Abstract
本发明公开了一种筛选急性放射性胃肠综合征治疗靶点的方法,属于生物医药技术领域。本发明利用CreERT‑loxP条件过表达转基因小鼠模型,在大剂量电离辐射照射后,有效促进肠隐窝静止期干细胞增殖,从而筛选在照射后18‑24hr仍然具有救治效果的、针对放射性胃肠综合征的治疗靶点。CreERT‑loxP条件过表达转基因小鼠只有在注射了他莫昔芬之后,才会在特定细胞中发生基因剪切,从而调控基因表达。本发明利用这一特性,可以很好地模拟核事故发生后,先照射、后治疗的实际情况,从而更具现实意义,将筛选得到的治疗靶点开发成药物用于核事故救治,可以为核事故的救治赢得宝贵时间。

技术领域
本发明涉及一种筛选急性放射性胃肠综合征治疗靶点的方法,属于生物医 药技术领域。
背景技术
人体不同组织在受到放射性射线照射后,损伤的程度不尽相同。通常认为, 细胞对辐射的敏感程度与细胞的增殖速度正相关,与细胞的分化程度负相关。 生理状态下,人体的肠上皮在肠道干细胞的增殖、驱动下快速新陈更替,由于 小肠隐窝干细胞在生理状态下处于高速增殖的状态,因此,其极易遭受辐射损 伤,从而失去原有的增殖、分裂能力。干细胞分裂的停滞将导致肠上皮失去细 胞更新的来源,从而导致肠上皮的完整性遭受严重破坏,肠绒毛断裂、脱落, 失去原有的屏障与吸收功能。小鼠腹部受到剂量大于15Gy的X射线(或γ射 线)照射,会在照射后一周内出现肠上皮损伤导致的严重腹泻、营养吸收障碍、体重减轻等症状,并在照射后10天内死于上述症状导致的放射性胃肠综合征。 可见,在大剂量的事故性照射条件下,人体的肠道组织是遭受辐射损伤的重要 靶组织。
尽管照射前预防性给药可以在一定程度上减轻辐射导致的干细胞损伤与肠 上皮破坏,但核事故与恐怖袭击通常具有不可预见性,而现有的照射后治疗措 施均难以逆转辐射导致的肠道干细胞不可逆死亡(因为增殖迅速的肠道干细胞 对电离辐射特别敏感,在照射后6-12hr就会不可逆地进入细胞凋亡程序,此时 大多数药物甚至还来不及在这些敏感的细胞内发挥作用),从而无法有效救治放 射性胃肠综合征患者。因此,迫切需要开发出可用于照射后救治的全新的治疗 策略。
随着精准医学的发展与科学界对肠隐窝干细胞研究的深入,发现肠隐窝中 除了增殖快速的干细胞群体(Lgr5+干细胞)外,还存在了一小群在生理状态下 处于“静止”状态的干细胞,“隐窝静止期干细胞”。这类干细胞在生理状态下增 殖十分缓慢,并不负责维持肠上皮的新陈更替,由于增殖缓慢,它们具有相对 较高的辐射抵抗性。在化学毒物或电离辐射引起Lgr5+干细胞大量死亡的条件 下,这类干细胞具有潜在的增殖能力,可以在一定程度上代替Lgr5+干细胞增殖、 分化为肠绒毛上皮细胞。但是在大剂量电离辐射导致的损伤条件下,肠上皮的 崩解往往发生在照射后3天之内,而隐窝静止期干细胞有限的增殖能力并不足 以在短时间内逆转肠上皮完整性的破坏,动物在照射后7-10天依然会发生死亡。因此,在电离辐射后短时间内加速隐窝静止期干细胞的增殖是极为重要的救治 手段,目前还未见针对隐窝静止期干细胞的放射性胃肠综合征救治措施,迫切 需要基础研究提供可靠的理论与实验依据。
在医学研究中,基于动物实验的研究结果通常较体外细胞实验更贴近人体 的实际情况,因而更具有科学意义和实用价值。但是,从动物伦理与实验成本 的角度考虑,动物实验不适宜用于大规模的药物筛查(成千上万种药物)和有 效治疗靶点的筛选。相反,基于一定理论基础的、针对特定基因的转基因动物 模型更具有疾病治疗的针对性和目的性。目前与放射性胃肠综合征研究相关的 转基因动物模型与其他现有技术主要包括:
