Description

Method for improving conception rate of folliculofloxacin cows
Technical Field
The invention relates to the technical field of livestock breeding obstacle control, in particular to a method for improving the conception rate of folliculofloxacin cows.
Background
The non-ruptured folliculitis flaviviridae syndrome (LUFS) is a common infertility of women of childbearing age, mainly refers to the process that dominant follicles are mature but not ruptured, oocytes are not discharged and luteinize in situ to form corpus luteum and secrete progesterone, and is clinically mainly manifested by normal menstrual cycle, similar ovulation manifestation but continuous infertility and a special type of anovulatory menstruation, but the LUFS is not reported at present on cows.
In the autumn of 2018, a B-type ultrasonic instrument is used by an animal breeding research laboratory of agriculture and forestry academy of sciences in Beijing suburban areas to successfully diagnose the LUFS of a multi-head cow for the first time. LUFS causes the delay of pregnancy of cows, prolongs the interval time of calving, and causes poor milk quality, so that the cows are eliminated, and great economic loss is caused to farmers. According to the current Chinese milk consortium report, extending the calving interval of one day will result in an economic loss of about 50 yuan, so that one estrus (21 days) will result in a loss of at least 1000 yuan.
As the cow has few reports of folliculofloxacin, no relevant treatment scheme is found. How to recover the function of the ovarian follicle flavonized and improve the breeding efficiency of the dairy cows is a technical problem which needs to be solved urgently.
Disclosure of Invention
The first purpose of the invention is to provide a method for improving the conception rate of the folliculofloxacin cows;
the second purpose of the invention is to provide a medicament combination for improving the conception rate of the folliculofloxacin cows.
The study of the invention finds that the conception rate is improved after LRH-A3 treats the cow with luteinized follicle, and the invention is completed. Specifically, the technical scheme of the invention is as follows:
1. a cow diagnosed as folliculofloxacin by B ultrasonic was given an intramuscular injection of LRH-A325. mu.g for three consecutive days. The function is as follows: stimulates the release of the luteinized follicle, and is beneficial to the formation of a new periodic corpus luteum and the normal development of the follicle wave in the next period.
After 2.24 days, 100 μ g of gonadotropin releasing hormone (GnRH) was injected intramuscularly. The effect is to stimulate the development of small follicles in the ovary and initiate the first follicular wave.
After 3.31 days, 0.6mg of chloroprostenol (PGF2a) was intramuscularly injected. The function is as follows: induce ovarian luteolysis.
After 4.33 days, 100. mu.g of gonadotropin releasing hormone (GnRH) was injected intramuscularly. The function is as follows: promoting the maturation of dominant follicles and timing ovulation.
5. The interval is 16-18 hours, and the semen is infused regularly.
Further, the invention provides a medicament combination for improving the conception rate of the folliculofloxacin cows, which comprises LRH-A3, GnRH and PGF2a which are stored separately.
Preferably, each dose of combination comprises 325-50 mug LHR-A, 100-500 ng GnRH and 0.6-1.8 mg PGF2a0.6-1.8 mg.
More preferably, each combination dose comprises 325 μ g LHR-A, 200ng GnRH, 0.6mg PGF2a 0.6.
The method for improving the conception rate of the luteinized ovarian cows is simple and easy to operate, and high-quality ova are generated by ovulation of the luteinized ovarian follicles and normal cycle recovery of ovaries, so that the conception rate is improved, the breeding efficiency of the cows is improved, the calving interval is shortened, the cost is reduced, and the economic benefit is increased, thereby having good application prospect.
Drawings
FIG. 1 is a flow chart of the operation of the method of the present invention.
The specific implementation mode is as follows:
the following examples are intended to further illustrate the invention but should not be construed as limiting it. Modifications and substitutions may be made thereto without departing from the spirit and scope of the invention. LHR-A3, GnRH and PGF2a were all available from Sanbo pharmaceutical Co., Ltd.
Example 1 diagnosis of folliculofloxacin in cows
A diagnostic instrument: b ultrasonic instrument for animals (Japanese Honda HS-1600 type).
