示从420到450的表面等离振子共振现象,表面等离子体共振现象是与光共振的电子在纳米粒子上的传导键中的激发。本实施例的AgNPs溶液的表面等离子体共振带保持接近421nm,这进一步证实了水相中的银纳米颗粒是单分散的。AgNPs的稳定性可能是由于辣木植物的加帽剂,例如多糖,蛋白质和酚类化合物。
以上结果证实了辣木籽提取物介导的AgNPs的合成与化学合成方法相比具有环保性和快速性,步骤单一简洁且不需要苛刻的条件。结论是,辣木籽的一级/二级代谢产物的变化是造成银纳米颗粒还原的原因。图8显示了辣木籽介导的AgNP生物合成的示意图。
(2)场发射扫描电子显微镜(FE-SEM)分析
利用SEM分析以可视化生物合成的AgNP的形态分析。将5μL生物合成的AgNPs涂在载玻片上,然后在烘箱中干燥15分钟。在分析之前,使用双面粘合碳将载玻片固定在铝制短管上,并使用铂的导电层覆盖样品。SEM图像在10Kv的加速电压下拍摄。
生物合成的AgNPs的FE-SEM图像显示出均匀的形状和球形表面形态(图3)使用image J软件计算出的生物合成AgNPs大小在12.4至33.7nm范围内,平均大小为15.7nm。辣木籽提取物介导的生物合成的AgNPs的化学分析通过使用EDX分析进行分析。EDX光谱显示存在Ag和覆盖Ag聚集体的其他元素,Ag的存在归因于AgNP的形成,而其他原子的存在归因于辣木籽提取物。此外,EDX分析表明,AgNPs为金属形式,不含其他杂质。
(3)透射电子显微镜(TEM)分析
为了进行TEM分析,将一滴AgNPs水溶液放在涂有碳的铜网上并干燥。通过200KV获得显微照片,并且通过在TEM显微照片上通过Image J软件测量至少50个颗粒来获得生物合成的AgNP的尺寸和直径。
透射电子显微镜(TEM)分析提供了对AgNPs的大小,形状和形态的深入分析。TEM图像(图4)显示了辣木籽提取物介导的生物合成AgNP以及小的聚集体。大多数生物合成的AgNPs呈球形,其大小(通过图像J测量)范围为5.05至24.2nm,与FE-SEM结果一致。此外,大多数AgNP的直径在11至15nm之间,而几乎60%的AgNP都在11nm以下,即使在聚集体内,AgNP也没有直接接触,这表明通过辣木籽提取物加帽分子使颗粒稳定。
(4)傅立叶变换红外(FT-IR)光谱
使用KBr沉淀与生物合成的AgNPs纯化粉末以4cm-1的分辨率和32的扫描频率混合,在4000-400cm-1的范围内进行FT-IR分析。
FTIR光谱法是确定辣木籽提取物介导的生物合成AgNP和辣木籽提取物中存在的各种官能团的相关技术,辣木籽提取物中的官能团负责生物合成的AgNP的形成和稳定。将辣木籽提取物介导的生物合成AgNPs的FTIR光谱与辣木籽水提取物进行了比较(图5),结果表明,在3387-3392.7cm-1(OH拉伸)对应于酚基团的波长,2927.9-2924cm-1(NH拉伸)对应于伯胺,1651-1637.5cm-1(-C=C-)拉伸对应于烯烃,1062-1058.9cm-1(CH拉伸)对应于碳水化合物,并在588.2cm-1(C-Cl拉伸)处达到峰值,其对应于AgNP和辣木籽提取物中均存在的卤素化合物。FTIR结果表明,辣木籽中存在的官能团也出现在生物合成的AgNPs中,并具有轻微的峰位移,这表明AgNP的形成。
因此,从以上讨论可以得出结论,多糖,酚,蛋白质和酰胺是还原,加帽和稳定生物合成的AgNP的主要负责基团。
(5)X射线衍射(XRD)分析
用X射线衍射(XRD)扫描分析了AgNPs的晶体性质,范围为20-60℃。简要地说,将AgNPs粉末研磨成细粉,然后安装到样品载体上。最后,使用XRD在40KV的电压下记录强度峰。
进行XRD分析以确认辣木籽介导的生物合成AgNP的晶体结构。XRD分析表明,在37.8°(2θ)处的峰表明纳米颗粒由纯银制成,生物合成的AgNPs本质上是晶体,此外,在42°(2θ),65°(2θ)和78°(2θ)处分别出现了与200、220和311个银平面相对应的峰。此外,XRD图案中以(*)标记的未分配峰可能是由AgNPs表面上发生的生物有机相的结晶所致(图6)。总体而言,X射线衍射图谱表明生物合成的AgNP具有立方结构。
(6)热重分析
通过在氮气氛压力下使用热重分析仪研究了生物合成的AgNPs的热稳定性。简而言之,将5mg AgNP以10℃/min的加热速率从环境温度加热至700℃,以获得单个光谱。
辣木籽提取物介导的AgNPs生物合成的热稳定性通过在氮气氛下从25-800℃加热的热重分析法进行评估(图7)。TGA结果表明存在三个主要的失重范围,由于吸收的水和挥发性残留物的流失,观察到的第一个失重(约5.2%),在200-420°之间发现第二次失重(大约11.4%),这是由于在生物合成的AgNPs表面发现的辣木籽植物化学化合物的热降解,在800℃或更高温度下,观察到相变也接近银的熔点。总体而言,首先失重归因于水,其余失重归因于辣木籽化合物的表面脱附。