1、研究发现p53基因依赖的PUMA(p53 upregulated modulator of apoptosis)会通过线粒体途径介导辐射后肠上皮细胞的凋亡。采用PUMA基因缺失型小鼠 (普通的敲基因小鼠)则呈现出对大剂量电离辐射的耐受性,并对肠隐窝Lgr5+干细胞具有一定保护效果。由于采用的是普通的敲基因小鼠,所以无法实现在 小鼠照射后对基因进行干预。
2、研究发现TLR3基因缺失的小鼠也可以抵抗大剂量电离辐射导致隐窝细 胞死亡及肠道损伤。作用机制上,p53依赖型细胞死亡会释放细胞内RNA,并 通过TLR3介导细胞凋亡。这一研究提示使用TLR3/dsRNA复合体抑制剂具有 缓解放射性胃肠综合征的潜在作用。同样的,由于采用的是普通的敲基因小鼠, 所以无法实现在小鼠照射后对基因进行干预,导致该研究结果无法预测照射后 干预TLR3对放射性胃肠综合征的救治效果,仅仅只能预测照射前干预TLR3 对放射性胃肠综合征的预防作用。
3、采用敲基因小鼠模型研究发现,当小鼠缺失双链脱氧核糖核苷酸(dsDNA) 损伤的感受器AIM2(Absent in melanoma 2)后,可有效缓解放射性胃肠综合征。 其肠道保护机制在于AIM2可参与招募Caspase-1使其活化,诱导隐窝干细胞发 生细胞焦亡,且此过程不依赖Caspase-3和Caspase-7相关的凋亡信号通路。同 样的,由于采用的是普通的敲基因小鼠,所以无法实现在小鼠照射后对基因进 行干预。
4、2019年研究团队发现过表达的URI(Unconventional prefoldin RPB5interactor)蛋白可以保护小鼠免受辐射引起的胃肠道综合征。而URI正常表达水 平的小鼠,则有高达70%的死亡率,完全敲除URI基因则会使小鼠全部死于放 射性胃肠综合征。URI蛋白的保护机制在于,其主要存在于肠隐窝静止期干细 胞群体中,该群体较慢的增殖速度使其免于受到辐射损伤。但当URI被敲除后, 原本被URI抑制的β-catenin-c-MYC信号通路则会被激活,细胞迅速增殖的同 时也更容易受到辐射损伤,进而导致小鼠死于放射性胃肠综合征。本研究尽管 研究了肠隐窝静止期干细胞,但是未进行照射后基因干预,所以无法预测辐射 救治中的有效性。
然而,在普通的敲基因小鼠或基因过表达小鼠中,目的基因已处在稳定的 敲除或过表达状态,无法在照射后再对基因进行表达调控。目前较为先进和成 熟的转基因动物模型是CreERT-loxP条件过表达小鼠模型,但是,尚未见利用CreERT-loxP条件过表达小鼠模型进行肠道辐射损伤救治的相关研究。
发明内容
为解决上述问题,本发明提供一种筛选急性放射性胃肠综合征治疗靶点的 方法。利用CreERT-loxP条件过表达转基因小鼠模型,在大剂量电离辐射照射 后,有效促进肠隐窝静止期干细胞增殖,从而筛选在照射后18-24hr仍然具有 救治效果的、针对放射性胃肠综合征的治疗靶点。
本发明的第一个目的是提供一种筛选急性放射性胃肠综合征治疗靶点的方 法,包括如下步骤:采用CreERT-loxP条件过表达小鼠模型,进行大剂量电离 辐射照射,筛选促进肠隐窝静止期干细胞增殖的治疗靶点。
进一步地,所述的CreERT-loxP条件过表达小鼠模型包括Bmi1-CreERT -loxP条件过表达小鼠模型。
进一步地,所述的方法具体包括如下步骤:
S1、将候选治疗靶点的基因插入loxP-STOP-loxP序列下游,将构建的序列 插入小鼠基因组中,构建带有候选治疗靶基因的loxP小鼠;