The diagnosis method comprises the following steps:
for lactating cows with long-term infertility, rectum is adopted to grasp B ultrasonic diagnosis, the frequency of B ultrasonic diagnosis is 7.5MHz, and the abnormal situation of ovaries is observed through a display screen.
And (3) diagnosis results:
on the day of oestrus and 2d later, the ovaries were well-defined and morphologically normal. Mature follicles exist on the ovary, but the follicles are oblate or irregular, the size is 19-24 mm, follicular fluid is not full, the follicular membrane is strong echo, the thickness is 1-2 mm, the inner wall of the follicles is not smooth, and punctate strong echoes are more in the follicular cavity. After 2d, follow-up examination is carried out again, and the same results of B-ultrasonic examination of the ovarian follicles of the oestrous cows are found.
Example 2 method for increasing conception rate of folliculofloxacin cows
Test cows: example 1 the diagnosed folliculofloxacin cows.
The treatment method comprises the following steps:
control group:
the diagnosis of folliculofloxacin cattle by B ultrasonic is only without any treatment.
The method A comprises the following steps:
a. intramuscular injection of gonadotropin releasing hormone (GnRH)100 μ g;
b.7 days later, 0.6mg of chloroprostenol (PGF2a) was intramuscularly injected;
c.9 days later, 100. mu.g gonadotropin releasing hormone (GnRH) was injected intramuscularly;
d. the interval is 16-18 hours, and the semen is infused regularly.
The method B comprises the following steps:
a. intramuscular injection of 325. mu.g of LRH-A to a cow diagnosed as folliculofloxacin by type-B ultrasonography for three consecutive days;
b.24 days later, 100. mu.g gonadotropin releasing hormone (GnRH) was injected intramuscularly;
c.31 days later, 0.6mg of chloroprostenol (PGF2a) was intramuscularly injected;
d.33 days later, 100 μ g gonadotropin releasing hormone (GnRH) was injected intramuscularly;
e. the interval is 16-18 hours, and the semen is infused regularly.
The method C comprises the following steps:
a. intramuscular injection of 325. mu.g of LRH-A to a cow diagnosed as folliculofloxacin by type-B ultrasonography for three consecutive days;
b. the last injection is performed at an interval of 16-18 hours, and semen is infused regularly.
The days mentioned above, except the last injection for regular insemination, refer to the number of days counted from the first injection for intramuscular injection.
Example 3 semen thawing, semen deposition method and early pregnancy diagnosis
1) Taking out 0.25mL of tubule frozen semen (sex-controlled frozen semen purchased from Beijing Dairy cow center) from the liquid nitrogen, and placing in a 38 ℃ water bath kettle for thawing for 30 seconds;
2) cutting off the thin tube seal, adding the frozen semen into a semen delivery gun, and preparing for semen delivery;
3) insemination method
Strictly disinfect the vulva of a cow. Adopts the deep insemination of the uterus, and the insemination part is required to be 2-5 cm deep in the uterine horn at the ovulation side, thus ensuring that enough sperms reach the insemination part of the ovum. And the effects of 'slight insertion, slow pushing and slow discharge' are achieved during insemination so as to avoid the damage to the uterus of the cow.
4) Early pregnancy diagnosis
And collecting 5ml of blood 28 days after the mating, separating serum, detecting the conception condition of the cow by using an Aide visual early pregnancy detection kit, and counting the result.
TABLE 1 conception rate after luteinizing follicular treatment of cows
Figure BDA0002395295590000041
The results show that: compared with the group A and the group C, the group B (LRH-A3+ GnRH + PG + GnRH) obviously improves the conception rate of the folliculoflavated cows after mating.
And (4) conclusion: from the results, the combined use of LRH-A3+ GnRH + PG + GnRH can improve the conception rate of the cow after insemination.
It will be appreciated by those skilled in the art that although LRH-A3 is used in an amount of 25 μ g in the present example, LRH-A3, preferably in an effective amount of 325-100 μ g, will have an effect once applied to folliculofloxacin cows.