检测结果显示:
纳米银材料的粒径范围为3.8~31.0nm,平均粒径为9.8nm,纳米银材料中粒径为11~15nm的纳米银颗粒占纳米银材料总颗粒数的30%,粒径为5~11nm(11nm以下)的纳米银颗粒占纳米银材料总颗粒数的58%。
实施例2
一种辣木籽提取物介导合成纳米银材料在刚果红的染料废水处理中的应用。
染料废水处理的具体方法如下:
S1.将NaBH4溶液与含刚果红的染料废水混合,其中刚果红的浓度为15μM;
S2.将辣木籽提取物介导合成纳米银材料加入上述混合溶液中,光催化反应,反应时间分别为15min、30min、45min、60min、75min,
其中S1中NaBH4溶液的浓度为0.03mol/L,S2中辣木籽提取物介导合成纳米银材料的浓度为400mg/L。
其中辣木籽提取物介导合成的纳米银材料通过辣木籽提取物和硝酸银合成得到,其中所述辣木籽提取物和硝酸银的质量比为100mg:7.64mg,具体合成方法与实施例1相同。
实施例3
一种辣木籽提取物介导合成纳米银材料在4-硝基苯酚的染料废水处理中的应用。
染料废水处理的具体方法如下:
S1.将NaBH4溶液与含4-硝基苯酚的染料废水混合,其中4-硝基苯酚的浓度为153.8μM;
S2.将辣木籽提取物介导合成纳米银材料加入上述混合溶液中,光催化反应,反应时间分别为15min、30min、45min、60min、75min,
其中S1中NaBH4溶液的浓度为0.03mol/L,S2中辣木籽提取物介导合成纳米银材料的浓度为400mg/L。
其中辣木籽提取物介导合成的纳米银材料通过辣木籽提取物和硝酸银合成得到,其中所述辣木籽提取物和硝酸银的质量比为100mg:7.64mg,具体合成方法与实施例1相同。
结果检测
本发明的辣木籽提取物介导合成纳米银材料对染料废水的处理效果通过检测亚甲基蓝(MB)和刚果红(CR)的光催化降解率来进行表征。
具体光催化降解活性检测如下:
将0.2mL AgNPs(1mg/mL)溶液与3mL含有NaBH4(25mM)的有机染料水溶液(15μM)混合,并通过UV-Vis光谱法监测吸光度的变化。最后,将反应混合物离心,并通过使用UV-Vis光谱在CR的490nm和MB的660nm下测量上清液的吸光度。
有机染料降解率(k)如下计算:
有机染料降解率(k)=[(A0-At)/A0]×100
k是有机染料的降解速率,A0是原始溶液的吸光度,At是随时的溶液吸光度。
检测了AgNP对MB和CR的催化活性。废水样品从中国广东省深圳市污水处理厂收集。
检测结果如下表1所示:

从上述表1的数据可以看出,将辣木籽提取物介导合成纳米银材料应用在染料废水处理中可以有较好的有机染料降解去除效果,尤其是对亚甲基蓝、刚果红和4-硝基苯酚的降解,均可以达到85~90左右的降解率,体现了辣木籽提取物作为生物介导合成纳米银材料的优势。
显然,本发明的上述实施例仅仅是为清楚地说明本发明所作的举例,而并非是对本发明的实施方式的限定。对于所属领域的普通技术人员来说,在上述说明的基础上还可以做出其它不同形式的变化或变动。这里无需也无法对所有的实施方式予以穷举。凡在本发明的精神和原则之内所作的任何修改、等同替换和改进等,均应包含在本发明权利要求的保护范围之内。

1.一种辣木籽提取物介导合成的纳米银材料在染料废水处理中的应用。
2.如权利要求1所述应用,其特征在于,所述染料废水为含有亚甲基蓝和/或刚果红和/或4-硝基苯酚的染料废水。
3.如权利要求2所述应用,其特征在于,所述染料废水处理的具体方法如下:
S1.将NaBH4溶液于染料废水混合;
S2.将辣木籽提取物介导合成纳米银材料加入上述混合溶液中,光催化反应,
其中S1中NaBH4溶液的浓度为0.01~0.18mol/L,S2中辣木籽提取物介导合成纳米银材料的浓度为20~6000mg/L。
4.如权利要求3所述应用,其特征在于,S1中NaBH4溶液的浓度为0.02~0.10mol/L。
5.如权利要求3所述应用,其特征在于,S2中辣木籽提取物介导合成纳米银材料的浓度为50~500mg/L。
6.如权利要求3所述应用,其特征在于,S2中所述光催化反应的反应时间为15~75min。
7.如权利要求1~6任意一下所述应用,其特征在于,所述辣木籽提取物介导合成纳米银材料为圆球形核壳结构,其内部为聚集的银离子,外部为覆盖的辣木籽提取物分子。
8.如权利要求7所述应用,其特征在于,所述辣木籽提取物介导合成纳米银材料的粒径范围为3.8~31.0nm,平均粒径为9.8nm。
9.如权利要求8所述应用,其特征在于,所述辣木籽提取物介导合成纳米银材料中粒径为11~15nm的纳米银颗粒占纳米银材料总颗粒数的30%,粒径为3.8~11nm的纳米银颗粒占纳米银材料总颗粒数的58%。
10.如权利要求7所述应用,其特征在于,所述辣木籽提取物介导合成纳米银化合物通过辣木籽提取物和硝酸银合成得到,其中所述辣木籽提取物和硝酸银的质量比为10:0.764。