S2、将带有候选治疗靶基因的loxP小鼠与CreERT-loxP条件过表达小鼠模 型进行合笼繁育,筛选带有候选治疗靶基因的CreERT-loxP条件过表达小鼠作 为实验组小鼠,以带有候选治疗靶基因的loxP小鼠作为对照组小鼠;
S3、采用大剂量电离辐射对实验组小鼠和对照组小鼠进行照射处理;
S4、照射后立即注射雌激素类似物,评价辐射救治效果,筛选促进肠隐窝 静止期干细胞增殖的治疗靶点。
本发明对照组小鼠和实验组小鼠均需要注射他莫昔芬,只是对照组小鼠体 内缺乏重组酶,所以即使注射他莫昔芬也无法诱导基因剪切,无法诱导治疗靶 基因过表达。
进一步地,在S1步骤中,将构建的序列插入小鼠基因组的H11或者ROSA26 位点。在本发明中,选用上述两个基因位点是常用的进行基因编辑的位点,在 这两个位点插入基因编辑的片段不易对原有的其他基因产生影响。
进一步地,所述大剂量电离辐射剂量为15-18Gy,剂量率为0.5-10Gy/min, 照射范围为全腹照射。
进一步地,所述的雌激素类似物为他莫昔芬。
进一步地,所述的他莫昔芬的注射剂量为4-5mg/20g小鼠体重。
进一步地,所述的辐射救治效果通过肠隐窝干细胞增殖效果及小鼠存活情 况进行评价。
进一步地,所述的肠隐窝干细胞增殖效果是照射后3-5天的肠隐窝干细胞 增殖情况。
进一步地,所述的小鼠存活率是照射后30天内小鼠的存活率。
本发明的有益效果:
本发明利用CreERT-loxP条件过表达转基因小鼠模型,在大剂量电离辐射 照射后,有效促进肠隐窝静止期干细胞增殖,从而筛选在照射后18-24hr仍然 具有救治效果的、针对放射性胃肠综合征的治疗靶点。CreERT-loxP条件过表达 转基因小鼠只有在注射了他莫昔芬之后,才会在特定细胞中发生基因剪切,从 而调控基因表达。本发明利用这一特性,可以很好地模拟核事故发生后,先照 射、后治疗的实际情况,从而更具现实意义,将筛选得到的治疗靶点开发成药 物用于核事故救治,可以为核事故的救治赢得宝贵时间。

具体实施方式
下面结合附图和具体实施例对本发明作进一步说明,以使本领域的技术人 员可以更好地理解本发明并能予以实施,但所举实施例不作为对本发明的限定。
Cre-loxP条件过表达动物模型的原理图如图1所示,在图1中,并没有标 明与诱导表达密切相关的雌激素受体(ER或ERT)。当Cre重组酶与雌激素受 体结合时,无法进入细胞核完成切割;只有当注射他莫昔芬等药物后,才能解 除Cre重组酶与雌激素受体的结合,从而完成基因剪切。正是利用上述特性, 才能实现在电离辐射后进行基因的诱导与调控。
本发明实施例中采用Bmi1-CreERT;loxP条件过表达小鼠为例,主要是由于 将编码重组酶Cre的基因按插在肠隐窝静止期干细胞特异性启动子(如Bmi1) 的下游,可以特异性地调控肠隐窝静止期干细胞内的基因,在时空特异性上最 大程度地模拟事故性照射后的治疗干预,从而对辐射损伤后促进肠隐窝静止期 干细胞增殖的基因进行有效的筛查。
实施例1:CreERT-loxP条件过表达转基因小鼠构建
为了有效促进隐窝静止期干细胞的增殖,本技术方案采用 Bmi1-CreERT;loxP条件过表达转基因小鼠,对小鼠隐窝静止期干细胞进行基因 干预,选用TIGAR作为目的基因,通过他莫昔芬诱导隐窝静止期干细胞内 TIGAR蛋白的表达,如图2所示。
在具有上述基因表型的小鼠中,通过Bmi1这一隐窝静止期干细胞特异性 的启动子,可以使TIGAR仅仅在隐窝静止期干细胞内过表达。
上述小鼠的书写方式为:Bmi1-creERT;H11-Tigar。具体来讲,Bmi1是隐窝 静止期干细胞的特异性启动子。cre是编码重组酶的基因,可以翻译成为重组酶, 从而对特定的基因序列进行切割。ERT编码雌激素受体,当creERT作为一个 整体被翻译后,重组酶与雌激素受体绑定,无法进入细胞核完成DNA剪切, 所以需要通过注射他莫昔芬将雌激素受体与重组酶解离,解离后的重组酶便可 以进入细胞核对特定DNA序列(loxP序列)进行剪切。剪切的结果是导致两 个loxP位点之间的基因序列从DNA序列中被移除,余下的两个断端拼接后变 成新的完整的DNA序列。就Bmi1-creERT;H11-Tigar小鼠而言,剪切的结果是: 导致原来位于Tigar上游的STOP位点(polyA)从DNA序列里移除,原来不 能被转录的Tigar基因(由于上游STOP位点的存在)在剪切后可以被转录、进 而被翻译,从而导致Tigar蛋白表达增加。从腹腔注射他莫昔芬到实现TIGAR 蛋白过表达需要18-24hr。
Tigar下游的EGFP基因可以翻译为绿色荧光蛋白起到示踪的作用,Tigar 与EGFP之间的2A是连接体,保证TIGAR与EGFP在翻译后不会相互融合而 导致蛋白质空间结构破坏、失去功能。由于Bmi1这一肠隐窝静止期干细胞特 异性启动子的存在,上述整个切割过程仅仅发生在肠隐窝静止期干细胞内。综 上所述,通过上述动物模型,可以实现在照射后18-24hr完成隐窝静止期干细 胞内TIGAR蛋白的过表达。(照射后即刻注射他莫昔芬)
并且只要更换Tigar基因的序列,替换为其他具有潜在治疗价值的基因, 便可以实现对急性放射性胃肠综合征治疗靶点的筛选,当然上述治疗靶点是针 对肠隐窝静止期干细胞的。
实施例2:电离辐射照射后诱导目的基因过表达
由于从药物注射,到TIGAR在隐窝静止期干细胞内过表达需要一定的时间 (对于CreERT-loxP动物模型而言,通常是18-24hr),所以我们在小鼠受到15 Gy X射线全腹照射(图3)后即刻进行药物腹腔注射(他莫昔芬,单次注射, 4.5mg/20g小鼠体重)。
小鼠照射后第1、3、5天,处死小鼠、将肠组织制成冰冻切片,观察TIGAR 蛋白在隐窝静止期干细胞内的表达情况,如图4所示。由于在转基因小鼠设计 与构建过程中,TIGAR与绿色荧光蛋白(EGFP)会同时表达,所以,可以用 绿色荧光蛋白的表达程度来指示TIGAR的表达水平。在照射后1天,可见隐窝 内只有1-2个绿色细胞,即隐窝静止期干细胞被成功过表达。由于隐窝静止期 干细胞具有分裂增殖能力,在照射后3天、5天可以形成大量子代细胞,且隐 窝静止期干细胞的子代细胞也具有绿色荧光,所以绿色荧光不仅反应了TIGAR蛋白的过表达状态,绿色荧光阳性的细胞也反应了隐窝静止期干细胞的子代细 胞。图中DAPI荧光用来指示细胞核,帮助更好地进行细胞定位与细胞计数。 可见,照射后3-5天,重建的隐窝几乎有隐窝静止期干细胞的子代细胞构成, 表明TIGAR过表达促进了隐窝静止期干细胞的增殖并加速了照射后隐窝的重 建。
实施例3:辐射救治效果的评价
为了评价TIGAR过表达的辐射救治效果,采用小鼠生存率、肠道组织切片 HE染色等方法进行评估。在生存率实验中,对照组小鼠(将Tigar基因插入 loxP-STOP-loxP序列下游,获得loxP-STOP-loxP-Tigar序列,将上述序列插入 小鼠基因组的H11位点,获得H11-Tigar小鼠)与肠隐窝静止期干细胞TIGAR 过表达小鼠均接受15Gy X射线全腹照射(图3),照射后即刻进行他莫昔芬腹 腔注射(单次注射,4.5mg/20g小鼠体重)。注射后继续饲养小鼠,观察小鼠的 存活情况,如图5所示。
可见,对照组小鼠(H11-Tigar小鼠,WT)在照射后7天全部死于放射性 胃肠综合征(生存率0%),而肠隐窝静止期干细胞TIGAR过表达组 (Bmi1-creERT;H11-Tigar)小鼠在照后30天依然有接近40%的生存率,救治效 果明显(p<0.01)。
在肠道组织切片HE染色实验中(如图6所示),在照射后1、3、5天分别 处死小鼠收获肠组织,制成组织切片并进行HE染色。可见,随着照射后肠隐 窝静止期干细胞的增殖,照射后3天、5天肠隐窝静止期干细胞TIGAR过表达 组小鼠肠道隐窝数目与隐窝大小均显著大于对照组小鼠,表明肠隐窝静止期干 细胞TIGAR过表达的确促进了照射后肠隐窝的增殖与重建,而作为肠绒毛更新 的来源,肠隐窝能否及时重建对于放射性胃肠综合征的救治具有决定性意义。
这里特别说明:尽管实验中在电离辐射后即刻进行了他莫昔芬腹腔注射, 但由于他莫昔芬诱导TIGAR在肠隐窝静止期干细胞内表达需要时间(18-24 hr),可以认为诱导后24hr,TIGAR才开始真正发挥作用。所以,证明了发明 目的中提到的:TIGAR在辐射暴露后24hr依然可以发挥促进肠隐窝干细胞增 殖、促进小鼠存活的作用。
以上所述实施例仅是为充分说明本发明而所举的较佳的实施例,本发明的 保护范围不限于此。本技术领域的技术人员在本发明基础上所作的等同替代或 变换,均在本发明的保护范围之内。本发明的保护范围以权利要求书为准。
Claims (10)
1.一种筛选急性放射性胃肠综合征治疗靶点的方法,其特征在于,包括如下步骤:采用CreERT-loxP条件过表达小鼠模型,进行大剂量电离辐射照射,筛选促进肠隐窝静止期干细胞增殖的治疗靶点。
2.根据权利要求1所述的方法,其特征在于,所述的CreERT-loxP条件过表达小鼠模型包括Bmi1-CreERT-loxP条件过表达小鼠模型。
3.根据权利要求1所述的方法,其特征在于,所述的方法具体包括如下步骤:
S1、将候选治疗靶点的基因插入loxP-STOP-loxP序列下游,将构建的序列插入小鼠基因组中,构建带有候选治疗靶基因的loxP小鼠;
S2、将带有候选治疗靶基因的loxP小鼠与CreERT-loxP条件过表达小鼠模型进行合笼繁育,筛选带有候选治疗靶基因的CreERT-loxP条件过表达小鼠作为实验组小鼠,以带有候选治疗靶基因的loxP小鼠作为对照组小鼠;
S3、采用大剂量电离辐射对实验组小鼠和对照组小鼠进行照射处理;
S4、照射后立即注射雌激素类似物,评价辐射救治效果,筛选促进肠隐窝静止期干细胞增殖的治疗靶点。
4.根据权利要求3所述的方法,其特征在于,在S1步骤中,将构建的序列插入小鼠基因组的H11或者ROSA26位点。
5.根据权利要求3所述的方法,其特征在于,所述大剂量电离辐射剂量为15-18Gy,剂量率为0.5-10Gy/min,照射范围为全腹照射。
6.根据权利要求3所述的方法,其特征在于,所述的雌激素类似物为他莫昔芬。
7.根据权利要求6所述的方法,其特征在于,所述的他莫昔芬的注射剂量为4-5mg/20g小鼠体重。
8.根据权利要求3所述的方法,其特征在于,所述的辐射救治效果通过肠隐窝静止期干细胞增殖效果及小鼠存活率进行评价。
9.根据权利要求8所述的方法,其特征在于,所述的肠隐窝静止期干细胞增殖效果是照射后3-5天的肠隐窝静止期干细胞增殖情况。
10.根据权利要求8所述的方法,其特征在于,所述的小鼠存活率是照射后30天内小鼠的存活率。
